To enhance the electrical safe operation area (eSOA) of laterally diffused metal oxide semiconductor (LDMOS) transistors, a novel reduced surface electric field (Resurf) structure in the n-drift region is proposed, which was fabricated by ion implantation at the surface of the LDMOS drift region and by drift region dimension optimization. Technology computer-aided design (TCAD) simulations show that the optimal value of Resurf ion implantation dose 1 × 1012 cm−2 can reduce the surface electric field in the n-drift region effectively, thereby improving the ON-state breakdown voltage of the device (BVon). In addition, the extended n-drift region length of the Ld design also improves device BVon significantly, and is aimed at reducing the current density and the electric field, and eventually suppressing the n-drift region impact ionization. The results show that the novel 60 V nLDMOS has a competitive BVon performance of 106.9 V, which is about 20% higher than that of the conventional one. Meanwhile, the OFF-state breakdown voltage of the device (BVoff) of 88.4 V and the specific ON-resistance (RON,sp) of 129.7 mΩ⋅mm2 exhibit only a slight sacrifice.

Abstract Identifying epitopes that T cells respond to is critical for understanding T cell-mediated immunity. Traditional multimer and other single cell assays often require large blood volumes and/or expensive HLA-specific reagents and provide limited phenotypic and functional information. Here, we present the Rapid TCR:Epitope Ranker (RAPTER) assay, a single cell RNA sequencing (scRNA-SEQ) method that uses primary human T cells and antigen presenting cells (APCs) to assess functional T cell reactivity. Using hash-tag oligonucleotide (HTO) coding and T cell activation-induced markers (AIM), RAPTER defines paired epitope specificity and TCR sequence and can include RNA- and protein-level T cell phenotype information. We demonstrate that RAPTER identified specific reactivities to viral and tumor antigens at sensitivities as low as 0.15% of total CD8+ T cells, and deconvoluted low-frequency circulating HPV16-specific T cell clones from a cervical cancer patient. The specificities of TCRs identified by RAPTER for MART1, EBV, and influenza epitopes were functionally confirmed in vitro. In summary, RAPTER identifies low-frequency T cell reactivities using primary cells from low blood volumes, and the resulting paired TCR:ligand information can directly enable immunogenic antigen selection from limited patient samples for vaccine epitope inclusion, antigen-specific TCR tracking, and TCR cloning for further therapeutic development.

Background: Accurate and convenient serological markers for prognosis after traumatic brain injury (TBI) are still lacking. We aimed to explore the predictive value of serum calcium for prognosing outcomes within 6 months after TBI. Methods: In this multicenter retrospective study, 1255 and 719 patients were included in development and validation cohorts, respectively, and their 6-month prognoses were recorded. Serum calcium was measured through routine blood tests within 24 h of hospital admission. Two multivariate predictive models with or without serum calcium for prognosis were developed. Receiver operating characteristics and calibration curves were applied to estimate their performance. Results: The patients with lower serum calcium levels had a higher frequency of unfavorable 6-month prognosis than those without. Lower serum calcium level at admission was associated with an unfavorable 6-month prognosis in a wide spectrum of patients with TBI. Lower serum calcium level and our prognostic model including calcium performed well in predicting the 6-month unfavorable outcome. The calcium nomogram maintained excellent performance in discrimination and calibration in the external validation cohort. Conclusions: Lower serum calcium level upon admission is an independent risk factor for an unfavorable 6-month prognosis after TBI. Integrating serum calcium into a multivariate predictive model improves the performance for predicting 6-month unfavorable outcomes.

Grain refinement occurs during manufacturing process, and grain size has a significant impact on mechanical properties. It is essential to establish the relationship between grain evolution and plastic behavior. The quasi-static and dynamic compression experiments for three kinds of oxygen-free high-conductivity (OFHC) copper with different grain sizes have been carried out. The true stress–strain curves were corrected to the isothermal curves, in which the temperature dependence of the specific heat capacity was used. It was found that the grain size has a significant influence on the yield stress, strain rate sensitivity, and thermal softening. A new phenomenon-based constitutive model considering the grain size and the coupling between strain hardening and strain rate was developed, and the results of the quasi-static and dynamic compression experiments were used to fit the parameters. The experimental data and the predicted results of the newly developed constitutive model agree well, which proves the constitutive model has a high prediction accuracy. The fitted constitutive model depicted that the strain rate sensitivity increases and thermal softening degree declines as the rise of the grain size of OFHC copper. The developed constitutive model could be used to the numerical simulation for the manufacturing process of OFHC copper considering the grain refinement.

Abstract The synthetic lethal association between BRCA deficiency and poly (ADP-ribose) polymerase (PARP) inhibition supports PARP inhibitor (PARPi) clinical efficacy in BRCA-mutated tumors. PARPis also demonstrate activity in non-BRCA mutated tumors presumably through induction of PARP1-DNA trapping. Despite pronounced clinical response, therapeutic resistance to PARPis inevitably develops. An abundance of knowledge has been built around resistance mechanisms in BRCA-mutated tumors, however, parallel understanding in non-BRCA mutated settings remains insufficient. In this study, we find a strong correlation between the epithelial-mesenchymal transition (EMT) signature and resistance to a clinical PARPi, Talazoparib, in non-BRCA mutated tumor cells. Genetic profiling demonstrates that SNAI2, a master EMT transcription factor, is transcriptionally induced by Talazoparib treatment or PARP1 depletion and this induction is partially responsible for the emerging resistance. Mechanistically, we find that the PARP1 protein directly binds to SNAI2 gene promoter and suppresses its transcription. Talazoparib treatment or PARP1 depletion lifts PARP1-mediated suppression and increases chromatin accessibility around SNAI2 promoters, thus driving SNAI2 transcription and drug resistance. We also find that depletion of the chromatin remodeler CHD1L suppresses SNAI2 expression and reverts acquired resistance to Talazoparib. The PARP1/CHD1L/SNAI2 transcription axis might be therapeutically targeted to re-sensitize Talazoparib in non-BRCA mutated tumors.

Abstract Objective The neutrophil to lymphocyte ratio (NLR) has been proposed to capture the inflammatory status of patients with various conditions involving the brain. This retrospective study aimed to explore the association between the NLR and the early growth of traumatic intracerebral haemorrhage (tICH) in patients with traumatic brain injury (TBI). Methods A multicentre, observational cohort study was conducted. Patients with cerebral contusion undergoing baseline computed tomography for haematoma volume analysis within 6 h after primary injury and follow‐up visits within 48 h were included. Routine blood tests were performed upon admission, and early growth of tICH was assessed. Prediction accuracies of the NLR for the early growth of tICH and subsequent surgical intervention in patients were analysed. Results There were a total of 1077 patients who met the criteria included in the study cohort. Univariate analysis results showed that multiple risk factors were associated with the early growth of tICH and included in the following multivariate analysis models. In comparison with single crystal scintillators, glass scintillators are more promising materials for their benefits of easy preparation, low cost, controllable size, and large-scale manufacture. The emission of Eu3+ ion at 612 nm matches well with the photoelectric detector, making it suitable for the activator in glass scintillators. Therefore, the research on Eu3+ doped glass scintillators attract our attention. The photoluminescent and scintillating properties of Eu3+-activated boroaluminosilicate glass scintillators prepared by the conventional melt-quenching method were investigated in this work. The glass samples present good internal quantum yield. Under X-ray radiation, the optimal sample reveals high X-ray excited luminesce (XEL), and its integrated intensity of XEL is 22.7% of that of commercial crystal scintillator Bi4Ge3O12. Furthermore, the optimal specimen possesses a spatial resolution of 14 lp/mm in X-ray imaging. These results suggest that Eu3+-doped boroaluminosilicate glass is expected to be applied in X-ray imaging.

The aged seeds have a significant influence on seed vigor and corn growth. Therefore, it is vital for the planting industry to identify aged seeds. In this study, hyperspectral reflectance imaging (1,000–2,000 nm) was employed for identifying aged maize seeds using seeds harvested in different years. The average spectra of the embryo side, endosperm side, and both sides were extracted. The support vector machine (SVM) algorithm was used to develop classification models based on full spectra to evaluate the potential of hyperspectral imaging for maize seed detection and using the principal component analysis (PCA) and ANOVA to reduce data dimensionality and extract feature wavelengths. The classification models achieved perfect performance using full spectra with an accuracy of 100% for the prediction set. The performance of models established with the first three principal components was similar to full spectrum models, but that of PCA loading models was worse. Compared to other spectra, the two-band ratio (1,987 nm/1,079 nm) selected by ANOVA from embryo-side spectra achieved a better classification accuracy of 95% for the prediction set. The image texture features, including histogram statistics (HS) and gray-level co-occurrence matrix (GLCM), were extracted from the two-band ratio image to establish fusion models. The results demonstrated that the two-band ratio selected from embryo-side spectra combined with image texture features achieved the classification of maize seeds harvested in different years with an accuracy of 97.5% for the prediction set. The overall results indicated that combining the two wavelengths with image texture features could detect aged maize seeds effectively. The proposed method was conducive to the development of multi-spectral detection equipment.

To improve the quality of products produced from microforming, various nanolubricants have been applied in the field of micromanufacturing in recent years. In this paper, the effects of glycerol-based lubricant containing TiO2 NPs (NPs) on micro deep drawing (MDD) of austenitic stainless steel (ASS) SUS301 were studied, and the lubrication mechanism involved was discussed. The MDD experiments were conducted with the SUS301 foils under dry, 1, 2, and 4 wt% TiO2 NP lubrication conditions. The results show that the use of the TiO2 nanolubricants can significantly improve the quality of the drawn cups in terms of decreased wrinkling and surface roughness. Besides, the concentration of TiO2 NPs influences lubricity by reducing friction during the MDD process. The peak drawing force is the lowest when 2 wt% nanolubricant is applied, which drops to 72.54 N from 77.38 N under dry conditions. The micro cup drawn under 2 wt% TiO2 nanolubricant has the best quality among those obtained under all the lubrication conditions. The lubrication mechanisms are derived from the mending effects of TiO2 NPs and the formation of thin lubricant films associated with the open lubricant pockets (OLPs) and close lubricant pocket (CLPs) theory in the MDD. The CLPs function as reservoirs that retain lubricants to counteract the load pressure, whereas the OLPs lead to lubricant leakage due to the higher flow resistance. It was found that the lubricant film and NPs are insufficient at a low concentration (1 wt%), while the lubrication performance can be enhanced with increased NP concentration. However, there exist apparent agglomerations on the surface of the produced micro cup when using 4 wt% nanolubricant, which greatly deteriorates the lubricant performance in the MDD process. It is concluded that the lubricant containing 2 wt% TiO2 NPs demonstrates the best lubrication performance during the MDD of ASS SUS301.

Background. Drug-eluting stent (DES) plus drug-coated balloon (DCB) is a safe and effective treatment strategy for coronary artery bifurcation lesions, but there is no report about this strategy being used for left main (LM) bifurcation lesions. We aim to explore the efficacy and safety of DES plus DCB in the treatment of LM bifurcation lesions. Methods. A total of 100 patients diagnosed with LM bifurcation lesions by coronary angiography were retrospectively enrolled at our center from January 2018 to December 2019. They received either a two-stent strategy or a main branch (MB) stenting plus side branch (SB) DCB strategy and were accordingly divided into the 2-DES group and the DES + DCB group. Patients treated with DES + DCB were compared with a cohort of matched patients treated with a 2-DES strategy. Clinical data was collected and quantitative coronary analysis was performed. Results. For immediate postoperative angiography, though the two groups had no differences in the minimal luminal diameter (MLD) and luminal stenosis of MB, the DES + DCB group had significantly lower SB ostial MLD and a higher degree of residual lumen stenosis than the 2-DES group (P

Aortic dissection (AD) is a cardiovascular disease characterized by high mortality and poor prognosis. Although FBN1 is associated with syndromic AD, its association with non-syndromic AD remains unclear. In this study, DNA samples from 90 Chinese individuals with non-syndromic AD (60 Stanford A, 30 Stanford B types) were analyzed to determine the relationship between diverse genotypes of the FBN1 gene and non-syndromic AD. Eleven pathogenic/likely pathogenic variants (1 novel) were identified in 12.2% of patients with non-syndromic AD. Patients with positive variants suffered from AD at a younger age than those in the negative variant group. Among the six positive missense mutations associated with cysteine residue hosts, four (66.7%) were Stanford A AD, whereas two (33.3%) were Stanford B AD. Three (100%) positive splicing/truncation variant hosts were Stanford A AD. The splicing/truncation variants and missense variants involving cysteine residues in the FBN1 gene increased the risk of Stanford A AD. Ten common SNPs that increased susceptibility to AD were identified. In particular, five SNPs were detected significantly in Stanford A AD, whereas another four SNPs were significantly detected in Stanford B AD. These significant variants can function as biomarkers for the identification of patients at risk for AD. Our findings have the potential to broaden the database of positive mutations and common SNPs of FBN1 in non-syndromic AD among the Chinese population.

Rubber materials are widely used in aerospace, automotive, smart devices and artificial skin. It is significant to address the aging susceptibility of conventional vulcanized rubber and to impart it rapid self-healing performance for destructive crack damage. Herein, a novel supramolecular rubber elastomer is prepared by introducing metal coordination between carboxyl-terminated polybutadiene and polystyrene-vinylpyridine copolymer. Based on the metal coordination interaction, the elastomer exhibits shape memory and self-healing properties. Moreover, a rapid closure-repair process of destructive cracks is achieved by presetting temporary shapes. This shape memory-assisted self-repair model is shown to be an effective means for rapid repair of severe cracks. An approach to enhance the mechanical and self-healing properties of elastomer was demonstrated by adding appropriate amounts of oxidized carbon nano-onions (O-CNO) into the system. The tensile strength of the elastomer with an O-CNOs content of 0.5 wt% was restored to 83 ± 10% of the original sample after being repaired at 85 °C for 6 h. This study confirms that metal coordination interaction is an effective method for designing shape memory self-healing rubber elastomer. The shape memory-assisted self-healing effect provides a reference for the rapid self-repairing of severe cracks.

At present, the apple grading system usually conveys apples by a belt or rollers. This usually leads to low hardness or expensive fruits being bruised, resulting in economic losses. In order to realize real-time detection and classification of high-quality apples, separate fruit trays were designed to convey apples and used to prevent apples from being bruised during image acquisition. A semantic segmentation method based on the BiSeNet V2 deep learning network was proposed to segment the defective parts of defective apples. BiSeNet V2 for apple defect detection obtained a slightly better result in MPA with a value of 99.66%, which was 0.14 and 0.19 percentage points higher than DAnet and Unet, respectively. A model pruning method was used to optimize the structure of the YOLO V4 network. The detection accuracy of defect regions in apple images was further improved by the pruned YOLO V4 network. Then, a surface mapping method between the defect area in apple images and the actual defect area was proposed to accurately calculate the defect area. Finally, apples on separate fruit trays were sorted according to the number and area of defects in the apple images. The experimental results showed that the average accuracy of apple classification was 92.42%, and the F1 score was 94.31. In commercial separate fruit tray grading and sorting machines, it has great application potential.

The construction of heavy polar icebreakers is usually done with special hull steels, which require comprehensive properties such as good low-temperature toughness, high strength, and superior fatigue resistance. Reasonable and satisfactory heat treatments should be investigated and applied to acquire the required high strength and superior low-temperature toughness, since this is deemed an effective approach to ameliorate the combined properties of high-strength hull steels. Regarding this, the present study specifically explores the effects of different laboratory-based quenching (850 to 930 °C) and tempering (580 to 660 °C) heat treatments on the final low-temperature toughness of the high-strength hull steels. The low-temperature toughness is eventually improved without significantly sacrificing the strength. The results show that a favourable combination of properties can be obtained in the specimens under 900 °C quenching and 660 °C tempering processes. Additionality, the specimens tempered at 620 °C present the highest hardness, owing to the higher percentage of tempered martensite. Detailed mechanisms of the enhanced properties of the typical specimens subjected to the corresponding quenching and tempering processing are analysed and explicated.

The hydrocarbon generation model and hydrocarbon potential are investigated in the Lishui Sag, based on gold-tube pyrolysis experiments of deeply buried type Ⅲ kerogen. From this, we discuss the classification of kerogen types of source rocks with mixed organic matter sources. The hydrocarbon generated from the source rocks of the Lingfeng Formation and Yueguifeng Formation is dominated by natural gases with little oil in the West subsag, and the hydrocarbon generation model of the Lingfeng Formation is similar to that of Yueguifeng Formation, but the gas potential of Lingfeng Formation is higher than that of Yueguifeng Formation. The hydrocarbon potential of the Yueguifeng Formation in the East subsag is much higher than the West subsag, and it has considerable oil potential. Macerals diversity of source rocks is responsible for the difference of hydrocarbon generation characteristics for type Ⅲ kerogen in the Lishui Sag. It is not rigorous to evaluate the hydrocarbon potential of kerogen only based on pyrolysis parameters. Application of kerogen type index (KTI) can improve the accuracy of the classification of kerogen types with mixed organic matter sources. According to the classical kerogen classification template, the selected samples belong to type III kerogen. In this article, the selected samples were further subdivided into type III and type II/III based on the KTI value. Type III kerogen (0.5 ≤ KTI < 1.5) mainly produces gas, and type II/III kerogen (1.5 ≤ KTI < 5) mainly produces gas, but its oil potential is higher than that of type III.

In order to comprehensively analyze the risk factors and accurately find the high risk factors related to accidents, an analysis model of risk factors of urban bus operation is proposed, in which the advantages of the structural analysis of the Fault Tree Analysis (FTA) and the correlation analysis of the Cumulative Logistic Regression (CLR) are combined. Firstly, based on the accident data in Northeast China, FTA is used to compile the urban bus operation fault tree. In the fault tree, 16 bus operation risk factors are classified, while the risk factors are sorted and compared from three aspects: structural importance, probability importance, and critical importance. And then, the 11 higher risk factors are selected according to the discriminant principle. Secondly, bus operation accidents are divided into fatal accidents, injury accidents, and major economic loss accidents. The CLR model is used to fit the much higher risk factors that lead to urban bus operation accidents from above 11 higher risk factors. Finally, the scientific rationality and applicability of the model are verified, through the goodness of fit test and the comparison test with the actual probability of occurrence.

Purpose. To explore the potential of monocyte-to-lymphocyte ratio (MLR) at hospital admission for predicting acute traumatic intraparenchymal hematoma (tICH) expansion in patients with cerebral contusion. Patients and Methods. This multicenter, observational study included patients with available at-hospital admission (baseline) and follow-up computed tomography for volumetric analysis (retrospective development cohort: 1146 patients; prospective validation cohort: 207 patients). Semiautomated software assessed tICH expansion (defined as ≥33% or 5 mL absolute growth). MLR was acquired from routine blood tests upon admission. We constructed two predictive models: basic combined model of clinical and imaging variables and MLR combined model of both MLR and other variables in the basic model. Receiver operating characteristic (ROC) analysis and decision curve analysis (DCA) were used to estimate the performance of MLR for predicting acute tICH expansion. Results. Abstract This paper is concerned with the issue of robust stability for quaternion-valued neural networks (QVNNs) with leakage, discrete and distributed delays by employing a linear matrix inequality (LMI) approach. Based on the homeomorphic mapping theorem, the quaternion matrix theorem and the Lyapunov theorem, some criteria are developed in the form of real-valued LMIs for guaranteeing the existence, uniqueness, and global robust stability of the equilibrium point of the delayed QVNNs. Two numerical examples are provided to demonstrate the effectiveness of the obtained results.

Based on the ABAQUS/Explicit code, A 3D elastic–plastic and coupled thermo-mechanical FE model of radial ring rolling of AZ31 Magnesium alloy has been proposed to analyze the influence of rotational speed of driver roll to study the inhomogeneity distribution of strain and temperature, fishtail coefficient, rolling force parameters. The results show that: (1) when the rotational speed of driver roll n increases, the strain distribution of the rolled ring becomes less homogeneous, and the temperature distribution more homogeneous yet, and leading to an optimal n value; (2) the fishtail coefficient firstly decreases, then increases with the increase of n; (3) the rolling force, contact area and rolling moment gradually descend with the increase of n.
