Anemoside B4 (AB4) is reported to prevent acute colitis when given via intraperitoneal injection by two recent studies. However, whether oral AB4 protects against chronic colitis which resembles the clinical phenotype of ulcerative colitis (UC) and its mechanism of action are largely unknown.To systemically investigate the effects of oral AB4 against chronic colitis and illustrate the underlying mechanism of action.The preventive, therapeutic, and dose-dependent effects of AB4 against UC were examined in mice with acute or chronic relapsing colitis induced by dextran sulfate sodium (DSS). The inflammatory responses, colonic transcriptome, and 16S rDNA sequencing of the intestinal content of mice were analyzed.Oral administration of AB4 alleviated disease severity and colon shortening in mice with chronic relapsing colitis in a dose-dependent manner. The effects of AB4 were comparable to those of two positive-control compounds: tofacitinib and berberine. Unlike tofacitinib, AB4 did not have a deleterious effect on DSS-induced splenic swelling and anemia. Furthermore, AB4 inhibited the inflammatory responses of colitis, as evidenced by in-vivo, ex-vivo, and in-vitro studies. Transcriptomics revealed that AB4 treatment reversed the DSS-mediated decrease in the expression of colonic Pelo, B3gat2 and Mir8010. In addition, AB4 reversed DSS-induced alterations in the intestinal microbiome in mice. Through fecal microbiota transplantation, we proved that AB4 partially exerted its anti-colitis effects by modulating the gut microbiota.We demonstrated for the first time that AB4 has dose-dependent therapeutic effects against chronic relapsing colitis by modulating the inflammatory response, colonic gene expression, and intestinal microbiota.

Anemoside B4 (AB4) is a representative component of Pulsatilla decoction that is used in traditional Chinese medicine for treating inflammatory conditions. It is not known whether AB4 has beneficial effects on multiple sclerosis (MS).In the present study, we examined the preventative and therapeutic effects of AB4, and the possible mechanism by which it protects female mice against experimental autoimmune encephalomyelitis (EAE).Preventative treatment with AB4 (given orally at 100 and 200 mg/kg for 18 days) reduced the clinical severity of EAE significantly (from 3.6 ± 1.3 to 1.8 ± 1.5 and 1.6 ± 0.6, respectively), and inhibited demyelination and inflammatory infiltration of the spinal cord. In the therapeutic protocol, oral administration of 200 mg/kg AB4 for 21 days after initiation of EAE significantly alleviated disease severity (from 2.6 ± 1.3 to 0.9 ± 0.6) and was as effective as the clinically used drug fingolimod (0.3 ± 0.6). Furthermore, both doses of AB4 significantly inhibited mRNA expression of TNF-α, IL-6, and IL-17, and STAT3 activation, in the spinal cord; and the ex vivo and iv vitro AB4 treatment markedly inhibited secretion of the three cytokines from lymphocytes of EAE mice upon in vitro restimulation. In addition, AB4 reversed the changes in the composition of the intestinal microbiome observed in EAE mice.We reveal for the first time that AB4 protects against EAE by modulating inflammatory responses and the gut microbiota, demonstrating that AB4 may have potential as a therapeutic agent for treating MS in humans.

In this research, the effects of main parameters such as speed of agitator, concentration and redium change ratio on the grinding effect were studied by the method of wet-grinding. The powder was characterized and analyzed by means of particle size analysis and scanning electron microscope. The results show that the mechanical grinding method can effectively prepare micro-nanosilicon carbide powder with particle size of about 130nm, the optimum parameter is the rotational speed 1000r/min, the concentration is 30%, the medium change ratio is 75%.

Pulsatilla Decoction (Bai-Tou-Weng-Tang) has been used medically in China for thousands of years for the treatment of diseases caused by bacteria. In recent decades, Pulsatilla Decoction is becoming a well-known formula prescription used for the treatment of ulcerative colitis in traditional Chinese medicine. Pulsatilla chinensis is the chief herbal source of Pulsatilla Decoction, and it is rich in triterpenoid saponins, such as anemoside B4, anemoside A3, and 23-hydroxybetulinic acid. Anemoside B4 is the most abundant of that group and has been used as a quality control marker for Pulsatilla chinensis. As the major active component of Pulsatilla chinensis, anemoside B4 has also received attention as a pure compound for its therapeutic potential. In this review, we systematically analyze the findings on triterpenoid saponins, especially anemoside B4, anemoside A3 and 23-hydroxybetulinic acid, included in Pulsatilla chinensis and Pulsatilla Decoction. We discuss the pharmacokinetics and tissue distribution of these triterpenoid saponins as well as their biological activities. We also summarize the pharmacological effects of anemoside B4 and its two possible metabolites, anemoside A3 and 23-hydroxybetulinic acid, as pure compounds. In summary, this review sketches a profile of the state of existing knowledge with regard to the pharmacological effects of anemoside B4, especially its anti-inflammatory and immunomodulatory effects. These findings point to the possibility that anemoside B4 has potential to be studied further as a natural compound-originated immunomodulatory agent for the treatment of inflammatory diseases such as ulcerative colitis and thus, may represent one of the most important active components of Pulsatilla Decoction responsible for its anti-ulcerative colitis efficacy.

Using high quality carbon raw material,high quality silica raw material , the β-SiC powder are prepared by high-temperature vacuum smelting method. The effects of different materials and electrical power to the product of β-SiC quality has been studied. The microstructure and phase analysis of β-SiC powder samples were observed by Scanning Electron Microscopy and X-ray diffraction. The result shows that the raw materials are better, the product quality are better. In addition, the result also indicates that the β-SiC powder have better quality when the power are 3000W.

Anthracite and bitumite were processed respectively at 1400°C,1700°C, 2000°C, 2200°C, 2400°C and 2600°C,and their chemical composition,resistivity,microstructure, phase composition,and the internal migration of molecular functional group were tested and characterized. The results indicate that moisture, ash and volatile in coal have gradually shifted and lost with the elevation of heat treatment temperature, while the higher temperature, the quicker and completer phase change. Heat treatment can make the coal transform from approximately insulative phase to conductive phase,. Furthermore, as the temperature increases, the conductive phase transformation effect is better. The higher the heat treatment temperature of coal, the more amorphous carbon transforming into crystalline carbon completely, but the less types of phases .

The distributed processing of β-SiC submicron powders were carried out by tetramethyl ammonium hydroxide (TMAH). The effects of dispersant on the dispersion stability of the β-SiC powders were investigated by the particle size, flow characteristics, morphology and other physical properties of the powder before and after dispersed treatment. The results show that the best dispersing state of SiC is obtained by using the ultrasonic 7mins. In these conditions, the suspension solid content of β-SiC is 8%, and the dosage of TMAH is 8wt.%. An optimum amount of TMAH is proved as 8wt.% when the solid content is kept as 8wt.%. The powder particle size has been reduced from 0.73μm to 0.45μm, and its flowability has improved significantly after low temperature drying. The dispersed powder has small particle size, uniform distribution and excellent dispersion stability.

βThe B4C and phenolic resin were used as sintering additives to study the different β-SiC（cubic silicon carbide) addition to the performance of α-SiC pressureless sintering ceramics in this paper, and the optimum sintering temperature and the best β-SiC addition amount were determined. By XRD, SEM,density testing and other tests showed that, all the β-SiC transform 6Hα-SiC during the sintering process ,the ceramics have a Vickers hardness of 18.04 GPa,a frature toughness of 4.51MPa ,a density of 3.13g/cm3 by adding 15wt% β-SiC powders.

The β-SiCp/Al electronic packaging composites with excellent performance were successfully fabricated by pressureless infiltration technology in air.The effects of alloying elements, infiltration temperature and time on infiltration process and application of -SiC were studied.The results show that by adding appropriate magnesium to aluminum matrix, a interface reaction between oxide films of SiC and magnesium occurs, and the interface reaction product MgAl2O4 is generated, the interface wettability of Al and SiC and pressureless infiltration are improved.The interface harmful phase Al4C3 can be inhibited by adding silicon to aluminum matrix.Identified 850°C for the best infiltration temperature, and the thickness with infiltration time and larger, infiltration rate is about 10mm/hour.Under the same parameter conditions, the thermal properties of β-SiCp/Al electronic packaging material are 4 ~ 6% higher than that of ɑ-SiCp/Al. The β-SiCp/Al electronic packaging materials with 66% SiC volume ratio has lower coefficiency of thermal expansion than those ɑ-SiCp/Al electronic packaging materials.And the thermal expansion coefficient and thermal conductivity of β-SiCp/Al electronic material can satisfy the requirements for electronic packaging materials.

Objective To probe into the intervening action of polysaccharides of Zhu Zi Shen (Rhizoma Panacis Majoris) (PZZS) on oxidative stress and hemodynamics in rats with adriamycin-induced chronic congestive heart failure (CHF). Methods After SD rats were successfully modeled with adriamycin, they were randomly divided into a normal control group, a model group, a PZZS group, and a captopril group, and were administrated respectively. At the end of experiment, the hemodynamic function, whole heart weight index, and the blood CK, SOD, MDA, NO, NOS were detected; and the myocardial morphological examinations were carried out. Results Compared with the normal control group, the arterial systolic pressure (SBP), diastolic pressure (DBP), mean arterial pressure (MAP), heart rate (HR), left ventricular systolic peak (LVSP), and left ventricular pressure change rate (dp/dt max ) significantly decreased, and left ventricular end diastolic pressure (LVEDP), whole heart weight index, the blood CK, MDA, NO, NOS significantly increased in the model group. PZZS significantly improved the hemodynamic function, lowered the MDA and NO levels, and decreased the CK and NOS activities in the CHF rats. Conclusion PZZS can improve the hemodynamic function, and alleviate the oxidative stress reaction in the CHF rat.

Using a synthetic, alpha satellite consensus DNA unit as a probe, we could show dicentrics as well as acentric fragmented chromosomes in metaphases of gamma-irradiated lymphocyte cells and that the number of appearance of dicentrics or acentric fragments seemed to be proportional to radiation doses. To make such examination with a large number of chromosomes of metaphases, a quantitative fluorescence measurement was performed using a fluorescent microscope digital image analysis system. The relative amounts of fluoresceinated probe hybridized to alpha satellite DNA varied with chromosomes to a certain extent (1-4% of total probe-fluorescence of one metaphase). However, we could score dicentrics and acentric fragments as dots with extraordinarily higher or low percentage in plots of relative amount of probe-fluorescence on metaphase chromosome in gamma-irradiated cells. The number of appearance of acentric fragments were proportional to radiation doses of 1, 2 and 4 Gy. In the case of dicentrics, the number of appearance seemed to be proportional to square doses. Further, to reduce the variation of relative amounts of probe-fluorescence, we tried to make probe-fluorescence reflect the content of alpha satellite DNA on each chromosome more exactly, using a DNA probe amplified by polymerase chain reaction.

A chymotrypsin inhibitor, CI-1, and its co-dominant allelic counterpart CI-2, detectable in certain strains of Bombyx mori each as an electrophoretic band close to the origin, were purified from the larval hemolymph by using ion-exchange and affinity chromatography. CIs 1 and 2 were monomeric neutral proteins with a pI of 7.12 and 7.00 and an M r of about 10,400 and 7900, respectively. Amino acid analysis showed that these inhibitors were rich in aspartic acid (or asparagine), glutamic acid (or glutamine) and lysine, but poor, or lacking, in valine, methionine, histidine and arginine. In the amino-terminal sequence, 14 out of 20 amino acid residues analyzed were common between CIs 1 and 2.

The value was the highest at the later half of the 5th instar, although some differences in pattern of developmental changes were seen between the sexes. 3. 3. The electrophoretic pattern in activity of the chymotrypsin inhibitors exhibited little variation according to the sex and developmental stage. 4. 4. Eggs were found to possess the chymotrypsin inhibitors. 5. 5. The transplantation of ovary as well as a mating experiment showed that the inhibitors in the eggs at early stages were of the maternal origin, and that zygote-dependent synthesis of chymotrypsin inhibitors occurred after day 5 of embryogenesis.
