Abstract Endo-periodontal lesions (EPLs) involve both the periodontium and pulp tissue and have complicated etiologies and pathogenic mechanisms, including unique anatomical and microbiological characteristics and multiple contributing factors. This etiological complexity leads to difficulties in determining patient prognosis, posing great challenges in clinical practice. Furthermore, EPL-affected teeth require multidisciplinary therapy, including periodontal therapy, endodontic therapy and others, but there is still much debate about the appropriate timing of periodontal therapy and root canal therapy. By compiling the most recent findings on the etiology, pathogenesis, clinical characteristics, diagnosis, therapy, and prognosis of EPL-affected teeth, this consensus sought to support clinicians in making the best possible treatment decisions based on both biological and clinical evidence.

Lettuce is a commonly consumed vegetable worldwide, due to its crisp edible organs and rich nutrition. Heat stress is known to seriously threaten its growth and development. However, research on lettuce response to heat stress focusing on molecular aspects is limited. Therefore, we aimed to understand the complex molecular mechanisms underlying the regulation of lettuce plant response to heat stress (37 °C), by transcriptomic analysis of a heat-resistant lettuce variety (K482) following heat stress treatment for 1.5 and 3 days. A total of 3798 and 4121 differentially expressed genes (DEGs) were identified at 1.5 and 3 days vs. the control after heat treatment, respectively; they were mostly enriched in protein processing in the systemic lupus erythematosus, alcoholism, neurotrophin signaling pathways, and the cell cycle. Most DEGs were NACs, HSPs, MYBs, bHLHs, and WRKYs. Transcriptome analysis showed that the NAC domain transcription factor LsNAC28, which is associated with heat response, is up-regulated during heat stress and LsNAC28 inhibited yeast growth under heat stress (37 °C). These findings provide a basis to comprehensively understand the complex molecular mechanism of lettuce response to heat stress.

Abstract The popular leafy vegetable lettuce (Lactuca sativa L.) is susceptible to cold stress during the growing season, which slows growth rate, causes leaf yellowing and necrosis, and reduced yield and quality. In this study, transcriptomic and metabolomic analyses of two cold-resistant lettuce cultivars (GWAS-W42 and F11) and two cold-sensitive lettuce cultivars (S13K079 and S15K058) were performed to identify the mechanisms involved in the cold response of lettuce. Overall, transcriptome analysis identified 605 differentially expressed genes (DEGs), including significant enrichment of genes involved in the flavonoid and flavonol (CHS, CHI, F3H, FLS, CYP75B1, HCT, etc.) biosynthetic pathways related to oxidation–reduction and catalytic activity. Untargeted metabolomic analysis identified fifteen flavonoid metabolites and 28 other metabolites potentially involved in the response to cold stress; genistein, quercitrin, quercetin derivatives, kaempferol derivatives, luteolin derivatives, apigenin and their derivatives accumulate at higher levels in cold-resistant cultivars. Moreover, MYBs, bHLHs, WRKYs and Dofs also play positive role in the low temperature response, which affected the expression of structural genes contributing to the variation of metabolites between the resistant and sensitive. These results provide valuable evidence that the metabolites and genes involved in the flavonoid biosynthetic pathway play important roles in the response of lettuce to cold stress.

Species of the family Polycephalomycetaceae grow on insects or entomopathogenic fungi and are distributed from tropical to subtropical regions. This study proposed four new species of hyperparasitic fungi from China based on six molecular markers (ITS, SSU, LSU, TEF-1α, RPB1 and RPB2) phylogenetic analyses and morphological characteristics. The four new species, i.e. Pleurocordyceps litangensis, Polycephalomyces jinghongensis, Po. multiperitheciatae and Po. myrmecophilus, were described and illustrated. Pl. litangensis, exhibiting a hyperparasitic lifestyle on Ophiocordyceps sinensis, differed from Pleurocordyceps other species in producing subulate β-phialides and ovoid or elliptic α-conidia. Po. jinghongensis was distinct from Polycephalomyces other species, being parasitic on Ophiocordyceps sp., as producing oval or long oval-shaped α-conidia and columns of β-conidia. Po. multiperitheciatae differed from Polycephalomyces other species as having synnemata with fertile head, linear β-conidia and parasitic on Ophiocordyceps multiperitheciata. Po. myrmecophilus was distinct from Polycephalomyces other species, being parasitic on the fungus Ophiocordyceps acroasca, as producing round or ovoid α-conidia and elliptical β-conidia without synnemata from the colonies. These four species were clearly distinguished from other species in the family Polycephalomycetaceae by phylogenetic and morphological characteristics. The morphological features were discussed and compared to relevant species in the present paper.

Abstract Background As the most common subtype of adult muscular dystrophy worldwide, large cohort reports on myotonic dystrophy type I (DM1) in China are still lacking. This study aims to analyze the genetic and clinical characteristics of Chinese Han DM1 patients. Methods Based on the multicenter collaborating effort of the Pan-Yangtze River Delta Alliance for Neuromuscular Disorders, patients with suspected clinical diagnoses of DM1 were genetically confirmed from January 2020 to April 2023. Peak CTG repeats in the DMPK gene were analyzed using triplet repeat-primed PCR (TP-PCR) and flanking PCR. Time-to-event analysis of onset age in females and males was performed. Additionally, detailed clinical features and longitudinal changes from the disease onset in 64 DM1 patients were retrospectively collected and analyzed. The Epworth Sleepiness Scale and Fatigue Severity Scale were used to quantify the severity of daytime sleepiness and fatigue. Results Among the 211 genetically confirmed DM1 patients, the mean age at diagnosis was 40.9 ± 12.2 (range: 12–74) with a male-to-female ratio of 124:87. The average size of CTG repeats was 511.3 (range: 92–1945). Among the DM1 patients with comprehensive clinical data (n = 64, mean age 41.0 ± 12.0), the age at onset was significantly earlier in males than in females (4.8 years earlier, p = 0.026). Muscle weakness (92.2%), myotonia (85.9%), and fatigue (73.4%) were the most prevalent clinical features. The predominant involved muscles at onset are hands (weakness or myotonia) (52.6%) and legs (walking disability) (42.1%). Of them, 70.3% of patients had daytime sleepiness, 14.1% had cataract surgery, 7.8% used wheelchairs, 4.7% required ventilatory support, and 1.6% required gastric tubes. Regarding the comorbidities, 4.7% of patients had tumors, 17.2% had diabetes, 23.4% had dyspnea, 28.1% had intermittent insomnia, 43.8% experienced dysphagia, and 25% exhibited cognitive impairment. Chinese patients exhibited smaller size of CTG repeats (468 ± 139) than those reported in Italy (613 ± 623), the US (629 ± 386), and Japan (625 [302, 1047]), and milder phenotypes with less multisystem involvement. Conclusion The Chinese Han DM1 patients presented milder phenotypes compared to their Caucasian and Japanese counterparts. A male predominance and an early age of onset were identified in male Chinese Han DM1 patients.

ObjectivesWater-saving and drought-resistance rice (WDR) plays a vital role in the sustainable development of agriculture. Nevertheless, the impacts and processes of water and nitrogen on grain yield in WDR remain unclear.MethodsIn this study, Hanyou 73 (WDR) and Hyou 518 (rice) were used as materials. Three kinds of nitrogen fertilizer application rate (NFAR) were set in the pot experiment, including no NFAR (nitrogen as urea applied at 0 g/pot), medium NFAR (nitrogen as urea applied at 15.6 g/pot), and high NFAR (nitrogen as urea applied at 31.2 g/pot). Two irrigation regimes, continuous flooding cultivation and water stress, were set under each NFAR. The relationships between root and shoot morphophysiology and grain yield in WDR were explored.ResultsThe results demonstrated the following: 1) under the same irrigation regime, the grain yield of two varieties increased with the increase of NFAR. Under the same NFAR, the reduction of irrigation amount significantly reduced the grain yield in Hyou 518 (7.1%–15.1%) but had no substantial influence on the grain yield in Hanyou 73. 2) Under the same irrigation regime, increasing the NFAR could improve the root morphophysiology (root dry weight, root oxidation activity, root bleeding rate, root total absorbing surface area, root active absorbing surface area, and zeatin + zeatin riboside contents in roots) and aboveground physiological indexes (leaf photosynthetic rate, non-structural carbohydrate accumulation in stems, and nitrate reductase activity in leaves) in two varieties. Under the same NFAR, increasing the irrigation amount could significantly increase the above indexes in Hyou 518 (except root dry weight) but has little effect on Hanyou 73. 3) Analysis of correlations revealed that the grain yield of Hyou 518 and Hanyou 73 was basically positively correlated with aboveground physiology and root morphophysiology, respectively.ConclusionThe grain yield could be maintained by water stress under medium NFAR in WDR. The improvement of root morphophysiology is a major factor for high yield under the irrigation regime and NFAR treatments in WDR.

Ophiocordyceps unilateralis sensu lato is a common pathogenic fungus of ants. A new species, O. fusiformispora, was described based on morphology and phylogenetic evidence from five genes (SSU, LSU, TEF1α, RPB1, and RPB2). The whole genomes of O. fusiformispora, O. contiispora, O. subtiliphialida, O. satoi, O. flabellata, O. acroasca, and O. camponoti-leonardi were sequenced and annotated and compared with whole genome sequences of other species in O. unilateralis sensu lato. The basic genome-wide characteristics of the 12 species showed that the related species had similar GC content and genome size. AntiSMASH and local BLAST analyses revealed that the number and types of putative SM BGCs, NPPS, PKS, and hybrid PKS-NRPS domains for the 12 species differed significantly among different species in the same genus. The putative BGC of five compounds, namely, NG-391, lucilactaene, higginsianin B, pyripyropene A, and pyranonigrin E were excavated. NG-391 and lucilactaene were 7-desmethyl analogs of fusarin C. Furthermore, the 12 genomes had common domains, such as KS-AT-DH-MT-ER-KR-ACP and SAT-KS-AT-PT-ACP-ACP-Te. The ML and BI trees of SAT-KS-AT-PT-ACP-ACP-Te were highly consistent with the multigene phylogenetic tree in the 12 species. This study provided a method to obtain the living culture of O. unilateralis sensu lato species and its asexual formed on the basis of living culture, which was of great value for further study of O. unilateralis sensu lato species in the future, and also laid a foundation for further analysis of secondary metabolites of O. unilateralis sensu lato.

Through backcrossing and marker-assisted selection, gene Pi9 for resistance to rice blast was introduced into the water-saving and drought-resistant rice variety, Hanhui 3. The genetic background identity between Hanhui 8200 and Hanhui 3 was 91.4%. The drought resistance and drought avoidance abilities of Hanhui 8200 were equivalent to those of Hanhui 3. The resistance to rice blast was improved from grade 7 to grade 1. The rice quality of Hanhui 8200 meets the Ministry of Agriculture’s grade 3 rice standards. The two-line and three-line hybrids formulated with Hanhui 8200 have high yield potential. Among them, the three-line hybrid Hanyou 8200 (Approval No.: Evaluated Rice 20210073), formulated with Huhan 7A, passed the Hubei Provincial approval in 2021, and the two-line hybrid Hanyouliangyou 8200 (Approval No.: Nationally Validated Rice 20210448), formulated with Huhan 82S, passed the national variety approval in 2021. Both hybrids demonstrated strong resistance to rice blast, moderate resistance to bacterial leaf blight, strong drought resistance, high quality, and high yield.

Red pandas evolved from carnivores to herbivores and are unique within Carnivora. Red pandas and carnivorous mammals consume milk during the suckling period, while they consume bamboo and meat during the adult period, respectively. Red pandas and carnivorous mammal ferrets have a close phylogenetic relationship. To further investigate the molecular mechanisms of dietary changes and nutrient utilization in red pandas from suckling to adult, comparative analysis of the whole transcriptome was performed on stomach tissues from red pandas and ferrets during the suckling and adult periods. The main results are as follows: (1) we identified ncRNAs for the first time in stomach tissues of both species, and found significant expression changes of 109 lncRNAs and 106 miRNAs in red pandas and 756 lncRNAs and 109 miRNAs in ferrets between the two periods; (2) up-regulated genes related to amino acid transport regulated by lncRNA-miRNA-mRNA networks may efficiently utilize limited bamboo amino acids in adult red pandas, while up-regulated genes related to amino acid degradation regulated by lncRNAs may maintain the balance of amino acid metabolism due to larger daily intakes in adult ferrets; and (3) some up-regulated genes related to lipid digestion may contribute to the utilization of rich nutrients in milk for the rapid growth and development of suckling red pandas, while up-regulated genes associated with linoleic acid metabolism regulated by lncRNA-miRNA-mRNA networks may promote cholesterol decomposition to reduce health risks for carnivorous adult ferrets. Collectively, our study offers evidence of gene expression adaptation and ncRNA regulation in response to specific dietary changes and nutrient utilization in red pandas during suckling and adult periods.

An animal model harboring pathogenic mitochondrial DNA (mtDNA) mutations is important to understand the biological links between mtDNA variation and mitochondrial diseases. DdCBE, a DddA-derived cytosine base editor, has been utilized in zebrafish, mice, and rats for tC sequence-context targeting and human mitochondrial disease modeling. However, human pathogenic mtDNA mutations other than the tC context cannot be manipulated. Here, we screened the combination of different DdCBE pairs at pathogenic mtDNA mutation sites with nC (n for a, g, or c) context and identified that the left-G1333C (L1333C) + right G1333N (R1333N) pair could mediate C⋅G-to-T⋅A conversion effectively at aC sites in rat C6 cells. The editing efficiency at disease-associated mtDNA mutation sites within aC context was further confirmed to be up to 67.89% in vivo. Also, the installed disease-associated mtDNA mutations were germline transmittable. Moreover, the edited rats showed impaired cardiac function and mitochondrial function, resembling human mitochondrial disease symptoms. In summary, for the first time, we expanded the DdCBE targeting scope to an aC motif and installed the pathogenic mutation in rats to model human mitochondrial diseases.

Abstract Background Deep rooting is an important factor affecting rice drought resistance. However, few genes have been identified to control this trait in rice. Previously, we identified several candidate genes by QTL mapping of the ratio of deep rooting and gene expression analysis in rice. Results In the present work, we cloned one of these candidate genes, OsSAUR11, which encodes a small auxin-up RNA (SAUR) protein. Overexpression of OsSAUR11 significantly enhanced the ratio of deep rooting of transgenic rice, but knockout of this gene did not significantly affect deep rooting. The expression of OsSAUR11 in rice root was induced by auxin and drought, and OsSAUR11-GFP was localized both in the plasma membrane and cell nucleus. Through an electrophoretic mobility shift assay and gene expression analysis in transgenic rice, we found that the transcription factor OsbZIP62 can bind to the promoter of OsSAUR11 and promote its expression. A luciferase complementary test showed that OsSAUR11 interacts with the protein phosphatase OsPP36. Additionally, expression of several auxin synthesis and transport genes (e.g., OsYUC5 and OsPIN2) were down-regulated in OsSAUR11-overexpressing rice plants. Conclusions This study revealed a novel gene OsSAUR11 positively regulates deep rooting in rice, which provides an empirical basis for future improvement of rice root architecture and drought resistance.

Abstract Rapeseed meal, a nutritious organic fertilizer (OF), contributes to improving soil environment and crop productivity. However, there are also problems, namely slow fertilizer efficiency and low nutrient utilization during the growing season. This 2‐year field trial was conducted to explore the effect of biochar addition on improving the nutrient availability of OF through a comparative study of various biochar application rates under rice‐rapeseed rotation conditions. The findings revealed that, compared to the individual application of chemical fertilizers (CF), OF alone decreased rice yield (2%/2%) and rapeseed yield (6%/10%) in 2019/2020. Compared with OF, combining biochar (15 t ha−1) with OF (OF + B15) significantly increased rice yield (17%/10%) and rapeseed yield (25%/20%) in the first/second year. Additionally, OF + B15 still increased rice yield (14%/7%) and rapeseed yield (12%/13%) for two consecutive years compared to CF. The co‐application of biochar and OF had positive impacts on soil physicochemical properties and enzymes. Compared to OF, OF + B15 elevated soil organic carbon (SOC) by 57%–81%, soil catalase 19%, invertase 14%–20%, urease 17%–19%, and phosphatase 13%–17% during rice season, and similarly increased SOC by 77%–90%, soil catalase 14%–16%, invertase 14%–20%, urease 18%–24%, and phosphatase 16%–17% in rapeseed season. Biochar addition improved soil conditions and enzymatic activities, and the available nutrient supply of OF. Also, the co‐application of biochar and rapeseed meal surpassed the effect of chemical fertilizer alone on the growth and yield of crops. Therefore, biochar coupling with organic fertilizer is an effective fertilization strategy based on resource recycling, which promotes both crop yield and sustainable agriculture.

Seed dormancy and germination play pivotal roles in the agronomic traits of plants, and the degree of dormancy intuitively affects the yield and quality of crops in agricultural production. Seed priming is a pre-sowing seed treatment that enhances and accelerates germination, leading to improved seedling establishment. Seed priming technologies, which are designed to partially activate germination, while preventing full seed germination, have exerted a profound impact on agricultural production. Conventional seed priming relies on external priming agents, which often yield unstable results. What works for one variety might not be effective for another. Therefore, it is necessary to explore the internal factors within the metabolic pathways that influence seed physiology and germination. This review unveils the underlying mechanisms of seed metabolism and germination, the factors affecting seed dormancy and germination, as well as the current seed priming technologies that can result in stable and better germination.

Several Pleurocordyceps species have been reported as hyperparasitic fungi. A new species, Pleurocordyceps fusiformispora, and a known species, Perennicordyceps elaphomyceticola, are described here based on morphology and phylogenetic evidence from six genes (ITS, SSU, LSU, TET1-α, RPB1, and RPB2). Pl. fusiformispora differed from the other Pleurocordyceps species by producing flaky colonies, ovoid or elliptic α-conidia, and fusiform or long fusiform β-conidia. Both full genomes of Pe. elaphomyceticola and Pl. fusiformispora were sequenced, annotated, and compared. The antiSMASH and local BLAST analyses revealed significant differences in the number and types of putative secondary metabolite biosynthetic gene clusters, i.e., NPPS, PKS, and hybrid PKS–NRPS domains, between the two species. In addition, the putative BGCs of six compounds, namely ε-poly lysine, 4-epi-15-epi-brefeldin A, Monorden D/monocillin IV/monocillin VII/pochonin M/monocillin V/monocillin II, Tolypyridone, Piperazine, and Triticone DABFC, were excavated in the present study. This study motivates the use of heterologous expression and gene knockout methods to discover novel biologically active SMs from Polycephalomycetaceae.

Background: Glucuronidation is an essential metabolic pathway for a variety of drugs. IMM-H004 is a novel neuroprotective agent against ischemic stroke, and its glucuronide metabolite IMM-H004G exhibits similar pharmacological activity. Despite possessing a higher molecular weight and polarity, brain exposure of IMM-H004G is much higher than that of IMM-H004. This study aimed to investigate the brain metabolism and transport mechanisms of IMM-H004 and IMM-H004G. Methods: First, the possibility of IMM-H004 glucuronidation in the brain was evaluated in several human brain cell lines and rat homogenate. Subsequently, the blood–brain barrier carrier-mediated transport mechanism of IMM-H004 and IMM-H004G was studied using overexpression cell models. In addition, intracerebroventricular injection, in situ brain perfusion model, and microdialysis/microinjection techniques were performed to study the distribution profiles of IMM-H004 and IMM-H004G. Results: IMM-H004 could be metabolized to IMM-H004G in both rat brain and HEB cells mediated by UGT1A7. However, IMM-H004G could not be hydrolyzed back into IMM-H004. Furthermore, the entry and efflux of IMM-H004 in the brain were mediated by the pyrilamine-sensitive H+/OC antiporter and P-gp, respectively, while the transport of IMM-H004G from the blood to the brain was facilitated by OATP1A2 and OATP2B1. Ultimately, stronger concentration gradients and OATP-mediated uptake played a critical role in promoting greater brain exposure of IMM-H004G. Conclusions: The active glucuronide metabolite of the brain protectant IMM-H004 with poor blood–brain barrier permeability demonstrates a high partition in the rat brain via multiple mechanisms, and our findings deepen the understanding of the mechanisms underlying the blood–brain barrier metabolism and transport of active glucuronide conjugates.

Developing tailored emission reduction strategies and estimating their potential is crucial for achieving low-carbon rice production in a specific region, as well as for advancing China’s dual carbon goals in the agricultural sector. By utilizing water-saving and drought-resistant rice (WDR) with enhanced water and nitrogen utilization efficiency, the mitigation strategies were constructed for rice production systems, and their potential for emission reduction was estimated in the southern rice propagation base of Hainan Province. This study revealed that the implementation of a reduction strategy, which involves dry direct seeding and dry cultivation, combined with a 53% reduction in nitrogen fertilizer, can effectively synergize the mitigation of methane (CH4) and nitrous oxide (N2O) emissions from rice paddies. Compared with traditional flooded rice cultivation, this integrated approach exhibits an impressive potential for reducing net greenhouse gas (GHG) emissions by 97% while simultaneously doubling economic benefits. Moreover, when combined with plastic film mulching, the strategy not only sustains rice yields but also achieves a remarkable emission reduction of 92%, leading to a fourfold increase in economic benefits. Our study provides a comprehensive low-carbon sustainable development strategy for rice production in the southern rice propagation base of Hainan Province and offers valuable insights for researching GHG emissions in other regions or crops. These emission reduction pathways and the assessment method could contribute to the realization of low-carbon agriculture.

The application of nitrogen fertilizer within a normal range has been found to increase the leaf nitrogen content and photosynthetic rate of canola plants (Brassica napus L.). Despite numerous studies on the separate effects of CO2 diffusion limitation and nitrogen allocation trade-off on photosynthetic rate, few have examined both these factors in relation to the photosynthetic rate of canola. In this study, two genotypes of canola with varying leaf nitrogen content were analyzed to determine the impact of nitrogen supply on leaf photosynthesis, mesophyll conductance, and nitrogen partitioning. The results showed that the CO2 assimilation rate (A), mesophyll conductance (gm), and photosynthetic nitrogen content (Npsn) increased with an increase in nitrogen supply in both genotypes. The relationship between nitrogen content and A followed a linear-plateau regression, while A had linear relationships with both photosynthetic nitrogen content and gm, indicating that the key to enhancing A is increasing the distribution of leaf nitrogen into the photosynthetic apparatus and gm, rather than just increasing nitrogen content. Under high nitrogen treatment, the genotype (QZ) with high nitrogen content had 50.7% more nitrogen than the other genotype (ZY21), but had similar A, which was primarily due to ZY21’s higher photosynthetic nitrogen distribution ratio and stomatal conductance (gsw). On the other hand, QZ showed a higher A than ZY21 under low nitrogen treatment as QZ had higher Npsn and gm compared to ZY21. Our results indicate that, in selecting high PNUE rapeseed varieties, it is important to consider the higher photosynthetic nitrogen distribution ratio and CO2 diffusion conductance.

Phosphate (Pi) is indispensable for the growth and development of plant, and low-Pi stress is a major limitation for crop growth and yield worldwide. The tolerance to low-Pi stress varied among rice germplasm resources. However, the mechanisms underlying the tolerance of rice to low-Pi stress, as a complex quantitative trait, are not clear. We performed a genome-wide association study (GWAS) through a diverse worldwide collection of 191 rice accessions in the field under normal-Pi and low-Pi supply in two years. Twenty and three significant association loci were identified for biomass and grain yield per plant under low-Pi supply respectively. The expression level of OsAAD as a candidate gene from a associated locus was significantly up-regulated after low-Pi stress treatment for five days and tended to return to normal levels after Pi re-supply in shoots. Suppression of OsAAD expression could improve the physiological phosphorus use efficiency (PPUE) and grain yields through affecting the expression of several genes associated with GA biosynthesis and metabolism. OsAAD would be a promising gene for increasing PPUE and grain yield in rice under normal- and low-Pi supply via genome editing.

Abstract Background Inflammation is a complex physiological and pathological process. Although many types of inflammation are well characterized, their physiological functions are largely unknown. tRNA aspartic acid methyltransferase 1 (TRDMT1) has been implicated as a stress‐related protein, but its intrinsic biological role is unclear. Methods We constructed a Trdmt1 knockout rat and adopted the LPS‐induced sepsis model. Survival curve, histopathological examination, expression of inflammatory factors, and protein level of TLR4 pathway were analyzed. Results Trdmt1 deletion had no obvious impact on development and growth. Trdmt1 deletion slightly increased the mortality during aging. Our data showed that Trdmt1 strongly responded in LPS‐treated rats, and Trdmt1 knockout rats were vulnerable to LPS treatment with declined survival rate. We also observed more aggravated tissue damage and more cumulative functional cell degeneration in LPS‐treated knockout rats compared with control rats. Further studies showed upregulated TNF‐α level in liver, spleen, lung, and serum tissues, which may be explained by enhanced p65 and p38 phosphorylation. Conclusions Our data demonstrated that Trdmt1 plays a protective role in inflammation by regulating the TLR4‐NF‐κB/MAPK‐TNF‐α pathway. This work provides useful information to understand the TRDMT1 function in inflammation.

Pheasants are widely distributed in the southwest of China, but many of them are endangered due to habitat fragmentation and environmental changes. Genetic diversity is crucial for species to maintain their evolutionary potential, and thus it is important to develop universal genetic markers for facilitating the assessment of genetic diversity and planning effective conservation actions in these endangered species. In this study, 471 microsatellite loci which are common among eight pheasant species were screened based on genome data, and 119 loci were selected to develop microsatellite markers. After PCR amplifications and reaction condition optimizations, and validation of microsatellite loci in 14 species of 11 genera within Phasianidae. Finally, 49 potentially universal microsatellite markers in pheasant species were obtained. These microsatellite markers were successfully applied to assess the genetic diversity of 3 pheasant species. The Sichuan hill partridge (Arborophila rufipectus), blood pheasant (Ithaginis cruentus), buff-throated partridge (Tetraophasis szechenyii) and Sichuan hill partridge had a relatively low genetic diversity level. These 49 microsatellite loci are potentially universal microsatellite loci for pheasants and are of great significance to establish a shared platform in population genetics study of pheasants.

The application of high-quality seeds ensures successful crop establishment, healthy growth, and improved production in both quantity and quality. Recently, biochar-based seed coating has been recognized as a new, effective, and environmentally friendly method to enhance seed quality, seedling uniformity, and nutrient availability. To study the impact of biochar coating on the surface mechanical properties of coated seeds, rice emergence and growth, and related physical and physiological metabolic events, laboratory experiments were performed on two water-saving and drought-resistance rice (WDR) varieties (Huhan1512 and Hanyou73) using biochar formulations with varying contents (20%–60%). The results showed that the appropriate concentration of biochar significantly improved emergence traits and seedling performance of the two rice varieties, compared to the uncoated treatment, and that the optimal percentage of biochar coating was 30% (BC30). On average, across both varieties, BC30 enhanced emergence rate (9.5%), emergence index (42.9%), shoot length (19.5%), root length (23.7%), shoot dry weight (25.1%), and root dry weight (49.8%). The improved germination characteristics and vigorous seedling growth induced by biochar coating were strongly associated with higher water uptake by seeds, increased α-amylase activity and respiration rate, and enhanced accumulation of soluble sugar and soluble protein. Moreover, the evaluation results of mechanical properties related to seed coating quality found that increasing the proportion of biochar in the coating blend decreased the integrity and compressive strength of the coated seeds and reduced the time required for coating disintegration. In conclusion, biochar coating is a cost-effective strategy for enhancing crop seed quality and seedling establishment.

Background: Myasthenia gravis (MG) is an autoimmune disease characterized by muscle weakness and fatigability. The fluctuating nature of the disease course impedes the clinical management. Objective: The purpose of the study was to establish and validate a machine learning (ML)–based model for predicting the short-term clinical outcome in MG patients with different antibody types. Methods: We studied 890 MG patients who had regular follow-ups at 11 tertiary centers in China from 1 January 2015 to 31 July 2021 (653 patients for derivation and 237 for validation). The short-term outcome was the modified post-intervention status (PIS) at a 6-month visit. A two-step variable screening was used to determine the factors for model construction and 14 ML algorithms were used for model optimisation. Results: The derivation cohort included 653 patients from Huashan hospital [age 44.24 (17.22) years, female 57.6%, generalized MG 73.5%], and the validation cohort included 237 patients from 10 independent centers [age 44.24 (17.22) years, female 55.0%, generalized MG 81.2%]. The ML model identified patients who were improved with an area under the receiver operating characteristic curve (AUC) of 0.91 [0.89–0.93], ‘Unchanged’ 0.89 [0.87–0.91], and ‘Worse’ 0.89 [0.85–0.92] in the derivation cohort, whereas identified patients who were improved with an AUC of 0.84 [0.79–0.89], ‘Unchanged’ 0.74 [0.67–0.82], and ‘Worse’ 0.79 [0.70–0.88] in the validation cohort. Both datasets presented a good calibration ability by fitting the expectation slopes. The model is finally explained by 25 simple predictors and transferred to a feasible web tool for an initial assessment. Conclusion: The explainable, ML-based predictive model can aid in forecasting the short-term outcome for MG with good accuracy in clinical practice.

Objective: To provide consultative information on selecting potential indications of stem cells in the treatment of non-tumorous and non-hematopoietic system conditions, we screened the clinical trials on website: http://www.clinicaltrials.gov. Methods: A literature search was conducted using the National Institute of Health (NIH) website (http://www.clinicaltrials.gov) from May 10th, 2012 to Oct 13th, 2020 with stem cells as an intervention in human trials for non-tumorous and non-hematological conditions. There was no restriction for language, research location, and research race. Results and conclusion: Search terms initially found a total of 3576 articles. Firstly, 138 terminated or suspended studies were excluded and further 24 repeated studies were excluded. Secondly, 987 tumorous and hematopoietic conditions-related studies were excluded. Lastly, 1218 studies were excluded without stem cell therapy as their primary purpose. A total of 1209 research studies were entered into the analysis and reviewed. The top 4 diseases were about motor system diseases 229 (19.28%). In addition, 206 (17.34%) studies were related to central nervous system (CNS) diseases. 140 (11.78%) were autoimmune diseases or graft-versus-host disease (GVHD) after organ transplantation. 129 (10.86%) were about respiratory system diseases, among them, 44.19% projects were about new coronary pneumonia (NCP). The main cell types used in various diseases are Mesenchymal Stem Cells (MSCs), bone marrow stem cells (BM-SCs), peripheral blood stem cells (PBSCs), hematopoietic stem cells (HSCs), adipose stem cell (ASCs) and precursor cells.

Abstract Background The two-line method based on the photoperiod and thermo-sensitive genic male sterile (PTGMS) lines is more cost-effective, simple, and efficient than the three-line system based on cytoplasmic male-sterility. Blast and drought are the most prevalent biotic and abiotic stress factors hampering rice production. Molecular techniques demonstrate higher efficacy in the pyramiding of disease resistance genes, providing green performance under the background of water-saving and drought-resistance rice. Results This study employed molecular marker-assisted selection, conventional hybridization, and high-intensity stress screening to integrate three broad-spectrum blast resistance genes Pi9, Pi5, and Pi54 into Huhan 1S. Subsequently, a novel water-saving and drought-resistance rice (WDR) PTGMS line Huhan 74S was developed. The drought resistance of the new PTGMS line Huhan 74S was comparable to that of Huhan 1S. Pathogenicity assays involving the inoculation of 14 blast prevalent isolates in the glasshouse showed that the blast resistance frequency of Huhan 74S was 85.7%. Further evaluation under natural blast epidemic field conditions showed that Huhan 74S and its hybrids were resistant to leaf and neck blast. The critical temperature point of fertility-sterility alteration of Huhan 74S was 23 °C daily mean temperature. The complete male sterility under natural growth conditions in 2017 at Shanghai lasted for 67 days. Also, both the agronomic and grain quality traits met the requirement for two-line hybrid rice production. Conclusion These results indicate that the newly bred PTGMS line Huhan 74S can be used to breed high-yielding, good-quality, disease-resistant two-line hybrid water-saving and drought-resistance rice (WDR), hence promoting sustainable rice production in China.

IntroductionThe lack of knowledge regarding the differences between Chinese and other ethnicities in the early manifestation of late-onset Pompe disease (LOPD) prohibits the development of an effective screening strategy. We conducted a multicenter screening study to determine LOPD prevalence in high-risk populations and define the early manifestation of LOPD in China.MethodsBetween August 2020 and April 2021, the participants were prospectively identified through medical examination at 20 centers from inpatient departments and outpatient neuromuscular clinics in China. The inclusion criteria were as follows: (1) age ≥ 1 year and (2) either one of the following conditions: (a) persistent hyperCKemia, (b) muscle weakness of the axial and/or limb-girdle muscles, or (c) unexplained restrictive respiratory insufficiency (RI). Enzymatic activity of acid α-glucosidase (GAA) was measured in a dried blood spot (DBS) using a tandem mass spectrometry (MS/MS) assay. Next-generation sequencing (NGS) was used to evaluate all samples with decreased GAA activity, searching for GAA mutations and pseudodeficiency alleles.ResultsAmong the 492 cases, 26 positive samples (5.3%) were detected in the DBS test. Molecular studies confirmed a diagnosis of LOPD in eight cases (1.6%). Using MS/MS assay, GAA activities in individuals with pseudodeficiency could be distinguished from those in patients with LOPD. The median interval from the onset of symptoms to diagnosis was 5 years. All patients also showed RI, with a mean forced vital capacity (FVC) of 48%, in addition to axial/proximal muscle weakness. The creatine kinase (CK) level ranged from normal to no more than 5-fold the upper normal limit (UNL). LOPD with isolated hyperCKemia was not identified.ConclusionLess frequent hyperCKemia and predominant RI depict a different early portrait of adult Chinese patients with LOPD. A modified high-risk screening strategy should be proposed for the early diagnosis of Chinese patients with LOPD.

Diuron (DU) abuse in weed removal and shipping pollution prevention always leads to pesticide residues and poses a risk to human health. In the current research, an innovative electrochemical sensor for DU detection was created using a glassy carbon electrode (GCE) that had been modified with chitosan-encapsulated multi-walled carbon nanotubes (MWCNTs-CS) combined with nitrogen-doped graphene quantum dots (NGQDs). The NGQDs were prepared by high-temperature pyrolysis, and the MWCNTs-CS@NGQDs composite was further prepared by ultrasonic assembly. TEM, UV-Vis, and zeta potential tests were performed to investigate the morphology and properties of MWCNTs-CS@NGQDs. CV and EIS measurements revealed that the assembly of MWCNTs and CS improved the electron transfer ability and effective active area of MWCNTs. Moreover, the introduction of NGQDs further enhanced the detection sensitivity of the designed sensor. The MWCNTs-CS@NGQDs/GCE electrochemical sensor exhibited a wide linear range (0.08~12 μg mL−1), a low limit of detection (0.04 μg mL−1), and high sensitivity (31.62 μA (μg mL−1)−1 cm−2) for DU detection. Furthermore, the sensor demonstrated good anti-interference performance, reproducibility, and stability. This approach has been effectively employed to determine DU in actual samples, with recovery ranges of 99.4~104% in river water and 90.0~94.6% in soil. The developed electrochemical sensor is a useful tool to detect DU, which is expected to provide a convenient and easy analytical technique for the determination of various bioactive species.

Mesocotyl elongation of rice seedlings is a key trait for deep sowing tolerance and well seedling establishment in dry direct sowing rice (DDSR) production. Subsets of the Rice Diversity Panel 1 (RDP1, 294 accessions) and Hanyou 73 (HY73) recombinant inbred line (RIL) population (312 lines) were screened for mesocotyl length (ML) via dark germination. Six RDP1 accessions (Phudugey, Kasalath, CA902B21, Surjamkuhi, Djimoron, and Goria) had an ML longer than 10 cm, with the other 19 accessions being over 4 cm. A GWAS in RDP1 detected 118 associated SNPs on all 12 chromosomes using a threshold of FDR-adjusted p < 0.05, including 11 SNPs on chromosomes 1, 4, 5, 7, 10, and 12 declared by −log10(P) > 5.868 as the Bonferroni-corrected threshold. Using phenotypic data of three successive trials and a high-density bin map from resequencing genotypic data, four to six QTLs were detected on chromosomes 1, 2, 5, 6, and 10, including three loci repeatedly mapped for ML from two or three replicated trials. Candidate genes were predicted from the chromosomal regions covered by the associated LD blocks and the confidence intervals (CIs) of QTLs and partially validated by the dynamic RNA-seq data in the mesocotyl along different periods of light exposure. Potential strategies of donor parent selection for seedling establishment in DDSR breeding were discussed.

Abstract Background An accurate prediction for prognosis can help in guiding the therapeutic options and optimizing the trial design for generalized myasthenia gravis (gMG). We aimed to develop and validate a predictive nomogram to assess the short-term outcome in patients with the anti-acetylcholine receptor (AChR) subtype gMG. Methods We retrospectively reviewed 165 patients with AChR subtype gMG who were immunotherapy naïve at the first visit from five tertiary centers in China. The short-term clinical outcome is defined as the achievement of minimal symptom expression (MSE) at 12 months. Of them, 120 gMG patients from Huashan Hospital were enrolled to form a derivation cohort (n = 96) and a temporal validation cohort (n = 24) for the nomogram. Then, this nomogram was externally validated using 45 immunotherapy naïve AChR subtype gMG from the other four hospitals. Multivariate logistic regression was used to screen independent factors and construct the nomogram. Results MSE was achieved in 70 (72.9%), 20 (83.3%), and 33 (73.3%) patients in the training, temporal validation, and external validation cohort, respectively. The duration ≤ 12 months (p = 0.021), ocular score ≤ 2 (p = 0.006), QMG score > 13 (p = 0.008), and gross motor score ≤ 9 (p = 0.006) were statistically associated with MSE in AChR subtype gMG. The nomogram has good performance in predicting MSE as the concordance indexes are 0.81 (95% CI, 0.72–0.90) in the development cohort, 0.944 (95% CI, 0.83–1.00) in the temporal validation cohort, and 0.773 (95% CI, 0.63–0.92) in the external validation cohort. Conclusion The nomogram achieved an optimal prediction of MSE in AChR subtype gMG patients using the baseline clinical characters.

The commercialization of hybrid rice has greatly contributed to the increase in rice yield, with the improvement of its seed production capacity having played an important role. The stigma exsertion rate (SER) is a key factor for improving the outcrossing of the sterile line and the hybrid rice seed production. We used the Zhenshan 97B × IRAT109 recombinant inbred population comprising 163 lines and a natural population of 138 accessions to decipher the genetic foundation of SER over 2 years in three environments. Additionally, we detected eight QTLs for SER on chromosomes 1, 2, and 8 via linkage mapping. We also identified seven and 19 significant associations for SER using genome-wide association study in 2016 and 2017, respectively. Interestingly, we located two lead SNPs (sf0803343504 and sf083344610) on chromosome 8 in the qTSE8 QTL region that were significantly associated with total SER. After transcriptomic analysis, quantitative real-time PCR, and haplotype analysis, we found 13 genes within this reliable region as important candidate genes. Our study results will be beneficial to molecular marker-assisted selection of rice lines with high outcrossing rate, thereby improving the efficiency of hybrid seed production.

Mitochondrial RNA editing plays crucial roles in the plant development and environmental adaptation. Pentatricopeptide repeat (PPR) genes, which are involved in the regulating mitochondrial RNA editing, are potential gene resources in the improvement of rice drought tolerance. In this study, we investigated genome-wide mitochondrial RNA editing in response to drought between upland and lowland rice. Responses of mitochondrial RNA editing to drought exhibit site-specific and genotype-specific patterns. We detected 22 and 57 ecotype-differentiated editing sites under well-watered and drought-treated conditions, respectively. Interestingly, the RNA editing efficiency was positively correlated with many agronomic traits, while it was negatively correlated with drought tolerance. We further selected two mitochondrial-localized PPR proteins, PPR035 and PPR406, to validate their functions in drought tolerance. PPR035 regulated RNA editing at rps4-926 and orfX-406, while PPR406 regulated RNA editing at orfX-355. The defectiveness in RNA editing at these sites had no apparent penalties in rice respiration and vegetative growth. Meanwhile, the knockout mutants of ppr035 and ppr406 show enhanced drought- and salt tolerance. PPR035 and PPR406 were under the balancing selection in upland rice and highly differentiated between upland and lowland rice ecotypes. The upland-dominant haplotypes of PPR035 and PPR406 shall contribute to the better drought tolerance in upland rice. They have great prospective in the improvement of rice drought tolerance.

ObjectiveTo compare the predictive performance of five handgrip strengths for cardiovascular disease (CVD) risk factors.MethodsA total of 804 Chinese middle-aged community residents' health medical examinations were collected. The absolute handgrip strength was denoted as HGS. HGS/body weight (HGS/BW), HGS/body mass index (HGS/BMI), HGS/lean body mass (HGS/LBM), and HGS/muscle mass (HGS/MM) represented relative handgrip strength (RHGS). To assess predictive performance, receiver operating characteristic (ROC) curves and the area under the curve (AUC) were constructed.ResultsHGS was not associated with most CVD risk biomarkers; however, RHGS showed a negative correlation trend after controlling for covariates (sex, age, smoking, and exercise). HGS/BMI and HGS/BW had better AUCs for predicting CVD risk factors than HGS/LBM or HGS/MM. HGS/BMI and HGS/BW can successfully predict all CVD risk factors in men with AUCs 0.55–0.65; similarly, women may effectively predict arteriosclerosis, hyperglycemia, hyperuricemia, and metabolic syndrome with AUCs 0.59–0.64, all p < 0.05. The optimal HGS/BW cut-off points for identifying different CVD risk factors were 0.59–0.61 in men and 0.41–0.45 in women, while the HGS/BMI were 1.75–1.79 in men and 1.11–1.15 in women.ConclusionsAlmost all CVD risk biomarkers and CVD risk factors were unrelated to HGS. There is, however, a significant inverse relationship between RHGS and CVD risk factors. HGS/BMI or HGS/BW should be recommended to be the best choice for predicting the risk of CVD risk factors in five expressions of handgrip strength. We also acquired the recommended optimal cut-off points of HGS/BMI and HGS/BW for predicting CVD risk factors.

Mosses are one of the earliest diverging land plants that adapted to living on land. The BURP domain-containing proteins (BURP proteins) are plant-specific proteins that appeared when plants shifted from aquatic environments to land. Phylogenetic analysis revealed that the BURP domain of higher plants is originated from lower land plants and divergent because of motif conversion. To discover the function of BURP protein in moss, rice transgenics with ectopic expression of PpBURP2 were subjected to different abiotic stresses treatments. The results revealed that the ectopic expression of PpBURP2 enhanced the tolerance to osmotic and saline stresses at the seedling stage and drought stress at the adult stage. Further ectopic expression of PpBURP2 improved the cadmium (2+) (Cd2+) tolerance and reduced Cd2+ accumulation in rice leaves. Transcriptomic analysis of the transgenic PpBURP2 plants showed that the differentially expressed genes were involved in the metabolism of secondary metabolites, energy, oxidation-reduction process, and defense-related genes. Further experiments showed that the photosynthetic efficiency and resistance against bacterial leaf blight were obviously improved in transgenic plants. Yeast two-hybrid and bimolecular fluorescence complementation (BiFC) assays revealed the physical interaction of BURP domain protein from rice and moss with mitogen-activated protein kinase kinase (MKK) from rice. Therefore, our findings demonstrate that overexpressing PpBURP2 in rice confers resistance to abiotic stresses and bacterial leaf blight. They also suggested that the regulatory role of BURP-like proteins across lower and higher plants was evolutionary conservation of responses of different classes of plants to different environmental challenges.

The amendment of biochar has been proposed to improve soil fertility and crop yield. However, consolidated information lacks explaining the role of biochar on soil and plant enzymatic activities involved in nutrients cycling in soil and accumulation in plants improving utilization of applied inorganic fertilizer and crop growth. In the current study, we evaluated the integral effects of biochar levels (B0:0, B15:15, B3:30, and B60:60 t ha–1) and nitrogen fertilizer levels (N0:0, N75:75, N225:225, and N450:450 kg ha–1) on soil physicochemical properties, enzymatic activities, nitrogen use efficiency (NUE) and grain yield of rapeseed for 2 years in the pots during 2020 and 2021. The findings revealed that compared to control (B0 + N0), a combination of B30 + N450 increased soil urease activity by 73 and 75%, and B60 + N450 increased activities of soil catalase by 17 and 16%, and B60 + N225 increased alkaline phosphatase by 17 and 19%, respectively, in the first and second year. Moreover, a single application of high nitrogen at 450 kg ha–1 reduced the activities of plant nitrogen metabolism-related enzymes, however; the integration of biochar at 30 t ha–1 compensated the high nitrogen toxicity and improved the activities of nitrate reductase (NR), nitrite reductase NIR, glutamate synthase (GS) and glutamine synthetase (GOGAT) at seedling stage (SS) and flowering stage (FS) in both years. The integration of biochar at 30 t ha–1 with nitrogen at 450 kg ha–1 induced synergetic effects on rapeseed growth through sorption of excessive nitrogen in soil and significantly improved the plant height up to 11 and 18%, pods plant–1 39 and 32% and grain yield plant–1 54 and 64%, respectively, during the first and second year. Moreover, biochar at 15 t ha–1 along with nitrogen at 225 kg ha–1 resulted in the highest NUE of 29% in both years suggesting that biochar can also offset the deficiency of lower nitrogen. This study highlighted the ameliorative effect of biochar suppressing high nitrogen toxicity and decreasing lower nitrogen deficiency effects on rapeseed growth by improving nitrogen use efficiency via enhancing soil conditions, enzymatic activities and soil nitrogen utilization potential and thus improving rapeseed growth and yield.

Abstract Background Drought-tolerance ensures a crop to maintain life activities and protect cell from damages under dehydration. It refers to diverse mechanisms temporally activated when the crop adapts to drought. However, knowledge about the temporal dynamics of rice transcriptome under drought is limited. Results Here, we investigated temporal transcriptomic dynamics in 12 rice genotypes, which varied in drought tolerance (DT), under a naturally occurred drought in fields. The tolerant genotypes possess less differentially expressed genes (DEGs) while they have higher proportions of upregulated DEGs. Tolerant and susceptible genotypes have great differences in temporally activated biological processes (BPs) during the drought period and at the recovery stage based on their DEGs. The DT-featured BPs, which are activated specially (e.g. raffinose, fucose, and trehalose metabolic processes, etc.) or earlier in the tolerant genotypes (e.g. protein and histone deacetylation, protein peptidyl-prolyl isomerization, transcriptional attenuation, ferric iron transport, etc.) shall contribute to DT. Meanwhile, the tolerant genotypes and the susceptible genotypes also present great differences in photosynthesis and cross-talks among phytohormones under drought. A certain transcriptomic tradeoff between DT and productivity is observed. Tolerant genotypes have a better balance between DT and productivity under drought by activating drought-responsive genes appropriately. Twenty hub genes in the gene coexpression network, which are correlated with DT but without potential penalties in productivity, are recommended as good candidates for DT. Conclusions Findings of this study provide us informative cues about rice temporal transcriptomic dynamics under drought and strengthen our system-level understandings in rice DT.

Abstract Introduction Transcriptomic divergence drives plant ecological adaptation. Upland rice is differentiated in drought tolerance from lowland rice during its adaptation to the drought‐prone environment. They provide a good system to learn the evolution of drought tolerance in rice. Methods and Results We estimate morphological differences between the two rice ecotypes under well‐watered and drought conditions, as well as their genetic and transcriptomic divergences by the high‐throughput sequencing. Upland rice possesses higher expression diversity than lowland rice does. Thousands of genes exhibit expression divergences between the two rice ecotypes, which contributes to their morphological differences in drought tolerance. These transcriptomic divergences contribute to drought adaptation of upland rice during its domestication. Mutations in transcriptional regulatory regions, which cause presence and absence of cis‐elements, are the cause of expression divergence. About 15.3% transcriptionally selected genes also receive sequence‐based selection in upland or lowland ecotype. Some highly differentiated genes promote the transcriptomic divergence between rice ecotypes via gene co‐expression network. In addition, we also detected transcriptomic trade‐offs between drought tolerance and productivity. Discussion Many key genes, which promote transcriptomic adaptation to drought in upland rice, have great prospective in breeding water‐saving and drought‐resistant rice. Meanwhile, appropriate strategies are required in breeding to overcome the potential transcriptomic trade‐off.

Abstract Background Plant miRNAs play crucial roles in responses to drought and developmental processes. It is essential to understand the association of miRNAs with drought-tolerance (DT), as well as their impacts on growth, development, and reproduction (GDP). This will facilitate our utilization of rice miRNAs in breeding. Results In this study, we investigated the time course of miRNA responses to a long-term drought among six rice genotypes by high-throughput sequencing. In total, 354 conserved miRNAs were drought responsive, representing obvious genotype- and stage-dependent patterns. The drought-responsive miRNAs (DRMs) formed complex regulatory network via their coexpression and direct/indirect impacts on the rice transcriptome. Based on correlation analyses, 211 DRMs were predicted to be associated with DT and/or GDP. Noticeably, 14.2% DRMs were inversely correlated with DT and GDP. In addition, 9 pairs of mature miRNAs, each derived from the same pre-miRNAs, were predicted to have opposite roles in regulating DT and GDP. This suggests a potential yield penalty if an inappropriate miRNA/pre-miRNA is utilized. miRNAs have profound impacts on the rice transcriptome reflected by great number of correlated drought-responsive genes. By regulating these genes, a miRNA could activate diverse biological processes and metabolic pathways to adapt to drought and have an influence on its GDP. Conclusion Based on the temporal pattern of miRNAs in response to drought, we have described the complex network between DRMs. Potential associations of DRMs with DT and/or GDP were disclosed. This knowledge provides valuable information for a better understanding in the roles of miRNAs play in rice DT and/or GDP, which can facilitate our utilization of miRNA in breeding.

Abstract In the present work, a tubular nano-copper sulfide was successfully synthesized by hydrothermal method. The physical and chemical properties of the prepared materials were characterized by XRD, SEM, TEM, and BET. The synthesized copper sulfide was used as an adsorbent for removing 17α-ethynyl estradiol (EE2) and exhibited excellent adsorption properties. At 25 °C, 15 mg of adsorbent was applied for 50 mL of 5 mg/L EE2 solution, adsorption equilibrium was reached after 180 min, and the adsorption rate reached nearly 90%. In addition, the kinetics, isothermal adsorption, and thermodynamics of the adsorption process were discussed on the basis of theoretical calculations and experimental results. The theoretical maximum adsorption capacity of copper sulfide was calculated to be 147.06 mg/g. The results of this study indicated that copper sulfide was a stable and efficient adsorbent with promising practical applications.

Of all the crops, rice is the one that consumes the most water. Rice yields and quality are significantly influenced by irrigation. However, it is still unknown how different irrigation practices would affect the grain yield and quality of water-saving and drought-resistant rice. Hyou 518 (high-yielding rice variety) and Hanyou 73 (water-saving and drought-resistant rice variety) were employed as materials. Three irrigation regimes were set up in the field: conventional flooding irrigation (CF), alternate wetting and moderate soil drying irrigation (AWD), and dry cultivation (D). It was investigated how various irrigation regimes affected the two varieties’ yield and quality. The results revealed the following: 1. D considerably increased water-use efficiency while drastically reducing the yield, compared to CF and AWD. In comparison to other irrigation regimes, the grain yield and water use efficiency of Hanyou 73 enhanced synergistically under AWD treatment. 2. In contrast to CF treatment, AWD and D (especially) treatments decreased perfect rice kernel, total starch content, amylose content, amylopectin content, amylose/amylopectin, gel consistency, and breakdown, but increased green rice kernel, chalky kernel, protein content, and setback. 3. After heading, AWD and D lowered, and D treatment decreased more, the activities of ADP-glucose pyrophosphorylase (AGP), soluble starch synthase (SSS), and starch branching enzyme (SBE). AGP, SSS, and SBE were strongly inversely linked with perfect rice kernel, amylopectin content, gel consistency, and breakdown, but significantly negatively correlated with green rice kernel, chalky kernel, protein content, and setback. The results indicate that with AWD treatment, Hanyou 73 might provide a synergistic boost grain production, water-use efficiency, and quality. D treatment could significantly improve water-use efficiency. Compared with Hyou518, Hanyou 73 could maintain higher AGP, SSS, and SBE activities, head milled rice, perfect rice kernel, amylopectin content, and gel consistency under AWD and D treatment.

A three-dimensional (3D) electric field sensing scheme is proposed and experimentally demonstrated based on an integrated lithium niobate on insulator (LNOI) platform. The 3D measurement is realized by packing three LNOI-based sensor chips in a triangular-prism-type clamp. For each sensor chip, the optical waveguide has an asymmetrical Michelson interferometer architecture, and the tapered dipole antenna is inclined to the optical waveguide. By finely placing the three sensor chips in the clamp, the three pairs of inclined tapered dipole antennas are mutually orthogonal and can be applied to measure the electric field in three orthogonal polarization directions. The volume of the packaged 3D sensor is 9.5 cm3. In the experiment, a flat response in the frequency range of 10 MHz to 3 GHz is demonstrated. In addition, a 3 × 3 response calibration matrix is obtained and utilized to reduce the measurement error. After calibration, the relative measurement error of the electric field amplitude is smaller than 5.1% for every polarization direction.

Recent studies showed that Escherichia coli (E. coli) strains isolated from captive giant pandas have serious resistance to antibiotics and carry various antibiotic resistance genes (ARGs). ARGs or virulence-associated genes (VAGs) carried by antibiotic-resistant E. coli are considered as a potential health threat to giant pandas, humans, other animals and the environment. In this study, we screened ARGs and VAGs in 84 antibiotic-resistant E. coli strains isolated from clinically healthy captive giant pandas, identified the association between ARGs and VAGs and analyzed the phylogenetic clustering of E. coli isolates. Our results showed that the most prevalent ARG in E. coli strains isolated from giant pandas is blaTEM (100.00%, 84/84), while the most prevalent VAG is fimC (91.67%, 77/84). There was a significant positive association among 30 pairs of ARGs, of which the strongest was observed for sul1/tetC (OR, 133.33). A significant positive association was demonstrated among 14 pairs of VAGs, and the strongest was observed for fyuA/iroN (OR, 294.40). A positive association was also observed among 45 pairs of ARGs and VAGs, of which the strongest was sul1/eaeA (OR, 23.06). The association of ARGs and mobile gene elements (MGEs) was further analyzed, and the strongest was found for flor and intI1 (OR, 79.86). The result of phylogenetic clustering showed that the most prevalent group was group B2 (67.86%, 57/84), followed by group A (16.67%, 14/84), group D (9.52%, 8/84) and group B1 (5.95%, 5/84). This study implied that antibiotic-resistant E. coli isolated from captive giant pandas is a reservoir of ARGs and VAGs, and significant associations exist among ARGs, VAGs and MGEs. Monitoring ARGs, VAGs and MGEs carried by E. coli from giant pandas is beneficial for controlling the development of antimicrobial resistance.

Dongxiang wild rice (DXWR) is one of the most valuable germplasm resources of rice. It is important to conserve the genetic diversity and uncover the population differentiation of DXWR. In this study, we analyzed the genetic diversity and population differentiation of DXWR based on whole-genome resequencing of 220 DXWR lines collected from nine natural populations in an ex situ conservation nursery. Almost half of the SNPs and Indels detected in these DXWR lines were absent in cultivated rice or other common wild rice, indicating the potential and importance of DXWR in rice breeding. Based on Structure and PCA analysis, these DXWR lines could be divided into two subpopulations, in which subpopulation G1 had more specific SNPs and Indels and was genetically more genetically diverse than subpopulation G2. The average Fst of regions with low relative genetic diversity between G1 and G2 were significantly lower than whole-genomic Fst, indicating directional selection in these regions. Some functional genes and QTLs were found to locate in highly differentiated regions between G1 and G2. Moreover, the deep root ratios of G2 were significantly higher than G1. Our results would be helpful to the conservation and utilization of DXWR germplasm.

Electromagnetic spectrum sensing is quite important for communication safety in commercial wireless communication, as well as for the equipment paralysis of rivals and self-protection in modern warfare. Specifically, a passive electric field sensor featured with broadband, remote sensing capabilities and small size is urgently needed to realize spectrum sensing. Here, we demonstrate a photonic electric field sensor operating in the frequency range of 10 MHz to 26.5 GHz. Based on the electric field sensor, distortion-free sensing of on–off keying signals centered at 3 GHz is achieved under a bit rate up to 40 Mb/s. In addition, remote electromagnetic spectrum sensing is also demonstrated up to 70 km, where the signal-to-noise ratio is measured to be larger than 20 dB.

To address the difficulty of early fault diagnosis of rolling bearings, this paper proposes a rolling bearing diagnosis method by combining the attention entropy and adaptive deep kernel extreme learning machine (ADKELM). Firstly, the wavelet threshold denoising method is employed to eliminate the noise in the vibration signal. Then, the empirical wavelet transform (EWT) is utilized to decompose the denoised signal, and extract the attention entropy of the intrinsic mode function (IMF) as the feature vector. Next, the hyperparameters of the deep kernel extreme learning machine (DKELM) are optimized using the marine predators algorithm (MPA), so as to achieve the adaptive changes in the DKELM parameters. By analyzing the fault diagnosis performances of the ADKELM model with different kernel functions and hidden layers, the optimal ADKELM model is determined. Compared with conventional machine learning models such as extreme learning machine (ELM), back propagation neural network (BPNN) and probabilistic neural network (PNN), the high efficiency of the method proposed in this paper is verified.

Seed germination and stand establishment are the first steps of crop growth and development. However, low seed vigor, improper seedbed preparation, unfavorable climate, and the occurrence of pests and diseases reduces the germination rate and seedling quality, resulting in insufficient crop populations and undesirable plant growth. Seed coating is an effective method that is being developed and applied in modern agriculture. It has many functions, such as improving seed vigor, promoting seedling growth, and reducing the occurrence of pests and diseases. Yet, during seed coating procedures, several factors, such as difficulty in biodegradation of coating materials and hindrance in the application of chemical ingredients to seeds, force us to explore reliable and efficient coating formulations. Biochar, as a novel material, may be expected to enhance seed germination and seedling establishment, simultaneously ensuring agricultural sustainability, environment, and food safety. Recently, biochar-based seed coating has gained much interest due to biochar possessing high porosity and water holding capacity, as well as wealthy nutrients, and has been proven to be a beneficial agent in seed coating formulations. This review presents an extensive overview on the history, methods, and coating agents of seed coating. Additionally, biochar, as a promising seed coating agent, is also synthesized on its physico-chemical properties. Combining seed coating with biochar, we discussed in detail the agricultural applications of biochar-based seed coating, such as the promotion of seed germination and stand establishment, the improvement of plant growth and nutrition, suitable carriers for microbial inoculants, and increase in herbicide selectivity. Therefore, this paper could be a good source of information on the current advance and future perspectives of biochar-based seed coating for modern agriculture.

Rice production is often affected by biotic and abiotic stressors. The breeding of resistant cultivars is a cost-cutting and environmentally friendly strategy to maintain a sustainable high production level. An elite water-saving and drought-resistant rice (WDR), Hanhui3, is susceptible to blast and bacterial blight (BB). This study was conducted to introgress three resistance genes (Pi2, xa5, and Xa23) for blast and BB into Hanhui3, using marker-assisted selection (MAS) for the foreground selection and a whole-genome single-nucleotide polymorphism (SNP) array for the background selection. As revealed by the whole-genome SNP array, the recurrent parent genome (RPG) recovery of the improved NIL was 94.2%. The resistance levels to blast and BB of the improved NIL and its derived hybrids were higher than that of the controls. In addition, the improved NIL and its derived hybrids retained the desired agronomic traits from Hanhui3, such as yield. The improved NIL could be useful to enhance resistance against biotic stressors and produce stable grain yields in Oryza sativa subspecies indica rice breeding programs.

Drought stress adversely affects crop growth and productivity worldwide. In response, plants have evolved several strategies in which numerous genes are induced to counter stress. High mobility group (HMG) proteins are the second most abundant family of chromosomal proteins. They play a crucial role in gene transcriptional regulation by modulating the chromatin/DNA structure. In this study, we isolated a novel HMG gene, OsHMGB707, one of the candidate genes localized in the quantitative trait loci (QTL) interval of rice drought tolerance, and examined its function on rice stress tolerance. The expression of OsHMGB707 was up-regulated by dehydration and high salt treatment. Its overexpression significantly enhanced drought tolerance in transgenic rice plants, whereas its knockdown through RNA interference (RNAi) did not affect the drought tolerance of the transgenic rice plants. Notably, OsHMGB707-GFP is localized in the cell nucleus, and OsHMGB707 is protein-bound to the synthetic four-way junction DNA. Several genes were up-regulated in OsHMGB707-overexpression (OE) rice lines compared to the wild-type rice varieties. Some of the genes encode stress-related proteins (e.g., DREB transcription factors, heat shock protein 20, and heat shock protein DnaJ). In summary, OsHMGB707 encodes a stress-responsive high mobility group protein and regulates rice drought tolerance by promoting the expression of stress-related genes.

The effect of preparation conditions (molar ratio of S/Mo, hydrothermal temperature and time) on the morphologies of MoS2 was investigated. The as-synthesized materials were characterized by X-ray diffraction (XRD), field emission scanning microscopy (FESEM), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), Raman spectra and Brunauer–Emmett–Teller’s (BET) surface area measurement. Results showed that the three kinds of morphologies of MoS2 were obtained. Morphologies varied gradually from the coral-like aggregated particles to flower-like spheres to wrapped nano-sheet structure with increasing hydrothermal temperature and time. It is meaningful for specific morphology demand in different fields. Moreover, the prepared materials were used as tentative adsorbents for the removal of bisphenol A (BPA) as a representative of endocrine disruptors. The results showed that the prepared MoS2 materials with wrapped nano-sheet structure had good adsorption capacity (39.03 mg/g) and fast adsorption rate (0.0053 g/(mg/min). The adsorption process was thermodynamically exothermic and spontaneous. The study indicated that electrostatic interaction is strong and π-π interaction between BPA and the MoS2 materials play an important role for BPA adsorption. Abstract Background Stigma exsertion rate (SER) is a key determinant of outcrossing in hybrid rice seed production. A quantitative trait locus (QTL) for stigma exsertion rate in rice, qSER-7, has previously been detected on chromosome 7 by using a F2 population derived from two indica cytoplasmic male sterility (CMS) maintainers, Huhan 1B and II-32B. Results The chromosomal location of qSER-7 was precisely delimited by fine-scale mapping. Near-isogenic lines (NILs) were established, one of which isolated the locus in the qSER-7 II-32B line, which contains an introgressed segment of II-32B in the Huhan 1B genetic background, and exhibits a significantly higher stigma exsertion rate than that of the recurrent parent. Using 3192 individuals from the BC4F2 segregation population, the QTL qSER-7 was narrowed down to a 28.4-kb region between the markers RM3859 and Indel4373 on chromosome 7. According to the rice genome annotation database, three genes were predicted within the target region. Real-time PCR analysis showed significantly higher expression levels of LOC_Os07g15370 and LOC_Os07g15390 in II-32B than in Huhan 1B. LOC_Os07g15370(OsNRAMP5) was a previously reported gene for Mn and Cd transporter. The stigma exertion rates of OsNRAMP5-overexpressing plants were significantly higher than that of wild type plants, in contrast, a T-DNA insertion mutant osnramp5 showed a lower stigma exertion rate. Conclusions In the present study, the QTL qSER-7 was isolated to a region between the markers RM3859 and Indel4373. Two candidate genes were selected based on the expression difference between the two parents, which can facilitate the further cloning of the gene underlying the quantitative trait associated with qSER-7 as well as the marker-assisted transfer of desirable genes for stigma exsertion rate improvement in rice.

Abstract Background Drought is a major abiotic stress factor that influences the yield of crops. Basic leucine zipper motif (bZIP) transcription factors play an important regulatory role in plant drought stress responses. However, the functions of a number of bZIP transcription factors in rice are still unknown. Results In this study, a novel drought stress-related bZIP transcription factor, OsbZIP62, was identified in rice. This gene was selected from a transcriptome analysis of several typical rice varieties with different drought tolerances. OsbZIP62 expression was induced by drought, hydrogen peroxide, and abscisic acid (ABA) treatment. Overexpression of OsbZIP62-VP64 (OsbZIP62V) enhanced the drought tolerance and oxidative stress tolerance of transgenic rice, while osbzip62 mutants exhibited the opposite phenotype. OsbZIP62-GFP was localized to the nucleus, and the N-terminal sequence (amino acids 1–68) was necessary for the transcriptional activation activity of OsbZIP62. RNA-seq analysis showed that the expression of many stress-related genes (e.g., OsGL1, OsNAC10, and DSM2) was upregulated in OsbZIP62V plants. Moreover, OsbZIP62 could bind to the promoters of several putative target genes and could interact with stress/ABA-activated protein kinases (SAPKs). Conclusions OsbZIP62 is involved in ABA signalling pathways and positively regulates rice drought tolerance by regulating the expression of genes associated with stress, and this gene could be used for the genetic modification of crops with improved drought tolerance.
