Abstract

In recent studies, it has been discovered that phonons can carry angular momentum, leading to a series of investigations into systems with 3-fold rotation symmetry. However, for systems with 2-fold screw rotational symmetry, such as $\alpha$-MoO$_3$, there has been no relevant discussion. In this paper, we investigated the pseudoangular momentum of phonons in crystals with 2-fold screw rotational symmetry. Taking $\alpha$-MoO$_3$ as an example, we explain the selection rules in circularly polarized Raman experiments resulting from pseudoangular momentum conservation, providing important guidance for experiments. This study of pseudoangular momentum in $\alpha$-MoO$_3$ opens up a new degree of freedom for its potential applications, expanding into new application domains., Comment: 12 pages,2 figures

Abstract

The APETALA2/ethylene-responsive factor (AP2/ERF) transcription factor family is one of the largest transcription factor families in plants. Its subfamily members activate or repress the expression of related genes by specifically binding to cis-acting elements such as the GCC box and DRE/CRT in the promoter regions of downstream interacting genes, and participate in the regulation of biological processes such as growth and development, maturation and senescence, and stress response in fruits and vegetables. In recent years, an increasing number of studies have shown that AP2/ERF can also regulate the quality and resistance of fruits and vegetables by regulating target genes and inducing various phytohormone responses such as ethylene, jasmonic acid and salicylic acid, or by acting as a response factor in response to phytohormone signals. This article provides an overview of the structural characteristics of AP2/ERF transcription factors and their classification and distribution in fruits and vegetables and reviews the latest research progress in the regulatory effect of AP2/ERF on stress resistance in fruits and vegetables through interaction with the major hormone signals. It is our hope that this review will provide a theoretical basis for enhancing stress resistance in fruits and vegetables from the perspective of hormonal regulation of AP2/ERF transcription factors.

14. A Polysaccharide Isolated from Dictyophora indusiata Promotes Recovery from Antibiotic-Driven Intestinal Dysbiosis and Improves Gut Epithelial Barrier Function in a Mouse Model

Abstract

Despite the tremendous biological activity of polysaccharides from the mushroom Dictyophora indusiata, its role in the restoration of gut microbiota has not yet been explored. The present study aimed to investigate whether D. indusiata polysaccharide (DIP) could modulate the recovery of gut microbiota composition and intestinal barrier function after broad-spectrum antibiotic-driven dysbiosis. Alteration and restoration in the microbial communities were elucidated by the Illumina MiSeq platform. Colon histology, expression of tight-junction associated proteins, and serum/tissue endotoxin and cytokine levels were evaluated. Two-week daily oral administration of clindamycin and metronidazole resulted in reduced bacterial diversity and richness, and perturbed the microbial flora at various taxonomic levels (altered Firmicutes/Bacteroidetes ratio and increased relative abundance of harmful flora (Proteobacteria, Enterococcus, and Bacteroides)), whereas DIP administration reversed the dysbiosis and increased beneficial flora, including Lactobacillaceae (lactic acid-producing bacteria), and Ruminococaceae (butyrate-producing bacteria). In addition, it resulted in the reduction of endotoxemia (through lipopolysaccharides (LPSs)) and pro-inflammatory cytokine (tumor necrosis factor alpha (TNF-α), interleukin 6 (IL-6), and interleukin 1β (IL-1β)) levels, with the increased expression of tight-junction associated proteins (claudin-1, occludin, and zonula occludens-1). These findings not only suggested a comprehensive understanding of the protective effects of a DIP in the restoration of gut microbiota but also highlighted its role in the enhancement of gut barrier integrity, reduction of inflammation and lowering of endotoxin levels in mice.

Abstract

Structured illumination microscopy (SIM) achieves doubled spatial resolution by exciting the specimen with a high-contrast, high-frequency sinusoidal pattern. Such an excitation pattern can be generated by interference between multiple laser beams, which are diffracted from a grating. In SIM, 2D imaging requires 9 patterns and 3D imaging requires 15 patterns. Compared to mechanical movement of gratings, opti-electro devices provide rapid switch of the excitation patterns, in which Digital Micro-mirror Device (DMD) is most common in industry. Here we model DMD as the blazed grating and report a fast and cost-efficient SIM. Our home-built DMD-based laser interference structured illumination microscopy (DMD-ISIM) system reveals the nuclear pore complex and microtubule in mammalian cells with doubled spatial resolution. We further proposed multi-color DMD-ISIM system with simulation, which could potentially exploit the full power of DMD-ISIM.

Abstract

Fluorescence polarization microscopy images both the intensity and orientation of fluorescent dipoles, which plays a vital role in studying the molecular structure and dynamics of bio-complex. However, it is difficult to resolve the dipole assemblies on the subcellular structure and their dynamics in living cells with super-resolution. Here we report polarized structured illumination microscopy (pSIM), which decouples the entangled spatial and angular structured illumination through interpreting the dipoles in spatio-angular hyperspace. We demonstrate its application on a series of biological filamentous systems such as cytoskeleton networks and lambda-DNA, and report the dynamics of short actin sliding through myosin-coated surface. Further, pSIM reveals "side-by-side" organization of the actin ring structure in the membrane-associated periodic skeleton in hippocampal neurons. It also images the dipole dynamics of green fluorescent proteins labeled to the microtubules in live U2OS cells. pSIM can be applied directly to a large variety of commercial or home-built SIM systems., Comment: 18 pages,5 figures, typos fixed
