19. Integrin‐Linked Kinase in the Development of Gastric Tumors Induced by Helicobacter pylori: Regulation and Prevention Potential.

Abstract

Background: Integrin‐linked kinase (ILK) is crucial in solid tumors by regulating the Hippo‐Yes‐associated protein 1 (YAP) pathway. This study aimed to uncover how Helicobacter pylori influences ILK levels and its role in regulating YAP during H. pylori‐induced gastric cancer. Materials and Methods: GES‐1 cells with stable Ilk knockdown and overexpression and a mouse carcinogenesis model for H. pylori infection were constructed. And ILK, the phosphorylated mammalian STE20‐like protein kinase 1 (MST1), large tumor suppressor 1 (LATS1; S909, T1079), and YAP (S109, S127) were detected in cells, and mice by western blotting, as well as fluorescence intensity of YAP were assayed by immunofluorescence. YAP downstream genes Igfbp4 and Ctgf, the pathological changes and tumor necrosis factor alpha (TNF‐α), interleukin‐6 (IL‐6), interleukin‐1beta (IL‐1β), and nitric oxide (NO) levels in mice gastric tissues were detected by real‐time PCR, H&E, and ELISA assays. Results: In this study, stable Ilk knockdown cells exhibited significantly higher phosphorylated levels of MST1, LATS1, and YAP, as well as increased YAP in the nuclei of GES‐1 cells. Conversely, cells with Ilk overexpression showed opposite results. H. pylori infection led to decreased ILK levels in gastric epithelial cells but increased ILK levels in gastric cancer cell lines (MGC803, SGC7901) and gastric cancer tissues in mice. Treatment with the ILK inhibitor OST‐T315 elevated the phosphorylated MST, LATS1, and YAP levels, and inhibited the mRNA levels of Igfbp4 and Ctgf at 44, 48 week‐aged mice. OST‐T315 also reduced the release of TNF‐α, IL‐6, IL‐1β, and NO, as well as the progression of gastric cancer caused by H. pylori and N‐Nitroso‐N‐methylurea (NMU) treatment. Conclusion: Upon initiation of gastric tumorigenesis signals, H. pylori increases ILK levels and suppresses Hippo signaling, thereby promoting YAP activation and gastric cancer progression. 34. High-Pressure Studies of Flexible Metal-Organic Frameworks and Their Performance for CO2 Adsorption using Infrared Spectroscopy

Abstract

Metal-organic frameworks (MOFs) are crystalline porous materials comprising metal ions/clusters and organic linkers. MOFs feature very large surface area and broad tunability, which distinguish them from traditional CO2 adsorbents. High external pressure can significantly modify the framework structures and CO2 adsorption properties of MOFs. MIL-53(Al) and NH2-MIL-53(Al) exhibit excellent CO2 affinity by forming hydrogen bonds between bridging OH groups and adsorbed CO2. We used in situ infrared spectroscopy to investigate the high-pressure performance of their framework structures and CO2 adsorption capacities. Diamond anvil cell was employed to apply high pressures in gigapascal level. For as-made MIL-53(Al), pressures-induced inter-framework hydrogen bonds between OH groups and free H2BDC molecules were observed. The IR spectra of activated MIL-53(Al) upon compression provided direct evidence of its extraordinary stability compared to as-made and CO2-loaded MIL-53(Al). Pressure-induced intra-framework hydrogen bonding interactions between OH groups and octahedral [AlO6] were observed in activated MIL-53(Al). Moreover, structural modifications of as-made and activated MIL-53(Al) were irreversible upon complete decompression. Pressure-enhanced CO2 adsorption in MIL-53(Al) was demonstrated. Upon complete decompression, considerable CO2 molecules remained in the framework. Activated NH2-MIL-53(Al) exhibited reversible pressure-enhanced intra-framework interactions via two types of hydrogen bonding: one was between -NH2 groups and octahedral [AlO6], the other was between OH groups and octahedral [AlO6]. For CO2-loaded NH2-MIL-53(Al), there were four different high-pressure adsorption sites co-existing upon compression: dimeric adsorption, large-pore adsorption, narrow-pore adsorption, and amino adsorption. We demonstrated that high pressures made the narrow-pore adsorption highly favored over the other three.

Abstract

Vibrio cholerae can be differentiated into epidemic and non-epidemic strains by sorbitol fermentation speed, but little research has been done on its mechanisms. In this study, we investigated differential protein expression of the two strains in response to sorbitol metabolism. V. cholerae strains were cultured in media with and without sorbitol, respectively. Proteins were separated by 2-DE, and those that showed different expression in the two media were identified by MALDI-TOF MS. Fifteen proteins in epidemic strains and 11 proteins in non-epidemic strains showed a different expression in sorbitol medium. Among them, 4 proteins were common to epidemic and non-epidemic strains. Gene sequence analysis showed that some mutations occurred in these proteins between the two strains. Potential functions of these proteins included sugar uptake, amino acid uptake, electron transport, sulfate and thiosulfate transport.

50. Enhancing CO2Adsorption in MIL-53(Al) through Pressure–Temperature Modulation: Insights from Guest–Host Interactions

Abstract

Metal–organic frameworks (MOFs) have garnered significant attention for their exceptional CO2adsorption capabilities. Among them, MIL-53(Al) is uniquely known for its “breathing effect”, a reversible phase transition between large-pore and narrow-pore phases. While previous studies have explored the structural changes in MIL-53(Al) under varying conditions, this work represents an innovative investigation into the simultaneous effects of high pressure and high temperature on the CO2adsorption performance of MIL-53(Al). Utilizing a diamond anvil cell as the high-pressure device, we employed in situ Fourier transform infrared spectroscopy to examine the structural changes in activated MIL-53(Al) under compression and its CO2adsorption performance under specific simultaneous high-pressure and high-temperature conditions. Our findings reveal that heating serves as an effective strategy to augment CO2adsorption by enhancing the mobility of the CO2molecules under high pressure. Remarkably, the CO2adsorption capacity of MIL-53(Al) surged when subjected to pressures from 0.20 to 1.24 GPa and temperatures up to the melting point of CO2. Detailed spectral analysis further elucidated the chemisorptive nature of host–guest interactions between the framework and CO2. These findings significantly advance our understanding of MOFs’ potential for carbon capture across a broad pressure–temperature spectrum.
