14. Post-translational regulation of transcription factor EB

Abstract

The bHLH-LZ transcription factor EB (TFEB) is a master regulator of lysosomal biogenesis and autophagy, it also plays an important role in lipid and glucose metabolism, mitochondrial biogenesis, integrated stress response and immunity. Dysregulation of TFEB family is implicated in cancer as well as neurodegenerative diseases. TFEB subcellular localisation and transcriptional activity are regulated by cellular nutrient and energy status. Elucidating the mechanism of TFEB regulation is the key to understanding its role in maintaining cellular homeostasis as well as providing therapeutic insights. In this study we showed that TFEB subcellular localisation is regulated by both amino acids and glucose availability. Low glucose levels activate mTORC2-AKT signalling, leading to the inactivation of GSK3β, and GSK3β kinase activity is required for TFEB cytoplasmic localisation. We also demonstrated that GSK3β phosphorylates TFEB at S138 only after a priming phosphorylation event at S142. The sequential phosphorylation is required for CRM1-depedent nuclear export of TFEB via a novel nuclear export signal. This mechanism is important for the rapid cytoplasmic relocalisation and inactivation of TFEB upon nutrient replenishment. Additionally, we performed a high-throughput screen of 1,600 FDA-approved drugs and identified around 100 chemical modulators of TFEB and TFE3, of which around 50 % exhibited lysosomal biogenesis promoting potential. In particular, trifluoperazine, pimozide and loperamide induce TFEB nuclear translocation, lysosomal biogenesis and autophagy in an mTORC1- and AMPK- independent manner.

Abstract

Fine root traits and their relationships reflect the ecological trade-off strategies of plants in resource investment and are important for understanding the life strategies and growth of plants in response to changes in the environment. We used 16-year-old Chinese fir (Cunninghamia lanceolata Lamb. Hook.) plantations with different slope positions as the research object to explore the morphological, anatomical, and chemical properties of fine roots and their relationships. With increasing root order levels, the morphological, anatomical, and chemical traits of the fine roots of Chinese fir at different slope positions showed similar change trends; however, at the same order level, the differences were large. Under the upper slope site conditions, the average diameter of the second- and third-order roots and the thickness of the third-order root cortex were the highest. However, specific surface area, vascular bundle diameter, and the ratio of third-order roots were higher under the middle-slope site conditions. Under the lower slope site conditions, the specific surface area and specific root length of first-order roots and the root ratio of second-order roots were the highest. The biomasses of the first- and third-order roots on the middle and lower slopes were higher than those on the upper slope. The contents of N and P in fine roots of grades 1–3 Chinese fir showed the order of lower slope > middle slope > upper slope; however, the changes in C/N and C/P ratios showed the opposite trend, indicating differences in the morphological, anatomical, and chemical properties as well as resource acquisition strategies of fine roots of grades 1–3 Chinese fir under different slope positions. There were negative correlations between fine root diameter, N and P contents, and specific root length, indicating an acquisition and conservative resource trade-off relationships between fine root morphological, anatomical, and chemical traits. There were also differences in the relationships between the morphological, anatomical, and chemical traits of Chinese fir fine roots at different slope positions, indicating that the relationships between these traits were affected by slope position change. Chinese fir varieties with root-foraging characteristics ranging from resource conservation to resource acquisition can be selected for planting to improve the productivity of C. lanceolata plantations. 32. Improving aetiological classification of transient ischaemic attack and ischaemic stroke

Abstract

Transient ischaemic attack (TIA) and ischaemic stroke is a heterogeneous disease with more than 150 known causes including different cardiac, arterial, hemodynamic and other systemic abnormalities. TIA and ischaemic stroke of different aetiology have been suggested to have different risk factor profiles, clinical manifestations and prognoses. Therefore categorization of patients into classes congruent with the aetiology is the key to both understanding and managing stroke. Such categorization requires assembling stroke features to create categories based on similarities and is known as aetiological classification. Accurate aetiological classification is indispensable in epidemiological and genetic studies and in clinical trials. However, all current stroke classification systems have limitations. Perhaps the most important limitation of all current stroke classification systems is that about one-third of TIA and ischaemic stroke are of undetermined aetiology despite standard diagnostic work-up, potentially undermining primary and secondary stroke prevention. Therefore, better understanding of stroke aetiology will help to improve the classification of TIA and ischaemic stroke hence ultimately improving the prevention strategy. The aim of my thesis has been to compare characteristics of currently used aetiological classification systems, to explore the underlying causes for TIA and ischaemic stroke of undetermined aetiology, and to study aetiological classification related aspects of TIA and ischaemic stroke, including risk factors, imaging characteristics, plasma biomarkers and prognoses. I have collected, collated and analysed data from the first ten years of the Oxford Vascular study (OXVASC), which is an ongoing prospective, population-based incidence study of vascular disease in all territories in Oxfordshire, UK. The study population comprises approximately 92,728 individuals registered with nine general practices and uses multiple overlapping “hot” and “cold” methods to identify all patients with acute vascular events. There are several clinically relevant findings in this thesis which address areas that could be improved for better aetiological classification of TIA and ischaemic stroke. First, I have validated the aetiological classification of TIA. Second, I have found that compared to the widely used TOAST aetiological classification system, the stringent requirements for adequate investigation in recently developed classification systems do not add subtype-specific information to reliably reduce the assignment to the undetermined subtype. Third, I have shown that neither occult atheroma nor traditional risk factors appear to account for undetermined TIA and ischaemic stroke. However, previous migraine may have a particular role in the aetiology of undetermined TIA and ischaemic stroke. Fourth, I have shown that compared to cases of non-small vessel disease, small vessel disease cases have higher long-term average systolic and diastolic blood pressure but less long-term blood pressure variability. Fifth, I have shown that in contrast to cases of small vessel disease, white matter changes on brain imaging are not independently associated with non-small vessel subtypes. Sixth, I have shown that applying haemostatic and inflammatory markers in stroke aetiological work-up would appear to have little potential to add very limited information regarding the specific underlying cause of different aetiological subtypes. Finally, I have highlighted the large clinical burden of TIA and ischaemic stroke of undetermined aetiology by showing that compared to cases of determined aetiology, undetermined TIA and ischaemic stroke appears to have non-benign short- and long-term prognoses.
