Abstract Background Goji (Lycium barbarum) is an important economic crop that is widely cultivated in Northwest China and is known as superfood. Goji plants are often affected by powdery mildew (Arthrocladiella mougeotii) in the field, resulting in considerable reduction in production and serious economic losses. The long-term reliance on agricultural chemicals to control powdery mildew not only leads to environmental pollution, but also causes excessive residues to accumulate in fruits, endangering human health. The plant epicuticular wax is the first physical barrier between land plants and the atmospheric environment, and plays an important role in the interactions of plants with pathogenic microorganisms. However, the relationship between epicuticular wax and powdery mildew resistance in goji plants is not well understood. Results The leaf epicuticular wax crystals of ‘Ningqi I’ goji were dense and small, and the wax load was 121.74 μg/cm2, which was 3.36 times greater than that of ‘Huangguo’ goji. The ratio of alcohol components in ‘Ningqi I’ goji was 121.18% greater than that in ‘Huangguo’ goji. The incidence rate and disease index of ‘Ningqi I’ goji inoculated with powdery mildew were significantly lower than those inoculated ‘Huangguo’ goji, showing high resistance and moderate susceptibility, respectively. After mechanical wax removal, the disease indices of the two goji varieties increased significantly, indicating extreme susceptibility. Seventeen differential expression genes showing significantly greater expression in ‘Ningqi’ goji leaves than in ‘Huangguo’ goji leaves were enriched in genes involved in fatty acid elongation and the wax biosynthesis pathway. The FAR activity and relative expression levels of LbaFAR and LbaWSD1 in ‘Ningqi I’ goji were significantly greater than those in ‘Huangguo’ goji. The relative expression levels of these genes significantly increased in the overexpression tobacco lines FAR-OE and WSD1-OE, and the wax load were augmented by 46.10% and 22.23%, respectively. Conclusions The increased expression of the LbaFAR and LbaWSD1 genes in ‘Ningqi I’ leaves led to increased FAR and WSD1 activity, increasing the wax load and alcohol content of the epicuticular of leaves, and improving leaf glossiness, resulting in strengthened resistance to powdery mildew. Therefore, the production of leaf epicuticular wax mediated by the LbaFAR and LbaWSD1 genes could be the main reason for the difference in powdery mildew resistance between the two goji varieties, which can provide a theoretical basis for the selection of resistant varieties to control goji powdery mildew. Objective: The aim of this study is to examine the perception, willingness to engage, and demand of community residents regarding the 'internet + nursing service' in a designated pilot area, aiming to offer insights for the widespread adoption of the 'internet + nursing service' throughout China. Methods: A survey pertaining to the 'internet + nursing service' was conducted from March to April 2022. The study specifically targeted residents within two sub-districts of a city in the Jiangsu province. The sampling technique employed in this study was stratified random sampling. Results: Out of a total of 400 community residents selected from two sub-districts in this region, 378 provided valid responses, resulting in an effective rate of 94.5%. Within the study cohort, 80 participants (21.16%) demonstrated familiarity with the concept of 'internet + nursing service.' Additionally, 231 participants (61.11%) conveyed their willingness to adopt such services. Regarding service preferences, the primary demands were for health guidance, vital sign monitoring, and basic care. Challenges in implementing the service were attributed to concerns related to medical risks, personal safety for both nurses and patients, and potential breaches of privacy. Conclusions: Residents in the pilot area exhibited a moderate awareness of the 'internet + nursing service,' with a relatively high willingness to embrace the program. There is a need for further refinement of pertinent laws, widespread dissemination of policies, and enhancements in the quality of nursing services. Slope instability under dynamic load is the technical difficulty in the engineering; the evaluation of slope stability under dynamic load and the control of dynamic load is particularly important. In this paper, taking the right side slope of K27+140 m~380 m typical section (K27 slope for short) in Chongqing Fuling-Fengdu-Shizhu expresses highway as an example to calculate and analyze. The K27 slope is under sinusoidal vehicle load and supported by anchor cable and antislide pile to resist downslide strength; at the same time, the combined effect of them is studied. Three-dimensional finite element methodology (FEM) is used to simulate the bedding slope with piles and anchor cable support; furthermore, the eigenvalue can be obtained. In order to reduce error of the elastic boundary conditions caused by the reflection effect of wavelengths, the combination of Lysmer surface viscous boundary and traditional ground support boundaries is utilized to analyze and calculate the time-histories during bedding slope under dynamic load. The dynamic response of pile anchor support to resist sliding force is obtained. The concept of the pile anchor supporting coordinate interval is put forward. Furthermore, it is verified that the pile anchor supporting coordinate interval can be used to evaluate the stability of the slope under dynamic load and provide a new method for the control of the dynamic load.

Autophagy and apoptosis are two different physiological phenomena, however they are closely related in function. This paper introduces the classification and process of autophagy and apoptosis, and discusses the effects of some key regulatory proteins in the formation of autophagosome and the dual role of apoptosis and anti-apoptosis proteins on autophagy and apoptosis, so as to explore the relationship between autophagy and apoptosis, and to provide theoretical basis for further study of autophagy and apoptosis related diseases.

Background: Dynamic and precise estimation of blood loss (EBL) is quite important for perioperative management. To date, the Triton System, based on feature extraction technology (FET), has been applied to estimate intra-operative haemoglobin (Hb) loss but is unable to directly assess the amount of blood loss. We aimed to develop a method for the dynamic and precise EBL and estimate Hb loss (EHL) based on artificial intelligence (AI)., Methods: We collected surgical patients' non-recycled blood to generate blood-soaked sponges at a set gradient of volume. After image acquisition and preprocessing, FET and densely connected convolutional networks (DenseNet) were applied for EBL and EHL. The accuracy was evaluated using R2, the mean absolute error (MAE), the mean square error (MSE), and the Bland-Altman analysis., Results: For EBL, the R2, MAE and MSE for the method based on DenseNet were 0.966 (95% CI: 0.962-0.971), 0.186 (95% CI: 0.167-0.207) and 0.096 (95% CI: 0.084-0.109), respectively. For EHL, the R2, MAE and MSE for the method based on DenseNet were 0.941 (95% CI: 0.934-0.948), 0.325 (95% CI: 0.293-0.355) and 0.284 (95% CI: 0.251-0.317), respectively. The accuracies of EBL and EHL based on DenseNet were more satisfactory than that of FET. Bland-Altman analysis revealed a bias of 0.02 ml with narrow limits of agreement (LOA) (-0.47 to 0.52 mL) and of 0.05 g with narrow LOA (-0.87 to 0.97 g) between the methods based on DenseNet and actual blood loss and Hb loss., Conclusions: We developed a simpler and more accurate AI-based method for EBL and EHL, which may be more fit for surgeries primarily using sponges and with a small to medium amount of blood loss., Competing Interests: Conflicts of Interest: All authors have completed the ICMJE uniform disclosure form (available at http://dx.doi.org/10.21037/atm-20-1806). Purpose: The aim of this study was to make a comparative analysis of the possible different expression of Th22 cells in two subtypes of autoimmune thyroid diseases (AITDs), i.e., Graves' disease (GD) and Hashimoto's thyroiditis (HT)., Methods: We recruited 61 AITDs patients (31 GD and 30 HT) and 22 controls. Serum level of IL-22 was measured by enzyme linked immunosorbent assay (ELISA). The proportion of Th22 cells in peripheral blood mononuclear cells (PBMCs) was analyzed by flow cytometry. The messenger RNA (mRNA) expressions of IL-22, its receptors (IL10R2, IL22R1) and key transcription factor (aryl hydrocarbon receptor, AHR) in PBMCs were assayed by quantitative reverse transcription polymerase chain reaction (qRT-PCR). Several cytokines of the cultured PBMCs were also measured under IL-22 stimulation., Results: The proportion of Th22 cells, serum IL22 level and IL-22 mRNA expression were significantly higher in patients with GD than in healthy controls. Additionally, AHR increased in GD patients compared to healthy controls. However, the elevation of Th22 cells and their relative cytokines was not found in patients with HT. Consistent with specific mRNAs expression of cultured PBMCs, IL-4 increment in supernatant was much higher in GD group than in control group, while IFN-γ levels were decreased under IL-22 stimulation., Conclusion: Th22 cells may participate in the pathogenesis of AITDs as a proinflammatory factor, especially in GD, through expressing and secreting IL-22.
