7. Reflection on Practice: Pre-Service Teachers' Reflection and Intentional Planning to Enhance Toddlers' Engagement during Free Play

Abstract

In this paper, early childhood teacher educators describe their work to provide opportunities for pre-service teacher candidates to engage in specific practices that facilitate reflection and planning related to enhancing toddlers' engagement during free play. The practices targeted were structured by the instructor and supported by the mentor teacher and included observation; guided written reflection; collective discussion and reflection; and intentional planning. Each practice and how it was implemented and completed by the instructor and students are discussed. As a result of participating in the practices, teacher candidates identified several factors they believed impacted toddlers' engagement during free play. They included location of centers, materials, and other people, variety of materials, adult interaction, and children's interests, all of which aligned with prior research. Implications for practice for pre-service teacher preparation programs and practicing teachers are provided.

Abstract

BACKGROUND: Autism spectrum disorder involves neurodevelopmental dysregulations that lead to visible symptoms at early stages of life. Many autism spectrum disorder-related mechanisms suggested by animal studies are supported by demonstrated improvement in autistic-like phenotypes in adult animals following experimental reversal of dysregulated mechanisms. However, whether such mechanisms also act at earlier stages to cause autistic-like phenotypes is unclear. METHODS: We used Shank2-/- mice carrying a mutation identified in human autism spectrum disorder (exons 6 and 7 deletion) and combined electrophysiological and behavioral analyses to see whether early pathophysiology at pup stages is different from late pathophysiology at juvenile and adult stages and whether correcting early pathophysiology can normalize late pathophysiology and abnormal behaviors in juvenile and adult mice. RESULTS: Early correction of a dysregulated mechanism in young mice prevents manifestation of autistic-like social behaviors in adult mice. Shank2-/- mice, known to display N-methyl-D-aspartate receptor (NMDAR) hypofunction and autistic-like behaviors at postweaning stages after postnatal day 21 (P21), show the opposite synaptic phenotype-NMDAR hyperfunction-at an earlier preweaning stage (∼P14). Moreover, this NMDAR hyperfunction at P14 rapidly shifts to NMDAR hypofunction after weaning (∼P24). Chronic suppression of the early NMDAR hyperfunction by the NMDAR antagonist memantine (P7-P21) prevents NMDAR hypofunction and autistic-like social behaviors from manifesting at later stages (∼P28 and P56). CONCLUSIONS: Early NMDAR hyperfunction leads to late NMDAR hypofunction and autistic-like social behaviors in Shank2-/- mice, and early correction of NMDAR dysfunction has the long-lasting effect of preventing autistic-like social behaviors from developing at later stages.

Abstract

Anti‐inflammatory protein (AIP)‐1 and AIP‐2, identified as parasite excretory/secretory (ES) proteins, play a crucial role in promoting the survival of the parasite and evading the host immunological response. Both proteins inhibit inflammatory reactions, induce apoptosis in effector cells, and influence the phenotype of the immune response. Numerous parasite‐derived proteins have shown promise as therapeutic targets for inflammatory and allergic diseases. Despite this, the precise biological roles, and molecular characteristics of many such proteins remain unclear. In this study, AIP‐1 and AIP‐2 were produced in the baculovirus‐insect cell expression system (BEVS). The multiplicity of infection (MOI) and the duration of infection conditions for AIP‐1 and AIP‐2 protein expression were successfully optimized to induce the highest expression levels of AIP‐1 and AIP‐2 proteins in insect cells. The insect cell‐derived AIP‐1 and AIP‐2 exhibited inhibitory functions against human matrix metalloproteinases (MMPs). This study demonstrates that insect cell‐derived AIP‐1 and AIP‐2 have the potential as therapeutic proteins for MMP‐TIMP (Metalloprotease‐Tissue inhibitor of metalloprotease) axis related disease. Abstract

Delpazolid (LCB01‐0371) is a novel oxazolidinone derivative with a good safety profile for treating gram‐positive pathogenic infections such as Mycobacterium abscessus, a highly pathogenic drug‐resistant Mycobacterium. In this study, we evaluated the pharmacokinetics (PK) and pharmacodynamics (PD) of delpazolid after 14 days of multiple oral administration, using data from adult patients with pulmonary tuberculosis. 800 mg once a day, 400 mg twice a day, 800 mg twice a day, and 1200 mg once a day delpazolid for 14 days were tested in 63 patients with pulmonary tuberculosis. For PK blood collection, inpatient and outpatient scheduling were separately implemented. Plasma concentrations of delpazolid were measured at visits 2, 4, 6, and 8 in outpatients, and four sparse blood samples were measured in inpatients. PD models were sequentially fitted using individual PK parameter estimates obtained from PK compartmental models. For PK modeling, 180 plasma concentrations of delpazolid from 56 patients were included. A two‐compartment mixed first‐ and zero‐order absorption model best described the time course of plasma concentration. For the PD model, 448 bacterial titer data from 60 patients were used. The time course of bacterial titers (log10 CFU/mL) was described by a model that consists of the growth and killing rate of bacteria with the sigmoid Emax model. The PK–PD simulation suggested that the bacterial titers are the lowest on the 800 mg bid regimen among the four, consistent with observed data, as all regimens substantially decrease.
