2. Synthia's Melody: A Benchmark Framework for Unsupervised Domain Adaptation in Audio

Abstract

Despite significant advancements in deep learning for vision and natural language, unsupervised domain adaptation in audio remains relatively unexplored. We, in part, attribute this to the lack of an appropriate benchmark dataset. To address this gap, we present Synthia's melody, a novel audio data generation framework capable of simulating an infinite variety of 4-second melodies with user-specified confounding structures characterised by musical keys, timbre, and loudness. Unlike existing datasets collected under observational settings, Synthia's melody is free of unobserved biases, ensuring the reproducibility and comparability of experiments. To showcase its utility, we generate two types of distribution shifts-domain shift and sample selection bias-and evaluate the performance of acoustic deep learning models under these shifts. Our evaluations reveal that Synthia's melody provides a robust testbed for examining the susceptibility of these models to varying levels of distribution shift.

Abstract

Mitochondrial dysfunction may precipitate intestinal dysfunction, while inflammatory bowel disease manifests as a chronic inflammatory ailment affecting the gastrointestinal tract. This condition disrupts the barrier function of the intestinal epithelium and alters metabolic products. Increasing mitochondrial adenosine triphosphate (ATP) synthesis in intestinal epithelial cells presents a promising avenue for colitis treatments. Nevertheless, the impact of cedrol on ATP and the intestinal barrier remains unexplored. Hence, this study is dedicated to examining the cedrol's protective effect on an inflammatory cocktail (IC)–induced intestinal epithelial barrier dysfunction in Caco‐2 cells. The finding reveals that cedrol enhances ATP content and the transepithelial electrical resistance value in the intestinal epithelial barrier. Moreover, cedrol mitigates the IC‐induced decrease in the messenger ribonucleic acid (mRNA) expression of tight junction proteins (ZO‐1, Occludin, and Claudin‐1), thereby ameliorating intestinal epithelial barrier dysfunction. Furthermore, nuclear magnetic resonance (NMR)–based metabolomic analysis indicated that IC‐exposed Caco‐2 cells are restored by cedrol treatments. Notably, cedrol elevates metabolites such as amino acids, thereby enhancing the intestinal barrier. In conclusion, cedrol alleviates IC‐induced intestinal epithelial barrier dysfunction by promoting ATP‐dependent proliferation of Caco‐2 cells and bolstering amino acid levels to sustain tight junction messenger ribonucleic acid expression. 6. Composition Characterization of Crossostephium chinense Leaf Essential Oil and Its Anti-Inflammatory Activity Mechanisms.

Abstract

This study investigates the composition characteristics and anti-inflammatory activity mechanisms of the essential oil from the leaves of Crossostephium chinense. C. chinense is a perennial herb commonly found in East Asia, traditionally used to treat various ailments. The essential oil extracted through water distillation, primarily contains 1,8-cineole (13.73%), santolina triene (13.53%), and germacrene D (10.67%). Three compounds were identified from the essential oil, namely 1-acetoxy-2-(2-hydroxypropyl)-5-methylhex-3,5-diene, 1-acetoxy-isopyliden-hex-5-en-4-one, and chrysanthemyl acetate, with the first two being newly discovered compounds. Then, the essential oil of C. chinense exhibits significant anti-inflammatory effects on RAW264.7 macrophages, effectively inhibiting the production of NO and ROS, with the IC50 value of 10.3 μg/mL. Furthermore, the essential oil reduces the expression of pro-inflammatory cytokines such as TNF-α, IL-6, and IL-1β. Mechanistic studies indicate that the essential oil affects the inflammatory response by inhibiting the expression of iNOS but has no significant impact on COX-2. Further analysis suggests that the essential oil may regulate the inflammatory response through the ERK protein in the MAPK pathway and IκBα in the NF-κB pathway, while also promoting the activity of the NRF2/HO-1 antioxidant pathway, enhancing the cell's antioxidant capacity, thereby achieving an effect of inhibiting the inflammatory response. 11. Studies on the effect of cryopreservation on gene expression in zebrafish blastomeres

Abstract

Cryopreservation is now a common practice in the fields of aquaculture, conservation and biomedicine. The success of cryopreservation is usually analysed in terms of cell survival, although there are other potential adverse effects that don’t necessarily result in cell death. These include DNA damage, which could result in altered gene expression. The aim of this study is to discover if cryopreservation has an impact on gene expression using zebrafish (Danio rerio) as the model organism. As the whole embryo cannot yet be successfully cryopreserved, isolated blastomeres from the embryos were used to investigate the impact of cryo-treatment on gene expression. This study sets out firstly to determine an optimum cryopreservation protocol for 50% epiboly blastomeres (Epiboly displaces the blastoderm margin to 50% of the distance between the animal and vegetal pole). Blastomeres had the highest survival level (70.2 ± 3.2%) when a mixture of 1.5 M dimethyl sulfoxide (DMSO) and 0.1 M sucrose were used as cryoprotectants. As quantitative analysis of gene expression using real-time PCR requires the use of a housekeeping gene as an internal control to normalize date, the second study aimed to identify and validate housekeeping genes for use in cryopreservation studies of zebrafish blastomeres. Seven potential housekeeping genes were analysed across a range of embryo stages and isolated blastomeres using the GeNorm and NormFinder software packages. Results indicated β-actin and EF1α housekeeping genes to be the most appropriate for cryopreservation studies on zebrafish embryos and blastomeres, and these housekeeping genes were used in the third study, the effect of cryopreservation on Pax gene expression. The results indicated that the trends (profile changes) in expression of Pax2a and Pax5 occurred to a lesser extent in frozen-thawed blastomeres than in fresh blastomeres whilst the opposite was true for Pax8. The trends in expression of Pax2b were delayed in frozen-thawed blastomeres compared to fresh blastomeres. Cryopreservation can therefore disrupt normal gene expression patterns in zebrafish embryos which could have a detrimental effect on embryo development. This is the first study on the stability of housekeeping and transcription factor genes in chilled and cryopreserved embryonic cells of the zebrafish. This work will significantly enhance future studies investigating the impact of cryopreservation on gene expression in zebrafish embryos.

Abstract

The uniform and smaller-sized (~3 μm) LiNi0.8Co0.1Mn0.1O2 (SNCM) particles are prepared via a fast nucleation process of oxalate co-precipitation, followed by a two-step calcination procedure. It is found that the fast nucleation by vigorous agitation enables us to produce oxalate nuclei having a uniform size which then grow into micron-particles in less than a few minutes. The impacts of solution pH, precipitation time, calcination temperature, and surface modification with ZrO2 on the structural, morphological, and electrochemical properties of SNCM are systematically examined to identify the optimal synthetic conditions. A novel bimodal cathode design has been highlighted by using the combination of the SNCM particles and the conventional large (~10 μm) LiNi0.83Co0.12Mn0.05O2 (LNCM) particles to achieve the high volumetric energy density of cathode. The volumetric discharge capacity is found to be 526.6 mAh/cm3 for the bimodal cathode L80% + S20%, whereas the volumetric discharge capacity is found to be only 480.3 and 360.6 mAh/cm3 for L100% and S100% unimodal, respectively. Moreover, the optimal bi-modal cathode delivered higher specific energy (622.4 Wh/kg) and volumetric energy density (1622.6 Wh/L) than the L100% unimodal (596.1 Wh/kg and 1402.1 Wh/L) cathode after the 100th cycle. Abstract

Chinese (N = 152) and White (N = 160) students reviewed a counseling interview transcript and provided perceptions on counselor credibility, counselor behaviors, and utilization intent. Utilization intent was defined as perceived willingness to use counseling after an initial hypothetical interview. In the proximal-distal model S. Sue and Zane (1987) proposed that achieving counselor credibility is more directly related to utilization intent than to culture-specific techniques, such as directiveness, when counselors are working with ethnic minority clients. For both ethnic samples, counselor credibility was the strongest predictor of utilization intent, and empathic involvement was the sole predictor of counselor credibility. Whereas the proximal-distal model was supported by the results obtained with the Chinese sample, a variant of this model best described the results for the White sample for which empathic involvement, as as well as counselor credibility, was found to be directly related to utilization intent.
