The corrosion behavior and mechanism of P110 steel tubing in in low CO2 particle pressure and high salinity formation water environment was investigated through high temperature and high pressure dynamic immersion tests, macro-morphology analysis, scanning electron microscopy(SEM) inspection, energy disperse spectroscopy(EDS) analysis and X-ray diffraction(XRD) analysis. The Results showed that the increasing of CO2 partial pressure accelerated the corrosion process of P110 tubing steel, and the corrosion rates of P110 tubing decreased with the increasing of temperature. Furthermore, the high content of Ca2+ in the formation water also had a big influence on the corrosion behavior of P110 tubing steel, for it could affect the precipitation quantity and morphology of CaCO3 at different temperature and CO2 particle pressure.

The effect of pre-stretching and under-aging treatment on microstructure and fatigue crack resistance of Al-4.1Cu-1.27Mg-0.47Mn-0.02Ti alloy has been investigated in present work. The results showed that the fatigue strength corresponding to 107 stress cycle numbers of the sample pre-stretched 4% and under-aged at for 2 hours reached up to 280MPa, 80MPa higher than that pre-stretched 0% and peak-aged at for 8 hours. Microstructural observations showed that fine and uniformly dispersed S’ plates were present in the sample treated by pre-stretching and under-aging, whereas, a coarse S’ plate was formed in the sample when treated by 0% pre-stretching and peak-aging. It is indicated that the pre-stretching and under-aging treatment promotes the dispersoid formation of strengthening particles, enhancing the fatigue crack resistance and resultant fatigue strength. In recent years, with the rapid development of modern petroleum industry, some conventional oil country tubular goods (OCTGs) have been unable to meet the requirements of ultra-deep oil and gas drilling and production. Titanium alloy OCTGs showed broad application prospects due to its high specific strength, Low elastic modulus and strong CO2+H2S+Cl- corrosion resistance. However, large-scale applications of titanium alloy OCTGs were seldom reported, which is mainly attributed to the lack of anti-corrosive resistance and mechanical performance of titanium alloy OCTGs in the working conditions. In the present work, the research progress on properties and application of titanium alloy OCTGs were reviewed. The resistance to uniform corrosion, stress corrosion cracking, pit and crevice corrosion, mechanical properties of the titanium alloy OCTGs in air and downhole corrosion media were comprehensively discussed. Application status and prospect of titanium alloy OCTGs in oil and gas exploration were also summarized.

New information of the genus Agalope Walker, 1854 from mainland China is presented. Three new species of the Agalope pica species-group are described: A. haoi S.-Y. Huang sp. n. from Weixi County, Yunnan, A. chayuensis S.-Y. Huang & Pan sp. n. from Chayu County, Southeastern Xizang and A. owadai S.-Y. Huang sp. n. from Bomi and Jiali Counties, Southeastern and Eastern Xizang. The little-known Agalope aurelia Oberthür, 1923 and A. lucia Oberthür, 1923 were rediscovered, with the male of the latter reported for the first time. Based on the newly discovered male, Agalope lucia is found to be a close relative of A. dejeani, hence it is excluded from the A. pica species-group and transferred to the A. bieti species-group. A new species of the Agalope hyalina species-group is also described: A. jianqingi S.-Y. Huang sp. n. from Pianma, Western Yunnan. Adults and genitalia of the aforementioned and related taxa are illustrated. An updated checklist of the genus is also provided.

Hypertension is the leading cause of morbidity and mortality globally among all cardiovascular diseases. Purinergic signalling plays a crucial role in hypertension through the sympathetic nerve system, neurons in the brain stem, carotid body, endothelium, immune system, renin-angiotensin system, sodium excretion, epithelial sodium channel activity (ENaC), and renal autoregulation. Under hypertension, adenosine triphosphate (ATP) is released as a cotransmitter from the sympathetic nerve. It mediates vascular tone mainly through P2X1R activation on smooth muscle cells and activation of P2X4R and P2YR on endothelial cells and also via interaction with other purinoceptors, showing dual effects. P2Y1R is linked to neurogenic hypertension. P2X7R and P2Y11R are potential targets for immune-related hypertension. P2X3R located on the carotid body is the most promising novel therapeutic target for hypertension. A1R, A2AR, A2BR, and P2X7R are all related to renal autoregulation, which contribute to both renal damage and hypertension. The main focus is on the evidence addressing the involvement of purinoceptors in hypertension and therapeutic interventions.

Acupuncture point specificity has been recognized as a key scientific issue in traditional Chinese medicine (TCM), but there is limited clinical trial or animal study to verify the characteristics of PC6, BL15, and ST36 in the protection from myocardial injury. We aimed to compare the effects among these three acupoints on the acute myocardial infarction mice model and to explore possible mechanisms for the first time. We found that PC6 is the most appropriate acupoint to deliver efficacy and safety to treat acute MI in mice. BL15 stimulation improved the systolic function, but increased the risk of arrhythmia. ST36 only slightly attenuated systolic function and had no effect on arrhythmia during MI. RNA profiles of skin tissue in local acupoints demonstrated that the most altered DEGs and related pathways may partly support its best effects of PC6 treatment on MI injury, and support the observed phenomenon of the acupoint specificity. New information on the genus Fossia Volynkin, Ivanova & S.-Y. Huang, 2019 from China is provided. A new subspecies of F. bachma (Volynkin & Černý, 2018) is described from Hainan, southern China: Fossia bachma hainanensis S.-Y. Huang & Volynkin ssp. nov., which is confirmed by morphological and molecular evidences. Fossia punicea (Moore, 1878) has been formally recorded from Yunnan, southwestern China, where it is represented by the subspecies F. punicea kachina (Volynkin & Černý, 2018). Adults and genitalia of the aforementioned taxa are illustrated.

Molecular phylogenetic analysis indicates that the genus Carterocephalus is not monophyletic. Based on combined molecular and morphological evidence, we propose a new genus, Pulchroptera Hou, Fan & Chiba, gen. nov., for Pamphila pulchra Leech, 1891. The adult, wing venation, and male genitalia of Pulchroptera pulchracomb. nov., Carterocephalus palaemon, and related genera are illustrated.

A new species of the genus Aberrasine Volynkin & S.-Y. Huang, 2019, Aberrasine tiani S.-Y. Huang, Volynkin, N. Singh & M. Wang, sp. nov. is described from southern Xizang, China. The species belongs to the A. strigivenata species-group having a characteristic reduced forewing pattern consisting of a medial transverse line and lengthwise strokes along veins. Adults and genitalia of the new and the similar species are illustrated.

Compared with aluminum alloy and alloy steel, titanium alloy has higher specific strength, lower modulus of elasticity, and better toughness, fatigue performance and corrosion resistance. In terms of oil well tubing, the development of titanium alloy lags behind that of aluminum alloy and alloy steel. Aluminum alloy tubing is sensitive to pitting, fatigue corrosion and stress corrosion cracking. At the same time, it is not suitable for ultra-deep wells due to temperature limitations. Easily interact with corrosive media to cause corrosion and cracking. Titanium alloy oil well tubing is expected to solve this corrosion problem, but its corrosion resistance research is still incomplete. Therefore, it is necessary to develop titanium alloy oil well tubing with good corrosion resistance to improve corrosion fatigue (CF), fatigue during deep oil well and natural gas drilling operations. Catastrophic brittle fracture caused by hydrogen induced cracking (HIC), pitting corrosion and sulfide stress cracking (SSC). In this paper, by investigating a large number of domestic and foreign documents, the corrosion types of titanium alloy oil well pipes are analyzed, and the research status of corrosion resistance of titanium alloy oil well pipes is reviewed from three aspects: oil pipes, casings and drill pipes.

Laser cladding is an effective way to improve the surface hardness and wear resistance of titanium alloy. The powder materials and laser cladding process parameters have great influence on the properties of the wear resistant coating. In order to reduce the common defects including the cracks and pores of the coating, titanium alloy powder and self-melting alloy powder are selected to ensure the good adhesion between titanium alloy and wear resistant coating. In addition, the strengthening phase and self-lubricating phase were added to the laser cladding powder material to improve the micro hardness and reduce the friction coefficient, which bring in the improvement of the wear resistance of the coating. In the present work, the selection principle and application effect of powder materials for wear-resistant coatings are reviewed. the research status of the influence of laser technological parameters on coating properties is discussed. Meanwhile, the methods to reduce the defect of wear resistance coating by laser cladding are proposed.

Epoxy coatings and Reduced Graphene Oxide (RGO) modified epoxy coatings were prepared on N80 tubing steel. The influence of temperature on the corrosion resistance of RGO modified epoxy coatings was studied. And the corrosion behavior of epoxy coatings and RGO modified epoxy coatings in 10.0 wt% NaCl solution at different temperatures was compared. The results showed that the corrosion resistance of both coatings decreased with the increase of temperature. However, the corrosion resistance of RGO modified epoxy coating was two to three orders of magnitude higher than that of epoxy coating at different test temperature. The addition of RGO nanosheets greatly enhanced the corrosion resistance of epoxy composite coatings in 10.0 wt% NaCl solution at high temperature due to their high impermeability and negative coefficient of thermal expansion.

BackgroundEarly gastric cancer (EGC) has a high survival rate, but it is difficult to diagnosis. Recently, artificial intelligence (AI) based on deep convolutional neural network (DCNN) has made significant progress in the field of gastroenterology. The purpose of this study was to establish a DCNN assist system to improve the detection of EGC.Methods3400 EGC and 8600 benign images were collected to train the DCNN to detect EGC. Subsequently, its diagnostic ability was compared to that of endoscopists using an independent internal test set (ITS, including 1289 images) and an external test set (ETS, including 542 images) come from three digestive center.ResultsThe diagnostic time of DCNN and endoscopists were 0.028s, 8.05 ± 0.21s, 7.69 ± 0.25s in ITS, and 0.028s, 7.98 ± 0.19s, 7.50 ± 0.23s in ETS, respectively. In ITS, the diagnostic sensitivity and accuracy of DCNN are 88.08%(95% confidence interval,95%CI,85.24%-90.44%), 88.60% (95%CI,86.74%-90.22%), respectively. In ETS, the diagnostic sensitivity and accuracy are 92.08% (95%CI, 87.91%- 94.94%),92.07%(95%CI, 89.46%-94.08%),respectively. DCNN outperformed all endoscopists in ETS, and had a significantly higher sensitivity than the junior endoscopists(JE)(by18.54% (95%CI, 15.64%-21.84%) in ITS, also higher than JE (by21.67%,95%CI, 16.90%-27.32%) and senior endoscopists (SE) (by2.08%, 95%CI, 0.75%-4.92%)in ETS. The accuracy of DCNN model was higher (by10.47%,95%CI, 8.91%-12.27%) than that of JE in ITS, and also higher (by14.58%,95%CI, 11.84%-17.81%; by 1.94%,95%CI,1.25%-2.96%, respectively) than JE and SE in ETS.ConclusionThe DCNN can detected more EGC images in a shorter time than the endoscopists. It will become an effective tool to assist in the detection of EGC in the near future.

In order to study the comprehensive effects of different types of dehydrating agents on the dewatering and solidification of dredged sediments, this study took the dredged sediments of Taihu Lake as the research object and selected microorganisms, polymeric iron aluminum salts, organic polymers, organic-inorganic composites, and aluminum salt microorganisms. These five types of composite dehydrating agents were used to conduct a three-month solidification test on the dredged sediment by means of geotechnical pipe bag solidification. The results of the study showed that the dehydration efficiency of organic polymers and organic-inorganic composite chemicals was better. After one month, the water content of sediment dropped to 61.78% and 63.26%, respectively, which then dropped to 40.56% and 32.16% after three months. Compared with that of the unsolidified sludge, the total nitrogen of the bottom sludge after solidification by the organic-inorganic composite agent was reduced by 74.82%, reaching 591 mg·kg

Pancreatic cancer remains clinically challenging because of the lack of efficient targeted therapies and high aggressiveness. NEDD8 activating enzyme (NAE) plays a critical role in various cellular functions in cancers. Herein, we report the synthesis, optimization, and evaluation of a new series of pyrido[2,3-d]pyrimidin-7(8H)-one derivative as highly selective and efficacious NAE inhibitors, enabling rapid degradation of related substrates and potent inhibition of BxPC-3 cell proliferation. Moreover, western blot assays demonstrated that compound 51 could inhibit NAE activity, resulting in apoptosis in BxPC-3 cells. Furthermore, compound 51 induced cell apoptosis in vitro and inhibited tumor growth in BxPC-3 xenograft models. Our work established that 51 was a highly potent and efficacious NAE inhibitor, representing an effective strategy and great potential as a new targeted therapy for pancreatic cancer.

Manganese chloride (MnCl2) has been shown to inhibit the Yes-associated protein (YAP) in high-fat diet-fed ApoE−/− mice. Although YAP has been implicated in atherogenesis, there are limited data on the effects of MnCl2 on cardiac remodeling. In this study, we discovered, by electrocardiography, that hyperlipidemia led to spontaneous supraventricular arrhythmia (SVA) in ApoE−/− (KO) mice, with 3 of 9 KO + MnCl2 mice (33%) exhibiting lower incidence of spontaneous SVA than KO mice (6 of 10 mice, 60%). Echocardiography revealed that reduced systolic function in KO mice was reversed by MnCl2 treatment. Oil Red O staining of the aortas and biochemical analysis of lipid levels showed that MnCl2 inhibited plaque formation in a lipid metabolism-independent manner. MnCl2 inhibited inflammatory cell infiltration and reduced fibrosis, as evidenced by hematoxylin and eosin, immunohistochemical and Masson's trichrome staining, respectively. Our findings demonstrate that spontaneous SVA and reduced systolic function were blocked by MnCl2. Our findings show that MnCl2 was useful in delaying cardiac remodeling and reducing susceptibility to spontaneous SVA in a mouse model of hyperlipidemia.

Depressive disorder is the leading cause of disability worldwide, yet the mechanisms underlying depression are not fully understood. Vesicle release is essential for synaptic neurotransmission, the abnormalities of vesicle release and synaptic plasticity are associated with various neuropsychiatric disorders. Neural circuits are ensembles of interconnected neurons that collectively perform specific functions. To some extent, depression may be caused by a disruption in the structural and functional connections of the neural circuits underlying emotion regulation. In this review, we summarized the role of abnormalities of vesicle release and synaptic transmission, as well as the related regulatory molecules and signal pathways in the regulation of depression.

M1 microglial activation is crucial for the pathogenesis of early brain injury (EBI) following subarachnoid hemorrhage (SAH), and there is growing evidence that glucose metabolism is frequently involved in microglial activation. However, the molecular mechanism of glycolysis and its role in M1 microglial activation in context of EBI are not yet fully understood. In this study, firstly, the relationship between aerobic glycolysis and M1 microglial activation as well as SAH-induced EBI was researched in vivo. Then, intervention on mammalian target of rapamycin (mTOR) was performed to investigate the effects on glycolysis-dependent M1 microglial activation and EBI, and its relationship with hypoxia-inducible factor-1α (HIF-1α) in vivo. Next, Hif-1α was inhibited to analyze its role in aerobic glycolysis, M1 microglial activation and EBI in vivo. Lastly, both in vivo and in vitro, mTOR inhibition and Hif-1α enhancement were administered simultaneously, and the combined effects were further confirmed again. The results showed that aerobic glycolysis and M1 microglial polarization were increased after SAH, and glycolytic inhibition could attenuate M1 microglial activation and EBI. Inhibition of mTOR reduced glycolysis-dependent M1 microglial polarization and EBI severity by down-regulating HIF-1α expression, while enhancement had the opposite effects. Blockading HIF-1α had the similar effects as suppressing mTOR, while HIF-1α agonist worked against mTOR antagonist when administered simultaneously. In conclusion, the present study showed new evidence that aerobic glycolysis induced by mTOR/HIF-1α might promote EBI after SAH by activating M1 microglia. This finding provided new insights for the treatment of EBI.

Background and purpose Anxiety disorder is a common mental health disorder. However, there are few safe and fast-acting anxiolytic drugs available that can treat anxiety disorder. We previously demonstrated that the interaction of neuronal NOS (nNOS) with its carboxy-terminal PDZ ligand (CAPON) is involved in regulating anxiety-related behaviours. Here, we further investigated the anxiolytic effects of nNOS-CAPON disruptors in chronic stress-induced anxiety in animals. Experimental approach Mice were intravenously treated with nNOS-CAPON disruptors, ZLc-002 or Tat-CAPON12C, at the last week of chronic mild stress (CMS) exposure. We also infused corticosterone (CORT) into the hippocampus of mice to model anxiety behaviours and also delivered ZLc-002 or Tat-CAPON12C on the last week of chronic CORT treatment via pre-implanted cannula. Anxiety-related behaviours were examined using elevated plus maze, open field, novelty-suppressed feeding and light-dark (LD) tests. The level of nNOS-CAPON interaction was determined by co-immunoprecipitation (CO-IP) and proximity ligation assay (PLA). The neural mechanisms underlying the behavioural effects of nNOS-CAPON uncoupling in anxiety animal models were assessed by western blot, immunofluorescence and Golgi-Cox staining. Key results ZLc-002 and Tat-CAPON12C reversed CMS- or CORT-induced anxiety-related behaviours. ZLc-002 and Tat-CAPON12C increased synaptogenesis along with improved dendritic remodelling in CMS mice or CORT-treated cultured neurons. Meanwhile, blocking nNOS-CAPON interaction significantly activated the cAMP response element-binding protein (CREB)-brain-derived neurotrophic factor (BDNF) pathway, which is associated with synaptic plasticity. Conclusion and implications Collectively, these results provide evidence for the anxiolytic effects of nNOS-CAPON uncouplers and their underlying mechanisms in anxiety disorders.

During acidizing process in a gas field, leakage of 80SS anti-sulfur tubing occurred.The main reasons for failure of the tubing have been investigated based on analysis techniques of chemical composition analysis,Optical Microscopy (OM), Scanning Electron Microscope (SEM), Energy Dispersive Spectrometer (EDS) , X-ray Diffraction (XRD) and mechanical test. The results show that composition and mechanical properties of the tubing were all in accordance with the corresponding requirements of API Spec 5CT standards. Furthermore,leakage of tubing from inner wall was caused by combined impacts of acid corrosion, H2S corrosion, low PH value, high temperature and a certain pressure. Meanwhile, Low concentration of corrosion inhibitor was also one of the factors of corrosion perforation.

Graphene modified coatings have attracted extensive attention in recent years due to their excellent corrosion resistance and broad application prospects in the field of anti-corrosion. However, large-scale applications of graphene coatings were seldom reported, which is mainly attributed to the lack of fundamental research on the anti-corrosive mechanism and the long-term service performance evaluation of graphene modified coatings in actual working conditions. In the present work, the influence of the characteristics of corrosive medium, the content of graphene, the structure and morphology of graphene and the external environmental conditions on the anti-corrosive performance of graphene modified coatings were systematically reviewed. The deficiencies in the research of anti-corrosive performance of graphene modified coatings were summarized. The future work were prospected for the anti-corrosive performance and applications of graphene modified coatings in oil and gas exploration.

Corrosion of oil country tubular goods not only causes huge economic loss, but also brings serious environmental pollution. Stress corrosion cracking of oil country tubular goods in the sulfur-containing environment is the principal corrosion failure form in the exploitation of oil and gas process. Therefore, extensive investigations on hydrogen sulfide corrosion of steels for oil country tubular goods have been carried out. The aim of this work is to review the research progress on the hydrogen sulfide corrosion performance of steels for oil country tubular goods. The influence of alloy elements, defects, environmental factors and mechanical factors on hydrogen sulfide corrosion resistance of steels for oil country tubular goods were discussed. The future work in hydrogen sulfide corrosion performance of oil country tubular good and new sulfur-resistant steels for oil country tubular goods development were prospected.

Corrosion leakage became serious due to cyclic sewage injection and continuous increase in water injection pressure. Corrosion behavior for N80 casing was studied during long-term service, using corrosion reaction kettle and weight loss method. The morphology and composition were characterized with laser confocal microscopy (LCM), scan electron microscopy (SEM) and energy disperse spectroscopy (EDS). These results showed that corrosion rate remained constantly at initial stage, and increased gradually, then sharply reached to 0.327mm/a at later stage, accompanying with inflection point under oxygen condition. Meanwhile, corrosion rate decreased significantly from 0.028mm/a to 0.020mm/a under oxygen-free condition. The corrosion of N80 steel was dominated by dissolved oxygen, and accelerated by chloridion. The micro-mechanism was suggested to be the transition of defect structure from crack with micron range to etch pit with millimeter range.

Background Anxiety is a common disorder with high social burden worldwide. Dysfunction of serotonin-1A receptor (5-HT1A receptor) in the dentate gyrus (DG) of the hippocampus has been predominantly implicated in the anxiety behavior. However, the molecular mechanism underlying the deficiency of postsynaptic 5-HT1A receptor in regulating anxiety behavior remains unclear. Methods Using pharmacological and genetic methods, we investigated the role of detate nNOS in 5-HT1A receptor decline and anxiety behavior induced by chronic mild stress (CMS) in mice. Results Here we showed that local elevation of glucocorticoids in the DG accounted for chronic stress-induced anxiety behavior. Neuronal nitric oxide synthase (nNOS) mediated chronic stress-induced downregulation of 5-HT1A receptor in the DG through peroxynitrite anion (ONOO•) pathway but not cyclic guanosine monophosphate (cGMP) pathway. By using pharmacological tool drugs and nNOS knockout mice, we found that nNOS in the DG played a key role in chronic stress-induced anxiety behavior. Conclusions These findings uncovered an important role of nNOS-5-HT1A receptor pathway in the DG of the hippocampus in chronic stress-induced anxiety. Accordingly, we developed a "dentate nNOS-5-HT1A receptor closed-loop" theory (stress-glucocorticoids-nNOS-Nitric oxide-ONOO•-5-HT1A receptor -nNOS) of stress-related anxiety.

Anxiety is recognized as primary clinical phenotype of psychiatric disorders. However, many patients with anxiety have not yet received effective treatment. Our previous study demonstrated that hippocampal nNOS-CAPON interaction is implicated in anxiety-related behaviors, and blocking nNOS-CAPON interaction in the hippocampus produces anxiolytic-like effects. Here, ZLc-002, a small molecule inhibitor of nNOS-CAPON coupling, was evaluated for anxiolytic-like properties after systemic administered using anxiety behavioral tests, including open-field (OF), elevated plus maze (EPM), novelty-suppressed feeding (NSF) and light-dark (LD) tests. We reported that ZLc-002 when administered intraperitoneally at the dose of 40 or 80 mg/kg/d for 14 days produces anxiolytic-like effects. Furthermore, the similar effects of ZLc-002 were observed when administered intravenously at the dose of 10, 20 or 40 mg/kg/d for 7 days. More importantly, the mice dosing with 80 mg/kg/d ZLc-002 intraperitoneally or 40 mg/kg/d ZLc-002 intravenously for 3 days exerted significant behavioral effects. However, intragastric administration with ZLc-002 was devoid of effect on anxiety behaviors, even at high doses. Furthermore, intraperitoneal or intravenous treatment of ZLc-002 significantly disrupted the interaction between nNOS and CAPON in the hippocampus of adult mice, and there was a significant anxiolytic-like effect of ZLc-002 at day 3 after intrahippocampal microinjection. Our results verified that systemic administration of putative small molecule inhibitor of nNOS-CAPON can be used for the treatment of anxiety disorders.

Singh, 2020 is described from Chayu County of Southeastern Xizang: Barsine mahakala S.-Y. Huang & Volynkin sp. n. Barsine germana (Rothschild, 1913) and B. atypicobarsine N. Singh, Kirti & Joshi, 2016 are newly recorded from China, with the female of the latter described and illustrated for the first time. Adults and genitalia of the aforementioned taxa are illustrated.

Elderly patients tend to have poor self-efficacy and poor confidence in postoperative rehabilitation for hip fractures, and are prone to negative emotions, which affect treatment compliance.To evaluate the effects of evidence-based intervention on postoperative fear, compliance, and self-efficacy in elderly patients with hip fractures.A total of 120 patients with hip fracture surgically treated from June 2018 to June 2020 at the orthopedic department of our hospital were selected and divided into intervention and routine groups (Before the intervention, there were no statistically significant differences in motion phobia and pain fear scores between the groups (allEvidence-based nursing intervention can alleviate fear of postoperative rehabilitation in elderly patients who underwent hip fracture surgery, and improve rehabilitation treatment compliance and patient self-efficacy, which promote hip function recovery.

Stroke is the leading cause of adult lifelong disability worldwide. A stroke is an acute cerebrovascular disease with a variety of causes and corresponding clinical symptoms. Around 75% of surviving stroke patients experience impaired nerve function, and some suffer from traumatic fractures, which can lead to special care needs.To determine the effect of timing theory continuous care, with resistance training, on the rehabilitation and mental health of caregivers and stroke patients with traumatic fractures.Between January 2017 to March 2021, we selected 100 hospital admissions with post-stroke hemiplegia complicated with a traumatic fracture. Two participant groups were created: (1) Control group: given resistance training; and (2) Observation group: given timing theory continuous care combined with resistance training. The degree of satisfaction and differences in bone and phosphorus metabolism indexes between the two groups were compared. The self-perceived burden scale (SPBS) and caregiver burden questionnaire were used to evaluate the psychological health of patients and caregivers. The Harris hip function score, ability of daily living (ADL) scale, and global quality of life questionnaire (GQOL-74) were used to evaluate hip function, ability of daily living, and quality of life.Data were collected prior to and after intervention. Alkaline phosphatase (ALP), osteocalcin, and vitamin D3 in the observation group and control group increased after intervention (Timing theory continuous nursing with resistance training can reduce hip dysfunction in stroke patients with a traumatic fracture and enhance quality of life and mental health of patients and caregivers.

Preliminary assessment of rabies virus neutralizing activity, safety and immunogenicity of a recombinant human rabies antibody (NM57) compared with human rabies immunoglobulin (HRIG) in Chinese healthy adults.Subjects were randomly (1:1:1) allocated to Groups A (20 IU/kg NM57), B (40 IU/kg NM57), or C (20 IU/kg HRIG). One injection was given on the day of enrollment. Blood samples were collected on days -7 to 0 (pre-injection), 3, 7, 14, 28, and 42. Adverse events (AEs) and serious AEs (SAEs) were recorded over a period of 42 days after injection.All 60 subjects developed detectable rabies virus neutralizing antibodies (RVNAs) (0.05 IU/mL) on days 3, 7, 14, 28, and 42. The RVNA levels peaked on day 3 in all three groups, with a geometric mean concentration (GMC) of 0.2139 IU/mL in Group A, 0.3660 IU/mL in Group B, and 0.1994 IU/mL in Group C. At each follow-up point, the GMC in Group B was significantly higher than that in Groups A and C. The areas under the antibody concentration curve over 0-14 days and 0-42 days in Group B were significantly larger than those in Groups A and C. Fifteen AEs were reported. Except for one grade 2 myalgia in Group C, the other 14 were all grade 1. No SAEs were observed.The rabies virus neutralizing activity of 40 IU/kg NM57 was superior to that of 20 IU/kg NM57 and 20 IU/kg HRIG, and the rabies virus neutralizing activity of 20 IU/kg NM57 and 20 IU/kg HRIG were similar. Safety was comparable between NM57 and HRIG.

Background The pathological process of myocardial ischemia (MI) is very complicated. Acupuncture at PC6 has been proved to be effective against MI injury, but the mechanism remains unclear. This study investigated the mechanism that underlies the effect of acupuncture on MI through full-length transcriptome. Methods Adult male C57/BL6 mice were randomly divided into control, MI, and PC6 groups. Mice in MI and PC6 group generated MI model by ligating the left anterior descending (LAD) coronary artery. The samples were collected 5 days after acupuncture treatment. Results The results showed that treatment by acupuncture improved cardiac function, decreased myocardial infraction area, and reduced the levels of cTnT and cTnI. Based on full-length transcriptome sequencing, 5083 differential expression genes (DEGs) and 324 DEGs were identified in the MI group and PC6 group, respectively. These genes regulated by acupuncture were mainly enriched in the inflammatory response pathway. Alternative splicing (AS) is a post-transcriptional action that contributes to the diversity of protein. In all samples, 8237 AS events associated with 1994 genes were found. Some differential AS-involved genes were enriched in the pathway related to heart disease. We also identified 602 new genes, 4 of which may the novel targets of acupuncture in MI. Conclusions Our findings suggest that the effect of acupuncture on MI may be based on the multi-level regulation of the transcriptome.

Candida albicans is the most common human fungal pathogen in immunocompromised individuals. With the emergence of clinical fungal resistance, there is an urgent need to develop novel antifungal agents. AMP-17, a novel antimicrobial peptide from Musca domestica, has an antifungal effect against C. albicans, but its mechanism of antifungal action remains unclear. In the current study, we performed a proteomics analysis in C. albicans using TMT technique under the treatment of AMP-17. A total of 3931 proteins were identified, of which 3600 included quantitative information. With a 1.5-fold change threshold and a t-test p-value 0.05 as standard, 423 differentially expressed proteins (DEPs) were up-regulated and 180 DEPs were down-regulated in the AMP-17/control. Notably, GO enrichment revealed that DEPs associated with the cell wall, RNA and oxidative stress were significantly up-regulated, while DEPs involved in ergosterol metabolism and membrane were significantly down-regulated in the AMP-17/control. KEGG pathway enrichment revealed that DEPs involved seven significant metabolic pathways, mainly involved oxidative phosphorylation, RNA degradation, propanoate metabolism and fatty acid metabolism. These results show that AMP-17 induces a complex organism response in C. albicans, indicating that AMP-17 may inhibit growth by affecting multiple targets in C. albicans cells. SIGNIFICANCE: Antimicrobial peptides (AMPs) are an important part of the innate immune system of organisms and having broad range of activity against fungi, bacteria and viruses. These AMPs are considered as probable candidate for forthcoming drugs, due to their broad range of activity, lesser toxicity and decreased resistance development by target cells. AMP-17, a novel antimicrobial peptide from M. domestica, has significant antifungal activity against C. albicans. It has been confirmed that AMP-17 can play an antifungal effect by destroying the cell wall and cell membrane of C. albicans in previous studies, but its mechanism of action at the protein level is currently unclear. In the current study, using the TMT-based quantitative proteomics method, 603 differentially expressed proteins were identified in the cells of C. albicans treated with AMP-17 for 12 h, and these DEPs were closely related to cell wall, cell membrane, RNA degradation and oxidative stress. The results provide new insights into the potential mechanism of action of AMP- 17 against C. albicans. Meanwhile, it provides certain technical support and theoretical basis for the research and development of novel peptide drugs.

The stretch bending of L-section variable-curvature SUS301L stainless-steel roof bending beams for metro vehicles was numerically simulated. The causes of defects such as wrinkling, section distortion, and poor contour accuracy were analysed, and the corresponding control methods were proposed. The simulation results demonstrated that wrinkling in the small-arc segment could be eliminated by setting the die clearance and adjusting the elongation reasonably. Owing to the sidewall shrinkage of the profile in the process of stretch bending, the die groove depth was correspondingly reduced. Each section of the profile was effectively supported by the bottom of the die groove, and the section distortion could be controlled. Springback was the main reason for the poor contour accuracy, which could be compensated by modifying the die surface based on the springback value. Using the above defect control methods, forming experiments were performed on a new type of stretch bending die with variable die clearance and groove depth developed in this work. Finally, high-quality components were obtained, which verified the efficacy of the defect control methods.

Pancreatic lipase is an interfacial enzyme. The kinetic parameters of pancreatic lipase derived from heat release in crowded surroundings have not been reported. In this work, the effect of crowded medium on the catalytic activity of porcine pancreatic lipase (PPL) during lipid digestion was investigated by Isothermal Titration Calorimetry (ITC). The results showed that dietary polysaccharide Ficoll400 tended to increase the catalytic efficiency of PPL for both glyceryl trioleate and glyceryl trioctanoate substrates, whereas Dextran20 decreased the activity of the enzyme. The change in the fluorescence intensity and secondary structural content of PPL parallels the change of kcat/Km, of PPL, which was further confirmed in PEG2000. The findings of this research improve understanding of macromolecular crowding effects on interfacial catalysis by lipases. Moreover, it provides a useful method for research on enzyme kinetics in crowed medium by ITC.

In the present work, a self-built device called “full scale tubular goods erosion-corrosion test system” was used to test the P110SS tubing with a total length of 10 m and a full scale PFF78-70 flat valve to study their erosion-corrosion behaviors in the acceleration simulation operating condition of a well in Sichuang oil field of China. The erosion-corrosion performance of the specimen was investigated by KH-7700 digital microscope, scanning electron microscopy (SEM), X-ray power diffraction (XRD) and energy-dispersive spectroscopy (EDS). The results indicated that the P110SS tubing suffered catastrophic erosion-corrosion; the maximum pitting corrosion rate of the P110SS in the gas phase fluid was 5.29mm/a; while that in the liquid phase fluid was 11.13mm/a; A double-layer corrosion products scale, inner FeCO3 layer and outer mixture layer mainly dominated by FeO, Fe3O4, Fe2O3 and FeO(OH), formed on the P110SS tubing. Erosion-corrosion only occurred in the stress concentration area of the valve plate hole of the PFF78-70 plate valve, and the size of the corrosion pits is about 0.5mm.

With the further deterioration of the oil and gas exploitation environment, the corrosion of tubing becomes an intractable problem. Therefore, choosing suitable anticorrosive coating is of great significance to the safe operation life of tubing and reduce economic cost. Effect addition of appropriate graphene in coatings can significantly improve its corrosion resistance of the coating. However, most of the applied research are focus on marine heavy corrosion condition; the relationship between the corrosion mechanism and performance of graphene coating for tubing is not clear, probably due to difficulty in the characterization of the organizational structure characterization of the graphene modified coating. In the present work, Typical methods of the graphene coating characterization: Scanning Electron Microscope (SEM) with EDS, Transmission Electron Microscope (TEM), Atomic Force Microscope (AFM), X-ray Diffraction (XRD) and X-ray Photoelectron Spectroscopy (XPS) will be introduced, and the application prospect of the coating will be discussed. It indicated that the single characteristic method cannot analysis the structure and morphology information of graphene anticorrosion coating comprehensively. Flexibly use of a variety of characterization methods to study the anticorrosion mechanism and performance of the graphene modified coating for tubing is a better choice. The graphene modified anticorrosive coating for tubing is with the potential of wide applications.

A new modified low-cost titanium alloy, Ti-Al-X, was designed for petroleum drilling applications. The alloy ingots were prepared by combination of vacuum consumable electrode arc melting, forging/hot rolling, homogenization, and solid-solution/aging treatments. The hot deformation behavior of Ti-Al-X alloy was investigated by a thermal simulation machine Gleeble 1500 at temperature range of 850~1000°C with the strain rate range of 0.001 s-1~1s-1. The deformation resistance significantly decreases with the increase of deformation temperature and the strain rate. The alloy exhibits flow instability under the deformation conditions of strain rates about 0.001 s-1 and temperature above 1000°C, which should be avoided during hot working. In addition, the instability area enlarged in processing map with the increasing of true strain.
