Abstract

In order to assess the influencing factors of the presence of mercury in a river within the Savanna biome (Cerrado), we surveyed total mercury (THg) in bottom sediment from 50 lakes along 750 km of the Middle Araguaia floodplain. The sampling sites included non-urban and urban surroundings over three distinct geomorphologies. We measured water physicochemical parameters at each site and tested statistically if land use nested within the geological formation influenced the THg concentration in bottom sediments and related water parameters. Multivariate results indicate that the interaction between geological groups and land use is statistically significant (p < 0.05). Nested ANOVA and Tukey HSD tests confirmed that the geological formation with its nested land use influences the THg, pH, DO, conductivity, and TDS (p < 0.05). THg was significantly lower in Quaternary terrains (p < 0.05) and differed significantly between non-urban and urban areas in Neoproterozoic terrains (p = 0.02). The spatial projections of the THg eigenvector on the main axes with the scoring factors of the Neoproterozoic/Paleoproterozoic terrains, and urban/non-urban, confirmed the spatial correlations. These results indicate that the association of land use and geology could be the main driver of THg in the bottom sediments of lakes from the Middle Araguaia floodplain.

Abstract

Pneumonia is a kind of inflammatory disease characterized by pathogen infection of lower respiratory track. Lipopolysaccharide (LPS) is the main bioactive component of Gram-negative bacteria responsible for inflammatory response. Recently, coniferyl aldehyde (CA) has been reported to play a crucial role because of its anti-inflammatory activity. However, the effect and mechanisms of CA in ameliorating symptoms of acute pneumonia remain unknown. Evaluating and identifying the value and exploring the mechanisms of CA on LPS-mediated WI-38 apoptosis and inflammation were the aims of this study. Here, CCK-8 cell viability assay was applied on WI-38 after treatment with or without LPS at different doses of CA to verify that CA can increase LPS-induced cell viability. Then, quantitative polymerase chain reaction (qPCR) and enzyme-linked-immunosorbent serologic assays (ELISA) suggested that LPS treatment dramatically decreased the expression level of IL-10 (anti-inflammatory factor) while strikingly increasing the expression levels of IL-1β, IL-6, and TNF-α (tumor necrosis factor-α; proinflammatory factor) whereas CA treatment attenuates LPS-induced inflammation of WI-38. Further, flow cytometry and Western blot assay verified that LPS treatment dramatically promoted apoptosis of WI-38 cells, while administration of CA notably inhibited apoptosis of WI-38 cells. Moreover, the Western blot assay hinted that CA could inactivate LPS-induced JAK2–STAT1 signaling pathway. These findings indicated that CA could alleviate LPS-mediated WI-38 apoptosis and inflammation injury through JAK2–STAT1 pathway in acute pneumonia.

Abstract

The clinical history is of importance in the investigation of allergic diseases but does have limitations. Many allergic conditions will be over-diagnosed if anamnesis alone is used for diagnostic criteria. Serum total immunoglobulin E (TIgE) quantification, as well as panels containing allergens prevalent in the studied population, may serve as screening tests and facilitate the diagnosis of allergic disease or its exclusion. We assessed the positivity of two versions of these tests, Phadiatop Europe® (PhEU) and Phadiatop Infant® (PhInf), as well as total IgE (TigE) values in patients with a medical diagnosis of allergic disease and non-allergic individuals. Methods A cross-sectional study performed in eleven Brazilian pediatric allergy centers with patients divided into groups according to the primary condition and a group of assessed control subjects. They were submitted to TIgE measurement and screening tests (PhEu and PhInf). Results TIgE mean serum levels were significantly higher among allergic patients, especially those with asthma/rhinitis or atopic dermatitis. The positivity of the screening tests, considering the total population, was 63.8% for PhEU and 72.6% for PhInf. These increased when we evaluated only the allergic subjects. The concordance index of the two tests was Kappa = 0.7 and higher among those of greater age. Conclusions In the assessed population, there were significantly higher levels among those with positive screening tests and PhInf showed better performance in the identification of sensitized individuals, regardless of age. This is the first study to evaluate Phadiatop and Phadiatop Infant in the same population.

Abstract

The Amazon rainforest and the Brazilian Cerrado both possess high phlebotomine diversity. The fragmentation of these habitats has resulted in the appearance of human cases of cutaneous leishmaniasis. In one altered area of mixed primary vegetation (forest and Cerrado) and its adjacent settlement in the northeast state of Maranhão, Brazil, evidence exists for the active transmission of cutaneous leishmaniasis. Accordingly, an entomological investigation was performed in both the forest and the settlement to compare the phlebotomine vector faunain each environment. The study was conducted from September 2009 to August 2010 in the municipality of Itapecuru Mirim in the state of Maranhão, Brazil. The phlebotomine species were captured using 24 light Center for Disease Control and Prevention traps that were placed in the forest and the settlement (peridomicile and intradomicile). The similarity between the phlebotomine compositions in the forest and those in the settlement was determined using a Principal Coordinate Analysis based on a dissimilarity matrix that was calculated using the Bray-Curtis index (relative abundance) and the Jaccard index (presence and absence of species). In total, 29 Lutzomyia species and one Brumptomyia species were collected. The phlebotomines were diverse and abundant in both the forest fragment (27 species, 4,606 specimens) and the settlement (22 species, 753 specimens). The most abundant species were L. infraspinosa (25%), L. davisi (21%), L. antunesi (21%), L. longipalpis (9%), L. saulensis (6%), L. flaviscutellata (5%), and L. wellcomei (4%). Some species were found strictly in the forest, other species were exclusive to the anthropic environment, and some species colonized both of the studied environments. The phlebotomines adaptation to these modified environments explains the autochthonous outbreak of cutaneous leishmaniasis.

Abstract Background: Infantile hemangiomas (IH) occur in approximately 4% to 10% of the pediatric population. The identification of clinical subtypes and conditions that indicate increased risk for complications is essential for therapeutic success. Objectives: To identify risk factors for complications, recurrence and unaesthetic sequelae. Methods: Retrospective cohort of patients with infantile hemangiomas undergoing follow-up at the Dermatology Service of Universidade Federal de Ciências da Saúde de Porto Alegre, between 2006 and 2018. Results: 190 patients were included; 24% had some type of complication, ulceration being the most frequent, and 86% required treatment. On correlation, ulceration was statistically related to mixed IH (p = 0.004), segmental IH (p < 0.01) and location in the gluteal region (p = 0.001). The mean time of treatment with propranolol was 12.7 months. Patients with PHACES syndrome and segmental infantile hemangioma required longer treatment (p < 0.001 and p = 0.0407, respectively), as well as those who started treatment after five months of life (p < 0.0001). Recurrence occurred in 16.6% of the treated patients, all-female; 94% were located on the head and neck (mainly on the upper eyelid, cyrano, S3 segment, and with parotid involvement); 61% and 38.8% were of the mixed and deep subtypes, respectively. Approximately 1/3 of the patients had some unaesthetic sequelae. Study limitations: As this is a retrospective study, data and photos of some patients were lost. Conclusions: Mixed and segmental hemangiomas are risk factors for ulceration and sequelae. Recurrence occurs more often in females and segmental hemangiomas. Segmental infantile hemangioma and PHACES syndrome require a longer time of treatment. Specific protocols are required for infantile hemangiomas with a high risk of recurrence.

Abstract

Introdução: A síndrome de Stevens-Johnson e a necrólise epidérmica tóxica (NET) são reações cutâneas severas, associadas a lesões mucosas e comprometimento sistêmico. Os glicocorticoides foram empregues por muitos anos no tratamento da síndrome de Stevens-Johnson/necrólise epidérmica tóxica, mas não há evidências que sustentem esta conduta. Objetivo: Avaliação da mortalidade em casos de síndrome de Stevens-Johnson/necrólise epidérmica tóxica em um grupo de pacientes que fez uso de glicocorticoides sistêmico e em outro grupo não fez uso. Objetivo secundário: Descrição da população estudada; avaliação de infecção secundária e complicações extra-cutâneas nos dois grupos de pacientes. Material e Métodos: Coorte retrospectiva de pacientes avaliados por meio de consultorias solicitadas ao Serviço de Dermatologia do Hospital Nossa Senhora da Conceição, bem como pelo diagnóstico na internação ou óbito, durante o período de Março de 2007 a Agosto de 2014. Resultados: Mortalidade foi de 63.2% no grupo que usou corticoide e 9,1% no grupo que não usou corticoide, com p valor de 0,005 (IC 95% 1.79-163.8). Análise multivariada ajustada pelo SCORTEN demonstrou apenas uma tendência, com odds ratio para mortalidade no grupo de pacientes que usou glicocorticoides sendo de 7.38 (IC 95% 0.87-161.43) e p valor de 0.06. Conclusão: Este estudo sugere uma tendência de malefício relacionado ao uso de glicocorticoides para o tratamento dessas doenças, principalmente no que diz respeito à mortalidade.
