Umbilical cord blood (UCB) is an advantageous source for hematopoietic stem/progenitor cell (HSPC) transplantation, yet the current strategies for large-scale and cost-effective UCB-HSPC preparation are still unavailable. To overcome these obstacles, we systematically evaluate the feasibility of our newly identified CH02 peptide for ex vivo expansion of CD34+ UCB-HSPCs. We herein report that the CH02 peptide is specifically enriched in HSPC proliferation via activating the FLT3 signaling. Notably, the CH02-based cocktails are adequate for boosting 12-fold ex vivo expansion of UCB-HSPCs. Meanwhile, CH02-preconditioned UCB-HSPCs manifest preferable efficacy upon wound healing in diabetic mice via bidirectional orchestration of proinflammatory and anti-inflammatory factors. Together, our data indicate the advantages of the CH02-based strategy for ex vivo expansion of CD34+ UCB-HSPCs, which will provide new strategies for further development of large-scale HSPC preparation for clinical purposes.

The comprehensive efficiency of the gear transmission directly restricts the development quality of transportation, construction, wind power and other fields, and the requirements of gear transmission power, load capacity and life expectancy improve gradually. However, the fatigue failure of gear greatly limits the performance and reliability of modern gear equipment. The present situation of fatigue crack initiation and propagation of gear is discussed comprehensively, and the main forms and causes of the gear failure are introduced. This research induces the factors affecting the fatigue strength of gears, summarizes three types of gear cracks, the crack initiation and extension method, and the existing gear fatigue crack simulation method. It is used to describe the microstructure mechanical behavior of gear fatigue, improve the understanding of the related characteristics and mechanism of gear fatigue, and study and analyze the remaining service life of gear fatigue tests. Finally, some suggestions on how to avoid gear cracks are put forward.

Simple Summary: Skeletal muscle growth and development are closely related to the growth performance of livestock and poultry, and miRNAs are involved in skeletal muscle growth and development by negatively regulating target genes. In this study, the Gannan yak and the Tianzhu white yak which have large differences in production performance were used as research subjects. The difference of muscle fibers was analyzed by fiber staining. A total of 254 differentially expressed (DE) miRNAs were screened by transcriptome sequencing. Functional enrichment analysis revealed that the target genes were significantly enriched in muscle growth-related terms and pathways. By constructing a DE miRNA- DE mRNA interaction network, 18 key miRNAs related to muscle growth and development such as miR-3968-z, miR-2478-z, novel-m0143-3p, novel-m0024-3p, novel-m0128-5p, and novel-m0026-3p were screened. These results help us to explore the key influencing factors of skeletal muscle development in different breeds of yaks, and lay a foundation for further research on the molecular regulatory mechanisms of muscle growth and development. The Gannan yak, a superior livestock breed found on the Tibetan Plateau, exhibits significantly enhanced body size, weight, and growth performance in comparison to the Tianzhu white yak. MiRNAs play a pivotal role in regulating muscle growth by negatively modulating target genes. In this study, we found the average diameter, area, and length of myofibers in Gannan yaks were significantly higher than those of Tianzhu white yaks. Further, we focused on analyzing the longissimus dorsi muscle from both Gannan yaks and Tianzhu white yaks through transcriptome sequencing to identify differentially expressed (DE)miRNAs that influence skeletal muscle development. A total of 254 DE miRNAs were identified, of which 126 miRNAs were up-regulated and 128 miRNAs were down-regulated. GO and KEGG enrichment analysis showed that the target genes of these DE miRNAs were significantly enriched in signaling pathways associated with muscle growth and development. By constructing a DE miRNA- DE mRNA interaction network, we screened 18 key miRNAs, and notably, four of the candidates (novel-m0143-3p, novel-m0024-3p, novel-m0128-5p, and novel-m0026-3p) targeted six genes associated with muscle growth and development (DDIT4, ADAMTS1, CRY2, AKIRIN2, SIX1, and FOXO1). Traditional wireless positioning technology has low accuracy, and UWB positioning technology can achieve centimeter-level high-precision positioning to meet the accuracy requirements of the fully mechanized working face positioning systems. However, the existing researches ignore the impact of position change of the base station installed on the hydraulic support on the positioning effect. In order to solve the above problems, laser ranging technology is applied to the mine environment and a positioning system of fully mechanized working face based on UWB and laser ranging is designed. The positioning base station is installed on the hydraulic support, and the position of the base station changes when the position of the hydraulic support changes. The mine intrinsically safe laser ranging sensor is used to measure the distance between the positioning base stations. The external antenna of the positioning base station transmits fixed-power UWB wireless signals. The positioning base station communicates with the positioning identification card worn on the personnel or installed on the equipment, and determines the positioning information of the moving target through the time-of-flight ranging method. The positioning base station uploads the positioning information data to the ground data center through the mine ring network. The system test is carried out on the E2308 working face of Gaohe Coal Mine, and the result show that the positioning accuracy of the system reaches 0.193 m and the positioning accuracy is high. Moreover, the system base station position calibration accuracy is about 0.011 m, which solves the problem of base station position calibration effectively.

Jeryak is the F1 generation of the cross between Gannan yak and Jersey cattle, which has the advantages of fast growth and high adaptability. The growth and development of skeletal muscle is closely linked to meat production and the quality of meat. However, the molecular regulatory mechanisms of muscle growth differences between Gannan yak and Jeryak analyzed from the perspective of chromatin opening have not been reported. In this study, ATAC-seq was used to analyze the difference of chromatin openness in longissimus muscle of Gannan yak and Jeryak. It was found that chromatin accessibility was more enriched in Jeryak compared to Gannan yak, especially in the range of the transcription start site (TSS) ± 2 kb. GO and KEGG enrichment analysis indicate that differential peak-associated genes are involved in the negative regulation of muscle adaptation and the Hippo signaling pathway. Integration analysis of ATAC-seq and RNA-seq revealed overlapping genes were significantly enriched during skeletal muscle cell differentiation and muscle organ morphogenesis. At the same time, we screened FOXO1, ZBED6, CRY2 and CFL2 for possible involvement in skeletal muscle development, constructed a genes and transcription factors network map, and found that some transcription factors (TFs), including YY1, KLF4, KLF5 and Bach1, were involved in skeletal muscle development. Overall, we have gained a comprehensive understanding of the key factors that impact skeletal muscle development in various breeds of cattle, providing new insights for future analysis of the molecular regulatory mechanisms involved in muscle growth and development. In view of problem that theoretical modeling and simulation experiment are mostly used in research of underground path loss characteristics of high -precision ultra wide band (UWB) signal, actual measurement research of UWB signal is less. The path loss measurement and analysis of UWB signal are carried out in two typical scenarios of running roadway of overhead passenger device (monkey car) with relatively common environmental conditions such as roadway size, roadway wall material and pipe facilities，and heading face with bad environment, high coal dust concentration and serious water accumulation. The results show that: ① The path loss index of UWB signal in two kinds of roadway environments is less than 2, and wave guide effect of roadway leads to better propagation characteristics of UWB signal than that of free space. The base station deployment interval of the UWB positioning system in roadway is larger than that of the ground free space propagation scene.② In the same mine, the roadway environment is different, and the path loss characteristics are also different. The path loss index and shadow fading variance of heading face are significantly different from those of monkey car running roadway. ③ From comparison of single test point in roadway of heading face, the path loss in NLOS environment is larger than that in LOS environment. More intensive positioning base stations need to be deployed in roadway in NLOS environment.The results can be used to guide deployment scheme design of mine UWB precise positioning system.

During the postnatal stages, skeletal muscle development undergoes a series of meticulously regulated alterations in gene expression. However, limited studies have employed chromatin accessibility to unravel the underlying molecular mechanisms governing muscle development in yak species. Therefore, we conducted an analysis of both gene expression levels and chromatin accessibility to comprehensively characterize the dynamic genome-wide chromatin accessibility during muscle growth and development in the Tianzhu white yak, thereby elucidating the features of accessible chromatin regions throughout this process. Initially, we compared the differences in chromatin accessibility between two groups and observed that calves exhibited higher levels of chromatin accessibility compared to adult cattle, particularly within ±2 kb of the transcription start site (TSS). In order to investigate the correlation between alterations in chromatin accessible regions and variations in gene expression levels, we employed a combination of ATAC-seq and RNA-seq techniques, leading to the identification of 18 central transcriptional factors (TFs) and 110 key genes with significant effects. Through further analysis, we successfully identified several TFs, including Sp1, YY1, MyoG, MEF2A and MEF2C, as well as a number of candidate genes (ANKRD2, ANKRD1, BTG2 and LMOD3) which may be closely associated with muscle growth and development. Moreover, we constructed an interactive network program encompassing hub TFs and key genes related to muscle growth and development. This innovative approach provided valuable insights into the molecular mechanism underlying skeletal muscle development in the postnatal stages of Tianzhu white yaks while also establishing a solid theoretical foundation for future research on yak muscle development. A hybrid offspring of Gannan yak and Jersey cattle, the Jeryak exhibits apparent hybrid advantages over the Gannan yak in terms of production performance and other factors. The small non-coding RNAs known as miRNAs post-transcriptionally exert a significant regulatory influence on gene expression. However, the regulatory mechanism of miRNA associated with muscle development in Jeryak remains elusive. To elucidate the regulatory role of miRNAs in orchestrating skeletal muscle development in Jeryak, we selected longissimus dorsi muscle tissues from Gannan yak and Jeryak for transcriptome sequencing analysis. A total of 230 (DE) miRNAs were identified in the longissimus dorsi muscle of Gannan yak and Jeryak. The functional enrichment analysis revealed a significant enrichment of target genes from differentially expressed (DE)miRNAs in signaling pathways associated with muscle growth, such as the Ras signaling pathway and the MAPK signaling pathway. The network of interactions between miRNA and mRNA suggest that some (DE)miRNAs, including miR-2478-z, miR-339-x, novel-m0036-3p, and novel-m0037-3p, played a pivotal role in facilitating muscle development. These findings help us to deepen our understanding of the hybrid dominance of Jeryaks and provide a theoretical basis for further research on the regulatory mechanisms of miRNAs associated with Jeryak muscle growth and development. The production performance of Jeryak, resulting from the F1 generation of the cross between Gannan yak and Jersey cattle, exhibits a significantly superior outcome compared with that of Gannan yak. Therefore, we used an RNA-seq approach to identify differentially expressed mRNAs (DEMs) and differentially expressed lncRNAs (DELs) influencing muscle growth and development in Gannan yaks and Jeryaks. A total of 304 differentially expressed lncRNAs and 1819 differentially expressed mRNAs were identified based on the screening criteria of |log 2 FC| > 1 and FDR < 0.05. Among these, 132 lncRNAs and 1081 mRNAs were found to be down-regulated, while 172 lncRNAs and 738 mRNAs were up-regulated. GO and KEGG analyses showed that the identified DELs and DEMs were enriched in the entries of pathways associated with muscle growth and development. On this basis, we constructed an lncRNA–mRNA interaction network. Interestingly, two candidate DELs (MSTRG.16260.9 and MSTRG.22127.1) had targeting relationships with 16 (MYC, IGFBP5, IGFBP2, MYH4, FGF6, etc.) genes related to muscle growth and development. These results could provide a basis for further studies on the roles of lncRNAs and mRNAs in muscle growth in Gannan yaks and Jeryak breeds. [ABSTRACT FROM AUTHOR]

The task of relation classification is an important pre-task in natural language processing tasks. Relation classification can provide a high-quality corpus for tasks such as machine translation, human–computer dialogue, and structured text generation. In the process of the digitalization of standards, identifying the entity relationship in the standard text is an important prerequisite for the formation of subsequent standard knowledge. Only by accurately labeling the relationship between entities can there be higher efficiency and accuracy in the subsequent formation of knowledge bases and knowledge maps. This study proposes a standard text relational classification model based on cascaded word vector attention and feature splicing. The model was compared and ablated on our labeled standard text Chinese dataset. At the same time, in order to prove the performance of the model, the above experiments were carried out on two general English datasets, SemEval-2010 Task 8 and KBP37.
