The rapid development of the global economy and population growth are accompanied by the production of numerous waste textiles. This leads to a waste of limited resources and serious environmental pollution problems caused by improper disposal. The rational recycling of wasted textiles and their transformation into high-value-added emerging products, such as smart wearable devices, is fascinating. Here, we propose a novel roadmap for turning waste cotton fabrics into three-dimensional elastic fiber-based thermoelectric aerogels by a one-step lyophilization process with decoupled self-powered temperature–compression strain dual-parameter sensing properties. The thermoelectric aerogel exhibits a fast compression response time of 0.2 s, a relatively high Seebeck coefficient of 43 μV·K−1, and an ultralow thermal conductivity of less than 0.04 W·m−1·K−1. The cross-linking of trimethoxy(methyl)silane (MTMS) and cellulose endowed the aerogel with excellent elasticity, allowing it to be used as a compressive strain sensor for guessing games and facial expression recognition. In addition, based on the thermoelectric effect, the aerogel can perform temperature detection and differentiation in self-powered mode with the output thermal voltage as the stimulus signal. Furthermore, the wearable system, prepared by connecting the aerogel-prepared array device with a wireless transmission module, allows for temperature alerts in a mobile phone application without signal interference due to the compressive strains generated during gripping. Hence, our strategy is significant for reducing global environmental pollution and provides a revelatory path for transforming waste textiles into high-value-added smart wearable devices.

Abstract The purpose of this study was to compare the predictive value of different lymph node staging systems and to develop an optimal prognostic nomogram for predicting distant metastasis in pancreatic ductal adenocarcinoma (PDAC). Our study involved 6364 patients selected from the Surveillance, Epidemiology, and End Results (SEER) database and 126 patients from China. Independent risk factors for distant metastasis were screened by univariate and multivariate logistic regression analyses, and a model-based comparison of different lymph node staging systems was conducted. Furthermore, we developed a nomogram for predicting distant metastasis using the optimal performance lymph node staging system. The lymph node ratio (LNR), log odds of positive lymph nodes (LODDS), age, primary site, grade, tumor size, American Joint Committee on Cancer (AJCC) 7th Edition T stage, and radiotherapy recipient status were significant predictors of distant metastasis in PDAC patients. The model with the LODDS was a better fit than the model with the LNR. We developed a nomogram model based on LODDS and six clinical parameters. The area under the curve (AUC) and concordance index (C-index) of 0.753 indicated that this model satisfied the discrimination criteria. Kaplan–Meier curves indicate a significant difference in OS among patients with different metastasis risks. LODDS seems to have a superior ability to predict distant metastasis in PDAC patients compared with the AJCC 8th Edition N stage, PLN and LNR staging systems. Moreover, we developed a nomogram model for predicting distant metastasis. Clinicians can use the model to detect patients at high risk of distant metastasis and to make further clinical decisions.

Based on the Dali I section of the Central Yunnan Diversion Dali Tunnel Project, an indoor model test of TBM traversing faulted geological conditions under high ground stress was carried out using the self-developed miniature TBM tunneling integrated test system to study the impact of faulting on the stability of TBM tunnels under high ground stress, analyze and summarize the change of stress and strain in the surrounding rock of the tunnel and the tunnel damage phenomenon, and reveal the disaster-causing mechanism of faulted geological jamming. The research results show that the TBM tunneling process first produces cracks in the surrounding rock at the upper part of the tunnel, and then a small drop in the vault when the TBM bores into the intersection of the two materials. The pressure on the surrounding rock in front of the palm face is mainly in the range of 0.43D～0.50D (D is the diameter of the hole), with the maximum increase of 1.19 times in the pressure on the surrounding rock. The internal rock displacement above the tunnel vault has four distinct phases of change with TBM tunneling. The closer the upper surrounding rock is to the tunnel contour line, the greater the deformation. When TBM passes through the fault interface, the deformation rate of the surrounding rock is the highest. The impact range on the internal displacement of the surrounding rock is mainly within 1.21D in front of the palm and 1.50D above it. The impact range of the fault is about 1.73H (H is the horizontal projection length of the fault interface), which is mainly caused by the loose sliding of the surrounding rock above the fault interface, and it is also easy to cause TBM jamming, Therefore, it is advisable to carry out advanced treatment at the fault interface during construction to reduce the risk of TBM operation.

Abstract Background The use of stem cell-derived exosomes (Exos) as therapeutic vehicles is receiving increasing attention. Exosome administration has several advantages over cell transplantation, thus making exosomes promising candidates for large-scale clinical implementation and commercialization. However, exosome extraction and purification efficiencies are relatively low, and therapeutic heterogeneity is high due to differences in culture conditions and cell viability. Therefore, in this study, we investigated a priming procedure to enhance the production and therapeutic effects of exosomes from human umbilical cord mesenchymal stem cells (hucMSCs). After preconditioning hucMSCs with agonists/inhibitors that target the Wnt/β-catenin pathway, we assessed both the production of exosomes and the therapeutic efficacy of the optimized exosomes in the context of diabetic wound healing, hoping to provide a safer, more stable and more effective option for clinical application. Results The Wnt signalling pathway agonist CHIR99021 increased exosome production by 1.5-fold without causing obvious changes in the characteristics of the hucMSCs or the size of the exosome particles. Further studies showed that CHIR99021 promoted the production of exosomes by facilitating exocytosis. This process was partly mediated by SNAP25. To further explore whether CHIR99021 changed the cargo that was loaded into the exosomes and its therapeutic effects, we performed proteomic and transcriptomic analyses of exosomes from primed and control hucMSCs. The results showed that CHIR99021 significantly upregulated the expression of proteins that are associated with cell migration and wound healing. Animal experiments confirmed that, compared to control hucMSC-derived exosomes, CHIR99021-pretreated hucMSC-derived exosomes (CHIR-Exos) significantly accelerated wound healing in diabetic mice, enhanced local collagen deposition, promoted angiogenesis, and reduced chronic inflammation. Subsequent in vitro experiments confirmed that the CHIR-Exos promoted wound healing by facilitating cell migration, inhibiting oxidative stress-induced apoptosis, and preventing cell cycle arrest. Conclusions The Wnt agonist CHIR99021 significantly increased exosome secretion by hucMSCs, which was partly mediated by SNAP25. Notably, CHIR99021 treatment also significantly increased the exosomal levels of proteins that are associated with wound healing and cell migration, resulting in enhanced acceleration of wound healing. All of these results suggested that pretreatment of hucMSCs with CHIR99021 not only promoted exosome production but also improved the exosome therapeutic efficacy, thus providing a promising option for large-scale clinical implementation and commercialization. Abstract The authors investigated the ingredient detection technique of silicone rubber based on the Terahertz spectrum. For this purpose, 18 diverse high‐temperature vulcanised silicone rubber (HTVSR) formulations were customised, 8 of which are used as calibration set while the rest 10 as prediction set. Based on the Beer‐Lambert Law, the partial‐least‐square (PLS) regression model and the least‐squares support‐vector machines (LS‐SVM) regression model were used to yield the relationships between the absorption spectrums and the content percentages of polydimethylsiloxane (PDMS), alumina trihydrate (ATH), and silica in HTVSR. The results showed that for the formulations tested, the prediction accuracy of all three main ingredients by the PLS regression model could be improved by changing the spectrum range from 0.2–4 to 0.5–2 THz. If the data were pre‐processed by the Savitzky–Golay smoothing method or multiplicative scatter correction method, the prediction accuracy of PDMS could be further enhanced. However, this would lead to a slight decrease in the prediction accuracy of ATH. For the LS‐SVM regression model, the radial basis function (RBF) kernel and the linear kernel were studied. It was found that the prediction accuracy of both kernels was better than that of the PLS regression model. With the LS‐SVM regression model using the RBF kernel, the correlated coefficients of PDMS and ATH in the prediction set could be up to 0.9915 and 0.9742, respectively.

This is an article in the field of mineral processing engineering. The opper polymetallic ore in a high-altitude mining area in Xizang has a high oxidation rate and complex ore properties. In order to make full, efficient and comprehensive use of mineral resources, this article analyzes and studies the mineral properties and characteristics of the raw ore based on theory and practical experience. Finally, the flotation process with JM-1 sulfide agent, ZJ-1 collector, copper, lead and zinc mixed flotation of "sulfur before oxygen" as the key technology is determined. At the condition that the copper oxidation rate of the raw ore is 38.58%, the mixed concentrate grade sum of lead and zinc is 41.17%, and the recovery of copper, lead and zinc is 58.74%, 80.71% and 70.82%, respectively, after two roughing, three cleaning and two scavenging processes, which greatly improves the utilization effect and technical level of dressing. It also provides technical support and favorable basis for the improvement and innovation of mineral processing technology in the later stage.

Abstract Fractures of the contralateral hip may easily occur in elderly patients after an initial hip fracture. The aim of this study was to investigate the clinical characteristics and major predisposing risk factors of contralateral hip fracture after initial hip fracture in the elderly, to provide a clinical basis for preventing contralateral hip fracture. The data of 1586 patients who had sustained first or second hip fractures and had been surgically treated in our department were retrospectively analyzed. Potential predictive factors for contralateral hip fracture and descriptive statistics associated with surgery (such as blood loss, operation time, and length of hospital stay) were recorded. Abstract Background Lung adenocarcinoma (LUAD) is a non-small cell carcinoma. Ribonuclease/angiogenin inhibitor 1 (RNH1) exerts multiple roles in virous cancers. E2F1 is a critical transcription factor involved in the LUAD development. Here, we analyze the expression of RNH1 in LUAD patients, investigate the biological function of RNH1 in LUAD, and demonstrate its potential mechanisms through E2F1 in LUAD. Methods In the present study, we presented the expression of RNH1 in LUAD based on the database and confirmed it by western blot detection of RNH1 in human LUAD tissues. Lentiviral infection was constructed to silence or overexpress RNH1 in NCI-H1395 and NCI-H1437 cells. We assess the role of RNH1 on proliferation in LUAD cells by MTT assay, colony formation assays, and cell cycle detection. Hoechst staining and flow cytometry were used to evaluate the effects of RNH1 on apoptosis of LUAD cells. The function of RNH1 in invasion and migration was investigated by Transwell assay. Dual luciferase assay, ChIP detection, and pull-down assay were conducted to explore the association of E2F1 in the maintenance of RNH1 expression and function. The regulation of E2F1 on the functions of RNH1 in LUAD cells was explored. Mouse experiments were performed to confirm the in-vivo role of RNH1 in LUAD. mRNA sequencing indicated that RNH1 overexpression altered the expression profile of LUAD cells. Results RNH1 expression in LUAD tissues of patients was presented in this work. Importantly, RNH1 knockdown improved the proliferation, migration and invasion abilities of cells and RNH1 overexpression produced the opposite effects. Dual luciferase assay proved that E2F1 bound to the RNH1 promoter (-1064 ∼ -1054, -1514 ∼ -1504) to reduce the transcriptional activity of RNH1. ChIP assay indicated that E2F1 DNA was enriched at the RNH1 promoter (-1148 ∼ -943, -1628 ∼ -1423). Pull-down assays also showed the association between E2F1 and RNH1 promoter (-1148 ∼ -943). E2F1 overexpression contributed to the malignant behavior of LUAD cells, while RNH1 overexpression reversed it. High-throughput sequencing showed that RNH1 overexpression induced multiple genes expression changes, thereby modulating LUAD-related processes. Conclusion Our study demonstrates that binding of E2F1 to the RNH1 promoter may lead to inhibition of RNH1 expression and thus promoting the development of LUAD.

Flexible thermoelectric materials play an important role in smart wearables, such as wearable power generation, self-powered sensing, and personal thermal management. However, with the rapid development of Internet of Things (IoT) and artificial intelligence (AI), higher standards for comfort, multifunctionality, and sustainable operation of wearable electronics have been proposed, and it remains challenging to meet all the requirements of currently reported thermoelectric devices. Herein, we present a multifunctional, wearable, and wireless sensing system based on a thermoelectric knitted fabric with over 600 mm·s−1 air permeability and a stretchability of 120%. The device coupled with a wireless transmission system realizes self-powered monitoring of human respiration through an mobile phone application (APP). Furthermore, an integrated thermoelectric system was designed to combine photothermal conversion and passive radiative cooling, enabling the characteristics of being powered by solar-driven in-plane temperature differences and monitoring outdoor sunlight intensity through the APP. Additionally, we decoupled the complex signals of resistance and thermal voltage during deformation under solar irradiation based on the anisotropy of the knitted fabrics to enable the device to monitor and optimize the outdoor physical activity of the athlete via the APP. This novel thermoelectric fabric-based wearable and wireless sensing platform has promising applications in next-generation smart textiles.

Abstract A key factor limiting the development of silicone rubber insulating materials is their short service life in harsh operating environments. The establishment of a self‐healing system is desired to improve the long‐term operational performance of silicone rubber and effectively extend its service life. In this study, poly(urea‐formaldehyde) microcapsules containing polydimethylsiloxane healing agents are prepared and a dual‐component microcapsule self‐healing system for silicone rubber material is established and optimised. In addition, the key factors affecting the self‐healing performance (such as microcapsule size, rupture rate, and microcapsule doping amount) are investigated. It is found that when SYLGARDTM 184 is used as the healing agent and matrix, with a 4:6 (A:B) doping ratio of dual‐component microcapsules and 20 phr doping amount, the self‐healing efficiency is approximately 51%. Furthermore, increasing the microcapsule rupture rate and the interface strength between the healing agent and the matrix can effectively improve the self‐healing performance. The relationships between key factors and self‐healing performance are theoretically revealed. Based on the relationships, the microcapsule self‐healing system can be appropriately designed. The results of this paper can provide a reference for the development of long‐life silicone rubber insulating materials.

Abstract Despite that surgical resection is widely regarded as the most effective approach to the treatment of liver cancer, its safety and efficacy upon centrally located hepatocellular carcinoma (HCC) remain unsatisfactory. In consequence, seeking an integrated treatment, like combined with adjuvant radiotherapy, to enhance the prognosis of patients is of critical importance. By recruiting patients undergoing surgical resection for centrally located HCC ranging from June 2015 to 2020, they were divided into liver resection combined with adjuvant radiotherapy (LR + RT) and mere liver resection (LR) groups. The calculation of propensity score and model of Cox proportional hazards regression were utilized. 193 patients were recruited in aggregation, containing 88 ones undergoing LR + RT, while 105 handled with LR. RT was verified to be an independent factor of prognosis for relapse (HR 0.60). In propensity-score analyses, significant association existed between adjuvant radiotherapy and better disease-free survival (DFS) (Matched, HR 0.60; Adjustment of propensity score, HR 0.60; Inverse probability weighting, HR 0.63). Abstract As a distributed learning paradigm, federated learning is supposed to protect data privacy without exchanging users’ local data. Even so, the gradient inversion attack, in which the adversary can reconstruct the original data from shared training gradients, has been widely deemed as a severe threat. Nevertheless, most existing researches are confined to impractical assumptions and narrow range of applications. To mitigate these shortcomings, we propose a comprehensive framework for gradient inversion attack, with well-designed algorithms for image and label reconstruction. For image reconstruction, we fully utilize the generative image prior, which derives from wide-used generative models, to improve the reconstructed results, by additional means of iterative optimization on mixed spaces and gradient-free optimizer. For label reconstruction, we design an adaptive recovery algorithm regarding real data distribution, which can adjust previous attacks to more complex scenarios. Moreover, we incorporate a gradient approximation method to efficiently fit our attack for FedAvg scenario. We empirically verify our attack framework using benchmark datasets and ablation studies, considering loose assumptions and complicated circumstances. We hope this work can greatly reveal the necessity of privacy protection in federated learning, while urge more effective and robust defense mechanisms.

Summary: Sensing is a critical function of artificial cells; however, this is challenging to realize using bottom-up approaches. Here, we present a protocol for building protocell membranes that sense cues important for redox biochemistry and signaling by combining synthetic phospholipids and natural lipids. BackgroundObservational data posits a correlation between reproductive traits and nonalcoholic fatty liver disease (NAFLD), but their causal inference is still unclear. This investigation seeks to elucidate the causal influence of reproductive traits on NAFLD and determine the intervening role of health condition and socioeconomic status in these connections.MethodsUtilizing a Mendelian Randomization (MR) approach, this research leveraged a comprehensive dataset from the Genome-wide Association Study (GWAS) database. The study incorporated body mass index, major depression, educational level, household income and Townsend deprivation index as intermediary variables. Initially, a bidirectional two-sample MR study was conducted to explore the genetic associations between reproductive traits and NAFLD. Then, two-step MR analyses were implemented to quantify the extent of mediation by these indicators. The weighted inverse variance method was the primary analytical approach, complemented by several sensitivity analyses to affirm the robustness of the MR assumptions. Finally, these findings were validated in the FinnGen research.ResultsThe bidirectional MR analysis indicated that earlier reproductive traits (age at menarche, age at first sexual intercourse, and age at first birth) were associated with an elevated risk of NAFLD, absent any evidence of the reverse relationship. Body mass index accounted for 35.64% of the association between premature menarche and NAFLD. Additionally, body mass index, major depression, educational level and household income mediated 41.65%, 14.35%, 37.88%, and 18.59% of the connection between early sexual intercourse and NAFLD, respectively. Similarly, these same variables elucidated 36.36%, 15.58%, 41.56%, and 22.73% of the correlation between younger age at first birth and NAFLD.ConclusionOur study elucidated the causal relationships between reproductive traits and NAFLD. Potential underlying mechanisms may involve factors such as body mass index, major depression, educational attainment and household income.

As the second part of the investigation on vibration characteristics of ± 800 kV converter transformers, this part focuses on load conditions. A full-load thermal stability test is carried out on 12 YY and YD converter transformers in a converter station. Vibration, voltage, current, and other important information are collected by online monitoring devices. Vibration amplitude, vibration spectrum, dominant frequency, vibration power ratio of frequency band, vibration power entropy, and odd–even harmonic ratio under different load conditions are calculated and studied to evaluate the vibration state. Theoretical analyses of winding electromagnetic force and vibration acceleration are carried out and results show that the harmonic currents make the dominant frequency shift to 300 Hz or 400 Hz. Most vibration characteristics are sensitive to load rate changes. The odd–even harmonic ratio is sensitive to filter disconnection and reconnection. The results of this study reveal the vibration regularity and corresponding mechanism and can contribute to further study on vibration assessment of ± 800 kV converter transformers under load conditions.

For lack of study on the vibration characteristics of converter transformers, this paper studied the vibration signal analysis method and investigated the vibration characteristics of converter transformers during the commissioning of a ±800 kV converter station. The study comprised two parts including no-load and load conditions. In Part I, converter transformers are under no-load conditions. Vibration data are collected under conditions of no-load closing, no-load voltage rise with converters, and no-load voltage rise with DC lines. The vibration characteristics and mechanisms are studied based on the collected data. Several different vibration characteristics are introduced. Vibration power entropy, vibration total harmonic distortion, and recurrence rate are calculated under no-load conditions. Results show that vibration amplitude, vibration total harmonic distortion, and recurrence rate are sensitive to no-load closing. Vibration power entropy is sensitive to no-load voltage rise with DC lines. All these vibration characteristics are sensitive to converter unlocking. These characteristics show different and significant changes under different no-load conditions. Thus, it is vital to choose appropriate vibration characteristics when assessing the vibration state of converter transformers under different operating conditions. The study examines the vibration characteristics’ physical meanings and their changing mechanisms. It also summarizes distribution intervals and typical values of vibration characteristics. This research reveals the vibration regularity and corresponding mechanism. It provides a reference for vibration state assessment and early fault detection of ±800 kV converter transformers under no-load conditions.

ABSTRACT Objective: Tibial plateau fractures are common intra-articular fractures that pose classification and treatment challenges for orthopedic surgeons. Objective: This study examines the value of 3D printing for classifying and planning surgery for complex tibial plateau fractures. Methods: We reviewed 54 complex tibial plateau fractures treated at our hospital from January 2017 to January 2019. Patients underwent preoperative spiral CT scans, with DICOM data processed using Mimics software. 3D printing technology created accurate 1:1 scale models of the fractures. These models helped subdivide the fractures into seven types based on the tibial plateau's geometric planes. Surgical approaches and simulated operations, including fracture reduction and plate placement, were planned using these models. Results: The 3D models accurately depicted the direction and extent of fracture displacement and plateau collapse. They facilitated the preoperative planning, allowing for precise reconstruction strategies and matching intraoperative details with the pre-printed models. Post-surgery, the anatomical structure of the tibial plateau was significantly improved in all 54 cases. Conclusion: 3D printing effectively aids in the classification and preoperative planning of complex tibial plateau fractures, enhancing surgical outcomes and anatomical restoration. Hepatocellular carcinoma (HCC) is the major cause of cancer-associated mortality worldwide. Despite great advances have been made on the treatment of HCC, the survival rate of patients remains poor. Spindle apparatus coiled-coil protein 1 (SPDL1) is involved in the development of various cancers in humans. However, the role of SPDL1 in HCC remains unclear. In this study, we found high expression of SPDL1 in HCC tissues as compared to normal samples. In vitro, silencing of SPDL1 induced HCC cell apoptosis, and suppressed HCC cell propagation and migration. In vivo, knockdown of SPDL1 inhibited the tumor growth of HCC cells. These findings indicated the tumor-promoting role of SPDL1 in HCC. Mechanistically, we identified farnesyltransferase-beta (FNTB) as the downstream target protein of SPDL1 based on immunoprecipitation and mass spectrometry, which were confirmed by western blotting. Rescue assay determined that FNTB played a tumor promoting role in SPDL1-trigger HCC cell growth. Overexpression of FNTB recovered HCC cell viability and migration in SPDL1 knockdown cells. We also found that silencing of SPDL1 increased the sensitivity of Huh7 cells to sorafenib and lenvatinib, suggesting that SPDL1 is a new therapeutic target in HCC. Collectivity, the present study identified a new axis SPDL1/FNTB involved in the progression of HCC. Hence, SPDL1/FNTB is a potential target for the treatment of HCC.

Background: Diffuse large B-cell lymphoma (DLBCL) is the predominant type of malignant B-cell lymphoma. Although various treatments have been developed, the limited efficacy calls for more and further exploration of its characteristics. Methods: Datasets from the Gene Expression Omnibus (GEO) database were used for identifying the tumor purity of DLBCL. Survival analysis was employed for analyzing the prognosis of DLBCL patients. Immunohistochemistry was conducted to detect the important factors that influenced the prognosis. Drug-sensitive prediction was performed to evaluate the value of the model. Results: VCAN, CD3G, and C1QB were identified as three key genes that impacted the outcome of DLBCL patients both in GEO datasets and samples from our center. Among them, VCAN and CD3G+ T cells were correlated with favorable prognosis, and C1QB was correlated with worse prognosis. The ratio of CD68 + macrophages and CD8 + T cells was associated with better prognosis. In addition, CD3G+T cells ratio was significantly correlated with CD68 + macrophages, CD4 + T cells, and CD8 +T cells ratio, indicating it could play an important role in the anti-tumor immunity in DLBCL. The riskScore model constructed based on the RNASeq data of VCAN, C1QB, and CD3G work well in predicting the prognosis and drug sensitivity. Conclusions: VCAN, CD3G, and C1QB were three key genes that influenced the tumor purity of DLBCL, and could also exert certain impact on drug sensitivity and prognosis of DLBCL patients. Introduction: Human papillomavirus (HPV) integration can induce gene expression dysregulation by destroying higher-order chromatin structure in cervical cancer. Objectives: We established a 13q22 site-specific HPV16 gene knock-in cell model to interrogate the changes in chromatin structure at the initial stages of host cell malignant transformation. Methods: We designed a CRISPR-Cas9 system with sgRNA targeting 13q22 site and constructed the HPV16 gene donor. Cells were cotransfected, screened, and fluorescence sorted. The whole genome sequencing (WGS) was used to confirm the precise HPV16 gene integration site. Western blot and qRT-PCR were used to measure gene expression. In vitro and in vivo analysis were performed to estimate the tumorigenic potential of the HPV16 knock-in cell model. Combined Hi-C, chromatin immunoprecipitation and RNA sequencing analyses revealed correlations between chromatin structure and gene expression. We performed a coimmunoprecipitation assay with anti-PIBF1 antibody to identify endogenous interacting proteins. In vivo analysis was used to determine the role of PIBF1 in the tumor growth of cervical cancer cells. Results: We successfully established a 13q22 site-specific HPV16 gene knock-in cell model. We found that HPV integration promoted cell proliferation, invasion and stratified growth in vitro, and monoclonal proliferation in vivo. HPV integration divided the affected topologically associated domain (TAD) into two smaller domains, and the progesterone-induced blocking factor 1 (PIBF1) gene near the integration site was upregulated, although PIBF1 was not enriched at the domain boundary by CUT-Tag signal analysis. Moreover, PIBF1 was found to interact with the cohesin complex off chromatin to reduce contact domain formation by disrupting the cohesin ring-shaped structure, causing dysregulation of tumorigenesis-related genes. Xenograft experiments determined the role of PIBF1 in the proliferation in cervical cancer cells. Conclusion: We highlight that PIBF1, a potential chromatin structure regulatory protein, is activated by HPV integration, which provides new insights into HPV integration-driven cervical carcinogenesis.

Monkeypox (mpox) outbreak in 2022 has caused more than 91,000 cases, has spread to 115 countries, regions, and territories, and has thus attracted much attention. The stability of poxvirus particles in the environment is recognized as an important factor in determining their transmission. However, few studies have investigated the persistence of poxviruses on material surfaces under various environmental conditions, and their sensitivity to biocides. Here, we systematically measured the stability of vaccinia virus (VACV) under different environmental conditions and sensitivity to inactivation methods via plaque assay, quantitative real-time polymerase chain reaction (qPCR), and Gaussia luciferase (G-luciferase) reporter system. The results show that VACV is stable on the surface of stainless steel, glass, clothing, plastic, towel, A4 paper, and tissue and persists much longer at 4 °C and −20 °C, but is effectively inactivated by ultraviolet (UV) irradiation, heat treatment, and chemical reagents. Our study raises the awareness of long persistence of poxviruses in the environment and provides a simple solution to inactivate poxviruses using common disinfectants, which is expected to help the control and prevention of mpox virus and future poxvirus outbreaks.

Composite insulators have been widely used in transmission lines. After being removed from transmission lines, their housing silicone material cannot degrade naturally. To tackle this problem, this paper proposes an effective method to recycle waste insulators by pyrolysis to obtain mullite $(\mathbf{3}\mathbf{Al}_{2}\mathbf{O}_{3}\cdot \mathbf{2}\mathbf{SiO}_{2})$ with high purity and compact structure. The recycling process studied will not generate toxic products. The thermal degradation process of housing material is investigated by analyzing its degradation products including the gas and residues in detail. The experimental results indicate that the colorant agent Fe2O3 inside the housing material is beneficial for the generation of mullite by decreasing the temperature of mullitization. Besides, since the transitional alumina generated by the dehydration of aluminum hydroxide (ATH) has a smaller diameter and can better dissolute into the silica phases, ATH is a better choice as the additional aluminum resource. By comparing the components, structure, and particle size of grains formed at different calcination temperatures, the proposed pyrolysis temperatures of the two stages are 1400°C and 1600°C, respectively.

Abstract Background We aimed to determine whether and how the combination of acetazolamide and remote ischemic preconditioning (RIPC) reduced the incidence and severity of acute mountain sickness (AMS). Methods This is a prospective, randomized, open-label, blinded endpoint (PROBE) study involving 250 healthy volunteers. Participants were randomized (1:1:1:1:1) to following five groups: Ripc (RIPC twice daily, 6 days), Rapid-Ripc (RIPC four times daily, 3 days), Acetazolamide (twice daily, 2 days), Combined (Acetazolamide plus Rapid-Ripc), and Control group. After interventions, participants entered a normobaric hypoxic chamber (equivalent to 4000 m) and stayed for 6 h. The primary outcomes included the incidence and severity of AMS, and SpO2 after hypoxic exposure. Secondary outcomes included systolic and diastolic blood pressure, and heart rate after hypoxic exposure. The mechanisms of the combined regime were investigated through exploratory outcomes, including analysis of venous blood gas, complete blood count, human cytokine antibody array, ELISA validation for PDGF-AB, and detection of PDGF gene polymorphisms. Results The combination of acetazolamide and RIPC exhibited powerful efficacy in preventing AMS, reducing the incidence of AMS from 26.0 to 6.0% (Combined vs Control: RR 0.23, 95% CI 0.07–0.70, P = 0.006), without significantly increasing the incidence of adverse reactions. Combined group also showed the lowest AMS score (0.92 ± 1.10). Mechanistically, acetazolamide induced a mild metabolic acidosis (pH 7.30 ~ 7.31; HCO3 − 18.1 ~ 20.8 mmol/L) and improved SpO2 (89 ~ 91%) following hypoxic exposure. Additionally, thirty differentially expressed proteins (DEPs) related to immune-inflammatory process were identified after hypoxia, among which PDGF-AB was involved. Further validation of PDGF-AB in all individuals showed that both acetazolamide and RIPC downregulated PDGF-AB before hypoxic exposure, suggesting a possible protective mechanism. Furthermore, genetic analyses demonstrated that individuals carrying the PDGFA rs2070958 C allele, rs9690350 G allele, or rs1800814 G allele did not display a decrease in PDGF-AB levels after interventions, and were associated with a higher risk of AMS. Conclusions The combination of acetazolamide and RIPC exerts a powerful anti-hypoxic effect and represents an innovative and promising strategy for rapid ascent to high altitudes. Acetazolamide improves oxygen saturation. RIPC further aids acetazolamide, which synergistically regulates PDGF-AB, potentially involved in the pathogenesis of AMS. Abstract Purpose Prolonged exposure to plasma free fatty acids (FFAs) leads to impaired glucose tolerance (IGT) which can progress to type 2 diabetes (T2D) in the absence of timely and effective interventions. High-fat diet (HFD) leads to chronic inflammation and oxidative stress, impairing pancreatic beta cell (PBC) function. While Didymin, a flavonoid glycoside derived from citrus fruits, has beneficial effects on inflammation dysfunction, its specific role in HFD-induced IGT remains yet to be elucidated. Hence, this study aims to investigate the protective effects of Didymin on PBCs. Methods HFD-induced IGT mice and INS-1 cells were used to explore the effect and mechanism of Didymin in alleviating IGT. Serum glucose and insulin levels were measured during the glucose tolerance and insulin tolerance tests to evaluate PBC function and insulin resistance. Next, RNA-seq analysis was performed to identify the pathways potentially influenced by Didymin in PBCs. Furthermore, we validated the effects of Didymin both in vitro and in vivo. Mitochondrial electron transport inhibitor (Rotenone) was used to further confirm that Didymin exerts its ameliorative effect by enhancing mitochondria function. Results Didymin reduces postprandial glycemia and enhances 30-minute postprandial insulin levels in IGT mice. Moreover, Didymin was found to enhance mitochondria biogenesis and function, regulate insulin secretion, and alleviate inflammation and apoptosis. However, these effects were abrogated with the treatment of Rotenone, indicating that Didymin exerts its ameliorative effect by enhancing mitochondria function. Conclusions Didymin exhibits therapeutic potential in the treatment of HFD-induced IGT. This beneficial effect is attributed to the amelioration of PBC dysfunction through improved mitochondrial function.

The reconstruction algorithms play an important role in acoustic thermometry and are a research hotspot. This paper proposes a novel reconstruction method to improve the accuracy of temperature field reconstruction methods based on acoustic tomography. First, three mesh models are constructed by using the finite element method, which overcomes the defects of oversimplified classical discretization models. Secondly, the coefficient matrix A of the acoustic thermometry forward problem is calculated offline, and then the temperature values of several points in the measurement are obtained by solving the inverse problem using the improved regularization method based on singular value decomposition. Finally, the temperature profile of the entire region is obtained through interpolation. The correctness and feasibility of the proposed method are validated through simulations and experimental testing with three different temperature fields. Results show that the proposed method has satisfactory reconstruction accuracy and provides a different solution for reconstructing the ultrasound temperature field.

Due to their outstanding anti-flashover characteristics, composite insulators have been extensively applied in power systems. A lot of research has investigated flashover characteristics of hydrophobic specimens with artificial water droplets. However, the phenomenon is not consistent with that of the contaminated composite insulators. On the test specimens covered with water droplets, there is no obvious leakage current before the flashover and no obvious relationship between flashover voltage and the conductivity of water droplets. On the contaminated composite insulator surface, there are short continuous arcs on the insulators before critical flashover, making insulators look like a luminous lantern. Considering that under these two conditions, the proportion of water along the insulation distance is different, the flashover characteristic of “dry band-water band” on a hydrophobicity surface is analyzed in the present study. The influence of the water band parameter (including length, width, and conductivity) as well as the length of dry band is studied. On this basis, the arc generation and development process of the surface covered with “dry band-water band” is analyzed. The research results improve the understanding of the flashover process on contaminated composite insulators.

Shallow underground explosive source localization technology is a key technology in the field of underground space localization. The existing approaches mainly aim to improve the localization accuracy, but need to deploy enormous sensors in the monitoring area, and rely on a large number of backend workstations to solve. These methods have the defects of considerable calculation and high time cost, and are hard to satisfy the precise and real-time requirements of onsite testing, ultimately resulting in slow localization speed and accurate localization failure. Fortunately, emerging deep reinforcement learning can effectively solve the problem of slow search policy by modeling the source localization as a Markov decision process (MDP). Therefore, a curiosity-driven deep dueling double Q-learning network (C-D3QN) is subsequently proposed to solve the above MDP. The overestimation problem is solved by decoupling selection and evaluation of the bootstrap action, and the action difference is effectively increased by introducing the dueling network that separately represents state values and action advantages. Meanwhile, the exploration is jointly reinforced by an intrinsic reward outputted from the curiosity module and an extrinsic reward supplied by the environment, guaranteeing the convergence to global optimal. Finally, extensive simulation results based on the outfield experiment data show that compared with other algorithms, the proposed scheme can significantly improve exploration ability and learning speed as well as generalization and robustness. In addition, compared to the baseline algorithm deep Q-learning network, the C-D3QN algorithm can offer an improved localization accuracy as high as 99.62% and an increased localization speed of 66.23%.

Abstract Early identification of gastric cancer (GC) is associated with a superior survival rate compared to advanced GC. However, the poor specificity and sensitivity of traditional biomarkers suggest the importance of identifying more effective biomarkers. This study aimed to identify novel biomarkers for the prognosis of GC and construct a risk score (RS) signature based on these biomarkers, with to validation of its predictive performance. We used multi-omics data from The Cancer Genome Atlas to analyze the significance of differences in each omics data and combined the data using Fisher's method. Hub genes were subsequently subjected to univariate Cox and LASSO regression analyses and used to construct the RS signature. The RS of each patient was calculated, and the patients were divided into two subgroups according to the RS. The RS signature was validated in two independent datasets from the Gene Expression Omnibus and subsequent analyses were subsequently conducted. Five immune-related genes strongly linked to the prognosis of GC patients were obtained, namely CGB5, SLC10A2, THPO, PDGFRB, and APOD. Introduction: The present study aimed to investigate whether and how normobaric intermittent hypoxic training (IHT) or remote ischemic preconditioning (RIPC) plus normoxic training (RNT) has a synergistic protective effect on lipid metabolism and vascular function compared with normoxic training (NT) in overweight or obese adults. Methods: A total of 37 overweight or obese adults (36.03 ± 10.48 years) were randomly assigned to 3 groups: NT group (exercise intervention in normoxia), IHT group (exercise intervention in normobaric hypoxic chamber), and RNT group (exercise intervention in normoxia + RIPC twice daily). All participants carried out the same 1-h exercise intervention for a total of 4 weeks, 5 days per week. Physical fitness parameters were evaluated at pre- and postexercise intervention. Results: After training, all three groups had a significantly decreased body mass index (p < 0.05). The IHT group had reduced body fat percentage, visceral fat mass (p < 0.05), blood pressure (p < 0.01), left ankle-brachial index (ABI), maximal heart rate (HRmax) (p < 0.05), expression of peroxisome proliferator-activated receptor-γ (PPARγ) (p < 0.01) and increased expression of SIRT1 (p < 0.05), VEGF (p < 0.01). The RNT group had lowered waist-to-hip ratio, visceral fat mass, blood pressure (p < 0.05), and HRmax (p < 0.01). Conclusion: IHT could effectively reduce visceral fat mass and improve vascular elasticity in overweight or obese individuals than pure NT with the activation of SIRT1-related pathways. And RNT also produced similar benefits on body composition and vascular function, which were weaker than those of IHT but stronger than NT. Given the convenience and economy of RNT, both intermittent hypoxic and ischemic training have the potential to be successful health promotion strategies for the overweight/obese population.

To propose a method for testing the dynamic change rate of the throat diameter based on the characteristics of the jet noise field, this study measured the effect of the size variation of the throat diameter on the jet noise during engine nozzle operation. Steady-state flow field simulation and transient noise simulation based on the Ffowcs Williams–Hawkings equation were carried out using large-eddy simulation for nozzles with different sizes of the throat diameter. The jet noise’s sound pressure level (SPL) is discussed and analyzed. By performing a Fourier transform on the noise signal obtained from the simulation, the influence rule of throat diameter size on the magnitude and frequency characteristics of jet noise is analyzed. The results show that the jet noise signal mainly manifests in the low-frequency region. With a gradual increase in frequency, the corresponding amplitude will first increase and then decrease. The effect of the change in the nozzle throat size on the structure of the jet wave system is more prominent; with the increase in the throat diameter, the SPL of the jet noise tends to increase, and the peak frequency of the SPL tends to decrease. The size of the nozzle throat diameter affects the change in the main frequency of the noise, which decreases as the nozzle throat diameter becomes larger.

A tremendous amount of research has witnessed the exploration of two-dimensional (2D) materials with intrinsic ferromagnetism and diverse physical properties. However, the low Curie temperature and deficient magnetic anisotropy hinder their practical applications in nanoscale spintronics. Based on first-principles calculations, we propose a new family of 2D ternary transition metal tellurides, CrX2Te4 (X = Al, Ga, and In), with both structural and magnetic stabilities at room temperature. Our calculations demonstrate that the 2D CrX2Te4 crystal exhibits the intrinsic 100% spin-polarized half-metallic feature with spin-up metallic and spin-down semi-conducting properties. With the remarkable magnetic moment of 4 μB per Cr atom, both 2D CrAl2Te4 and CrGa2Te4 crystals perform robust ferromagnetism with the out-of-plane magnetic anisotropy, while the 2D CrIn2Te4 crystal prefers the in-plane easy magnetization axis. The Monte Carlo simulation based on the 2D Heisenberg model shows that the critical Curie temperatures of the 2D CrAl2Te4, CrGa2Te4, and CrIn2Te4 crystals could reach 466, 431, and 536 K, respectively. Moreover, the magnetic exchange strength and magnetic anisotropy could be further enhanced by the in-plane biaxial strain. The novel electronic and magnetic features promote 2D CrX2Te4 (X = Al, Ga, and In) crystals as a new family of two-dimensional intrinsic ferromagnetic materials for next-generation advanced spintronics.

AbstractTumour-associated endothelial cells (TECs) are a critical stromal cell type in the tumour microenvironment and play central roles in tumour angiogenesis. Notably, TECs have phenotypic plasticity, as they have the potential to transdifferentiate into cells with a mesenchymal phenotype through a process termed endothelial-to-mesenchymal transition (EndoMT). Many studies have reported that EndoMT influences multiple malignant biological properties of tumours, such as abnormal angiogenesis and tumour metabolism, growth, metastasis and therapeutic resistance. Thus, the value of targeting EndoMT in tumour treatment has received increased attention. In this review, we comprehensively explore the phenomenon of EndoMT in the tumour microenvironment and identify influencing factors and molecular mechanisms responsible for EndoMT induction. Furthermore, the pathological functions of EndoMT in tumour progression and potential therapeutic strategies for targeting EndoMT in tumour treatment are also discussed to highlight the pivotal roles of EndoMT in tumour progression and therapy.

Based on data of surface sediment observed in Changdao County, August, 2018, the temporal and spatial characteristic and ecological risk assessment of pollutants in surface sediment were analyzed. The results showed that the contents the average concentrations of sulfide, oil, Cr, Zn, Pb, Cu, As, Cd, Hg and organic carbon were (40.0, 86.8, 38.4, 36.3, 11.2, 15.9, 13.5, 0.146, 0.0211) mg/kg and 0.359 %, respectively. The single factor pollution index p was shown as As, Cr, Cu, Cd, Zn, Pb, organic carbon, oil, sulfide, Hg, and the pollution of heavy metals was more seriously than that of physical and chemical indexes. All stations potential ecological risk were low. Cadmium was determined as the principal ecological risk factor. The contribution rate of Cd to the Hakanson index was 47.0%. Heavy metals and metalloid pollutants of the studied areas mainly came from atmospheric deposition, maritime traffic pollution, non-point breeding sources, domestic sewage pollution, and exploitation of coal and gold resources, etc.

Abstract Metabolic dysfunction-associated fatty liver disease (MAFLD) is frequently linked to type 2 diabetes mellitus (T2DM), and both conditions exacerbate the progression of the other. However, there is currently no standardized treatment or drug for MAFLD. In this study, A MAFLD animal model through a high-fat diet (HFD) along with administration of streptozotocin (STZ), and palmitic acid (PA)-induced AML12 cells were treated by puerarin. The objective of this study was to assess the therapeutic effect of puerarin, a flavonoid substance that possesses various pharmacological properties, on MAFLD. The results showed that puerarin administration enhanced glucose tolerance and insulin sensitivity, while also mitigating liver dysfunction and hyperlipidemia in MAFLD mice. Moreover, puerarin attenuated oxidative stress levels and inflammation in the liver. Transmission electron microscopy and Western blot analysis indicated that puerarin inhibited ferroptosis in vivo. Further mechanistic investigations revealed that puerarin upregulated SIRT1 expression, increased nuclear factor erythroid 2-related factor 2 (Nrf2) protein levels, and facilitated translocation into the nucleus. The protective effect of puerarin on PA-induced AML12 cells was diminished by the utilization of EX-527 (a SIRT1 inhibitor) and Nrf2 siRNA. Overall, the results demonstrate that puerarin ameliorates MAFLD by suppressing ferroptosis and inflammation via the SIRT1/Nrf2 signaling pathway. The results emphasize the possible medicinal application of puerarin for managing MAFLD.

This paper analyzed the horizontal distribution and limitation of DIN, PO4-P and SiO3-Si based on the four surveys of Miaodao Archipelago sea area in August and October of 2018, April and June of 2019. The results showed that the concentration of DIN ranged from 0.82 μmol/L to 95.14 μmol/L with an average value of 5.27 μmol/L and the maximum was in August of 2018. The concentration of PO4-P ranged from "not detected" to 2.12 μmol/L with an average value of 0.19 μmol/L and the maximum was in October of 2018. The concentration of SiO3-Si ranged from 0.25 μmol/L to 48.93 μmol/L with an average value of 5.38 μmol/L and the maximum was in August of 2018. In general, the concentration of nutrients was higher in summer and was relatively scarce in spring. PO4-P was the limiting factor in August and October of 2018, June of 2019, while SiO3-Si and PO4-P were the limiting factors in April of 2019. The PO4-P limitation would limit the primary productivity of sea area and the aquaculture could be influenced. The low content of SiO3-Si was not conducive to the growth of diatoms, which indirectly promoted the reproduction of dinoflagellates, so that this area was easy to cause red tide in spring.

Abstract Thermoelectric sensors have attracted increasing attention in smart wearables due to the recognition of multiple signals in self‐powered mode. However, present thermoelectric devices show disadvantages of low durability, weak wearability, and complex preparation processes and are susceptible to moisture in the microenvironment of the human body, which hinders their further application in wearable electronics. Herein, we prepared a new thermoelectric fabric with thermoplastic polyurethane/carbon nanotubes (TPU/CNTs) by combining vacuum filtration and electrospraying techniques. Electrospraying TPU microsphere coating with good biocompatibility and environmental friendliness made the fabric worn directly and exhibits preferred water resistance, mechanical durability, and stability even after being bent 4000 times, stretched 1000 times, and washed 1000 times. Moreover, this fabric showed a Seebeck coefficient of 49 μV K−1 and strain range of 250% and could collect signals well and avoided interference from moisture. Based on the biocompatibility and safety of the fabric, it can be fabricated into devices and mounted on the human face and elbow for long‐term and continuous collection of data on the body's motion and breathing simultaneously to provide collaborative support information. This thermoelectric fabric‐based sensor will show great potential in advanced smart wearables for health monitoring, motion detection, and human–computer interaction.

In the theoretical calculation of atmospheric wind fields using the cross-correlation analysis method of Medium-Frequency radar, it is necessary to compute a series of correlation parameters from the received echo signals, such as autocorrelation and cross-correlation functions, within the main lobe range of the antenna array to retrieve atmospheric parameters. However, both theoretical analysis and practical applications have shown that the shape of correlation functions can be affected by atmospheric conditions and receiver noise, leading to significant biases in the estimated correlation parameters within the main lobe range. In this study, we theoretically analyze the influence of noise on the amplitude of autocorrelation and cross-correlation functions. We propose a noise reduction method based on the characteristics of correlation functions at the zero-delay point to calculate the noise factor and process the correlation functions within the main lobe range. Furthermore, we conduct simulation analysis to evaluate the performance of this noise reduction method and summarize the effects of the number of fitting points and fitting methods on the noise reduction performance.

With the development of UHV transmission technology, large-tonnage disc-type suspension porcelain insulators have been widely used. Insulators with cylindrical heads have the advantages of light weight and high strength compared with traditional insulators with conical heads. Cement is an important part of disc-type suspension porcelain insulators. The drying shrinkage of cement can lead to sliding between cement and porcelain. It can also transform the stress distribution of the porcelain, thus influencing the overall long-term mechanical performance of the insulators. In this paper, the effect of the shrinkage of cement on the stress distribution of porcelain insulators was studied by simulation. In addition, the effect of the drying shrinkage rates of cement on insulators with cylindrical and conical heads were compared. The simulation results show that cylindrical-head insulators can better resist the stress change caused by cement shrinkage than the conical-head insulators, which was beneficial to the long-term performance of the insulators. Based on the tensile stress of porcelain parts, the recommended range of the drying shrinkage rate of cement for insulators with cylindrical heads was presented. The research results are of practical value for the selection of cement.

Background: Scarce evidence exists for clinical target volume (CTV) definitions of regional lymph nodes (LNs) in intrahepatic cholangiocarcinoma (iCCA) or combined hepatocellular-cholangiocarcinoma (cHCC-CCA). We investigated the mapping pattern of nodal recurrence after surgery for iCCA and cHCC-CCA and provided evidence for the nodal CTV definition. Methods: We retrospectively reviewed the medical records of patients with iCCA or cHCC-CCA who underwent surgery between 2010 and 2020. Eligibility criteria included patients pathologically diagnosed with iCCA or cHCC-CCA after surgery and a first recurrent event in regional LNs during follow-up. All recurrent LNs were registered onto reference computed tomography images based on the vascular structures to reconstruct the node mapping. Fifty-three patients were eligible. LN regions were classified into four risk groups. Results: Hepatic hilar and portal vein-vena cava were the most common recurrent regions, with recurrence rates of 62.3 % and 39.6 % (high-risk regions), respectively. Recurrence rates in the left gastric, diaphragmatic, common hepatic, superior mesenteric vessels, celiac trunk, and paracardial regions ranged from 15.1 % to 30.2 % (intermediate-risk regions). Background & Aims: This study aims to analyze the prognosis of null-margin (≤1.0 mm) hepatectomy (NH) in patients with hepatocellular carcinoma (HCC) adhering to the major vessels and explore the value of postoperative radiotherapy (RT) in these patients. Methods: HCC patients who underwent null-margin or wide-margin (≥1.0 cm) hepatectomy (WH) by our team from January 2008 to March 2016 were recruited and analyzed retrospectively. The patients were divided into the NH, NH + RT, and WH groups. Propensity score matching (PSM) was performed to balance baseline characteristics. Results: A total of 357 patients were recruited. Of these, 84, 49, and 224 patients were given NH alone, NH plus RT, and WH, respectively. After PSM, the 5-year overall survival (OS) and disease-free survival (DFS) rates of the NH group were significantly worse than those of the WH group (51.5 % vs. 71.4 %, P = 0.003; 32.2 % vs. 50.9 %, P = 0.005). The OS and DFS rates of the NH + RT group were significantly higher than those of the NH group (75.6 % vs. 56.1 %, P = 0.012; 46.6 % vs. 30.2 %, P = 0.015) and similar to those of the WH group (75.6 % vs. 75.1 %, P = 0.354; 46.6 % vs. 56.6 %, P = 0.717). In addition, patients in the NH + RT group experienced significantly lower early (P = 0.023) and intrahepatic (P = 0.015) recurrences than those in the NH group. Conclusions: Patients with HCC adhering to the major vessels who underwent NH alone had a poorer prognosis, and the addition of RT to NH provide a significant survival benefit for these patients, which may yield outcomes comparable to the efficacy of WH.

ABSTRACTIn China, 45% of adolescents with obesity develop fatty liver disease, a condition that increases the long-term risk of developing cirrhosis and liver cancer. Although the factors triggering nonalcoholic fatty liver disease (NAFLD) vary in children, the composition of intestinal microflora has been found to play an increasingly important role. However, evidence is limited on the prevalence of nonalcoholic fatty liver (NAFL) and nonalcoholic steatohepatitis (NASH) in Chinese children. Therefore, this study aimed to evaluate the fecal microbiome of Chinese children with NAFLD and further analyze the potential of flora in regulating NAFLD-related symptoms and metabolic functions. Specifically, the study applied a 16S rRNA and metagenomic sequencing to the fecal samples of pediatric patients with NAFLD, NASH, and NAFL, as well as healthy controls, to explore the correlation among NAFLD-related indexes, metabolic pathways, and gut flora. The findings showed that some fecal microbiota had a negative correlation with body mass index, and various NAFLD-related bacteria, including Lachnoclostridium, Escherichia-Shigella, and Faecalibacterium prausnitzii, were detected. Consequently, the study concluded that the variation in gut microbiota might be more important in improving NAFLD/NASH compared with single species, providing a microbiota diagnostic profile of NAFLD/NASH.IMPORTANCEThis study aims to characterize the gut microbiota in Chinese children with nonalcoholic fatty liver disease (NAFLD) through 16S rRNA and metagenomic sequencing. The results highlight the association between fecal microbiota and NAFLD in Chinese children, demonstrating distinct characteristics compared to adults and children from other countries. Based on the sequencing data from our cohort’s fecal samples, we propose a microbiota model with a high area under the curve for distinguishing between NAFLD and healthy individuals. Furthermore, our follow-up study reveals that changes in the relative abundance of microbial biomarkers in this model are consistent with variations in patients’ body mass index. These findings suggest the potential utility of the microbiota model and microbial biomarkers for diagnosing and treating NAFLD in children.

Inspired by the biological visual attention mechanism, we propose a visual attention convolutional neural network to solve the problem of identifying aircraft skin, but showing that the aircraft s kin image collected by the camera is rarely illuminated, and the contrast between defects and background is low and difficult to identify. Firstly, the U-Net with the encoder-decoder structure is used to initially segment the skin image that are provided by Airlines. Then, the residual block is introduced into the U-Net to enhance the propagation ability of the features and extract more defect detail feature information. Finally, the visual attention mechanism is used to increase the weight of the defect area to reduce the influence of uneven illumination on the model. The experimental results show that the proposed model has better defect segmentation effects in both visual effects and objective evaluation indexes. Our algorithm not only enhances the accuracy and effectiveness of skin defect identification but also provides management personnel with a more relevant tool, promoting the intelligence and information of air maintenance. This contributes to excellent management support for aviation companies and related industries.

In practical applications involving robotic arms, particularly in tasks such as manipulating door handles, improper strategies often lead to excessive contact forces. Such forces not only jeopardize the integrity of the robotic arm’s joints but also pose a risk of damaging the door handle. This paper delves into a meticulous study aimed at refining the opening techniques employed by manipulators, enhancing their adaptability across various environments. A novel method is introduced, amalgamating force information feedback with the deep deterministic policy gradient algorithm, fostering a more nuanced approach in trajectory planning. This innovative strategy is meticulously evaluated through simulations and physical experiments, proving instrumental in guiding the robotic arm toward the successful completion of the door-opening task. The findings from the experiments underscore the algorithm’s prowess in cultivating a compliant door-opening strategy, harmonizing with the force applied at the manipulator’s end. A comparative analysis with conventional methods reveals a notable reduction in the end force of the manipulator, facilitating a more efficient and secure execution of door-opening operations.

Abstract Background Although the efficacy and safety of intraoperative radiotherapy (IORT) in the treatment of malignant tumours, such as breast cancer, have been documented, it remains unclear whether this treatment is effective for centrally located hepatocellular carcinoma (HCC) with microvascular invasion (MVI). Aims This study aimed to explore the efficacy and safety of IORT in the treatment of centrally located HCC with MVI. Methods and results Patients with centrally located HCC, who underwent surgery between January 2016 and January 2020, were enrolled. The patient cohort was then allocated to two groups: those who underwent IORT combined with liver resection (IORT+LR); or LR alone (LR). Propensity score matching and Cox proportional hazards regression analyses were performed. The Kaplan–Meier method was used to estimate recurrence‐free survival (RFS), and the log‐rank test was used to determine whether RFS differed between the groups. Subgroup analysis was performed to evaluate differences in RFS and early recurrence rates in patients with different MVI grades. E‐values were generated to measure the sensitivity to unmeasured confounding factors. In total, 97 patients were enrolled, 27 of whom underwent IORT+LR and 70 underwent LR alone. The 1‐, 3‐, and 5‐year RFS rates in the IORT+LR group were 66%, 50%, and 32%, respectively, whereas those in the LR group were 54%, 37%, and 26%, respectively. After matching analysis, 23 patients were successfully matched, and RFS was found to be significantly different between the two groups (p = .04). IORT was an independent prognostic factor for RFS (hazard ratio 0.46 [95% confidence interval 0.21–0.99]). ABSTRACT: The recombinant plasmid pCI-IL-4-IL-2-EGFP containing fusion genes of chicken IL-4 and IL-2 can be used as an adjuvant to enhance the anticoccidiosis effect of the chicken coccidia live vaccine. The chickens were divided into 3 groups: blank control group, vaccine + pCI-IL-4-IL-2-EGFP adjuvant coimmunization group, and vaccine-only group to investigate the immune synergy mechanism of recombinant plasmid adjuvant pCI-IL-4-IL-2-EGFP. The expressions of IL-2, IL-4, TNF-α, and IFN-γ in chicken sera and tissues were detected by ELISA and RT-qPCR, and the proliferation of T and B lymphocytes and antigen presenting cells (APC) in chicken immune organs and intestines were detected by acid alpha-naphthalase (ANAE) staining, methyl green pyronine (MGP) staining, and immunofluorescence (IF) staining, respectively. Results showed that the mRNA expression of IL-2, IL-4, IFN-γ and the number of activated T and B lymphocytes were significantly upregulated in the spleen and cecum tonsils of chickens in vaccine + pCI-IL-4-IL-2-EGFP group compared with the vaccine-only group on 7 d after vaccination (P < 0.05). Protein contents of IL-2, IL-4 and TNF-α in vaccine + pCI-IL-4-IL-2-EGFP group were significantly increased compared to vaccine-only group on 28 d of inoculation (P < 0.05). The number of T and B lymphocytes and APC in chickens of the vaccine+ pCI-IL-4-IL-2-EGFP group was significantly higher than that of the vaccine-only group in cecum tonsils, thymus and spleen after 14 and 28 d of inoculation (P < 0.05). All results revealed that pCI-IL-4-IL-2-EGFP adjuvant enhanced the immune response of chicken coccidia live vaccine by upregulating the expression of IL-2, IL-4, TNF-α, and IFN-γ and promoting the proliferation of T, B lymphocytes and APCs in chicken intestines and immune organ sites. Moreover, our study provides a theoretical basis for the clinical application of cytogenic plasmids as adjuvants.

Mesenchymal stem cells (MSCs) have tumor-homing ability and play critical roles in tumor treatment, but their dual influences on tumor progression limit their therapeutic applications. Exosomes derived from MSCs (MSC-exosomes) exhibit great potential in targeted tumor treatment due to their advantages of high stability, low immunogenicity, good biocompatibility, long circulation time and homing characteristics. Furthermore, the artificial modification of MSC-exosomes could amplify their advantages and their inhibitory effect on tumors and could overcome the limit of tumor-promoting effect. In this review, we summarize the latest therapeutic strategies involving artificially modified MSC-exosomes in tumor treatment, including employing these exosomes as nanomaterials to carry noncoding RNAs or their inhibitors and anticancer drugs, and genetic engineering modification of MSC-exosomes. We also discuss the feasibility of utilizing artificially modified MSC-exosomes as an emerging cell-free method for tumor treatment and related challenges.

BackgroundCumulative fatigue without intervention will seriously threaten the physical and mental health of workers. Shift work and life satisfaction are strongly associated with fatigue accumulation. ObjectiveTo explore the effects of life satisfaction, shift work, and their interaction on cumulative fatigue in petrochemical employees, and to provide a scientific basis for preventing cumulative fatigue. MethodsAll staff of a petrochemical enterprise were selected by cluster sampling for a cross-sectional study from July to October 2021 in Jiangsu Province. A questionnaire designed by the project team was used to collect information on shift work; and life satisfaction and cumulative fatigue were investigated by the World Health Organization Five-item Well-Being Index and the Self-diagnosis Checklist for Assessment of Worker’s Fatigue Accumulation respectively. A logistic regression model was used to analyze the influences of life satisfaction and shift work on cumulative fatigue. Multiplicative and additive models were applied to analyze the interaction effect of life satisfaction and shift work. ResultsA total of 4066 questionnaires were returned, of which 3763 were valid, with an effective recovery rate of 92.5%. The percentage of cumulative fatigue in the petrochemical employees was 63.2% (2377/3763), and the percentages of low life satisfaction and shift work in the petrochemical employees were 53.6% (2016/3763) and 54.2% (2041/3763), respectively. The results of univariate analysis showed no significant difference in cumulative fatigue among different marital status groups (P=0.176), and there were statistically significant differences in cumulative fatigue among the petrochemical employees in different groups of age, gender, educational level, average monthly income, job title, length of service, working hours, night shift, smoking, drinking, physical exercise, life satisfaction, and shift work (P
