Abstract Background Excavation of key molecules can help identify therapeutic targets and improve the prognosis of pancreatic cancer. This study evaluated the roles of SUMO3 in cell viability, glycolysis, gemcitabine (GEM) sensitivity, and the antitumor activity of butyric acid (BA) in pancreatic cancer. Methods The mRNA and protein levels of SUMO3 were detected by qRT-PCR, Western blot, and immunohistochemical assay. SUMO3 was silenced or overexpressed in pancreatic cancer cells with or without Wnt/β-catenin pathway inhibitor, glycolysis inhibitor, GEM, or BA treatment. Cell viability was measured using the Cell Counting Kit-8 assay. Glycolysis was measured by determining the extracellular acidification rate, ATP level, and lactate content. Apoptosis was measured by flow cytometry, and TUNEL staining was used to examine in vitro and in vivo sensitivity to GEM chemotherapy. Luciferase reporter and chromatin immunoprecipitation assays were conducted to detect the binding of the SUMO3 promoter and NF-κB p65. Results SUMO3 was increased and associated with poor survival in pancreatic cancer. SUMO3 knockdown decreased cell viability and glycolysis in vitro and inhibited tumor growth in vivo. SUMO3 overexpression increased cell viability and glycolysis in vitro through the β-catenin pathway. SUMO3 knockdown increased GEM sensitivity, whereas SUMO3 overexpression decreased GEM sensitivity and inhibited the antitumor activity of BA. BA promoted histone acetylation and p-IκBα expression to inhibit NF-κB p65-mediated SUMO3 transcription. Conclusion SUMO3 acted as an active molecule in cell survival and growth by enhancing glycolysis in response to either GEM or BA. The mechanism was related to the constitutive IκBα/NF-κB/SUMO3/β-catenin signaling pathway.

Abstract The joint made of cast steel is frequently utilized within a treelike column structure to ensure a smooth transition. It is of great significance in ensuring the overall structural safety, but currently, the mechanical property and bearing capacity of this type of joint cannot be fully understood. This study investigates the load characteristics of three-forked cast steel joints through concrete experiments, finite element analysis, and regression method formula derivation, filling the gap in mechanical properties and calculation formulas of forked cast steel joints. Initially, a comprehensive model of the cast-steel joint, sourced from a practical engineering, underwent vertical load testing. Detailed scrutiny of stress distribution and vertical displacement of the tested joint was conducted based on the experimental outcomes. Subsequently, a finite element model of the tested joint was constructed using SolidWorks and subjected to analysis via ANSYS. The numerical findings were juxtaposed with experimental data and extrapolated to encompass other parametric scenarios. Ultimately, a regression analysis method was employed to derive a calculation formula for the load-carrying capacity of branch-bearing cast-steel joints. The regression analysis method can accurately obtain the load-bearing capacity calculation formula for tree-shaped joint models and can be extended to determine corresponding branch and main pipe dimensions, as well as the deviation angle between branches and the main pipe, under known load conditions. This improves design efficiency and accuracy. Comparative analysis reveals a substantial concurrence among experimental, finite element analysis, and formula-based predictive outcomes. The maximum error between experimental results and those obtained from finite element analysis is 9.02%. The maximum error between the results calculated using the load-bearing capacity formula derived from regression methods and those from finite element analysis is only 1.9%.

Abstract Background Evidences of comparison of sex difference in Chinese irritable bowel syndrome (IBS) patients were few. We aim to compare gender difference in the biopsychosocial characteristics of Chinese patients of IBS predominant with diarrhea (IBS-D). Methods IBS-D patients meeting Rome III criteria were enrolled. We administered IBS symptom questionnaires, evaluation of psychological status (HAMD and HAMA scales) and IBS quality of life (IBS-QOL), dietary habits, healthcare seeking behaviors, and compared biopsychosocial characteristics between male and female patients. Results Four hundred and ninety patients were enrolled including 299 males and 191 females. More female patients reported abdominal pain associated with defecation (84.3% vs. 74.9%, P = 0.014) while males reported more abdominal discomfort (39.8% vs. 26.7%, P = 0.003). Females had higher IBS symptom score (9.7 ± 1.7 vs. 9.4 ± 1.4, P = 0.025) and more of females had severe abdominal pain/discomfort (17.8% vs. 12.4%, P = 0.013) while there were no significant differences of other bowel symptoms. Females reported higher incidence of comorbid anxiety state (64.9% vs. 52.8%, P = 0.008) and depression state (35.6% vs. 19.7%, P

Abstract Rare earth elements have excellent catalytic effects on improving hydrogen storage properties of the Mg2Ni-based alloys. This study used a small amount of Y to substitute Mg partially in Mg2Ni0.9Co0.1 and characterized and discussed the effects of Y on the solidification and de-/hydrogenation behaviors. The Mg2−xYxNi0.9Co0.1 (x = 0, 0.2, 0.3, and 0.4) hydrogen storage alloys were prepared using a metallurgy method. The phase composition of the alloys was studied using X-ray diffraction (XRD). Additionally, their microstructure and chemical composition were studied using scanning electron microscopy and energy-dispersive X-ray spectroscopy, respectively. The hydrogen absorption and desorption properties of the alloys were studied using pressure-composition isotherms and differential scanning calorimetric (DSC) measurements. The structure of the as-cast Mg2Ni0.9Co0.1 alloy was composed of the peritectic Mg2Ni, eutectic Mg–Mg2Ni, and a small amount of pre-precipitated Mg–Ni–Co ternary phases, and was converted into the Mg2NiH4, Mg2Ni0.9Co0.1H4, and MgH2 phases after hydrogen absorption. Furthermore, the XRD patterns of the alloys showed the MgYNi4 phase and a trace amount of the Y2O3 phase along with the Mg and Mg2Ni phases after the addition of Y. After hydrogen absorption, the phase of the alloys was composed of the Mg2NiH4, MgH2, MgYNi4, YH3, Y2O3, and Mg2NiH0.3 phases. With the increase of Y addition, the area ratios of the peritectic Mg2Ni matrix phase in the Mg2−xYxNi0.9Co0.1 (x = 0, 0.2, 0.3, and 0.4) alloys gradually decreased until they disappeared. However, the eutectic structure gradually increased, and the microstructures of the alloys were obviously refined. The addition of Y improves the activation performance of the alloys. The alloy only needed one cycle of de-/hydrogenation to complete the activation for x = 0.4. The DSC curves showed that the initial dehydrogenation temperatures of Mg2Ni0.9Co0.1 and Mg1.8Y0.2Ni0.9Co0.1 were 200 and 156 °C, respectively. The desorption activation energies of the hydrides of the Mg2Ni0.9Co0.1 and Mg1.8Y0.2Ni0.9Co0.1 alloys calculated using the Kissinger method were 94.7 and 56.5 kJ/mol, respectively. Moreover, the addition of Y reduced the initial desorption temperature of the alloys and improved their kinetic properties.

Steel truss bridges are especially vulnerable in the event of a sudden loss of a load-carrying element, which can trigger a chain of failures. This paper describes a unique case study of a steel truss bridge under construction subjected to sudden member breakages with an extensive monitoring system. The failures occurred during the dismantlement of temporary members that had been used to transform a three-span simply supported steel truss bridge into a three-span continuous structure during incremental launching. These temporary members needed to be removed once the bridge reached its final position. The robustness of the bridge was assessed using computer simulations of various failure scenarios to evaluate its capacity to effectively activate alternative load paths (ALPs). The results demonstrated the structural redundancy of the steel truss bridge. However, the dynamic response resulting from the failure of the temporary upper chord, due to the initially high tension in the rods, should not be overlooked. To mitigate this issue, a structural retrofitting method was proposed, involving jacking the truss girder above the side pier to reduce the tension in the temporary upper chord above the middle pier. The effectiveness of this method was demonstrated through both simulated and formal experimental tests.

Objective: Asthma, a chronic inflammatory disease in which type 2 T helper cells (Th2) play a causative role in the development of T2 asthma. N6-methyladenosine (m6A) modification, an mRNA modification, and methyltransferase-like 3 (METTL3) is involved in the development of T2 asthma by inhibiting Th2 cell differentiation. Sex determining region Y-box protein 5 (SOX5) is involved in regulating T cell differentiation, but its role in T2 asthma was unclear. The objective of this study was to explore the role of METTL3 and SOX5 in T2 asthma and whether there is an interaction between the two. Materials and methods: Adults diagnosed with T2 asthma (n = 14) underwent clinical information collection and pulmonary function tests. In vivo and in vitro T2 asthma models were established using female C57BL/6 mice and human bronchial epithelial cells (HBE). The expressions of METTL3 and SOX5 were detected by Western blot and qRT-PCR and Western blot. Th2 cell differentiation was determined by flow cytometry and IL-4 level was detected by ELISA. m6A methylation level was determined by m6A quantitative assay. The relationship between METTL3 expression and clinical parameters was determined by Spearman rank correlation analysis. The function of METTL3 and SOX5 genes in asthma was investigated in vitro and in vivo. The RNA immunoprecipitation assay detected the specific interaction between METTL3 and SOX5. Results: Patients with T2 asthma displayed lower METTL3 levels compared to healthy controls. Within this group, a negative correlation was observed between METTL3 and Th2 cells, while a positive correlation was noted between METTL3 and clinical parameters as well as Th1 cells. In both in vitro and in vivo models representing T2 asthma, METTL3 levels decreased significantly, while SOX5 levels showed the opposite trend. Overexpression of METTL3 gene in HBE cells significantly inhibited Th2 cell differentiation and increased m6A methylation activity. From a mechanism perspective, low METTL3 negatively regulates SOX5 expression through m6A modification dependence, while high SOX5 expression is positively associated with T2 asthma severity. Cell transfection experiments confirmed that METTL3 regulates Th2 cell differentiation and IL-4 release through SOX5. Conclusions: Overall, our results indicate that METTL3 alleviates Th2 cell differentiation in T2 asthma by modulating the m6A methylation activity of SOX5 in bronchial epithelial cells. This mechanism could potentially serve as a target for the prevention and management of T2 asthma.

The rapid concentration of tailings is a crucial initial step in the cemented paste backfill (CPB) process, as it ensures the stability of the backfilling slurry concentration and the continuity of the backfilling process. The structure of flocs plays a pivotal role in tailings concentration, directly contributing to such critical technical parameters as settling speed and bottom flow concentration. However, most of the current studies focus on flocculant addition rather than the tailings slurry itself. Also, there is a lack of uniform characterization of the flocculant structure in the flocculation and settling system. This study establishes a semi-quantitative assessment system for evaluating the fine structure of tailings flocs based on indicators such as the exposure state of coarse particles, floc size, and floc compactness (porosity). The findings reveal that, during the settling process, the higher true density of tailings (3.156 g/cm3) and intermediate particles (40 wt% of 38 μm–74 μm particles) attenuates the clamping effect of turbulence on settling, leading to a faster settling rate to 0.65 cm/s and 0.29 cm/s respectively. Moreover, in the thickening process, tailings located at the bottom of the mud layer expedite the precipitation of interstitial water within the floc structure through the combined influence of gravity and rake frame shear. Consequently, a denser floc structure forms, as evidenced by a higher concentration of bottom flow at a macroscopic level. This conclusion provides a theoretical basis for faster preparation of highly concentrated tailings slurry in CPB.

Thioredoxin (TRX) is a small protein with REDOX activity that plays a crucial role in a plant’s growth, development, and stress resistance. The TRX family has been extensively studied in Arabidopsis, rice, and wheat, and so it is likely that its members have similar biological functions in Liriodendron that have not been reported in Liriodendron. In this study, we performed the genome-wide identification of the TRX gene family based on the Liriodendron chinense genome, leading to a total of 42 LcTRX gene members. A phylogenetic analysis categorized these 42 LcTRX proteins into 13 subfamilies. We further characterized their chromosome distributions, gene structures, conserved protein motifs, and cis-elements in the promoter regions. In addition, based on the publicly available transcriptome data for Liriodendron hybrid and following RT-qPCR experiments, we explored the expression patterns of LhTRXs to different abiotic stressors, i.e., drought, cold, and heat stress. Notably, we found that several LhTRXs, especially LhTRX-h3, were significantly upregulated in response to abiotic stress. In addition, the subcellular localization assay showed that LhTRX-h3 was mainly distributed in the cytoplasm. Subsequently, we obtained LhTRX-h3 overexpression (OE) and knockout (KO) callus lines in Liriodendron hybrid. Compared to the wild type (WT) and LhTRX-h3-KO callus proliferation of LhTRX-h3-OE lines was significantly enhanced with reduced reactive oxygen species (ROS) accumulation under drought stress. Our findings that LhTRX-h3 is sufficient to improve drought tolerance. and underscore the significance of the TRX gene family in environmental stress responses in Liriodendron.

Primarily generated at the interface between the wheel and the rail, railroad vibrations then propagate through the supporting soil. If these vibrations reach nearby bridges and buildings, they amplify the vibration nuisance and cause ground noise, which has detrimental effects on nearby residents, sensitive equipment, and historic structures. By analyzing measured data from metro vibration field vibration experiments, this article attempts to contribute to the body of knowledge on environmental vibration propagation patterns by offering insightful conclusions. Before analyzing the deformation response of the metro jet system (MJS) vibration isolation piles to the structure and the ground, we investigated the effect of MJS vibration isolation piles in the ground of the existing subway tunnel structure on the control of vibration of the proximate structure and conducted dynamic tests on the vibration of bridges without vibration isolation measures caused by operating subway trains. The tests determined that the acceleration of the bridge’s lateral vibration exceeded the code limit; one of the contributing factors was that the bridge’s structure had already sustained damage. The utilization of MJS isolation piles was also discovered to safeguard the extant bridge pile foundations. The paper presents an innovation in the form of economically viable vibration mitigation strategies that were implemented subsequent to the identification that the lateral vibration acceleration of the preexisting bridge surpassed the prescribed code standards. Considerable insight is gained regarding the design and implementation of vibration control systems for structures situated near caverns, encompassing deep foundation works.

There has been much controversy over the effects of music tempo on movement flow. In this study, a single-factor repeated-measurement design was used to explore the effect of music tempo (fast, slow, and no music control) on movement flow by measuring both subjective experiences and objective electroencephalographic (EEG) characteristics during brisk walking. In the experiment, 20 college students walked briskly on a treadmill using EEG equipment. Each participant walked for 10 min on three different days. Their brain waves were recorded during brisk walking on a treadmill. After each walk, the participants completed a form of short flow state scale-2 (S FSS-2), which covered nine major aspects of flow. The results showed that music tempo had a significant effect on subjective experiences and objective physiological characteristics; that is, a higher subjective flow level for fast-tempo music in brisk walking and a significant enhancement of mean power values in the subconscious brain waves of the delta, theta, alpha, and beta bands for fast tempo music were observed. A fast tempo facilitated the movement flow. The findings of this study can be instructive for the use of music in exercises to improve sports training outcomes.

Abstract Background The relationship between obesity and osteoporosis is complex, with contradictory findings reported. Our aim was to evaluate the association between waist circumference (WC), as an easy-to-determine clinical index of abdominal obesity, and femoral neck bone mineral density (BMD) among older adults, using the National Health and Nutrition Examination Survey (NHANES) database. Methods Data of five NHANES cycles (2005–2010, 2013–2014, and 2017–2018), including 5801 adults aged ≥ 60 years, were used in the analysis. Weighted multiple regression analyses were performed to evaluate the association between WC and femoral neck BMD. Weighted generalized additive models and smooth curve fitting were further performed to characterize nonlinearities in the association. Results There was a positive association between WC and femoral neck BMD in non-adjusted models. After adjusting for body mass index (BMI), the association became negative. On subgroup analysis stratified by sex, this negative association only existed for men. An inverted U-shaped curve relationship between WC and femoral neck BMD was further identified, with an inflection point at a WC of 95 cm for both men and women. Conclusions Abdominal obesity is a negative predictor of bone health among older adults, independent of BMI. The association between WC and femoral neck BMD followed an inverted U-shaped curve.

Characterized by soil moisture content and plant growth, agricultural drought occurs when the soil moisture content is lower than the water requirement of plants. Microwave remote sensing observation has the advantages of all-weather application and sensitivity to soil moisture change. However, microwave remote sensing can only invert 0~5 cm of soil surface moisture, so it cannot effectively reflect the drought situation of farmland. Therefore, this study took Henan Province as the study area, used soil moisture active and passive (SMAP) satellite soil moisture data, employed NDVI, LST, and ET as the independent variables, and took the drought grade on the sample as the dependent variable. Using the 2017–2019 data as the training set and the 2020 data as the testing set, a random forest drought monitoring model with comprehensive influence of multiple factors was constructed based on the training set data. In the process of model training, the cross-validation method was employed to establish and verify the model. This involved allocating 80% of the sample data for model construction and reserving 20% for model verification. The results demonstrated an 85% accuracy on the training set and an 87% accuracy on the testing set. Additionally, two drought events occurring during the winter wheat growing period in Henan Province were monitored, and the validity of these droughts was confirmed using on-site soil moisture and the vegetation supply water index (VSWI). The findings indicated a high incidence of agricultural drought in the southwestern part of Henan Province, while the central and northern regions experienced a lower incidence during the jointing to heading and filling stages. Subsequently, leveraging the results from the random forest drought monitoring, this study conducted a time series analysis using the Mann–Kendall test and a spatial analysis employing Moran’s I index to examine the temporal and spatial distribution of agricultural drought in Henan Province. This analysis aimed to unveil trends in soil moisture changes affecting agricultural drought, as observed via the SMAP satellite (NASA). The results suggested a possible significant spatial auto-correlation in the occurrence of agricultural drought.

Soil moisture (SM) is a crucial environmental variable, and it plays an important role in energy and water cycles. SM data retrieval based on microwave satellite remote sensing has garnered significant attention due to its spatial continuity, wide observational coverage, and relatively low cost. Validating the accuracy of satellite remote sensing SM products is a critical step in enhancing data credibility, which plays a vital role in ensuring the effective application of satellite remote sensing data across various fields. Firstly, this study focused on Henan Province and evaluated the accuracy of the SMAP Enhanced L3 Radiometer Global and Polar Grid Daily 9 km EASE-Grid Soil Moisture (SPL3SMP_E) product along with its application in agriculture. The evaluation was based on in situ SM data from 55 stations in Henan Province. The assessment metrics used in this study include mean difference (MD), root mean square error (RMSE), unbiased root mean square error (ubRMSE), and the Pearson correlation coefficient (R). The time span of this study is from 2017 to 2020. The evaluation results indicated that the SPL3SMP_E soil moisture product performs well, as reflected by an ubRMSE value of 0.045 (m3/m3), which was relatively close to the product’s design accuracy of 0.04 (m3/m3). Moreover, the accuracy of the product was unaffected by temporal factors, but the product exhibited strong spatial aggregation, which was closely related to land use types. Then, this study explored the response of the SPL3SMP_E product to irrigation signals. The precipitation and irrigation data from Henan Province were employed to investigate the response of the SPL3SMP_E soil moisture product to irrigation. Our findings revealed that the SPL3SMP_E soil moisture product was capable of capturing over 70% of irrigation events in the study area, indicating its high sensitivity to irrigation signals in this region. In this study, the SPL3SMP_E product was also employed for monitoring agricultural drought in Henan Province. The findings revealed that the collaborative use of the SPL3SMP_E soil moisture product and machine learning algorithms proves highly effective in monitoring significant drought events. Furthermore, the integration of multiple indices demonstrated a notable enhancement in the accuracy of drought monitoring. Such an evaluation holds significant implications for the effective application of satellite remote sensing SM data in agriculture and other domains.

Calcineurin B-like (CBL) proteins are a class of important Ca2+ receptors that play key roles in plant stress response. CBLs have been shown to participate in responses to abiotic stresses such as drought, salt, and cold in many plant species, including Arabidopsis and rice. However, little is known about their potential functions in the desert halophyte Nitraria tangutorum. Here, we have identified 11 CBL genes distributed across six chromosomes of N. tangutorum and categorized them into four groups through phylogenetic analysis. Synteny analysis showed that they have strong collinear relationships and have undergone purifying selection during their evolution. NtCBL promoter regions contain a variety of cis-acting elements related to hormone and environmental stress responses. Real-time quantitative PCR showed that the expression of NtCBLs differed significantly among various tissues and was upregulated by salt and drought stress. We chose NtCBL1-1 for an in-depth functional characterization and observed that transgenic Arabidopsis plants expressing NtCBL1-1 exhibited increased tolerance to both drought and salt stress. Compared to wild-type Arabidopsis, transgenic lines showed higher germination rates, slower chlorophyll degradation, more soluble proteins, and reduced accumulation of the oxidative stress marker malondialdehyde. These findings indicate that NtCBL1-1 plays a significant role in responding to drought and salt stress, laying the foundation for further investigations into the functional mechanisms of NtCBL genes in N. tangutorum.

Heat shock factors (Hsfs) play a crucial role in plant defense processes. However, the distribution and functional characteristics of Hsf genes in the relict plant Liriodendron chinense are still unclear. In this study, a total of 19 LcHsfs were identified and divided into three separate subgroups, comprising 10 LcHsfA, 7 LcHsfB, and 2 LcHsfC genes, respectively, based on their phylogenetic tree and the presence/absence of conserved protein domains. Whole-genome duplication and segmental duplication led to an expansion of the LhHsf gene family. The promoters of LcHsf genes are enriched for different types of cis-acting elements, including hormone responsive and abiotic-stress-responsive elements. The expression of LcHsfA3, LcHsfA4b, LcHsfA5, LcHsfB1b, and LcHsfB2b increased significantly as a result of both cold and drought treatments. LcHsfA2a, LcHsfA2b, and LcHsfA7 act as important genes whose expression levels correlate strongly with the expression of the LcHsp70, LcHsp110, and LcAPX genes under heat stress. In addition, we found that transiently transformed 35S:LcHsfA2a seedlings showed significantly lower levels of hydrogen peroxide (H2O2) after heat stress and showed a stronger thermotolerance. This study sheds light on the possible functions of LcHsf genes under abiotic stress and identifies potentially useful genes to target for molecular breeding, in order to develop more stress-resistant varieties.

In order to investigate the equivalent complex refractive index of atmospheric aerosols near the Earth’s surface, we conducted measurements in the Hefei region from March to April 2022. These measurements utilized a micro-pulse lidar, an Aethalometer, and a Portable Optical Particle Profiler. These measurements encompassed aerosol particle size distribution as well as standard meteorological parameters including temperature, humidity, atmospheric pressure, and wind speed. Subsequently, this dataset was employed to develop an optimization algorithm for retrieving the equivalent complex refractive indices of near-surface aerosols. The methodology relies on lookup tables containing data for extinction efficiency and absorption efficiency factors. It operates on the premise of aerosol property stability within a defined time frame, utilizing measured extinction and absorption coefficients as simultaneous constraints during this period to inversely derive both the real and imaginary parts of the aerosol complex refractive index. Results from the simulation analysis reveal that the newly optimized retrieval algorithm, which relies on lookup tables, exhibits reduced sensitivity to instrument errors when compared to single-point constraint algorithms. This enhancement results in a more efficient and dependable approach for retrieving the aerosol complex refractive index. Empirical inversion and simulation studies were carried out to determine the aerosol equivalent complex refractive index in the Hefei region, utilizing measured data. This inversion process yielded an average complex refractive index of 1.48-i0.017 for aerosols in the Hefei region throughout the experimental period. Correlation analysis unveiled a positive association between the real part of the aerosol complex refractive index and the single-scattering albedo (SSA), whereas the imaginary part displayed a linear negative correlation with the SSA. The mathematical relationship between the real part and the SSA is y=0.19x+0.62, and the corresponding relationship between the imaginary part and the SSA is y=−5.3x+0.99. This research offers a novel method for the retrieval of the aerosol equivalent complex refractive index.

Background: Studying the effects of traffic vibration on adjacent structures has produced fruitful results, but there is a lack of systematic research on the protection, assessment, and ambient vibration effects on cultural relics, and the majority of the studies focus on above-ground buildings, with less research conducted on underground cultural relic sites. Objective: In order to investigate the effects of road-traffic-induced vibration on nearby underground sites, the distance between them was precisely determined. Methodology/approach: The site of Chengshang Village in Jurong City, Nanjing, China, was chosen as the research object, and the vibration of the underground site caused by traffic volume was measured on-site. Based on statistical analysis of experimental data, the vibration velocity was deduced as a function of the vehicle’s speed and the vibration source’s distance. Results: The excellent frequency band for traffic load vibration is between 0 and 40 Hz, and the attenuation speed of high-frequency vibration is faster than that of low-frequency vibration; the vibration speed is positively correlated with the speed of the vehicle, and the distance from the vibration source is exponentially attenuated; and under the condition of the determined limit value of the load and the vibration speed, the safety distance increases. Conclusions: This research utilizes the collected data to describe the relationship between the vibration velocity and the distance from the vibration source. Additionally, it estimates the appropriate distance at which cultural relics should be placed from the road to ensure their safety. The study’s findings may serve as a valuable point of reference for traffic planning and the preservation of underground cultural monuments.

Abstract Exercise intolerance is one of the major symptoms of chronic obstructive pulmonary disease (COPD). Exercise training can benefit COPD patients, but the underlying mechanism remains unclear. The modified Total Body Recumbent Stepper (TBRS, Nustep-T4) can benefit patients with stroke, spinal cord injury and amyotrophic lateral sclerosis. Nevertheless, the effect of TBRS training alone on pulmonary rehabilitation (PR) in COPD patients remains largely unknown. We aimed to explore the effect of TBRS training on exercise capacity and the thioredoxin system (TRXS) in COPD patients to provide a novel rehabilitation modality and new theoretical basis for PR of COPD patients. Ninety stable COPD patients were randomly divided into a control group (NC group) and a TBRS training group (TBRS group), with 45 cases in each group. Subjects in the TBRS training group were scheduled to undergo TBRS endurance training triweekly for 12 weeks under the guidance of a rehabilitation therapist. We assessed the primary outcome: exercise capacity (6-min walking distance, 6MWD); and secondary outcomes: perception of dyspnoea (mMRC, Borg), the COPD assessment test (CAT), the BODE index, pulmonary function, the number of acute exacerbations of COPD and oxidative stress (TRXS) at one-year follow-up. The accelerator-driven subcritical system has a strong transmutation ability and high inherent safety, and it is internationally recognized as the most promising long-life nuclear waste disposal device. This study involves the construction of a Visual Hydraulic ExperimentaL Platform (VHELP) for the purpose of evaluating the applicability of Reynolds-averaged Navier–Stokes (RANS) models and analyzing the pressure distribution within the fuel bundle channel of China initiative accelerator-driven system (CiADS). Measurements of thirty differential pressures in edge subchannels within a 19-pin wire-wrapped fuel bundle channel were obtained under different conditions using deionized water. The pressure distribution in the fuel bundle channel at Reynolds numbers of 5000, 7500, 10,000, 12,500, and 15,000 was simulated using Fluent. The results show that RANS models obtained accurate results, and the shear stress transport k-ω model provided the most accurate prediction of the pressure distribution. The difference between the results of the Shear stress transport (SST) k-ω model and experimental data was the smallest, and the maximum difference was ±5.57%. Moreover, the error between the experimental data and numerical results of the axial differential pressure was smaller than that of the transverse differential pressure. The pressure periodicity in axial and transverse directions (one pitch) and a relatively three-dimensional pressure measurements were studied. The static pressure fluctuated and decreased periodically as the z-axis coordinate increased. These results can facilitate research on the cross-flow characteristics of liquid metal-cooled fast reactors.

Background: Tied-arch bridges’ hangers are crucial load-bearing parts, and their excellent condition directly influences bridge safety. However, assessing in-service hangers’ continuing functional performance is irrelevant and incomplete, particularly for unique hangers covered by outer steel tubes. Objective: This research uses a case study of an under-bearing tied-arch bridge with substantial hanger damage to determine the origin of the damage and analyze the hanger’s remaining operational ability. Methodology/approach: This study presents a set of assessment methodologies and procedures for in-service hangers’ remaining functioning performance using field inspection and indoor tests. First, an appearance inspection of the full bridge hanger’s upper and lower anchor heads was carried out, and the categories of anchor head damage and distribution rules are summarized. The causes of major water damage and the lower anchor head’s water infiltration channel were explored. Then, a full interior test was performed on the disassembled sick hanger to establish its present mechanical qualities. Finally, field inspection and indoor test findings assessed the bridge hanger’s operational performance. The findings suggest that the anchor box drainage prevention system should be improved to prevent rainfall and condensation from pooling in the lower anchor box and causing anticorrosive grease failure and anchor head corrosion. Results: The hanger’s mechanical qualities have deteriorated and no longer meet usage standards. Most of the water accumulated in the anchor head of the conventional construction hanger enters from the bridge deck or rope surface, but because of the outer steel pipe, rainwater can flow into the lower anchor box through the upper anchor box along the gap between the hanger and the outer steel pipe, so the waterproof system of the upper anchor box should be checked. Conclusions: This research may be used for safety evaluation and maintenance of the same hanger in service.

Abstract Background Liriodendron chinense (Lchi) is a tree species within the Magnoliaceae family and is considered a basal angiosperm. The too low or high temperature or soil drought will restrict its growth as the adverse environmental conditions, thus improving L. chinense abiotic tolerance was the key issues to study. WRKYs are a major family of plant transcription factors known to often be involved in biotic and abiotic stress responses. So far, it is still largely unknown if and how the LchiWRKY gene family is tied to regulating L. chinense stress responses. Therefore, studying the involvement of the WRKY gene family in abiotic stress regulation in L. chinense could be very informative in showing how this tree deals with such stressful conditions. Results In this research, we performed a genome-wide analysis of the Liriodendron chinense (Lchi) WRKY gene family, studying their classification relationships, gene structure, chromosomal locations, gene duplication, cis-element, and response to abiotic stress. The 44 members of the LchiWRKY gene family contain a significant amount of sequence diversity, with their lengths ranging from 525 bp to 40,981 bp. Using classification analysis, we divided the 44 LchiWRKY genes into three phylogenetic groups (I, II, II), with group II then being further divided into five subgroups (IIa, IIb, IIc, IId, IIe). Comparative phylogenetic analysis including the WRKY families from 17 plant species suggested that LchiWRKYs are closely related to the Magnolia Cinnamomum kanehirae WRKY family, and has fewer family members than higher plants. We found the LchiWRKYs to be evenly distributed across 15 chromosomes, with their duplication events suggesting that tandem duplication may have played a major role in LchiWRKY gene expansion model. A Ka/Ks analysis indicated that they mainly underwent purifying selection and distributed in the group IId. Motif analysis showed that LchiWRKYs contained 20 motifs, and different phylogenetic groups contained conserved motif. Gene ontology (GO) analysis showed that LchiWRKYs were mainly enriched in two categories, i.e., biological process and molecular function. Two group IIc members (LchiWRKY10 and LchiWRKY37) contain unique WRKY element sequence variants (WRKYGKK and WRKYGKS). Gene structure analysis showed that most LchiWRKYs possess 3 exons and two different types of introns: the R- and V-type which are both contained within the WRKY domain (WD). Additional promoter cis-element analysis indicated that 12 cis-elements that play different functions in environmental adaptability occur across all LchiWRKY groups. Heat, cold, and drought stress mainly induced the expression of group II and I LchiWRKYs, some of which had undergone gene duplication during evolution, and more than half of which had three exons. LchiWRKY33 mainly responded to cold stress and LchiWRKY25 mainly responded to heat stress, and LchiWRKY18 mainly responded to drought stress, which was almost 4-fold highly expressed, while 5 LchiWRKYs (LchiWRKY5, LchiWRKY23, LchiWRKY14, LchiWRKY27, and LchiWRKY36) responded equally three stresses with more than 6-fold expression. Subcellular localization analysis showed that all LchiWRKYs were localized in the nucleus, and subcellular localization experiments of LchiWRKY18 and 36 also showed that these two transcription factors were expressed in the nucleus. Conclusions This study shows that in Liriodendron chinense, several WRKY genes like LchiWRKY33, LchiWRKY25, and LchiWRKY18, respond to cold or heat or drought stress, suggesting that they may indeed play a role in regulating the tree’s response to such conditions. This information will prove a pivotal role in directing further studies on the function of the LchiWRKY gene family in abiotic stress response and provides a theoretical basis for popularizing afforestation in different regions of China.

Profiling the spatial distributions and tissue changes of characteristic compounds with interspecific differences is critical to elucidate the complex species identification during tree species traceability, wood anti-counterfeiting verification and timber trade control. In this research, in order to visualize the spatial position of characteristic compounds in two species with similar morphology (Pterocarpus santalinus and Pterocarpus tinctorius), a high coverage MALDI-TOF-MS imaging method was used to found the mass spectra fingerprints of different wood species. 2-Mercaptobenzothiazole matrix was used to spray wood tissue section to enhance the detection effect of metabolic molecules, and the mass spectrometry imaging data were obtained. Based on this technology, the spatial location of fifteen potential chemical markers with remarkable interspecific differences in 2 Pterocarpus timber species were successfully obtained. Distinct chemical signatures obtained from this method can promote rapid identification at the wood species level. Thus, matrix-assisted laser desorption/time-of-flight/ionization mass spectrometry imaging (MALDI-TOF-MSI) provides a spatial-resolved way for traditional wood morphological classification and breaking through the limitations of traditional wood identification technology.

The calcineurin B-like (CBL)-interacting protein kinases (CIPKs), a type of plant-specific genes in the calcium signaling pathway, function in response to adverse environments. However, few halophyte derived CIPKs have been studied for their role in plant physiological and developmental adaptation during abiotic stresses, which inhibits the potential application of these genes to improve environmental adaptability of glycophytes. In this study, we constructed Nitraria billardieri CIPK25 overexpressing Arabidopsis and analyzed the seedling development under salt treatment. Our results show that Arabidopsis with NbCIPK25 expression exhibits more vigorous growth than wild type plants under salt condition. To gain insight into the molecular mechanisms underlying salt tolerance, we profiled the transcriptome of WT and transgenic plants via RNA-seq. GO and KEGG analyses revealed that upregulated genes in NbCIPK25 overexpressing seedlings under salt stress are enriched in photosynthesis related terms; Calvin-cycle genes including glyceraldehyde-3-phosphate dehydrogenases (GAPDHs) are significantly upregulated in transgenic plants, which is consistent with a decreased level of NADPH (GAPDH substrate) and increased level of NADP+. Accordingly, NbCIPK25 overexpressing plants exhibited more efficient photosynthesis; soluble sugar and proteins, as photosynthesis products, showed a higher accumulation in transgenic plants. These results provide molecular insight into how NbCIPK25 promotes the expression of genes involved in photosynthesis, thereby maintaining plant growth under salt stress. Our finding supports the potential application of halophyte-derived NbCIPK25 in genetic modification for better salt adaptation.

Thin-shell dome structures are conventionally built with concrete. However, the building process is time-consuming and of high labor cost. Composite Structural Insulation Panels (CSIPs) have recently been regarded to be suitable for large-span civil structures, in terms of its light weight, easy processing, and mass production. Hence, this paper explores the feasibility of CSIPs domes applied in large-span dome structures. Compared with the performance of thin-shell concrete domes, the stiffness, strength, and stability of CSIPs domes are studied by means of refined finite element analysis. Results demonstrate CSIPs dome possesses good dynamic performance, bears larger nonlinear loads, and even copes with stress loads under earthquakes. Moreover, the stiffness should be handled in the onsite applications to fulfil the safety requirements.

In this study, on-site monitoring was used to obtain the temperature field distribution and cable force values of the bridge at various times, the temperature change pattern of the bridge elements, the temperature sensitivity of the cable force of different parts of the bridge, and the trend of the cable force under temperature load were all analyzed. The static response of the wires was studied under various operating circumstances. The total cable force of the bridge is inversely related to the temperature rise and fall, and the temperature sensitivity of the cable force in different parts of the bridge is different under the same temperature effect. The change in cable force is uneven. They vary according to the loading circumstances. This project's cable force changes are the most substantial and consistent with observed circumstances and should be appropriately considered in comparable projects. Thermal impacts must be addressed in strand cable safety design.

Accurate irrigation water-use data are essential to agricultural water resources management and optimal allocation. The obscuration presented by ground cover in farmland and the subjectivity of irrigation-related decision-making processes mean that effectively identifying regional irrigation water use remains a critical problem to be solved. In view of the advantages of satellite microwave remote sensing in monitoring soil moisture, previous studies have proposed a method for estimating irrigation water use using the satellite microwave remote sensing of soil moisture. However, the method is affected by false irrigation signals from soil moisture increases caused by non-irrigation factors, causing irrigation water use to be overestimated. Therefore, the purpose of this study is to improve the estimation of irrigation water use in drylands by using irrigation signals from SMAP soil moisture data. In this paper, the irrigation water use in Henan Province is estimated by using the irrigation signals from SMAP (soil moisture active and passive) soil moisture data. Firstly, a method for recognizing irrigation signals in soil moisture data obtained by microwave satellite remote sensing was used. Then, an estimation model of the amount of irrigation water (SM2Rainfall model) was built on each data pixel of the satellite microwave remote sensing of soil moisture. Finally, the amount of irrigation water utilized in Henan Province was estimated by combining the irrigation signals and irrigation water-use estimation model, and the results were evaluated. According to the findings, this study improved the estimation accuracy of irrigation water use by using the irrigation signals in Henan Province. The result of this study is of great importance to accurately obtain irrigation water use in the region.

This study addresses the problem of restricted ability for large-scale monitoring due to the limited cruising time of unmanned aerial vehicles (UAV) by identifying an optimal leaf ChlorophyII content (LCC) inversion machine learning model at different scales and under different parameterization schemes based on simultaneous observations of ground sampling, UAV flight, and satellite imagery. The following results emerged: (1) The correlation coefficient between most remote sensing features (RSFs) and LCC increased as the remote scale expanded; thus, the scale error caused by the random position difference between GPS and measuring equipment should be considered in field sampling observations. (2) The LCC simulation accuracy of the UAV multi-spectral camera using four machine learning algorithms was ExtraTree > GradientBoost > AdaBoost > RandomForest, and the 20- and 30-pixel scales had better accuracy than the 10-pixel scale, while the accuracy for three feature combination schemes ranked combination of extremely significantly correlated RSFs > combination of significantly correlated and above RSFs > combination of all features. ExtraTree was confirmed as the optimal model with the feature combination of scheme 2 at the 20-pixel scale. (3) Of the Sentinel-2 RSFs, 27 of 28 were extremely significantly correlated with LCC, while original band reflectance was negatively correlated, and VIs were positively correlated. (4) The LCC simulation accuracy of the four machine learning algorithms ranked as ExtraTree > GradientBoost > RandomForest > AdaBoost. In a comparison of two parameterization schemes, scheme 1 had better accuracy, while ExtraTree was the best algorithm, with 11 band reflectance as input RSFs; the RMSE values for the training and testing data sets of 0.7213 and 1.7198, respectively.

The plant-specific module of calcineurin B-like proteins (CBLs) and CBL-interacting protein kinases (CIPKs) play a crucial role in plant adaptation to different biotic and abiotic stresses in various plant species. Despite the importance of the CBL-CIPK module in regulating plant salt tolerance, few halophyte CIPK orthologs have been studied. We identified NbCIPK25 in the halophyte Nitraria billardieri as a salt-responsive gene that may improve salt tolerance in glycophytes. Sequence analyses indicated that NbCIPK25 is a typical CIPK family member with a conserved NAF motif, which contains the amino acids: asparagine, alanine, and phenylalanine. NbCIPK25 overexpression in salt-stressed transgenic Arabidopsis seedlings resulted in enhanced tolerance to salinity, a higher survival rate, longer newly grown roots, more root meristem cells, and less damaged root cells in comparison to wild-type (WT) plants. H2O2 accumulation and malondialdehyde (MDA) content were both deceased in NbCIPK25-transgenic plants under salt treatment. Furthermore, their proline content, an important factor for scavenging reactive oxygen species, accumulated at a significantly higher level. In concordance, the transcription of genes related to proline accumulation was positively regulated in transgenic plants under salt condition. Finally, we observed a stronger auxin response in salt-treated transgenic roots. These results provide evidence for NbCIPK25 improving salt tolerance by mediating scavenging of reactive oxygen species, thereby protecting cells from oxidation and maintaining plant development under salt stress. These findings suggest the potential application of salt-responsive NbCIPK25 for cultivating glycophytes with a higher salt tolerance through genetic engineering.

Among all kinds of journal bearings, the tilting pad bearing has the characteristics of high stability. In order to further reduce the vibration and improve the stability of rotor-bearing systems, a novel double layers flexible support tilting pad bearing (DL-FSTPB) is presented in this paper. The bearing utilizes two layers of springs, which have different functions. The offset springs control the swing angle of the tilting pad to ensure the formation of a convergent oil film, and the symmetrical springs are used to change the bearing stiffness coefficient, increase the bearing elasticity, and improve the ability to accommodate misalignment. Experiments were carried out to study the vibration characteristics of the rotor system. The results were analyzed using wavelet analysis of acceleration and displacement, waterfall plots, a wavelet analysis, a run-up process and shaft center orbits areas. The results show that the DL-FSTPB exhibits excellent vibration attenuation characteristics, which can effectively reduce the amplitude and the system vibration level.

The van der Pauw method is an important technique in measuring the resistivity of flat materials by means of the van der Pauw formula. The formula is derived analytically from a rectangular material by solving the two-dimensional Laplace equation of electric potential. The input and output current densities are two Neumann boundary conditions. The use of the δ function as the current boundary condition, in comparison with the rect function in the literature, makes the deduction simple. The van der Pauw measurement is simulated with the finite element method by using two arbitrarily shaped materials of different resistivities and four point contacts around the periphery. The input current is used as the boundary condition at one contact and electric potential as the variable to be solved. The potential differences between two voltage contacts are employed to calculate the two resistances for the formula. With the reduction of mesh size, the simulated resistivity approaches the actual value progressively. The minimum relative error is on the order of ppm. In addition, one circular hole is produced in the irregular material, and the relative error is investigated in response to the hole position and radius. The equipotential curves and current pattern are displayed for demonstration. This confirms that the van der Pauw method is valid for measurement of arbitrarily shaped materials without an inner hole.

Liriodendron chinense is a tree species of the Magnoliaceae family, an ancient relict plant mainly used for landscaping and timber production due to its excellent material properties and ornamental value. The cytokinin oxidase/dehydrogenase (CKX) enzyme regulates cytokinin levels and plays an important role in plant growth, development, and resistance. However, too-high or too-low temperatures or soil drought can limit the growth of L. chinense, representing a key issue for research. Here, we identified the CKX gene family in the L. chinense genome and examined its transcriptional responses to cold, drought, and heat stresses. A total of five LcCKX genes, distributed on four chromosomes and divided into three phylogenetic groups, were identified across the whole L. chinense genome. Further analysis showed that multiple hormone- and stress-responsive cis-acting elements are located in the promoter regions of LcCKXs, indicating a potential role of these LcCKXs in plant growth, development, and response to environmental stresses. Based on existing transcriptome data, LcCKXs, especially LcCKX5, were found to transcriptionally respond to cold, heat, and drought stresses. Furthermore, quantitative reverse-transcription PCR (qRT-PCR) showed that LcCKX5 responds to drought stress in an ABA-dependent manner in stems and leaves and in an ABA-independent manner in roots. These results lay a foundation for functional research on LcCKX genes in the resistance breeding of the rare and endangered tree species of L. chinense.

Water quality is directly linked to drinking water safety for millions of people receiving the water. The Danjiangkou Reservoir is the main water source for the Middle Route of the South-to-North Water Diversion Project (MR-SNWDP), located in the vicinity of Henan and Hubei provinces in China. Aquatic microorganisms are key indicators of biologically assessing and monitoring the water quality of the reservoir as they are sensitive to environmental and water quality changes. This study aimed to investigate the spatiotemporal variations in bacterioplankton communities during wet (April) and dry (October) seasons at eight monitoring points in Hanku reservoir and five monitoring points in Danku reservoir. Each time point had three replicates, labeled as wet season Hanku (WH), wet season Danku (WD), dry season Hanku (DH), and dry season Danku (DD) of Danjiangkou Reservoir in 2021. High-throughput sequencing (Illumina PE250) of the 16S rRNA gene was performed, and alpha (ACE and Shannon) and beta (PCoA and NDMS) diversity indices were analyzed. The results showed that the dry season (DH and DD) had more diverse bacterioplankton communities compared to the wet season (WH and WD). Proteobacteria, Actinobacteria, and Firmicutes were the most abundant phyla, and Acinetobacter, Exiguobacterium, and Planomicrobium were abundant in the wet season, while polynucleobacter was abundant in the dry season. The functional prediction of metabolic pathways revealed six major functions including carbohydrate metabolism, membrane transport, amino acid metabolism, signal transduction, and energy metabolism. Redundancy analysis showed that environmental parameters greatly affected bacterioplankton diversity during the dry season compared to the wet season. The findings suggest that seasonality has a significant impact on bacterioplankton communities, and the dry season has more diverse communities influenced by environmental parameters. Further, the relatively high abundance of certain bacteria such as Acinetobacter deteriorated the water quality during the wet season compared to the dry season. Our findings have significant implications for water resource management in China, and other countries facing similar challenges. However, further investigations are required to elucidate the role of environmental parameters in influencing bacterioplankton diversity in order to devise potential strategies for improving water quality management in the reservoir.

Carotenoid cleavage oxygenase (CCO) is an enzyme that can catalyze carotenoids to volatile aromatic substances and participate in the biosynthesis of two important phytohormones, i.e., abscisic acid (ABA) and strigolactone (SL). However, the genome-wide identification and analysis of the CCO gene family in the rare and endangered woody plant Liriodendron chinense has not been reported. Here, we performed a genome-wide analysis of the CCO gene family in the L. chinense genome and examined its expression pattern during different developmental processes and in response to various abiotic stresses. A total of 10 LcCCO genes were identified and divided into 6 subfamilies according to the phylogenetic analysis. Subcellular localization prediction showed that most of the LcCCO proteins were located in the cytoplasm. Gene replication analysis showed that segmental and tandem duplication contributed to the expansion of this gene family in the L. chinense genome. Cis-element prediction showed that cis-elements related to plant hormones, stress and light response were widely distributed in the promoter regions of LcCCO genes. Gene expression profile analysis showed that LcNCED3b was extensively involved in somatic embryogenesis, especially the somatic embryo maturation, as well as in response to heat and cold stress in leaves. Furthermore, qRT-PCR analysis showed that LcNCED3b obviously responded to drought stress in roots and leaves. This study provides a comprehensive overview of the LcCCO gene family and a potential gene target for the optimization of the somatic embryogenesis system and resistance breeding in the valuable forest tree L. chinense.

The ability to obtain an accurate measure of irrigation water use is urgently needed in order to provide further scientific guidance for irrigation practices. This investigation took soil moisture and precipitation as the study objects and quantitatively analyzed their relationship by establishing four models: a linear model, a logarithmic model, a soil water balance model, and a similarity model. The results from building models on every site clearly revealed the relationship between soil moisture and precipitation and confirmed the feasibility of estimating irrigation water use when soil moisture data are known. Four models combined with soil moisture data were used to estimate irrigation water use. First, the 16 sites which monitor soil moisture conditions in Hebi City were identified as study objects, from which everyday meteorological data (temperature, precipitation, atmospheric pressure, wind speed, sunshine duration) and soil moisture data from 2015 to 2020 (totaling six years) were collected. Second, the eligible data from the first four years in the date range were used to create four kinds of models (linear model, logarithmic model, soil water balance model, and similarity model) to estimate the amount of water input to the soil surface based on soil moisture. Third, the eligible data from the last two years in the established date range were used to verify the established models on every site and then judge the accuracy of the models. For example, for site 53990, the RMSE of the linear model, logarithmic model, soil water balance model, and similarity model was 10,547, 10,302, 8619, and 7524, respectively. The results demonstrate that the similarity model proposed in this study can express the quantitative relationship between soil moisture and precipitation more accurately than the other three models. Based on this conclusion, the eligible soil moisture data known in the specific site were ultimately used to estimate the irrigation water use in the field by the relationship expressed in the similarity model. Compared with the amount of irrigation water data recorded, the estimated irrigation water use yielded by the similarity model in this study was 18.11% smaller. In a future study, microwave satellite remote sensing of soil moisture data, such as SMAP and SMOS soil moisture data, will be used to evaluate the performance of estimated regional irrigation water use.

Cunninghamia lanceolata (Lamb.) Hook. (Chinese fir) is an important timber species that is widely cultivated in southern China. However, the shallow root system and weak drought resistance of Chinese fir are not enough to cope with high temperature and drought. In recent years, molecular biology has been used to modify plants to make them more resilient. Therefore, improving heat and drought resistance of Chinese fir by molecular biology technology is one of the best choices, whereas fewer genetic information resources for C. lanceolata limit more comprehensive molecular studies. In this study, single-molecule full-length transcriptome (SMRT) sequencing technology was used to obtain full-length transcriptome data on Chinese fir. A total of 21,331 transcripts were obtained via co-assembly, and 11,094 gene sets were obtained via further de-redundancy. In addition, gene function annotation and gene structure analysis were performed. We also used these data to identify nine heat shock transcription factors (Hsfs) in Chinese fir, and heat stress transcriptome and real-time quantitative polymerase chain reaction (PCR) analyses revealed expression changes in response to heat stress, indicating that these may play roles in heat resistance. These studies have enriched the genetic information resources of Chinese fir, which may be utilized for further species promotion, improvement, and application.

Superoxide dismutase (oxide dismutase) is an enzyme synthesized via the expression of SOD-related genes. It is the first line of defense against oxygen free radicals, and it widely exists in the cytoplasm, chloroplasts, and mitochondria. However, the SOD gene family of woody plant Liriodendron chinense has not been studied. To reveal the potential role of SOD genes, we systematically identified and analyzed the SOD gene family of L. chinense, and investigated the transcriptional responses of LcSOD genes to several abiotic stresses, including cold, heat, and drought. A total of eight SOD genes were identified, namely, five Cu/Zn-SODs, two Fe-SODs, and one Mn-SOD, and they were divided into two subgroups according to phylogenetic analysis, gene structure, and conserved motifs. Cis-acting element analysis reveals various hormones and stress respond as cis-acting elements in the promoters of LcSODs. Gene expression analysis shows that most LcSOD genes were in response to abiotic stresses at the transcriptional level. These results help in clarifying the function of LcSOD genes under abiotic stresses, and provide potential targets for the improvement in abiotic stress tolerance in the endangered L. chinense.

In this study, we devised a constraint method, called multi-model ensemble pattern (MEP), to estimate the refractive index structure parameter (Cn2) profiles based on observational data and multiple existing models. We verified this approach against radiosonde data from field campaigns in China’s eastern and northern coastal areas. Multi-dimensional statistical evaluations for the Cn2 profiles and integrated astronomical parameters have proved MEP’s relatively reliable performance in estimating optical turbulence in the atmosphere. The correlation coefficients of MEP and measurement overall Cn2 in two areas are up to 0.65 and 0.76. A much higher correlation can be found for a single radiosonde profile. Meanwhile, the difference evaluation of integrated astronomical parameters also shows its relatively robust performance compared to a single model. The prowess of this reliable approach allows us to carry out regional investigation on optical turbulence features with routine meteorological data soon.

Abstract Background Pulmonary arterial smooth muscle cell (PASMC) proliferation plays a crucial role in hypoxia-induced pulmonary hypertension (HPH). Previous studies have found that resistin-like molecule β (RELM-β) is upregulated de novo in response to hypoxia in cultured human PASMCs (hPASMCs). RELM-β has been reported to promote hPASMC proliferation and is involved in pulmonary vascular remodeling in patients with PAH. However, the expression pattern, effects, and mechanisms of action of RELM-β in HPH remain unclear. Methods We assessed the expression pattern, mitogenetic effect, and mechanism of action of RELM-β in a rat HPH model and in hPASMCs. Results Overexpression of RELM-β caused hemodynamic changes in a rat model of HPH similar to those induced by chronic hypoxia, including increased mean right ventricular systolic pressure (mRVSP), right ventricular hypertrophy index (RVHI) and thickening of small pulmonary arterioles. Knockdown of RELM-β partially blocked the increases in mRVSP, RVHI, and vascular remodeling induced by hypoxia. The phosphorylation levels of the PI3K, Akt, mTOR, PKC, and MAPK proteins were significantly up- or downregulated by RELM-β gene overexpression or silencing, respectively. Recombinant RELM-β protein increased the intracellular Ca2+ concentration in primary cultured hPASMCs and promoted hPASMC proliferation. The mitogenic effects of RELM-β on hPASMCs and the phosphorylation of PI3K, Akt, mTOR, PKC, and MAPK were suppressed by a Ca2+ inhibitor. Conclusions Our findings suggest that RELM-β acts as a cytokine-like growth factor in the development of HPH and that the effects of RELM-β are likely to be mediated by the Ca2+-dependent PI3K/Akt/mTOR and PKC/MAPK pathways.

Abstract WHSC1 is a histone methyltransferase that facilitates histone H3 lysine 36 dimethylation (H3K36me2), which is a permissive mark associated with active transcription. In this study, we revealed how WHSC1 regulates tumorigenesis and chemosensitivity of colorectal cancer (CRC). Our data showed that WHSC1 as well as H3K36me2 were highly expressed in clinical CRC samples, and high WHSC1 expression is associated with poorer prognosis in OS patients. WHSC1 reduction promoted colon cancer cell apoptosis both in vivo and in vitro. We found that B cell lymphoma-2 (BCL2) expression, an anti-apoptotic protein, is markedly decreased in after WHSC1 depletion. Mechanistic characterization indicated that WHSC1 directly binds to the promoter region of BCL2 gene and regulate its H3K36 dimethylation level. What’s more, our study indicated that WHSC1 depletion promotes chemosensitivity in CRC cells. Together, our results suggested that WHSC1 and H3K36me2 modification might be optimal therapeutic targets to disrupt CRC progression and WHSC1-targeted therapy might potentially overcome the resistance of chemotherapeutic agents.

Cunninghamia lanceolata (Lamb.)Hook is an important economic timber tree in China. However, its genome characteristics have not been extensively assessed. To better understand its genome information, the bacterial artificial chromosome (BAC) library of chinese fir was constructed. A total of 422 BAC clones were selected and divided into 10 pools and sequenced, and with an average insert size of 121 kb, ranging from 97 to 145 kb. A total of 61,902,523 bp of reference sequences were sequenced and assembled, and based on an estimated genome size of 11.6 Gb for Chinese fir, the BAC library was estimated to have a total coverage of 0.53% genome equivalents. Bioinformatics analyses were also performed for repeated sequences, tRNAs, coding gene prediction, and functional annotation. The results of this study provide insights into the brief structure of the Chinese fir genome and has generated gene data that will facilitate molecular investigations on the mechanisms underlying tree growth.

Abstract Background The doctor-patient relationship in China has deteriorated in recent years, and poor doctor-patient communication is one of the main reasons. How to effectively carry out doctor-patient communication training originated from the West among Chinese medical students still to be studied. In the past decade, Peking Union Medical College has adopted clinical scenario drama to teach doctor-patient relationship and clinical communication skills. The aim of this study was to introduce clinical scenario dramas and evaluate its effectiveness in promoting doctor-patient relationships and clinical communication skills through students’ self-perceptions in Chinese medical students. Methods This study was a retrospective, self-controlled study and conducted from March 2009 to October 2018. Doctor-patient relationship and communication skills training were administered to all sixth-year medical students, which involved lectures and various clinical scenario dramas. The program totaled 24 h, of which each class session was 3 h, with 8 sessions in total. All students were requested to complete an anonymous 5 likert self-rating survey including self-confidence in using communication skills and self-perceived learning attitude and ability before and at the end of the course. In addition, they were requested to evaluate the curriculum after completion of the course. Most colorectal cancer (CRC) are characterized by allele loss of the genes located on the short arm of chromosome 17 (17p13.1), including the tumor suppressor p53 gene. Although important, p53 is not the only driver of chromosome 17p loss. In this study, we explored the biological and prognostic significance of genes around p53 on 17p13.1 in CRC. The Cancer Genome Atlas (TCGA) were used to identify differentially expressed genes located between 1000 kb upstream and downstream of p53 gene. The function of CLDN7 was evaluated by both in vitro and in vivo experiments. Quantitative real-time PCR, western blot, and promoter luciferase activity, immunohistochemistry were used to explore the molecular drivers responsible for the development and progression of CRC. The results showed that CLDN7, located between 1000 kb upstream and downstream of p53 gene, were remarkably differentially expressed in tumor and normal tissues. CLDN7 expression also positively associated with p53 level in different stages of the adenoma-carcinoma sequence. Both in vitro and in vivo assays showed that CLDN7 inhibited cell proliferation in p53 wild type CRC cells, but had no effects on p53 mutant CRC cells. Mechanistically, p53 could bind to CLDN7 promoter region and regulate its expression. Clinically, high CLDN7 expression was negatively correlated with tumor size, invasion depth, lymphatic metastasis and AJCC III/IV stage, but was positively associated with favorable prognosis of CRC patients. Collectively, our work uncovers the tumor suppressive function for CLDN7 in a p53-dependent manner, which may mediate colorectal tumorigenesis induced by p53 deletion or mutation.

The Mg _1.8 Y _0.2 Ni _1−y Co _y (y = 0, 0.05, 0.1, 0.15, 0.2) hydrogen storage alloys were prepared following the principles of metallurgy, the phase composition and microstructure of the alloys were studied using XRD and SEM/EDS techniques, and the hydrogen absorption and desorption properties of the alloys were studied using PCT and DSC techniques. The results showed that the addition of Co did not affect the phase composition of Mg _1.8 Y _0.2 Ni _1−y Co _y alloys in the as-cast state and after hydrogen absorption. The Co addition could help refine the microstructure of the alloys to a certain extent. The de-/hydrogenation kinetics of Mg _1.8 Y _0.2 Ni _1−y Co _y (y = 0, 0.1, and 0.2) alloys were improved by adding Co, and the best results were obtained at y = 0.1. The onset decomposition temperature of Mg _1.8 Y _0.2 Ni _1−y Co _y (y = 0, 0.1, and 0.2) alloys were recorded to be 180 °C, 156 °C, and 210 °C, respectively, which were significantly lower than that of Mg _2 Ni (253 °C). The results revealed that the addition Co could improve the thermodynamic performance of the dehydrogenation process.

The development and application of new Fiber Reinforced Polymer (FRP) material and 3D printing construction technology provide a basis for making up for the shortcomings of traditional thin-shell structures and building new thin-shell structures with better performance. In this paper, a new 3D Printing Composite (3DPC) thin-shell structure is proposed, which is prepared using a FRP plate as a permanent base mold and combining it with 3D printing cement technology. Both the typical experiment and finite element numerical simulation analysis of the 3DPC thin-shell structure are carried out. The results show that the maximum load capacity of the 3DPC thin-shell structure is increased by 53.3% as compared with the corresponding traditional concrete thin-shell structure. The presence of the FRP sheet effectively delays the generation of initial cracks and enhances the ductility of components.

The Salt Overly Sensitive (SOS) signaling pathway is key in responding to salt stress in plants. SOS2, a central factor in this pathway, has been studied in non-halophytes such as Arabidopsis and rice, but has so far not been reported in the halophyte Nitraria tangutorum. In order to better understand how Nitraria tangutorum acquires its tolerance for a high salt environment, here, the NtSOS2 was cloned from Nitraria tangutorum, phylogenetic analyses showed that NtSOS2 is homologous to the SOS2 of Arabidopsis and rice. Gene expression profile analysis showed that NtSOS2 localizes to the cytoplasm and cell membrane and it can be induced by salt stress. Transgenesis experiments showed that exogenous expression of NtSOS2 reduces leaf mortality and improves the germination rate, biomass and root growth of Arabidopsis under salt stress. Also, exogenous expression of NtSOS2 affected the expression of ion transporter-related genes and can rescue the phenotype of sos2-1 under salt stress. All these results revealed that NtSOS2 plays an important role in plant salt stress tolerance. Our findings will be of great significance to further understand the mechanism of salt tolerance and to develop and utilize molecular knowledge gained from halophytes to improve the ecological environment.

Plants in the genus Melaleuca have been widely used as traditional medicine mainly because of their broad spectrum antimicrobial activity. In this study, we reported the complete chloroplast genome of Melaleuca cajuputi subsp. cumingiana. The chloroplast genome of this species is 158,855 bp in length, including a pair of inverted repeat regions (IRs) (26,727 bp) that is divided by a large single-copy (LSC) area (87,338 bp) and a small single-copy (SSC) area (18,063 bp). The circular chloroplast genome of M. cajuputi subsp. cumingiana contains 135 unique genes, composing of 87 protein-coding genes, 40 tRNA genes, and eight rRNA genes. Phylogenetic analysis indicates that M. cajuputi subsp. cumingiana was clustered with species in the tribe Melaleuceae. This complete chloroplast genome of M. cajuputi subsp. cumingiana will provide a powerful tool to accelerate breeding, biotechnological and phylogenetic study.

Cantilever brackets have been widely used in buildings to provide support for all kinds of pipes. In order to improve the bearing capacity of cantilever brackets, two types of reinforcement schemes are proposed, one is to sleeve a pipe, and the other is to add a haunch. Their mechanical properties are studied by numerical simulation and experimental investigation. First, the non-linear finite element (FE) simulation analysis was carried out, and the structural bearing capacity, stress distribution, and failure modes were discussed. Then, the full-scale model tests were completed to provide validation of the FE analysis. On this basis, a comparison of the FE results and test results of three kinds of cantilever brackets was discussed in detail. The results show that two reinforcement schemes can enhance the bearing capacity of the cantilever bracket significantly by 38.3% and 25.9%, respectively, and they are applicable for the reinforcement of existing cantilever brackets.

Nitraria sibirica is a shrub that can survive in extreme drought environments. The auxin-response factors (ARFs) are a class of transcription factors that are widely involved in plant growth and development, as well as in the regulation of stress resistance. However, the genome-wide identification of the ARF gene family and its responses to environmental stresses, especially drought stress, in N. sibirica has not yet been reported. Here, we identified a total of 12 ARF genes in the genome of N. sibirica, which were distributed over 10 chromosomes and divided into three clades. Intragenome synteny analysis revealed one collinear gene pair in the ARF gene family, i.e., NsARF9a and NsARF9b. Cis-acting element analysis showed that multiple hormones and stress-responsive cis-acting elements were found in the promoters of NsARFs, suggesting that NsARFs may be involved in multiple biological processes. Quantitative real-time PCR (qRT-PCR) showed that many NsARFs had tissue-specific expression patterns, with the highest expression of NsARF16 in the seedlings of N. sibirica. In addition, most of the NsARFs that were upregulated under drought were independent of endogenous ABA biosynthesis, whereas the response of NsARF5 and NsARF7a to drought was disrupted by the ABA-biosynthesis inhibitor fluridone. These studies provide a basis for further research into how NsARFs in N. sibirica respond to hormonal signaling and environmental stresses.

The calcineurin B-like-interacting protein kinase (CIPK) protein family plays a key role in the plant calcium ion-mediated signal transduction pathway, which regulates a plant’s response to abiotic stress. Nitraria sibirica pall. (N. sibirica) is a halophyte with a strong tolerance for high salt environments, yet how it is able to deal with salt stress on a molecular level is still unknown. Due to their function as described in other plant species, CIPK genes are prime candidates for a role in salt stress signaling in N. sibirica. In this study, we identified and analyzed the phylogenetic makeup and gene expression of the N. sibirica CIPK gene family. A total of 14 CIPKs were identified from the N. sibirica genome and were clustered into seven groups based on their phylogeny. The promoters of NsCIPK genes contained multiple elements involved in hormonal and stress response. Synteny analysis identified a total of three pairs of synteny relationships between NsCIPK genes. Each gene showed its own specific expression pattern across different tissues, with the overall expression of CIPK6 being the lowest, and that of CIPK20 being the highest. Almost all CIPK genes tended to respond to salt, drought, and cold stress, but with different sensitivity levels. In this study, we have provided a general description of the NsCIPK gene family and its expression, which will be of great significance for further understanding of the NsCIPK gene family function.

Liriodendron chinense (Lchi) is a Magnoliaceae plant, which is a basic angiosperm left behind by the Pleistocene and mainly distributed in the south of the Yangtze River. Liriodendron hybrids has good wood properties and is widely used in furniture and in other fields. It is not clear if they can adapt to different environmental conditions, such as drought and high and low temperatures, and the molecular mechanisms for this adaptation are unknown. Among plant transcription factors (TFs), the MYB gene family is one of the largest and is often involved in stress or adversity response signaling, growth, and development. Therefore, studying the role of MYBTFs in regulating abiotic stress signaling, growth, and development in Lchi is helpful to promote afforestation in different environments. In our research, a genome-wide analysis of the LchiMYB gene family was performed, including the phylogenetic relationship tree, gene exon-intron structure, collinearity, and chromosomal position. According to the evolutionary tree, 190 LchiMYBs were divided into three main branches. LchiMYBs were evenly distributed across 19 chromosomes, with their collinearity, suggesting that segment duplication events may have contributed to LchiMYB gene expansion. Transcriptomes from eight tissues, 11 stages of somatic embryogenesis, and leaves after cold, heat, and drought stress were used to analyze the function of the MYB gene family. The results of tissue expression analysis showed that most LchiMYB genes regulated bark, leaf, bud, sepal, stigma, and stamen development, as well as the four important stages (ES3, ES4, ES9, and PL) of somatic embryogenesis. More than 60 LchiMYBs responded to heat, cold, and drought stress; some of which underwent gene duplication during evolution. LchiMYB3 was highly expressed under all three forms of stress, while LchiMYB121 was strongly induced by both cold and heat stress. Eight genes with different expression patterns were selected and verified by quantitative real-time PCR (qRT-PCR) experiments. The results suggested that these LchiMYBs may regulate Lchi growth development and resistance to abiotic stress. This study shows the cross-regulatory function of LchiMYBs in the growth and development, asexual reproduction, and abiotic resistance of Lchi. This information will prove pivotal to directing further studies on the biological function of Lchi MYBTFs in genetic improvement and abiotic stress response.
