This study aimed to investigate the effects of solid-state fermentation products of yeast (SFPY) on liver and intestinal health and disease resistance of common carp (Cyprinus carpio). A total of 200 common carp with an initial average weight of 2.55 ± 0.004 g were divided into 5 groups (4 replications per group and 10 fish per replication), and were fed with one of five diets, including a control diet and 4 diets supplemented with 2‰ (Y2), 3‰ (Y3), 4‰ (Y4), or 5‰ (Y5) SFPY, respectively, for 8 weeks. Results indicated that, the addition of SFPY to the diet of common carp did not affect the growth performance or survival rate of fish (P = 0.253). Interestingly, with the addition of SFPY, the triacylglycerol (TAG) content of the liver presented a linear decreasing tendency (P = 0.004), with significantly decreased in Y4 and Y5 groups (P = 0.035) compared with control. The purpose of this study was to evaluate the potential of a host-associated Bacillus subtilis 1-C-7 as a probiotic for Chinese perch (Siniperca chuatsi). Four test diets were formulated to contain graded levels of B. subtilis 1-C-7 at 0 (CY), 0.85 × 108 (Y1), 0.95 × 109 (Y2) and 0.91 × 1010 (Y3) CFU/kg diet. The test fish with initial weight 30.0 ± 1.2 g were fed the 4 test diets with 3 replicates in an indoor water-flow aquaculture system with 12 net cages (40 fish/cage) for 10 wk. Abstract Background We have previously reported that extracellular vesicles (EVs) derived from osteoblastic, osteoclastic and mixed prostate cancer cells promote osteoclast differentiation and inhibit osteoblast differentiation via transferring miR-92a-1-5p. In the present study, we focused on engineering miR-92a-1-5p into EVs and determining any therapeutic roles and mechanisms of the engineered EVs. Methods A stable prostate cancer cell line (MDA PCa 2b) overexpressing miR-92a-1-5p was constructed by lentivirus, and EVs were isolated by ultracentrifugation. The overexpression of miR-92a-1-5p in both cells and EVs was tested using qPCR. Osteoclast function was evaluated by Trap staining, mRNA expression of osteoclastic markers ctsk and trap, immunolabeling of CTSK and TRAP and microCT using either in vitro and in vivo assays. Target gene of miR-92a-1-5p was proved by a dual-luciferase reporter assay system. siRNAs were designed and used for transient expression in order to determine the role of downstream genes on osteoclast differentiation. Results Stable overexpression cells of miRNA-92a-5p was associated with EVs upregulating this microRNA, as confirmed by qPCR. Further, miR-92a-1-5p enriched EVs promote osteoclast differentiation in vitro by reducing MAPK1 and FoxO1 expression, associated with increased osteoclast function as shown by TRAP staining and mRNA expression of osteoclast functional genes. siRNA targeting MAPK1 or FoxO1 resulted in similar increase in osteoclast function. In vivo, the miR-92a-1-5p enriched EVs given via i.v. injection promote osteolysis, which was associated with reduction of MAPK1 and FoxO1 expression in bone marrow. Conclusion These experiments suggest that miR-92a-1-5p enriched EVs regulate osteoclast function via reduction of MAPK1 and FoxO1. Graphical Abstract

Vegetable and fruit classification can help all links of agricultural product circulation to better carry out inventory management, logistics planning and supply chain coordination, and improve the efficiency and response speed of the supply chain. However, the current classification of vegetables and fruits mainly relies on manual classification, which inevitably introduces the influence of human subjective factors, resulting in errors and misjudgments in the classification of vegetables and fruits. In response to this serious problem, this research proposes an efficient and reproducible novel model to classify multiple vegetables and fruits using handcrafted features. In the proposed model, preprocessing operations such as Gaussian filtering, grayscale and binarization are performed on the pictures of vegetables and fruits to improve the quality of the pictures; statistical texture features representing vegetable and fruit categories, wavelet transform features and shape features are extracted from the preprocessed images; the feature dimension reduction method of diffusion maps is used to reduce the redundant information of the combined features composed of statistical texture features, wavelet transform features and shape features; five effective machine learning methods were used to classify the types of vegetables and fruits. In this research, the proposed method was rigorously verified experimentally and the results show that the SVM classifier achieves 96.25% classification accuracy of vegetables and fruits, which proves that the proposed method is helpful to improve the quality and management level of vegetables and fruits, and provide strong support for agricultural production and supply chain.

Bamboo leaves contain high concentrations of various biologically active compounds, such as polyphenols and volatiles, making them attractive as raw resources for antioxidant additives in the food industry. Here, we investigated the total phenolic content (TPC) and total flavonoid content (TFC) of four bamboo leaf extracts from two species (Phyllostachys edulis and Chimonocalamus delicatus) at two growth stages (first and second years). Antioxidant capacity was determined based on the radical-scavenging capacity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+). We also assessed the antifungal capacity based on mycelial growth inhibition of Colletotrichum musae (C. musae), Botrytis cinerea (B. cinereain), and Alternaria alternata (A. alternata). Pearson’s correlation coefficients showed that the TPC was significantly (p < 0.01) negatively correlated with the half-maximal inhibitory concentrations against DPPH and ABTS+, whereas the TFC was positively correlated with C. musae and B. cinereain growth inhibition, which suggest that TPC and TFC might be the major contributors to the antioxidant and antifungal capacities of bamboo leaves, respectively. The volatile organic compounds (VOCs) of bamboo leaves were also analyzed using gas chromatography–ion mobility spectrometry. The VOCs included twenty-four aldehydes, eleven alcohols, four furans, seven esters, fifteen terpenes, three ketones, one pyrazine, and thirty unidentified compounds. Principal component analysis, partial least squares discriminant analysis, and hierarchical cluster analysis were performed to assess the differences in the volatile profiles of the four bamboo leaf samples, from which 23 discriminatory VOCs with variable importance in the projection values > 1 were screened, and part of them were impacted by species or growth stage. These findings provide a theoretical foundation for the use of bamboo leaves.

Yanzhiguo [Prunus napaulensis (Ser.) Steud] belongs to Rosaceae family and is consumed as wild fruit, pulp and juice. However, its potential for extracting natural pigment has not yet been explored. Herein, the components in the fresh Yanzhiguo pulp were preliminarily analyzed by liquid chromatography coupled to mass spectrometry. And, the optimal pre-treatment conditions were established for further extraction of Yanzhiguo pigment based on the a* value. Then, by combining the data from single-factor experiments and response surface methodology, the optimal extraction process was established as: 35% EtOH, a liquid-solid ratio of 200:1 mL g−1, an extraction time of 65 min, and an extraction temperature of 100 °C. Moreover, it was found that the a* value and yield had high fitness except when extracted into ethanol (EtOH) with different concentrations. Meanwhile, our result demonstrated Yanzhiguo pigment had high stability in general environments with carmine (a synthetic pigment) as control, except for extreme environments such as direct (hot) sunlight, high temperature (75 °C) and strong alkaline (pH $\geq$≥ 11). Also, Yanzhiguo pigment exhibited good antioxidant activity. Our results contribute to more information on Yanzhiguo pigment and promote its application by providing efficient extraction technology.

Dried instant rice noodles are a convenience food, and they have shown rapid development in recent years. However, there are many disadvantages of dried instant rice noodles, such as long rehydration time, high turbidity of soup, high break rate, poor taste, and disappointing textural properties. In this study, to overcome these disadvantages, freeze-dried rice noodles were prepared at freezing temperatures of −10, −20, and −40 ℃ by the vacuum freeze-drying process, which were called FDRN10, FDRN20, and FDRN40, respectively. The factors that affected the rehydration quality of freeze-dried rice noodles were investigated by analyzing the rehydration speed, microstructure, starch retrogradation, and texture characteristics at different water temperatures. The results showed that FDRN10 had the highest water content of the three freeze-dried rice noodles for the same water temperature and rehydration time. This indicated that the rehydration speed of FDRN10 was the fastest. The microstructures of the external surface and cross section were observed. FDRN10 had the loosest structure and largest aperture. The diameter of the aperture of FDRN10 was 72.58 µm, which was four times larger than that of FDRN40. This microstructure led to FDRN10 showing good water absorption. Differential scanning calorimetry showed that the enthalpy of FDRN10 was 92.98 J/g. The enthalpy of FDRN40 was the lowest (12.39 J/g). Thus, FDRN10 had the highest level of starch retrogradation among the three freeze-dried rice noodle samples, and the lowest level of starch water-absorbing swelling. Hardness analysis showed that the hardness value of FDRN10 decreased with increasing water temperature for the same rehydration time. The texture characteristics of FDRN10 were the closest to those of fresh rice noodles after rehydrating with water at 70 ℃ for 8 min. The above results indicate that the freezing temperature and rehydration temperature play important roles in the quality of freeze-dried rice noodles. This study provides an innovative process for improvement of instant-rice-noodle production.

Summary: The impairment of antibody-mediated immunity is a major factor associated with fatal cases of severe fever with thrombocytopenia syndrome (SFTS). By collating the clinical diagnosis reports of 30 SFTS cases, we discovered the overproliferation of monoclonal plasma cells (MCP cells, CD38+cLambda+cKappa−) in bone marrow, which has only been reported previously in multiple myeloma. The ratio of CD38+cLambda+ versus CD38+cKappa+ in SFTS cases with MCP cells was significantly higher than that in normal cases. MCP cells presented transient expression in the bone marrow, which was distinctly different from multiple myeloma. Moreover, the SFTS patients with MCP cells had higher clinical severity. Further, the overproliferation of MCP cells was also observed in SFTS virus (SFTSV)-infected mice with lethal infectious doses. Together, SFTSV infection induces transient overproliferation of monoclonal lambda-type plasma cells, which have important implications for the study of SFTSV pathogenesis, prognosis, and the rational development of therapeutics.

Peach fruit softening is the result of a series of complex physiological and biochemical reactions that influence shelf life and consumer acceptance; however, the precise mechanisms underlying softening remain unclear. We conducted a metabolomic study of the flesh and peel of the honey peach (Prunus persica L.) to identify critical metabolites before and after fruit softening. Compared to the pre-softening profiles, 155 peel metabolites and 91 flesh metabolites exhibited significant changes after softening (|log2(FC)| > 1; p < 0.05). These metabolites were mainly associated with carbohydrate metabolism, respiratory chain and energy metabolism (citrate cycle, pantothenate and CoA biosynthesis, nicotinate and nicotinamide metabolism, and pentose and glucuronate interconversions), reactive oxygen species (ROS) metabolism, amino acid metabolism, and pyrimidine metabolism. During peach fruit softening, energy supply, carbohydrate and amino acid metabolism, oxidative damage, and plant hormone metabolism were enhanced, whereas amino acid biosynthesis and cell growth declined. These findings contribute to our understanding of the complex mechanisms of postharvest fruit softening, and may assist breeding programs in improving peach fruit quality during storage.

Heating, ventilation, and air-conditioning (HVAC) systems consume approximately 60% of the total energy consumption in public buildings, and an effective way to reduce HVAC energy consumption is to provide accurate load forecasting. This paper proposes a load forecasting model CEEMDAN-Conv1D-BiLSTM-AM which combines empirical mode decomposition and neural networks. The load data are decomposed into fifteen sub-sequences using complete ensemble empirical mode decomposition with adaptive noise (CEEMDAN). The neural network inputs consist of the decomposition results and five exogenous variables. The neural networks contain a one-dimensional convolutional layer, a BiLSTM layer, and an attention mechanism layer. The Conv1D is employed to extract deep features from each input variable, while BiLSTM and the attention mechanism layer are used to learn the characteristics of the load time series. The five exogenous variables are selected based on the correlation analysis between external factors and load series, and the number of input steps for the model is determined through autocorrelation analysis of the load series. The performance of CEEMDAN-Conv1D-BiLSTM-AM is compared with that of five other models and the results show that the proposed model has a higher prediction accuracy than other models.

Cinnamon protects against irritable bowel syndrome with diarrhea (IBS-D) in humans, but its efficacy and underlying mechanism of action remain poorly understood. Maternally separated (MS) IBS-D rat model and 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced post-inflammatory IBS-D rat model are characterized by visceral hyperalgesia and diarrhea. This study used the two models to evaluate the effect of cinnamon extract (CE) on bowel symptoms. The MS rat model was also used to explore its underlying anti-IBS mechanism. cinnamon extract reduced defecation frequency and visceral hyperalgesia in MS rats in a dose-dependent manner and effectively improved visceral hyperalgesia in TNBS rats. The efficacy of cinnamon extract was comparable to the positive drug serotonin receptor 3 (5-HT3) selective antagonist, Ramosetron. Excessive 5-HT, a well-known pathogenic factor for IBS, in the colon and circulation of IBS rats was reduced after cinnamon extract intervention. Both, gene and protein levels of the colonic 5-HT synthetase, Tryptophan Hydroxylase 1 (Tph1), were also decreased in CE-treated IBS rats. In addition, a luciferase assay revealed that cinnamon extract and its major components, catechin, procyanidin B1/2, cinnamic acid, and cinnamyl alcohol, significantly inhibited Tph1 transcription activity in vitro. These findings illustrated that aqueous cinnamon extract partially attenuated bowel symptoms in IBS models by directly inhibiting Tph1 expression and controlling 5-HT synthesis. This provides a scientific viewpoint for the use of cinnamon as a folk medication to treat IBS.

Objectives: Androgen deprivation therapy (ADT) is a standard treatment for advanced prostate cancer (PCa). However, after 2–3 years ADT treatment, prostate cancer inevitably transits from androgen-dependent PCa (ADPC) to androgen-independent PCa (AIPC), which has a poor prognosis owing to its unclear mechanism and lack of effective therapeutic targets. Small extracellular vesicles (sEVs) play a vital role in the development of cancer. However, the role of PCa sEVs in the transformation of AIPC remains poorly understood. Materials and methods: Two different cell models were employed and compared. sEVs from ADPC cells (LNCaP) and AIPC cells (LNCaP-AI + F cells) were isolated and characterized. After co-culture of LNCaP-AI + F sEVs with LNCaP cells and of LNCaP sEVs with LNCaP-AI + F cells, androgen-independent transformation was determined respectively. Mechanically, small RNA sequencing was performed. Androgen-independent transformation was examined by the upregulation and downregulation of miRNA and downstream pathways were analyzed. Results: LNCaP-AI + F sEVs promoted the androgen-independent transformation of LNCaP cells. Interestingly, LNCaP sEVs exhibited a capacity to reverse the process.Let-7a-5p transfer was demonstrated. Furthermore, let-7a-5p overexpression promotes the androgen-independent transformation and let-7a-5p down-regulation reverses the process. Androgen receptor (AR) and PI3K/Akt pathways were identified and demonstrated by both let-7a-5p regulation and PCa sEVs coculture. Conclusions: PCa sEVs are intimately involved in the regulation of androgen-independent transformation of prostate cancer by transferring the key sEVs molecular let-7a-5p and then activating the AR and PI3K/Akt signaling pathways. Our results provide new perspectives for the development of sEVs and sEVs molecular targeted treatment approaches for AIPC patients.

This experiment was conducted to determine the effect of dietary paeonol levels on growth performance, serum biochemical parameters, antioxidant enzyme activities and transcriptional expression of antioxidant- and immune-related genes in Genetic Improvement of Farmed Tilapia (GIFT) juveniles. Six diets supplemented with 0, 60, 120, 240, 480 and 960 mg kg−1 paeonol were formulated, and each diet was randomly assigned to three replicate groups of 25 fish with an average initial weight of 23.10 ± 0.90 g for 53 days. Our results showed that weight gain (WG) increased first and then decreased with the increase of dietary paeonol levels. Serum alkaline phosphatase (ALP), albumin (ALB) and total protein (TP) were significantly increased as the increasing dietary paeonol levels. Lactate dehydrogenase (LDH) contents, alanine aminotransferase (ALT) and aspartate transaminase (AST) activities displayed a trend of decreasing or significantly decreasing in the paeonol-added groups. Supplemental paeonol in diets significantly elevated the activities of catalase (CAT), glutathione peroxidase (GPx) and Lysozymes (LYZ) in both serum and liver, while decreased the malondialdehyde (MDA) values. Furthermore, the levels of dietary paeonol had significant effects on the mRNA expression of antioxidant genes (including CAT, GPx, Maf, HMOX1) and immune genes (including irak1, IL-8, CC1, LYZ). These data suggested that dietary paeonol could improve the growth performance and some serum biochemical indices of GIFT, enhanced the antioxidant enzyme activities, and induced the expression of antioxidant- and immune-related genes. Analysis based on broken-line regression, 90.55 mg kg−1 paeonol would be suitable for tilapia juveniles.

It is no doubt that the improvement of flesh quality of Pacific white shrimp (Litopenaeus vannamei) reared in freshwater contributes to its development potential in aquaculture. In this study, we aimed to explore the effect of arginine supplementation on the flesh quality of L. vannamei reared in freshwater and its mechanism. L. vannamei were randomly fed with three diets for 56 days, of which arginine level was 10.15 g kg–1 (arginine-deficient diet), 21.82 g kg–1 (arginine-optimal diet), and 32.46 g kg–1 (arginine-excessive diet), respectively. Each diet was randomly assigned to triplicate tanks, and each tank was stocked with 35 shrimps (initial weight: 1.70 ± 0.02 g). Results showed the arginine-optimal diet increased the weight gain, flesh percentage, crude protein and flavor amino acid contents in muscle, and improved the flesh hardness by conversing fast myofibers to slow myofibers, increasing myofiber density and myofibrillar length, and promoting ornithine and collagen synthesis. The arginine-optimal diet influenced the purine metabolic pathway by reducing hypoxanthine, xanthine, and inosine contents. Ornithine, citrulline, and glutamate were identified as the key metabolites affecting flesh quality traits after arginine treatments. Only increasing the levels of dietary arginine did not result in an increase in endogenous creatine synthesis in muscle and hepatopancreas. Overall, the arginine-optimal diet improved the flesh quality traits of L. vannamei reared in freshwater due to the enhanced muscular hardness, protein deposition, and flavor, which may be contributing to the transformation of muscle fiber type and increase in protein synthesis by the metabolites of arginine (ornithine, citrulline, and glutamate).

Bone marrow (BM)-derived CD45 (BM45) cells were demonstrated to exhibit an improved antifibrotic effect on the treatment of CCL4-induced liver fibrosis by significantly increasing the level of matrix metalloproteinase 9 (MMP-9). In this study, we aimed to validate the therapeutic effect of BM45 on the treatment of liver cirrhosis and to further investigate the molecular mechanism underlying the effect of growth arrest-specific transcript 5 (GAS5) on BM45. Accordingly, GAS5 significantly suppressed miR-222 and miR-21 expression but enhanced p27 and MMP-9 expression in HepG2 and LX2 cells. Additionally, GAS5 obstructed transforming growth factor (TGF)-β-induced dysregulation of miR-222, p27, and α-smooth muscle actin (α-SMA) in mice. GAS5 showed a considerable potential to enhance the capability of BM45 in restoring the normal expression of CCL4, miR-222, miR-21, MMP-9, p27, and α-SMA that was dysregulated by alanine aminotransferase (ALT), albumin, and fibrosis. In summary, our study validated the regulatory relationship between miR-21 and MMP-9, as well as between miR-222 and p27. The overexpression of GAS5 upregulated the expression of MMP-9 and p27 via respectively reducing the miR-222 and miR-21 expression, resulting in higher BM45-induced activation of hepatic stellate cells (HSCs). Accordingly, same results were obtained in an animal model, indicating that GAS5 may exert a positive effect on the treatment of BM45 of liver cirrhosis.

Postmenopausal osteoporosis (PMOP) is a systemic metabolic bone disease in postmenopausal women. It has been known that long non-coding RNAs (lncRNAs) play a regulatory role in the progression of osteoporosis. However, the mechanism underlying the effects of exosome-derived lncRNA on regulating the occurrence and development of PMOP remains unclear. Exosomes in the serum of patients PMOP were collected and identified. RNA sequencing was performed to obtain the expression profile of exosome-derived lncRNAs in the serum of PMOP patients. RNA sequencing identified 26 differentially expressed lncRNAs from the exosomes between healthy people and PMOP patients. Among them, the expression of TCONS_00072128 was dramatically down-regulated. A co-location method was employed and searched its potential target gene caspase 8. TCONS_00072128 knockdown notably decreased the expression of caspase 8, while the osteogenic differentiation of BMSCs was also reduced. Reversely, TCONS_00072128 overexpression enhanced caspase 8 expression and osteogenic differentiation of BMSCs. Moreover, the continuous expression of caspase 8 regulated by TCONS_00072128 significantly activated inflammation pathways including NLRP3 signaling and NF-κB signaling. Simultaneously, RIPK1 which has emerged as a promising therapeutic target for the treatment of a wide range of human neurodegenerative, autoimmune, and inflammatory diseases, was also phosphorylated. The results of the present study suggested that exosome-derived lncRNA TCONS_00072128 could promote the progression of PMOP by regulating caspase 8. In addition, caspase 8 expression in BMSCs was possible to be a key regulator that balanced cell differentiation and inflammation activation.

Background: Mesenchymal stem cells (MSCs) can be easily expanded without losing the ability of multilineage differentiation, including oesteogenic, chondrogenic and adipogenic differentiation. These characters make MSCs a promising cell resource for cartilage defect repair. MSCs could be recruited by inflammatory stimulation, then home to the injury tissues. However, its capacity of homing is extremely limited. Thus, it has become extremely necessary to develop an agent or a method, which can be used to enhance the efficiency of MSCs homing. This study investigates the effect of stearic acid methyl ester (SAME) on MSCs mobilisation and cartilage regeneration. Methods: MSCs were isolated from femurs of Sprague-Dawley (SD) rats. MTT assay was used to detect effect of SAME on viability of MSCs. Transwell assay and wound healing assay were used to detect effect of SAME on migration of MSCs. RNA-seq, quantitative real-time PCR and western blot were performed to analyze the expression of RNAs and proteins. Colony forming assay and flow cytometry were used to evaluate the effect of SAME on MSCs mobilisation in vivo. A rat cartilage defect model was created to evaluate the effect of SAME on cartilage regeneration. Results: We found that SAME could promote the migration of MSCs. Interestingly, we found SAME significantly increased the expression levels of Vav1 in MSCs. On the other hand, the enhanced migration ability of MSCs induced by SAME was retarded by Vav1 small interfering RNA (siRNA) and Rho-associated protein kinase 2 (ROCK2) inhibitor. In addition, we also checked the effect of SAME on mobilisation of MSCs in vivo. The results showed that SAME increased the number of MSCs in peripheral blood and enhanced the capacity of colony formation. Finally, using a cartilage defect model in rats, we found SAME could improve cartilage repair. Conclusion: Our study demonstrates that SAME can enhance MSCs migration ability mainly through the Vav1/ROCK2 signaling pathway, which could contribute to the accelerated cartilage regeneration. The translational potential of this article: These findings provide evidence that SAME could be used as a therapeutic reagent for MSCs mobilisation and cartilage regeneration.

Snow pea is a very important vegetable, and its postharvest storage characteristics vary with species. Few studies on the differences in its storage characteristics are available. In this study, postharvest changes in metabolic rate (respiration rate and water loss rate), membrane permeability (relative conductivity), nutrient contents (total sugar, amino acids, starch), lignin, cellulose, β-Glucosidase (β-GC) enzyme activity, texture properties, PG enzyme activity and their relationship were analyzed in large sweet broad peas and small snow peas. On the 8th day of storage, we found that the respiration rate and water loss rate were increased, total sugars and total amino acids were decreased significantly in these two legume vegetables, and that metabolic rate was slower with less nutrients consumed in large sweet broad peas than in small snow peas. Throughout the 8-day whole storage, the lignin and cellulose contents were always lower in large sweet broad peas than in small snow peas. With the increasing storage time, small snow peas were more susceptible to lignification and fibrosis, which was observed in their texture properties. The enzyme activities related to cellulose and pectin degradation (β-GC, PG) also showed the same trend during the storage. At the late stage of storage, the taste of large sweet broad peas was better than that of small snow peas. In conclusion, the storage period of large sweet broad peas was longer than that of the small snow peas, and its lignification degree was lower than that of the small snow peas. Meanwhile, senescence and lignin accumulation led to hardening of snow pea during postharvest storage. Our findings provide a theoretical reference for improving the postharvest storage quality of snow pea and extending the shelf life.

To solve the problems of the impact of carbon emission reduction and low-carbon advertising on the supply chain of fresh agricultural products, a three-level low-carbon supply chain system composed of a manufacturer, a retailer and a third-party logistics service provider is taken as the research object. The profit functions of each party under the three contracts of the manufacturer bearing, the retailer bearing and the two parties jointly bearing the advertising cost are, respectively, established to solve the optimal pricing, advertising level preservation efforts, service levels and carbon emission reduction decisions. The numerical analysis shows that, with the increase in wholesale price and the decrease in fresh-keeping price coefficient and low-carbon cost coefficient, manufacturers will choose better fresh-keeping effort level and low-carbon service level. When the proportion of advertising cost borne by the supplier increases, the benefits of all parties in the supply chain will decrease; however, when the retailer bears the advertising cost alone, the profit of the supply chain system is the largest.

Owing to the competitive advantages of fast response speed, large pushing force, high reliability, and high precision, the permanent magnet linear synchronous motor (PMLSM) has played an increasingly vital role in various high-speed and high-precision control systems. However, PMLSM exhibits nonlinear behavior in actual operation, and position tracking precision is negatively affected by friction, load changes, and other external disturbances. To meet the growing demand and solve the position tracking control problem for the PMLSM, the control system is critical. Sliding-mode control (SMC) has been used extensively in nonlinear control systems due to its superior performance characterized by simplicity, good dynamic response and insensitivity to parameter perturbation and external disturbances, and has been implemented in PMLSMs to track practical position. The objective of this article is to classify, scrutinize and review the major sliding-mode control approaches for position control of PMLSM. The three different conventional SMC methods, namely the boundary layer approach, the reaching law approach and the disturbance observer-based SMC, are discussed in detail. The four advanced forms of SMC, namely terminal SMC, super-twisting SMC, adaptive SMC and intelligent SMC, are also presented. A comparison of these approaches is given, in which the advantages and disadvantages of each approach are presented; additionally, they are presented in table form in order to facilitate reading. It is anticipated that this work will serve as a reference and provide important insight into position control of PMLSM systems.

An 8 week feeding study was conducted to investigate the effects of dietary rapeseed meal levels on growth, serum biochemical indices and flesh quality of juvenile genetically improved farmed tilapia, Oreochromis niloticus (GIFT) (initial body weight 26.81 g). Five isonitrogenous and isoenergetic diets (Diets RM1, RM2, RM3, RM4 and RM5) were formulated containing 0%, 11.6 %, 23.2 %, 34.8 % and 46.4 % of rapeseed meal, respectively. The values for weight gain, specific growth rates and feed efficiency of fish fed with RM4 and RM5 diets were significantly lower than those of fish fed with other diets. Fish fed with RM4 and RM5 diets had significantly lower serum alkaline phosphatase activity and total protein content than those fed with RM1 diet. The highest serum triglyceride concentration was observed in fish fed with RM5 diet. Dietary treatments had no significant influence on whole body and muscle moisture, crude protein, and ash contents. However, crude lipid content in the whole body and muscle of fish fed with RM4 and RM5 diets was significantly higher than that of fish fed with other diets. Lysine concentrations in the muscle decreased with increasing levels of RM. Similar trends were observed in hardness, cohesiveness, chewiness and springiness of fish. Broken-line analysis revealed that the appropriate dietary RM supplementation level of GIFT is 22.08 % based on weight gain.

Background The purpose of this study was to investigate an association between EGFR mutation status and 18F‐fluorodeoxyglucose positron emission tomography‐computed tomography (18F‐FDG PET‐CT) image features in lung adenocarcinoma. Methods Retrospective analysis of the data of 139 patients with lung adenocarcinoma confirmed by surgical pathology who underwent preoperative 18F‐FDG PET‐CT was conducted. Correlations between EGFR mutation status, clinical characteristics, and PET‐CT parameters, including the maximum standardized uptake value (SUVmax), the mean of the SUV (SUVmean), the peak of the SUV (SUVpeak) of the primary tumor, and the ratio of SUVmax between the primary tumor and the mediastinal blood pool (SUVratio), were statistically analyzed. Multivariate logistic regression analysis was performed to identify predictors of EGFR mutation. Receiver operating characteristic curves of statistical quantitative parameters were compared. Results EGFR mutations were detected in 74 (53.2%) of the 139 lung adenocarcinomas and were more frequent in non‐smoking patients. Univariate analysis showed that the SUVmax, SUVmean, SUVpeak, and SUVratio were lower in EGFR‐mutated than in wild‐type tumors. The receiver operating characteristic curves showed no significant differences between their diagnostic efficiencies. Multivariate logistic regression analysis showed that being a never smoker was an independent predictor of EGFR mutation. Conclusion Quantitative parameters based on 18F‐FDG PET‐CT have modest power to predict the presence of EGFR mutation in lung adenocarcinoma; however, when compared to smoking history, they are not good or significant predictive factors.

Statistical shape models (SSMs) have been established as robust anatomical priors for medical image segmentation, registration and anatomy modelling. To construct an SSM which accurately models the inter-subject anatomical variations, it is crucial to compute accurate shape correspondence between the training samples. To achieve this goal, the state-of-the-art shape correspondence computation methods always require tedious segmentation of the training images, while they seldom pay enough attention to the correspondence accuracy of key anatomical landmarks like the bone joints, vessel bifurcations, etc. As a result, the computation of shape correspondence is time-consuming and the correspondence accuracy is imperfect. To solve these problems, this paper proposes a novel shape correspondence computation approach which eliminates the need for image segmentation by registering an organ shape template to the training images. This method allows the human expert to specify key anatomical landmarks in the training images to define the correspondence of the crucial landmarks. An intensity-and-landmark-combined strategy is implemented to utilized both the image intensity and expert landmarks to obtain accurate shape correspondence. This method is evaluated for the construction of head anatomy SSM and spine SSM based on computed tomography (CT) images. The SSMs constructed using the proposed method demonstrates better shape correspondence accuracy than other state-of-the-arts correspondence methods. In particular, this method obtains pixel-level surface correspondence accuracy (1.38 mm) for the skull and sub-pixel level accuracy (0.92 mm) for the spine. The generalisability and specificity of the SSMs constructed using our method are also superior to SSMs constructed using other compared correspondence methods. With this method, we propose a novel approach which takes less human intervention and produces higher quality SSM with better shape modelling accuracy.

Since permanent magnet linear synchronous motors are widely used in fatigue testing, in this paper, the thrust of a permanent magnet linear synchronous motor (PMLSM) is amplified more than 10 times by the method of resonance. Firstly, the air gap magnetic field is analyzed by the equivalent magnetization current method (EMC), the electromagnetic thrust is calculated, and the expression is given by the Maxwell tensor method. The vibration analysis of the whole machine is used to obtain the conditions under which the motor resonates. The dynamic and static characteristics of the motor are analyzed through finite element simulation, the results of the motor design are judged to be reasonable, and the theoretical calculation results are compared with the simulation results to verify the accuracy of the theoretical calculation. The accuracy of the simulation results is verified by static force experiments. Finally, the rated thrust of the motor was enlarged more than 10 times by resonance experiments.

Honey peach (Prunus persica L.) is highly nutritious; it is an excellent source of sugars, proteins, amino acids, vitamins, and mineral elements. However, it is a perishable climacteric fruit that is difficult to preserve. In this study, “Feicheng” honey peach fruit was used as a test material to investigate the synergistic preservation effect of 1-methylcyclopropene (1-MCP) and laser microporous film (LMF). The peach fruits were fumigated for 24 h with 2 μL L−1 1-MCP, then packed in LMF. In comparison with the control treatment, 1-MCP + LMF treatment markedly decreased the respiration rate, weight loss, and rot rate of peach fruits. Moreover, the combination of 1-MCP and LMF suppressed the increase in soluble solids (SS) and reducing sugars (RS), as well as the decrease in titratable acid (TA) and ascorbic acid (AsA). The combined application also maintained a high protopectin content and low soluble pectin content; it reduced the accumulation of superoxide anions (O2−) and hydrogen peroxide (H2O2). Except in a few samples, the catalase (CAT) and ascorbate peroxidase (APX) activities were higher when treated by 1-MCP + LMF. Conversely, the phenylalanine deaminase (PAL), peroxidase (POD), lipase, lipoxygenase (LOX), polygalacturonase (PG), β-glucosidase, and cellulase (Cx) activities were lower than in the control. Furthermore, 1-MCP + LMF treatment reduced the relative abundances of dominant pathogenic fungi (e.g., Streptomyces, Stachybotrys, and Issa sp.). The combined treatment improved the relative abundances of antagonistic fungi (e.g., Aureobasidium and Holtermanniella). The results indicated that the co-application of 1-MCP and LMF markedly reduced weight loss and spoilage, delayed the decline of nutritional quality, and inhibited the physiological and biochemical metabolic activities of peach during storage. These changes extended its shelf-life to 28 days at 5 °C. The results provide a reference for the commercial application of this technology.

Abstract In patients with prostate cancer (PCa), bone lesions appear osteoblastic in radiographs; however, pathological fractures frequently occur in PCa patients, and bone resorption is observed in all metastatic lesions under histopathologic assessment. The mechanisms that balance the activities of osteoblasts and osteoclasts in PCa patients remain unclear. We unexpectedly discovered that PCa exosomes are critical mediators in the regulation of bone homeostasis that results in osteoclastic lesions and thereby promotes tumor growth in bone. We evaluated how exosomes derived from osteoblastic, osteoclastic, and mixed PCa cell lines affect osteoblast and osteoclast differentiation, revealing that all three types of PCa exosomes promoted osteoclastogenesis in vitro and induced osteolysis in vivo. Mechanistically, microRNAs (miRNAs) delivered by PCa exosomes were found to play several key roles in bone homeostasis. Among the delivered miRNAs, miR‐92a‐1‐5p, the most abundant miRNA, downregulated type I collagen expression by directly targeting COL1A1, and thus promoting osteoclast differentiation and inhibiting osteoblastogenesis. Furthermore, PCa exosomes also markedly reduced type I collagen expression in vivo. Our findings not only offer a novel perspective on tumor bone metastasis, where—contrary to our initial hypothesis—exosomes derived from an osteoblastic tumor induce osteoclast differentiation, but also suggest potential therapeutic targets for PCa bone metastasis.

This study was conducted to evaluate the effect of dietary protein levels on growth performance, muscle composition, digestive enzymes activities, hemolymph biochemical indices and ovary development of pre-adult red swamp crayfish Procambarus clarkii (initial weight = 12.86 ± 0.10 g). Six isoenergetic diets (fish meal, soybean meal, gluten, and shrimp powder as main protein sources) were formulated to contain graded levels of 20.70 %, 24.70 %, 30.56 %, 35.77 %, 41.12 % and 44.64 % crude protein. Each diet was assigned to triplicate groups of 15 experimental crayfish (10 female and 5 male) for 8 weeks. Our results showed that weight gain (WG) increased with dietary protein supplementation from 20.7% to 35.77%, and then decreased with further increases in dietary protein. Broken-line regression analysis based on WG showed that the optimal dietary protein level for maximum growth performance of crayfish was 39.60 %. Feed conversion ratio (FCR) exhibited an opposite trend as observed in WG, with the lowest in crayfish fed the 30.56 % or 35.77 % protein diet. Protein efficiency ratio (PER) in crayfish fed the 41.12 % or 44.64 % protein diet was significantly lower than that in crayfish fed the 20.70–30.56 % protein diet. Hepatosomatic index (HSI) value of crayfish decreased with dietary protein supplementation and achieved minimum values at 35.77 % or 41.12 % protein diet. The highest crude protein content of tail muscle was found in crayfish fed the diet containing 35.77 % or 41.12 % protein. Activities of protease and lipase in both hepatopancreas and intestine were significantly affected by dietary protein levels, while amylase activity showed no significant difference. Dietary protein also significantly induced the contents of hemolymph total protein, albumin, globulin, glucose, total cholesterol and low-density lipoprotein, along with aspartate transaminase and alanine transaminase activities. Moreover, histological analysis revealed that the ovary development of most female crayfish fed the 35.77–41.12 % protein diet were at stage V, which had a higher frequency of mature oocytes than that of females fed other protein diets. Thus, these results provide the theoretical basis for feed formulation to determine the appropriate protein requirement of pre-adult red swamp crayfish.

Rice (Oryza sativa L.) is one of the most important food crops, feeding half of the world’s population. However, rice production is affected by cadmium (Cd) toxicity. Due to an increase in Cd-contaminated soil and rice grains, and the serious harm to human health from Cd, research on Cd uptake, transport and resistance in rice has been widely conducted, and many important advances have been made. Rice plants absorb Cd mainly from soil through roots, which is mediated by Cd absorption-related transporters, including OsNramp5, OsNramp1, OsCd1, OsZIP3, OsHIR1,OsIRT1 and OsIRT2. Cd uptake is affected by soil’s environmental factors, such as the concentrations of Cd and some other ions in soil, soil properties, and other factors can affect the bioavailability of Cd in soil. Then, Cd is transported within rice plants mediated by OsZIP6, OsZIP7, OsLCD, OsHMA2, CAL1, OsCCX2, OsLCT1 and OsMTP1, from roots to shoots and from shoots to grains. To resist Cd toxicity, rice has evolved many resistance strategies, including the deposition of Cd in cell walls, vacuolar Cd sequestration, Cd chelation, antioxidation and Cd efflux. In addition, some unresolved scientific questions surrounding Cd uptake, transport and resistance in rice are proposed for further study.

Pharmacological doses of niacin can improve lipid metabolism in humans and some homothermal model organisms, but flushing and hepatotoxicity are the main side-effects. Our previous study indicated that genetically improved farmed tilapia (GIFT, Oreochromis niloticus) fed the diet with 1000 mg/kg niacin decreased the lipid content in serum and liver, suggesting that high niacin intake also affects lipometabolism in fish. Thus, to investigate the liver health status of GIFT fed high level of niacin, a 2 × 2 factorial experiment with two niacin (100 or 1000 mg/kg diet) and two energy (16 or 18 MJ/kg diet) levels was carried out for 8 weeks. The 276 fish (mean body weight, 24.45 ± 0.07 g) were divided into four treatments, and each treatment had three replicates. Each replicate was stocked with 23 fish and fed to apparent satiety thrice daily. Neither dietary niacin nor energy level affected weight gain, specific growth rate, feed conversion ratio, condition factor, viscerosomatic index, mesenteric fat index, or liver histology and apoptosis rate of hepatocytes. The high niacin intake reduced the hepatosomatic index, alleviated glycogen and lipid accumulation in the liver, and decreased the hepatic mRNA expression level of claudin-7a, and the activities of glutathione peroxidase (GPx) and catalase (CAT), raised the hepatic glycogen distribution, and mRNA expression levels of zonula occludens-1 and tumour necrosis factor-alpha (TNF-α). In fish fed high rather than normal energy diets, high niacin intake enhanced superoxide dismutase activity, lowered GPx, CAT activities and inhibition of hydroxyl radicals capacity, and upregulated interleukin-1-beta and TNF-α mRNA expression levels in the liver. These results suggested that feeding a diet containing 1000 mg/kg niacin had no adverse effects on liver health, and the effects of high niacin intake on the hepatic antioxidative capacity and inflammatory factors mRNA expression in GIFT depended on dietary energy. Acetylshikonin, a natural naphthoquinone derivative compound from Lithospermum erythrorhyzon, has been reported to kill bacteria, suppress inflammation, and inhibit tumor growth. However, the effect of acetylshikonin on human chronic myelocytic leukemia (CML) cells apoptosis and its detailed mechanisms remains unknown. The purpose of the present study was to investigate whether acetylshikonin could inhibit proliferation or induce apoptosis of the K562 cells, and whether by regulating the NF-κB signaling pathway to suppress the development of CML. K562 cells were treated with serial diluted acetylshikonin at different concentrations. Our data showed that K562 cell growth was significantly inhibited by acetylshikonin with an IC50 of 2.03 μM at 24 h and 1.13 μM at 48 h, with increased cell cycle arrest in S-phase. The results of annexin V-FITC/PI and AO/EB staining showed that acetylshikonin induced cell apoptosis in a dose-dependent manner. K562 cells treated with acetylshikonin underwent massive apoptosis accompanied by a rapid generation of reactive oxygen species (ROS). Scavenging the ROS completely blocked the induction of apoptosis following acetylshikonin treatment. The levels of the pro-apoptotic proteins Bax, cleaved caspase-9, cleaved PARP and cleaved caspase-3 increased with increased concentrations of acetylshikonin, while the level of the anti-apoptotic protein Bcl-2 was downregulated. The levels of Cyt C and AIF, which are characteristic proteins of the mitochondria-regulated intrinsic apoptotic pathway, also increased in the cytosol after acetylshikonin treatment. However, the mitochondrial fraction of Cyt C and AIF were decreased under acetylshikonin treatment. In addition, acetylshikonin decreased Bcr-Abl expression and inhibited its downstream signaling. Acetylshikonin could lead to a blockage of the NF-κB signaling pathway via decreasing nuclear NF-κB P65 and increasing cytoplasmic NF-κB P65. Moreover, acetylshikonin significantly inhibited the phosphorylation of IkBα and IKKα/β in K562 cells. These results demonstrated that acetylshikonin significantly inhibited K562 cell growth and induced cell apoptosis through the mitochondria-regulated intrinsic apoptotic pathway. The mechanisms may involve the modulating ROS accumulation, inhibition of NF-κB and BCR-ABL expression. The inhibition of BCR-ABL expression and the inactivation of the NF-κB signaling pathway caused by acetylshikonin treatment resulted in K562 cell apoptosis. Together, our results indicate that acetylshikonin could serve as a potential therapeutic agent for the future treatment of CML.

In the paper, spatial meshing theory of involute spiroid gear drive with line contact (ISGDWLC), which is a new type of worm drive with large ratio, crossed axes and high load capacity, is presented completely for the further researches of load capacity, wear and efficiency. The methods of modeling and motion simulation are also brought forward on CATIA. Firstly, based on the spatial meshing theory, the theoretical model of ISGDWLC is proposed, including equations of flanks, meshing equation, nonundercutting conditions, conjugate zone, sliding angle and induced normal curvature. Secondly, a set of formulas for the design of ISGDWLC is provided, which has solved the problems such as undercutting, interference and redundant line of contact. In addition, in the light of simultaneous equations method the length of contact is calculated accurately, easily and rapidly. Finally, a numerical example is supplied and its sliding angle, induced normal curvature and length of contact are analyzed utilizing MATLAB. The calculations are done in a second and the analytic solutions are got. The results show that ISGDWLC not only has a simple calculation and stable instantaneous transmission ratio but also has a slight edge in local meshing performances compared to other spiroid gear drive. It has the potential to be widely used. At present, a prototype has been established according to the above work.

Abstract Background Bushen Huoxue decoction (BHD) has a significant effect on fracture rehabilitation, yet its underlying mechanism is still unknown. The purpose of this study was to explore whether BHD promotes bone marrow mesenchymal stem cell (BMSC) migration through the Wnt5a signalling pathway. Methods BHD was extracted by petroleum, and its composition was analysed. Cell viability in the presence of various concentrations of BHD for 24, 48 and 72 h was measured using a Cell Counting Kit-8 assay. Transwell assays and wound healing assays were used to observe the migration ability of BMSCs. Lentiviral vectors were used to knock down Wnt5a. Polymerase chain reaction and Western blot analyses were used to further compare Wnt5a signalling components at the mRNA and protein levels between groups. Results BHD treatment groups showed increased migration ability and Wnt5a expression. Knocking down Wnt5a using a lentivirus significantly inhibited the effects of BHD, which implies that BHD promotes BMSC migration ability through activation of Wnt5a. Conclusions BHD can enhance BMSC migration, possibly by activating Wnt5a signalling.

To date, many digital atlases of the human head have been developed, but few incorporate the anatomical variations of the population. In this paper, for the first time, we constructed two digital head atlases (DHAs) of Chinese male and female adults, which can be deformed to simulate the inter-subject anatomical variations. The atlases were developed to assist with personalized medical treatment, population based radio magnetic simulation, and medical imaging device design. The statistical shape model approach was used to construct the atlases based on a training set of 65 computed tomography (CT) images (including those of 46 males and 19 females). To evaluate the accuracy of anatomy modeling, we compared the atlas morphometry parameters with the reference values of the Chinese population and observed the reasonable agreement between them. Compared with the existing digital human head atlases, a unique feature of the DHA is the capability of emulating individual head anatomy via model deformation. This feature is validated by registering the DHAs to patient CT images. The registered atlases provide complete anatomy of the target subjects, including the structures that are not clearly visible in the CT images.

Background The pro-tumoral action of the cluster of differentiation 147 (CD147), which is associated with the chemotherapy resistance of lung adenocarcinoma, is partly due to accelerated tumor cell glycolysis. 18F-fluorodeoxyglucose positron emission tomography (18F-FDG PET) metabolic parameters included maximal standardized uptake value (SUVmax), mean standardized uptake value (SUVmean), metabolic tumor volume (MTV), and total lesion glycolysis (TLG), which are non-invasive markers of the glucose metabolism of tumor cells in vivo. This study aimed to clarify the correlation between PET metabolic parameters and CD147 expression, and to evaluate the prognostic value of CD147 expression in resectable lung adenocarcinoma patients. Methods A total of 89 lung adenocarcinoma chemotherapy-naive patients who underwent 18F-fluorodeoxyglucose positron emission tomography and computerized tomography scan before pulmonary surgery were retrospectively analyzed. The PET metabolic parameters were calculated by 18F-FDG PET imaging, and CD147 expression was analyzed by immunohistochemistry. SUVmax, SUVmean, MTV, and TLG compared for their performance in predicting the expression of CD147 were illustrated with statistical analysis. All patients were then followed-up for survival analysis. Results The SUVmax was significantly correlated with the CD147 expression and was the primary predictor for the CD147 expression of lung adenocarcinoma. A cut-off value of the SUVmax, 9.77 allowed 85.1% sensitivity and 64.3% specificity for predicting the CD147 positive lung adenocarcinoma. CD147 expression was correlated with tumor differentiation and metastasis. Univariate survival analysis showed that CD147 expression was significantly associated with a shorter overall survival (OS) time. Multivariate analysis revealed that CD147 was an independent prognostic factor of lung adenocarcinoma patients. Conclusion The SUVmax of a primary tumor measured with 18F-FDG PET may be a simple and non-invasive marker for predicting CD147 expression in lung adenocarcinoma. CD147 is an independent prognostic factor related to OS of postoperative lung adenocarcinoma patients.

Objectives: Increasing interests have been focused on using artificial intelligence (AI) to extend prognostic value of medical imaging. Feature extraction is a critical step for successful application of AI. The aim of this study was to explore several metabolic parameters measured by 18 F-fluorodeoxyglucose positron emission tomography/computed tomography (PET/CT) as potential AI features in predicting the effectiveness of chemotherapy in patients with non-small cell lung cancer (NSCLC). Methods: A set of metabolic parameters of PET/CT and clinical characteristics were detected from 137 patients with NSCLC treated with at least 1 cycle of chemotherapy. Survival receiver–operating characteristic (ROC) analysis was used to define the more significant parameters chosen for the following survival analysis. Patient survival was analyzed by Kaplan-Meier method, log-rank test, and Cox regression. Results: Survival ROC showed that maximum standardized uptake value (SUVmax), metabolic tumor volume 50% (MTV50), and total lesion glycolysis 50% (TLG50) had larger area under the curve, and the optimal cutoff values were 11.72, 4.04, and 34.55, respectively. Univariate and multivariate analyses synergistically showed that late PET/CT stage and MTV50 >4.04 were independent factors of poor survival in patients with NSCLC who received chemotherapy. Conclusions: Several potential prognostic biomarkers of PET/CT imaging have been extracted for predicting survival and selecting patients with NSCLC who are more likely to benefit from chemotherapy. The identification may accelerate the development of AI methods to improve treatment outcome for NSCLC.

Background and objective Mediastinal involvement in lung cancer is a highly significant prognostic factor for survival, and accurate staging of the mediastinum will correctly identify patients who will benefit the most from surgery. Positron emission tomography/computed tomography (PET/CT) has become the standard imaging modality for the staging of patients with lung cancer. The aim of this study is to investigate 18-fluoro-2-deoxy-glucose (18F-FDG) PET/CT imaging in the detection of mediastinal disease in lung cancer. Methods A total of 72 patients newly diagnosed with non-small cell lung cancer (NSCLC) who underwent preoperative whole-body 18F-FDG PET/CT were retrospectively included. All patients underwent radical surgery and mediastinal lymph node dissection. Mediastinal disease was histologically confirmed in 45 of 413 lymph nodes. PET/CT doctors analyzed patients’ visual images and evaluated lymph node’s short axis, lymph node’s maximum standardized uptake value (SUVmax), node/aorta density ratio, node/aorta SUV ratio, and other parameters using the histopathological results as the reference standard. The optimal cutoff value for each ratio was determined by receiver operator characteristic curve analysis. Results Using a threshold of 0.9 for density ratio and 1.2 for SUV ratio yielded high accuracy for the detection of mediastinal disease. The lymph node’s short axis, lymph node’s SUVmax, density ratio, and SUV ratio of integrated PET/CT for the accuracy of diagnosing mediastinal lymph node was 95.2%. The diagnostic accuracy of mediastinal lymph node with conventional PET/CT was 89.8%, whereas that of PET/CT comprehensive analysis was 90.8%. Conclusion Node/aorta density ratio and SUV ratio may be complimentary to conventional visual interpretation and SUVmax measurement. The use of lymph node’s short axis, lymph node’s SUVmax, and both ratios in combination is better than either conventional PET/CT analysis or PET/CT comprehensive analysis in the assessment of mediastinal disease in NSCLC patients.

This work aims to investigate the methane emissions from integrated vertical-flow constructed wetlands (IVCWs) when ethanol is added as an external carbon source. In this study, a gradient of ethanol (0, 2, 4, 8, 16 and 32 mmol/L) was added as the carbon source in an IVCW planted with Cyperus alternifolius L. The results showed that the methane emission flux at an ethanol concentration of 32 mmol/L was 32.34 g CH4 m−2 day−1 less than that of the control experiment (0 mmol/L) and that the methane emission flux at an ethanol concentration of 16 mmol/L was 5.53 g CH4 m−2 day−1 less than that at 0 mmol/L. In addition, variations in the water quality driven by the different ethanol concentrations were found, with a redox potential range of −64 mV to +30 mV, a pH range of 6.6−6.9, a chemical oxygen demand (COD) removal rate range of 41% to 78%, and an ammonia nitrogen removal rate range of 59% to 82% after the ethanol addition. With the average CH4-C/TOC (%) value of 35% driven by ethanol, it will be beneficial to understand that CH4-C/TOC can be considered an ecological indicator of anthropogenic methanogenesis from treatment wetlands when driven by carbon sources or carbon loading. It can be concluded that adding ethanol as an external carbon source can not only meet the water quality demand of the IVCW treatment system but also stimulate and increase the average CH4 emissions from IVCWs by 23% compared with the control experiment. This finding indicates that an external carbon source can stimulate more CH4 emissions from IVCWs and shows the importance of carbon sources during sewage treatment processes when considering greenhouse emissions from treated wetlands.

The study objective was to retrospectively analyze the metabolic variables derived from 18 F-fluorodeoxyglucose (18F-FDG) positron emission tomography/computed tomography (PET/CT) as predictors of progression-free survival (PFS) and overall survival (OS) in advanced lung adenocarcinoma stratified by epidermal growth factor receptor (EGFR) mutation status. A total of 176 patients (91, EGFR mutation; 85, wild-type EGFR) who underwent 18F-FDG PET/CT before treatment were enrolled. The main 18F-FDG PET/CT-derived variables: primary tumor maximum standardized uptake value (SUVmaxT), primary tumor total lesion glycolysis (TLGT), the maximum SUVmax of all selected lesions in whole body determined using the Response Evaluation Criteria In Solid Tumors (RECIST) 1.1 criteria (SUVmaxWBR), and whole-body total TLG determined using the RECIST 1.1 criteria (TLGWBR) were measured. Survival analysis regarding TLGWBR, and other factors in advanced lung adenocarcinoma patients stratified using EGFR mutation status, were evaluated. The results indicated that high TLGWBR (≥259.85), EGFR wild-type, and high serum LDH were independent predictors of worse PFS and OS in all patients with advanced lung adenocarcinoma. Among patients with wild-type EGFR, only TLGWBR retained significance as an independent predictor of both PFS and OS. Among patients with the EGFR mutation, high serum LDH level was an independent predictor of worse PFS and OS, and high TLGWBR (≥259.85) was an independent predictor of worse PFS but not worse OS. In conclusion, TLGWBR is a promising parameter for prognostic stratification of patients with advanced lung adenocarcinoma and EGFR status; however, it cannot be used to further stratify the risk of worse OS for patients with the EGFR mutation. Further prospective studies are needed to validate our findings.

The aim of this study was to investigate the predictive value of the numbers of metabolically positive lymph nodes (MPLN) detected by 18F-fluorodeoxyglucose (18F-FDG) positron emission tomography (PET)/computed tomography (CT) in patients with locally advanced gastric cancer (LAGC).We retrospectively analyzed the records of 50 patients with LAGC (stage T2-T4) who had undergone pre-operative PET/CT examination and laparotomy (total gastrectomy, n = 11; subtotal gastrectomy, n = 13; distal gastrectomy, n = 22; and bypass with gastrojejunstomy, n = 4). The numbers of MPLN were determined by combining visual observations with semi-quantitative measurements of the maximized standardized uptake value (SUVmax). Performance was investigated in terms of predicting post-surgical overall survival (OS).The median post-surgical OS was 32.57 months (range 3.0-94 months). The numbers of MPLN were moderately correlated with the numbers of histological positive LN (r = 0.694, p = 0.001). In univariate analyses, the numbers of MPLN (≤ 2 vs. ≥3), PET/CT LN (positivity vs. negativity), SUVmax of LN (< 2.8 vs. ≥ 2.8), TNM stage (I, II vs. III, IV), and surgery type (R0 vs. non-R0) were significantly associated with OS. In multivariate analysis, surgery type (R0 vs. non-R0) and numbers of MPLN (≤ 2 vs. ≥ 3) were both independent factors for poor OS.This explored study indicates that the number of MPLN could provide additional information for LAGC prognosis. Patients with MPLNs ≥ 3 may be at the risk of the more bad outcomes, and the further clinical trials are needed.

Background and objective PET-CT have emerged as powerful imaging tools in radiotherapy to add information that might lead to a change in GTV definition in patients with non-small cell lung cancer (NSCLC). The aim of this study was to investigate the impact of co-registering 18F-FDG-PET-CT images on the gross tumor volume (GTV), the planning target volume (PTV), and critical organ dose in radiation therapy planning of NSCLC. Methods Thirty patients with stage III NSCLC, referred for radical radiation therapy, underwent PET-CT scanning. GTV was delineated on the CT imaging and PET-CT fused imaging respectively. The two images were used to develop the radiotherapy planning at the GE Advantage Sim 6.0 workstation. Dose-volume histograms (DVH) parameters were selected to be paralleled to compare the quality of the two plans and the impact of PET-CT on radiation plan (RP) was further analyzed. Results (1) PET-CT image changed the clinical stage: 3 patients (10%) upstaged and 3 patients (10%) downstaged. (2) PET-CT image altered GTV and PTV: GTV and PTV was decreased by PET-CT image fusion in 18 patients (60%) and was increased in 12 patients (40%). A significant (>25%) treatment volume modification was observed in 17 patients (56.67%). (3) PET-CT image changed the parameters of DVH: to deliver the same target dose of 60 Gy in 30 fractions, all dose-volume characteristics decreased in treatment planning for the 18 patients with decreased PTV based on PET-CT; the issue was the contrary for the 12 patients who had enlarged PTV based on PET-CT. Conclusion The use of the co-registration of PET-CT significantly reduced inaccurate outlining of the GTV and/or to geographic misses.

Background and objective Glut1 is the main carrier which intervenes the glucose uptake of cell, and the expression of Glut1 was positively correlated to the uptake of FDG. The aim of this study is to select the CT characteristics and clinicopathological parameters which can reflect the levels of glucose metabolism of human non-small lung cancer (NSLC). Methods Thirty-three cases of the NSLC (squamous carcinoma 11 cases and adenocarcinoma 22 cases) have undergone the scanning of FDG PET/CT before operation. The tumor pathological classification, degree of differentiation and lymph node metastasis were collected. The CT characteristics which included the diameter, deep lobulation, thin and short spiculation, spinous protuberance, cavity sign, vessel convergence sign and pleural retraction sign were described. Tracer uptake of suspected lesions was assessed by visual assessment and quantitatively by the maximum standardized uptake values (SUVmax). The expression of Glut1 which were examined by means of SP immunohistochemical technique was also analyzed. Results The positive rate of Glut1 was 66.67% (squamous carcinoma was 90.91% and adenocarcinoma was 54.55%). The positive rate of Glut1 and SUVmax of squamous carcinoma were significant higher than adenocarcinoma (Pbilateral=0.037, Punilateral=0.045). Sleep deprivation (SD) plays a complex role in central nervous system (CNS) diseases. Recent studies indicate that short-term SD can affect the extent of ischemic damage. The aim of this study was to investigate whether short-term SD could stimulate hippocampal neurogenesis in a rat model of global cerebral ischemia/reperfusion (GCIR).One hundred Sprague-Dawley rats were randomly divided into Sham, GCIR and short-term SD groups based on different durations of SD; the short-term SD group was randomly divided into three subgroups: the GCIR+6hSD*3d-treated, GCIR+12hSD-treated and GCIR+12hSD*3d-treated groups. The GCIR rat model was induced via the bilateral occlusion of the common carotid arteries and hemorrhagic hypotension. The rats were sleep-deprived starting at 48 h following GCIR. A Morris water maze test was used to assess learning and memory ability; cell proliferation and differentiation were analyzed via 5-bromodeoxyuridine (BrdU) and neuron-specific enolase (NSE), respectively, at 14 and 28 d; the expression of hippocampal BDNF was measured after 7 d.The different durations of short-term SD designed in our experiment exhibited improvement in cognitive function as well as increased hippocampal BDNF expression. Additionally, the short-term SD groups also showed an increased number of BrdU- and BrdU/NSE-positive cells compared with the GCIR group. Of the three short-term SD groups, the GCIR+12hSD*3d-treated group experienced the most substantial beneficial effects.Short-term SD, especially the GCIR+12hSD*3d-treated method, stimulates neurogenesis in the hippocampal dentate gyrus (DG) of rats that undergo GCIR, and BDNF may be an underlying mechanism in this process.

This study was undertaken to evaluate the efficacy of infliximab (IFX) in treatment of Crohn’s disease (CD) patients. 106 CD patients were undergoing treatment with IFX from five hospitals in Shanghai, China. Clinical remission to IFX induction therapy was defined as Crohn’s disease activity index (CDAI) < 150. Clinical response was assessed by a decrease in CDAI ≥ 70, and the failure as a CDAI was not significantly changed or increased. Ten weeks after therapy, 61 (57.5%) patients achieved clinical remission, 17 (16.0%) had clinical response, and the remaining 28 (26.4%) were failed. In remission group, significant changes were observed in CDAI, the Simple Endoscopic Score for Crohn’s Disease (SES-CD), and serum indexes. Patients with short disease duration (22.2 ± 23.2 months) and luminal lesions showed better effects compared to those with long disease duration (71.0 ± 58.2 months) or stricturing and penetrating lesions. IFX markedly downregulated Th1/Th17-mediated immune response but promoted IL-25 production in intestinal mucosa from remission group. No serious adverse events occurred to terminate treatment. Taken together, our studies demonstrated that IFX is efficacious and safe in inducing clinical remission, promoting mucosal healing, and downregulating Th1/Th17-mediated immune response in short course CD patients with luminal lesions.

OBJECT: The aim of this study was to determine the suitability of magnetic resonance spectroscopy (MRS) for screening brain tumors, based on a systematic review and meta-analysis of published data on the diagnostic performance of MRS. METHODS: The PubMed and PHMC databases were systematically searched for relevant studies up to December 2013. The sensitivities and specificities of MRS in individual studies were calculated and the pooled diagnostic accuracies, with 95% confidence intervals (CI), were assessed under a fixed-effects model. RESULTS: Twenty-four studies were included, comprising a total of 1013 participants. Overall, no heterogeneity of diagnostic effects was observed between studies. The pooled sensitivity and specificity of MRS were 80.05% (95% CI = 75.97%-83.59%) and 78.46% (95% CI: 73.40%-82.78%), respectively. The area under the summary receiver operating characteristic curve was 0.78. Stratified meta analysis showed higher sensitivity and specificity in child than adult. CSI had higher sensitivity and SV had higher specificity. Higher sensitivity and specificity were obtained in short TE value. CONCLUSION: Although the qualities of the studies included in the meta-analysis were moderate, current evidence suggests that MRS may be a valuable adjunct to magnetic resonance imaging for diagnosing brain tumors, but requires selection of suitable technique and TE value.
