Abstract

RNA‐based therapeutics have emerged as a promising strategy for cancer immunotherapy, encompassing the silencing of immune checkpoint genes, chimeric antigen receptor T (CAR‐T) cell production, and antitumor vaccines. Despite their tremendous potential, the urgent need for the development of clinically applicable delivery systems remains paramount. In this study, vesicle‐like nanoparticles (VNPs) are devised using clinically approved amphiphilic polymers and lipids as the delivery system for siRNA and mRNA. Through meticulous formulation adjustments of cationic lipids and ionizable lipids, a VNP formulation with exceptional transfection efficiency is identified. Notably, the VNPs maintained their remarkable transfection efficiency even after 6 months of storage. When loaded with siPD‐L1 and siCD47, these VNPs effectively silenced two critical immune checkpoint genes, enabling successful cancer immunotherapy. Moreover, when employed as a delivery system for mRNA vaccines, the VNPs induced a robust population of antigen‐specific CD8+ T cells in immunized mice. This led to the successful suppression of tumor growth (5 out of 8 subjects becoming tumor‐free) and nearly complete inhibition of lung metastasis. In summary, this lipid‐ and polymer‐based VNPs offer a long shelf life, excellent loading and transfection efficiency, versatility for various RNA types, and hold great promise as a delivery system for clinical applications. 21. Production of Accurate Skeletal Models of Domestic Animals Using Three-Dimensional Scanning and Printing Technology

Abstract

Access to adequate anatomical specimens can be an important aspect in learning the anatomy of domestic animals. In this study, the authors utilized a structured light scanner and fused deposition modeling (FDM) printer to produce highly accurate animal skeletal models. First, various components of the bovine skeleton, including the femur, the fifth rib, and the sixth cervical (C6) vertebra were used to produce digital models. These were then used to produce 1:1 scale physical models with the FDM printer. The anatomical features of the digital models and three-dimensional (3D) printed models were then compared with those of the original skeletal specimens. The results of this study demonstrated that both digital and physical scale models of animal skeletal components could be rapidly produced using 3D printing technology. In terms of accuracy between models and original specimens, the standard deviations of the femur and the fifth rib measurements were 0.0351 and 0.0572, respectively. All of the features except the nutrient foramina on the original bone specimens could be identified in the digital and 3D printed models. Moreover, the 3D printed models could serve as a viable alternative to original bone specimens when used in anatomy education, as determined from student surveys. This study demonstrated an important example of reproducing bone models to be used in anatomy education and veterinary clinical training.

35. Novel Area-Changed Capacitive Methods for Simultaneous Displacement Transducing and Force Balance in a Nano-g MEMS Accelerometer

Abstract

High-precision MEMS accelerometers with nano-g resolution are emergent instruments for geophysical applications and proved their competence in terms of functionality. The electromagnetic actuator, which serves as an auxiliary component in nano-g MEMS accelerometers for improving the dynamic response, faces the challenges of process incompatibility, temperature sensitivity, and large form factor. Thereby, this paper proposes an area-changed capacitive method for both displacement transducing and force balance in a nano-g MEMS accelerometer, aiming to address those posed challenges and provide favourable performance. Thanks to the allowed large displacement range in the sensitive direction of the proposed device, the area-changed capacitive mechanism is able to be integrated with a highly-sensitive quasi-zero stiffness spring-mass structure. As a result, the fabricated force-balance MEMS accelerometer attains a calibrated self-noise of 1.3 ng/ $\surd $ Hz, which is one of the most sensitive MEMS-based accelerometers reported to date. The settling time, on the other hand, is reduced to 0.5 s with the electrostatic closed-loop control featuring the proposed subject, compared to 15.7 s in the open-loop configuration. In addition, the critical acceleration input at the boundary of the “pull-in” is calculated as 5.4 g, which is adaptable to most geophysical applications. This work is of considerable potential in geophysical applications such as earthquake monitoring or gravity measurements, and promising a high-performance closed-loop MEMS accelerometer. [2023-0161]

Abstract

Since complex barriers need to be passed through during the process of drilling long-distance horizontal freezing boreholes, the stability of the borehole wall at the soft and hard stratum interface is the key in controlling the deflection of the horizontal boreholes. The author puts forward to establishing the stability mechanical model of horizontal freezing borehole walls with fluid loss as the control index, starting from the stability mechanism of horizontal freezing boreholes and combined with the working conditions of horizontal freezing boreholes; selects two optimal fluid loss additives from the perspective of the molecular structure of bentonite materials, and determines the optimal formula by formulation optimization by virtue of orthogonal tests; applies the optimal formula into the long-distance pile and borehole penetration test, and validates the reliability of wall protection slurry formula materials for horizontal boreholes via comparing the water loss and soil erosion at the pile interface and analyzing the deflection rules, which is of certain value in engineering promotion and application.

41. Accuracy Check and Comparative Analysis of Horizontal Fiber-Optic Gyro Inclinometer in Freezing Hole

Abstract

Based on the principle of fiber-optic gyro inclinometer, the characteristics of horizontal freezing hole and the working conditions, it is analyzed that the horizontal hole fiber-optic gyroscope tilting technology for freezing holes is one of the most reliable methods. In order to verify the azimuth measurement error of the horizontal fiber optic-gyro inclinometer, the simulation test under the 1:1 working condition was carried out with the actual working conditions on site. Compared with the total station measurement data, it was concluded that 85.2% of the data is in measurement error within ±2°, 55.6% of the data is within ±1°, which verifies the reliability of the measurement data of the horizontal fiber-optic gyro inclinometer. At the test level of α=0.05, the azimuth test error accords with the normal distribution of μ=0.54 and σ=0.73. In the actual use process, the algorithm can be used to optimize the measurement data by taking multiple measurements and taking the average value in combination with the field conditions, so that the data can be closer to the true value.

The occurrence of VL was recorded, and treatment responses were assessed according to the modified response evaluation criteria in solid tumor (mRECIST).In terms of treatment response, the total response status (including CR, PR, SD and PD), objective response rate (ORR) and disease control rate (DCR) were elevated in VL patients compared to non-VL patients as well as in VL nodules compared to non-VL nodules. Liver function indexes and adverse events were assessed. Progression-free survival (PFS) and overall survival (OS) were evaluated with the last follow-up date of March 2020.&#x00A0Results: The patient-based and nodule-based VL occurrence rates were 17.1% and 16.4%, respectively. Larger tumor size, pseudocapsules and smaller bead size were independently associated with VL occurrence. PFS and OS were more prolonged in VL patients than in non-VL patients, and VL independently correlated with better PFS and OS. For liver function, the liver function indexes before and after DEB-TACE were of no difference between VL patients and non-VL patients. Additionally, the incidences of adverse events were similar between VL patients and non-VL patients.Conclusion:  VL occurs in 17.1% of HCC patients treated with DEB-TACE, and it is correlated with larger tumor size, pseudocapsule, smaller bead size, more favorable treatment response and better survival.Keywords: hepatocellular carcinoma, drug-eluting bead transarterial chemoembolization, vascular lake, related factors, efficacy, safety
