2. Nivolumab Plus 5-Azacitidine in Pediatric Relapsed/Refractory Acute Myeloid Leukemia (AML): Phase I/II Trial Results from the Therapeutic Advances in Childhood Leukemia and Lymphoma (TACL) Consortium.

Abstract

Improvements in survival have been made over the past two decades for childhood acute myeloid leukemia (AML), but the approximately 40% of patients who relapse continue to have poor outcomes. A combination of checkpoint-inhibitor nivolumab and azacitidine has demonstrated improvements in median survival in adults with AML. This phase I/II study with nivolumab and azacitidine in children with relapsed/refractory AML (NCT03825367) was conducted through the Therapeutic Advances in Childhood Leukemia & Lymphoma consortium. Thirteen patients, median age 13.7 years, were enrolled. Patients had refractory disease with multiple reinduction attempts. Twelve evaluable patients were treated at the recommended phase II dose (established at dose level 1, 3 mg/kg/dose). Four patients (33%) maintained stable disease. This combination was well tolerated, with no dose-limiting toxicities observed. Grade 3-4 adverse events (AEs) were primarily hematological. Febrile neutropenia was the most common AE ≥ grade 3. A trend to improved quality of life was noted. Increases in CD8+ T cells and reductions in CD4+/CD8+ T cells and demethylation were observed. The combination was well tolerated and had an acceptable safety profile in pediatric patients with relapsed/refractory AML. Future studies might explore this combination for the maintenance of remission in children with AML at high risk of relapse.

Abstract

Avalanche photodiode (APD) has been intensively investigated as a promising candidate to replace photomultiplier tubes (PMT) for weak light detection. However, in conventional APDs, a large portion of carrier energy drawn from the electric field is thermalized, and the multiplication efficiencies of electron and hole are low and close. In order to achieve high gain, the device should work under breakdown bias, where carrier multiplication proceeds bi-directionally to form a positive feedback multiplication circle. However, breakdown is hard to control, in practice, APDs should work under Geiger mode as a compromise between sustainable detection and high gain. The complexity of system seriously restricts the application. Here, we demonstrate an avalanche photodiode holding high gain without breakdown, which means no quenching circuit is needed for sustainable detection. The device is based on a GaN/AlN periodically-stacked-structure (PSS), wherein electron holds much higher efficiency than hole to draw energy from the electric field, and avalanche happens uni-directionally with high efficiency. and a recorded high gain (10^4) tested under constant bias is obtained in a prototype device, wherein the stable gain can be determined by the periodicity of the GaN/AlN PSS. This work not only brings a new light into avalanche multiplication mechanism, but also paves a technological path with high commercial value to realize highly sensitive avalanche devices working under constant bias like PMT.

Abstract

In order to achieve energy saving and environmental protection, the research and utilization of solid waste polyurethane is realized. Using monocomponent di-propylene glycol (PG) and polyethylene glycol (PEG) as the alcoholysis agent of the two-component diols, alkali metal sodium hydroxide as the metal catalyst, and montmorillonite clay (MMT) as the modifier, the degradation products were utilized to re-prepare montmorillonite/polyurethane nanocomposites with excellent performance, and to re-prepare nano-foam materials. And a series of tests and analyses were carried out on the degradation products and nanofoam materials. The results show that when m(PG): m(PEG) = 40: 60, sodium hydroxide is 1.0 g, montmorillonite addition is 0.3 g, the polyurethane nanomaterials can achieve the best degradation effect, the apparent density of the newly prepared nano-foam material is 0.05 g/cm3, the compressive strength is 0.41 MPa. At this time, the nano-foam material has a smaller thermal conductivity and better cold-retaining performance, and meets the requirements of the national standard. [ABSTRACT FROM AUTHOR]

Abstract

The chemical epigenetic modifier 5-azacitidine (5-Aza C), a DNA methyltransferase inhibitor, was used to manipulate the endophytic fungus Penicillium sp. KMU18029. From its rice fermentation extract, a new polyketone compound (3S,4R)-3,4,8-trihydroxy-6-methyl-3,4-dihydronaphthalen-1(2H)-one (1), along with 13 known compounds, 3,4,8-trihydroxy-6-(hydroxymethyl)-3,4-dihydronaphthalen-1(2H)-one (2), decaturin B (3), 15-hydroxydecaturin A (4), oxalicine A (5), pileotin A (6), pyrandecarurin A (7), decaturenol A (8), decaturenoid (9), penisarins A (10), oxaline (11), (4E,8E)-N-D-2'-hydroxyocta-decanoyl-1-O-β-D-glycopy-ranosyl-9-methyl-4,8-sphingadienine (12), ergosterol (13) and stigma-5-en-3-O-β-glucoside (14), were separated. Among the known compounds, 2, 7, 12 and 14 were not found in our previous research on this strain. The structure of the new compound was identified by spectroscopic techniques such as HR-ESIMS, 1D NMR, 2D NMR and CD. Furthermore, all the isolated compounds were tested for their antimicrobial activities, and only compounds 1, 2 and 11 showed weak activities against S. aureus, with MICs of 128 μg/mL. Abstract

Tetrapanax papyrifer, the rice‐paper plant, is an evergreen shrub and is traditionally used as a herbal medicine in China. In October 2021, a serious leaf spot disease was extensively observed on T. papyrifer in Guizhou, China. Given the limited information available about the aetiology of this disease, isolations were performed, the morphological characteristics and multi‐locus phylogenetic analysis of the fungus associated with the disease were described, and the pathogenicity tests and Koch's postulates were completed. To our knowledge, this is the first report of leaf spot on T. papyrifer caused by Alternaria tenuissima in China. Among 11 fungicides tested, 10% difenoconazole WDG had the best inhibition effect in vitro with EC50 values at 0.1 μg mL−1.
