1. Data-Driven Identification unravels multiaxial mechanical response of a carbon-black filled elastomer during ageing

Abstract

Under environmental exposure, the mechanical properties of elastomers change due to ageing, all while enduring mechanical service loading conditions. The influence of ageing on the multiaxial mechanical response of elastomers remains an understudied question, lacking exploration in both experimental evidence and modelling proposals. The present study describes an experimental/numerical approach to characterize the multiaxial behaviour of elastomers with consideration of ageing. This technique associates complex experimental tests conducted with a hexapod device, with a Data-Driven Identification (DDI) algorithm. Practically, heterogeneous strain fields are measured by Digital Image Correlation (DIC), and the corresponding stress and energy fields are calculated by DDI. These fields are visualized through three-dimensional maps, encompassing kinematical quantities and strain energy density. These maps convincingly capture the stiffening induced by ageing, in different deformation modes. Finally, the coupling between ageing and multiaxiality is foregathered in a material database that can be fitted for further modelling purposes.

Abstract

Permeability is a fundamental property of porous media. It quantifies the ease with which a fluid can flow under the effect of a pressure gradient in a network of connected pores. Porous materials can be natural, such as soil and rocks, or synthetic, such as a densified network of fibres or open-cell foams. The measurement of permeability is difficult and time-consuming in heterogeneous and anisotropic porous media; thus, a numerical approach based on the calculation of the tensor components on a 3D image of the material can be very advantageous. For this type of microstructure, it is important to perform calculations on large samples using boundary conditions that do not suppress the transverse flows that occur when flow is forced out of the principal directions. Since these are not necessarily known in complex media, the permeability determination method must not introduce bias by generating non-physical flows. A new finite element-based method proposed in this study allows us to solve very high-dimensional flow problems while limiting the biases associated with boundary conditions and the small size of the numerical samples addressed. This method includes a new boundary condition, full permeability tensor identification based on the multiscale homogenization approach, and an optimized solver to handle flow problems with a large number of degrees of freedom. The method is first validated against academic test cases and against the results of a recent permeability benchmark exercise. The results underline the suitability of the proposed approach for heterogeneous and anisotropic microstructures.

Abstract

Abstract The COVID-19 pandemic has affected all domains of human life, including the economic and social fabric of societies. One of the central strategies for managing public health throughout the pandemic has been through persuasive messaging and collective behaviour change. To help scholars better understand the social and moral psychology behind public health behaviour, we present a dataset comprising of 51,404 individuals from 69 countries. This dataset was collected for the International Collaboration on Social & Moral Psychology of COVID-19 project (ICSMP COVID-19). This social science survey invited participants around the world to complete a series of moral and psychological measures and public health attitudes about COVID-19 during an early phase of the COVID-19 pandemic (between April and June 2020). The survey included seven broad categories of questions: COVID-19 beliefs and compliance behaviours; identity and social attitudes; ideology; health and well-being; moral beliefs and motivation; personality traits; and demographic variables. We report both raw and cleaned data, along with all survey materials, data visualisations, and psychometric evaluations of key variables.

11. Model-Free Data-Driven Methods in Mechanics: Material Data Identification and Solvers

Abstract

This paper presents an integrated model-free data-driven approach to solid mechanics, allowing to perform numerical simulations on structures on the basis of measures of displacement fields on representative samples, without postulating a specific constitutive model. A material data identification procedure, allowing to infer strain-stress pairs from displacement fields and boundary conditions, is used to build a material database from a set of mutiaxial tests on a non-conventional sample. This database is in turn used by a data-driven solver, based on an algorithm minimizing the distance between manifolds of compatible and balanced mechanical states and the given database, to predict the response of structures of the same material, with arbitrary geometry and boundary conditions. Examples illustrate this modelling cycle and demonstrate how the data-driven identification method allows importance sampling of the material state space, yielding faster convergence of simulation results with increasing database size, when compared to synthetic material databases with regular sampling patterns., Comment: Revised version

17. Unified numerical process modeling of forming and consolidation for thermoplastic composites with prepreg patches

Abstract

The main idea behind “Quilted Stratum Process” (QSP®) is to create a flat blank made of unidirectional/woven thermoplastic prepreg patches instead of using uniformly shaped prepreg stack as is the case with standard thermostamping process. Thus, using QSP®; one can manufacture components with complex geometries by using nearly rectangular patches while still maintaining a short cycle time similar to the standard thermostamping process. The use of near-rectangular patches results in a significant material saving which is a necessity for a sustainable product development. During standard thermostamping and/or QSP®; the consolidation phase plays a key role in the strength and quality of the final product. This becomes even more important in the case of unidirectional thermoplastic prepregs where mechanisms such as transverse squeeze flow can impact not only the in-plane dimensions of the prepreg but also the fibre orientations within the prepreg. This work presents a unified modeling approach that combines a novel pinching shell element, a new elasto-plastic constitutive model for pinching shell in order to provide a unified solution to simulate both forming and consolidation-flow using a shell-based approach. This unique unified approach of simulating forming and consolidation provides a set of additional outputs such as the through-thickness stress, final deformed shape of the plies including the squeeze flow effect and the changes in the orientation of fibres within the plies during and after the process. This work finally demonstrates how this information can help the manufacturers to design better tooling based on the outcomes of the numerical process simulation in order to achieve a desired product quality. Additionally, one can also steer the final fibre orientation which results from the initial position of the patch, its forming and squeeze flow.

19. Generation of prolactin-inducible protein (Pip) knockout mice by CRISPR/Cas9-mediated gene engineering

Abstract

Prolactin-inducible protein (PIP) is a multifunctional glycoprotein that is highly expressed and found in the secretions of apocrine glands such as salivary, lacrimal, and sweat glands including the mammary glands. PIP has been implicated in various diseases, including breast cancer, gross cystic disease of the breast, keratoconus of the eye, and the autoimmune Sjogren's syndrome. Here we have generated a Pip knockout (KO) mouse using the Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/CRSPR-associated (Cas)9 system. The Cas9 protein and two single guide RNAs targeting specific regions for both exons 1 and 2 of the Pip gene were microinjected into mouse embryos. The deletions and insertions promoted by CRISPR/Cas9 system on the Pip gene successfully disrupted Pip protein coding, as confirmed by PCR genotyping, sequencing, and ultimately Western blot analysis. This mouse model was generated in the inbred C57Bl/6J mouse, which exhibits lower genetic variation. This novel CRISPR Pip KO mouse model will not only be useful for future studies to interrogate the multifunctional role of PIP in physiological processes but will facilitate a broader understanding of the function of PIP in vivo while providing unprecedented insight into its role in a spectrum of diseases attributed to the deregulation of the PIP gene. La proteine inductible par la prolactine (PIP) est une glycoproteine multifonctionnelle hautement exprimee et recueillie dans les secretions de glandes apocrines, comme les glandes salivaires lacrymales et sudoripares, y compris les glandes mammaires. On a montre que la PIP joue un role dans diverses maladies, y compris le cancer du sein, les kystes macroscopiques mammaires, le keratocone oculaire et le syndrome de Sjogren (auto immun). Dans les presents travaux, nous avons obtenu une souris avec repression (KO pour<< knockout>>) de la Pip a l'aide du systeme CRISPR/Cas9. Nous avons injecte de la proteine Cas9 et deux ARN guides uniques ciblant des regions specifiques des exons 1 et 2 du gene Pip a la fois dans des embryons de souris. Les deletions et les insertions favorisees par le systeme CRISPR/Cas9 sur le gene Pip ont permis de rompre le codage de la proteine Pip avec succes, ce qui a ete confirme a l'aide du genotypage par PCR, du sequenqage et enfin de l'immunobuvardage de Western. Nous avons obtenu ce modele de souris chez la souris consanguine C57Bl/6J, laquelle presente un tres faible taux de variations genetiques. Ce nouveau modele de souris KO CRISPR Pip sera non seulement utile pour de futures etudes portant sur le role multifonctionnel de la PIP dans les processus physiologiques, mais il permettra aussi de mieux comprendre le fonctionnement de la PIP in vivo tout apportant des renseignements sans precedent sur son role dans un eventail de maladies attribuees au dereglement du gene PIP. Abstract

Breast cancer (BC) is the most common cancer in women, with metastatic BC being responsible for the highest number of deaths. A frequent site for BC metastasis is the brain. Brain metastasis derived from BC involves the cooperation of multiple genetic, epigenetic, angiogenic, and tumor–stroma interactions. Most of these interactions provide a unique opportunity for development of new therapeutic targets. Potentially targetable signaling pathways are Notch, Wnt, and the epidermal growth factor receptors signaling pathways, all of which are linked to driving BC brain metastasis (BCBM). However, a major challenge in treating brain metastasis remains the blood–brain barrier (BBB). This barrier restricts the access of unwanted molecules, cells, and targeted therapies to the brain parenchyma. Moreover, current therapies to treat brain metastases, such as stereotactic radiosurgery and whole-brain radiotherapy, have limited efficacy. Promising new drugs like phosphatase and kinase modulators, as well as BBB disruptors and immunotherapeutic strategies, have shown the potential to ease the disease in preclinical studies, but remain limited by multiple resistance mechanisms. This review summarizes some of the current understanding of the mechanisms involved in BC brain metastasis and highlights current challenges as well as opportunities in strategic designs of potentially successful future therapies.

Abstract

The title compound, C9H7NO3, crystallizes in the monoclinic (P21) space group. In the crystal, the almost planar molecules display a flattened herringbone arrangement. Stacking molecules are slipped in the lengthwise and widthwise directions and are linked by π–π interactions [d(Cg...Cg = 3.6640 (11) Å]. The structure is characterized by strong C—H...N and weak C—H...O hydrogen bonds, and further stabilized by C–O...π interactions.

Abstract

Permeability is a fundamental property of porous media. It quantifies the ease with which a fluid can flow under the effect of a pressure gradient in a network of connected pores. Porous materials can be natural, such as soil and rocks, or synthetic, such as a densified network of fibres or open-cell foams. The measurement of permeability is difficult and time-consuming in heterogeneous and anisotropic porous media; thus, a numerical approach based on the calculation of the tensor components on a 3D image of the material can be very advantageous. For this type of microstructure, it is important to perform calculations on large samples using boundary conditions that do not suppress the transverse flows that occur when flow is forced out of the principal directions. Since these are not necessarily known in complex media, the permeability determination method must not introduce bias by generating non-physical flows. A new finite element-based method proposed in this study allows us to solve very high-dimensional flow problems while limiting the biases associated with boundary conditions and the small size of the numerical samples addressed. This method includes a new boundary condition, full permeability tensor identification based on the multiscale homogenization approach, and an optimized solver to handle flow problems with a large number of degrees of freedom. The method is first validated against academic test cases and against the results of a recent permeability benchmark exercise. 43. Claudin 1 Is Highly Upregulated by PKC in MCF7 Human Breast Cancer Cells and Correlates Positively with PKCε in Patient Biopsies

Abstract

Recent studies provide compelling evidence to suggest that the tight junction protein claudin 1, aberrantly expressed in several cancer types, plays an important role in cancer progression. Dysregulation of claudin 1 has been shown to induce epithelial mesenchymal transition (EMT). Furthermore, activation of the ERK signaling pathway by protein kinase C (PKC) was shown to be necessary for EMT induction. Whether PKC is involved in regulating breast cancer progression has not been addressed. The PKC activator 12-O-tetradecanoylphorbol 13-acetate (TPA) was used to investigate the effect of PKC activity on claudin 1 transcription and protein levels, subcellular distribution, and alterations in EMT markers in human breast cancer (HBC) cell lines. As well, tissue microarray analysis (TMA) of a large cohort of invasive HBC biopsies was conducted to investigate correlations between claudin 1 and PKC isomers. TPA upregulated claudin 1 levels in all HBC cell lines analyzed. In particular, a high induction of claudin 1 protein was observed in the MCF7 cell line. TPA treatment also led to an accumulation of claudin 1 in the cytoplasm. Additionally, we demonstrated that the upregulation of claudin 1 was through the ERK signaling pathway. In patient biopsies, we identified a significant positive correlation between claudin 1, PKCα, and PKCε in ER+ tumors. A similar correlation between claudin 1 and PKCε was identified in ER− tumors, and high PKCε was associated with shorter disease-free survival. Collectively, these studies demonstrate that claudin 1 and the ERK signaling pathway are important players in HBC progression.

Abstract

Circulating cell-free methyl-DNA (mcfDNA) contains promising cancer markers but its low abundance and possibly diverse origin pose challenges toward the accurate diagnosis of early stage cancers. By whole-genome bisulfite sequencing (WGBS) of cell-free DNA (cfDNA) from about 0.5 mL plasma of mice xenografted with human tumors, we obtained and aligned the reads to the human genome, filtered out the mouse and carrier bacterial sequences, and confirmed the tumor origin of methyl-cfDNA (mctDNA) by methylation-sensitive restriction enzyme digestion prior to species-specific PCR. We estimated that human tumor-specific reads (ctDNA) or mctDNA comprised about 0.29 or 0.01%, respectively of the xenograft mouse cfDNA, and about 0.029 or 0.001% of the cfDNA of human early stage cancer patients. Similar WGBS of early stage (0-II, node- and metastasis-free) breast, lung or colorectal cancer samples identified hundreds of specific DMRs (differentially methylated regions) compared to healthy controls. Their association with tumourigenesis was supported by stage-dependent methylation, tumor suppressor or oncogene clusters, and genes also identified in the xenograft samples. Using 20 three-cancer-common and 17 colorectal cancer-specific DMRs in combination (top 0.0018% of the WGBS methylation clusters) was sufficient to distinguish the stage I colorectal cancers from breast and lung cancers and healthy controls. Our data thus confirmed the tumor origin of mctDNA by sequence specificity, and provide a selection threshold for authentic tumor mctDNA markers toward precise diagnosis of early stage cancers solely by top DMRs in combination.
