AbstractThe mechanical properties of the graphite electrode will dynamically change during the charge-discharge cycle, which will affect the propagation characteristics of guided waves in lithium-ion batteries. Meanwhile, lithium-ion batteries experience fluctuations in operating temperature during cycling, which will also influence the propagation of guided waves. This research aims to investigate the guided wave propagation problem of porous electrode material with electrolyte versus temperature. The problem is under the framework of Green-Naghdi theory and Biot theory, and a numerical approach is presented to solve the thermoelastic wave propagation characteristics in such a multi-layered porous electrode. A frequency domain simulation for a multi-layered porous anode will be established. The simulation results confirm the feasibility and accuracy of the proposed approach. The porosity of the anode material shows obvious dynamic variation during cycling, which illustrates a strong correlation with the elastic modulus of the solid skeleton. Then, the effects of porosity and temperature variations on the propagation characteristics of thermoelastic guided waves in multi-layered porous graphite electrodes will be analyzed in detail. The phase velocity of fundamental modes appears regularly shift in the low frequency range. This may provide theoretical support for the acoustic nondestructive characterization of the operating states of the lithium-ion batteries. [ABSTRACT FROM AUTHOR]

Introduction: Pulmonary embolism (PE) is among the most severe cardiovascular disorders worldwide. Timely and appropriate diagnosis of PE remains an important step in reducing PE related mortality and morbidity. Methods: In this retrospective single‐center cohort study, we comprehensively compared the screening performances of several clinical scoring systems (Wells score [WS], Revised Geneva score [RGS], WS + d‐Dimer [D‐D], RGS + D‐D, WS + PE rule‐out criteria [PERC] and RGS + PERC) among PE suspected patients. Failure rates across different PE severity grades as well as overall sensitivity/specificity were considered in evaluating each screening strategy. Results: A total of 3437 patients were included in this study and 698 of them were diagnosed with PE. Patients with and without PE were similar in demographics, while significantly different in respiration‐related characteristics. Compared with WS or RGS alone, Integrating PERC or D‐D with WS or RGS significantly decreased the failure rates across all PE severity grades, and increased the overall sensitivity from 88.5% and 87.2% to 96.3% and 94.8% (D‐D) to 99.4% and 99.6% (PERC), respectively. However, compared with other four scoring approaches, using WS or RGS alone increased the specificity from 8.3% and 7.2%, 38.3% and 21.3%, to 63.5% and 34.8%, respectively, and increased the AUC from 0.54 to 0.54, 0.70 and 0.69, to 0.8 and 0.76, respectively. In general, all screening approaches achieved better performances among PE patients with respiratory distress compared to those without respiratory distress. Conclusion: Combining PERC or D‐D with WS or RGS, and the presence of respiratory distress provide significantly better PE rule‐out performances. [ABSTRACT FROM AUTHOR]

The common terminology criteria for adverse events by the National Cancer Institute has greatly facilitated the revolution of drug development and an increasing number of Phase I trials have started to collect multiple-grade toxicity endpoints. Appropriate and yet transparent Phase I statistical designs for multiple-grade toxicities are therefore in great needs. In this article, we propose a quasi-toxicity probability interval (qTPI) design that incorporates a quasi-continuous measure of the toxicity probability (q T P) into the Bayesian theoretic framework of the interval based designs. Multiple-grade toxicity outcomes of each patient are mapped to q T P according to a severity weight matrix. Dose–toxicity curve underlying the dosing decisions in the qTPI design is continuously updated using accumulating trial data. Numerical simulations investigating the operating characteristics of qTPI show that qTPI achieved better safety, accuracy and reliability compared to designs that rely on binary toxicity data. Furthermore, parameter elicitation in qTPI is simple and does not involve multiple hypothetical cohorts specification. Finally, a hypothetical soft tissue sarcoma trial with six toxicity types and grade 0 to grade 4 severity grades is illustrated with patient-by-patient dose allocation under the qTPI design. Administration of glucocorticoids induces ocular hypertension in some patients. If untreated, these patients can develop a secondary glaucoma that resembles primary open-angle glaucoma (POAG). The underlying pathology of glucocorticoid-induced glaucoma is not fully understood, due in part to lack of an appropriate animal model. Here, we developed a murine model of glucocorticoid-induced glaucoma that exhibits glaucoma features that are observed in patients. Treatment of WT mice with topical ocular 0.1% dexamethasone led to elevation of intraocular pressure (IOP), functional and structural loss of retinal ganglion cells, and axonal degeneration, resembling glucocorticoid-induced glaucoma in human patients. Furthermore, dexamethasone-induced ocular hypertension was associated with chronic ER stress of the trabecular meshwork (TM). Similar to patients, withdrawal of dexamethasone treatment reduced elevated IOP and ER stress in this animal model. Dexamethasone induced the transcriptional factor CHOP, a marker for chronic ER stress, in the anterior segment tissues, and Chop deletion reduced ER stress in these tissues and prevented dexamethasone-induced ocular hypertension. Furthermore, reduction of ER stress in the TM with sodium 4-phenylbutyrate prevented dexamethasone-induced ocular hypertension in WT mice.
