Abstract

Item does not contain fulltext, Microtubules are dynamic cytoskeletal elements coordinating and supporting a variety of neuronal processes, including cell division, migration, polarity, intracellular trafficking, and signal transduction. Mutations in genes encoding tubulins and microtubule-associated proteins are known to cause neurodevelopmental and neurodegenerative disorders. Growing evidence suggests that altered microtubule dynamics may also underlie or contribute to neurodevelopmental disorders and neurodegeneration. We report that biallelic mutations in TBCD, encoding one of the five co-chaperones required for assembly and disassembly of the alphabeta-tubulin heterodimer, the structural unit of microtubules, cause a disease with neurodevelopmental and neurodegenerative features characterized by early-onset cortical atrophy, secondary hypomyelination, microcephaly, thin corpus callosum, developmental delay, intellectual disability, seizures, optic atrophy, and spastic quadriplegia. Molecular dynamics simulations predicted long-range and/or local structural perturbations associated with the disease-causing mutations. Biochemical analyses documented variably reduced levels of TBCD, indicating relative instability of mutant proteins, and defective beta-tubulin binding in a subset of the tested mutants. Reduced or defective TBCD function resulted in decreased soluble alpha/beta-tubulin levels and accelerated microtubule polymerization in fibroblasts from affected subjects, demonstrating an overall shift toward a more rapidly growing and stable microtubule population. These cells displayed an aberrant mitotic spindle with disorganized, tangle-shaped microtubules and reduced aster formation, which however did not alter appreciably the rate of cell proliferation. Abstract

Children with autism were examined to determine amounts of mercury (Hg) in their baby hair and the factors that might affect Hg body burden. US children with autism (n = 78) and matched controls (n = 31) born between 1988 and 1999 were studied. Hg in first-cut baby hair was determined using cold vapor atomic fluorescence spectrometry (CVAFS). Twenty samples were split and also measured with Neutron Activation Analysis (NAA). Logistic regression analysis showed that compared to children with higher levels of mercury (above 0.55 mcg g-1), children with lower levels of mercury in their hair (below 0.55 mcg g-1) were 2.5-fold more likely to manifest with autism. Children with autism had similar mercury exposure as controls from maternal seafood and maternal dental amalgams. Children with autism also had 2.5-fold higher incidence of oral antibiotic use during their first 18 months of life. Their mothers were possibly more likely to use oral antibiotics during pregnancy or nursing. The amount of Hg in the baby hair of children with autism showed a significant correlation with the number of maternal dental amalgams. The lower level of Hg in the baby hair of children with autism indicates an altered metabolism of Hg, and may be due to a decreased ability to excrete Hg. This is consistent with usage of higher amounts of oral antibiotics, which are known to inhibit Hg excretion in rats due to alteration of gut flora, and may exert a similar effect in humans. Higher usage of oral antibiotics in infancy may also partially explain the high incidence of chronic gastrointestinal problems seen in individuals with autism. [ABSTRACT FROM AUTHOR]

Abstract

This study was conducted at a large midwestern university to examine the effects of short (approximately seven minutes in length) and longer (approximately 15 minutes in length) training on the number of ideas generated by nominal and brainstorming groups. A main effect for group type was found such that nominal groups outperformed brainstorming groups. A main effect was found for training. Trained groups outperformed untrained groups. Longer training helped brainstorming groups more than short training. Quality of ideas was evaluated. Neither group type nor training had an impact on the quality of ideas. [ABSTRACT FROM AUTHOR]

42. Distinguishing differences in pathways to resilience among Canadian youth.

Abstract

As part of an 11-country qualitative study of resilience among at-risk youth, 19 Aboriginal and non-Aboriginal Canadian adolescents were interviewed. In this paper, we report on the Canadian youths' culturally specific and generic strategies to cope with adversity. Findings suggest that the youths' resilience, or capacity to cope under stress, reflects different degrees of access to 7 mental health-enhancing experiences (we term these 'tensions'): access to material resources; access to supportive relationships; development of a desirable personal identity; experiences of power and control; adherence to cultural traditions; experiences of social justice; and experiences of a sense of cohesion with others. Abstract

This article investigates the mercury content in commercially available processed tuna products and unprocessed fish fillet and seafood products. Thermal decomposition, amalgamation and atomic absorption spectrometry were used to analyze the mercury level. The mercury content in the products ranged from 0.00351 to 1.85 mg/kg. The determined mercury concentrations for the catfish fillet, tilapia fillet, shrimp and scallop samples were all relatively low compared to the other unprocessed seafood products. The tuna and swordfish fillet samples had higher mercury content relative to the other test samples.

Abstract

We studied the practice patterns regarding intravenous (i.v.) ondansetron in children receiving stem cell transplants (SCT) at The Children’s Hospital, Boston to identify cost efficiencies. The pharmacy provided information on material and preparation costs on 36 patients who received i.v. ondansetron during 41 SCT in 1995. We examined the effects of frequency, duration, and route of administration on costs. There were 498 days of ondansetron administration costing $49 083 (95$). Tremendous variation existed in frequency and duration with one third receiving i.v. ondansetron once daily, despite published evidence of equivalence of once a day and divided dosing. A switch to once daily i.v. dosing for all patients would have resulted in 28% savings. The median duration of use was 11 days (range 1–48); placing a cap for 7–10 days based on the length of SCT conditioning regimens, would produce savings of 48–60% over current use. By shifting administration route from i.v. to oral, a savings of 67% over current use, without a cap on duration, would be realized. Identifying areas for cost savings can be achieved after thorough analysis of all the component costs. We demonstrated that significant cost reductions could be realized by simple changes in prescribing practices without jeopardizing efficacy. These savings are achieved by standardizing dosing interval, route of administration and duration of treatment without altering daily dosage or access to an effective antiemetic. Abstract

In this research the influence of mixed and consistent chemisorption of differently sized molecules of quaternary ammonium compounds (alkamon and triamon) on disperse aluminum on its properties. As a part of the research the lubricants containing modified aluminum were tested. It was discovered that as the hydrophobicity of Al-additives increases, the force of friction (Ffr) and coefficient of friction (f) decrease. It was discovered that Al-additives with external layer of ethylhydridesiloxane and only one underlayer of triamon has the best anti-frictional effect. The addition more T-underlayers (from 1 to 3) did not give a significant improvement.
