Abstract Developments in data mining techniques have significantly influenced the progress of Intelligent Water Systems (IWSs). Learning about the hydraulic conditions enables the development of increasingly reliable predictive models of water consumption. The non-stationary, non-linear, and inherent stochasticity of water consumption data at the level of a single water meter means that the characteristics of its determinism remain impossible to observe and their burden of randomness creates interpretive difficulties. A deterministic model of water consumption was developed based on data from high temporal resolution water meters. Seven machine learning algorithms were used and compared to build predictive models. In addition, an attempt was made to estimate how many water meters data are needed for the model to bear the hallmarks of determinism. The most accurate model was obtained using Support Vector Regression (8.9%) and the determinism of the model was achieved using time series from eleven water meters of multi-family buildings.

The relationship between a cover of four grass living mulches in an apple orchard and the weed cover as well as its impact on the fruit tree yield, growth, and fruit quality was estimated at the Fruit Experimental Station of the Wrocław University of Environmental and Life Sciences in Wrocław (Poland). The experiment was established in a young orchard of ‘Chopin’ cv. apple trees grafted on rootstock MM 106, i.e. one of the strongest semi-dwarf rootstock. One year after, the tree planting soil was covered by blue fescue, red fescue, Kentucky bluegrass, and perennial ryegrass in tree rows and tractor alleys. The apple trees showed a similar degree of adaptation to the changing cultivation conditions in four different living mulches despite the varied share of the area covered by weeds. However, they did not avoid competition from different grass sods and their weediness, which was reflected in their low yield and low crop efficiency coefficient. The perennial species composition and the percentage share of the most common weeds differentiated the sod of the studied grasses. Among several perennial weed species, Trifolium repens L. was found to be the most competitive in all grass living mulches. The dynamic development of this species in time was stimulated by a rapid increase in precipitation in the orchard. The lowest soil surface cover by the total weeds was noted soon after the perennial ryegrass emergence due to the rapid development of the grass. Red fescue spread the most efficiently among all the studied grasses, and its coverage allowed effective reduction of the presence of weeds. This cover crop also maintained high purity of grass sod, especially in the tree rows, until the end of the experiment period.

Abstract Systemic sclerosis (SSc) is an autoimmune, inflammatory and fibrotic disease with limited treatment options. Developing new therapies is therefore crucial to address patient needs. To this end, we focused on galectin-3 (Gal-3), a lectin known to be associated with several pathological processes seen in SSc. Using RNA sequencing of whole-blood samples in a cross-sectional cohort of 249 patients with SSc, Gal-3 and its interactants defined a strong transcriptomic fingerprint associated with disease severity, pulmonary and cardiac malfunctions, neutrophilia and lymphopenia. We developed new Gal-3 neutralizing monoclonal antibodies (mAb), which were then evaluated in a mouse model of hypochlorous acid (HOCl)-induced SSc. We show that two of these antibodies, D11 and E07, reduced pathological skin thickening, lung and skin collagen deposition, pulmonary macrophage content, and plasma interleukin-5 and -6 levels. Moreover, E07 changed the transcriptional profiles of HOCl-treated mice, resulting in a gene expression pattern that resembled that of control mice. Similarly, pathological pathways engaged in patients with SSc were counteracted by E07 in mice. Collectively, these findings demonstrate the translational potential of Gal-3 blockade as a therapeutic option for SSc.

NDM-1 (New-Delhi-Metallo-β-lactamase-1) is an enzyme developed by bacteria that is implicated in bacteria resistance to almost all known antibiotics. In this study, we deliver a new, curated NDM-1 bioactivities database, along with a set of unifying rules for managing different activity properties and inconsistencies. We define the activity classification problem in terms of Multiple Instance Learning, employing embeddings corresponding to molecular substructures and present an ensemble ranking and classification framework, relaying on a k-fold Cross Validation method employing a per fold hyper-parameter optimization procedure, showing promising generalization ability. The MIL paradigm displayed an improvement up to 45.7 %, in terms of Balanced Accuracy, in comparison to the classical Machine Learning paradigm. Moreover, we investigate different compact molecular representations, based on atomic or bi-atomic substructures. Finally, we scanned the Drugbank for strongly active compounds and we present the top-15 ranked compounds.

Research was carried out to assess the yield and quality of fruits from the new Polish apple tree ‘Chopin’—a ‘green peel’, scab-resistant cultivar under grass living mulch management. Blue fescue and red fescue were tested in experiment no. 1. In experiment no. 2, meadow grass and perennial ryegrass were used. Every species of grass was sown in two doses of 50 and 150 kg per ha. Herbicide fallow was introduced as a control in both experiments. Strongly variable temperatures and precipitations in the years of evaluation made it possible to estimate year—a function of variable environmental conditions—as an additional experimental factor. An unexpected effect of the presence of living mulch was its significant impact on the appearance of blush on the ‘green peel’ fruit. However, living mulches had little effect on weight and fruit size. Cool days during apple ripening enhanced the process of fruit skin red coloration. The effect of both agrotechnical and environmental factors on fruit quality was more visible in the case of less vigorous trees, which were more susceptible to experimental, stressful conditions. However, increasing the sowing dose of each grass seed did not influence red blushing, weight, or fruit size. An additional difficulty for the trees was the competition caused by the early germination of these living grass mulches, reinforced by the presence of Trifolium repens L.

The long-term influence of several living mulches on apple tree growth, nutrient status, yield, and fruit quality concerning the year of cover crop sowing in tree rows was evaluated in the Lower Silesia region in the southwestern part of Poland. The estimation was conducted in six apple tree ‘Ligol’ populations differentiated by the rootstock: M.26, M.9, P 60, P 2, P 16, and P 22. In experiment no. 1, one-year-old nursery stock was planted, and perennial living mulches were sown: colonial bent grass (Agrostis vulgaris With.) and white clover (Trifolium repens L.) in the same year and blue fescue (Festuca ovina L.) in the following year. In experiment no. 2, which involved two-year-old stock, the sowing of blue fescue was delayed until the second, third, fourth, and fifth years following orchard planting. The presence of the cover crop significantly affected young tree growth when one-year-old trees were planted. The sowing of the living mulch in the first or the second year after tree planting led to a significant reduction in the cumulative yield obtained from the young trees. The living mulch improved the red coloration of the fruit skin in the young orchard, but it caused a reduction in the mean fruit weight and size. However, it also contributed to decreased nitrogen concentration in the leaves of the apple trees, and the tree tolerance to living mulch increased as the orchard aged. A choice of semidwarf rootstock, postponed sowing of cover crop, and high-quality nursery stock were recognized as the most important factors for fostering apple tree tolerance to living mulches in tree rows.

Study region: A network of 25 rain gauges roughly covering an area of 517.2 km2 of Warsaw, in Poland. Study focus: Scaling and intermittency of data series of rainfall intensity are investigated for each of the network rain gauges using five multifractal framework methods: spectral density analysis, functional box-counting, trace moment, probability distribution/multiple scaling, and double trace moment. Synthetic precipitation series are then generated by means of continuous universal random cascade models. Cascade generators are defined by universal parameters calculated for selected Warsaw rain gauges, or rain gauge clusters displaying similarity of multifractal parameters. The generated synthetic precipitation time series are subject to statistical evaluation by comparing the complementary cumulative distribution function and the intermittency calculated for synthetic versus observed data. Finally, a novel filtering algorithm is proposed to correct the intermittency characteristics of synthetic precipitation time series. New hydrological insights: Based on our analyses, the temporal structure of the recorded Warsaw precipitation time series is found to be a multifractal set characterized by a scaling behaviour over a wide range of scales. Furthermore, it has been observed that most Warsaw rain gauges, have a distinct similarity of multifractal properties. It has also been demonstrated that, for the first time in Poland, the universal continuous cascades could be used in practice for the generation of synthetic rainfall series at fine (1-min) temporal resolution

The European Commission Flood Risk Directive review shows that while many nations have embraced the concepts of flood risk management, there is still quite more to do in delineating risk–cost-effective measures and developing cost estimates and financing of those measures. Not mentioned are the necessary changes to existing design standards and protocols which will have to change in order to properly encompass climate change and variability, with associated uncertainties. Adjustments in engineering design standards and changes in hazards are examined, based on trend detection in observational records and projections for the future. Issues of urban and transport (motorways and railways) drainage design are also examined. Furthermore, risk reduction strategies are discussed. Finally, a way of accounting for non-stationarity in determining design precipitation and design floods is tackled. Climate change adjustments in engineering design standards, such as design precipitation and design floods, are reviewed via examples from Europe. HIGHLIGHTS Intense precipitation and high river flows have been changing and will change, even if trend detection may not yet consistently show a ubiquitous, and statistically significant, change.; European nations have embraced the concepts of flood risk management in line with the common Floods Directive, yet existing design standards have to change in order to properly encompass climate change and variability, with associated uncertainties.; It is necessary to prepare for the existing climate variability, but this is not likely to be sufficient for the future. Current water management practices may be inadequate to reduce the adverse impacts of climate change.; There is a clear gap between results of scientific studies and needs of practitioners in the domain of climate change adjustments in engineering design. Scientific results are publishable but not necessarily actionable. Both science and practice should try to improve the interface.;

The durability of four grass living mulches, population of annual and perennial weeds, and their biodiversity in tree rows and tractor alleys were evaluated in the first 4 years after cover crop sowing. The experiment was established in a young semi-dwarf apple orchard 1 year after the planting of the tree ‘Chopin’ cv. Soil coverage was satisfactory for the three cover crops: red fescue, Kentucky bluegrass, and perennial ryegrass. The sods of the grasses exhibited 100% soil surface cover in the tree rows and drive alleys, starting from 1 year after sowing to the end of the study period. Only the maintenance of the blue fescue resulted in low average soil development. Annual and perennial weed populations were lower in the tree rows than in the drive alleys. It was determined meticulous and manual soil preparation prior to all grass sowing under tree crowns, followed by precise mowing of grasses and nitrogen fertilization in subsequent years. The highest soil surface cover by the total annual taxa was observed immediately after the emergence of the living mulch. Total perennial weed populations increased in the following vegetation seasons. Trifolium repens L. and Taraxacum offcinale (L.) Web. dominated all the grass living mulches evaluated. Only the red fescue sod effectively limited the infestation of perennial weed after 4 years of living mulch maintenance in the tree row. More than half of the annual and perennial weed taxa occurred sporadically in all living mulches. In both the tree rows and tractor alleys, the soil surface cover was not more than 1%, and these species contributed to the increase in orchard biodiversity.

This study was conducted to estimate the yield, and to identify and quantify primary and secondary metabolites in fruit of Malus domestica Borkh. cv. ‘Sampion’ under two agrotechnical factors: the floor management (herbicide fallow and living mulch) and the dose of nitrogen (50, 80, 110, and 140 kg ha−1). Compared to herbicide fallow, living mulch did not decrease yield. Research showed a rich composition of phenolic and volatile organic compounds in apples, which varied with the evaluated factors, as well as with the weather conditions during the vegetation season. The precipitation deficit and high summer temperatures did not contribute to proper fruit growth and development and led to a higher content of phenolic compounds in the fruit flesh from trees in herbicide fallow compared to living mulch. Living mulch, which could be a factor regulating the availability of nitrogen to trees, stimulated the synthesis of anthocyanins, which was also potentiated by low average temperatures at harvest time, resulting in a large area of fruit skin red blush.

A highly frequented beach in Marseille, France, was monitored on an hourly basis during a summer day in July 2018, to determine possible water and sand fecal pollution, in parallel with influx of beach users from 8 a.m. to 8 p.m. Fecal indicator bacteria were enumerated, together with four host-associated fecal molecular markers selected to discriminate human, dog, horse, or gull/seagull origins of the contamination. The antimicrobial resistance of bacteria in water and sand was evaluated by quantifying (i) the class 1, 2, and 3 integron integrase genes intI, and (ii) blaTEM, blaCTX–M, and blaSHV genes encoding endemic beta-lactamase enzymes. The number of beach users entering and leaving per hour during the observation period was manually counted. Photographs of the beach and the bathing area were taken every hour and used to count the number of persons in the water and on the sand, using a photo-interpretation method. The number of beach users increased from early morning to a peak by mid-afternoon, totaling more than 1,800, a very large number of users for such a small beach (less than 1 ha). An increase in fecal contamination in the water corresponded to the increase in beach attendance and number of bathers, with maximum numbers observed in the mid-afternoon. The human-specific fecal molecular marker HF183 indicated the contamination was of human origin. In the water, the load of Intl2 and 3 genes was lower than Intl1 but these genes were detected only during peak attendance and highest fecal contamination. The dynamics of the genes encoding B-lactamases involved in B-lactams resistance notably was linked to beach attendance and human fecal contamination. Fecal indicator bacteria, integron integrase genes intI, and genes encoding B-lactamases were detected in the sand. This study shows that bathers and beach users can be significant contributors to contamination of seawater and beach sand with bacteria of fecal origin and with bacteria carrying integron-integrase genes and beta lactamase encoding genes. High influx of users to beaches is a significant factor to be considered in order to reduce contamination and manage public health risk.

In this study, conducted at the Research Station of the Wrocław University of Environmental and Life Sciences, Poland, we sought to determine the communities of weed species and their temporal occurrence in a young apple orchard mulched with spent mushroom compost or Miscanthus straw applied to tree rows. A herbicide fallow treatment was used as a control. During the first year of application, both organic mulches protected against the germination of weed seeds stored within the soil. In subsequent years, however, annual weeds occurred in the mulched tree rows, the most aggressive of which was Chenopodium album L. Annual increases in population densities were noted over the 4 subsequent years of research, particularly in the rows receiving spent mushroom compost. Perennial species in the family Poaceae and genus Malva were present in soil receiving both organic mulches. Trifolium repens L. was the perennial weed most often noted in the Miscanthus mulch, whereas Taraxacum officinale Web. was more characteristic of the spent mushroom compost. Commencing from the spring of the third year following apple tree planting, weed infestation associated with the spent mushroom compost was similar to that observed in the herbicide fallow, thus necessitating three annual applications of herbicide to further maintain the orchard. The insufficient weed suppression obtained with this mulch precludes its recommendation as an effective weed management system. Although Miscanthus straw provided extended tree row protection from weed infestation, herbicide intervention was also required. Notably, however, despite the fact that Miscanthus straw provided conditions more favorable to tree growth, apple tree yields and fruit quality tended to be similar under the three investigated orchard soil management systems.

The living mulch permanence along with the succession of their weed infestation in an apple orchard were evaluated at the Research Station, Wrocław University of Environmental and Life Sciences. The perennial cover crops: white clover and colonial bent grass, as well as the annual dwarf nasturtium, were sown as living mulches in apple tree rows, in the year of establishing the orchard. Blue fescue was sown one year later to replace the dwarf nasturtium. The percent of covers and temporal dominance dynamics of weeds were estimated during the first 13 years of the orchard maintenance. The occurrence of annual weeds, which had been abundant in all the living mulches in the year of their sowing, decreased in the following years of orchard maintenance. Conversely, the dominance of several perennial weed species increased as the orchard reached the full cropping period. White clover exhibited the lowest permanence. Dynamic spreading of Elymus repens (L.) Gould and other species from the Poaceae family was the direct cause of this cover crop disappearance. The presence of perennial dicotyledonous weeds, primarily Taraxacum officinale Web. and Convolvulus arvensis L., also contributed to the diminished sod of all the living mulches. Blue fescue maintained satisfactory dominance relative to colonial bent grass for nearly the entire first decade of the research. Nevertheless, both grass living mulches were present on less than half of the tree row soil surface area, in the thirteenth year after planting of the apple trees.

To investigate the capacities of persistence and dissemination of blaNDM-5 within Escherichia coli and in aquatic environment, we characterized E. coli (sequence type 636) strains B26 and B28 isolated one month apart from the same urban river in Montpellier, France. The two isolates carried a pTsB26 plasmid, which sized 45,495 Kb, harbored blaNDM-5 gene and belonged to IncX-3 incompatibility group. pTsB26 was conjugative in vitro at high frequency, it was highly stable after 400 generations and it exerted no fitness cost on its host. blaNDM-5harboring plasmids are widely dispersed in E. coli all around the world, with no lineage specialization. The genomic comparison between B26 and B28 stated that the two isolates probably originated from the same clone, suggesting the persistence of pTsB26 in an E. coli host in aquatic environment.

An accurate air-temperature prediction can provide the energy consumption and system load in advance, both of which are crucial in HVAC (heating, ventilation, air conditioning) system operation optimisation as a way of reducing energy losses, operating costs, as well as pollution and dust emissions while maintaining residents’ thermal comfort. This article presents the results of an outdoor air-temperature time-series prediction for a multifamily building with the use of artificial neural networks during the heating period (October–May). The aim of the research was to analyse in detail the created neural models with a view to select the best combination of predictors and the optimal number of neurons in a hidden layer. To meet that task, the Akaike information criterion was used. The most accurate results were obtained by MLP 3-3-1 (r = 0.986, AIC = 1300.098, SSE = 4467.109), with the ambient-air-temperature time series observed 1, 2, and 24 h before the prognostic temperature as predictors. The AIC proved to be a useful method for the optimum model selection in a machine-learning modelling. What is more, neural network models provide the most accurate prediction, when compared with LR and SVR. Additionally, the obtained temperature predictions were used in HVAC applications: entering-water temperature and indoor temperature modelling.

Carbapenems are β-lactams antimicrobials presenting a broad activity spectrum and are considered as last-resort antibiotic. Since the 2000s, carbapenemase producing Enterobacterales (CPE) have emerged and are been quickly globally spreading. The global dissemination of carbapenemase encoding genes (CEG) within clinical relevant bacteria is attributed in part to its location onto mobile genetic elements. During the last decade, carbapenemase producing bacteria have been isolated from non-human sources including the aquatic environment. Aquatic ecosystems are particularly impacted by anthropic activities, which conduce to a bidirectional exchange between aquatic environments and human beings and therefore the aquatic environment may constitute a hub for CPE and CEG. More recently, the isolation of autochtonous aquatic bacteria carrying acquired CEG have been reported and suggest that CEG exchange by horizontal gene transfer occurred between allochtonous and autochtonous bacteria. Hence, aquatic environment plays a central role in persistence, dissemination and emergence of CEG both within environmental ecosystem and human beings, and deserves to be studied with particular attention.

Understanding the variability of precipitation at small scales is fundamental in urban hydrology. Here we consider the case study of Warsaw, Poland, characterized by a precipitation-monitoring network of 25 gauges and microcanonical cascade models as the instrument of investigation. We address the following issues partially investigated in literature: (1) the calibration of microcanonical cascade model generators in conditions of short time series (i.e., 2.5–5 years), (2) the identification of the probability distribution of breakdown coefficients (BDCs) through ranking criteria and (3) the variability among the gauges of the monitoring network of the empirical distribution of BDCs. In particular, (1) we introduce an overlapping moving window algorithm to determine the histogram of BDCs and compare it with the classic non-overlapping moving window algorithm; (2) we compare the 2N–B distribution, a mixed distribution composed of two normal (N) and one beta (B), with the classic B distribution to represent the BDCs using the Akaike information criterion; and (3) we use the cluster analysis to identify patterns of BDC histograms among gauges and timescales. The scarce representation of the BDCs at large timescales, due to the short period of observation (~ 2.5 years), is solved through the overlapping moving window algorithm. BDC histograms are described by a 2N–B distribution. A clear evolution of this distribution is observed, in all gauges, from 2N–B for small timescales, N–B for intermediate timescales and B distribution for large timescales. The performance of the microcanonical cascades is evaluated for the considered gauges. Synthetic time series are analyzed with respect to the intermittency and the variability of intensity and compared to observed series. BDC histograms for each timescale are compared with the 25 gauges in Warsaw and with other gauges located in Poland and Germany.
