Meagre (Argyrosomus regius) is a fast-growing marine teleost and a promising candidate for the diversification of the Mediterranean aquaculture industry. Information on specific dietary requirements is necessary to improve the competitiveness of meagre farming. While the essential fatty acids (EFA) requirement of meagre fingerlings was previously estimated, knowledge regarding the EFA requirement of on-growing meagre juveniles is still lacking. To assess the EFA requirement of juvenile meagre, six isolipidic, isoproteic, semi-purified diets with different levels of n-3 long-chain polyunsaturated fatty acid (n-3 LC-PUFA) contents 0.0%, 0.3%, 0.6%, 0.9%, 1.2%, and 1.5% of diet dry matter (DM) were formulated and fed three times daily to triplicate groups of juvenile meagres of 35.6 ± 0.3 g for twelve weeks. Growth rates, whole body composition and fatty acid profile, liver lipid content as well as fatty acid profiles were assessed. In addition, the incidence of hepatic steatosis and granulomatosis was evaluated. Juvenile meagre fed the 0.0% n-3 LC-PUFA diet showed significantly lower growth and final body weight compared to all other treatments. There was an increase in growth of meagre with increasing dietary n-3 LC-PUFA levels and weight gain plateaued at levels above 0.9% n-3 LC-PUFA. Substitution of marine ingredients (FM-FO) by plant protein and oil sources can modify selenium (Se) levels in feeds. Se plays an important role in the antioxidative defence by forming part of selenoproteins. Se requirements of gilthead sea bream are not accurately determined; therefore, this study was conducted to define Se supplementation levels in low FM-FO practical diets for sea bream fingerlings. A plantbased diet containing 0.45 mg Se/kg diet was used as the basal diet. Four other diets were supplemented to contain 0.68, 0.86, 1.00 or 1.70 mg Se/kg diet, supplied as sodium selenite. Sea bream, weighing 12.6 ± 1.4 g, were distributed in triplicate groups per diet and fed for 42 days. Se supplementation up to 1.00 mg Se/kg significantly improved the growth of sea bream, whereas further increase up to 1.70 mg Se/kg diet reduced growth. The results of this study suggest that the optimum dietary levels of sodium selenite in diets with low FM-FO with basal levels of 0.45 mg Se/kg are around 0.94 mg Se/kg to promote growth of gilthead sea bream juveniles. On the contrary, dietary levels of 1.70 mg Se/kg were found to be excessive and caused growth reduction, increased catalase expression and hydropic degeneration in the liver.

Fish meal is increasingly substituted in diets for gilthead sea bream with ingredients of terrestrial origin which may affect the mineral content and availability. Among these minerals, copper (Cu) is an essential trace element whose excess may have a potential toxic effect. Since ingredients of terrestrial origin have higher Cu levels than marine ones it is important to define the optimal dietary supply of Cu. Therefore, the aim of this study was to evaluate optimal dietary inclusion level of Cu in low FM-FO diets for gilthead sea bream fingerlings. Five practical diets with low FM (10%) and FO (6%) contents were respectively supplemented with 5 levels of CuSO4 to provide 5.5, 7.4, 9.3, 11.0 and 32.0 mg Cu kg−1 diet. Sea bream fingerlings (12.6 ± 1.4 g, mean ± SD) were distributed in 15 tanks with 30 fish per tank in triplicates and randomly assigned one of the dietary treatments. The fish were fed three times a day until apparent visual satiation for 42 days. Growth was recorded at the end of the trial and samples were taken for biochemical, mineral, histology, X-ray and hepatic gene expression analyses. The results obtained suggest that gilthead sea bream fed practical diets based on plant protein sources that provide at least 5.5 mg Cu kg−1 need no additional Cu supplementation, whereas dietary contents of 11–32 mg Cu kg−1 negatively affected gilthead sea bream performance by reducing growth, increasing oxidative risk and inducing hepatic damage and cholestasis. Dietary Cu levels did not affect body weight, SGR, TGC or FCR, denoting that the level in the non-supplemented diet (5.5 mg Cu kg−1) was enough to cover the requirements for growth. However, increasing dietary Cu levels from 5.5 to 9.3 mg/kg-1 up-regulated cat gene expression. On the contrary elevation of dietary Cu levels up to 11.0 and 32.0 mg Cu kg−1 tended to reduce growth and increased liver steatosis, broken cell margin, peripheral nuclei and sinusoid dilatation which are the markers of hepatic damage and cholestasis denoting potential toxic effects of Cu.

The present study aimed to determine the effects of dietary inclusion of selenium (Se) in the form of either inorganic Se (sodium selenite, NaSe) or organic Se (hydroxy-selenomethionine, OH-SeMet) on gilthead seabream (Sparus aurata). Triplicate groups of 150 fish each (6.2 ± 0.04 g initial body weight) were distributed in 15 tanks and fed one of five experimental diets for 63 days. A control diet (C) without Se supplementation (0.8 mg Se/kg diet), was formulated and served as a basal diet for the other 4 experimental feeds, supplemented with either 0.2 or 0.5 mg Se/kg supplied in the form of NaSe or OH-SeMet. Fish growth performance, biochemical composition of liver and muscle and Se content, malondialdehyde (MDA) concentration in muscle and hepatopancreas, hepatocyte morphology and lysozyme activity in the serum were studied. Furthermore, expression of related hepatic genes, such as manganese superoxide dismutase (Mn-sod), catalase (cat), heat shock protein 70 (hsp70), and glutathione peroxidase (gpx) was also analyzed. After the trial, fish were exposed to an acute and chronic confinement stress. Blood samples for cortisol analysis were taken at 0 and 2 h after the acute stress and after 7 days of chronic stress. The highest growth rate was observed in fish fed OH-SeMet at a level of 0.2 mg/kg, but with no significant difference with fish fed the control diet with no-added Se. The lowest growth was observed in fish fed NaSe up to 0.5 mg/kg. Increase in dietary Se, particularly in the form of OH-SeMet, led to an increase in Se contents in liver and muscle. Furthermore, dietary inclusion of OH-SeMet, led to a significant (p In summary, supplementation of Se up to 0.2 mg/kg (1–1.1 mg/kg analyzed dietary Se), particularly in the form of OH-SeMet, led to a beneficial effect on growth, maintenance of hepatic morphology and improved protection of juvenile gilthead seabream from acute or chronic stress. Besides, OH-SeMet was found to be more effective than NaSe in protection against oxidative stress in fish muscle.

Meagre (Argyrosomus regius) is a promising new aquaculture species, however with some bottlenecks to high-scale commercial production. One of those constraints are the scarce information about its nutritional requirements, which difficult the production of optimized diets and can lead to pathological alterations in liver, such as hepatic steatosis. Furthermore, meagre production is widely affected by granulomatosis, an infectious or non-infectious disease that may compromise fish performance and appearance. The cause of non-infectious granulomatosis is still unclear, but nutritional deficiencies are potential explanations. Since essential fatty acids are nutrients required to sustain vital functions, growth, but also immune status and disease resistance, the present work evaluated, in a 30-day feeding trial, the effect of 5 dietary n-3 long-chain polyunsaturated fatty acid levels (0.8, 1.4, 2.0, 2.6 and 3.5% in dry weight of the diet) on liver health status of meagre fingerlings. Fish fed the lowest n-3 LC-PUFA diets (0.8 and 1.4%) showed a higher hepatosomatic index, as well as a higher hepatic lipid infiltration, which resulted in severe hepatic steatosis. Oleic acid and docosahexaenoic acid were the most important fatty acids driving the variability of meagre hepatic fatty acid composition and, consequently, hepatic histological alterations. Although fatty acid composition of phospholipids was less affected by the dietary n-3 LC-PUFA, meagre fed 0.8% n-3 LC-PUFA tended to present lower levels of essential fatty acids and higher levels of oleic acid in polar lipids. Moreover, the diet with the lowest n-3 LC-PUFA content (0.8%) led to a higher incidence of hepatic granulomas, suggesting a possible relation between essential fatty acid deficiency and hepatic granulomatosis in meagre. These results suggest that the inclusion of vegetable oils in diets for meagre fingerlings should be taken with careful, guaranteeing that, at least, 2.0% DW of n-3 LC-PUFA is available in feeds, in order to maintain the normal hepatic function in meagre.

The aim of this work is the optimization of phenolic compound extraction from three by-products of banana crops (rachis, discarded banana, and banana’s pseudostem pulp), as a way to valorize them through a green extraction process. The influence of the temperature and aqueous ethanol concentration (Et-OH) on extract properties (total phenol content (TPC) and antioxidant activity) was firstly analyzed. 78 ℃ and ethanol concentrations close to 50% yielded the best results for the three materials. The equations obtained by the response surface methodology gave a satisfactory description of the experimental data, allowing optimizing the extraction conditions. Under optimized conditions, time influence was then assessed, although this parameter seemed not influence results. Among the three by-products, rachis extract (60% Et-OH, 78 ℃, and 30 min) presented the highest TPC (796 mg gallic acid/100 g of dried material) and antioxidant activity (6.51 mg Trolox equivalents/g of dried material), followed by discarded banana, and pseudostem pulp. Under the optimal conditions, experiments were performed at a larger scale, allowing to determine the extraction yields (EY) and to characterize the extracts. The highest EY was obtained for the rachis (26%), but the extract with the highest activity was obtained for discarded banana (50% Et-OH, 78 ℃, and 60 min), which presented a TPC of 27.26 mg/g extract corresponding to 54.59 mg Trolox equivalents/g extract. This study contributes to the valorization of banana crops residues as a source of polyphenolic compounds with bioactive functions that can be extracted under economic extraction conditions. The supplementation of fish diets with OH-SeMet reduces oxidative stress and modulates immune response against bacterial infection. However, despite the importance of essential polyunsaturated fatty acids in fish nutrition and their high risk of oxidation, the potential protective effect of OH-SeMet on these essential fatty acids has not been studied in detail. Moreover, while viral infection is very relevant in seabream production, no studies have focused the Se effects against viral infection. The aim of the present study was to assess the impact of dietary supplementation with OH-SeMet on gilthead seabream fatty acid profiles, growth performance and response against viral infection. Gilthead seabream juveniles (21.73 ± 0.27 g) were fed for 91 days with three experimental diets, a control diet without supplementation of Se (0.29 mg Se kg diet−1) and two diets supplemented with OH-SeMet (0.52 and 0.79 mg Se kg diet−1). A crowding stress test was performed at week 7 and an anti-viral response challenge were conducted at the end of the feeding trial. Selenium, proximate and fatty acid composition of diets and body tissues were analyzed. Although fish growth was not affected, elevation in dietary Se proportionally raised Se content in body tissues, increased lipid content in the whole body and promoted retention and synthesis of n-3 polyunsaturated fatty acids. Specifically, a net production of DHA was observed in those fish fed diets with a higher Se content. Additionally, both monounsaturated and saturated fatty acids were significantly reduced by the increase in dietary Se. Despite the elevation of dietary Se to 0.79 mg kg−1 not affecting basal cortisol levels, 2 h post-stress plasma cortisol levels were markedly increased. Finally, at 24 h post-stimulation, dietary OH-SeMet supplementation significantly increased the expression of the antiviral response myxovirus protein gene, showing, for the first time in gilthead seabream, the importance of dietary Se levels on antiviral defense.

A preliminary approximation to characterizes the banana culture by-products, like banana flowers and banana pseudo-stem, including a comprehensive analysis of the phenolic compounds by HPLC-MS was performed in this study. Systemic granulomatosis is a growing disease with a high morbidity, which affects to the majority of farmed meagre (Argyrosomus regius). The impossibility of isolating any infectious agents has hypothesized a nutritional origin of the disease. In order to try to elucidate the nutritional origin of granulomas, juvenile meagre were fed for 15 weeks with six diets containing different levels of vitamin E and C and with or without addition of vitamin K: Diet 0 (basal premix, no K, 150 mg kg‐−1 E, 20 mg kg‐−1 C), K (added 23 mg kg‐−1 K), EC (300 mg kg‐−1 E, 70 mg kg‐−1 C), KEC (23 mg kg‐−1 K, 300 mg kg‐−1 E, 70 mg kg‐−1 C), EECC (450 mg kg‐−1 E, 230 mg kg‐−1 C) and KEECC (23 mg kg‐−1 K, 450 mg kg‐−1 E, 230 mg kg‐−1 C). The diet EC significantly increased meagre growth in terms of final weight and length. Fish fed the highest levels of vitamin E and C presented lower percentage of granulomas in liver and heart than fish fed diet 0. The scored severity of granulomatosis in liver and kidney (main affected organs) tended to be lower with dietary increase of vitamin E, C and addition of vitamin K (from 1.83 diet 0 to 1.3 diet KEECC and from 0.91 diet 0 to 0.39 diet KEECC). In liver, the diet KEECC significantly increased catalase expression compared with diet 0. In kidney tnfα expression was significantly up-regulated in fish fed diet EECC and KEECC. In heart, low vitamin E and C levels (300 and 70 mg kg‐−1, respectively) significantly increased superoxide dismutase and glutathione peroxidase expression and high addition increased the expression of tnfα and cox-2 (0 or 23 mg kg‐−1 K, 450 mg kg‐−1 E and 230 mg kg‐−1 C, diet EECC and KEECC). The results show that combination of high dietary content of vitamin K and antioxidant vitamins E and C (23, 450 and 230 mg kg‐−1, respectively) influenced in the incidence of the granulomatosis, which suggests that this pathology could be mediated by nutritional factors.

The aim of this study was to evaluate the effects of increasing dietary arachidonic acid (ARA) levels (from 1 up to 6 % of total fatty acids) on European sea bass (Dicentrarchus labrax) juveniles growth performance, tissue fatty acid profile, liver morphology as well as long chain polyunsaturated fatty acids (LC-PUFA) biosynthesis, triglyceride and cholesterol synthesis and lipid transport. A diet with total fish oil (FO) replacement and defatted fish meal (FM) containing a 0.1 g ARA g-1 diet was added to the experimental design as a negative control diet. Dietary ARA inclusion levels below 0.2 g ARA g-1 diet significantly worsened growth even only 30 days after the start of the feeding trial, whereas dietary ARA had no effect on fish survival. Liver, muscle and whole body fatty acid profile mainly reflected dietary contents and ARA content increased accordingly with ARA dietary levels. Tissue eicosapentaenoic acid (EPA), docosapentaenoic acid (DPA) and docosahexaenoic acid (DHA) levels were positively correlated among them. Hepatic lipid vacuolization increased with reduced dietary ARA levels. Expressions of fatty acyl desaturase 2 and 3-hydroxy-3-methylglutaryl-coenzyme genes were up-regulated in fish fed the negative control diet compared to the rest of the dietary treatments denoting the influence of ARA on lipid metabolism. Results obtained highlight the need to include adequate n-6 levels and not only n-3 LC-PUFA levels in European sea bass diets. Among culture conditions, rearing temperature is a key factor for achieving optimum growth and maintaining fish welfare. However, little is known about the optimum culture conditions of greater amberjack (Seriola dumerili). Thus, the aim of this study was to determine the effect of three different rearing temperatures (17, 22 and 26°C) during 120 days on growth performance, body morphometry, biochemical composition, gut transit and liver morphology of greater amberjack (Seriola dumerili) juveniles. After 120 days of rearing, fish raised at 26°C showed higher (p < .05) body weight and specific growth rate than fish held at lower temperatures, as well as improved feed utilization, protein efficiency and nutrient retention percentages. Fish stomach emptying was faster (p < .05) in fish raised at 26°C than in fish held at 22°C and 17°C. Similar results were obtained for gut transit time, being gut emptying faster (p < .05) in fish reared at 26°C than in fish cultured at lower temperatures. Rearing temperature also induced changes in fish morphology which resulted in a higher (p < .05) caudal propulsion efficiency index for fish reared at 26°C. The aim of this study was to evaluate the combination of maximum replacement of fishmeal (FM) and fish oil (FO) by alternative meals (AM) and oils (AO) to develop nutritious, but more eco-friendly diets for European sea bass ( Dicentrarchus labrax ) juveniles. European sea bass of 9.8 ± 1.5 g and 9.1 ± 0.5 cm were fed nine isonitrogenous (45% crude protein) and isolipidic (21%) diets containing gradually reduced levels of FM and FO as follows (%FM/%FO): 58/15, 20/6, 20/3, 10/6, 10/3, 5/6, 5/3 and 0/0. Another diet, 0/0 + , was similar to the 0/0 but supplemented with LC-PUFA from microbial and marine sources. After 90 days of feeding, European sea bass fed the 0/0 diet showed reduced (P + diet). Reduction of FM/FO down to 5/3 did not significantly affect fish survival or condition factor, whereas reduction to 5/6 did neither affect sea bass length or feed conversion and reduction down to 10/3 did not affected body weight, DGI and perivisceral fat or visceral weight. FM reductions down to 5% significantly reduced growth rate. FO substitution by vegetable oils (VO) led to reduced growth and increased liver to body weight ratios, liver lipid deposition and hepatocellular size. In liver and muscle, FO substitution increased n-6 fatty acids (FA) and n-6/n-3 ratio and reduced saturated FA and n-3 LC-PUFA, whereas monounsaturated FA were not increased in proportion to dietary contents. Moreover, FO reduction increased the products from Δ6 desaturase and elongases, Elovl6 and Elovl5. In conclusion, the results of this study show that it is possible to reduce dietary FM and FO levels down to 10% and 3% in mostly plant-based diets without affecting growth performance of European sea bass juveniles.

Nutrition during periconception and early development can modulate metabolic routes to prepare the offspring for adverse conditions through a process known as nutritional programming. In gilthead sea bream, replacement of fish oil (FO) with linseed oil (LO) in broodstock diets improves growth in the 4-month-old offspring challenged with low-FO and low-fishmeal (FM) diets for 1 month. The present study further investigated the effects of broodstock feeding on the same offspring when they were 16 months old and were challenged for a second time with the low-FM and low-FO diet for 2 months. The results showed that replacement of parental moderate-FO feeding with LO, combined with juvenile feeding at 4 months old with low-FM and low-FO diets, significantly (Pα-linolenic acid and linoleic acid in broodstock diets lead to a significant down-regulation of hepatic lipoprotein lipase (PPn-3, 18 : 3n-6, 20 : 5n-3, 22 : 6n-3 and 22 : 5n-6. Thus, this study demonstrated the long-term nutritional programming of gilthead sea bream through broodstock feeding, the effect of feeding a ‘reminder’ diet during juvenile stages to improve utilisation of low-FM/FO diets and fish growth as well as the regulation of gene expression along the fish’s life-cycle.

The main objective of this study was to assess the effects of graded levels of replacement of fishmeal (FM) and fish oil (FO) by terrestrial meals (TM) and vegetable oils (VO) on gut health of European sea bass (Dicentrarchus labrax) based on gut morphology, gut-associated immune system (GALT) and gut microbiota composition. Reducing the dietary FM and FO levels down to 10 and 3%, respectively did not affect the growth of European sea bass. Increase in TM and VO markedly affected anterior and posterior gut morphology and fatty acid composition, which mirrored the dietary fatty acid profile. FO replacement by VO increased lipid deposition in anterior gut lamina propria, denoting an effect in lipid metabolism and lipoprotein synthesis. In posterior gut, FM replacements by TM induced an engrossment of the submucosa, an up-regulation of the proinflammatory cytokines IL-1β and TNFα genes, a relatively high mucus production and changes in mucosal microbiota profiles. Finally, supplementation of 0%FM-0%FO diets with LC-PUFA contributed to maintain gut homeostasis in terms of GALT response and microbiota profiles alterations. Seven experimental isonitrogenous (50%) and isolipidic (16%) diets with different levels of α-tocopherol acetate (16, 100, 190, 285, 430, 880 and 1300 mg kg − 1 ) were tested during 72 days to evaluate growth performance, tissue composition, fillet oxidation and liver histology in meagre juveniles, Argyrosomus regius . Growth performance, feed conversion ratio (FCR) and tissue composition were similar among treatments ( P > 0.05). In the liver, no major differences were recorded in lipid and fatty acid composition but higher lipid vacuolization were observed in diets E100, E190 and E880. Muscle fatty acid profiles showed an increment of the highly unsaturated fatty acid (HUFA) and a decrease of the saturated fatty acid with the increase of dietary vitamin E, which was accompanied with a reduction of the muscle TBARS responses. Therefore, is suggested that diets for this species should be supplemented with 451 mg kg − 1 of DL-α-tocopherol acetate (496 UI of vitamin E), as determine by broken-line regression analysis of muscle TBARS, to provide good overall growth performance and improved fish quality and storage stability. Moreover, results suggest that vitamin E deficiency or excess may deteriorate fish health. Statement of relevance The optimization of the dietary vitamin E level will contribute to formulate a suitable diet for meagre, a candidate for European aquaculture diversification, that until now is being fed with diets specific for other species. This study will narrow the knowledge gap that exists regarding meagre nutritional needs.

Environmental factors such as nutritional interventions during early developmental stages affect and establish long-term metabolic changes in all animals. Diets used during the spawning period causes a nutritional programming effect in offspring of gilthead sea bream and affects long-term metabolism. Studies showed modulation of genes such as fads2 which is considered to be a rate-limiting step in the synthesis of n-3 long chain polyunsaturated fatty acids (n-3 LC-PUFA). However, it is still unknown whether this adaptation is related to the presence of precursors or limitations in the pre-formed products, n-3 LC-PUFA, contained in the diets used during nutritional programming. This study investigates the combined effects of nutritional programming through broodstock diets during the spawning period and broodfish showing higher or lower fads2 expression levels in the blood after 1-month feeding with a diet containing high levels of plant protein sources and vegetable oils (VM/VO). Broodfish showing high fads2 expression had a noticeable improvement in the spawning quality parameters as well as on the growth of 6 months old offspring when challenged with a high VM/VO diet. Further, nutritional conditioning with 18:3n-3 rich diets had an adverse effect in comparison to progeny obtained from fish fed high fish meal and fish oil (FM/FO) diets, with a reduction in growth of juveniles. Improved growth of progeny from the high fads2 broodstock combined with similar muscle fatty acid profiles is an excellent option also for tailoring and increasing the flesh n-3 LC-PUFA levels to meet the recommended dietary allowances for human consumption.

Among of the actual challenges for the sustainable aquaculture development are to lower the trophic level of the cultured species, and to increase the use of by-products from the primary sector. The golden mullet (Liza aurata) is a marine low trophic level consumer present in the Mediterranean and in the Canary Islands, where there was a significant consumption in the past. On the other hand, Aloe vera plant, which production in Canary Islands is highly world representative, contains more than 70 biologically active components which have aroused the interest for its use in aquaculture, although the enormous quantity of the generated by-products has no use at all, and has never been tested for aquaculture purposes. The objective of this study was to run the first controlled feeding test with Liza aurata in the Canary Islands, as a target species to promote its cultivation, and to evaluate the use of pure Canarian Aloe vera against different levels of the by-product, to determine the effects in growth, health, and quality parameters. Therefore, 5 diets were formulated to contain 0% of aloe inclusion (diet control), 2% of pure form of aloe (diet P2), and 2, 4 and 6% of aloe by-product (diets BP2, BP4 and BP6). At 91 days of feeding, growth, proximal and fatty acid composition of liver, muscle and whole body, serum lysozyme, serum antibacterial activity, and malonaldheyde content of liver and whole body, were measured. According to obtained results, up to 6% of the aloe by-product could be included in diets for this species, without any rejections in growth or quality parameters, although no improved results compared to the control fish could be observed. Further studies are on the way to determine the sustainability and bioeconomic impact of present results, to gain knowledge on their direct industrial applications.

Manganese (Mn) is an essential metal for fish and requirements have been established for several finfish but not for gilthead sea bream. Thus, the present study aims to establish the optimal dietary supplementation level of Mn in gilthead sea bream fingerlings fed vegetable based diets. Gilthead sea bream fingerlings (weight 12.6 ± 1.5 g, mean ± S.D.) were fed five practical diets high in vegetable ingredients (fish meal: 10%, fish oil: 6%). The diets were supplemented to contain 19, 27, 30, 41 and 66 mg Mn kg−1 as MnSO4. Four hundred and fifty sea bream fingerlings were randomly distributed in 15 tanks and fed one of the five diets until apparent satiation three times per day for 42 days. Growth parameters including feed intake, thermal growth coefficient and feed conversion ratio were calculated. At the end of the trial, samples were taken for biochemical, mineral, histological and gene expression analyses. After the feeding trial, fish almost tripled their weight, but dietary Mn levels did not affect growth parameters or survival. The high fish meal substitution levels led to high Mn contents in the basal diet (19 mg Mn kg−1 diet), that seemed to be sufficient to promote sea bream growth. Body lipid composition, protein and ash were not affected by the dietary Mn. Similarly, whole body, liver and vertebrae mineral contents were not affected by Mn supplementation. Morphological characteristics of liver had no significant differences among dietary Mn levels. However, increase of Mn contents beyond 30 mg Mn kg−1 down-regulated mnsod expression. Expression of cat gene was not affected. Overall, results suggest that the Mn content present in the basal diet (19 mg Mn kg−1) was sufficient to cover the requirements in juvenile gilthead sea bream fed practical plant-based diets, although results from oxidative status markers might point out the need to increase supplementation levels beyond this point when fish are under conditions that may affect their oxidative status.

Fish and feed waste provide most of the nutrients required by the plants in aquaponics if the optimum ratio between daily fish feed inputs and the plant growing area is sustained. Thus, the fish feed needs to fulfil both the fish’s and plant’s nutritional requirements in an aquaponic system. A controlled fish waste production strategy where the nitrogen, phosphorus and mineral contents of fish diets are manipulated and used provides a way of influencing the rates of accumulation of nutrients, thereby reducing the need for the additional supplementation of nutrients. To optimize the performance and cost-effectiveness of aquaponic production, fish diets and feeding schedules should be designed carefully to provide nutrients at the right level and time to complement fish, bacteria and plants. To achieve this, a species-specific tailor-made aquaponic feed may be optimized to suit the aquaponic system as a whole. The optimal point would be determined based on overall system performance parameters, including economic and environmental sustainability measures. This chapter thus focuses on fish diets and feed and reviews the state of the art in fish diets, ingredients and additives, as well as the nutritional/sustainable challenges that need to be considered when producing specific aquaponic feeds.

Traditionally, a major part of aquaculture technology requires fish oil (FO) and fish meal (FM) to produce the aquafeed for farmed species. FO is the main source of persistent organic pollutants (POPs) in fish feed. In recent years, the use of vegetable-origin ingredients in fish feeds has been increasingly studied as an alternative to reduce the levels of these lipophilic pollutants in farmed species. The aim of this study was to evaluate the effect of the use of dietary vegetable oils in the farming of European sea bass (Dicentrarchus labrax) on the contents in persistent - polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) - and semi persistent pollutants - polycyclic aromatic hydrocarbons (PAHs) and polybrominated diphenyl ethers (BDEs) - of their edible parts. A total of 60 seabass muscle pools were obtained from fish farmed employing six experimental diets, which contained different percentages of FO (6 vs. 3%) and FM (20%, 10% and 5%). We did not observe differences in the contamination level of seabass muscle in relation to the percentage of FM in their diet. However, the fish farmed using feed which had lower levels of FO (3%) showed significantly lower muscle levels of ΣPCBs and carcinogenic PAHs (Σc-PAHs), with a reduction of 25.6% and 95.11% (respectively), as compared with those fished raised with feed with higher levels of FO (6%). Also much lower levels were found in OCPs such as sum of DDTs (30.88% of reduction), sum of chlordanes (42.85% of reduction), and sum of BDEs (48.16% of reduction) in those seabass fed with a lower percentage of FO. The results of this study indicate that the use of alternative feed ingredients that allow the employment of low percentage of FO in feeds help to reduce the load of several toxic pollutants in the fillets of European seabass.

To date, European abalone aquaculture relies mostly on locally harvested fresh seaweeds which nutritional quality and abundance vary greatly, hence affecting abalone growth. Abalone artificial diets generally include fishmeal, limiting their utilization in ecologically sustainable aquaculture and affecting abalone quality and acceptance by the consumers. A six month feeding trial was conducted to assess the nutritional value of four different dried seaweed meals: Ulva lactuca (Chlorophyta), Gracilaria cornea (Rhodophyta), Laminaria digitata (Phaeophyta) and Palmaria palmata (Rhodophyta), as ingredients to all-vegetable-based formulated feeds for abalone Haliotis tuberculata coccinea (33.1 ± 0.8 mm and 4.7 ± 0.6 g). A mixed fresh algae diet of G. cornea and U. Ulva rigida, reared in an Integrated Multi-Trophic Aquaculture (IMTA) system, served as control. Survival rates were very high (95–98%), regardless of the diet fed. Enriched fresh algae produced a significantly higher growth for H. tuberculata coccinea (169% weight gain) than all the artificial diets (49–84% WG). Comparison among abalone fed the different formulated diets showed that the inclusion of P. palmata improved growth, condition index and dietary protein utilization. On the contrary, the use of L. digitata markedly reduced the efficiency of dietary protein since the protein-related nutritional index (PER), the percentage of protein deposited in the foot muscle as well as the meat to shell ratio recorded for animals fed this diet were the lowest, despite a higher feed intake. Large differences were found in the FA profile of fresh algae as compared with the three formulated diets. The n-3/n-6 ratio was much higher in the fresh algae and, consequently, in the foot tissues of abalone fed this diet in comparison to the one of those fed the formulated ones. The elevated contents of 20:4n-6 in the abalone fed the experimental diets and 20:5n-3 in abalone fed the fresh algae, as well as their respective metabolites, suggest that abalone have the ability to desaturate and elongate LA to ARA and ALA to EPA. Further studies are required to improve the growth obtained with these vegetable based diets, especially concerning the use of different seaweed combinations and inclusion levels, as well as the diet processing methods to improve diet water stability. The aim of this study was to assess the effects of low levels of dietary fish meal (FM) and fish oil (FO) on disease resistance and gut associated lymphoid tissue (GALT) response after an experimental intestinal infection with V. anguillarum in European sea bass (Dicentrarchus labrax) For that purpose, sea bass juveniles were fed one of four diets containing combined levels of FO and FM as follows: 20%FM/6%FO, 20%FM/3%FO, 5%FM/6%FO and 5%FM/3%FO during 153 days. At the end of the feeding trial, fish were subjected to either an in vivo exposure to a sub-lethal dose of V. anguillarum via anal inoculation or to an ex vivo exposure to V. anguillarum. Additionally, inducible nitric oxide synthase (iNOS) and tumor necrosis factor α (TNFα) gut patterns of immunopositivity were studied. Growth performance was affected by dietary FM level, however ex vivo gut bacterial translocation rates and survival after the in vivo challenge test were affected by dietary FO level. After 5 months of feeding, low dietary FM levels led to a posterior gut up-regulation of interleukin-1β (IL-1β) and TNFα, major histocompatibility complex-II (MHCII) and cyclooxygenase-2 (COX2), which in turn reduced the gut associated lymphoid tissue (GALT) capacity of response after 24 h post infection and conditioned European sea bass capacity to recover gut homeostasis 7 days post infection. Immunoreactivity to anti-iNOS and anti-TNFα presented a gradient of increased immunopositivity towards the anus, regardless of the dietary FM/FO fed. Strong positive anti-TNFα isolated enterocytes were observed in the anterior gut in relation to low levels of dietary FM/FO. Submucosa and lamina propria immunoreactivity grade was related to the amount of leucocyte populations infiltrated and goblet cells presented immunopositivity to anti-iNOS but not to anti-TNFα. Thus, reducing FO content from 6% to a 3% by VO in European sea bass diets increases ex vivo and in vivo gut bacterial translocation rates, whereas reducing FM content from 20% down to 5% up-regulates the expression of several posterior gut inflammation-related genes conditioning fish growth and GALT capacity of response after bacterial infection.

The short-snouted seahorse (Hippocampus hippocampus) is a benthic fish that bases its survival on camouflage with the environment; therefore, both in wild and in captivity specimens, differences in coloration have been observed. This study was conducted to study the effect of five different diets, based on Artemia and frozen mysis, on fish survival, growth, skin colour parameters and biochemical composition. After 102 days of feeding different diets, no effect was observed on fish survival, growth, protein, ash or humidity content, except for colour changes on yellow and red fish. Differences on seahorse lipid and total carotenoids content were obtained in the experiment because of experimental diets. Fish eating Artemia showed the highest colour pattern response during the trial, with a maximum yellowness attained of 21. Additionally, a plateau of the skin colour expression for Artemia-feeding fish was reached on day 180. A reversion of the yellowish skin colour was determined after a change of diet.

Two trials were conducted to evaluate the nutrient digestibility of river crab (RC) ( Procambarus clarkii ) and marine crab (MC) ( Chaceon affinis ) meals in diets for red porgy ( Pagrus pagrus ) and the effect on ammonia-N excretion and P and Ca retentions. In trial I, the apparent digestibility coefficient (ADC) of nutrients and energy contents in RC and MC meals were determined. Results showed that, the ADC values for protein, lipids, ash, dry matter and gross energy were significantly higher for MC than for RC meal. Both crab meals were efficiently digested despite their high chitin and ash contents. In trial II, red porgy were fed over 6 months with five diets: a control diet (CD) based on high quality fish meal (FM), and four diets where FM protein was replaced at a rate of 10% or 20% by protein derived from either RC or MC meal (RC10, RC20, MC10 and MC20). Replacement of the FM protein by RC or MC proteins had no effect both on protein utilisation and ammonia-N excretion rates, which ranged from 105 to 119 mg N–NH 4 + kg − 1 day − 1 for all diets. A linear increase was observed in the dietary Ca/P ratio by the increase of the dietary RC and MC meal inclusions, resulting in a reduction in the whole fish body content of P, Ca and ash with interesting similar whole body Ca/P ratio (1.59–1.63) for all treatments. Accordingly, Ca, P and ash retention rates were significantly reduced by increasing the dietary proportion of both crab meals. Moreover, a negative relationship between fish growth and final whole body Ca, P and ash contents was obtained. However, no significant differences were observed in total P excretion value (kg P t − 1 ).

The present study was conducted to evaluate the effect of feeding marine crab (SMC) meal and sea urchin (SU) meal on growth performance, ammonia-N excretion, skin colouration and final flesh quality and lipid oxidation of red porgy ( Pagrus pagrus ). Fish were fed with five diets: a control-diet (CD); two diets replacing 10% and 20% fish meal protein by protein from SMC meal (SMC10, SMC20), and two diets with 8% and 16% inclusion of SU meal (SU8, SU16). The inclusion of both SMC and SU meals increased fish appetite response reflected in a significantly higher feed intake and final weight. Results were even better for fish fed on SU diets, with improved feed conversion (FCR) and protein utilization (PER). Ammonia-N excretion rates decreased in fish fed on SMC20, SU8 and SU16 diets. Fish skin colouration improved by SMC meal inclusion, while SU meal promoted yellowness colouration in skin. Higher muscle content of arachidonic fatty acid (ARA), was concomitant with the increased level of SMC in diets. The same trend was also found in fillet of fish fed on SU meal diets, with the highest values in ARA content. Results of TBARS in raw fillets showed that the dietary inclusion of SMC meal delayed lipid oxidation compared to FM and SU diets during refrigerated storage.

A 13-week feeding trial was carried out with juvenile rainbow trout to test two diets: a control diet without astaxanthin (AX) supplementation (CTRL diet), and a diet supplemented with 100 mg/kg of synthetic AX (ASTA diet). During the last week of the feeding trial, fish were exposed to episodic hyperoxia challenge for 8 consecutive hours per day. Episodic hyperoxia induced physiological stress responses characterized by a significant increase in plasma cortisol and hepatic glycogen and a decrease in plasma glucose levels. The decrease of plasma glucose and the increase of hepatic glycogen content due to episodic hyperoxia were emphasized with the ASTA diet. Hyperoxia led to an increase in thiobarbituric acid-reactive substances in the muscle, diminished by dietary AX supplementation in both liver and muscle. Muscle and liver AX were increased and decreased respectively after 7-day episodic hyperoxia, leading to an increase in flesh redness. This augment of muscle AX could not be attributed to AX mobilization, since plasma AX was not affected by hyperoxia. Moreover, hyperoxia decreased most of antioxidant enzyme activities in liver, whereas dietary AX supplementation specifically increased glutathione reductase activity. A higher mRNA level of hepatic glutathione reductase, thioredoxin reductase, and glutamate-cysteine ligase in trout fed the ASTA diet suggests the role of AX in glutathione and thioredoxin recycling and in de novo glutathione synthesis. Indeed, dietary AX supplementation improved the ratio between reduced and oxidized glutathione (GSH/GSSG) in liver. In addition, the ASTA diet up-regulated glucokinase and glucose-6-phosphate dehydrogenase mRNA level in the liver, signaling that dietary AX supplementation may also stimulate the oxidative phase of the pentose phosphate pathway that produces NADPH, which provides reducing power that counteracts oxidative stress. The present results provide a broader understanding of the mechanisms by which dietary AX is involved in the reduction of oxidative status.

Terrestrial protein and lipid sources are commonly used as substitutes for marine fishery-derived raw ingredients in fish diets. However, their use is related with several side-effects on marine fish performance, health, or disease resistance. Physical barriers of the skin, gills, and gut constitute the primary defense mechanism of fish. Skin mucus mucosal mucins, water, proteins, ions, and lipids determine the physical, chemical, and protective characteristics of skin mucus. Very little is known about the influence of diet composition on fish skin mucus fatty acid profile. Gilthead sea bream skin mucus contained 10% of total lipids (TL), which consisted of 50⁻60% neutral (NL) and 40⁻50% polar lipids (PL) fractions. Σn−3 long chain polyunsaturated fatty acids (LC-PUFA) deposition was preferential in the NL fraction, whereas Σn−6LC-PUFA accumulation was similar in both lipid classes. Docosahexaenoic acid (DHA; 22:6n−3) was the main LC-PUFA stored in skin mucus (14% TL) in relation to eicosapentaenoic acid (EPA; 20:5n−3) (2⁻3% TL) and arachidonic acid (ARA; 20:4n−6) (2% TL). This study denotes the importance of DHA as component of skin mucus lipids compared to other essential fatty acids, such as EPA and ARA, as well as importance of maintaining an adequate Σn−3/ Σn−6 ratio, regardless of dietary intake.

The present study was conducted to evaluate the effect of using river crab ( Procamburus clarkii ) meal (RC) and marine crab ( Chaceon affinis ) meal (MC) on the red porgy ( Pagrus pagrus ) fillet fatty acid profile and quality. Red porgy were fed during 193 days with five diets: a control diet based on high-quality fish meal (CD); and four diets replacing 10% and 20% fish meal protein by protein from both RC and MC meals (RC10, RC20 and MC10, MC20). Fillet fat content resulted significantly higher in MC20 fish than the rest of diets. Feeding with both RC and MC meals slightly increased muscle contents of polyunsaturated (PUFA), arachidonic (ARA), eicosapentaenoic (EPA), and docosahexaenoic (DHA) fatty acids and n − 3/n − 6 ratio as well as decreased n − 9 fatty acids, although difference is only significant for the ARA content. After inclusion of both crab meals, values of atherogenicity index (AI) resulted similar among treatments while thrombogenecity index (TI) was reduced, but not significantly, reflecting the higher n − 3 fatty acid content in fillets from crab diets. Sensory analysis showed that red porgy fillets of all treatments were very well appreciated with some differences detected in only MC20 fillets, which had significantly higher scores in the attributes of oily odour and flavour, cohesiveness aspect and firmness texture compared to the rest of diets. RC meal dietary inclusion did not promote any changes in sensory evaluation panel. Instrumental muscle texture did not revealed differences between treatments. Results from TBARS of raw fillets showed that the inclusion of both crab meals clearly delays lipid oxidation compared to a FM based diet, during refrigerated storage at 4 °C. Thus, for 4 and 7 days of storage, fillets of RC or MC diets, attained significantly lower TBARS values than those fed on CD.

The main objective of this study was to assess the effects of graded levels of dietary arachidonic acid (ARA), supplemented from alternative sources, on fatty acid composition of plasma and head kidney leucocytes of European sea bass (Dicentrarchus labrax). For that purpose, sea bass juveniles were fed four diets containing graded levels of ARA as follows: 0.5% (ARA0.5), 1% (ARA1), 2% (ARA2) and 4% (ARA4) during 60 days. At the end of the feeding trial fatty acid profiles of plasma and head kidney leucocytes were analyzed. Besides, plasma prostaglandins levels, head kidney leucocytes respiratory burst activity; peroxidase activity and phagocytic index were assayed. Reducing dietary ARA levels below 1% markedly reduced European sea bass growth performance. However, fish fed diet ARA0.5 tried to compensate this dietary ARA deficiency by a selective deposition of ARA on plasma and head kidney leucocytes, reaching similar levels to those fish fed diet ARA1 after 60 days of feeding. Nevertheless, head kidney phagocytic capacity was reduced as dietary ARA content in relation not only to variations on membrane composition but also to changes on fish basal prostaglandins levels. Results obtained demonstrated the importance to supply the necessary quantity n-6 LC-PUFA, and not only n-3 LC-PUFA levels, in European sea bass diets, in relation to not only growth performance but also immune system function.

Aquaponics is the combination of aquaculture (fish) and hydroponic cultivation of plants. This review examines fish welfare in relation to rearing water quality, fish feed and fish waste and faeces to develop a sustainable aquaponic system where the co-cultured organisms, fish, bacteria in biofilters and plants, should be considered holistically in all aquaponics operations. Water quality parameters are the primary environmental consideration for optimizing aquaponic production and for directly impacting fish welfare/health issues and plant needs. In aquaponic systems, the uptake of nutrients should be maximised for the healthy production of the plant biomass but without neglecting the best welfare conditions for the fish in terms of water quality. Measures to reduce the risks of the introduction or spread of diseases or infection and to increase biosecurity in aquaponics are also important. In addition, the possible impacts of allelochemicals, i.e., chemicals released by the plants, should be taken into account. Moreover, the effect of diet digestibility, faeces particle size and settling ratio on water quality should be carefully considered. As available information is very limited, research should be undertaken to better elucidate the relationship between appropriate levels of minerals needed by plants, and fish metabolism, health and welfare. It remains to be investigated whether and to what extent the concentrations of suspended solids that can be found in aquaponic systems can compromise the health of fish. The objective of this study was to evaluate three products derived from krill as sources of essential fatty acids, protein and, particularly, phospholipids in microdiets for larval gilthead seabream (Sparus aurata). Their effect on larval performance, biochemical composition and histological development was investigated. The addition of krill phospholipids, rich in highly unsaturated fatty acids, improved larval sea bream growth in terms of weight and length, enhanced hepatic utilization of dietary lipids and reduced the incidence of enterocyte injuries. These results confirm the higher nutritional value of marine phospholipids for the early development of marine fish larvae in comparison with soybean phospholipids.

River crab (RC) meal (Procambarus clarkii) and marine crab (MC) meal (Chaceon affinis) were tested as a partial replacement for fish meal in diets for red porgy (Pagrus pagrus), and their effects on growth performance, fish proximate composition and skin colouration were evaluated. Red porgy were fed during 165 days with five diets. High-quality fish meal diet was used as a control diet (CD). Protein of fish meal in the control was replaced by increasing the dietary levels of protein derived from RC and MC by up to 10% and 20% of each of them (RC10, RC20, MC10 and MC20). Fish fed on MC20 showed the highest values in feed intake, weight gain and growth (%). No differences were found in FCR and protein efficiency ratio among the treatments. Inclusion of both crab meals in diets significantly decreased the lipid content in whole fish compared with the control animals. On the other hand, no differences in muscle composition were found between the diets. Feeding both crab meals resulted in colour improvement compared with that of the control fish, with better hue values for the RC meal group than those for the MC meal group. The crab meals tested in the present study are suitable as a partial replacement for fish meal in diets for the red porgy, with the MC meal improving growth and both crabs meals improving skin colour, with further improvements in skin colour produced in fish-fed diets containing the RC meal.

Despite carotenoids and essential fatty acids seem to play important roles in fish reproduction, no studies have yet been conducted to determine the effect of dietary carotenoids on gilthead seabream broodstock performance and their relation as antioxidants with dietary n-3 highly unsaturated fatty acid (HUFA) levels. In addition, the high cost of synthetic sources of carotenoids is leading to the search for new natural carotenoid sources such as paprika oleoresin. Four experimental diets containing two combined levels of carotenoids from paprika oleoresin (40 and 60 mg kg−1) and n-3 HUFA (25 and 40 g kg−1) were respectively fed to triplicate groups of gilthead seabream (Sparus aurata) broodfish. Elevation of n-3 HUFA dietary levels from 25 to 40 g kg−1 significantly improved gilthead seabream broodstock performance in terms of egg viability, hatching rates and fecundity. Besides, it markedly increased egg contents in HUFAs which play important energetic and structural roles and improve embryo development. Both arachidonic acid and eicosapentaenoic acid (20:5n-3) egg contents were more readily affected by dietary n-3 HUFA than docosahexaenoic acid. HUFA levels did not caused any negative effect suggesting an optimized content of antioxidants in broodstock diets. Increase in dietary carotenoids from 40 to 60 mg kg−1 increased carotenoid contents in eggs and significantly improved egg fertilization rates suggesting an important sperm cell’s protective role by reducing the risk of lipid peroxidation which is detrimental for sperm motility. The increased inclusion of dietary paprika oleoresin enhanced egg carotenoid deposition and improved fish reproductive performance, denoting the high nutritional value of this product as a source of carotenoids for broodstock of this species.

Increased energy content in fish feeds has led to an enhanced fat deposition, particularly in European sea bass, concerning fish farmers. Inclusion of conjugated linoleic acid (CLA) could reduce fat deposition as in other vertebrates. The objective of this study was to determine the effect of two levels of inclusion of mannan oligosaccharides derived from the outer cell wall of a select strain of Saccharomyces cerevisiae (Bio-Mos, Alltech Inc, USA) on growth, feed utilization, immune status and disease resistance of European sea bass (Dicentrarchus labrax). Specimens of 35 g at initial density of 3 kg/m3 were fed during 67 days at 0 per thousand, 2 per thousand and 4 per thousand dietary MOS level of inclusion in a commercial sea bass diet. The Delta6 and Delta5 desaturases and elongases show only very limited activity in marine fish, and little is known of the possibility of enhancing Delta6 desaturase gene expression in these fish. The use of plant oils in marine fish diets is limited by their lack of n-3 highly unsaturated fatty acids (HUFA) despite an abundant content of the 18C fatty acid precursor linoleic and alpha-linolenic acids. The objective of the present study was to determine the ability of larval gilthead seabream to utilize vegetable oils and assess the nutritional regulation of Delta6 desaturase gene expression. Seventeen-day-old gilthead seabream larvae were fed during a 17-day period with one of four different microdiets formulated with either sardine fish oil (FO), soybean, rapeseed or linseed oils, respectively, or a fifth diet containing defatted squid meal and linseed oil. Good larval survival and growth, both in terms of total length and body weight, were obtained by feeding the larvae either rapeseed, soybean or linseed oils. The presence of vegetable oils in the diet increased the levels of 20:2n-9 and 20:2n-6, 18:2n-9, 18:3n-6, 20:3n-6 and 20:4n-6, in larvae fed rapeseed and soybean oils in comparison to those fed FO. In addition, a sixfold increase in the relative expression of Delta6 desaturase-like gene was found in larvae fed rapeseed and soybean oils, denoting the nutritional regulation of desaturase activity through its gene expression in this fish species. However, feeding linseed oil did not increase the expression of the Delta6 desaturase gene to such a high extent.
