In this review, we discuss the mechanisms of generation of membrane-bound protons using different energy sources in model and natural systems. Analysis of these mechanisms revealed that all three types of reactions include the same principal stage, which is dissociation of electrically neutral Brønsted acids at the interface during transition from the hydrophobic phase to water with a low dielectric constant. Special attention is paid to the fact that in one of the analyzed model systems, membrane-bound protons provide energy for the reaction of ATP synthesis. Similar mechanism for the generation of membrane-bound protons has been found in natural membranes involved in oxidative phosphorylation, in particular, on the membranes of mitoplasts and mitochondria. The energy of oxidative reactions required for ATP synthesis, is stored at the intermediate stage not only in the form of transmembrane electrochemical potential of protons, but also and perhaps mostly, as protons attached to the inner mitochondrial membrane. The process of energy storage in mitochondria is linked to the transfer of protons that simultaneously perform two functions. In this review, we discuss the principles of regulation and synchronization of metabolic processes in mammalian cells using a two-component model of cell metabolism consisting of a controlling signaling system that regulates major enzymatic cascades and executive metabolic system that directly performs biosynthetic reactions. This approach has allowed us to distinguish two transitional metabolic states (from catabolism to anabolism and vice versa) accompanied by major rearrangements in the signaling system. The signaling system of natural amino acids was selected, because amino acids are involved in both signaling and executive metabolic subsystems of general cell metabolism. We have developed a graphical representation of metabolic events that allowed us to demonstrate the succession of processes occurring in both metabolic subsystems during complete metabolic cycle in a non-dividing cell. An important revealed feature of the amino acid signaling system is that the signaling properties of amino acid are determined not only by their molecular structure, but also by the location within the cell. Four major signaling groups of amino acids have been identified that localize to lysosomes, mitochondria, cytosol, and extracellular space adjacent to the plasma membrane. Although these amino acids groups are similar in the composition, they have different receptors. We also proposed a scheme for the metabolism regulation by amino acids signaling that can serve as a basis for developing more complete spatio-temporal picture of metabolic regulation involving a wide variety of intracellular signaling cascades.

The structure of the ATP synthase of rat’s heart mitochondria is determined by cryoelectron tomography (cryo-ET). The experiments are carried out on fragments of mitochondrial membranes without the additional purification of samples, which allows us to study the mitochondrial enzymes in the natural phospholipid and protein environment. The use of subtomographic averaging yields the structure of the ATP synthase with a resolution of about 13 A, as well as the structure of a stable dimer of ATP synthases with a resolution of about 15 A. The classification of the obtained subtomograms containing ATP synthases showed that this enzyme is always located on the bends of the phospholipid bilayer, while there are two groups of ATP synthase dimers that differ in the membrane curvature in the regions of their localization. On the obtained three-dimensional structures, both extramembrane and intramembrane synthase subunits are quite clearly distinguishable, which indicates the prospects of the chosen methodological approach for studying polyenzyme systems in the natural environment.

The 'standard' fluid-mosaic membrane model can provide a framework for the operation of the photosynthetic and respiratory electron transport systems, the generation of the proton motive force (pmf) and its utilization for ATP synthesis according to the chemiosmotic theory. However, this model, with the bilayer organization of all lipid molecules, assigns no function to non-bilayer lipids - while in recent years it became clear that the two fundamental energy transducing membranes of the biosphere, chloroplast thylakoid membranes (TMs) and inner mitochondrial membranes (IMMs), contain large amounts of non-bilayer (non-lamellar) lipid phases. In this review, we summarize our understanding on the role of non-lamellar phases in TMs and IMMs: (i) We propose that for these membrane vesicles the dynamic exchange model (DEM) provides a more suitable framework than the 'standard' model; DEM complements the 'standard' model by assuming the co-existence of bilayer and non-bilayer phases and their interactions, which contribute to the structural dynamics of the membrane systems and safe-guard the membranes' high protein:lipid ratios. (ii) Non-bilayer phases play pivotal roles in membrane fusion and intermembrane lipid exchanges - essential processes in the self-assembly of these highly folded intricate membranes. (iii) The photoprotective, lipocalin-like lumenal enzyme, violaxanthin de-epoxidase, in its active state requires the presence of non-bilayer lipid phase. (iv) Cardiotoxins, water-soluble polypeptides, induce non-bilayer phases in mitochondria. (v) ATP synthesis, in mammalian heart IMMs, is positively correlated with the amount of non-bilayer packed lipids with restricted mobility. (vi) The hypothesized sub-compartments, due to non-lamellar phases, are proposed to enhance the utilization of pmf and might contribute to the recently documented functional independence of individual cristae within the same mitochondrion. Further research is needed to identify and characterize the structural entities associated with the observed non-bilayer phases; and albeit fundamental questions remain to be elucidated, non-lamellar lipid phases should be considered on a par with the bilayer phase, with which they co-exist in functional TMs and IMMs.

Toxic effect of 2-ethylnorbornane (2-ethyl(bicyclo[2.2.1]heptane) (EBH)) on bacteria has been studied using the E. coli pRecA-lux and E. coli pKatG- lux cells as lux-biosensors. It was shown that the addition of EBH to the incubation medium leads to death and growth retardation, high level oxidative stress and DNA damage in E. coli cells. The existence of a complete oxidative phosphorylation system (OXPHOS) supercomplex including both electron transport system and ATP synthases has long been assumed based on functional evidence. However, no structural confirmation of the docking between ATP synthase and proton pumps has been obtained. In this study, cryo-electron tomography was used to reveal the supramolecular architecture of the rat heart mitochondria cristae during ATP synthesis. Respirasome and ATP synthase structure in situ were determined using subtomogram averaging. The obtained reconstructions of the inner mitochondrial membrane demonstrated that rows of respiratory chain supercomplexes can dock with rows of ATP synthases forming oligomeric ordered clusters. These ordered clusters indicate a new type of OXPHOS structural organization. It should ensure the quickness, efficiency, and damage resistance of OXPHOS, providing a direct proton transfer from pumps to ATP synthase along the lateral pH gradient without energy dissipation.

Here we present the study of the genotoxic effect of a 2,2’-bis(bicyclo[2.2.1]heptane) (BBH), which is promising as a fuel component for liquid rocket engines. The use ofEscherichia coli lux-biosensors showed that in addition to DNA damage causing SOS-response, there is also an oxidative effect on cells. The greatest toxicity is determined by the mechanism of formation of superoxide anion radical and is detected by the lux biosensorE. colipSoxS-lux, in which the genes of bacterial luciferases are transcriptionally crosslinked with the promoter of thesoxSgene. It is assumed that the oxidation of BBH leads to the formation of reactive oxygen species, which should give the main contribution to the toxicity of this substance.

The toxic effect of strained hydrocarbon 2,2'-bis (bicyclo[2.2.1]heptane) (BBH) was studied using whole-cell bacterial lux-biosensors based on Escherichia coli cells in which luciferase genes are transcriptionally fused with stress-inducible promoters. It was shown that BBH has the genotoxic effect causing bacterial SOS response however no alkylating effect has been revealed. In addition to DNA damage, there is an oxidative effect causing the response of OxyR/S and SoxR/S regulons. The most sensitive to BBH lux-biosensor was E. coli pSoxS-lux which reacts to the appearance of superoxide anion radicals in the cell. It is assumed that the oxidation of BBH leads to the generation of reactive oxygen species, which provide the main contribution to the genotoxicity of this substance.

In this work systematization (classification) of biochemical and physiological processes that cause disorders in the human body during the development of diabetes mellitus is carried out. The development of the disease is considered as the interaction and mutual reinforcement of two groups of parallel processes. The first group has a molecular nature and it is associated with impairment of ROS-regulation system which includes NADPH oxidases, RAGE receptors, mitochondria, cellular peroxireductase system and the immune system. The second group has a pathophysiological nature and it is associated with impairment of microcirculation and liver metabolism. The analysis of diabetes biochemistry based on different published references yields a creation of a block diagram evaluating the disease development over time. Two types of autocatalytic processes were identified: autocatalysis in the cascade of biochemical reactions and "cross-section" catalysis, in which biochemical and pathophysiological processes reinforce each other. The developed model has shown the possibility of using pharmacologically active natural metabolite glycine as a medicine inhibiting the development of diabetes. Despite the fact that glycine is a substitute amino acid the drop in the glycine blood concentration occurs even in the early stages of diabetes development and can aggravate the disease. It is shown that glycine is a potential blocker of key autocatalytic cycles, including biochemical and pathophysiological processes. The analysis of the glycine action based on the developed model is in complete agreement with the results of clinical trials in which glycine has improved blood biochemistry of diabetic patients and thereby it prevents the development of diabetic complications.

Background: Electron transport chain (ETC) complexes, pyruvate dehydrogenase complex (PDC), and α-ketoglutarate dehydrogenase complex (KGDC) are important elements in mitochondrial metabolism. The localization of the aforementioned protein complexes differs since oxidative phosphorylation complexes are membrane proteins, while dehydrogenase complexes (DCs) are contained in the mitochondrial matrix. Our previous cryo-electron tomography (cryo-ET) studies showed the existence of a full oxidative phosphorylation system supercomplex consisting of ETC complexes and ATP synthases (Nesterov et al., 2021). Literature data also shows the binding of fatty acid oxidation enzymes to ETC complex I (Wang et al., 2010). Although it has long been shown that PDCs can bind to complex I (Sumegi et al., 1984) in vitro, this has not been visualized directly in mitochondria and the binding mechanisms are still unknown. Methods: The mitochondria were isolated from Wistar rat heart ventricles according to a standard procedure (Nesterov et al., 2021). The dense mitochondrial suspension was diluted to ~0.3mg/ml in a respiration medium. Phosphorylation was started 10 minutes prior to vitrification. Experimental data was obtained by cryo-ET using Titan Krios and processed with IMOD and RELION. Results: The tomograms show that the significant part of DCs is localized near the inner membrane of partially destroyed mitochondria in an array-like fashion. Sole PDCs and KGDCs can be identified on the images and their position appears to be close to ETC complex I. Subtomogram averaging of close to the membrane DCs showed that there is no specific density between them, suggesting that they are not linked with identical proteins or that this link may be soft. Significant damage to the mitochondrial membrane leads to the formation of membrane-unbound DCs fraction. It suggests that coupling of DCs with ETC complexes can be controlled in vivo by the topology of the inner mitochondrial membrane and the volume of the mitochondrial matrix. Conclusion: The obtained results show a possibility of unprecedentedly large multienzyme complex formation, including almost all main mitochondrial metabolic systems. Although cryo-ET of partially destroyed mitochondria showed close localization of PDC and KGDC to complex I, further studies are required in intact mitochondria. The mechanism of their binding also remains an open question.

According to previous studies, ADP/ATP carrier (AAC) can possibly exist as a monomer or in a dimer state in the inner mitochondrial membrane; however, the question on its functional oligomeric state is still open. The aim of the present work is to establish the external factors that could control the functional oligomeric state of AAC (i.e., monomer or dimer). The study is based on the results of our previous work, which revealed that the volume regulation system of mitochondria (MVRS) affects the oxidative phosphorylation (OXPHOS) system: MVRS could transfer OXPHOS system functioning in a state of supercomplex. Consequently, one may expect that the volume regulation system could also control the functional state of AAC during phosphorylation. Here, on rat liver mitochondria we show that, depending on the incubation medium tonicity, AAC functions in two different ways: either as a monomer (in hypotonic and isotonic media) or as a dimer (in a hypertonic medium). Thus, the transition between the monomeric and dimeric forms of AAC is regulated by MVRS, as well as by functioning of OXPHOS. We conclude that the structural reorganization of AAC is associated with the entire OXPHOS reorganization into a supercomplex. It was also found that dimerization of AAC can occur not only due to the action of MVRS (in hypotonic media) but also under hypoxic conditions.

This work is focused on the physicochemical nature and functional meaning of structural alterations in mitochondrial membranes. These alterations are observed at subnormal temperatures for homeothermic animals. Using pyrene as a fluorescent probe, some structural transitions in annular lipids were detected at 19 and 25°C, which are identified as the pre-transition and main phase transition temperatures. The observed changes are consistent with abrupt alterations in the efficiency of tryptophan fluorescence quenching by pyrene that occur at the same temperatures. The data imply considerable changes in the protein–lipid contact area. The effects are observed under low-amplitude mitochondrial swelling in media with lowered tonicity. Since the transition of the phosphorylating system into the supercomplex state has been previously shown under these conditions, it allows us to assume a relationship between the observed membrane structural alterations and the supercomplex formation. Measurements of the respiration rate in mitochondria in a hypoosmotic medium show that the activation energy of the rate-limiting step in the process of ATP synthesis changes abruptly at the temperature of the phase transition of annular lipids (25°C). Analysis of the literature data indicates that a similar abnormal low-temperature abrupt change in activation energy in the reaction of ATP synthesis around 25°C is observed for a variety of animal species. Hence, the low-temperature structural alterations in membranes of warm-blooded animals should have general biological importance. A comparison of the results obtained in our study and the literature data led, therefore, to a qualitative description of the physiochemical nature of the observed membrane alterations. Similar to the membrane-raft model, a model of supercomplex formation in mitochondria is proposed.

The purpose of this work was to study the regulative role of the glutamate receptor found earlier in the brain mitochondria. In the present work a glutamate-dependent signaling system with similar features was detected in mitochondria of the heart. The glutamate-dependent signaling system in the heart mitochondria was shown to be suppressed by γ-aminobutyric acid (GABA). The GABA receptor presence in the heart mitochondria was shown by golding with the use of antibodies to α- and β-subunits of the receptor. The activity of glutamate receptor was assessed according to the rate of synthesis of hydrogen peroxide. The glutamate receptor in mitochondria could be activated only under conditions of hypoxic stress, which in model experiments was imitated by blocking Complex I by rotenone or fatty acids. The glutamate signal in mitochondria was shown to be calcium- and potential-dependent and the activation of the glutamate cascade was shown to be accompanied by production of hydrogen peroxide. It was discovered that H2O2 synthesis involves two complexes of the mitochondrial electron transfer system – succinate dehydrogenase (SDH) and fatty acid dehydrogenase (ETF:QO). Thus, functions of the glutamate signaling system are associated with the system of respiration-glycolysis switching (the Pasteur-Crabtree) under conditions of hypoxia.

The purpose of this study is to demonstrate the presence of three more receptors in mitochondria. Two N-methyl- d -aspartate receptor (NMDAR) subunits (NR1 and NR2B) are found by protein immunoblot and immunogold labeling in mitochondria fraction isolated from rat heart. These data allow supposing NMDAR presence and functioning in the inner mitochondrial membrane. There are no signs of receptor presence obtained in heart tissue lysate, that indicates the receptor localization exactly in mitochondria. The possible receptor functions discussed are its participation in calcium transport and in excitation-metabolism coupling. Besides, preliminary evidence is obtained of GABAA and GABAB receptors presence in heart mitochondria. One can surmise their role in metabolism regulation and their possible co-operation with NMDAR just as in the nervous system.

Continuous generation of hydrogen peroxide catalyzed by low concentrations of 1,1-dimethylhydrazine (heptyl)--a rocket fuel component--in air saturated water was shown by the method of enhanced chemiluminescence in the system of luminol-p-iodophenol-peroxidase. The concentration dependence and the influence of heat and light on the formation of hydrogen peroxide in the water under the influence of dimethylhydrazine at concentrations considerably lower than maximum allowable concentrations were studied, and the physical-chemical mechanism of this process was considered. It is supposed that dimethylhydrazine at ultra-low concentrations is associated with air nanobubbles and represents a long-lived complex performing catalysis of hydrogen peroxide formation under the influence of heat and light. We put forward the new concept of.toxicity of dimethylhydrazine at very low concentrations due to violation of homeostasis of reactive oxygen species formation in aqueous solutions entering the body of humans and animals.

Membranotropic properties of the surface-active reagent 2,4,6-trichloro-3-pentadecylphenol (TCP-C15) have been studied. In preliminary investigations it was found that TCP-C15 acts as an uncoupler of oxidative phosphorylation, selectively interacting with a fraction of non-equilibrium membrane-bound protons that forms during the work of proton pumps. In this work we obtained the following results: (1) additional validation of the conclusions drawn from the previous experiments was acquired on modified preparations of liver mitochondria, in which the fraction of non-equilibrium membrane-bound protons did not form; (2) it was shown that binding of TCP-C15 to the liposome surface increases its negative ζ-potential; this proves that TCP-C15 binds to the surface of membranes in the negatively charged state; (3) on the BLM model it was found that under constant electric field (90 mV) TCP-C15 not only induces the transmembrane transfer of charge, but also induces a specific hydrogen ion affinity to the surface of lipid bilayer; (4) strong evidence of the ionophore activity of TCP-C15 was obtained on the same model; this allowed us to get a full scheme of uncoupling mechanism of TCP-C15 on mitochondria and to explain the action of a catalyst (HEPES, 20 mM), which blocks this mechanism.

Cardiolipin (CL) is an anionic phospholipid at the inner mitochondrial membrane (IMM) that facilitates the formation of transient non-bilayer (non-lamellar) structures to maintain mitochondrial integrity. CL modulates mitochondrial functions including ATP synthesis. However, the biophysical mechanisms by which CL generates non-lamellar structures and the extent to which these structures contribute to ATP synthesis remain unknown. We hypothesized that CL and ATP synthase facilitate the formation of non-bilayer structures at the IMM to stimulate ATP synthesis. By using 1H-NMR and 31P-NMR techniques, we observed that increasing the temperature (8 °C to 37 °C), lowering the pH (3.0), or incubating intact mitochondria with CTII - an IMM-targeted toxin that increases the formation of immobilized non-bilayer structures - elevated the formation of non-bilayer structures to stimulate ATP synthesis. The F0 sector of the ATP synthase complex can facilitate the formation of non-bilayer structures as incubating model membranes enriched with IMM-specific phospholipids with exogenous DCCD-binding protein of the F0 sector (DCCD-BPF) elevated the formation of immobilized non-bilayer structures to a similar manner as CTII. Native PAGE assays revealed that CL, but not other anionic phospholipids, specifically binds to DCCD-BPF to promote the formation of stable lipid-protein complexes. Mechanistically, molecular docking studies identified two lipid binding sites for CL in DCCD-BPF. We propose a new model of ATP synthase regulation in which CL mediates the formation of non-bilayer structures that serve to cluster protons and ATP synthase complexes as a mechanism to enhance proton translocation to the F0 sector, and thereby increase ATP synthesis.

Mitochondrial reactive oxygen species regulate many important biological processes. We studied H2O2 formation by nonsynaptic brain mitochondria in response to the addition of low concentrations of glutamate, an excitatory neurotransmitter. We demonstrated that glutamate at concentrations from 10 to 50 μM stimulated the H2O2 generation in mitochondria up to 4-fold, in a dose-dependent manner. The effect of glutamate was observed only in the presence of Ca2+ (20 μM) in the incubation medium, and the rate of calcium uptake by the brain mitochondria was increased by up to 50% by glutamate. Glutamate-dependent effects were sensitive to the NMDA receptor inhibitors MK-801 (10 μM) and D-AP5 (20 μM) and the inhibitory neurotransmitter glycine (5 mM). We have shown that the H2O2 formation caused by glutamate is associated with complex II and is dependent on the mitochondrial potential. We have found that nonsynaptic brain mitochondria are a target of direct glutamate signaling, which can specifically activate H2O2 formation through mitochondrial respiratory chain complex II. The H2O2 formation induced by glutamate can be blocked by glycine, an inhibitory neurotransmitter that prevents the deleterious effects of glutamate in brain mitochondria.

According to the Williams model, the work of mitochondrial respiratory H+ pumps gives rise to a fraction of membrane-bound protons (R-protons) that have excess free energy, which is used in the reaction of ATP synthesis. We have earlier managed to detect such a fraction in mitochondria and mitoplasts and to rigorously show (for mitoplasts) that the non-equilibrium R-proton fraction is localized on the surface of the inner membrane. Here we show that a surface-active compound 2,4,6-trichloro-3-pentadecylphenol anion (TCP-C15) selectively interacts with the R-proton fraction, and describe in detail its influence on mitochondrial respiration under conditions of R-proton generation. We also report endogenous regulation of the R-proton fraction volume, which is performed by the phosphate transport system. The results are discussed in terms of the local coupling model.

Glycine stabilizes energetics of brain mitochondria under conditions of brain hypoxia in vivo modeled by ligation of the common carotid artery in rats. Hypoxia reduced respiratory control in brain cortex mitochondria from 7.7 ± 0.5 to 4.5 ± 0.3. Preliminary oral administration of glycine almost completely prevented this decrease. In both in vitro models of hypoxia, similar phosphorylation disturbances were detected in both cortical slices and isolated brain mitochondria; they were effectively prevented by glycine. Hypoxia activates H(2)O(2) generation in mitochondrial suspension. The process is significantly reduced in the presence of 5 mM glycine. It is concluded that both in the model of hypoxia in vivo and during in vitro modeling of hypoxia in cortical slices and mitochondria, glycine acts as a protector inhibiting generation of reactive oxygen species in mitochondria and preventing energetics disturbances in brain mitochondria.

In our previous studies we have described the non-equilibrium binding of hydrogen ions to the membranes upon induction of transmembrane proton flux in the model system (BLM) and in mitochondria. In 2009 the investigation program aimed to find uncouplers selectively interacting with the non-equilibrium bound protons was started. This study is devoted to a new representative of this class of uncouplers. The effect of respiratory stimulation evoked by this compound can be suppressed by 50–90% through the elimination of non-equilibrium fraction of protons associated with the outer side of the inner mitochondrial membrane. The peculiarity of this compound results from its belonging to the class of quinones with a high affinity to lipid membranes. It becomes a weak acid (hydroquinone) only after reduction. The formation of hydroquinone in the mitochondria determines the observed effect of respiratory stimulation. Thus, the artificial induction of interaction between redox-reactions and reactions of proton association and dissociation occurring on the surface of the inner mitochondrial membrane was demonstrated for the first time.

The existing high-resolution methods for obtaining structural parameters of biological membrane objects are associated, as a rule, with strong physical and chemical actions on the soft membrane structure. Therefore the results of such experiments have an ambiguity arising from the changes in the structure of objects in the process of measurements. Investigations of living membrane systems retaining their main biological functions are a considerable challenge. Our object of study was the structure of mitochondria by small-angle neutron scattering (SANS). Functioning mitochondria carries out the ATP synthesis (source of energy for cellular activities) that is realized by the oxidative phosphorylation system located in the inner mitochondrial membrane. In the 1970s, small-angle X-ray scattering was applied to investigate the structure of intact mitochondria [1–2], but there are no neutron investigations. As compared to X-rays, thermal neutrons have less damaging effect on biological objects and are sensitive...

A 5-min exposure of air-saturated bidistilled water to low-intensity laser infrared radiation at the wavelength of the electronic transition of dissolved oxygen to the singlet state ((3)∑(g)(-)→ (1)Δ(g)) induces, after a long latent period, auto-oscillations of water luminescence in the blue-green region, which last many hours. Laser irradiation causes the accumulation of hydrogen peroxide, which depends on the concentration of dissolved oxygen. The auto-oscillations do not arise if water is irradiated beyond the oxygen absorption band and if the oxygen is removed from water. The wavelet transform analysis of luminescence records indicates that there are two characteristic periods of pulsations of about 300 and 1150 s. The results obtained suggest that auto-oscillations are triggered by photoinduced singlet oxygen (1)Δ(g), and this phenomenon is closely related to formation of hydrogen peroxide.

A unique phenomenon of mitochondria-targeted protonophores is described. It consists in a transmembrane H + -conducting fatty acid cycling mediated by penetrating cations such as 10-(6’-plastoquinonyl)decyltriphenylphosphonium (SkQ1) or dodecyltriphenylphosphonium (C 12 TPP). The phenomenon has been modeled by molecular dynamics and directly proved by experiments on bilayer planar phospholipid membrane, liposomes, isolated mitochondria, and yeast cells. In bilayer planar phospholipid membrane, the concerted action of penetrating cations and fatty acids is found to result in conversion of a pH gradient (ΔpH) to a membrane potential (Δ ψ ) of the Nernstian value (about 60 mV Δ ψ at ΔpH = 1). A hydrophobic cation with localized charge (cetyltrimethylammonium) failed to substitute for hydrophobic cations with delocalized charge. In isolated mitochondria, SkQ1 and C 12 TPP, but not cetyltrimethylammonium, potentiated fatty acid-induced ( i ) uncoupling of respiration and phosphorylation, and ( ii ) inhibition of H 2 O 2 formation. In intact yeast cells, C 12 TPP stimulated respiration regardless of the extracellular pH value, whereas a nontargeted protonophorous uncoupler (trifluoromethoxycarbonylcyanide phenylhydrazone) stimulated respiration at pH 5 but not at pH 3. Hydrophobic penetrating cations might be promising to treat obesity, senescence, and some kinds of cancer that require mitochondrial hyperpolarization.

Under the conditions of low-amplitude mitochondrial swelling, the oxidative phosphorylation system functions in a local coupling mode postulated by Williams in 1961. The proton pumps activation leads to the formation of non-equilibrium membrane bounded proton fraction (nef-H+), which is sorbed on the outer side of the inner mitochondrial membrane under these conditions. This proton fraction is crucial for the ATP synthesis. The present work is devoted to the development of the methods allowing investigations of the properties of nef-H+. For this purpose, a new membranotropic highly hydrophobic uncoupler 2,4,6-trichloro-3-pentadecylphenol was synthesized. In accordance with our results, it can be referred to as a new type of transmemebrane proton carriers, which interact specifically only with nef-H+ outer side of the inner membrane. A new method of the nef-H+ removal from the outer side of the inner mitochondrial membrane under the conditions when the proton pumps are active has been developed. The method is based on neutralization of nef-H+ by hydroxyl anions transferred by H2PO 4 − /OH−-antiporter to interfacial border. It was demonstrated that the membrane proton fraction is heterogeneous. Two components of the fraction were identified. And at the same time, it was shown that nucleotide translocator participates in the formation of one component. Also, in this series of experiments it was found that a well-known effect of respiration inhibition by high concentrations of uncoupler under certain conditions can be completely explained by the nef-H+ formation. We presume that one of the most important results of the studies carried out is a new independent evidence of the existence of nef-H+, which is formed under the conditions of proton pump activation.

The ultrastructure of cristae of intact mitochondria, in which the activity of the oxidative phosphorylation system was retained, was investigated by small-angle neutron scattering. Osmotic swelling of organelles was found to cause ultrastructural rearrangements of the inner mitochondrial membrane in rat liver and heart mitochondria. This process was accompanied by the appearance of diffraction peaks indicative of the formation of ordered structures. The lamellar packing of the membranes in heart mitochondria, which is observed under normal conditions, was found to undergo a transition to a nonlamellar hexagonal packing, whose scattering curve shows two peaks with the scattering vectors characterized by a spacing ratio of 1:√3. The electron-microscopy results confirm the possibility of the formation of a hexagonal packing of the inner membrane in heart mitochondria.

A brief description of the principal directions for searching and investigating the model of local coupling between respiration and phosphorylation proposed by R. Williams is given in this paper. We found conditions where it was possible to reveal typical functional special features of the mitochondrial phosphorylating system. According to the theory, such special features should be observed experimentally if the mitochondrial phosphorylating system operated in the state of a supercomplex. It was proved that the phosphorylating system is able to operate in two states: P. Mitchell state and R. Williams state. It was demonstrated that in the ATP synthesis reaction, ATP-synthase (F1F0) was able to use thermodynamic potential of Bronsted acids as a source of energy. It was shown using a double-inhibitor titration technique that when the phosphorylating system operated in the supercomplex state, the electron transfer system and ATP-synthesis system were docked rigidly. A model system of chemical synthesis of membrane-bound proton fraction (Bronsted acids), carrying a free energy excess, was developed on the model of bilayer lipid membrane. Catalysts selectively accelerating proton detachment of this fraction were also found. The formation of a Bronsted acids fraction carrying free energy excess was recorded during the operation of proton pumps on mitochondrial and mitoplastic membranes. In the experimental part of the work, a brief description is given of studies on new uncouplers that transfer the phosphorylation system from the local coupling state to the state of transmembrane proton transfer. Thus, they accelerated the respiration of mitochondria and decreased the ADP/O parameter.

Ordered and amorphous protein aggregation causes numerous diseases. Tobacco mosaic virus coat protein for many decades serves as the classical model of ordered protein aggregation ("polymerization"). It was also found to be highly prone to heat-induced amorphous aggregation and the rate of this aggregation could be easily manipulated by changes in solution ionic strength and temperature. Here, we report that rapid amorphous aggregation of this protein can be induced at 25 degrees C in phosphate buffer by low micromolar (start at about 15 microM) concentrations of cationic surfactant cetyltrimethylammonium bromide. At equilibrium four surfactant molecules bound to the protein subunit. As judged by circular dichroism and fluorescence spectroscopy data, the coat protein molecules retained their native structure upon the cetyltrimethylammonium bromide induced aggregation. No aggregation was observed at the higher surfactant concentrations (above 300 microM). Micromolar concentrations of anionic surfactant sodium dodecylsulfate rapidly reversed the cetyltrimethylammonium bromide induced aggregation of the coat protein due to formation of mixed surfactant-surfactant micelles. Cetyltrimethylammonium bromide (100-300 microM) also induced the reversible intact tobacco mosaic virus virion aggregation. The possible liability to the cetyltrimethylammonium bromide induced amorphous aggregation of other ordered aggregate-producing proteins has been discussed.

Crystallization of F1F0-ATP synthase from thermophilic bacterium Chloroflexus aurantiacus was carried out in the present work. A new crystallization method based on slow detergent removal in a two-phase system, consisting of decane and protein solution, was developed. Three-dimensional protein crystals were grown by this method. Two-dimensional molecular lattice with unit cell parameters of a=28 nm, b=23 nm and φ =104° was observed by atomic force microscopy on their surface. Thickness of protein surface layer of the crystals was determined also. Model of the protein molecular packing of the crystals is suggested basing on the experimental data.

Effects of low SDS concentrations on amorphous aggregation of tobacco mosaic virus (TMV) coat protein (CP) at 52 degrees C and on the protein structure were studied. It was found that SDS completely inhibits the TMV CP (11.5 microM) unordered aggregation at the detergent/CP molar ratio of 15 : 1 (0.005% SDS). As judged by fluorescence spectroscopy, these SDS concentrations did not prevent heating-induced disordering of the large-distance part of the TMV CP subunit, including the so-called "hydrophobic girdle". At somewhat higher SDS/protein ratio (40 : 1) the detergent completely disrupted the TMV CP hydrophobic girdle structure even at room temperature. At the same time, these low SDS concentrations (15 : 1, 40 : 1) strongly stabilized the structure of the small-distance part of the TMV CP molecule (the four alpha-helix bundle) against thermal disordering as judged by the far-UV (200-250 nm) CD spectra. Possible mechanisms of TMV CP heating-induced unordered aggregation initiation are discussed.

In the presence of cyanide and various respiratory substrates (succinate or pyruvate + malate) addition of high concentrations of lucigenin (400 microM; Luc2+) to rat liver mitochondria can induce a short-term flash of high amplitude lucigenin-dependent chemiluminescence (LDCL). Under conditions of cytochrome oxidase inhibition by cyanide the lucigenin-induced cyanide-resistant respiration (with succinate as substrate) was not inhibited by uncouplers (FCCP) and oligomycin. Increase in transmembrane potential (Deltaphi) value by stimulating F0F1-ATPase functioning (induced by addition of MgATP to the incubation medium) caused potent stimulation of the rate of cyanide-resistant respiration. At high Deltaphi values (in the presence of MgATP) cyanide resistant respiration of mitochondria in the presence of succinate or malate with pyruvate was insensitive to tenoyltrifluoroacetone (TTFA) or rotenone, respectively. However, in both cases respiration was effectively inhibited by myxothiazol or antimycin A. Mechanisms responsible for induction of LDCL and cyanide resistant mitochondrial respiration differ. In contrast to cyanide-resistant respiration, generation of LDCL signal, that was suppressed only by combined addition of Complex III inhibitors, antimycin A and myxothiazol, is a strictly potential-dependent process. It is observed only under conditions of high Deltaphi value generated by F0F1-ATPase functioning. The data suggest lucigenin-induced intensive generation of superoxide anion in mitochondria. Based on results of inhibitor analysis of cyanide-resistant respiration and LDCL, a two-stage mechanism of autooxidizable lucigenin cation-radical (Luc*+) formation in the respiratory chain is proposed. The first stage involves two-electron Luc2+ reduction by Complexes I and II. The second stage includes one-electron oxidation of reduced lucigenin (Luc(2e)). Reactions of Luc(2e) oxidation involve coenzyme Q-binding sites of Complex III. This results in formation of autooxidizable Luc*+ and superoxide anion generation. A new scheme for lucigenin-dependent electron pathways is proposed. It includes formation of fully reduced form of lucigenin and two-electron-transferring shunts of the respiratory chain. Lucigenin-induced activation of superoxide anion formation in mitochondria is accompanied by increase in ion permeability of the inner mitochondrial membrane.

Apoptosis in myocardial tissue slices was induced by extended incubation under anoxic conditions. Mitochondria were isolated from the studied tissue. A new method of isolation of mitochondria in special conditions by differential centrifugation at 1700, 10,000, and 17,000g resulted in three fractions of mitochondria. According to the data of electron microscopy the heavy mitochondrial fraction (1700g) consisted of mitochondrial clusters only, the middle mitochondrial fraction (10,000g) consisted of mitochondria with typical for isolated mitochondria ultrastructure, and the light fraction consisted of small mitochondria (2 or 3 cristae) of various preservation. The heavy fraction contained unusual structural elements that we detected earlier in apoptotic myocardial tissue—small electron-dense mitochondria incorporated in bigger mitochondria. The structure of small mitochondria from the light fraction corresponded to that of the small mitochondria from these unusual elements—“mitochondrion in mitochondrion”. The most important functions of isolated mitochondria are strongly inhibited when apoptosis is induced in our model. The detailed study of the activities of the two fractions of the apoptotic mitochondria showed that the system of malate oxidation is completely altered, the activity of cytochrome c as electron carrier is partly inhibited, while succinate oxidase activity is completely preserved (complexes II, III, and IV of the respiration chain). Succinate oxidase activity was accompanied by high permeability of the internal membrane for protons: the addition of uncoupler did not stimulate respiration. ATP synthesis in mitochondria was inhibited. We demonstrated that in our model of apoptosis cytochrome c remains in the intermembrane space, and, consequently, is not involved in the cascade of activation of effector caspases. The possible mechanisms of induction of apoptosis during anoxia are discussed.

In aqueous medium etiolated wheat seedlings release superoxide anion (O2*-). Interaction of a synthetic antioxidant, butylated hydroxytoluene (BHT, ionol), with oxygen in the aqueous medium is accompanied by O2*- formation. This suggests that under certain conditions BHT behaves as a prooxidant. A natural antioxidant, superoxide dismutase (SOD), and also a wound healing preparation, emulsified denatured placenta (EDP), do not exhibit the prooxidant properties. In contrast to BHT, they reduce O2*- production by the etiolated wheat seedling system.

It was found that production of superoxide (O2– ·) is crucial for normal morphogenesis of etiolated wheat seedlings in the early stages of plant development. The development of etiolated wheat seedlings was shown to be accompanied with cyclic changes in the rate of O2– · production both in the entire intact seedling and in its separated organs (leaf, coleoptile). First increase in the rate of O2– · production was clearly observed in the period from two to four days of seedling development, then the rate of O2– · production decreased to the initial level, and then it increased again for two days to a new maximum. An increase in O2– · production in the period of the first four days of seedling development correlates with an increase in DNA and protein contents in the coleoptile. The second peak of increased rate of O2– · production observed on the sixth or seventh day of seedling development coincides with a decrease in DNA and protein contents and apoptotic internucleosomal nuclear DNA fragmentation in the coleoptile. Incubation of seedlings in the presence of the antioxidant BHT (ionol) strongly affects their development but it does not influence the increase in DNA and protein contents for the initial four days of seedling life, and it slows down the subsequent age-dependent decrease in protein content and fully prevents the age-dependent decrease in DNA content in the coleoptile. A decrease in the O2– · amount induced by BHT distorts the seedling development. BHT retards seedling growth, presumably by suppression of cell elongation, and it increases the life span of the coleoptile. It seems that O2– · controls plant growth by cell elongation at the early stages of seedling development but later O2– · controls (induces) apoptotic DNA fragmentation and protein disintegration.

Electrically silent hydrogen ion fluxes across a planar bilayer lipid membrane (BLM) induced by an addition of monocarboxylic acid at one sive of BLM were studied by measuring pH changes in the unstirred layers near the BLM surface. The pH changes were assayed by recording protonophore-vepenvent potentials as well as by direct measurements of pH shifts in the unstirred layers close to the membrane by the pH microelectrove. It was shown that the mechanism of the acid transport changed qualitatively upon the increase of the hydrophobic chain length of the acid. In the case of short-chain acids at pH < pKa, the total transport was limited by diffusion of the anionic form of the acid across the unstirred layers, while at the alkaline pH (pH > > pKa) the transport was limited by diffusion of the neutral form across the membrane. In the alkaline pH range the pH shifts induced by short-chain acids were sensitive to the presence of cholesterol in the BLM as well as to the stirring conditions in the cell. However, in the case of long chain acids (more than 8 carbonic atoms) the transport was limited by diffusion of the anionic form of the acid in the whole range of pH studied. In the latter case, pH changes in the unstirred layers did not vepend on the presence of cholesterol in the membrane, and moreover pH shifts were not vepenvent on the thickness of the unstirred layer. It was proposed that the peculiarities of the long-chain acid-induced proton transport were associated with the formation of micelles of the acid in bathing solutions.

Effects of the coenzyme Q analog (MitoQ10) carrying a positively charged decyltetraphenylphosphonium group on functional activity of phosphorylating liver mitochondria were studied. Using inhibitory analysis it was found that at micromolar concentrations this quinone is reduced by NADH-dependent DT-diaphorase. Under conditions of malate oxidation, MitoQ10 stimulates electron transfer from NADH to oxygen by shunting the block of rotenone-induced electron transport in Complex I. Steady-state mitochondrial respiration induced by rotenone and MitoQ10 (1 μM), as well as K3 shunt are both blocked by the DT-diaphorase inhibitor dicumarol, the Complex III inhibitor myxothiazole, and the cytochrome oxidase inhibitor cyanide. The electron transport chain induced in liver mitochondria by MitoQ10 in the presence of rotenone appears as follows: NADH → DT-diaphorase → MitoQ10 → Complex III → Complex IV → O2. Under conditions of malate (but not succinate) oxidation, MitoQ10 and high concentrations of vitamin K3 induce in mitochondria cyanide-resistant respiration and opening of the nonspecific pore eventually resulting in inhibition of oxidative phosphorylation. It is concluded that MitoQ10 should be regarded as an analog of hydrophilic quinones (vitamin K3, duroquinone, etc.) widely known as substrates for mitochondrial DT-diaphorase not interacting with CoQ10 rather than as a natural CoQ10 analog.

Synthesis of cationic plastoquinone derivatives (SkQs) containing positively charged phosphonium or rhodamine moieties connected to plastoquinone by decane or pentane linkers is described. It is shown that SkQs (i) easily penetrate through planar, mitochondrial, and outer cell membranes, (ii) at low (nanomolar) concentrations, posses strong antioxidant activity in aqueous solution, BLM, lipid micelles, liposomes, isolated mitochondria, and cells, (iii) at higher (micromolar) concentrations, show pronounced prooxidant activity, the "window" between anti- and prooxidant concentrations being very much larger than for MitoQ, a cationic ubiquinone derivative showing very much lower antioxidant activity and higher prooxidant activity, (iv) are reduced by the respiratory chain to SkQH2, the rate of oxidation of SkQH2 being lower than the rate of SkQ reduction, and (v) prevent oxidation of mitochondrial cardiolipin by OH*. In HeLa cells and human fibroblasts, SkQs operate as powerful inhibitors of the ROS-induced apoptosis and necrosis. For the two most active SkQs, namely SkQ1 and SkQR1, C(1/2) values for inhibition of the H2O2-induced apoptosis in fibroblasts appear to be as low as 1x10(-11) and 8x10(-13) M, respectively. SkQR1, a fluorescent representative of the SkQ family, specifically stains a single type of organelles in the living cell, i.e. energized mitochondria. Such specificity is explained by the fact that it is the mitochondrial matrix that is the only negatively-charged compartment inside the cell. Assuming that the Deltapsi values on the outer cell and inner mitochondrial membranes are about 60 and 180 mV, respectively, and taking into account distribution coefficient of SkQ1 between lipid and water (about 13,000 : 1), the SkQ1 concentration in the inner leaflet of the inner mitochondrial membrane should be 1.3x10(8) times higher than in the extracellular space. This explains the very high efficiency of such compounds in experiments on cell cultures. It is concluded that SkQs are rechargeable, mitochondria-targeted antioxidants of very high efficiency and specificity. Therefore, they might be used to effectively prevent ROS-induced oxidation of lipids and proteins in the inner mitochondrial membrane in vivo.

In rat liver mitochondria the amylrhodamine is responsible for uncoupling (respiratory stimulation in state 4) by two distinct processes. Immediately after amylrhodamine addition (2-12 microM) stimulation of respiration takes place. Respiration rate for this phase is constant in time, it is independent of the potassium or inorganic phosphate content in the medium, not inhibited by oligomycin, ruthenium red, cyclosporine A, N-ethyl-maleimide and EGTA. The second phase of the respiratory stimulation is not linear in time. Respiration rate within this phase increases with rising of potassium and phosphate content in the medium. This effect is abolished by oligomycin, ruthenium red, cyclosporine A, N-ethylmaleimide and EGTA. The beginning of respiratory increment coincides with the second phase of Ca2+ release from mitochondria.

Stroke is known to induce massive cell death in the ischemic brain. Either necrotic or apoptotic types of cell death program were observed in neurons in zone of ischemia. We suggest that spatial heterogeneity of glucose and oxygen distribution plays a crucial role in this phenomenon. In order to elucidate the role of glucose and oxygen in ischemic neurons choice of cell death pathway, conditions corresponding to different areas of insult were reproduced in vitro in the model of surviving brain cortex tissue slices. Three zones were modeled in vitro by varying glucose and oxygen concentration in surviving slices incubation media. Modeled ischemic area I (MIA I) was corresponded to the center of suggested ischemic zone where the levels of glucose and oxygen were considered to be extremely low. MIA II was assigned as intermediate area where oxygen concentration was still very low but glucose was present (this area was also divided into two sub-areas MIA IIa and MIA IIb with physiologically low (5 mM) and normal (10 mM) level of glucose respectively). MIA III was considered as a periphery area where glucose concentration was close to physiological level and high level of ROS production had been induced by reoxygenation after anoxia. Analysis of molecular mechanisms of cell death in MIA I, IIa, IIb and III was carried out. Cell death in MIA I was found to proceed by necrotic manner. Apoptosis characterized by cyt c release, caspase 3 activation and internucleosomal DNA fragmentation was observed in MIA III. Cell death in MIA II was accompanied by several (not all) hallmarks of apoptosis. Mechanisms of cell death in MIA IIa and MIA IIb were found to be different. Internucleosomal DNA fragmentation in MIA IIa but not in MIA IIb was sensitive to glycine (5 mM), inhibitor of NMDA receptor MK-801 (10 μM) and PTP inhibitor cyclosporine A (10 μM). Activation of caspase 3 was detected in MIA IIb but not in MIA IIa. However cytochrome c release was observed neither in MIA IIa nor in MIA IIb. In MIAs II–III apoptosis was accompanied by uncoupling of oxidative phosphorylation, which was induced by rise of intracellular Ca2+ and intensive ROS production. Results obtained in present study allow us to propose existence of at least four molecular pathways of cell death development in brain ischemic zone. The choice of cell death pathway is determined by oxygen and glucose concentration in the particular area of the ischemic zone.

A new approach was applied for the measurements of ion transport through bilayer lipid membranes (BLM) induced by electrically neutral cation/H+ exchangers. This is an improved version of the method of the measurements of the cation/H+ exchange rate based on recording pH shifts in the unstirred layers near the BLM. Using this approach, the pH gradient in the unstirred layers induced by the cation/H+ exchanger was reduced by successive addition of the acetate on one side of the BLM until the pH shift reached zero. The difference in acetate concentration across the membrane is a measure of the cation/H+ exchange rate. In the second part of the work we found that the changes in cation concentration in the unstirred layers under the conditions imposed when measuring cation selectivity (according to Antonenko, Yu.N. and Yaguzhinsky, L.S., Biochim. Biophys. Acta 1988; 938, 125–130) can significantly decrease the apparent value of cation selectivity. It was shown that more accurate results can be obtained if low concentrations of the carrier are used. The values of nigericin cation selectivity for the alkali metals were measured (K+/Rb+ 19 ± 1, Rb+/Na+ 1.9 ± 0.2, Na+/Cs+ 8 ± 0.5, Cs+/Li+ 1.8 ± 0.3).

The measurements of pH profiles in the unstirred layers (USLs) near planar bilayer lipid membranes (BLM) were applied for the evaluation of the hydrogen ion fluxes which were induced by nigericin in the presence of potassium ion gradients. It was shown that at high concentrations of KCl the increase in the concentration of citrate buffer caused an anomalous effect, namely, an increase in the local pH shifts in the USLs. The hydrogen ion flux rose 50 times upon the increase in the citrate concentration from 1 mM to 20 mM. Phosphate stimulated the flux 7 times under these conditions. In agreement with our previous results, at low KCl concentrations, when the process is limited by the K+-nigericin interaction, an increase in the buffer concentration led to a reduction of the local pH shifts, under these conditions the usual concentration dependence was observed. The data obtained favor the model implying the existence of the kinetic barrier for proton transfer at the membrane-water interface.
