5. Distinguishing Between Internal and External Defects Based on Stepped Array Sensor in Thick-Walled Ferromagnetic Components

Abstract

Distinguishing between internal and external defects is extremely important for defect evaluation, but it is difficult for traditional methods to distinguish between internal and external defects in thick-walled ferromagnetic components effectively. In this article, a method based on a stepped array sensor is proposed for distinguishing internal and external defects in thick-walled ferromagnetic components. Theoretical modeling and simulation analysis show that the magnetic permeability perturbation testing (MPPT) signals of internal and external defects show different trends as the sensor size increases. Experiments show that the method can distinguish between $1 \times 1$ mm internal and external defects in a ferromagnetic plate with a thickness of 12 mm. In addition, this method has shown good performance when applied to ferromagnetic components of different thicknesses (including 8, 10, and 12 mm), and the magnetization state has no effect on the change trend. It is worth noting that it can distinguish defects with a depth of 8.3% of the total plate thickness, ensuring accurate and reliable detection. This method has no need for prior knowledge of the parameters of the ferromagnetic components and the optimum magnetization strength, which is of greater practical value.

Abstract

There is a critical unmet need for safe and efficacious neoadjuvant treatment for cisplatin-ineligible patients with muscle-invasive bladder cancer. Here we launched a phase 1b study using the combination of intravesical cretostimogene grenadenorepvec (oncolytic serotype 5 adenovirus encoding granulocyte–macrophage colony-stimulating factor) with systemic nivolumab in cisplatin-ineligible patients with cT2-4aN0-1M0 muscle-invasive bladder cancer. The primary objective was to measure safety, and the secondary objective was to assess the anti-tumor efficacy as measured by pathologic complete response along with 1-year recurrence-free survival. No dose-limiting toxicity was encountered in 21 patients enrolled and treated. Combination treatment achieved a pathologic complete response rate of 42.1% and a 1-year recurrence-free survival rate of 70.4%. Pathologic response was associated with baseline free E2F activity and tumor mutational burden but not PD-L1 status. Although T cell infiltration was broadly induced after intravesical oncolytic immunotherapy, the formation, enlargement and maturation of tertiary lymphoid structures was specifically associated with complete response, supporting the importance of coordinated humoral and cellular immune responses. Together, these results highlight the potential of this combination regimen to enhance therapeutic efficacy in cisplatin-ineligible patients with muscle-invasive bladder cancer, warranting additional study as a neoadjuvant therapeutic option. Abstract

In the Chinese society, where power distance is high, leaders’ attitudes and behavior toward employees determine their career development as well as affect the entire team’s performance. Therefore, exploring the kind of employees that leaders expect in China is essential. Based on implicit followership theory perspective, this study considers leaders’ positive implicit followership (LPIF) as the main research variable and examines its influence on employees’ innovative behavior (EIB). Moreover, it explores the multiple mediation effect of the leader–member exchange (LMX) relationship and psychological empowerment (PE) in this influence mechanism. The study sample comprised 389 leaders and their direct employees at 45 large- and medium-sized enterprises in Shandong, Beijing, Hebei, Shanghai, Shanxi, Zhejiang, and other regions of China. We used the leader–employee 1:1 matching questionnaire, and the longitudinal research design was adopted to avoid homology variance, making the study results more realistic and reliable. This study used the SPSS 26.0 and AMOS 26.0 statistical software to verify the hypotheses. Our findings show that LPIF has a significant positive effect on EIB, and LMX and PE have multiple mediation effects on the relationship between LPIF and EIB. When the level of LPIF is high, LMX and PE are also enhanced, which in turn promotes the increase in EIB. This study provides a new perspective for subsequent research on the psychological mechanism of employees and suggests an important method for understanding leadership and following processes in an organization. It plays a guiding role for the management practice of an enterprise, selection of leaders, and training of employees.

Abstract

Special geometric features and complex working environments render the internal defects of small-diameter thick-walled steel pipes "easy to expand, difficult to detect". In this paper, a magnetic permeability perturbation testing (MPPT) method is proposed to assess the internal axial cracks of small-diameter thick-walled steel pipes. The mechanism of the MPPT method and its corresponding probe and magnetizer are introduced, and its feasibility is verified through a series of simulations and experiments. Experiments conducted using different sizes of small-diameter thick-walled pipes show that this method offers good performance with respect to the detection of internal axial cracks. Additionally, both diameter and wall thickness significantly affect the MPPT signal. To a certain extent, a greater wall thickness or a smaller diameter brings about a weaker signal. This method does not benefit from the lift-off effect, nor is it limited by the skin effect, which has great practical value as a supplement to the evaluation of thick-walled steel pipes. 28. OPTIMIZING PROTEIN PURIFICATION FOR RNA-BINDING RECOMBINANT FUSION PROTEINS

Abstract

There has been great interest in delivering short interfering RNA (siRNA) and microRNA (miRNA) for therapeutic applications. However, the delivery of small RNAs remains challenging due to its inefficient cellular uptake and instability under physiological conditions. Here, we engineered a CXCR4-targeting RNA-protein nanoplex that consists of a CXCR4-targeting single-chain variable fragment (scFv) antibody, which is fused to an RNA-binding protamine peptide (RSQSRSRYYRQRQRSRRRRRRS). To obtain a functional RNA-binding protein, the removal of external nucleic acids is essential. This study aims to optimize the purification process of RNA-binding fusion proteins to free up RNA-binding domains and study if protein/siRNA complexes could successfully protect and deliver siRNA to silence cellular genes. After testing out different nucleic-acid removal methods, the high-salt-wash assisted immobilized metal affinity column purification method showed a great reduction of bound nucleic-acid contaminants. The purified fusion proteins showed a successful complexation with siRNAs and could deliver siRNA to the targeted cells.
