Abstract Background Digital speech assessment has potential relevance in the earliest, preclinical stages of Alzheimer’s disease (AD). We evaluated the feasibility, test-retest reliability, and association with AD-related amyloid-beta (Aβ) pathology of speech acoustics measured over multiple assessments in a remote setting. Methods Fifty cognitively unimpaired adults (Age 68 ± 6.2 years, 58% female, 46% Aβ-positive) completed remote, tablet-based speech assessments (i.e., picture description, journal-prompt storytelling, verbal fluency tasks) for five days. The testing paradigm was repeated after 2–3 weeks. Acoustic speech features were automatically extracted from the voice recordings, and mean scores were calculated over the 5-day period. We assessed feasibility by adherence rates and usability ratings on the System Usability Scale (SUS) questionnaire. Test-retest reliability was examined with intraclass correlation coefficients (ICCs). We investigated the associations between acoustic features and Aβ-pathology, using linear regression models, adjusted for age, sex and education. Results The speech assessment was feasible, indicated by 91.6% adherence and usability scores of 86.0 ± 9.9. High reliability (ICC ≥ 0.75) was found across averaged speech samples. Aβ-positive individuals displayed a higher pause-to-word ratio in picture description (B = -0.05, p = 0.040) and journal-prompt storytelling (B = -0.07, p = 0.032) than Aβ-negative individuals, although this effect lost significance after correction for multiple testing. Conclusion Our findings support the feasibility and reliability of multi-day remote assessment of speech acoustics in cognitively unimpaired individuals with and without Aβ-pathology, which lays the foundation for the use of speech biomarkers in the context of early AD.

Abstract Visualizing the internal structure of museum objects is a crucial step in acquiring knowledge about the origin, state, and composition of cultural heritage artifacts. Among the most powerful techniques for exposing the interior of museum objects is computed tomography (CT), a technique that computationally forms a 3D image using hundreds of radiographs acquired in a full circular range. However, the lack of affordable and versatile CT equipment in museums, combined with the challenge of transporting precious collection objects, currently keeps this technique out of reach for most cultural heritage applications. We propose an approach for creating accurate CT reconstructions using only standard 2D radiography equipment already available in most larger museums. Specifically, we demonstrate that a combination of basic X-ray imaging equipment, a tailored marker-based image acquisition protocol, and sophisticated data-processing algorithms, can achieve 3D imaging of collection objects without the need for a costly CT imaging system. We implemented this approach in the British Museum (London), the J. Paul Getty Museum (Los Angeles), and the Rijksmuseum (Amsterdam). Our work paves the way for broad facilitation and adoption of CT technology across museums worldwide.

Urban ecological communities are shaped by an array of environmental and physical factors that include climate, biogeography, species interactions, dispersal, and ecological filters at the habitat and landscape scales. In addition, many social dynamics, decision-making processes, and other social characteristics, such as development, policy, government actions, as well as socioeconomic status of residents – the so-called ‘luxury’ effect – may also play strong roles in shaping which species occur within the urban context. This interaction between ecological and social drivers may impact species richness and may also influence functional traits and functional diversity, with important implications for ecosystem services provided within urban spaces. Urban agroecosystems provide food and other well-being benefits to urban residents, and are valuable green spaces in the city that provide refuges for biodiversity. Despite this fact, one of the biggest risks to gardens is insecure land tenure. While plant communities within gardens may be shaped by gardener demographics, food cultures, and ecological processes, little is known about how factors such as luxury and land tenure security may impact plant diversity, plant functional traits, and functional diversity, as well as species and trait composition. In this study, we ask how garden physical features, luxury, and land tenure security influence plant species richness, functional diversity, and species and trait composition within gardens in the California central coast. We surveyed vegetation, assessed plant traits related to growth form, plant defense, and floral characteristics, and collected information on multiple factors associated with land tenure and luxury. We found that land tenure secure gardens had higher plant and crop richness. Variables associated with garden luxury boosted ornamental plant richness, flower abundance and height, lowered crop richness, and strongly impacted plant species composition. Garden plot size negatively correlated with plant species and functional richness and relative abundance of trees. Weed species richness was not impacted by any social or physical feature examined. Thus overall, in community gardens, cultivated plants, and their traits, are strongly shaped by the socioeconomic factors of land tenure security and luxury as well as the spatial distribution and size of garden plots, with important implications for both conservation and ecosystem services provided by garden habitats within cities.

Cities are sometimes characterized as homogenous with species assemblages composed of abundant, generalist species having similar ecological functions. Under this assumption, rare species, or species observed infrequently, would have especially high conservation value in cities for their potential to increase functional diversity. Management to increase the number of rare species in cities could be an important conservation strategy in a rapidly urbanizing world. However, most studies of species rarity define rarity in relatively pristine environments where human management and disturbance is minimized. We know little about what species are rare, how many species are rare, and what management practices promote rare species in urban environments. Here, we identified which plants and species of birds and bees that control pests and pollinate crops are rare in urban gardens and assessed how social, biophysical factors, and cross-taxonomic comparisons influence rare species richness. We found overwhelming numbers of rare species, with more than 50% of plants observed classified as rare. Our results highlight the importance of women, older individuals, and gardeners who live closer to garden sites in increasing the number of rare plants within urban areas. Fewer rare plants were found in older gardens and gardens with more bare soil. There were more rare bird species in larger gardens and more rare bee species for which canopy cover was higher. We also found that in some cases, rarity begets rarity, with positive correlations found between the number of rare plants and bee species and between bee and bird species. Overall, our results suggest that urban gardens include a high number of species existing at low frequency and that social and biophysical factors promoting rare, planned biodiversity can cascade down to promote rare, associated biodiversity.

Purpose: This paper aims to understand the debate about what constitutes good education in the Netherlands. The meaning of the concept of democracy in these public debates is divergent and rather diffuse. If teachers, citizens, advisory councils, and the Dutch government agree that democracy ought to be anchored in future education, we first and foremost need to understand its meaning within these current debates. Approach: In this study we conducted a frame analysis of eleven key documents from three relevant domains. The diagnostic frame shows that on the whole the authors of these documents view 'neoliberalism' and a 'culture of measurement' as undermining forces in education. Findings: The prognostic frame shows that all authors frame democracy as a prognosis, but with four different meanings: 1) democracy as organizational structure, 2) democracy as governmental policy, 2) democracy as knowledge and skill, and 4) democracy as a practice. We argue that these can be interpreted as four dimensions of a democratic solution, constructed as a response to neoliberal tendencies in Dutch education

Friction in the classroom may create useful tension for teachers when they attempt to discuss sensitive topics as part of democratic learning. Due to the openness and indeterminacy of these topics, students can experience what it is like to be (political) subjects in a diverse society and become aware of other people's subjectness in a charged classroom. To better understand how to handle educational friction in the classroom, we observed and interviewed nine Dutch expert teachers and analyzed the empirical data by using our Educational Friction Modelling Framework as a heuristic lens. This study shows how teachers allowed extreme statements and used the subsequent friction during their lessons, challenged and provoked their students, made room for their pupils in several ways to enhance their participation, and made a distinction between rationality and emotions in the classroom. We argue that our framework sheds light on what charges the classroom and contributes to the further development of contained risk-taking as part of democratic education.

Abstract Biological pest control relies on interactions between herbivores and their natural enemies. Maintaining this ecosystem service requires considering herbivore and natural enemy interactions and their response to anthropogenic change at multiple scales. In this study, we used ecological networks to quantify the network structure of interactions between herbivorous insects and their parasitoids. We examined how herbivore host abundance, parasitism rates, and shifts in network structure relate to changes in local habitat management and landscape context. We sampled herbivores and parasitoids in Brassica oleracea plants at 22 urban gardens in the Central Coast of California. At each site, we measured local management characteristics (e.g., vegetation, ground cover, canopy cover) and quantified surrounding landscape composition (e.g., urban, natural, open, and agricultural cover). For the eight sites with large enough networks, we calculated three network structure metrics (interaction richness, vulnerability, and functional complementarity). We then used generalized linear and mixed models to examine relationships between herbivore host abundance, parasitism rates, garden management and landscape characteristics, and network metrics. We found that both local management and landscape composition influenced parasitism, while only local factors affected host abundance and network structure. Higher network interaction richness was marginally associated with enhanced parasitism rates for two host species and lower parasitism rates for one host species. Our results suggest that local garden management decisions may shift the structure of host–parasitoid networks, which may subsequently affect host parasitism rates, but outcomes for biological pest control will likely vary across host species.

Abstract Alzheimer’s disease (AD) is characterized by the accumulation of β-amyloid peptide (Aβ). It affects cognition and leads to memory impairment. The mitochondrial translocator protein (TSPO) plays an essential role in maintaining mitochondrial homeostasis and has been implicated in several neuronal disorders or neuronal injuries. Ligands targeting the mitochondrial translocator protein (18 kDa), promote neurosteroidogenesis and may be neuroprotective. To study whether the TSPO ligand XBD173 may exert early neuroprotective effects in AD pathology we investigated the impact of XBD173 on amyloid toxicity and neuroplasticity in mouse models of AD. We show that XBD173 (emapunil), via neurosteroid-mediated signaling and delta subunit-containing GABAA receptors, prevents the neurotoxic effect of Aβ on long-term potentiation (CA1-LTP) in the hippocampus and prevents the loss of spines. Chronic but not acute administration of XBD173 ameliorates spatial learning deficits in transgenic AD mice with arctic mutation (ArcAβ). The heterozygous TSPO-knockout crossed with the transgenic arctic mutation model of AD mice (het TSPOKO X ArcAβ) treated with XBD173 does not show this improvement in spatial learning suggesting TSPO is needed for procognitive effects of XBD173. The neuroprotective profile of XBD173 in AD pathology is further supported by a reduction in plaques and soluble Aβ levels in the cortex, increased synthesis of neurosteroids, rescued spine density, reduction of complement protein C1q deposits, and reduced astrocytic phagocytosis of functional synapses both in the hippocampus and cortex. Our findings suggest that XBD173 may exert therapeutic effects via TSPO in a mouse model of AD.

Abstract Like other urban green spaces, urban community gardens can act as biodiversity refugees, especially for small organisms like arthropods. In turn, arthropods can provide important ecosystem pest control services to these agroecosystems. Thus, an often‐asked question among urban gardeners is how to improve gardens and surrounding areas for natural enemies and associated pest control services. We examine how local vegetation and garden characteristics, as well as the surrounding landscape composition, affect ground‐dwelling beetles (Coleoptera: Carabidae and Staphylinidae), spiders (Araneae), opilionids (Opiliones), and ladybird beetles (Coleoptera: Coccinellidae), all of which are important predators. In the summer of 2019, we collected predators, vegetation, ground cover, and garden and landscape characteristic data from 10 community gardens in the city of Seattle, Washington. We found that different groups of natural enemies are associated with different environmental variables and at different scales; probably related to differences in their dispersal capabilities, habits, and diets. Floral variables (number of flowers and number of species in flower) had a negative effect on nonflying natural enemies (spiders, opilionids, and ground‐dwelling beetles), but not on flying ones (ladybird beetles). The only taxon that was significantly affected by a landscape‐scale variable was Opiliones, the only group examined that exclusively disperses by ground. Our results show contrasting results to similar studies in different regions and highlight the need to expand the taxa and regions of study.

Abstract Although X-ray imaging is used routinely in industry for high-throughput product quality control, its capability to detect internal defects has strong limitations. The main challenge stems from the superposition of multiple object features within a single X-ray view. Deep Convolutional neural networks can be trained by annotated datasets of X-ray images to detect foreign objects in real-time. However, this approach depends heavily on the availability of a large amount of data, strongly hampering the viability of industrial use with high variability between batches of products. We present a computationally efficient, CT-based approach for creating artificial single-view X-ray data based on just a few physically CT-scanned objects. By algorithmically modifying the CT-volume, a large variety of training examples is obtained. Our results show that applying the generative model to a single CT-scanned object results in image analysis accuracy that would otherwise be achieved with scans of tens of real-world samples. Our methodology leads to a strong reduction in training data needed, improved coverage of the combinations of base and foreign objects, and extensive generalizability to additional features. Once trained on just a single CT-scanned object, the resulting deep neural network can detect foreign objects in real-time with high accuracy.

Ecological networks can provide insight into how biodiversity loss and changes in species interactions impact the delivery of ecosystem services. In agroecosystems that vary in management practices, quantifying changes in ecological network structure across gradients of local and landscape composition can inform both the ecology and function of productive agroecosystems. In this study, we examined natural-enemy-herbivore co-occurrence networks associated with Brassica oleracea (cole crops), a common crop in urban agricultural systems. Specifically, we investigated how local management characteristics of urban community gardens and the landscape composition around them affect (1) the abundance of B. oleracea herbivores and their natural enemies, (2) the natural-enemy : herbivore ratio, and (3) natural-enemy-herbivore co-occurrence network metrics. We sampled herbivores and natural enemies in B. oleracea plants in 24 vegetable gardens in the California, USA central coast region. We also collected information on garden characteristics and land-use cover of the surrounding landscape (2 km radius). We found that increased floral richness and B. oleracea abundance were associated with increased parasitoid abundance, non-aphid herbivore abundance, and increased network vulnerability; increased vegetation complexity suppressed parasitoid abundance, but still boosted network vulnerability. High agricultural land-use cover in the landscape surrounding urban gardens was associated with lower predator, parasitoid, and non-aphid herbivore abundance, lower natural-enemy : herbivore ratios, lower interaction richness, and higher trophic complementarity. While we did not directly measure pest control, higher interaction richness, higher vulnerability, and lower trophic complementarity are associated with higher pest control services in other agroecosystems. Thus, if gardens function similarly to other agroecosystems, our results indicate that increasing vegetation complexity, including trees, shrubs, and plant richness, especially within gardens located in intensively farmed landscapes, could potentially enhance the biodiversity and abundance of natural enemies, supporting ecological networks associated with higher pest control services.

The importance of COVID-19 surveillance from wastewater continues to grow since case-based surveillance in the general population has been scaled back world-wide. In Berlin, Germany, quantitative and genomic wastewater monitoring for SARS-CoV-2 is performed in three wastewater treatment plants (WWTP) covering 84% of the population since December 2021. The SARS-CoV-2 Omicron sublineage JN.1 (B.2.86.1.1), was first identified from wastewater on 22 October 2023 and rapidly became the dominant sublineage. This change was accompanied by a parallel and still ongoing increase in the notification-based 7-day-hospitalization incidence of COVID-19 and COVID-19 ICU utilization, indicating increasing COVID-19 activity in the (hospital-prone) population and a higher strain on the healthcare system. In retrospect, unique mutations of JN.1 could be identified in wastewater as early as September 2023 but were of unknown relevance at the time. The timely detection of new sublineages in wastewater therefore depends on the availability of new sequences from GISAID and updates to Pango lineage definitions and Nextclade. We show that genomic wastewater surveillance provides timely public health evidence on a regional level, complementing the existing indicators.

Neuroactive steroids are known neuroprotective agents and neurotransmitter regulators. We previously found that expression of the enzymes synthesizing 5α-dihydroprogesterone (5α-DHP), allopregnanolone (ALLO), and dehydroepiandrosterone sulfate (DHEAS) were reduced in the substantia nigra (SN) of Parkinson's Disease (PD) brain. Here, concentrations of a comprehensive panel of steroids were measured in human post-mortem brains of PD patients and controls. Gas chromatography-mass spectrometry (GC/MS) was used to measure steroid levels in SN (involved in early symptoms) and prefrontal cortex (PFC) (involved later in the disease) of five control (CTR) and nine PD donors, divided into two groups: PD4 (PD-Braak stages 1–4) and PD6 (PD-Braak stages 5–6).In SN, ALLO was increased in PD4 compared to CTR and 5α-DHP and ALLO levels were diminished in PD6 compared to PD4. The ALLO metabolite 3α5α20α-hexahydroprogesterone (3α5α20α-HHP) was higher in PD4 compared to CTR. In PFC, 3α5α20α-HHP was higher in PD4 compared to both CTR and PD6.The effects of 5α-DHP, ALLO and DHEAS were tested on human post-mortem brain slices of patients and controls in culture. RNA expression of genes involved in neuroprotection, neuroinflammation and neurotransmission was analysed after 5 days of incubation with each steroid. In PD6 slices, both 5α-DHP and ALLO induced an increase of the glutamate reuptake effector GLAST1, while 5α-DHP also increased gene expression of the neuroprotective TGFB. In CTR slices, ALLO caused reduced expression of IGF1 and GLS, while DHEAS reduced the expression of p75 and the anti-apoptotic molecule APAF1.Together these data suggest that a potentially protective upregulation of ALLO occurs at early stages of PD, followed by a downregulation of progesterone metabolites at later stages that may exacerbate the pathological changes, especially in SN. Neuroprotective effects of neurosteroids are thus dependent on the neuropathological stage of the disease.

Abstract: Whether an ecological community is controlled from above or below remains a popular framework that continues generating interesting research questions and takes on especially important meaning in agroecosystems. We describe the regulation from above of three coffee herbivores, a leaf herbivore (the green coffee scale, Coccus viridis), a seed predator (the coffee berry borer, Hypothenemus hampei), and a plant pathogen (the coffee rust disease, caused by Hemelia vastatrix) by various natural enemies, emphasizing the remarkable complexity involved. We emphasize the intersection of this classical question of ecology with the burgeoning field of complex systems, including references to chaos, critical transitions, hysteresis, basin or boundary collision, and spatial self-organization, all aimed at the applied question of pest control in the coffee agroecosystem.

Human land use threatens global biodiversity and compromises multiple ecosystem functions critical to food production. Whether crop yield-related ecosystem services can be maintained by a few dominant species or rely on high richness remains unclear. Using a global database from 89 studies (with 1475 locations), we partition the relative importance of species richness, abundance, and dominance for pollination; biological pest control; and final yields in the context of ongoing land-use change. Pollinator and enemy richness directly supported ecosystem services in addition to and independent of abundance and dominance. Up to 50% of the negative effects of landscape simplification on ecosystem services was due to richness losses of service-providing organisms, with negative consequences for crop yields. Maintaining the biodiversity of ecosystem service providers is therefore vital to sustain the flow of key agroecosystem benefits to society.

Abstract Covered tightly by a thin leather skin, three early seventeenth-century cornetts from the collection of the Rijksmuseum were examined with the focus on their construction and manufacturing. One cornett of the three unexpectedly turned out to have a peculiar construction and to be made out of two sections of different wood species. The question arose whether this could be original or is the result of an extensive restoration. As the internal structure is not accessible for analysis and examination, multi-scale Computed Tomography (CT) scanning was employed to identify the exact regions of interest (ROI) and subsequently perform scans at a sufficiently high resolution in those areas. 3D images of the hollow spaces such as the tunnelling structure caused by the common furniture beetle (Anobium punctatum) criss-crossing the wood species could be computed from the 3D x-ray tomography reconstruction. This allowed to place the occurrence of the insect infestation after the joining of the two sections. Fine tool-marks, signs of construction and potential indications of earlier treatments could be visualized. These results were compared with the other two instruments of the same group and cross-referenced to instruments in other collections, in an attempt to answer questions about the instrument’s authenticity and originality. While the unusual construction out of two wood species might be the result of an extensive repair, another possible hypothesis—based on the combination of the results—is that this unique choice of original manufacturing was intentional, possibly to avoid splitting of the wood when inserting the mouthpiece or to counteract undesired vibrations when played.

Research shows that teachers and educators struggle to act when conflict appears in the classroom. This article argues that (political) conflict should not be avoided or eradicated. Teachers should enable conflict and attend to their pupils in the process, in order to enable further understanding of each other and their differences, as part of living together in a plural and diverse society. Scholars and educators often take a deliberative approach to citizen education by focusing on problem solving and consensus seeking. This article explores how conflict can be educational if we accept that antagonisms are inherent parts of human relations. The aim of this paper is not to propose moral boundaries to conflicts. Instead, it wants to contribute to a shift from teaching citizenship as conflict-free space towards learning democracy, in which educational conflict, or "friction," is seen as an important part of the political education of pupils. This paper uses democratic theory, narrative theory, and the cultural-historical theory of play as described by Dutch historian Johan Huizinga, to construct analytical tools to further understand conflict in the classroom.

Urban gardens are vital green spaces, providing food for residents and space for engaged citizenry and community development. In California, climate change conditions (heat and drought) are becoming more extreme, threatening the resilience of urban gardens. Water use restrictions limit the timing and amount of water that gardeners can access, exacerbating these climate challenges for urban food production. Together with volunteer gardeners, we examined how ambient temperature, water use, vegetation, ground cover, and soil management affect rates of soil moisture gain and loss in urban gardens for a six-week period in the summer of 2017, during the hottest part of the growing season. We found that plot-level management of soils is essential for creating urban garden plots that maintain stable levels of water within garden soils. Although plots with better soil quality (i.e. water holding capacity) experienced slower rates of soil moisture gain after a watering event, they also experienced slower rates of soil moisture loss after the event, leading to soils with more stable, less fluctuating moisture profiles over time. This may benefit gardeners because under extreme climates (such as heat and drought) and water use restrictions, maintaining more stable soils for their plants means that the soils will retain water over a longer period after each watering event. Overall, such results highlight that better soil management that improves soil quality measures such as water holding capacity are potential solutions for maintaining soil moisture and reducing water use under changing climate conditions.

The main estrogen, 17β-estradiol (E2), exerts several beneficial vascular actions through estrogen receptor α (ERα) in endothelial cells. However, the impact of other natural estrogens such as estriol (E3) and estetrol (E4) on arteries remains poorly described. In the present study, we report the effects of E3 and E4 on endothelial healing after carotid artery injuries in vivo. After endovascular injury, which preserves smooth muscle cells (SMCs), E2, E3, and E4 equally stimulated reendothelialization. By contrast, only E2 and E3 accelerated endothelial healing after perivascular injury that destroys both endothelial cells and SMCs, suggesting an important role of this latter cell type in E4’s action, which was confirmed using Cre/lox mice inactivating ERα in SMCs. In addition, E4 mediated its effects independently of ERα membrane-initiated signaling, in contrast with E2. Consistently, RNA sequencing analysis revealed that transcriptomic and cellular signatures in response to E4 profoundly differed from those of E2. Thus, whereas acceleration of endothelial healing by estrogens had been viewed as entirely dependent on endothelial ERα, these results highlight the very specific pharmacological profile of the natural estrogen E4, revealing the importance of dialogue between SMCs and endothelial cells in its arterial protection.

Abstract Renewable energy generation curtailment increases due to more frequently occurring congestions in power system operation. Post‐contingency curative congestion management actions can reduce the necessity of renewable energy curtailment by enabling a more efficient utilization of transmission capacities. In this research, the potential of curative actions to substitute renewable energy curtailment is studied considering technical and economic criteria. Therefore, a novel pricing methodology for the market‐based provision of curative actions is introduced. The method is based on the security constraint optimal power flow technique. Simulations are carried out on a modified version of the IEEE 14‐bus network and a real‐world 110 kV distribution network. Battery energy storage systems are implemented as an exemplary technology to provide curative actions. The developed method achieves a positive power system impact by reducing operational costs and maximizing renewable energy integration. Also, novel business models for merchant‐owned battery energy storage systems are unveiled. The provision of curative actions further proves to be competitive to established battery storage applications. Additionally, results of different grid expansion scenarios of the 110 kV network reveal the need to coordinate power system planning and operation more extensively in the future.

Citizenship education is an important part of current debates about future education. In contrast, educational philosopher Gert Biesta calls for a shift "from teaching citizenship to learning democracy". This study wants to contribute to this shift. A Delphi-method is applied on a Dutch case study, presenting a way to collect and analyze data from experts that can be applied in different political, cultural and geographical contexts. The Delphi panel consists of experts from three categories of expertise in the field of democracy and Dutch education. A thematic analysis of the data collected through two succeeding rounds showed four emerging themes when turning the scope from citizenship to democracy: the distribution of responsibility of teachers and school leaders, the questions revolving around the "freedom of education" secured by the Dutch constitution, the nature of "citizenship education", and most importantly, "the school as a playground to practice democracy". This final theme was singled out by the experts as a higher purpose of democracy in education. This study argues that the concept of the school as a playground to practice democracy should be the focus to make the shift from teaching citizenship to learning democracy.
