13. Interfacial engineering of suppressing Li dendrite growth in all solid-state Li-metal batteries.

Abstract

Solid-state electrolytes, as the key component of all solid-state Li metal batteries, address thermal runaway and safety issues of current Li-ion batteries but still face the challenge of Li dendrites. Experimental and theoretical studies demonstrated that Li dendrites grow along the interior cracks of SSEs, in which the conduction of electrons within solid-state electrolytes plays a pivotal role. In this review, the effects of cracks and electron conduction on Li dendrite growth were revealed through advanced characterization techniques and theoretical calculations and the strategies for suppressing Li dendrite growth are summarized, focusing on blocking electron conduction and suppressing the formation of cracks within solid-state electrolytes. This work presents a systematic review of recent progress in Li dendrite growth and provides a deep insight into its causes and solutions, which are crucial for the application of solid-state electrolytes. Abstract

The limited infiltration and persistence of chimeric antigen receptor (CAR)-T cells is primarily responsible for their treatment deficits in solid tumors. Here, we present a three-dimensional scaffold, inspired by the physiological process of T-cell proliferation in lymph nodes. This scaffold gathers the function of loading, delivery, activation and expansion for CAR-T cells to enhance their therapeutic effects on solid tumors. This porous device is made from poly(lactic-co-glycolic acid) by a microfluidic technique with the modification of T-cell stimulatory signals, including anti-CD3, anti-CD28 antibodies, as well as cytokines. This scaffold fosters a 50-fold CAR-T cell expansion in vitro and a 15-fold cell expansion in vivo. Particularly, it maintains long-lasting expansion of CAR-T cells for up to 30 days in a cervical tumor model and significantly inhibits the tumor growth. Abstract

In situtumor vaccination has aroused tremendous interest with its capability for eliciting strong and systemic antitumor immune responses. Unlike traditional cancer vaccines, in situtumor vaccination avoids the laborious process of tumor antigen identification and can modulate tumor immunosuppressive microenvironment at the same time. In recent years, bacteria have been used as both efficient tumor-targeted delivery vehicles and potent adjuvants. Regarding the rapid development in this area, in this review, we summarize recent advances in the application of bacteria for in situcancer vaccination. We illustrate the mechanisms of bacteria as both efficient tumor immunogenic cell death inducers and tumor-targeted delivery platforms. Then we comprehensively review the engineering strategies for designing bacteria-based in situvaccination, including chemical modification, nanotechnology, and genetic engineering. The current dilemma and future directions are discussed at the end of this review.

40. Impact of climate change on wheat security through an alternate host of stripe rust.

Abstract

In the 21st century, stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is still the most devastating disease of wheat globally. Despite the critical roles of the alternate host plants, the Berberis species, in the sexual reproduction and spread of Pst, the climate change impacts on the redistribution of Berberis plants, and their potential risk of incurring wheat stripe rust are scarcely discussed in the literature. This study evaluated the potential risk of Pst in wheat cultivation in Sichuan Province, China, for the first time, by analyzing the dynamics of Berberis plants and the change in Pst oversummering range under climate change scenarios. Ensembles of small models (ESMs) in combination with four algorithms were used to project the future (i.e., 2041–2060, 2061–2080, and 2081–2100) distributions of 54 Berberis species in Sichuan Province based on two shared socioeconomic pathways (SSP) scenarios. The species richness of Berberis was predicted to increase in northwestern Sichuan but decreased in southern and eastern Sichuan under both SSP245 and SSP585 scenarios. The mean temperature of the driest quarter was found to be the determinant of the distributional shifts for the majority of Berberis plants. Our overlapping assessments indicated that the risk of wheat stripe rust would be reduced in southwestern Sichuan. In contrast, it would be aggravated in the northwest due to the predicted shifts of Berberis plants northwards and stronger shrinkage of Pst oversummering range occurring in the south than in the north. In the "hotspots" of the stripe rust pathogen, risk assessment in advance could provide a scientific basis for decision‐makers and local communities in effectively controlling wheat stripe rust, thus further sustaining food security through adapting to future climate change.
