Purpose: To study the influence of cordycepin (Cor) on cisplatin insensitivity in bladder carcinoma, and its underlying mechanism of action.Methods: The effects of cisplatin and Cor treatments on the viability of T24-sensitive and T24/DDPinsensitive bladder carcinoma cells were investigated by using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) method to assess selectivity index. Flow cytometry was employed to evaluate the apoptosis of T24/DDP-resistant bladder cancer cells treated with cisplatin and Cor. The concentrations of PTEN, p-AKt and Akt in T24/DDP-resistant bladder cancer cells treated with cisplatin and Cor were determined by western blot assay.Results: Compared with T24-sensitive cells, the sensitivity of T24/DDP-resistant bladder cancer cells to cisplatin was significantly decreased, along with significant increase in half-inhibitory concentration (IC50) value, resulting in 10.56-fold increase in resistance (p < 0.05). The median effective concentration (EC50) value of Cor for DDP reversal was 1.03 ± 0.15 μM, and it had a high selectivity index for normal cells (> 48.5). The results from flow cytometry showed that Cor significantly enhanced the apoptosisinducing capacity of DDP in T24/DDP-resistant cells (p < 0.05), while Western blot data indicate that PTEN protein expression increased and phosphorylated Akt protein expression decreased in T24/DDPresistantcells after Cor treatment when compared with control group (p < 0.05).Conclusion: Cordycepin significantly improves the sensitivity of T24/ DDP-resistant bladder cancer cells to cisplatin via a mechanism related to the activation of PTEN/AKt signaling pathway, thus indicating that it is a potential candidate reversing DDP-resistance in bladder cancer. Although it is well established that the Kiss1/GPR54 system stimulates the reproductive axis in mammals, its functional roles, especially in male reproduction of non-mammalian species, is less clear. In this study, we have isolated the full-length kiss2 and gpr54 cDNAs from black porgy (Acanthopagrus schlegelii). The Kiss2 precursor expressed from kiss2 comprises 124 amino acids and contains a highly conserved 10-amino acid sequence, Kiss2-10 (FNFNPFGLRF). GPR54 comprises 375 amino acid residues and contains distinct characteristics of G protein-coupled receptors. Real-time PCR analysis indicated that kiss2 and gpr54 were expressed highly in the brain regions. Moreover, intraperitoneal injection of porgy Kiss2-10 could stimulate genes expression of the gpr54, gnrh1, gnrh3, fshβ, lhβ, p450c17, star, and ar, and the serum testerone level in male black porgy. Our findings demonstrate that the Kisspeptin stimulates the male reproductive axis in black porgy.

Thick plate structures are widely used in large deep-sea scientific detection equipment, such as deep-sea human-occupied vehicles, but they are prone to local damage caused by fatigue due to the complex working environment load. To prevent structural failure due to damage accumulation, it is necessary to carry out structural health monitoring for thick plate structures. In this work, two half-ring thickness poled d15 mode omnidirectional shear horizontal (SH) wave piezoelectric transducers are used to form an exciter/receiver network, and the SH2 mode wave packet is selected as the damage location wave packet. Then, the reconstruction algorithm for the probabilistic inspection of damage and an algorithm based on the time of guided wave flight are applied for damage detection in a 30 mm-thick aluminium plate. Experimental results show that structural damage detection based on SH waves can effectively identify and locate thick plate structural damage and evaluate the damage depth.

2D van der Waals materials are crystals composed of atomic layers, which have atomic thickness scale layers and rich distinct properties, including ultrafast optical response, surface effects, light-mater interaction, small size effects, quantum effects and macro quantum tunnel effects. With the exploration of saturable absorption characteristic of 2D van der Waals materials, a series of potential applications of 2D van der Waals materials as high threshold, broadband and fast response saturable absorbers (SAs) in ultrafast photonics have been proposed and confirmed. Herein, the photoelectric characteristics, nonlinear characteristic measurement technique of 2D van der Waals materials and the preparation technology of SAs are systematically described. Furthermore, the ultrafast pulsed fiber lasers based on classical 2D van der Waals materials including graphene, transition metal chalcogenides, topological insulators and black phosphorus have been fully summarized and analyzed. On this basis, opportunities and directions in this field, as well as the research results of ultrafast pulsed fiber lasers based on the latest 2D van der Waals materials (such as PbO, FePSe3, graphdiyne, bismuthene, Ag2S and MXene etc), are reviewed and summarized.

Traditional settlement is usually the original place of urban development, also as the most important part of the city. Courtyard presents significant transformations in the architectural history of the traditional settlement as extended reflected the focal point of families, villages, towns, and cities form. The historic district of Fuzhou three lanes and seven alleys is the best case to study characteristics of urban form transformation. It built in the Jin Dynasty, has a long history, and the traditional courtyard space layout is to preserve the shape of flexible, rich layers, with important culture and social value. Based on the spatial morphological perspective, this paper depending on the cultural background and regional characteristics of Fuzhou, analyzed the characteristics of layout plans, architectural components, enclosed patterns, settlement’s structures, space orders in the traditional courtyard space of Fuzhou three lanes and seven alleys. Through the calculation of the courtyard space units and street space integration, that took space syntax to analyze in the potential relation between spatial structure and social activity, such as connectivity, control value, mean depth, integration value, Intelligibility, Synergy, therefore to summary the principle of the courtyard form. Purpose of this paper is hoping that it will provide a feasible way to inherit the characteristics of traditional settlements in the modern architecture. It could serve as the reference for the urban design and the spread control of UHI in the future.

Background: UVB radiation is an increasing threat to aquatic organisms, and also a potential driving force for zooplankton population dynamics. The bdelloid rotifer, for its parthenogenesis and high resistance to stress, can be used as an excellent model in ecology and evolutionary biology. Given the strong tolerance of bdelloid rotifer to stressors, we hypothesized that it could hold the ability to tolerate significant UV radiation, and this capacity may be associated with the trade-off strategies among life historic components. With this study, to estimate the ecological effects and responses of antioxidant system against UV radiation, the acute lethal, life history strategies, population growth, and antioxidant enzyme activities were assessed in the bdelloid Rotaria rotatoria after exposure to UV-B radiation. Results: The persistence of tolerance (evaluating by LT50) in rotifer to stress was playing a more vital role than the radiation dose (LD50) in survival, with a significant negative correlation between the radiation intensity and LT50. The larger the culture volume, the weaker the lethal effect. Rotifers prolonged their first reproductive time (JP), and shortened their reproductive period (RP) and longevity with the increasing of radiation dose, and the fecundity was significant inhibited by UV-B radiation. These responses can be taken as energy trade-off to retard their mortality. The population density of rotifers increased at the lowest dose of radiation, and then descended with the increasing of UVR dose, and this pattern was also corroborated by detecting the content of SOD and CAT, which suggested that hormesis also applies to R. rotatoria under UV-B radiation stress. The enzyme SOD has higher level of content and more sensitive to low UVR than CAT.Conclusions: The lethal effects of UV-B radiation to organisms depends not only on the conditions they exposed to, like as intensity and time of exposure, but also combination of factors namely, the type and effectiveness of the strategy they employ to cope with the changed environment. The species specificity, ages and physiological status also could be the regulating factors under the stresses, also in this case there was the strategy of energy trade-off between reproduction and survival in rotifers.

Clear cell renal cell carcinoma (ccRCC) is one of the most common types of malignant adult kidney tumor. Tumor recurrence and metastasis is the primary cause of cancer‑associated mortality in patients with ccRCC. Therefore, identification of efficient diagnostic and prognostic molecular markers may improve survival times. The GSE46699, GSE36895, GSE53000 and GSE53757 gene datasets were downloaded from the Gene Expression Omnibus database and contained 196 ccRCC samples and 164 adjacent normal kidney samples. Bioinformatics analysis was used to integrate the four microarray datasets to identify and analyze differentially expressed genes. Functional analysis revealed that there were 12 genes associated with cancer, based on the tumor‑associated gene database. Erb‑B2 receptor tyrosine kinase 4, centrosomal protein 55 (CEP55) and vascular endothelial growth factor A are oncogenes, all of which were associated with tumor stage, whereas only CEP55 was significantly associated with survival time as determined by Gene Expression Profiling Interactive Analysis. The mRNA expression levels of CEP55 in ccRCC samples were significantly higher than those observed in adjacent normal kidney tissues based on The Cancer Genome Atlas data and reverse transcription‑polymerase chain reaction results. The receiver operating characteristic curve analysis revealed that CEP55 may be considered a diagnostic biomarker for ccRCC with an area under the curve of >0.85 in the training and validation sets. High CEP55 expression was strongly associated with sex, histological grade, stage, T classification, N classification and M classification. Univariate and multivariate Cox proportional hazards analyses demonstrated that CEP55 expression was an independent risk factor for poor prognosis. In addition, gene set enrichment analysis indicated that high CEP55 expression was associated with immunization, cell adhesion, inflammation, the Janus kinase/signal transducer and activator of transcription signaling pathway and cell proliferation. In conclusion, CEP55 was increased in ccRCC samples, and may be considered a potential diagnostic and prognostic biomarker for ccRCC.

Mechanical model and simulation model of shaper quick-return mechanism are introduced based on ADAMS. The number of rigid bodies is reduced by using composite joints constraint . It doesn’t need any other softwares and data interfaces in the simulation.The results prove the correctness of the simplified model.It improves the design efficiency and speed of simulation and provides a theoretical basis for the design and analysis of other six-bar mechanism.

A dynamic scheduling method for FJSP is put forward based on real-time adjustment of controllable margin, aiming at uncertain information and dynamic characteristic during practical job-shop scheduling process. Strategy and methods of dynamic scheduling adjustment are presented, which respond to real-time event. In view of practical operation condition and accuracy demands, two methods are proposed. One is rescheduling that condenses domain, the other is scheduling updating based on adjustment. Such strategy obtains dynamic robust scheduling of FJSP to ensure the real-time of algorithm, continuity and stability of scheduling schemes.

Combined with the stage-related characteristics in solving process of the Flexible Job-shop Scheduling Problem (FJSP) and the evolution characteristics of Genetic Algorithm (GA), a modified Adaptive Genetic Algorithm based on iterative generation and analysis of fitness values distribution is presented in this paper, which has both methods advantages. Instance simulation verifies that the FJSPs own characteristics are utilized in its solution by using the modified AGA, which overcomes traditional GAs limitation that initial stage of evolution is early and random search of medium-late stage is slow. Such methods are verified to accelerate convergence process, enhance searching efficiency and solving precision as well as avoid low efficiency and local optimum.

Aiming at uncertain information and dynamic characteristic during flexible job-shop scheduling process, some kind of dynamic scheduling method for flexible job-shop scheduling problem (FJSP) is put forward based on real-time adjustment. A dynamic simulation solution mode framework is presented for FJSP. This framework is inspired by adaptive control, combined with the robust scheduling and foreseeing scheduling. It has both advantages of such two scheduling methods, and its stable and highly efficient. Preliminary scheme generation method based on foreseeing dynamics scheduling is introduced then. Foreseeing function is realized by fault-handling algorithm and dynamic simulation solver on the basis of Adaptive Genetic Algorithm (AGA).

To study the dispersion characteristic of photonic crystal, the eigen-equations matrix of the light transmitting in is gotten in plane-wave expansion method, then the dispersion curves are calculated for two dimensional air-hole-type square photonic crystal respectively. And the influence on the band structures caused by oblique incidence also is studied by change incidence angle. The results of calculations demonstrate that: For two dimensional square structure photonic crystals, the shape of the band structures of TE mode is very similar to that of TM mode, but their positions of band gaps are different, especially the first band gap of TE mode in the square structure in direction is completely staggered with that of the TM mode. Moreover, we get the conclusion that the width of band gap enlarges with the increasing radius of the hole in square photonic crystal in direction.

In order to fully understand the physiological functions of adipose tissue, especially its secretory functions, and to provide a basis for the identification of novel obesity related genes, the gene expression profiling of human visceral adipose tissue was established by using cDNA array. 33 P -labelled cDNA, derived from visceral adipose tissue total RNA, was hybridized to a cDNA array containing over 16,000 expressed sequence-tagged clones which represent human singleton genes. The expressed sequence tag (EST) was considered to be expressed in visceral adipose tissue when the ratio of signal to noise was greater than or equal to 2. The results were analyzed with bioinformatics. Totally, 8230 genes were found to be expressed in visceral adipose tissue with 5200 known genes and 3030 known ESTs. Most of 84 secretory proteins, 120 receptors, and 74 transcription factors expressed in adipose tissue were newly identified. Many appetite-regulating related peptides or receptors and some reproduction-related genes were first found to be expressed in adipose tissue. Eight autocrine/paracrine systems were described for the first time in the visceral adipose tissue. These results clearly demonstrate that the visceral adipose tissue has important secretory functions and there is a complex local autocrine/paracrine regulatory network. The present work suggests that the visceral adipose tissue is an important component of the neuroendocrine-immune network and plays an important role in regulating appetite not only via endocrine but also via autocrine/paracrine systems. The visceral adipose tissue might also play a role in regulating reproduction and sexual function.

To investigate the allergen composition in the species of shrimp and crab.The main allergens of shrimps and crabs were analyzed by SDS-PAGE and Western blot assay.The molecule weight of main allergic protein in shrimps was near to 36 000, and the main allergic proteins in different kinds of crabs were about 29 000, 36 000, 66 000, 89 000 molecule weight.There are common allergens among different kinds of shrimps and crabs.

The full length cDNA sequence for neuronal nitric oxide synthase (nNOS) gene from red drum (Sciaenops ocellatus) has been cloned, subjected to bioinformatic analysis, and examined for expression in different tissues. Red drum nNOS showed high identity to nNOS of mammals and other fish species. Notably, a unique 7-aa insertion was found in the important catalytic sites of the NO synthase domain, possibly affecting the function of red drum nNOS. Furthermore, this nNOS was expressed not only in brain but also in most of the internal organs including liver, intestine, spleen, head kidney and thymus.

To investigate the electrophysiological effects of 17β-estradiol on pacemaker cells in sinoatrial (SA) nodes of rabbits and the underlying mechanism, intracellular microelectrode technique was used to record action potential (AP) in SA node cells of rabbits. The results showed that: (1) 17β-estradiol (1, 10, 100 μmol/L) not only significantly decreased the amplitude of action potential (APA) and the maximal rate of depolarization (V(max)), but also decreased the velocity of diastolic (phase 4) depolarization (VDD) and rate of pacemaker firing (RPF) in a concentration-dependent manner. The AP duration at 50% repolarization (APD(50)) and at 90% repolarization (APD(90)) were prolonged. But the maximal diastolic potential (MDP) was not affected. (2) Pretreatment with tamoxifen (10 μmol/L), an inhibitor of estrogen receptor, did not block the electrophysiological effects of 17β-estradiol (10 μmol/L) on SA node cells. (3) Pretreatment with N(G)-nitro-L-arginine methyl ester (L-NAME, 100 μmol/L), a nitric oxide (NO) synthase inhibitor, completely abolished the electrophysiological effects of 17β-estradiol (10 μmol/L) on SA node cells. The results suggest that 17β-estradiol inhibits the electrophysiological activity of pacemaker cells in SA nodes of rabbits in a concentration-dependent manner possibly through a non-genomic mechanism related with NO.

C-reactive protein (CRP) is considered as one of the most sensitive markers of inflammation. The aim of the present study is to investigate the effects of CRP on the production of adiponectin in 3T3-L1 adipocytes. Northern and western blot analysis revealed that CRP treatment inhibited adiponectin mRNA expression and secretion in a dose- and time-dependent manner. Co-incubation of adipocytes with rosiglitazone and CRP decreased induction of adiponectin gene expression by rosiglitazone. However, luciferase reporter assays did not show that CRP affected the activity of ~2.1 kb adiponectin gene promoter, which was increased by rosiglitazone alone. Pharmacological inhibition of phosphatidylinositol (PI)-3 kinase by LY294002 partially reversed inhibition of adiponectin gene expression by CRP. These results collectively suggest that CRP suppresses adiponectin gene expression partially through the PI-3 kinase pathway, and that decreased production of adiponectin might represent a mechanism by which CRP regulates insulin sensitivity.

Adipose tissue is not simply a depot of energy, but is an active endocrine organ. The adipokines play an important role in the pathogenesis of metabolic syndrome. The proinflammatory adipokines secreted from expanded visceral adipose tissue directly induce insulin resistance and vascular injuries. A better understanding of the endocrine function of adipose tissue may lead to more rational therapy for metabolic syndrome.

To investigate the expression of feeding-related peptide receptors mRNA in GT1-7 cell line and roles of leptin and orexins in the control of GnRH secretion.Receptors of bombesin3, cholecystokinin (CCK)-A, CCK-B, glucagon-like peptide (GLP)1, melanin-concentrating hormone (MCH)1, orexin1, orexin2, neuromedin-B, neuropeptide Y (NPY)1 and NPY5, neurotensin (NT)1, NT2, NT3, and leptin receptor long form mRNA in GT1-7 cells were detected by reversed transcriptase-polymerase chain reaction. GT1-7 cells were treated with leptin, orexin A and orexin B at a cohort of concentrations for different lengths of time, and GnRH in medium was determined by radioimmunoassay (RIA).Receptors of bombesin 3, CCK-B, GLP1, MCH1, orexin1, neuromedin-B, NPY1, NPY5, NT1, NT3, and leptin receptor long form mRNA were expressed in GT1-7 cells, of which, receptors of GLP1, neuromedin-B, NPY1, and NT3 were highly expressed. No amplified fragments of orexin2, NT2, and CCK-A receptor cDNA were generated with GT1-7 RNA, indicating that the GT1-7 cells did not express mRNA of them. Leptin induced a significant stimulation of GnRH release, the results being most significant at 0.1 nmol/L for 15 min. In contrast to other studies in hypothalamic explants, neither orexin A nor orexin B affected basal GnRH secretion over a wide range of concentrations ranging from 1 nmol/L to 500 nmol/Lat 15, 30, and 60 min.Feeding and reproductive function are closely linked. Many orexigenic and anorexigenic signals may control feeding behavior as well as alter GnRH secretion through their receptors on GnRH neurons.

To investigate the circadian rhythmicity of adiponectin in patients with Cushing's syndrome and obesity, as well as in normal subjects. The effects of glucocorticoids, insulin and leptin on the secretion of adiponectin were also explored.15 patients with Cushing's syndrome and 10 with obesity as well as 9 normal subjects were recruited and their serum adiponectin, glucocorticoids, insulin and leptin levels were determined 6 times a day (8:00, 12:00, 16:00, 20:00, 24:00, 3:00).Circadian rhythmicity of adiponectin was not observed, but the adiponectin levels in patients with obesity were lower than those in the normal subjects at all 6 time spots. The area under curve (AUC) of serum adiponectin concentrations were significantly negatively correlated with body mass index (BMI, r = 0.46, P0.01), waist circumference (r = -0.33, P0.05) and insulin AUC (r = -0.36, P0.05). A multiple linear regression analysis revealed that BMI and waist circumference were two significant independent parameters of the plasma adiponectin level. The adiponectin levels did not manifest any change after short term dexamethasone administration with both high and low doses, but were obviously decreased after surgery.Neither long-term endogenous hyperglucocorticoidism nor short-term dexamethasone administration affects the serum adiponectin levels and the serum adiponectin concentrations remain unchanged even with elevated postprandial insulin levels.

To investigate the effect of acupuncture and moxibustion on epithelial cell apoptosis and expression of Bcl-2, Bax, fas and FasL proteins in rat ulcerative colitis.A rat model of ulcerative colitis was established by immunological methods and local stimulation. All rats were randomly divided into model control group (MC), electro-acupuncture group (EA), herbs-partition moxibustion group (HPM). Normal rats were used as normal control group (NC). Epithelial cell apoptosis and expression of Bcl-2, Bax, fas and FasL proteins were detected by TUNEL and immunohistochemical method respectively.The number of epithelial cell apoptosis in MC was significantly higher than that in NC, and was markedly decreased after the treatment with herbs-partition moxibustion or electro-acupuncture. The expression of Bcl-2, Bax, fas and FasL in colonic epithelial cells in MC was higher than that in NC, and was markedly down- regulated by herbs-partition moxibustion or electro-acupuncture treatment.The pathogenesis of ulcerative colitis in rats involves abnormality of apoptosis. Acupuncture and moxibustion can regulate the expression of Bcl-2, Bax, fas and FasL proteins and inhibit the apoptosis of epithelial cells of ulcerative colitis in rats by Bcl-2/Bax, fas/FasL pathways.

To observe the effect of berberine on the differentiation of 3T3-L1 preadipocytes into adipocytes and to elucidate its mechanism.3T3-L1 preadipocytes were cultured and then divided into 7 groups into whose media were added berberine of the concentrations of 0, 0.1, 1, 10, and 100 micro mol/L, 100 nmol/Linsulin, and 10 micro mol/L berberine + 100 nmol/L insulin. The proliferation of 3T3-L1 preadipocytes was detected by MTT method. The accumulation of lipid in the cytoplasm of differentiated adipocytes was observed by oil red O staining. The peroxisome proliferation activated receptor gamma2 (PPARgamma2) mRNA and protein were detected by RT-PCR and Western blotting respectively.MTT method showed that the absorbance at 570 nm of 3T3-L1 preadipocytes increased by 17% (P0.01), 36% (P0.001), and 22% (P0.05) in the groups of 1, 10, and 100 micro mol/L berberine, by 53% (P0.0001)in the group of 100 nmol/L insulin, and by 66% in the group of 10 micro mol/L berberine + 100 nmol/L insulin. There were less and smaller lipid droplets in the 3T3-L1 adipocytes treated with berberine as compared with the untreated control cells and only 10% - 20% of the treated cells displayed big lipid drops. RT-PCR showed that berberine significantly reduced the expression of PPARgamma2 mRNA by 48% (P0.01) in the course of 3T3-L1 adipocyte differentiation. Western blotting showed that berberine inhibited the expression of PPARgamma2 protein.Berberine promotes the proliferation of 3T3-L1 preadipocytes, decreases the accumulation of lipid drops therein, and inhibits the terminal differentiation of adipocyte, which may be associated with its effect on decreasing the expression of PPARgamma2 mRNA and protein, suggesting that berberine has advantages in the treatment of obesity patients with type 2 diabetes.

Now many countries have developed cancer therapy with heavy ions, especially in GSI (Gesellschaft f r Schwerionenforschung mbH, Darmstadt, Germany), remarkable results have obtained, but due to the complexity of particle track structure, the basic theory still needs further researching. In this paper, the genotoxic effects of heavy ions irradiation on SMMC-7721 cells were measured using the single cell gel electrophoresis (comet assay). The information about the DNA damage made by other radiations such as X-ray, gamma-ray, UV and fast neutron irradiation is very plentiful, while little work have been done on the heavy ions so far. Hereby we tried to detect the reaction of liver cancer cells to heavy ion using comet assay, meanwhile to establish a database for clinic therapy of cancer with the heavy ions.The human hepatoma cells were chosen as the test cell line irradiated by 80 Mev/u (20)Ne(10+) on HIRFL (China), the radiation-doses were 0, 0.5, 1, 2, 4 and 8 Gy, and then comet assay was used immediately to detect the DNA damages, 100-150 cells per dose-sample (30-50 cells were randomly observed at constant depth of the gel). The tail length and the quantity of the cells with the tail were put down. EXCEL was used for statistical analysis.We obtained clear images by comet assay and found that SMMC-7721 cells were all damaged apparently from the dose 0.5 Gy to 8 Gy (t-test: P0.001, vs control). The tail length and tail moment increased as the doses increased, and the number of cells with tails increased with increasing doses. When doses were higher than 2 Gy, nearly 100 % cells were damaged. Furthermore, both tail length and tail moment, showed linear equation.From the clear comet assay images, our experiment proves comet assay can be used to measure DNA damages by heavy ions. Meanwhile DNA damages have a positive correlation with the dose changes of heavy ions and SMMC-7721 cells have a great radiosensitivity to (20)Ne(10+). Different reactions to the change of doses indicate that comet assay is a useful tool to detect DNA damage induced by heavy ions.

AIM: To observe the therapeutic effect of moxibustion on ulcerative colitis and its influence on the colonic mucosal morphology. METHODS: Forty-six patients with ulcerative colitis were randomly divided into the moxibustion with herbal medicine underneath group and the western medicine group. Thirty patients were treated with the above moxibustion and 16 patients with Salicylaye fapyridine (SASP). The colonic mucosa of 13 patients in the moxibustion group was observed by colonoscopy before and after the treatment. Mucin was also analyzed by H.E and AB-PAS staining. RESULTS: Seventeen patients were clinically cured, 12 were improved and 1 unchanged in the moxibustion group. In the control group, 5 patients were clinically cured, 7 improved and 4 unchanged. Thirteen patients with active UC were taken as the subjects for histopathologic analysis in this study. The colonic mucosal lesions were remarkably improved and the characteristic of the mucin also changed. In most sections, the chronic inflammation of mucosa was geatly ameliorated (P < 0.01). The inflammatory cell infiltratation much decreased and neutrophils, disapeared in most sections (P < 0.001). The goblet cells significantly increased (P < 0.001); crypt paracrypt abscess or mucosal ulceration was seen (P < 0.001). CONCLUSION: The rate of cure of ulcerative colitis by moxibustion with herbal medicine beneath is superior to that by SASP. This sort of moxibustion can effectively improve the colonic mucosal lesions and restore the proportion of mucoprotein to near normal.

AIM:To observe the effect of acupuncture and moxibustion on the expression of IL-1beta and IL-6 mRNA in ulcerative colitis rats.METHODS:The SD rat ulcerative colitis model was created by immunological method associated with local stimulation. Colonic mucosa was prepared from human fresh surgical colonic specimens, homogenized by adding appropriate amount of normal saline and centrifuged at 3000r/min. The supernatant was collected for measurement of protein conentration and then mixed with Freund adjuvant. This antigen fluid was first injected into the plantae of the model group rats, and then into their plantae, dorsa, inguina and abdominal cavities (noFreund adjuvant for the last injection) again on the 10th, 17th, 24th and 31st day. When a certain titer of serum anti colonic antibody was reached, 2% formalin and antigen fluid (no Freund adjuvant) were administered separately by enema. The ulcerative colitis rat model was thus set up. The animals were randomly divided into four groups: model control group (MC, n = 8), electro acupuncture group (EA, n = 8), herbs partition moxibustion group (HPM 8), normal control group (NC,n = 8). HPM: Moxa cones made of refined mugwort floss were placed on the medicinal pad (medicinal pad dispensing: Radix Aconiti praeparata, cortex Cinnamomi, etc) for Qihai (RN 6) and Tianshu (ST 25, bilateral) and ignited. Two moxa cones were used for each acupoint once a day and 14 times in all. EA: Tianshu (bilateral) and Qihai were stimulated by the intermittent pulse with 2Hz frequency, 4mA intensity for 20 minutes once a day and 14 times in all. After treatment, rats of all four groups were killed simultaneously. The spleen was separated and the distal colon was dissected. Total tissue RNA was isolated by the guanidinium thiocyanate phenol chloroform extraction method. RT-PCR technique was used to study the expression of IL-1 beta and IL-6 mRNA.RESULTS:IL-1 beta and IL-6 mRNAs were not detected in the spleen and colonic mucosa of the NC rats, whereas they were significantly expressed in that of the MC rats.IL-1 beta and IL-6 mRNAs were markedly lower in the EA and HPM rats than that in MC rats. There was no significant difference between the levels of IL-1 beta and IL-6 mRNAs in the EA and HPM rats. The expressions of IL-1 beta and IL-6 mRNAs were nearly the same in the spleen and colon of all groups.CONCLUSION:Acupuncture and moxibustion greatly inhibited the expression of IL-1 beta and IL-6 mRNA in the experimental ulcerative colitis rats.
