17. Long-Term Stable and Multifeature Microfluidic Impedance Flow Cytometry Based on a Constricted Channel for Single-Cell Mechanical Phenotyping

Abstract

The microfluidic impedance flow cytometer (m-IFC) using constricted microchannels is an appealing choice for the high-throughput measurement of single-cell mechanical properties. However, channels smaller than the cells are susceptible to irreversible blockage, extremely affecting the stability of the system and the throughput. Meanwhile, the common practice of extracting a single quantitative index, i.e., total cell passage time, through the constricted part is inadequate to decipher the complex mechanical properties of individual cells. Herein, this study presents a long-term stable and multifeature m-IFC based on a constricted channel for single-cell mechanical phenotyping. The blockage problem is effectively overcome by adding tiny xanthan gum (XG) polymers. The cells can pass through the constricted channel at a flow rate of 500 μL/h without clogging, exhibiting high throughput (∼240 samples per second) and long-term stability (∼2 h). Moreover, six detection regions were implemented to capture the multiple features related to the whole process of a single cell passing through the long-constricted channel, e.g., creep, friction, and relaxation stages. To verify the performance of the multifeature m-IFC, cells treated with perturbations of microtubules and microfilaments within the cytoskeleton were detected, respectively. It suggests that the extracted features provide more comprehensive clues for single-cell analysis in structural and mechanical transformation. Overall, our proposed multifeature m-IFC exhibits the advantages of nonclogging and high throughput, which can be extended to other cell types for nondestructive and real-time mechanical phenotyping in cost-effective applications.

Abstract

Determination of the accurate astrophysical S factor of {$^{12}$C($\alpha,\gamma$)$^{16}$O} reaction has been regarded as a holy grail of nuclear astrophysics for decades. In current stellar models, a knowledge of that value to better than 10\% is desirable. Due to the practical issues, tremendous experimental and theoretical efforts over nearly 50 years are not able to reach this goal, and the published values contradicted with each other strongly and their uncertainties are 2 times larger than the required precision. To this end we have developed a Reduced R-matrix Theory, based on the classical R-matrix theory of Lane and Thomas, which treats primary transitions to ground state and four bound states as the independent reaction channels in the channel spin representation. With the coordination of covariance statistics and error propagation theory, a global fitting for almost all available experimental data of $^{16}$O system has been multi-iteratively analyzed by our powerful code. A reliable, accurate and self-consistent astrophysical S factor of {$^{12}$C($\alpha,\gamma$)$^{16}$O} was obtained with a recommended value $S_{tot}$ (300) = 162.7 $\pm$ 7.3 keV b (4.5\%) which could meet the required precision., Comment: 23 pages, 44 figures, 6 tables

Abstract

Using the geological modeling software, based on the production data of 138 CBM wells in No.3 coal seam of western Sihe minefield from 2007 to 2017, the production capacity of the study area can be divided into three zones: A, B and C. The plane and profile characteristics of coal structure and the temporal and spatial variation characteristics of productivity were compared and analyzed. The results show that the study area is mainly composed of primary structure and contains a small amount of structure coal, It basically contains no broken grain structure coal and mylonite structure coal. Regional distribution of broken structure coal content is C>B>A. Take C as the center, the broken structure coal content gradually decreases to zone A and zone B. For the capacity distribution, the average daily CBM production is A>B>C. Most wells with good CBM production capacity are distributed in the A and B zones. The C-zone has a large distribution of production-attenuated CBM wells. Although the content of broken structure coal content has a great contribution to the early production capacity of CBM wells, it is bad to the sustainability of production capacity. In the C zone with broken structure coal content of about 18%, the early gas production is higher and the decay is faster.
