ObjectivesThe COVID-19 pandemic has taken a significant toll on people worldwide for more than 2 years. Previous studies have highlighted the negative effects of COVID-19 on the mental health of healthcare workers (HCWs) more than the positive changes, such as post-traumatic growth (PTG). Furthermore, most previous studies were cross-sectional surveys without follow-ups. This study draws on PTG follow-up during the COVID-19 outbreak at 12-month intervals for 2 years since 2020. The trajectories and baseline predictors were described.MethodsA convenience sampling method was used to recruit frontline nurses or doctors at the COVID-19-designated hospital who were eligible for this study. A total of 565 HCWs completed the 2 years follow-up and were used for final data analysis. The latent growth mixture models (GMM) was used to identify subgroups of participants with different PTG trajectories. Multinomial logistic regression model was used to find predictors among sociodemographic characteristics and resilience at baseline.ResultsFour trajectory PTG types among HCWs were identified: ‘Persistent, “Steady increase”, “High with drop”, and “Fluctuated rise.” Comparing the “Persistent low” type, the other three categories were all associated with older age, higher education. Furthermore, “Persistent low” was also negatively associated with resilience at baseline.ConclusionThe PTG of HCWs with different characteristics showed different trends over time. It is necessary to increase the measure frequency to understand the PTG status in different times. Improving HCW’s resilience could help improve staff PTG.

The traditional Mongolian medicine Erdun-Uril is a conventional combination of 29 herbs commonly used for the treatment of cerebrovascular ailments. It has the effects of reducing inflammation, counteracting oxidative stress, and averting strokes caused by persistent cerebral hypoperfusion. Prior research on Erdun-Uril has predominantly concentrated on its pharmacodynamics and mechanism of action; however, there has been a lack of systematic and comprehensive investigation into its chemical constituents. Therefore, it is crucial to establish an efficient and rapid method for evaluating the chemical constituents of Erdun-Uril. In this study, Erdun-Uril was investigated using UHPLC-Q-Exactive Orbitrap MS combined with parallel reaction monitoring for the first time. Eventually, a total of 237 compounds, including 76 flavonoids, 68 phenolic compounds, 19 alkaloids, 7 amino acids, etc., were identified based on the chromatographic retention time, bibliography data, MS/MS2 information, neutral loss fragments (NLFs), and diagnostic fragment ions (DFIs). And of these, 225 were reported for the first time in this study. This new discovery of these complex components would provide a reliable theoretical basis for the development of pharmacodynamics and quality standards of the Mongolian medicine Erdun-Uril. The programmed cell death 4 (PDCD4) tumor-suppressor gene regulates cell apoptosis, protein translation, signal transduction, and induction of mediators of inflammation. However, the mechanism by which PDCD4 is down-regulated and regulates tumor growth remains elusive. In this study, we showed that PDCD4 is down-regulated in glioma cells and acts as a tumor suppressor. Based on the TCGA data, we confirmed that AKT2, but not AKT1 or AKT3, interacts with PDCD4, thus leading to the suppression of PDCD4 in glioma cells. Moreover, the analysis suggested that PDCD4 regulates the expression of IL-5, CCL-5, VEGF, and CXCL10 via the NF-kB pathway. Additionally, depletion of levels of PDCD4 promoted angiogenic activity of glioma cells via the VEGF-STAT3 pathway. When tumor cells over-expressing PDCD4 were injected into nude mice, the increased expression of PDCD4 blocked tumorigenesis and prolonged overall survival. Our study indicates the need to develop drugs that can modulate the expression of PDCD4 and test their efficacy in clinical trials.

Background: Tinea of vellus hair is a dermatophyte infection with or without localised infection of adjacent skin. The epidemiology and pathogenesis of this disorder have insufficiently been established. Objectives: To gain insight into the epidemiology and analysis dissection of patterns of vellus hair infection. Methods: A comprehensive examination of our series of cases and existing literature, clinical information was performed. Microsporum species were assayed for levels of keratinase and SUB1‐3 level after different keratin‐induced cultures. Results: The infection affects both children and adults, with a male‐to‐female ratio of 1:1.3. The most afflicted areas are the face, neck and upper limbs. Both typical (ringworm‐like) and atypical (tinea incognito) tinea lesions can be observed. The disorder is caused primarily by the zoophilic Microsporum canis (40.39%). Topical antifungal drugs alone are ineffective; mycological cure is achieved in combination with systemic antifungal therapy for 4–8 weeks. In vitro, M. canis exhibited significantly elevated levels of secreted protease and SUB1 and SUB3 transcripts in cat hair cultures. Conversely, the findings of anthropophilic dermatophytes M. audouinii and M. ferrugineum demonstrated the opposite trend, suggesting that there are associations between keratinase activity and expression of SUB1 and SUB3 induced by different keratin substrates among M. canis complex members. Conclusions: The epidemiological characteristics of tinea of vellus hair help us to understand its transmission patterns and develop effective prevention strategies. There are associations between keratinase activity and expression of SUB1‐3 induced by different substrates, which may explain the host shift and adaptation from pet animals to particular micro‐habitats on the human host. To present a consecutive 20-year series of blood blister-like aneurysms (BBAs) to show that clip-on-wrapping with a Y-shaped autologous dura mater enables treatment of BBAs with a low complication rate and a satisfactory curative result. A retrospective review was performed from patients with BBAs of the internal carotid artery (ICA) at the Affiliated Hospital of Qingdao University from 1999 to 2019. Diagnosis and treatment options were analyzed. Outcome was assessed using the modified Rankin scale (mRS). A total of 30 patients with BBAs of the ICA were included. Among these patients, 20 patients underwent microsurgical treatment (15 patients were treated by clip-on-wrapping with a Y-shaped autologous dura mater), the other 10 patients underwent endovascular treatment. All patients presented with subarachnoid hemorrhage (SAH). Four angiograms were initially negative. For all patients, intraoperative rupture occurred in five cases, but no postoperative aneurysm rupture occurred in this series. Three cases with clinical or radiologic cerebral infarctions were observed. The outcome was favorable in 26 patients. Clip-reinforced wrapping technique using a Y-shaped autologous dura mater may be an effective method for treating BBAs. [ABSTRACT FROM AUTHOR]

Long noncoding RNA (lncRNA) growth arrest‐specific 5 (GAS5) is implicated in several cancers via modulating microRNAs (miRs). However, little information is available about the correlation between GAS5 and miR‐10b. Therefore, we sought out to investigate the biological role of GAS5‐miR‐10b node mainly in glioma cells. We artificially modulated GAS5 to explore its roles in viability assayed by cell counting kit‐8 (CCK‐8), motile activities by 24‐Transwell assay, as well as apoptosis by a flow cytometer and Western blot assay. miR‐10b and Sirtuin 1 (Sirt1) were quantified by qRT‐PCR. After co‐transfection, we analyzed the viability, migration, invasion, apoptosis, and Sirt1 expression. Western blot was implemented to detect the phosphorylated forms of PTEN, PI3K, AKT, MEK, and ERK. GAS5 inhibited proliferation and motile behaviors, and fortified apoptosis. As for the viability and motile activities, the property of GAS5 was reversed in miR‐10b‐replenished U251 and A172 cells, while maintained in miR‐10b‐deficient cells. Additionally, GAS5‐induced apoptosis was abolished by miR‐10b overexpression while fortified by miR‐10b silence. Besides, GAS5 negatively modulated Sirt1 via miR‐10b. Moreover, Sirt1 negatively modulated PTEN and positively mediated the abovementioned regulators. GAS5 represses the process of glioma cells by decreasing miR‐10b, which as accompanied by Sirt1 silence‐induced inactivation of PTEN/PI3K/AKT and MEK/ERK cascades. [ABSTRACT FROM AUTHOR]

The present study aimed to explore the application of diffusion tensor tractography (DTT) in the preoperative planning and prognostic evaluation of tumors located in the functional areas of the brain. A total of 42 patients diagnosed with intracranial tumors were randomly assigned to either the trial or the control group. DT imaging (DTI) was performed on the basis of preoperative conventional magnetic resonance imaging (MRI) and analyzed for patients in the trial group. Patients in the control group underwent only routine MRI scans. The effect of DTT on the prognosis of patients was evaluated by tumor resection rate and quality of life evaluation using Karnofsky performance score (KPS) comparison between the trial and control groups. There were no significant differences for total tumor removal rate in the trial group (85.71%) compared with that in the control group (71.43%) (P>0.05). The rate of postoperative symptom improvement in the trial group (85.71%) was significantly higher compared with that in the control group (47.62%) (P<0.05). The KPS value of the trial group was significantly higher postoperatively (78.57±17.40) compared with that preoperatively (66.67±16.23) (P<0.05). The KPS value of the control group postoperatively (72.38±19.21) was significantly higher compared with that preoperatively (66.67±16.00) (P<0.05). The postoperative KPS improvement rate [postoperative value-preoperative value)/preoperative value] of the trial group was significantly higher compared with that in the control group. In conclusion, the use of DTT is an effective supplement to traditional MRI, with particular relevance in preoperative planning, particularly for tumors in the functional area of the brain, and can significantly improve the prognostic function of patients.
