Abstract

Two-component signaling (TCS) is the primary means by which bacteria sense and respond to the environment. TCS involves two partner proteins working in tandem, which interact to perform cellular functions whereas limiting interactions with non-partners (i.e., cross-talk). We construct a Potts model for TCS that can quantitatively predict how mutating amino acid identities affect the interaction between TCS partners and non-partners. The parameters of this model are inferred directly from protein sequence data. This approach drastically reduces the computational complexity of exploring the sequence-space of TCS proteins. As a stringent test, we compare its predictions to a recent comprehensive mutational study, which characterized the functionality of 204 mutational variants of the PhoQ kinase in Escherichia coli We find that our best predictions accurately reproduce the amino acid combinations found in experiment, which enable functional signaling with its partner PhoP. These predictions demonstrate the evolutionary pressure to preserve the interaction between TCS partners as well as prevent unwanted cross-talk. Further, we calculate the mutational change in the binding affinity between PhoQ and PhoP, providing an estimate to the amount of destabilization needed to disrupt TCS.

The study further explores whether four variables: 1) number of units of college credit completed, 2) anticipated grade in the class, 3) gender, and 4) age are related to students' preference for mode of instruction.

10. Fluctuation-induced instabilities in front propagation up a comoving reaction gradient in two dimensions

Abstract

We study two-dimensional (2D) fronts propagating up a comoving reaction rate gradient in finite number reaction-diffusion systems. We show that in a 2D rectangular channel, planar solutions to the deterministic mean-field equation are stable with respect to deviations from planarity. We argue that planar fronts in the corresponding stochastic system, on the other hand, are unstable if the channel width exceeds a critical value. Furthermore, the velocity of the stochastic fronts is shown to depend on the channel width in a simple and interesting way, in contrast to fronts in the deterministic mean-field equation. Thus fluctuations alter the behavior of these fronts in an essential way. These effects are shown to be partially captured by introducing a density cutoff in the reaction rate. Moreover, some of the predictions of the cutoff mean-field approach are shown to be in quantitative accord with the stochastic results.

Abstract

The Advanced Plant Habitat (APH) was installed on the International Space Station (ISS) in October 2017. Following a successful EVT (Experiment Verification Test) study at Kennedy Space Center (KSC), using Arabidopsis lines with varying levels of lignin, two inaugural studies were carried out on ISS in 2018 under the same experimental design, with the corresponding ground controls at KSC. The APH for this study deploys a substrate-based root module designed for plant growth in microgravity. Upon experiment initiation (such as for the EVT), the root module is primed (liquid imbibition) by flooding the root zone to initiate seed germination and to remove air from the porous tubing and particulate media. In the APH ISS inaugural study, the speed of supplying water to initially dry media was found to adversely affect the overall moisture distribution within the root module in microgravity (but not at 1g). Non-destructive estimations of Arabidopsis plant growth were carried out by monitoring changes in rosette leaf area on a daily basis. These data indicated that the original priming procedure caused patchy moisture distribution that affected plant growth and survival. An improved methodology for priming the second root module of PH-01 was devised and implemented in the second experiment. Leaf area and color estimates suggested that the modified priming scheme improved moisture distribution and plant growth. These data, when compared with the EVT study, suggest that nondestructive measurements of plant growth can aid towards optimization of plant growth conditions in microgravity.

Item does not contain fulltext, Biomedical science is rapidly developing in terms of more transparency, openness and reproducibility of scientific publications. This is even more important for all studies that are based on results from basic semen examination. Recently two concordant documents have been published: the 6th edition of the WHO Laboratory Manual for the Examination and Processing of Human Semen, and the International Standard ISO 23162:2021. With these tools, we propose that authors should be instructed to follow these laboratory methods in order to publish studies in peer-reviewed journals, preferable by using a checklist as suggested in an Appendix to this article.

2017 © The Authors Quantum entanglement involving coherent superpositions of macroscopically distinct states is among the most striking features of quantum theory, but its realization is challenging because such states are extremely fragile. Using a programmable quantum simulator based on neutral atom arrays with interactions mediated by Rydberg states, we demonstrate the creation of “Schrödinger cat” states of the Greenberger-Horne-Zeilinger (GHZ) type with up to 20 qubits. Our approach is based on engineering the energy spectrum and using optimal control of the many-body system. We further demonstrate entanglement manipulation by using GHZ states to distribute entanglement to distant sites in the array, establishing important ingredients for quantum information processing and quantum metrology.

Abstract

Dynamic stabilization of an array Large-scale systems comprising one-dimensional chains and two-dimensional arrays of excited atoms held in a programmable optical lattice are a powerful platform with which to simulate emergent phenomena. Bluvstein et al. built an array of up to 200 Rydberg atoms and subjected the system to periodic excitation. Under such driven excitation, they found that the array of atoms stabilized, freezing periodically into what looked like time crystals. Understanding and controlling the dynamic interactions in quantum many-body systems lies at the heart of contemporary condensed matter physics and the exotic phenomena that can occur. Synthesizing topological order Topologically ordered matter exhibits long-range quantum entanglement. However, measuring this entanglement in real materials is extremely tricky. Now, two groups take a different approach and turn to synthetic systems to engineer the topological order of the so-called toric code type (see the Perspective by Bartlett). Satzinger et al . used a quantum processor to study the ground state and excitations of the toric code. Semeghini et al . detected signatures of a toric code–type quantum spin liquid in a two-dimensional array of Rydberg atoms held in optical tweezers. —JS

30. Growth Chambers on the International Space Station for Large Plants

Abstract

The International Space Station (ISS) now has platforms for conducting research on horticultural plant species under LED lighting, and those capabilities continue to expand. The 'Veggie' vegetable production system was deployed to the ISS as an applied research platform for food production in space. Veggie is capable of growing a wide array of horticultural crops. It was designed for low power usage, low launch mass and stowage volume, and minimal crew time requirements. The Veggie flight hardware consists of a light cap containing red (630 nm), blue, (455 nm) and green (530 nm) LEDs. Interfacing with the light cap is an extendable bellows/baseplate for enclosing the plant canopy. A second large plant growth chamber, the Advanced Plant Habitat (APH), is will fly to the ISS in 2017. APH will be a fully controllable environment for high-quality plant physiological research. APH will control light (quality, level, and timing), temperature, CO2, relative humidity, and irrigation, while scrubbing any cabin or plant-derived ethylene and other volatile organic compounds. Additional capabilities include sensing of leaf temperature and root zone moisture, root zone temperature, and oxygen concentration. The light cap will have red (630 nm), blue (450 nm), green (525 nm), far red (730 nm) and broad spectrum white LEDs (4100K). There will be several internal cameras (visible and IR) to monitor and record plant growth and operations. Veggie and APH are available for research proposals.

Abstract

Epithelial-mesenchymal transition (EMT) encompasses dynamic changes in cellular organization from epithelial to mesenchymal phenotypes, which leads to functional changes in cell migration and invasion. EMT occurs in a diverse range of physiological and pathological conditions and is driven by a conserved set of inducing signals, transcriptional regulators and downstream effectors. With over 5,700 publications indexed by Web of Science in 2019 alone, research on EMT is expanding rapidly. This growing interest warrants the need for a consensus among researchers when referring to and undertaking research on EMT. This Consensus Statement, mediated by 'the EMT International Association' (TEMTIA), is the outcome of a 2-year-long discussion among EMT researchers and aims to both clarify the nomenclature and provide definitions and guidelines for EMT research in future publications. We trust that these guidelines will help to reduce misunderstanding and misinterpretation of research data generated in various experimental models and to promote cross-disciplinary collaboration to identify and address key open questions in this research field. While recognizing the importance of maintaining diversity in experimental approaches and conceptual frameworks, we emphasize that lasting contributions of EMT research to increasing our understanding of developmental processes and combatting cancer and other diseases depend on the adoption of a unified terminology to describe EMT. In this Consensus Statement, the authors (on behalf of the EMT International Association) propose guidelines to define epithelial-mesenchymal transition, its phenotypic plasticity and the associated multiple intermediate epithelial-mesenchymal cell states. Clarification of nomenclature and definitions will help reduce misinterpretation of research data generated in different experimental model systems and promote cross-disciplinary collaboration.
