Abstract

The purpose of this work was to analyze and evaluate the functional properties of a new nanoemulsion that contained cordycepin. Droplet size and distribution were measured, and the stability of the nanoemulsion in different storage conditions was evaluated. We also tested skin irritation caused by the nanoemulsion, cordycepin stability in the nanoemulsion, anti-aging effect, and microbial challenge. The nanoemulsion was homogeneous with proper dispersion stability, and cordycepin was stable for eight months within the cosmetic nanoemulsion. The nanoemulsion did not irritate human hand and face skin, and the cosmetic nanoemulsion was effective in preventing microbial contamination of the product during use and storage. The new nanoemulsion containing cordycepin satisfied provisions of the 2015 Safety and Technical Standards for Cosmetics of the Ministry of Health, People’s Republic of China.

Abstract

The interaction between hyperoside and human serum albumin was studied in vitamin C (VC ) and VC -free environments using ultraviolet (UV)-vis absorption, fluorescence, circular dichroism spectra, and molecular docking techniques under simulated physiological conditions. The two environments had different influences on the secondary structure of human serum albumin (HSA). The α-helix content was slightly increased from 50% to 51% in the VC environment and increased from 50% to 55% in the VC -free environment. The thermodynamic parameters were ΔH° = -30.7 kJ⋅mol-1 and ΔS° = -23.4 mol-1 ⋅K-1 in the VC environment and ΔH° = -25.4 kJ⋅mol-1 and ΔS° = -11.4 J⋅mol-1 ⋅K-1 in the VC -free environment. Through thermodynamics parameters, hydrophobic force played a dominant role in the whole environment. The binding constants were calculated to be 7.25 × 105 mol⋅L-1 and 9.76 × 105 mol⋅L-1 at 298 K and they declined with the rise in temperature. The two binding distances were 2.6 nm and 2.5 nm respectively at 298 K, indicating that fluorescence energy transfer occurred. The UV-vis spectra indicated that fluorescence quenching of the HSA-hyperoside complex was a static quenching process. Hyperoside could spontaneously bind to HSA at site I (subdomain IIA). Molecular docking elucidated the way to binding basically through hydrophobic and van der Waals force interactions. Moreover, molecular docking showed that the VC environment could influence binding of HSA and hyperoside by more H-binding and less hydrophobic forces.

Abstract

Acanthopanax sessiliflorus (Rupr. &amp, Maxim.) Seem. (Araliaceae) is one of the most abundant species of genus Acanthopanax. The fruits of A. sessiliflorus are used in traditional medical protocols as an analgesic, tonic, antidiabetic, antihypertensive, anti-inflammatory, antitumor, and immune-stimulating agent. In this work, we carried out a comprehensive investigation into the anthocyanin components in the fruits of A. sessiliflorus. The anthocyanin content in the fresh fruits of A. sessiliflorus was determined by high performance liquid chromatography-diode array detection (HPLC/DAD), and the anthocyanin component was isolated from these using high-speed counter-current chromatography (HSCCC) and elucidated by electro-spray ionization-mass spectrometry (ESI/MS), 1H- and 13C-NMR. Its antioxidant activity was evaluated by ferric-reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH). We found that A. sessiliflorus contained a gross anthocyanin content of 121.35 mg/100 g. HSCCC was successfully used for separation and purification of the primary anthocyanin component, cyanidin 3-xylosyl-galactoside. The antioxidant and radical scavenging tests indicated that cyanidin 3-xylosyl-galactoside is a potent antioxidant.

Abstract

Cordyceps militaris, Chinese traditional medicinal fungus, has many bioactive properties. Cordycepin (3'-deoxyadenosine) is a major bioactive component of C. militaris. Various methods can significantly elevate cordycepin production, which suggests a diverse set of metabolic regulatory mechanisms. Thus, we aimed to identify transcription factors that regulate cordycepin biosynthesis pathways. Transcriptome analysis of wild-type C. militaris, C. militaris GYS60, a cordycepin high-producing strain, and C. militaris GYS80, a low-producing strain, were used to measure expression and function of genes related to cordycepin biosynthesis. The transcriptome expression data were confirmed by quantitative real-time polymerase chain reaction. We identified 155 relevant transcription factors in 19 families that included Fork head/winged helix factors, other C4 zinc finger-type factors, C2H2 zinc finger factors, tryptophan cluster factors, nuclear receptors with C4 zinc fingers, homeodomain factors, and Rel homology region factors. Energy generation and amino acid conversion pathways were activated in GYS60 so that abundance of cordycepin precursors was increased. Genes and transcription factors for rate-limiting enzymes in these pathways were identified. Overexpression of two key transcription factors, Kruppel-like factor 4 (Klf4) and Retinoid X receptor alpha (Rxra), promoted high cordycepin production in GYS60. In GYS60, Klf4 and Rxra were responsible for upregulation of genes in cordycepin biosynthesis, namely an oxidoreductase, 3',5'-cyclic AMP phosphodiesterase, a transferase, and adenylate cyclase. Upregulation of these genes increased 3'-AMP content, thereby elevating cordycepin synthesis.
