The apparently high yield of testis tumors (25%) in rats exposed long-term to Ochratoxin A (OTA) is uninterpretable without data on tumor yield in unexposed rats. Conversely, our demonstration that prenatal exposure to OTA induces DNA adducts in the testes of newborn mice and the absence of these adducts in the testes of mice not exposed prenatally to OTA, is evidence for the presumptive carcinogenicity of OTA in the testis. Together with recent data showing that prenatal exposure to OTA depresses expression of DMRT1, a tumor suppressor gene in the testis, our findings suggest that OTA may be a cause of testicular cancer.

The aim of this study was to determine the cytotoxic and genotoxic combined effects of three mycotoxins : citrinin CIT, ochratoxin A OTA and fumonisin FB. The studies were conducted (i) in cells culture and (ii) in vivo on rat or pig fed OTA and/or CIT ; and/or FB. The decrease of viability (measured by MTT test) of human kidney cells (HK2) induced by OTA is considerably enhanced by the simultaneous presence of CIT & FB. Expression of biotransformation enzymes (CYP, COX, LIPOX) in HK2 is differently modulated depending of the combination. Induction of COX2 and LIPOX by CIT increased genotoxicity of OTA measured by DNA adducts formation detected by P32 post-labeling. DNA adducts patterns of rat kidney after a 3-weeks feeding by either CIT alone or OTA alone or both together, are similar to those obtained on cell cultures. The main OTA DNA-adduct, found in human tumours, identified as C8 dG-OTA is increased by simultaneous presence of CIT and OTA. In the same way, in in vivo studies on rat and pig fed simultaneously by OT and FB in feed we observed increased OTA specific DNA adducts including C-C8dG OTA adduct and the both OTHQ related adduct. These specific adducts are found in human urothelial tumours, notably in Balkan regions. This data is particularly interesting as we previously shown that families suffering BEN eat food more frequently contaminated by OTA, CIT and FB. In blood cell and kidney of human and rat we isolated OTA and several OTA derivatives identified by HPLC ms/ms, including quinone OTA (OTHQ), GSH-OTA, GSH-OTHQ, DC-OTHQ. The data indicate clearly that exposure to low concentration of mycotoxin which is considered as safe when they are present together can lead to dramatic effect. Until now, regulation does not take into account co-contamination.

The aim of this paper was to identify OTA-DNA adduct in blood and/or kidney of Serbian, Croatian, Bulgarian individuals exposed to OTA via food. OTA metabolites in blood and urine were analysed. DNA adduct were analysed by classical postlabeling technique and using the new high sensitive method allowing us to identified by LC/ms/ms the C8-2’-deoxyguanosine OTA (1 ; 2). In parallel, kidney DNA adduct and OTA metabolites in fluids biologic of rat fed four weeks with increasing amount of OTA were analysed. In families having forebears suffering BEN, OTA contamination occurrence was higher compared to non BEN families (3, 4). C8dG OTA adduct was detected in rat kidney as well as in human samples. OTB, OP-OA, 4-OH OTA, 10 OH OTA, two OTHQ metabolites (quinone form), DC OTHQ and GSH-OTA derivatives were identified in blood and urine. Female rat and women excreted faster and more efficiently OTA and OTA metabolites, whereas male rat and man accumulated OTA derivatives in blood. Simultaneous presence of other mycotoxins increases OTA-DNA adducts. Thus determination of OTA-DNA adducts and metabolites are relevant biomarkers for BEN.

Les champignons filamenteux se développent de manière ubiquitaire sur de nombreux substrats organiques. Les spores se retrouvent dans les poussières et sont souvent inhalables. De plus en plus de données scientifiques pointent l’inhalation de moisissures comme cause d’allergie respiratoire ou cutanée aussi bien dans l’environnement (maisons insalubres) que dans le milieu du travail, notamment dans les industries de compostage de matière organique ou de fabrication de bioéthanol. La contamination peut aussi avoir lieu via les systèmes d’air conditionné. Les spores par elles-mêmes engendrent des effets toxiques à l’origine d’allergie. De plus, elles produisent des toxines. Les risques de développement de moisissures dans l’air inspiré sont bien connus en milieu hospitalier et plus particulièrement chez les personnes immunodépressives. Néanmoins, la présence des ces éléments fongiques en milieu de travail est responsable de rhinites, toux et problèmes dermatologiques, chez les travailleurs non immunodéprimés. Notre note portera sur la contamination de l’air en milieu hospitalier et dans un laboratoire de mycologie ainsi que sur les risques liés à l’air conditionné. Le dernier article relatera les effets subis par les travailleurs lors du compostage et de la fabrication de bioéthanol.

Les cancérogènes environnementaux sont convertis, par des enzymes de métabolisation dits de phase I ou de fonctionnalisation) et des enzymes de phase II ou de conjugaison), en métabolites réactifs pouvant se fixer à l’ADN. La plupart de ces enzymes sont polymorphes : le polymorphisme1 génétique est la base de la susceptibilité individuelle. Il est défini comme une différence par rapport au gène de référence, observée chez au moins 1 % des individus. Pour des expositions équivalentes, deux individus de même poids, même âge, même sexe peuvent avoir des taux plasmatiques2 de toxiques ou de métabolites variant de 1 à 1000 s’expliquant par des capacités d’absorption, de métabolisation et d’élimination différentes régulées génétiquement. La réponse toxique d’un individu vis-à-vis d’un contaminant va dépendre de ses capacités à transporter la substance, à la métaboliser et à réparer les lésions initiales sur l’ADN. Les enzymes de réparation sont aussi polymorphes. L’identification de certains variants alléliques3 de gène comme marqueurs de susceptibilité4 peut constituer un outil précoce de diagnostic et de pronostic.

The rejection of cyclophosphamide (CP) by nanofiltration (NF) and reverse osmosis (RO) membranes from ultrapure (Milli-Q) water and membrane bioreactor (MBR) effluent was investigated. Experimental results showed that the RO membrane provided excellent rejection (>90%) under all operating conditions. Conversely, efficiency of CP rejection by NF membrane was poor: in the range of 20-40% from Milli-Q water and around 60% from MBR effluent. Trans-membrane pressure, initial CP concentration and ionic strength of the feed solution had almost no effect on CP retention by NF. On the other hand, the water matrix proved to have a great influence: CP rejection rate by NF was clearly enhanced when MBR effluent was used as the background solution. Membrane fouling and interactions between the CP molecule and water matrix appeared to contribute to the higher rejection of CP.

The growing use of antineoplastic drugs in cancer therapy is an emerging issue in environmental research. The presence of the anticancer drug cyclophosphamide (CP) in municipal wastewater raises several environmental problems. Besides its cytotoxic effects, CP possesses teratogenic and mutagenic properties and is a known human carcinogen. The application of membrane bioreactor (MBR) technology was investigated with the aim of evaluating its potential for cytostatic drug bioremoval. The toxicity removal was assessed from biomarkers tests and related to the choice of the reactor operating conditions. The influence of sludge retention times (SRT) on CP removal was suited but not significant effects were found for variation of SRT from 50 days to 70 days. CP removal up to 80% was achieved under studied operating conditions. In front of such pollution, evidence has been made about the use of MBR. Our study proofed that advances wastewater treatment using a MBR provides a suitable process for lowering CP concentrations before discharge into the aqueous environment. However, a tertiary treatment is necessary for the complete elimination of toxicity.

La présence de médicaments cytostatique a été évaluée en sortie d’un service d’hématologie, par l’analyse des effets cyto et genotoxiques des effluents. Une quantification chimique est en cours. Cette caractérisation a servi pour mettre au point un effluent modèle de ce type de pollution. Grace au fonctionnement en parallèle de deux bioréacteurs à membrane, l’un traitant une eau domestique classique, l ‘autre traitant l’eau modèle contenant les molécules cytostatiques, la pertinence de la technologie bioréacteur à membrane a été démontrée dans le cas d’une traitement d’une telle pollution : la présence de telles molécules aux effets biologiques semble en effet entrainer une réaction de protection de la part des microorganismes épurateurs, induisant une diminution de leur capacités de séparation d’avec l’eau traitée. La présence des membranes est alors toute indiquée. Cette étude permet de démonter la faisabilité de bioréacteur à membrane dans le cas d’un pollution qui entraine des perturbations dans la biodégradation.

International audience; In the present study, the effect of temperature and water activity on fungal growth and ochratoxin production on coffee-based medium was assessed. Optimal growth of three Aspergillus strains was observed in the same ecological conditions, namely 30 degrees C and 0.99 water activity. Maximal daily growth is 11.2, 6.92, and 7.22 mm/day for Aspergillus niger, Aspergillus carbonarius, and Aspergillus ochraceus, respectively. However, ecological conditions for optimal ochratoxin production vary according to the toxinogenic strain, with water activity as a limiting factor. Such an ochratoxin A production is inhibited at 42 degrees C and 0.75 water activity. Correspondence between laboratory tested water activity and that measured on a sun-dried ripe cherry batch shows that the first 5 days of drying are critical for fungal growth and ochratoxin A production. Accordingly, artificial drying of cherries at temperatures above 42 degrees C will impede not only fungal growth but also contamination with ochratoxin A.

Faisabilité d’un bioréacteur à membrane pour la dégradation de la toxicité d’un médicament anticancéreux.

Le concept de susceptibilité génétique au cancer n’est pas nouveau. Denombreux exemples ont montré que la transmission héréditaire de certainesmutations était associée à un très haut risque de cancers tels que le rétinoblastome,le syndrome de Li-Frauméni, la polypose adénomateuse ou certainscancers du sein. Cependant, ces mutations sont très rares dans la populationgénérale et leur contribution à l’incidence des cancers est faible. Par ailleurs,on sait qu’un grand nombre de cancers sont dus à des facteurs environnementaux.De nombreuses études épidémiologiques ont montré le rôle des expositionsprofessionnelles (hydrocarbures aromatiques polycycliques, amiante,amines aromatiques, benzène, chlorure de vinyle{) dans le développement decancers. La plupart de ces substances n’ont pas d’effet cancérogène direct, c’estau cours des étapes de leur métabolisme qu’apparaissent des métabolitesréactifs susceptibles de léser l’ADN. Certains gènes codant les enzymes impliquéesdans le métabolisme des xénobiotiques (EMX) sont polymorphes. Cespolymorphismes peuvent être associés à des activités enzymatiques variables.Des études épidémiologiques ont recherché l’association entre ces polymorphismeset certains cancers.Les interactions entre facteurs génétiques et environnementaux constituentun axe de recherche en plein essor. Dans ce domaine, la majorité des étudesépidémiologiques sur le cancer ont porté sur les polymorphismes des gènescodant des EMX. Il est possible que d’autres gènes soient impliqués dans lasusceptibilité au cancer (par exemple ceux qui interviennent dans la réparationde l’ADN, la transduction du signal et la régulation du cycle cellulaire).Cependant, les variants de ces gènes ont fait l’objet de très peu d’étudesjusqu’à ce jour.Certaines pathologies allergiques comme l’asthme ont une composante génétiquequi a été suggérée par les études familiales et les études de jumeaux. Desrecherches sont actuellement en cours pour mettre en évidence les gènes,probablement nombreux, qui sont concernés. Jusqu’à présent, les études génétiquesont principalement concerné les gènes impliqués dans la réponse immunitaireet les processus inflammatoires, alors que les gènes des EMX n’ontfait l’objet que de peu d’études. L’asthme et ses phénotypes intermédiairesassociés, hyperréactivité bronchique et atopie, résultent vraisemblablementdes interactions de multiples facteurs génétiques et environnementaux.
