The γ-tubulin ring complex (γTuRC) is the principal nucleator of cellular microtubules, and the microtubule-nucleating activity of the complex is stimulated by binding to the γTuRC-mediated nucleation activator (γTuNA) motif. The γTuNA is part of the centrosomin motif 1 (CM1), which is widely found in γTuRC stimulators, including CDK5RAP2. Here, we show that a conserved segment within CM1 binds to the γTuNA and blocks its association with γTuRCs; therefore, we refer to this segment as the γTuNA inhibitor (γTuNA-In). Mutational disruption of the interaction between the γTuNA and the γTuNA-In results in a loss of autoinhibition, which consequently augments microtubule nucleation on centrosomes and the Golgi complex, the two major microtubule-organizing centers. This also causes centrosome repositioning, leads to defects in Golgi assembly and organization, and affects cell polarization. Remarkably, phosphorylation of the γTuNA-In, probably by Nek2, counteracts the autoinhibition by disrupting the γTuNA‒γTuNA-In interaction. Together, our data reveal an on-site mechanism for controlling γTuNA function.

Natural gas pipelines are an essential part of the economy. Natural gas pipelines may leak after aging, strong vibration signals may be generated in the pipeline when leakage occurs, and vibration signals may be noisy. Traditional variational mode decomposition (VMD) noise reduction methods need to set parameters in advance, and so may not achieve the best decomposition effect. To solve this problem, this paper proposes a method for pipeline leakage location based on the sparrow search algorithm (SSA) optimization of VMD combined with generalized quadratic cross-correlation. The method first calculates the original signal-to-noise ratio (SNR), and if the SNR is low, wavelet threshold denoising is used to process the signal. Then, SSA optimization is used to refine the two key parameters of VMD (penalty parameter α and mode decomposition number K) based on sample entropy. Subsequently, the signal undergoes decomposition into K intrinsic mode function (IMF) components through VMD according to the obtained analysis parameter combination. Then, the IMF components are screened to obtain the reconstructed signal. Finally, the noise reduction signal is obtained. The signal delay after noise reduction is obtained through a generalized quadratic cross-correlation and the accurate leakage position is obtained using the delay. Experiments showed that the minimum relative error of this method could reach 0.6%, which was more accurate than the traditional VMD method, and effectively improved the accuracy of noisy signals in pipeline leakage locations. Pond culture is the primary method for cultivating Chinese mitten crab (Eriocheir sinensis), with phytoplankton significantly influencing their growth. Green algae benefit crab growth by serving as supplementary food, while cyanobacteria, particularly during blooms, hinder it and pose health risks. Environmental changes in nutrient levels, temperature, and light significantly affect phytoplankton communities in ponds, impacting both ecosystem stability and crab growth. However, there is a limited understanding regarding the patterns of phytoplankton changes within adult Chinese mitten crab culture ponds. This study conducted monthly collection and analysis of phytoplankton throughout the culture cycle in typical adult Chinese mitten crab culture ponds, concurrently measuring physical and chemical water parameters to explore the correlation between phytoplankton changes and environmental factors. The results revealed distinct seasonal variations in phytoplankton composition. Chlorophyta and Bacillariophyta, such as Chlorella, Pediastrum, and Cocconeis, predominated in spring, while Chlorophyta and cyanobacteria, such as Volvox, Anabaena, and Microcystis, dominated in summer, and cyanobacteria and Bacillariophyta, such as Microcystis, Dolichospermum, and Cocconeis, prevailed in autumn. Total phytoplankton density consistently increased throughout the culture period. Microcystis constituted the majority of cyanobacteria biomass throughout most months. Although the total phytoplankton biomass fluctuated, cyanobacteria biomass consistently rose each month, reaching a peak of 61.7 mg/L in October. Water temperature and pH emerged as the primary environmental drivers influencing changes in phytoplankton community structure. Cyanobacteria density reached its peak between 18 and 26 °C and at a pH range of 7.5–8.5. These findings highlight the need for environmental regulation and cyanobacteria control in Chinese mitten crab culture ponds, thus promoting the health and sustainability of the Chinese mitten crab culture.
