Abstract

The sensitivity of direct terahertz detectors based on self-mixing of terahertz electromagnetic wave in field-effect transistors is being improved with noise-equivalent power close to that of Schottky-barrier-diode detectors. Here we report such detectors based on AlGaN/GaN two-dimensional electron gas at 77~K are able to sense broadband and incoherent terahertz radiation. The measured photocurrent as a function of the gate voltage agrees well with the self-mixing model and the spectral response is mainly determined by the antenna. A Fourier-transform spectrometer equipped with detectors designed for 340, 650 and 900~GHz bands allows for terahertz spectroscopy in a frequency range from 0.1 to 2.0~THz. The 900~GHz detector at 77~K offers an optical sensitivity about $1~\mathrm{pW/\sqrt{Hz}}$ being comparable to a commercial silicon bolometer at 4.2~K. By further improving the sensitivity, room-temperature detectors would find applications in active/passive terahertz imaging and terahertz spectroscopy., Comment: 4.5 pages, 5 figures

Abstract

The advance of terahertz science and technology yet lays wait for the breakthrough in high-efficiency and high-power solid-state terahertz sources applicable at room temperature. Plasmon in two-dimensional electron gas (2DEG) has long been pursued as a type of promising active medium for terahertz emitters. However, a high wall-plug efficiency sufficient for high-power operation has not been achieved. Here we report spontaneous terahertz emission tunable in a frequency range from 0.3 to 2.5 THz through electrical excitation of plasmon or plasmon-polariton modes in a grating-gate coupled AlGaN/GaN 2DEG at 7 K. A wall-plug efficiency ranging from $10^{-6}$ to $10^{-1}$ is found by tuning the energy (4.5 eV to 0.5 eV) of tunneling electrons between the grating gates and the 2DEG. The total emission power, albeit below 15 nW, is already detectable by using a direct terahertz field-effect transistor detector based on the same type of 2DEG. The findings indicate that both high-efficiency and high-power terahertz plasmon-polariton emission could be obtained by a proper injection of tunneling electrons with low energy and high current density., Comment: 4.5 pages, 4 figures

27. Silencing NUPR1 down-regulates autophagy and promotes migration and invasion in human multiple myeloma U266 cells

Abstract

Objective To explore the effect of silencing nuclear protein 1 (NUPR1) on autophagy, migration and invasion in human multiple myeloma (MM) U266 cells and its possible mechanism. Methods NUPR1-shRNA was transfected into MM U266 cells, and the transfection rate was detected by flow cytometry. Then the cells were divided into negative control cells (transfected with unrelated sequence RNA lentiviral vector) and NUPR1 knockdown cells (transfected with NUPR1-shRNA lentiviral vector). The interference effect of NUPR1 was detected by qRT-PCR, and the expression levels of autophagy related proteins (Beclin1, ATG5, LC3Ⅱ/LC3Ⅰ, P62), migration and invasion associated proteins (MMP9, CXCR4) and pathway proteins (p-AKT/T-AKT, p-mTOR/T-mTOR) were measured by Western blotting. Transmission electron microscopy (TEM) was used to observe autophagosomes. Transwell test was employed to detect cell migration and invasion. Results The expression of NUPR1 at mRNA and protein level was significantly decreased in NUPR1 knockdown cells (P < 0.05). The protein levels of autophagy related proteins, ATG5, Beclin1 and LC3Ⅱ/LC3Ⅰwere significantly lower, while those of f MMP9, CXCR4, P62, p-AKT/T-AKT and p-mTOR/T-mTOR were increased obviously in the NUPR1 knockdown cells than the negative control cells (P < 0.05). There were significantly reduced autophagosomes in NUPR1 knockdown cells. While, migratory and invasive abilities were enhanced in NUPR1 knock down group, and rapamycin reduced the promoting effects. Conclusion Silencing NUPR1 may promotes migration and invasion by down-regulating autophagy in U266 cells, which may be related to activation of the AKT/mTOR pathway.

Abstract

Introduction: Acquired aplastic anemia (AA), a heterogeneous bone marrow (BM) failure disease, is mainly mediated by the immune destruction of hematopoietic stem cells (HSCs). Given the predominant role of immunosuppressive therapy (IST) in AA, it is sensible to theorize that variants of cytokine genes might affect the outcome of IST. Methods: In this study, we analyzed three single nucleotide polymorphisms (SNPs) of interleukin (IL)‐10 gene in promoter region to clarify their relationship with susceptibility, clinical efficacy and prognosis of AA. Results: We observed that CT genotype of IL‐10 rs1800896 was associated with a decreased risk of AA (adjusted OR = 0.541 [95% CI 0.295–0.993], p =.047). Besides, the disease severity differed considerably by IL‐10 gene promoter genotypes and alleles. Furthermore, IL‐10 SNPs influenced efficacy of IST, with unfavorable response exhibited by rs1800871 and rs1800872 in dominant models (GG + AG vs. AA, adjusted OR = 0.409 [95% CI 0.178–0.943, p =.036] for rs1800871 and GG + GT vs. TT, adjusted OR = 0.396 [95% CI 0.173–0.909, p =.028] for rs1800872, respectively). Conclusion: The polymorphisms of IL‐10 promoter region were informatively genetic risk factors which might be conducive to the insights into the mechanisms of AA and the design of individual regimens.
