13. Liver enzyme and risk of vascular dementia: A univariable and multivariable Mendelian randomization of European descent

Abstract

Observational studies have indicated a link between liver enzymes and dementia, but the causal relationship remains uncertain. We conducted a two‐sample Mendelian randomization (MR) study to investigate potential causal links between liver function markers (alanine aminotransferase [ALT], aspartate aminotransferase [AST], alkaline phosphatase [ALP], and γ‐glutamyltransferase [GGT]) and various forms of dementia (all‐cause dementia, Alzheimer's disease [AD], vascular dementia [VaD], and frontotemporal dementia [FTD]). Genome‐wide association study (GWAS) data of liver enzyme levels with 517 single nucleotide polymorphisms from 315,572 individuals of European descent were considered as exposures. Additional GWAS data on dementia from the FinnGen consortium and the UK Biobank were used as outcomes. The causal relationship was evaluated using univariable MR (UVMR) and multivariable MR (MVMR) methods. UVMR approaches such as inverse variance weighting (IVW), MR‐Egger, weighted median, simple mode, and weighted mode were used, with IVW as primary. MVMR techniques, such as extended versions of IVW, MR‐Egger, and Q‐minimization methods, were used to assess causal effects. The robustness of the MR analysis findings was verified through heterogeneity, horizontal pleiotropy, and leave‐one‐out analyses. MVMR analysis demonstrated that a genetically determined one standard deviation rise in blood GGT levels was associated with an increased risk of VaD (IVW: odds ratio = 1.007, 95% confidence interval = 1.002–1.011, p= 0.010). These findings remained consistent after adjusting for confounding variables in MVMR analysis. Sensitivity analyses further supported the causal relationship. However, no significant links were observed between ALT, AST, ALP, and all‐cause dementia, VaD, AD, or FTD. Our study suggests clinical implications, demonstrating that high blood GGT concentrations are potential causal risk factors for VaD in European populations. Further research is needed to uncover the underlying biological mechanisms between GGT and VaD and validate the clinical relevance of early prevention and intervention strategies. 15. Adipose Kiss1 controls aerobic exercise‐related adaptive responses in adipose tissue energy homeostasis.

Abstract

Kisspeptin signaling regulates energy homeostasis. Adiposity is the principal source and receiver of peripheral Kisspeptin, and adipose Kiss1 metastasis suppressor (Kiss1) gene expression is stimulated by exercise. However, whether the adipose Kiss1 gene regulates energy homeostasis and plays a role in adaptive alterations during prolonged exercise remains unknown. Here, we investigated the role of Kiss1 role in mice and adipose tissues and the adaptive changes it induces after exercise, using adipose‐specific Kiss1 knockout (Kiss1adipoq−/−) and adeno‐associated virus‐induced adipose tissue Kiss1‐overexpressing (Kiss1adipoq over) mice. We found that adipose‐derived kisspeptin signal regulates lipid and glucose homeostasis to maintain systemic energy homeostasis, but in a sex‐dependent manner, with more pronounced metabolic changes in female mice. Kiss1 regulated adaptive alterations of genes and proteins in tricarboxylic acid (TCA) cycle and oxidative phosphorylation (OxPhos) pathways in female gWAT following prolonged aerobic exercise. We could further show that adipose Kiss1 deficiency leads to reduced peroxisome proliferator‐activated receptor gamma co‐activator 1 alpha (PGC‐1α) protein content of soleus muscle and maximum oxygen uptake (VO2 max) of female mice after prolonged exercise. Therefore, adipose Kisspeptin may be a novel adipokine that increases organ sensitivity to glucose, lipids, and oxygen following exercise. [ABSTRACT FROM AUTHOR]

Abstract

The void phenomenon behind the tunnel lining has become the main cause of tunnel disease, easily triggering tunnel lining structure damage, shedding, water seepage, and other diseases that seriously threaten the normal operation of the tunnel. The void area behind the lining seriously reduces the seismic capacity of the tunnel. In this paper, the mechanical parameters of the surrounding rock are calculated by optimizing the inversion of the improved system identification sensitivity analysis method, and the numerical model of the tunnel's dynamic response is optimized using the improved inversion results to study the repair effect and seismic capacity of the Longmenshan tunnel using polymer grouting to repair the void area behind the lining under the action of a seismic load. The results show that the displacement and stress at the top of the tunnel secondary lining void area are significantly reduced and close to normal after the repair of polymer grouting; however, the stress and displacement at the top of the secondary lining are significantly different under the action of seismic waves at the same peak at different site conditions, which indicates that the dynamic response of this tunnel model has obvious sensitivity to the seismic wave spectrum.
