Abstract Streets are an important component of urban public spaces and also a high-incidence area for urban crime. However, current research mainly involves adult crime, or fails to distinguish between adult and juvenile crime, which poses a severe challenge to the prevention of juvenile delinquency. Juveniles have lower self-control abilities and are more likely to be influenced by external environmental factors to trigger criminal behavior compared to adults. Therefore, this study uses New York’s Manhattan district as an example, based on CPTED and social disorganization theories, and utilizes street view data and deep learning techniques to extract street environment indicators. The GWR model is used to explore the influence mechanism of urban street environment on juvenile crime. The results of this study, considering spatial heterogeneity, demonstrate the impact of various physical environmental indicators of urban streets on juvenile delinquency, and reveal that some street indicators have differentiated effects on crime in different areas of the city. Overall, our research helps to uncover the relationship between juvenile delinquency and the built environment of streets in complex urban settings, providing important references for future urban street design and juvenile delinquency prevention.

Thermal comfort is critical for ensuring the health and productivity of occupants and knowing their thermal demand could help creat a satisfying environment with the least energy waste. Theory of thermal comfort is built based on experimental studies in uniform and stable environments, while there is still a gap when applying in evaluating human thermal comfort in non-uniform thermal environments. Therefore, an insight into human thermal comfort in non-uniform thermal environments is taken. The fundamental studies of thermal comfort in non-uniform thermal environments are explicated, including thermal comfort theory applied in non-uniform thermal environments, types of non-uniform thermal environments and corresponding studies, physiological and psychological thermal responses in the environments. Besides, the evaluation indices of non-uniform thermal environments are classified according to their definitions and the comfort models are reviewed. Finally, future works in this research field are discussed. In general, the overall thermal comfort and local thermal comfort should be both taken into account in non-uniform thermal environments where skin temperatures and psychological thermal responses of occupants are different among local body segments. Moreover, as studies of local thermal comfort are mainly conducted in thermally neutral condition, the limits considering the state deviating thermal neutrality (slightly warm or cool), which is frequently found in reality, are encouraged to be studied. Finally, a well-defined comprehensive index should be proposed considering heat exchange of human body with their microenvironments and thus the comfort range of the index could be provided for designing of non-uniform thermal environments.

Abstract DSS1, essential for BRCA2-RAD51 dependent homologous recombination (HR), associates with the helical domain (HD) and OB fold 1 (OB1) of the BRCA2 DSS1/DNA-binding domain (DBD) which is frequently targeted by cancer-associated pathogenic variants. Herein, we reveal robust ss/dsDNA binding abilities in HD-OB1 subdomains and find that DSS1 shuts down HD-OB1’s DNA binding to enable ssDNA targeting of the BRCA2-RAD51 complex. We show that C-terminal helix mutations of DSS1, including the cancer-associated R57Q mutation, disrupt this DSS1 regulation and permit dsDNA binding of HD-OB1/BRCA2-DBD. Importantly, these DSS1 mutations impair BRCA2/RAD51 ssDNA loading and focus formation and cause decreased HR efficiency, destabilization of stalled forks and R-loop accumulation, and hypersensitize cells to DNA-damaging agents. We propose that DSS1 restrains the intrinsic dsDNA binding of BRCA2-DBD to ensure BRCA2/RAD51 targeting to ssDNA, thereby promoting optimal execution of HR, and potentially replication fork protection and R-loop suppression.

Abstract In the future, novel and highly pathogenic viruses may re-emerge, leading to a surge in healthcare demand. It is essential for urban epidemic control to investigate different cities’ spatiotemporal spread characteristics and medical carrying capacity during the early stages of COVID-19. This study employed textual analysis, mathematical statistics, and spatial analysis methods to examine the situation in six highly affected Chinese cities. The findings reveal that these cities experienced three phases during the initial outbreak of COVID-19: “unknown-origin incubation”, “Wuhan-related outbreak”, and “local exposure outbreak”. Cities with a high number of confirmed cases exhibited a multicore pattern, while those with fewer cases displayed a single-core pattern. The cores were distributed hierarchically in the central built-up areas of cities’ economic, political, or transportation centers. The radii of these cores shrank as the central built-up area’s level decreased, indicating a hierarchical decay and a core–edge structure. It suggests that decentralized built environments (non-clustered economies and populations) are less likely to facilitate large-scale epidemic clusters. Additionally, the deployment of designated hospitals in these cities was consistent with the spatial distribution of the epidemic; however, their carrying capacity requires urgent improvement. Ultimately, the essence of prevention and control is the governance of human activities and the efficient management of limited resources about individuals, places, and materials through leveraging IT and GIS technologies to address supply–demand contradictions.

Abstract Drugs targeting the DNA damage response (DDR) are widely used in cancer therapy, but resistance to these drugs remains a major clinical challenge. Here, we show that SYCP2, a meiotic protein in the synaptonemal complex, is aberrantly and commonly expressed in breast and ovarian cancers and associated with broad resistance to DDR drugs. Mechanistically, SYCP2 enhances the repair of DNA double-strand breaks (DSBs) through transcription-coupled homologous recombination (TC-HR). SYCP2 promotes R-loop formation at DSBs and facilitates RAD51 recruitment independently of BRCA1. SYCP2 loss impairs RAD51 localization, reduces TC-HR, and renders tumors sensitive to PARP and topoisomerase I (TOP1) inhibitors. Furthermore, our studies of two clinical cohorts find that SYCP2 overexpression correlates with breast cancer resistance to antibody-conjugated TOP1 inhibitor and ovarian cancer resistance to platinum treatment. Collectively, our data suggest that SYCP2 confers cancer cell resistance to DNA-damaging agents by stimulating R-loop-mediated DSB repair, offering opportunities to improve DDR therapy.

Abstract Background Cross-species transmission of zoonotic IAVs to humans is potentially widespread and lethal, posing a great threat to human health, and their cross-species transmission mechanism has attracted much attention. miRNAs have been shown to be involved in the regulation of IAVs infection and immunity, however, few studies have focused on the molecular mechanisms underlying miRNAs and mRNAs expression after IAVs cross-species infection. Methods We used tree shrews, a close relative of primates, as a model and used RNA-Seq and bioinformatics tools to analyze the expression profiles of DEMs and DEGs in the nasal turbinate tissue at different time points after the newly emerged swine influenza A virus SW2783 cross-species infection with tree shrews, and miRNA-mRNA interaction maps were constructed and verified by RT-qPCR, miRNA transfection and luciferase reporter assay. Results 14 DEMs were screened based on functional analysis and interaction map, miR-760-3p, miR-449b-2, miR-30e-3p, and miR-429 were involved in the signal transduction process of replication and proliferation after infection, miR-324-3p, miR-1301-1, miR-103-1, miR-134-5p, miR-29a, miR-31, miR-16b, miR-34a, and miR-125b participate in negative feedback regulation of genes related to the immune function of the body to activate the antiviral immune response, and miR-106b-3p may be related to the cross-species infection potential of SW2783, and the expression level of these miRNAs varies in different days after infection. Conclusions The miRNA regulatory networks were constructed and 14 DEMs were identified, some of them can affect the replication and proliferation of viruses by regulating signal transduction, while others can play an antiviral role by regulating the immune response. It indicates that abnormal expression of miRNAs plays a crucial role in the regulation of cross-species IAVs infection, which lays a solid foundation for further exploration of the molecular regulatory mechanism of miRNAs in IAVs cross-species infection and anti-influenza virus targets.

In the development of silicon photonics, the continued downsizing of photonic integrated circuits will further increase the integration density, which augments the functionality of photonic chips. Compared with the traditional design method, inverse design presents a novel approach for achieving compact photonic devices. However, achieving compact, reconfigurable photonic devices with the inverse design that employs the traditional modulation method exemplified by the thermo-optic effect poses a significant challenge due to the weak modulation capability. Low-loss phase change materials (PCMs) exemplified by Sb2Se3 are a promising candidate for solving this problem benefiting from their high refractive index contrast. In this work, we first developed a robust inverse design method to realize reconfigurable silicon and phase-change materials hybrid photonic devices including mode converter and optical switch. The mode converter exhibits a broadband operation of >100 nm. The optical switch shows an extinction ratio of >25 dB and a multilevel switching of 41 (>5 bits) by simply changing the crystallinity of Sb2Se3. Here, we experimentally demonstrated a Sb2Se3/Si hybrid integrated optical switch for the first time, wherein routing can be switched by the phase transition of the whole Sb2Se3. Our work provides an effective solution for the design of photonic devices that is insensitive to fabrication errors, thereby paving the way for high integration density in future photonic chips.

Offshore wind-to-hydrogen production is an effective means of solving the problems of large-scale grid-connected consumption and high power transmission costs of offshore wind power. Site selection is a core component in planning offshore wind-to-hydrogen facilities, involving careful consideration of multiple factors, and is a classic multi-criteria decision-making problem. Therefore, this study proposes a multi-criteria decision-making method based on the two-dimensional linguistic cloud model to optimize site selection for offshore wind-to-hydrogen bases. Firstly, the alternative schemes are evaluated using two-dimensional linguistic information, and a new model for transforming two-dimensional linguistic information into a normal cloud is constructed. Then, the cloud area overlap degree is defined to calculate the interaction factor between decision-makers, and a multi-objective programming model based on maximum deviation-minimum correlation is established. Following this, the Pareto solution of criteria weights is solved using the non-dominated sorting genetic algorithm II, and the alternatives are sorted and selected through the cloud-weighted average operator. Finally, an index system was constructed in terms of resource conditions, planning conditions, external conditions, and other dimensions, and a case study was conducted using the location of offshore wind-to-hydrogen production bases in Shanghai. The method proposed in this study demonstrates strong robustness and can provide a basis for these multi-criteria decision-making problems with solid qualitative characteristics.

The ever-growing demand for miniaturized optical systems presents a significant challenge in revolutionizing their core element – the varifocal lens. Recent advancements in ultra-thin, tunable metasurface optics have introduced new approaches to achieving zoom imaging. However, current varifocal metalens have faced challenges such as low focusing efficiency, limited tunability, and complicated designs. Here, we employ the high-contrast transmit arrays (HCTA) structures to design and fabricate a polarization-independent, single-layer flexible metalens that operates at a wavelength of 940 nm. Using a uniform stretching system, we characterized its optical performance to achieve over 60 % focusing efficiency within a 0 %–25 % stretch range, while the focal length changes align with theoretical predictions. Furthermore, our research also successfully demonstrated the capacity of a metalens with a numerical aperture (NA) of 0.5 to efficiently adjust imaging magnification within a 2× range, achieving imaging results that approach the diffraction limit. This research offers promising prospects for the practical use of compact and miniaturized optoelectronic devices in fields like photography, mixed reality, microscopy, and biomedical imaging.

Purpose: Hydronephrosis is a common pediatric urological condition, characterized by dilation of the renal collecting system. Accurate identification of the severity of hydronephrosis is crucial in clinical management, as high-grade hydronephrosis can cause significant damage to the kidney. In this pilot study, we demonstrate the feasibility of machine learning in differentiating between high and low-grade hydronephrosis in pediatric patients. Materials and Methods: We retrospectively reviewed 592 images from 90 unique patients ages 0–8 years diagnosed with hydronephrosis at the University of Chicago’s Pediatric Urology Clinic. The study included 74 high-grade hydronephrosis (145 images) and 227 low-grade hydronephrosis (447 images). Patients were excluded if they had less than 2 studies prior to surgical intervention or had structural abnormalities. We developed a radiomic-based artificial intelligence algorithm incorporating computerized texture analysis and machine learning (support-vector machine) to yield a predictor of hydronephrosis grade. Results: Receiver operating characteristic analysis of the classifier output yielded an area under the curve value of 0.86 (95% CI 0.81–0.92) in the task of distinguishing between low and high-grade hydronephrosis using a five-fold cross-validation by kidney. Abstract The roles of R-loops and RNA modifications in homologous recombination (HR) and other DNA double-stranded break (DSB) repair pathways remain poorly understood. Here, we find that DNA damage-induced RNA methyl-5-cytosine (m5C) modification in R-loops plays a crucial role to regulate PARP1-mediated poly ADP-ribosylation (PARylation) and the choice of DSB repair pathways at sites of R-loops. Through bisulfite sequencing, we discover that the methyltransferase TRDMT1 preferentially generates m5C after DNA damage in R-loops across the genome. In the absence of m5C, R-loops activate PARP1-mediated PARylation both in vitro and in cells. Concurrently, m5C promotes transcription-coupled HR (TC-HR) while suppressing PARP1-dependent alternative non-homologous end joining (Alt-NHEJ), favoring TC-HR over Alt-NHEJ in transcribed regions as the preferred repair pathway. Importantly, simultaneous disruption of both TC-HR and Alt-NHEJ with TRDMT1 and PARP or Polymerase θ inhibitors prevents alternative DSB repair and exhibits synergistic cytotoxic effects on cancer cells, suggesting an effective strategy to exploit genomic instability in cancer therapy.

High beam quality, large-area output, and small footprint are significant pursuing goals for vertical-cavity surface-emitting lasers (VCSELs), which impose strict requirements on tight light confinements with minimized radiation losses. To achieve this, bound states in the continuum (BICs) have been demonstrated as an effective way of trapping light. Here, we combine BICs and photonic bandgaps to realize a quasi-BIC single-mode photonic crystal (PhC) laser on a colloidal quantum dots (CQDs)/silicon oxide (SiO2) hybrid integrated platform. The PhC cavity is a defect-free hexagonal heterostructure with three regions, and the thin CQDs film is embedded within the SiO2 nanopillar planar array as both an optical gain material and a backbone for the PhC. The mode gaps between different regions provide the lateral confinement while the quasi-BICs near the Γ-point generate the small-divergence vertical radiation coupling, resulting in a well-defined emission concentrating within ±1.85° of the normal surface direction and an optical pumping energy density threshold of 216.75 μJ/cm2. Our results demonstrate the design flexibility and versatility of the quasi-BIC laser even with a low contrast of a refractive index between the PhC slab and the substrate, which has potential applications in cavity quantum electrodynamics, nonlinear optics, and integrated photonics.

Type 2 diabetes mellitus (T2DM) is closely related to the occurrence and progress of Parkinson's disease (PD), in which neurotoxicity caused by hyperglycemia and cerebral insulin resistance may play important roles, and the specific mechanism is not certain. Chronic hyperglycemia may lead to the injury of substantia nigra dopaminergic neuron by promoting oxidative stress, neuroinflammation, the formation of advanced glycation end products and endogenous neurotoxin. Moreover, insulin resistance may be involved in several pathways. This article mainly reviews the mechanism of T2DM increasing the risk of PD and provides guidance for the prevention and treatment of PD in patients with T2DM.

Digital transformation is the engine to promote the high-quality development of the advanced manufacturing industry. This paper constructs a measurement model for the development level of digital transformation in the advanced manufacturing industry and proposes a measurement method for the development level of digital economy and digital transformation in the advanced manufacturing industry. The data are preprocessed through text mining, the vertical and horizontal pull-out grade method is used to determine the weights and calculate the final measurement value, and the time weights are obtained with the help of the nonlinear programming method to realize the measurement of the development level of the digital economy. Use the maximum-minimum value method to process the data related to digital transformation, utilize the entropy value method to calculate the index weights, and calculate the digital transformation development level through the linear weighting function method. The representative provinces of each region are selected to analyze the digital transformation of the advanced manufacturing industry in the east, middle, and west regions of China. The composite index of the digital economy development level in the East, Central, and West is 0.62, 0.45, and 0.37, respectively. The value of the digital transformation development level of Guangdong, Shandong, and Sichuan in 2023 reaches 0.9857, 0.3949, and 0.3747, respectively, ranking in the top three, whereas Jilin and Guizhou are only 0.1479 and 0.1128, and the level of digitalization of China’s advanced manufacturing industry is marked by obvious geographical imbalance.

A dormitory is a place where students live and live for a long time, and the safety of the dormitory can directly affect the quality of students’ studies and lives. In light of this, the study constructs a dynamic security planning system for college dormitory areas, enhancing their security and intelligence through the application of intelligent technology and multi-directional management coordination. Taking the access control system and fire monitoring system in the dormitory security prevention system as an example, based on the neural network perspective, we design the face recognition method of HOG-YOLO and the fire detection algorithm of improved YOLOv5 to realize, train, and test the two models, and evaluate the face recognition performance and fire target detection effect of the models. The results show that the face recognition method established in this paper is highly accurate and rapid, with a recognition accuracy of 95.7%, and it can complete recognition and detection under different facial angles. The fire detection algorithm’s FPS has been improved by 14.75%. The overall accuracy has improved by more than 1%, which has a good effect on fire monitoring. Planning the security system for the dormitory areas in higher education is a significant area of research for ensuring campus security, and access control and fire monitoring systems have significant application potential.

Summary: Fanconi anemia (FA) is characterized by congenital abnormalities, bone marrow failure, and cancer susceptibility. The central FA protein complex FANCI/FANCD2 (ID2) is activated by monoubiquitination and recruits DNA repair proteins for interstrand crosslink (ICL) repair and replication fork protection. Defects in the FA pathway lead to R-loop accumulation, which contributes to genomic instability. Here, we report that the splicing factor SRSF1 and FANCD2 interact physically and act together to suppress R-loop formation via mRNA export regulation. We show that SRSF1 stimulates FANCD2 monoubiquitination in an RNA-dependent fashion. In turn, FANCD2 monoubiquitination proves crucial for the assembly of the SRSF1-NXF1 nuclear export complex and mRNA export. Importantly, several SRSF1 cancer-associated mutants fail to interact with FANCD2, leading to inefficient FANCD2 monoubiquitination, decreased mRNA export, and R-loop accumulation. We propose a model wherein SRSF1 and FANCD2 interaction links DNA damage response to the avoidance of pathogenic R-loops via regulation of mRNA export.

With the rapid development of a social economy, the yearly increase in air conditioning load in the winter and summer seasons may bring serious challenges to the safe and economic operation of the power grid during the peak period of electricity consumption. So, how we reasonably adjust the set temperature of air conditioning so as to cut down the load during peak periods is very important. In this paper, considering the thermal inertia of air-conditioned buildings and the adaptability of human thermal comfort to temperature changes, the air conditioning load is regarded as virtual energy storage, the air conditioning temperature adjustment range for different users is determined based on the adaptive thermal comfort model of different geographic locations and climatic conditions, and a compensation mechanism is set up based on air conditioning users’ level of participation. Then, an optimal scheduling strategy for a microgrid was constructed with the objectives of user satisfaction, carbon emissions, and microgrid operation benefits, as well as regulating the users’ electricity consumption behavior, and the strategy was solved by using a multi-objective JAYA algorithm. Finally, winter and summer are used as case studies to analyze the results, which demonstrate that regulating the virtual energy storage of air conditioning can effectively improve the economy and environmental friendliness of a microgrid operation and reduce the cost of electricity consumption for the users, taking into account the comfort of the users.

Abstract Cerium manganese bimetallic catalysts have become the focus of current research because of their excellent catalytic performance for soot combustion. Two series of cerium manganese catalysts (Na-free catalysts and Na-containing catalysts) were prepared by coprecipitation method and characterized using XRD, N2 adsorption–desorption, SEM, Raman, XPS, H2-TPR, O2-TPD, Soot-TPR-MS and in situ IR. The effects of abundant oxygen vacancies and surface highly dispersed MnO x on soot catalytic combustion of cerium manganese catalysts prepared by different precipitants were analyzed. The activity test results show that the active oxygen species released by a large number of oxygen vacancies in the cerium manganese catalyst are more favorable to the soot catalytic combustion than MnO x which is highly dispersed on the surface of the catalyst and has good redox performance at low temperature. Because the catalytic effect of MnO x on the surface of Na-free catalysts is more dependent on the contact condition between the catalyst and the soot, this phenomenon can be observed more easily under the loose contact condition than under the tight contact condition. The activity cycle test results show that these two series of catalysts show good stability and repeated use will hardly cause any deactivation of the catalysts.

Objective To investigate the effect of nursing program in reducing the rate of unplanned extubation (UEX)of nasogastric tube. Methods Totally 80 patients with placement of nasogastric feeding tube before implement of nursing program(Dec, 2019 to Dec, 2020) were included as the control group, and another 84 patients with placement of nasogastric feeding tube after implement of nursing program(Jan, 2021 to Jan, 2022) were recruited as the observation group. The potential reasons of UEX were retrospectively analyzed, and a standard procedure of maintenance of nasogastric feeding tube was established. The rate of UEX was observed and compared between two groups. Results No case of UEX was reported in the observation group, and 4 cases(7. 49‰) of UEX were observed in the control group, there was significant difference in the rate of UEX between two groups (P＜0. 05). Nurses had achieved a higher score of theory test and practical assessment after implementation of nursing program(P＜0. 05). There was no significant difference in the indwelling time of nasogastric feeding tube between two groups(P＞0. 05). Conclusion The nursing program is potentially effective to reduce the UEX rate of nasogastric feeding tube, ensure the safety of patients, improve the quality control and comprehensive ability of nurses. (目的 探讨护理专案模式在降低胃管非计划拔管(UEX)发生率中的应用效果。方法 将2019 年12月—2020 年 12月护理专案实施前收治的胃管留置患者80例设为对照组, 2021年1 月—2022年 1月护理专案实施后收治的胃管留置患者84例设为观察组。运用护理专案改善方法分析胃管非计划脱管的原因, 制定标准化方案。比较两组胃管UEX发生率。结果 观察组未发生胃管UEX, 对照组发生胃管UEX4(7. 49‰)例, 两组比较差异有统计学意义(P＜0. 05)。护理专案模式实施后, 护士胃管管理理论和操作成绩较实施前提高, 差异有统计学意义(P＜0. 05)。两组食道异物患者和喉癌患者的胃管留置时间比较, 差异无统计学意义(P＞0. 05)。结论 护理专案模式的应用可降低住院患者胃管UEX发生率, 保障患者安全, 提高科室质量管理水平, 有助于护士综合素质和科室凝聚力的提升。)

In order to explore the influence of high temperature in deep well on coal dust moisture evaporation characteristics, evaporation tests were conducted with four coal dust samples in the temperature of 25 ℃, 30 ℃, 35 ℃, 40 ℃, 45℃ and 50 ℃, respectively. The effects of temperature on the evaporation characteristics of the moisture in coal dust were obtained by analyzing the evaporation parameters of four coal dust samples, mainly including evaporation rate, the maximum water loss and evaporation balance time. The results show that the evaporation process of coal dust moisture can be divided into two stages, i.e., the evaporation water loss stage and water loss balance stage; with the increasing of temperature, the evaporation rate and the maximum water loss rate of coal dust increased linearly, and the evaporation balance time decreased in a logarithmic trend; after the temperature increased from 25 ℃ to 50 ℃, The average evaporation rate of coal dust in 4 groups increased by 2 times, the maximum water loss rate increased by 24.57%, and the evaporation balance time was reduced from over 200 min to about 80 min with an average decrease of 63.3%.

Acute myocardial infarction(AMI) is a serious threat to human health due to its high morbidity and mortality. Persistent myocardial injury remains after AMI, although the mortality of AMI has been greatly reduced by drugs, interventional therapy and surgery. Astragaloside IV(AST) is one of the main active components of Astragali Radix. However, the hydrophobicity of AST limits its bioavailability. In this experiment, we developed a platelet membrane-modified poly(d, L-lactic-Co-Glycolic Acid) -block(PLGA-b) -poly(Ethylene Glycol)(PEG) Triphenylphosphonium(TPP)(PLGA-b-PEG-TPP) polymer nanomicelles for encapsulating AST to achieve the dual targeting effect of ' heart-mitochondria ' from the ' organ-organelle ' level. In this study, the drugloaded nanomicelles were characterized by transmission electron microscope, dynamic light scattering, drug release and stability investigation. The cardiac targeting effect of drug loaded nanomicelles was confirmed by in vivo imaging. The effects of drug-loaded nanomicelles on cardiac function and myocardial injury after I/R were investigated by echocardiography, histology and serum enzymology. The effects of drug-loaded nanomicelles on cardiac mitochondrial function and inflammation were evaluated by examining mitochondrial morphology, membrane potential, reactive oxygen species(ROS)and levels of inflammatory factors in serum and heart. Compared with the free AST, the results showed that drug-loaded nanomicelles significantly improved cardiac function, myocardial injury, mitochondrial damage and cardiac inflammation after I/R . It was suggested that the drug loaded nanomicelles significantly improved the bioavailability and cardioprotective activity of AST. This experiment provides a new strategy for the delivery of hydrophobic active monomer of traditional Chinese medicine.

Mounting evidence supports that long-term exposure to fine particle pollutants (PM2.5) is closely implicated in cardiovascular diseases, especially atherosclerosis. Amygdalin is reported to attenuate external stimuli-induced cardiovascular diseases. However, the underlying mechanisms are still not understood. In this study, we aim to explore the protective effects of amygdalin on PM2.5-induced human umbilical vein endothelial cell (HUVEC) injury and unravel the specific mechanisms by MTT, DCFH-DA, biochemical, immunofluorescence, ELISA, RT-qPCR, flow cytometry, TUNEL and western blot analysis. The results reveal that amygdalin reverses PM2.5-induced cytotoxicity and attenuates intracellular ROS production. Moreover, amygdalin increases the levels of SOD and GSH and alleviates the MDA content. Additionally, amygdalin causes a decline of IL-6, IL-1β, TNF-α and COX-2 levels. Moreover, amygdalin inhibits NF-κB p50 and TLR4 protein expressions and NF-κB p65 nuclear translocation. Concomitantly, a decline of phospho-NF-κB p65/NF-κB p65 and phospho-IκB-α/IκB-α is detected. Meanwhile, amygdalin pretreatment reduces HUVEC apoptosis. In addition, amygdalin triggers an upregulation of Bcl-2 and a downregulation of Bax after stimulation with PM2.5. Collectively, these results suggest that amygdalin suppresses PM2.5-induced HUVEC injury by regulating the TLR4/NF-κB and Bcl-2/Bax signaling pathways, indicating that amygdalin may be a novel target for atherosclerosis treatments.

Time-related cognitive function refers to the capacity of the brain to store, extract, and process specific information. Previous studies demonstrated that the cerebellar cortex participates in advanced cognitive functions, but the role of the cerebellar cortex in cognitive functions is unclear. We established a behavioral model using classical eyeblink conditioning to study the role of the cerebellar cortex in associative learning and memory and the underlying mechanisms. We performed an investigation to determine whether eyeblink conditioning could be established by placing the stimulating electrode in the middle cerebellar peduncle. Behavior training was performed using a microcurrent pulse as a conditioned stimulus to stimulate the middle cerebellar peduncle and corneal blow as an unconditioned stimulus. After 10 consecutive days of training, a conditioned response was successfully achieved in the Delay, Trace-200-ms, and Trace-300-ms groups of guinea pigs, with acquisition rates of >60%, but the Trace-400-ms and control groups did not achieve a conditioned stimulus-related blink conditioned response. It could be a good model for studying the function of the cerebellum during the establishment of eyeblink conditioning.

AimsTo verify whether the oral insulin N11005 is administered as a prandial insulin by assessing the pharmacokinetics (PK), pharmacodynamics (PD), and safety profiles of N11005 with a short-acting biosynthetic human insulin (Novolin R) as reference.MethodsThis was a randomized, open-label, single-dose, crossover hyperinsulinemic-euglycemic clamp study in healthy Chinese male subjects. A total of 12 subjects were enrolled in the test (T) group (N11005, 300 IU, p.o.) and the reference (R) group (Novolin R, 0.1 IU/Kg, i.h.) with a washout period of 14 days. All subjects were administered on the same day of the clamp study. Glucose Infusion Rates (GIR), serum insulin, and C-peptide concentration were determined during every 8-hour clamp cycle. Trial registration: Clinicaltrials.gov identifier NCT04975022.ResultsAfter administration, the ratios of mean serum C-peptide concentration to baseline concentration in both T and R groups were lower than 50%, which confirmed the stability of the clamp platform. T group (N11005) showed a more rapid onset of action (tGIR10%max≈11 min) and a comparable duration of action to the R group, which was basically in line with the characteristics of prandial insulins. No adverse events (AEs) occurred throughout the study, which demonstrated that N11005 and Novolin R are safe and well-tolerated.ConclusionsThe PD profiles of the single-dose N11005 in the human body are similar to those of prandial insulins, with an excellent safety profile.Clinical trial registrationClinicaltrials.gov, identifier NCT04975022.

BackgroundSystemic inflammation is associated with the prognosis of colorectal cancer (CRC). The current study aimed to construct a comprehensively inflammatory prognostic scoring system named risk score (RS) based on eosinophil- and basophil-related markers and assess its prognostic value in patients with stage II and stage III CRC.Patients and methodsA total of 3,986 patients were enrolled from January 2007 to December 2013. The last follow-up time was January 2019. They were randomly assigned to the training set and testing set in a 3:2 split ratio. Least absolute shrinkage and selection operator (LASSO)–Cox regression analysis was performed to select the optimal prognostic factors in the construction of RS. The Kaplan–Meier curve, time-dependent receiver operating characteristic (ROC), and Cox analysis were used to evaluate the association between RS and overall survival (OS).ResultsIn the training set, all inflammatory markers showed certain prognostic values. Based on LASSO-Cox analysis, nine markers were integrated to construct RS. The Kaplan–Meier curve showed that a higher RS (RS > 0) had a significantly worse prognosis (log-rank p< 0.0001). RS (>0) remained an independent prognostic factor for OS (hazard ratio (HR): 1.70, 95% confidence interval (CI), 1.43–2.03, p< 0.001). The prognostic value of RS was validated in the entire cohort. Time-dependent ROC analysis showed that RS had a stable prognostic effect throughout the follow-up times and could enhance the prognostic ability of the stage by combination. Nomogram was established based on RS and clinicopathological factors for predicting OS in the training set and validated in the testing set. The area under the curve (AUC) values of the 3-year OS in the training and testing sets were 0.748 and 0.720, respectively. The nomogram had a satisfactory predictive accuracy and had better clinical application value than the tumor stage alone.ConclusionsRS might be an independent prognostic factor for OS in patients with stage II and III CRC, which is helpful for risk stratification of patients. Additionally, the nomogram might be used for personalized prediction and might contribute to formulating a better clinical treatment plan.

Rationale and Objectives: Thyroid nodules are a common occurrence and up to 30% are indeterminate on Fine Needle Aspiration (FNA) biopsy. Use of machine learning (ML) algorithms as computer aids, i.e., computer-aided diagnosis (CADx), may provide accessible, accurate methods of diagnosing thyroid cancer on ultrasound (US) images of thyroid nodules. We conducted a systematic review to synthesize the current knowledge and potential of ML in assisting physicians in diagnosing indeterminate thyroid nodules. Materials and Methods: We searched PubMed for studies without a year constraint. Inclusion criteria were as follows: (1) publication of the study as a full-text manuscript in a peer-reviewed journal, (2) original work (e.g., not a review). Studies were excluded if they (1) did not perform FNA biopsy, (2) excluded all indeterminate lesions, (3) focused on a non-cancer thyroid pathology, (4) described only non-diagnostic uses of their model(s) (e.g. calcification quantification), or (5) did not use grayscale ultrasound. We used the Joanna Briggs Institute Appraisal Checklist for Diagnostic Test Accuracy Studies to synthesize and present quality data and PRISMA guidelines for reporting. Results: Our search yielded only 6 articles published between 2013 and 2022. All studies used either only fine-needle aspiration (FNA) biopsy or a combination of FNA and surgery as the reference standard. Of these, one study included only indeterminate nodules, while 5 studies included other nodules in categories as well. Results varied greatly and studies were inconsistent in which performance metrics they reported. Physician performances yielded areas under the ROC curve (AUC) ranging from 0.68 to 0.83, while ML models gave AUCs from 0.666 to 0.954. One study showed that their model had a greater AUC than physicians, while another study showed no difference in AUCs of their ML model and physicians. Another study demonstrated that their ML-assisted approach had a greater AUC (0.917) than either an American College of Radiology (ACR) Thyroid Imaging Reporting & Data System (TI-RADS)-only approach (AUC 0.68) or a ML only approach (AUC 0.77). One study measured the performance of ML models without physician comparison, with AUCs ranging from 0.85 to 0.90. One study found no difference between their ML model and molecular testing (AUC 0.88 and 0.81, respectively). Conclusion: The growing field of CADx and increasing availability of commercial models suggests an increasing interest toward utilization of ML-assisted diagnosis in the clinical setting. However, there is a significant lack of research on the role of ML the diagnosis of indeterminate thyroid nodules. Traditional centralized transactions require a control center for user demand matching, settlement and other processes. However, with the increase in the penetration rate of distributed energy in the community, the explosive increase in the number of transactions leads to a decrease in efficiency and it is difficult to guarantee user privacy and information security. The smart contract technology based on blockchain technology has the characteristics of decentralization, traceability and tamper resistance, and these key factors show unique advantages in distributed energy transactions. This paper explores Ethereum and smart contract technology, designs a peer-to-peer energy sharing mechanism with reward and punishment incentives and establishes a smart contract trading platform for smart community-based virtual power plant (CVPP). This paper verifies the functionality and effectiveness of smart contract. The results show that when the supply and demand ratio changes, the user can conduct energy transactions according to the contract without a third-party organization, which solves the problem of trust between the two parties and achieves the expected effect and runs successfully. In addition, the simulation results show that the peer-to-peer transaction based on smart contracts reduces the energy cost per household and increases the total benefit of CVPP.

Objective To investigate the role of miR-155 in the development and lung metastasis of osteosarcoma, and to explore its target proteins and related mechanisms. Methods We detected differential miR-155 expression in osteosarcoma and its lung metastasis process through relevant database analysis combined with qPCR detection of clinical tissue and cell samples. iTRAQ quantitative proteomics was used to screen the target protein of miR-155 in osteosarcoma and its lung metastasis. The selected miR-155 target protein was verified by Western blot in clinical tissue and cell samples. Results miR-155 expression was significantly higher in osteosarcoma tissue and lung metastatic tumor tissue than that in the control group. Compared with human normal osteoblasts, miR-155 expression in human osteosarcoma cell line was also significantly increased. A total of 3714 proteins were obtained by iTRAQ assay, and 253 differentially-expressed proteins were screened out, 144 of which were up-regulated and 109 were down-regulated. The prediction analysis and experiment verified that C/EBP β was a potential target protein of miR-155. Conclusion miR-155 expression is significantly increased in osteosarcoma and its lung metastasis process, and C/EBP β is a potential target of miR-155.

Background: The clinical application of music therapy and research into its use and effectiveness are common in Western countries. The physiological role of this type of therapy is to stimulate the central nervous system through music, which may have a sedative, analgesic effect, and reduce negative emotions. Previous studies have confirmed that music can be effective for a range of psychological disorders, including post-stroke depression (PSD). There is, however, a lack of systematic evaluation of its effectiveness, and variability in sample size and in the quality of research has detracted from the persuasiveness of findings. Methods: Based on PRISMA 2020, articles on music therapy intervention in post-stroke depression were identified through the Web of Science, PubMed, EMBASE, CNKI, Weipu, and Wanfang databases. The retrieval time was taken from the establishment of the database to October 18, 2022. Two researchers conducted a stringent evaluation of the quality of the articles and extracted the data. They then used RevMan5.3 software for meta-analysis. Results: Twenty articles were listed, involving 1625 patients. Meta-analysis results showed that music therapy could lower scores on the Hamilton Depression Rating Scale (HDRS/Ham-D), the National Institutes of Health stroke scale and self-rated depression scale for patients with PSD. Music therapy was also shown to improve the Barthel Index for Activities of Daily Living and treatment efficacy of PSD patients. However, music therapy did not reduce the incidence of adverse reactions in PSD patients. Conclusion: Music therapy has benefits in improving HDRS/Ham-D score and symptoms of PSD patients, and could be more widely applied.

Objective: To perform a bibliometric analysis of published research related to the use of traditional Chinese medicine (TCM) for the treatment of Coronavirus Disease 2019 (COVID-19). Methods: Research documents related to the use of TCM for prevention and treatment of COVID-19 published up to September 19, 2021, were retrieved from the Web of Science database. Bibliometrix R 4.0 software package was used to analyze data, including countries of publication, research institutions, journals, citations, and keywords. Further analysis was conducted to identify co-occurrence of keywords in the documents, including their titles and abstracts. Cooperative network analyses of authors, institutions, and countries of publication were also conducted. The classification types were statistically analyzed and the research progress of key TCMs was reviewed. Results: A total of 417 documents were included in our analysis. Of these, 85.13% originated in China. Of the 417 documents, 148 (35.5%) were published in journals with impact factors in quartile 1 and 164 (39.3%) in journals in quartile 2. The documents were mainly published in journals categorized as Medicine. The results of network analysis showed close cooperation between institutions and countries. Excluding disease- and drug-related keywords, the top four keywords were ‘Systematic review’, ‘Network pharmacology’, ‘Medicine’ and ‘Molecular docking’. Keyword co-occurrence analysis showed 4 main keywords association groups. Statistical analysis of the TCM studies showed that Lianhua Qingwen capsule, Qingfei Paidu decoction, Shufeng Jiedu capsules and ReDuNing injection were the most studied Chinese medicines. Lianhua Qingwen capsules, Qingfei Paidu decoction, ReDuNing injection, and Shufeng Jiedu capsules were used in clinical, bioinformatics, and basic research. Toujie Quwen granule, Jinhua Qinggan granule, Shuanghuanglian oral liquid, Tanreqing injection, and Xuanfei Baidu decoction were used in clinical and bioinformatics research, although basic research on their mechanisms of action is lacking. Conclusion: Research intensity and recognition, as well as cooperation, in the field of Chinese medicine for the prevention and treatment of COVID-19 has increased. Research types are generally comprehensive, and investigated several TCM formulations that are specifically recommended by Chinese COVID-19 guidelines. However, comprehensive, in-depth research on their molecular mechanisms of action is still lacking. More basic research is thus needed to identify therapeutic mechanisms to standardize and validate the use of TCM in the prevention and treatment of COVID-19.

Abstract Background Lung cancer is the most prevalent cancer, and the leading cause of cancer-related deaths in China. The aim of this study was to estimate the direct medical expenditure incurred for lung cancer care and analyze the trend therein for the period 2002–2011 using nationally representative data in China Methods This study was based on 10-year, multicenter retrospective expenditure data collected from hospital records, covering 15,437 lung cancer patients from 13 provinces diagnosed during the period 2002–2011. All expenditure data were adjusted to 2011 to eliminate the effects of inflation using China’s annual consumer price index. Results The direct medical expenditure for lung cancer care (in 2011) was 39,015 CNY (US$6,041) per case, with an annual growth rate of 7.55% from 2002 to 2011. Drug costs were the highest proportionally in the total medical expenditure (54.27%), followed by treatment expenditure (14.32%) and surgical expenditure (8.10%). Medical expenditures for the disease varied based on region, hospital level, type, and stage. Conclusion The medical expenditure for lung cancer care is substantial in China. Drug costs and laboratory test are the main factors increasing medical costs.

Tb3+ and Yb3+ co-doped Y(PO3)3 upconversion luminescence phosphors were prepared by coprecipitation method. The structure and optical properties of the as-synthesized Y(PO3)3: Tb3+,Yb3+ samples were investigated by XRD, SEM, FI-IR and upconversion luminescence spectrophotometer (UPL), respectively. XRD results show that the as-synthesized samples are Tb3+ and Yb3+ co-doped Y(PO3)3 crystals with monoclinic structure (space group P21/c). UPL results display that the obtained Tb3+ and Yb3+ co-doped Y(PO3)3 samples emit the Tb3+ characteristic upconversion blue-green luminescence under 980 nm excitation. The doping content of Tb3+ ions affects the upconversion luminescence properties of the as-prepared Y(PO3)3: x Tb3+, 20%(mole fraction,the same below) Yb3+ samples. When the doping amount of Tb3+ is 2%-10%, the 5D4→7F6 emission peak of the Y(PO3)3: x Tb3+, 20% Yb3+ samples splits into two emission peaks centered at 481 nm and 491 nm. The dominant upconversion peak is located at 547 nm for the Y(PO3)3: x Tb3+, 20% Yb3+ samples when the Tb3+ doping content is 10%-20%. The concentration quenching can be observed when the doping amount of Tb3+ is over 20%. The doping ratio of Tb3+ to Yb3+ and the excitation power density also exhibit effect on the upconversion luminescence properties of the prepared samples. The upconversion blue-green emission color of the as prepared Tb3+ and Yb3+ co-doped Y(PO3)3 samples can be tuned by changing the doping amount ratio of Tb3+ to Yb3+.The upconversion luminescence mechanism of Y(PO3)3: Tb3+, Yb3+ samples was also discussed. The 5D3→7FJ (J=6,5,4) and 5D4→7FJ (J=6,5,4,3) upconversion luminescence are attributed to a three- and two-photon absorption mechanisms, respectively.

Objective To find out the risk genes related to lymph node metastasis of colon cancer and construct a nomogram model to predict lymph node metastasis. Methods Genome sequencing data were downloaded from TCGA and GEO databases, and candidate genes were screened by differential expressed gene analysis and LASSO regression. AIC was used to determine the optimal nomogram model. ROC curve, calibration curve and Hosmer-Lemeshow test were used to evaluate the accuracy of the model. Decision curve analysis was used to evaluate the clinical utility. Results Eleven genes which could effectively predict lymph node metastasis of colon cancer were obtained through LASSO regression. According to the results of stepwise regression, the model composed of age, pathological T stage, TH, CDH4, PNMA6A, TNNC1, KIR2DL4, STUM and SFTA2 had the minimum AIC value (440.4). The AUC value of internal evaluation was 0.800, and that of external verification was 0.664. In model evaluation, the calibration and Hosmer-Lemeshow test showed favorable performance. Decision curve analysis showed nomogram model could bring clinical benefits for predicting lymph nodes metastasis. Conclusion Eleven risk genes of lymph node metastasis of colon cancer are selected and a nomogram model is constructed. The model has favorable performance in discriminative and calibration abilities to help evaluate the status of lymph node metastasis of colon cancer patients.

Circadian genes are a group of genes in cells that can modulate circadian rhythm by self-expression and regulation of self-excited oscillations. These genes can maintain and regulate many physiological and biochemical activities of the human body to operate according to a certain periodic rule and order, so as to be better adapt to the surrounding environment. Period1 (Per1) gene is an important circadian rhythm gene and is at the core of circadian rhythm system. Studies have shown that the abnormal expression of Per1 and rhythm change are highly correlated with the occurrence and development of a variety of malignant tumors, but the mechanism is not fully clear. This article will review the effect of Per1 on the invasion, metastasis and apoptosis of malignant tumors and the related mechanism.

Post-translational modifications of proteins at DNA damage sites can facilitate the recruitment of DNA repair factors. Here, the authors show that mRNA is locally modified with m5C at sites of DNA damage by the RNA methyltransferase TRDMT1 to promote homologous recombination repair.

Background We aimed to obtain a set of health state utility scores of patients with esophageal cancer (EC) and precancerous lesions in China, and to explore the influencing factors of health‐related quality of life (HRQoL). Methods A hospital‐based multicenter cross‐sectional study was conducted. From 2013 to 2014, patients with EC or precancerous lesions were enrolled. HRQoL was assessed using a European quality of life‐5 dimension (EQ‐5D‐3L) instrument. Multivariable linear regression analysis was performed to explore the influencing factors of the EQ‐5D utility scores. Results A total of 2090 EC patients and 156 precancer patients were included in the study. The dimension of pain/discomfort had the highest rate of self‐reported problems, 60.5% in EC and 51.3% in precancer patients. The mean visual analog scale (VAS) score for EC and precancer patients were 68.4 ± 0.7 and 64.5 ± 3.1, respectively. The EQ‐5D utility scores for EC and precancer patients were estimated as 0.748 ± 0.009 and 0.852 ± 0.022, and the scores of EC at stage I, stage II, stage III, and stage IV were 0.693 ± 0.031, 0.747 ± 0.014, 0.762 ± 0.015, and 0.750 ± 0.023, respectively. According to the multivariable analyses, the factors of region, occupation, household income in 2012, health care insurance type, pathological type, type of therapy, and time points of the survey were statistically associated with the EQ‐5D utility scores of EC patients. Conclusions There were remarkable decrements of utility scores among esophageal cancer patients, compared with precancer patients. The specific utility scores of EC would support further cost‐utility analysis in populations in China.

Abstract Telomeric DNA are TTAGGG tandem repeats, which are susceptible for oxidative DNA damage and hotspot regions for formation of DNA secondary structures such as t-loop, D-loop, G-quadruplexes (G4), and R-loop. In the past two decades, unique DNA or RNA secondary structures at telomeres or some specific regions of genome have become promising therapeutic targets. G-quadruplex and R-loops at telomeres or transcribed regions of genome have been considered as the potential targets for cancer therapy. Here we discuss the potentials to target the secondary structures (G4s and R-loops) in genome as therapy approaches.

In recent years, the dramatic increase of the prices of Pd and Rh brings about increasingly heavier cost pressure for the exhaust gas after-treatment industry, thus it is of great significance to develop advanced Pd-Rh bimetallic catalysts with low metal loading. In this work, on the basis of conventional catalyst composed of Al2O3-supported Pd and CZ (ceria-zirconia-based oxide)-supported Rh, an advanced Pd-Rh catalyst was obtained by adjusting the synthesis procedure. That was, a small portion of Rh was co-impregnated with Pd on Al2O3, while the other portion was still supported on CZ. In this way, the co-impregnated Rh may act as an auxiliary for Pd/Al2O3 component. As revealed by the various characterization results, the agglomeration of Pd species is well suppressed, and larger amount of PdO species is maintained for the modified catalyst after hydrothermal aging treatment. On the other hand, the state of Rh species also appears to be somewhat modified. Consequently, after hydrothermal aging treatment, the modified catalyst behaves much better for the catalytic conversions of CO, HC and NO.

Objectives To determine the incidence and intensity of household impoverishment induced by cancer treatment in China.Design Average income and daily consumption per capita of the households and out-of-pocket payments for cancer care were estimated. Household impoverishment was determined by comparing per capita daily consumption against the Chinese poverty line (CPL, US$1.2) and the World Bank poverty line (WBPL, US$1.9) for 2015. Both pre-treatment and post-treatment consumptions were calculated assuming that the households would divert daily consumption money to pay for cancer treatment.Participants Cancer patients diagnosed initially from 1 January 2015 to 31 December 2016 who had received cancer treatment subsequently. Those with multiple cancer diagnoses were excluded.Data sources A household questionnaire survey was conducted on 2534 cancer patients selected from nine hospitals in seven provinces through two-stage cluster/convenience sampling.Findings 5.89% (CPL) to 12.94% (WBPL) households were impoverished after paying for cancer treatment. Abstract Background Quantitative ultrasound has been used for the assessment of cancellous bone status. The attenuation mechanisms of cancellous bone, however, have not been well understood, because the microstructure of cancellous bone is significantly inhomogeneous and the interaction between ultrasound and the microstructure of cancellous bone is complex. In this study, a theoretical approach was applied to investigate the influence of the microstructure of cancellous bone on ultrasonic attenuation. Results The scattering from a trabecular cylinder was significantly angle dependent. The dependencies of the ultrasonic attenuation on frequency, scatterer size, and porosity were explored from the theoretical calculation. Prediction results showed that the ultrasonic attenuation increased with the increase of frequency and decreased linearly with the increase in porosity, and the broadband ultrasound attenuation decreased with the increase in porosity. All these predicted trends were consistent with published experimental data. In addition, our model successfully explained the principle of broadband ultrasound attenuation measurement (i.e., the attenuation over the frequency range 0.3–0.65 MHz was approximately linearly proportional to frequency) by considering the contributions of scattering and absorption to attenuation. Conclusion The proposed theoretical model may be a potentially valuable tool for understanding the interaction of ultrasound with cancellous bone.

Tumor tissues are often composed of not only tumor cells but also non-tumor cells such as immune cells, stromal cells and mesenchymal cells. They play an important role in tumor growth and progress together. Tumor purity is significantly associated with clinical features, gene expression and tumor biology, indicating that we need to fully consider the potential confounding effect of tumor purity on tumor analyses. This article reviews the concept, calculate methods and impact of tumor purity on genomic analysis.

Background: Spontaneous intracerebral hemorrhage (ICH) is a devastating disease with high mortality rate. This study aimed to predict hematoma expansion in spontaneous ICH from routinely available variables by using support vector machine (SVM) method. Methods: We retrospectively reviewed 1157 patients with spontaneous ICH who underwent initial computed tomography (CT) scan within 6 h and follow-up CT scan within 72 h from symptom onset in our hospital between September 2013 and August 2018. Hematoma region was manually segmented at each slice to guarantee the measurement accuracy of hematoma volume. Hematoma expansion was defined as a proportional increase of hematoma volume > 33% or an absolute growth of hematoma volume > 6 mL from initial CT scan to follow-up CT scan. Univariate and multivariate analyses were performed to assess the association between clinical variables and hematoma expansion. SVM machine learning model was developed to predict hematoma expansion. Findings: 246 of 1157 (21.3%) patients experienced hematoma expansion. Multivariate analyses revealed the following 6 independent factors associated with hematoma expansion: male patient (odds ratio [OR] = 1.82), time to initial CT scan (OR = 0.73), Glasgow Coma Scale (OR = 0.86), fibrinogen level (OR = 0.72), black hole sign (OR = 2.52), and blend sign (OR = 4.03). The SVM model achieved a mean sensitivity of 81.3%, specificity of 84.8%, overall accuracy of 83.3%, and area under receiver operating characteristic curve (AUC) of 0.89 in prediction of hematoma expansion. Interpretation: The designed SVM model presented good performance in predicting hematoma expansion from routinely available variables. The artificial intelligence calculation method can effectively solve various nonlinear mapping relationships. The strength of these nonlinear solvers is exploited for the evaluation of power grid investment risk using back propagation (BP) neural network optimized by genetic algorithm. The mathematical model of the problem is constructed by selecting the transfer function of the neural network and defining the fitness function of genetic algorithm. BP neural network has good ability of self-learning, self-adaptation and generalization, which can overcome the drawbacks of traditional evaluation methods relying on experts' experience. For the characteristics of genetic algorithm global optimization, the genetic algorithm is used to optimize the weight and threshold of BP neural network, and BP neural network is trained to obtain the optimal evaluation model. The model fully exploits the local search ability of BP neural network and the global search ability of genetic algorithm. It has obtained good evaluation accuracy for the processing of multi-dimensional influence factor problem. And the model can be adapted to different power grids by changing the training data. However, the method cannot describe the specific relationship between each impact factor and the investment risk of the grid. The case study shows that the method can accurately and effectively evaluate power grid investment risk and improve the fault tolerance of the power grid investment risk evaluation.

Abstract Background Expression patterns of many laticifer-specific gens are closely correlative with rubber yield of Hevea brasiliensis (para rubber tree). To unveil the mechanisms underlying the rubber yield, transcript levels of nine major latex metabolism-related genes, i.e., HMG-CoA synthase (HMGS), HMG-CoA reductase (HMGR), diphosphomevalonate decarboxylase (PMD), farnesyl diphosphate synthase (FPS), cis-prenyltransferase (CPT), rubber elongation factor (REF), small rubber particle protein (SRPP), dihydroxyacid dehydratase (DHAD) and actin depolymerizing factor (ADF), were dertermined, and the relationship between rubber yield with their expression levels was analysed. Results Except HbHMGR1, HbPMD and HbDHAD, most of these genes were predominantly expressed in latex, and bark tapping markedly elevated the transcript abundance of the analyzed genes, with the 7th tapping producing the greatest expression levels. Both ethephon (ETH) and methyl jasmonate (MeJA) stimulation greatly induced the expression levels of the examined genes, at least at one time point, except HbDHAD, which was unresponsive to MeJA. The genes’ expression levels, as well as the rubber yields and two yield characteristics differed significantly among the different genotypes examined. Additionally, the latex and dry rubber yields increased gradually but the dry rubber content did not. Rubber yields and/or yield characteristics were significantly positively correlated with HbCPT, HbFPS, HbHMGS, HbHMGR1 and HbDHAD expression levels, negatively correlated with that of HbREF, but not significantly correlated with HbPMD, HbSRPP and HbADF expression levels. In addition, during rubber production, significantly positive correlations existed between the expression level of HbPMD and the levels of HbREF and HbHMGR1, between HbSRPP and the levels of HbHMGS and HbHMGR1, and between HbADF and HbFPS. Conclusions The up-regulation of these genes might be related to the latex production of rubber trees under the stress of bark tapping and latex metabolism. The various correlations among the genes implied that there are differences in their synergic interactions. Thus, these nine genes might be related to rubber yield and yield-related traits in H. brasiliensis, and this work increases our understanding of their complex functions and how they are expressed in both high-and medium-yield rubber tree varieties and low-yield wild rubber tree germplasm.
