Differences in how individuals cope with stressful conditions (e.g. novel/unfamiliar environment, social isolation and increases in human contact) can explain the variability in data collection from nutrient digestibility trials. We used the collared peccary (Pecari tajacu), which is under process of domestication and shows high individual behavioral distinctiveness in reactions toward humans, to test the hypothesis that behavioral differences play a role in nutrient digestibility. We assessed the individual behavioral traits of 24 adult male collared peccaries using both the ‘behavioral coding’ and the ‘subjective ratings’ approaches. For the behavioral coding assessment, we recorded the hourly frequency of behaviors potentially indicative of stress during the 30-day habituation period to the experimental housing conditions. The subjective ratings were performed based on the individuals’ reactions to three short-term challenge tests (novel environment, novel object and threat from a capture net) over a period of 56 days. During the last 26 days, the collared peccaries were fed diets either high (n = 12) or low (n = 12) in dietary fiber levels, and we determined the total tract apparent digestibility of nutrients. The individual subjective ratings showed consistency in the correlated measures of ‘relaxedness’, ‘quietness’ and ‘satisfaction’ across the three challenge tests, which were combined to produce z score ratings of one derived variable (‘calmness’). Individual frequency of BPIS/h and calmness scores were negatively correlated and both predicted the total tract digestibility of acid detergent fiber (ADF), which ranged from 0.41 to 0.79. The greater the calmness z scores (i.e. calmer individuals), the greater the total tract digestibility of ADF. In contrast, the higher the frequency of BPIS/h, the lower the total tract digestibility of ADF. Therefore, our results provide evidence that by selecting calmer collared peccaries, there will be an increase in their capacity to digest dietary fiber.

Many small herbivores practice cecotrophy - the ingestion of special feces enriched in microbial protein by colonic separation mechanisms (CSM). In digesta passage experiments, secondary marker excretion peaks in feces are considered indicative of marker-reingestion via cecotrophy, but corroboration by behavioral observation was lacking so far. The paca (Cuniculus paca), a Neotropical hystricomorph rodent, produces two different kinds of feces (hard and soft) and practices cecotrophy either directly (from the anus) or indirectly (from a pile of defecated feces, mostly when hard and soft feces are defecated together). To investigate effects of diet on cecotrophy, we monitored cecotrophy behavior and digesta passage marker excretion of solute and particle markers in four adult pacas, at constant food intake, on four diets varying in protein and fiber content. Marker excretion patterns suggested a 'mucus-trap' CSM typical for hystricomorph rodents, and showed secondary peaks of a similar time lag after cecotrophy as the primary marker peak after marker feeding. However, not every cecotrophy event was followed by a secondary marker peak. On higher fiber/lower protein diets, the number of cecotrophy bouts, the duration of cecotrophy, the number of secondary marker peaks and the difference between solute and particle marker retention increased, whereas the proportion of indirect cecotrophy decreased, indicating a higher degree of digesta phase separation on these diets. Compared to hard feces, soft feces were particularly enriched in solute marker concentration. Cecotrophy depends on a CSM that varies in its efficiency with the nutrient composition of the diet.

The colonic separation mechanism in lagomorphs and some rodents, associated with caecotrophy, serves to retain microbial protein. As tropical fruits have low protein concentrations, caecotrophy could be an important microbial protein source in frugivorous rodents such as the paca (Cuniculus paca). Due to conflicting reports on the occurrence of caecotrophy in this species, we obtained digestive tracts of three adult animals and observed the behaviour of four specimens maintained on a diet of a pelleted feed (P) and a supplemental mix of fruits and sweet potato (S, a source of structural fibre). In a Latin square design, P and S were both offered either in the morning (M) or in the afternoon (A), or with one item in the morning and the other in the afternoon (SP or PS). The paca's proximal colon is characterized by a distinct furrow typical for the colonic separation mechanism of hystricomorph rodents. Caecotrophy, both "direct" (from the anus) and "indirect" (from a pile of defecated faeces), was a regular component of the paca's behavioural repertoire, and caecotrophs contained more nitrogen and less fibre than hard faeces. Higher food intake led to less overall caecotrophy. With afternoon feeding of S, the onset of caecotrophy was delayed and the proportion of indirect caecotrophy increased, with hard faeces and caecotrophs often defecated together. No indirect caecotrophy occurred on treatment M. The results suggest that the time available after ingestion of structural fibre determines the efficiency of the colonic mechanism for the separation of hard faeces and caecotrophs.

Triatomines are vectors of Trypanosoma cruzi, the etiologic agent of Chagas disease. Although the evolutionary process in triatomine is considered as disruptive, cryptic speciation and homoploid hybridization also are possible modes of speciation. Several analyses suggested Triatoma brasiliensis macromelasoma as a product of hybridization between T. brasiliensis and Triatoma juazeirensis. Thus, we analyzed genetic characteristics (chromosomal analysis, genetic distance for the mitochondrial ND1 gene, and the pattern of bands of internal transcribed spacer [ITS]-1) of these species, with emphasis on the phenomenon of homoploid hybridization. All species showed the same cytogenetic characteristics, low genetic distance for ND1 gene, and the same pattern of ITS-1 bands. We consider that these genetic characteristics, together with the large chromatic polymorphism and the viability of experimental crosses possibly are due to the processes of introgression that these species suffered during the process of homoploid hybridization.

Triatoma vitticeps is a Chagas disease vector that was found infected with Trypanosoma cruzi in homes. As this species is endemic from Brazil (Bahia, Espírito Santo, Minas Gerais, and Rio de Janeiro) and no study comparing the specimens from different Brazilian states was conducted, we analyzed the genetic distance (16S rDNA, Cyt b, and COI mitochondrial genes) and the chromosomal characteristics for T. vitticeps from Minas Gerais, Rio de Janeiro, and Espírito Santo. All specimens showed the same cytogenetic characteristics. On the other hand, the different mitochondrial genes demonstrated high intraspecific variation between the genetic distances of T. vitticeps from different states ranging from 2.3% to 7.2%. Based on this, our results suggest that possibly what is characterized as T. vitticeps is a complex of cryptic species (or subspecies).

The brasiliensis complex is composed of five triatomine species, and different approaches suggest that Triatoma lenti and Triatoma petrochiae may be the new members. Therefore, this study sought to analyze the phylogenetic relationships within this complex by means of the D2 region of the 28S RNA gene, and to analyze the degree of polymorphism and phylogenetic significance of this gene for South American triatomines. Phylogenetic analysis by using sequence fragments of the D2 domain did not allow to perform phylogenetic inferences on species within the brasiliensis complex, because the gene alignment composed of a matrix with 37 specimens exhibited only two variable sites along the 567 base pairs used. Furthermore, if all South American species are included, only four variable sites were detected, reflecting the high degree of gene conservation. Therefore, we do not recommend the use of this gene for phylogenetic reconstruction for this group of Chagas disease vectors.

Penicillium echinulatum 9A02S1 is a filamentous fungus with a cellulolytic system that may be used in the processes involved in the degradation of lignocellulosic materials for ethanol production. In this study, the 9A02S1 strain of P. echinulatum was grown in submerged cultures with various carbon sources to assess the activities of cellulases and β-glucosidases and their enzymatic activities were assessed utilizing polyacrylamide gel electrophoresis. Cellulose was used as the inducer. Carboxymethyl cellulose proved to be an efficient carbon source for the production of endoglucanase and filter paper activity. Cellobiose, glucose and sorbitol stimulated the production of β-glucosidases. The presence of multimeric forms of the enzymes or their post-translational modifications were assessed.

Chagas disease is most frequently transmitted to humans through contact with feces of insects from the Triatominae subfamily. In Brazil, there are 65 species of triatomines distributed throughout the country's 27 states. Among the species in the state of Rio Grande do Sul, Triatoma rubrovaria, Triatoma oliveirai, Triatoma pintodiasi, Triatoma klugi, Triatoma carcavalloi, and Triatoma circummaculata (with the addition Triatoma limai, which is endemic to Argentina) form the T. rubrovaria subcomplex. The last species described and grouped into this subcomplex was T. pintodiasi. Thus, this study characterized the genetic distance between T. pintodiasi and of the other members of the T. rubrovaria subcomplex to evaluate the specific status of T. pintodiasi. The genetic distance observed between T. pintodiasi and the other species of the T. rubrovaria subcomplex was large, a finding which highlights the specific status of the species considered to be cryptic of T. circummaculata.

Triatoma bahiensis SherlockSerafim, 1967, T. lenti SherlockSerafim, 1967, and T. pessoai SherlockSerafim, 1967 were described based on material collected in the Brazilian state of Bahia. These species were later included in the T. brasiliensis complex based on their geographic distribution. Triatoma bahiensis and T. pessoai were subsequently synonymized with T. lenti. However, the phylogenetic position of T. lenti within the T. brasiliensis complex has remained doubtful. This study aims to assess the taxonomic status of T. bahiensis and to infer the phylogenetic relationships between T. lenti, T. bahiensis and the other members of the T. brasiliensis species complex. The identities of the species in concern were confirmed by comparisons with high resolution photos of the respective type materials; lectotypes are designated for T. pessoai and T. bahiensis. Morphological, morphometric, molecular, and cytogenetic approaches as well as experimental crosses were used. The low viability of experimental crosses combined with morphological and morphometric data allow the differentiation of T. bahiensis and T. lenti. Pairwise cyt b sequence divergence between T. lenti and T. bahiensis was 2.5%. Cytogenetic and molecular analyses grouped T. lenti and T. bahiensis as members of the T. brasiliensis complex. These results revalidate the specific status of T. bahiensis.

The Brasiliensis subcomplex is a monophyletic group formed by the species Triatoma brasiliensis brasiliensis, T. b. macromelasoma, T. juazeirensis, T. melanica, and T. sherlocki. However, using cytogenetic data and experimental hybrid crosses, T. lenti and T. petrochiae were also grouped into this subcomplex. This study aims to analyze the properties of hotspot in the D2 domain of the nuclear gene 28S in all species of the Brasiliensis subcomplex as well as T. lenti and T. petrochiae. These species show two transversions at position 385 (G↔C and T↔G). We suggest that this mutation in haplotype 4 may be an initial molecular tool that supports the relationship of these species with the subcomplex. In addition to the transversion at haplotype 4, these species, aside from T. melanica, also possess a transversion at position 385 (G↔T) in haplotype 1. Thus, we describe the hotspot mutations of the D2 domain of the nuclear gene 28S for species in Brasiliensis subcomplex as follows: three transversions are present at position 385 of haplotypes 1 and 4, which are shared by members of the subcomplex as well as T. lenti and T. petrochiae. These transversions may be considered a synapomorphy between these species. However, we emphasize that new phylogenetic studies should be conducted to evaluate whether T. lenti and T. petrochiae are truly members of the Brasiliensis subcomplex.

The triatomines are vectors of the protozoan Trypanosoma cruzi, etiologic agent of Chagas disease. These insects are sexually active after the imaginal molt. Some aspects have been studied in Triatoma brasiliensis during the imaginal molt, such as autogeny in virgin females and the relationship between blood ingestion by fifth instar nymph and the realization of the imaginal molt. Thus, to aid in the understanding of reproductive biology and developmental physiology of these vectors, this article analyzes the spermatogenesis of T. brasiliensis during the imaginal molt. The analysis of the seminiferous tubules from males in the fifth instar during imaginal molt has demonstrated that T. brasiliensis has only a few spermatids and a plentiful quantity of sperm. Thus, we suggest that during imaginal molt the cell division is disrupted aiming to reduce energy costs and the differentiation into sperm is stimulated to ensure the paternity of the adult male.

Cytogenetic analyses of triatomines are considered to be important taxonomic tools. Thus, we analyzed the pattern of constitutive heterochromatin in 7 species of triatomine with fragmentation of the sex chromosome X, focusing on the cytotaxonomy of these triatomines. The species analyzed included Triatoma vitticeps, Triatoma melanocephala, Triatoma tibiamaculata, Triatoma protracta, Meccus pallidipennis, Panstrongylus megistus, and Panstrongylus lignarius. The seminiferous tubules of the adult males were subjected to C-banding. P. megistus and P. lignarius showed differences in chromosome number and disposition of constitutive heterochromatin, as only P. lignarius showed C-blocks in autosomes. C-banding can differentiate these species, since one of the sex chromosome (X) is heterochromatic in T. vitticeps. T. protracta showed C-blocks in both ends of all autosomes, T. tibiamaculata showed terminal C-dots in some autosomal pairs and M. pallidipennis did not show constitutive heterochromatin in autosomes. Thus, we confirmed the heterochromatic pattern of 7 species of insects and emphasized the importance of cytogenetic techniques for C-banding for taxonomy studies of the triatomines, which are important vectors of Chagas disease.

Isso ocorre devido ao aumento do pH sanguíneo, desencadeado pelo aumento da frequência respiratória do animal, levando a uma alcalose respiratória. Para minimizar esse problema é possível manipular a dieta das aves favorecendo o balanço e a relação eletrolítica a fim de evitar os distúrbios acido-básicos e, consequentemente, diminuir as perdas, principalmente aquelas relacionadas à mortalidade e fragilidade de casca do ovo. Sendo assim, com o presente estudo avaliou-se o desempenho, a qualidade de ovos, composição de excretas e viabilidade econômica de poedeiras comerciais criadas em condições de alta temperatura e submetidas a três balanços eletrolíticos (140, 210 e 280 mEq.kg -1 ) e 3 relações eletrolíticas (2:1; 3:1 e 4:1) na dieta, utilizando-se para isso diferentes níveis de sódio (Na), potássio (K) e cloro (Cl) em rações formuladas por programação não linear em software livre. Foram utilizadas 216 poedeiras, distribuídas em um delineamento experimental inteiramente casualizado em arranjo fatorial 3 x 3 (Balanço eletrolítico x Relação eletrolítica) com 4 repetições compostas de 6 aves em cada parcela experimental. O experimento teve duração de 3 ciclos de 28 dias cada. Ao final do último ciclo foi realizado um ensaio metabólico para determinar a umidade das excretas e a retenção de nitrogênio, cálcio e fósforo. Os balanços e relações eletrolíticas apresentaram pouca interferência sobre os parâmetros produtivos e de qualidade dos ovos, demonstrando que as poedeiras são mais resistentes às situações adversas do que os frangos de corte. Entretanto pode-se inferir que em rações com BE de 140 e 210 mEq.kg-1 a relação 4:1 apresenta-se antieconômica, encarece a produção em 4,36%, e além disso essa relação associada ao do BE de 140 mEq.kg-1 ... High values of temperature and relative humidity can cause heat stress in birds. This is due to the increased blood pH, triggered by the increase in the respiratory rate of the animal, leading to respiratory alkalosis. To alleviate this problem it is possible to manipulate the diet of the birds, and promoting electrolyte balance relation in order to prevent the acid - base disturbances and hence reduce losses, especially those related to mortality and fragility of the egg shell. Thus, the present study evaluated the performance, egg quality, composition of excreta and economic feasibility of laying hens reared in conditions of high temperature and subjected to three electrolyte balance (140, 210 and 280 mEq.kg -1 ) 3 and electrolytic relations (2:1, 3:1 and 4:1) in the diet using it for varying levels of sodium (Na), potassium (K) and chlorine (Cl) in diets for nonlinear programming free software. 216 laying hens, distributed in a completely randomized factorial 3 x 3 (electrolyte balance x electrolytic Value) with 4 replicates composed of 6 birds in each experimental plot were used. The experiment lasted for 3 cycles of 28 days each. At the end of the last cycle a metabolic assay was performed to determine the moisture of faeces and retention of nitrogen, calcium and phosphorus. The balance sheets and electrolytic contact showed little influence on production parameters and egg quality, demonstrating that the layers are more resistant to adverse conditions than broilers. However it can be inferred that in diets with and BE 140 210 mEq.kg-1 presents 4:1 wasteful, costly to produce at 4.36%, and besides that associated with the ratio of BE 140 mEq . kg-1 determines a lower calcium retention, whereas the combination of BE-1 280 mEq.kg 2:1 ratio with the best possible utilization of dietary nitrogen

Variation in the size and morphological differences intraspecific of Nesotriatoma flavida led to the description of N. bruneri. However, two years later the same author proposed the synonymization N. bruneri with N. flavida. N. bruneri was revalidated through morphological analysis after 35 years. Thus, given the existing taxonomic questioning between these Cuban triatomines, we analyzed new parameters such as genetic distance from the mitochondrial 16S rDNA deposited in Genbank and cytogenetic characterization, through the constitutive heterochromatin pattern, in order to reassess the specific status of N. bruneri. The analysis of the disposition of constitutive heterochromatin in the genome of these triatomines allowed observing that only the sex chromosome Y is heterochromatic, and the autosomes and the sex chromosomes X are euchromatic. These characteristics are identical to those described for N. flavida. By means of analysis of genetic distance matrix, we found that the genetic distance between N. bruneri and N. flavida was only 0.04%. Thus, by means of extremely low genetic distance and identical cytogenetic characteristics, we suggest that N. bruneri should be back again synonymized with N. flavida. However, we recommend that experimental hybrid crosses and new molecular analysis should be conducted, focusing mainly in the genetic distance based on other genes, on the rate of fertility of eggs and viability of hybrids to confirm the proposed of synonymization.

The genus Macrobrachium (Bate, 1868) belongs to the Palaemonidae family. These species are commonly found in lakes, floodplains and rivers in tropical and subtropical regions of South America. The Macrobrachium genus encompasses nearly 210 species of ecological and economic importance. In this study, three species of Macrobrachium (Macrobrachium jelskii, Macrobrachium amazonicum and Macrobrachium brasiliense) were studied in order to characterize the esterase patterns in the hepatopancreas, which were still unknown. Esterases are enzymes which catalyze the hydrolysis of esters. In the hepatopancreas, these enzymes play important roles in several metabolic processes involved in some functions of this organ, such as detoxification and digestion. Twelve esterase bands (EST1 to EST12) were detected in these species, and a comparison among them showed no qualitative differences in interspecific bands, or between males and females. Inhibitors were used to classify the esterase bands. The results indicated seven acetylesterases, two carboxylesterases, one arylesterase, and one cholinesterase. The EST11 band was not detected in these procedures because of its lower frequency. Statistical analyses showed no variability among the species, in either interspecific or intraspecific assays. These results support the hypothesis of a high evolutionary conservation of esterases in the hepatopancreas of these crustaceans. The data enabled us to assess the genetic structure of these species through the use of esterasic enzymes. It also contributes to our knowledge about the biology of these poorly studied species. Knowledge on the genetic structure of populations and species are essential when defining priorities for their management and conservation.

In triatomines, the constitutive heterochromatin pattern is species-specific and allows, in many cases, for the grouping of species. Thus, we cytogenetically analyzed T. sherlocki and T. lenti using C-banding, and we compared the results with previous ones obtained in other species of the brasiliensis subcomplex. Both species were found to have a male diploid chromosome number of 22 chromosomes (2n = 20A. +. XY) with heterochromatic blocks at one or both chromosomal ends of all autosomal pairs. During early meiotic prophase, they showed a large heteropycnotic chromocenter constituted by the association of both sex chromosomes plus two autosomal pairs and many heterochromatic blocks dispersed inside the nucleus. All of these cytogenetic characteristics are similar to those observed in other species of brasiliensis subcomplex, results which confirm the grouping of T. sherlocki and T. lenti within this subcomplex. However, we emphasize the importance of other approaches, such as molecular analysis, to confirm the placement of T. lenti within the brasiliensis subcomplex. © 2012 Elsevier B.V.
