Poly(N-isopropylmethacrylamide) (PNIPMAM) is a thermoresponsive polymer, exhibiting lower critical solution temperature (LCST) behavior in an aqueous solution. We investigate the temperature-dependent phase behavior of PNIPMAM solutions in D2O using turbidimetry, differential scanning calorimetry (DSC), small-angle and very small angle neutron scattering (SANS and VSANS), and Raman spectroscopy, covering a large concentration range, and compare the results from PNIPMAM with the findings from its analogue poly(N-isopropylacrylamide) (PNIPAM). We find that the PNIPMAM chains only dehydrate 2-3 °C above the macroscopic cloud point temperature, TCP. Even in the one-phase state, loosely packed, large-scale inhomogeneities and physical cross-links are observed, and the chain conformation of PNIPMAM is more compact than the one of PNIPAM. This is attributed to the attractive intermolecular interactions between the hydrophobic moieties. The phase transition of PNIPMAM is broader than the one of PNIPAM. Upon heating to the two-phase state, the PNIPMAM chains collapse and form mesoglobules. These are larger and more hydrated than those for PNIPAM. This is attributed to the steric hindrance caused by the additional methyl groups, which weaken the intrapolymer interactions in the two-phase state. Thus, the methyl groups in the backbone of the PNIPMAM chains have a significant impact on the hydration and the structural behavior around the phase transition.

The thermoresponsive behavior of a poly(2-oxazoline)-based molecular brush is investigated in aqueous solution. The molecular brush under study, PiPOx100-g-PEtOx17, has a poly(2-isopropenyl-2-oxazoline) (PiPOx) backbone grafted with thermoresponsive poly(2-ethyl-2-oxazoline) (PEtOx) side chains. Since the backbone degree of polymerization is only a factor of ~ 6 higher than the ones of the side chains, it features an architecture between a star-like polymer and a comb-like polymer. Its aqueous solution exhibits lower critical solution temperature (LCST) behavior with a cloud point temperature Tcp = 40.5 °C at 30 g L−1. The temperature-dependent structural evolution is disclosed using dynamic light scattering (DLS) and small-angle neutron scattering (SANS). An increase of the molecular brush size is found upon heating from room temperature to Tcp, which is attributed to the extension of the backbone resulting from the dehydration and collapse of the side chains. Above Tcp, the size decreases again, which indicates the collapse of the whole molecular brush. Large aggregates are found to be present in the solution in the temperature range 25–50 °C. These become more compact, as the temperature is increased across Tcp.

Recent pre-clinical studies have demonstrated the potential of combining chemotherapy and sonodynamic therapy for the treatment of pancreatic cancer. Oxygen-loaded magnetic microbubbles have been explored as a targeted delivery vehicle for this application. Despite preliminary positive results, a previous study identified a significant practical challenge regarding the co-alignment of the magnetic and ultrasound fields. The introduction of “water‐in‐salt” electrolyte (WiSE) concept opens a new horizon to aqueous electrochemistry that is benefited from the formation of a solid‐electrolyte interphase (SEI). However, such SEI still faces multiple challenges, including dissolution, mechanical damaging, and incessant reforming, which result in poor cycling stability. Here, we report a polymeric additive, polyacrylamide (PAM) that effectively stabilizes the interphase in WiSE. With the addition of 5 molar % PAM to 21 mol kg−1 LiTFSI electrolyte, a LiMn2O4∥L‐TiO2 full cell exhibits enhanced cycling stability with 86 % capacity retention after 100 cycles at 1 C. The formation mechanism and evolution of PAM‐assisted SEI was investigated using operando small angle neutron scattering and density functional theory (DFT) calculations, which reveal that PAM minimizes the presence of free water molecules at the anode/electrolyte interface, accelerates the TFSI− anion decomposition, and densifies the SEI., Polyacrylamide (PAM) was introduced to “water‐in‐salt” electrolyte (21 mol kg−1 LiTFSI) to realize the formation of an effective solid‐electrolyte interphase (SEI) in LiMn2O4∥L‐TiO2 full cells by lowering the cathodic limit in the electrochemical stability window (ESW) and suppressing the hydrogen evolution.

Magnetic microflow cytometers integrate magnetic field sensors and microfluidic channels to detect cell suspensions labeled with magnetic beads. As the magnetic beads are superparamagnetic, commercial permanent magnets (PMs) are typiacally positioned bellow the microchip for an out-of-plane magnetization. Though, they require careful alignment as their magnetic field can impact the sensors sensitivity. Magnetic nanoparticles (MNPs) are key elements in several biomedical applications, e.g., in cancer therapy. Here, the MNPs are remotely manipulated by magnetic fields from outside the body to deliver drugs or generate heat in tumor tissue. The efficiency and success of these approaches strongly depend on the spatial distribution and quantity of MNPs inside a body and interactions of the particles with the biological matrix. These include dynamic processes of the MNPs in the organism such as binding kinetics, cellular uptake, passage through cell barriers, heat induction and flow. While magnetic measurement methods have been applied so far to resolve the location and quantity of MNPs for therapy monitoring, these methods can be advanced to additionally access these particle–matrix interactions. By this, the MNPs can further be utilized as probes for the physical properties of their molecular environment. In this review, we first investigate the impact of nanoparticle–matrix interactions on magnetic measurements in selected experiments. With these results, we then advanced the imaging modalities magnetorelaxometry imaging and magnetic microsphere tracking to spatially resolve particle–matrix interactions.

Advanced materials 33(24), 2008683 (2021). doi:10.1002/adma.202008683, Self‐assembly of iron oxide nanoparticles (IONPs) into 1D chains is appealing, because of their biocompatibility and higher mobility compared to 2D/3D assemblies while traversing the circulatory passages and blood vessels for in vivo biomedical applications. In this work, parameters such as size, concentration, composition, and magnetic field, responsible for chain formation of IONPs in a dispersion as opposed to spatially confining substrates, are examined. In particular, the monodisperse 27 nm IONPs synthesized by an extended LaMer mechanism are shown to form chains at 4 mT, which are lengthened with applied field reaching 270 nm at 2.2 T. The chain lengths are completely reversible in field. Using a combination of scattering methods and reverse Monte Carlo simulations the formation of chains is directly visualized. The visualization of real‐space IONPs assemblies formed in dispersions presents a novel tool for biomedical researchers. This allows for rapid exploration of the behavior of IONPs in solution in a broad parameter space and unambiguous extraction of ​the parameters of the equilibrium structures. The thermoresponsive behavior of two molecular brushes having poly(propylene oxide)–poly(ethylene oxide) copolymer side chains is investigated in aqueous solution, where the side chains have longer contour lengths than the backbone. The brushes differ in the side-chain architecture; they are either a diblock (PbE) or a random copolymer (PrE). For both types of brushes, an overall rodlike shape at room temperature is revealed by cryo-electron microscopy. A polymer-rich central part and a water-rich outer part are identified by small-angle neutron scattering (SANS). Upon heating up to the cloud point, the shell of PbE weakly dehydrates, while the one of PrE severely dehydrates along with a cylinder-to-disk transformation. Moreover, the aggregates formed above the cloud points feature significantly different inner structures: PbE aggregates are composed of strongly interpenetrating brushes, while PrE aggregates consist of loosely packed brushes. These differences indicate the important role of the side-chain architecture in the thermal dehydration behavior and the associated structural changes.

Polyunsaturated omega-3 fatty acid docosahexaenoic acid (DHA) is found in very high concentrations in a few peculiar tissues, suggesting that it must have a specialized role. DHA was proposed to affect the function of the cell membrane and related proteins through an indirect mechanism of action, based on the DHA-phospholipid effects on the lipid bilayer structure. In this respect, most studies have focused on its influence on lipid-rafts, somehow neglecting the analysis of effects on liquid disordered phases that constitute most of the cell membranes, by reporting in these cases only a general fluidifying effect. In this study, by combining neutron reflectivity, cryo-transmission electron microscopy, small angle neutron scattering, dynamic light scattering and electron paramagnetic resonance spectroscopy, we characterize liquid disordered bilayers formed by the naturally abundant 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine and different contents of a di-DHA glycero-phosphocholine, 22:6-22:6PC, from both a molecular/microscopic and supramolecular/mesoscopic viewpoint. We show that, below a threshold concentration of about 40% molar percent, incorporation of 22:6-22:6PC in the membrane increases the lipid dynamics slightly but sufficiently to promote the membrane deformation and increase of multilamellarity. Notably, beyond this threshold, 22:6-22:6PC disfavours the formation of lamellar phases, leading to a phase separation consisting mostly of small spherical particles that coexist with a minority portion of a lipid blob with water-filled cavities. Concurrently, from a molecular viewpoint, the polyunsaturated acyl chains tend to fold and expose the termini to the aqueous medium. Smart, fully orthogonal switching was realized in a highly biocompatible diblock copolymer system with variable trigger-induced aqueous self-assembly. The polymers are composed of nonionic and zwitterionic blocks featuring lower and upper critical solution temperatures (LCSTs and UCSTs). In the system investigated, diblock copolymers from poly( N-isopropyl methacrylamide) (PNIPMAM) and a poly(sulfobetaine methacrylamide), systematic variation of the molar mass of the latter block allowed for shifting the UCST of the latter above the LCST of the PNIPMAM block in a salt-free condition. Thus, successive thermal switching results in "schizophrenic" micellization, in which the roles of the hydrophobic core block and the hydrophilic shell block are interchanged depending on the temperature. Furthermore, by virtue of the strong electrolyte-sensitivity of the zwitterionic polysulfobetaine block, we succeeded to shift its UCST below the LCST of the PNIPMAM block by adding small amounts of an electrolyte, thus inverting the pathway of switching. This superimposed orthogonal switching by electrolyte addition enabled us to control the switching scenarios between the two types of micelles (i) via an insoluble state, if the LCST-type cloud point is below the UCST-type cloud point, which is the case at low salt concentrations or (ii) via a molecularly dissolved state, if the LCST-type cloud point is above the UCST-type cloud point, which is the case at high salt concentrations. Systematic variation of the block lengths allowed for verifying the anticipated behavior and identifying the molecular architecture needed. The versatile and tunable self-assembly offers manifold opportunities, for example, for smart emulsifiers or for sophisticated carrier systems.

The pore structure of organic rich mudrocks is associated with both inorganic and organic constituents. The contribution of organic matter to the pore structure has been investigated on Posidonia and Bossier Shale samples having different organic carbon content and thermal maturity. Development and distribution of organic matter pores were studied by using small angle neutron scattering technique at a broad pore scale size investigation, from 2 nm to 2 µm. The pore structure of the mudrocks studied is highly complicated at which total pore volume and specific surface area are not significantly affected by thermal maturation, however, the maturity attribute contributes to different pore size distribution on meso- and macro-pores. Thermal maturation is likely to be the factor of amalgamating small organic matter pores into larger pores in overmature organic rich mudrocks, potentially causing an increase in pore volume at macroscale pores. Although not considerably, the increased macroporosity can enhance the permeability of pore network for viscous gas flow in organic rich mudrocks.

Small angle neutron scattering has been combined with supercritical methane sorption resulting in a collection of isothermal scattering curves for Opalinus Clay, Posidonia Shale and Eagleford Shale. Scattering data have been analyzed with respect to sorbed phase behavior for which a two- and three-phase model have been used. The two-phase model clearly indicates the formation of a sorbed phase of which properties like density and volume fraction change with pore size. Application of the three-phase model yields sorbed phase densities higher than the bulk density of methane as predicted by the equation of state provided by NIST. Current work focuses on implementing localized density calculations in order to quantify the effect of pore size on the sorbed phase properties.

The self-assembly of the two anomeric forms of n-hexadecyl-d-maltopyranoside (denoted α-C16G2 and β-C16G2) has been studied in dilute aqueous solution by means of surface tension measurements, scattering methods (dynamic light scattering, static light scattering, and small-angle X-ray and neutron scattering), and cryo-transmission electron microscopy at different surfactant concentrations and temperatures. Surface tension measurements demonstrate differences in the surfactant adsorption at the air–water interface, where α-C16G2 shows a lower CMC than β-C16G2. Similarly, micelle morphology was found to profoundly depend on anomerism. β-C16G2 preferentially forms very elongated micelles with large persistence lengths, whereas α-C16G2 assembles into smaller micelles for which the structure varies with concentration and temperature. The differences between the two surfactant anomers in terms of self-assembly can be attributed to the interaction between neighboring headgroups. Specifically, β-C16G2 allows for a closer packing in the palisade layer, hence reducing the micelle curvature and promoting the formation of more elongated micelles. Strong intermolecular headgroup interactions may also account for the observed rigidity of the micelles.

Low gradient magnetic holding fields are required for maintaining the polarization of polarized 3He, as diffusion though non-ideal gradients can cause total relaxation rates much greater than the intrinsic total 3He relaxation rate of a particular 3He cell in optimal conditions. For neutron scattering applications we often rely on µ-metal cavities to provide a degree of shielding from the many other sources of magnetic fields and gradient experienced on a typical neutron instrument. The JCNS utilizes two concepts for such cavities, one based on inexpensive plastic-bonded magnets to provide magnetic flux, and the other based on field coils wound on the sides of the µ-metal cavity. 2 different sized of permanent magnet cavities and three different geometries of coil-based cavities have been produced. Both types of boxes will be presented with magnetic design as well as mechanical construction details along with the achieved performance of the constructed devices.

Small angle neutron scattering (SANS) and nitrogen low-pressure adsorption (LPS) have been used to characterise the pore structure of two organic lean mudrocks, Opalinus Clay, Mont Terri, Switzerland and Carmel Claystone, Utah. This was done in order to obtain a better understanding of H2 and CO2 transport, reaction and sorption related to radioactive waste disposal and carbon storage, respectively. The pore structure information derived by SANS and LPS are comparable and the results have revealed a vast heterogeneity from 2 nm to 2 µm, which can be related to the high clay contents. Due to the high clay contents, pores smaller than 10 nm constitute a large fraction of total porosity (25-30 %) and most of specific surface area (up to 80 %) in the sample mudrocks. Accordingly, these interplays contribute to a pore network of few-to-several nano-Darcy permeability in which pore size dependent transport mechanisms can vary from high sorptive diffusional fluid flow in small pores to low sorptive slip flow regime at progressively larger pores.

We report the design and characterization of a lateral and vertical hydrodynamic focusing feature for whole cell detection on a miniaturized flow cytometer. The developed system, based on magnetic sensing, incorporates spin valve sensors on the bottom of the microfluidic channels that detect cells labeled with magnetic beads. An adaptable 3D hydrodynamic focusing system was developed that pushes labeled cells towards the bottom of the microchannel, closer to the sensors, allowing increased signal amplitude for cells labeled with magnetic beads and enhanced discrimination of labeled cells. Fluorescence microscopy indicates that the lateral and vertical hydrodynamic focusing effect was adequately implemented, consistent with simulation predictions. The sensitivity of the system to detect labeled cells was improved by at least two-fold. By estimating the coverage of magnetic beads on cells, the signal from labeled cells could be predicted using a mathematical model, which also demonstrated the sensitivity of the signal to the height of the cells relative to the sensor. The system is versatile allowing interchangeable flow rates for cells with different diameters.

Magnetically responsive microbubbles (MagMBs), consisting of an oxygen gas core and a phospholipid coating functionalised with Rose Bengal (RB) and/or 5-fluorouracil (5-FU), were assessed as a delivery vehicle for the targeted treatment of pancreatic cancer using combined antimetabolite and sonodynamic therapy (SDT). MagMBs delivering the combined 5-FU/SDT treatment produced a reduction in cell viability of over 50% when tested against a panel of four pancreatic cancer cell lines in vitro. The aim of this study was to characterize the behaviour of superparamagnetic particles in magnetic drug targeting (MDT) schemes. A 3-dimensional mathematical model was developed, based on the analytical derivation of the trajectory of a magnetized particle suspended inside a fluid channel carrying laminar flow and in the vicinity of an external source of magnetic force. Semi-analytical expressions to quantify the proportion of captured particles, and their relative accumulation (concentration) as a function of distance along the wall of the channel were also derived. These were expressed in terms of a non-dimensional ratio of the relevant physical and physiological parameters corresponding to a given MDT protocol. The ability of the analytical model to assess magnetic targeting schemes was tested against numerical simulations of particle trajectories. The semi-analytical expressions were found to provide good first-order approximations for the performance of MDT systems in which the magnetic force is relatively constant over a large spatial range. The numerical model was then used to test the suitability of a range of different designs of permanent magnet assemblies for MDT. The results indicated that magnetic arrays that emit a strong magnetic force that varies rapidly over a confined spatial range are the most suitable for concentrating magnetic particles in a localized region. By comparison, commonly used magnet geometries such as button magnets and linear Halbach arrays result in distributions of accumulated particles that are less efficient for delivery. The trajectories predicted by the numerical model were verified experimentally by acoustically focusing magnetic microbeads flowing in a glass capillary channel, and optically tracking their path past a high field gradient Halbach array.

Acoustically-responsive microbubbles have been widely researched as agents for both diagnostic and therapeutic applications of ultrasound. Recently, there has also been considerable interest in magnetically functionalised microbubbles as multi-modality imaging agents and carriers for magnetically targeted drug delivery. The latter application in particular requires simultaneous application of magnetic and acoustic fields to a target region. This can present a significant practical challenge, especially in vivo where access is typically limited. In this paper, we present a design for an integrated device capable of generating co-aligned magnetic and acoustic fields in order to accumulate microbubbles at a specific location and then to activate them acoustically. For the purposes of this proof of concept study, the magnetic component of the device was designed to concentrate microbubbles at a distance of 10 mm from the probe’s surface, commensurate with relevant tissue depths in preclinical small animal models. The ultrasound transducer was designed to maximise the acoustic intensity in the same region in order to induce cavitation of the magnetically captured microbubbles. Previous studies have indicated that both microbubble concentration and duration of cavitation activity are positively correlated with therapeutic effect. The ability of the device to trap and activate microbubbles was therefore assessed by a series of in vitro tests in a tissue mimicking phantom containing a single vessel of 1.2 mm diameter. At a flow rate of 4.2 mm/s magnetic trapping produced an increase in intensity under B-mode ultrasound imaging consistent with the predicted accumulation profile. When the microbubbles were exposed to the ultrasound field from the probe, the resulting cavitation activity was sustained for a period more than 4 times longer than that achieved with an identical acoustic field but in the absence of a magnet. The feasibility of developing a larger scale device for human applications is discussed.

A key challenge in the development of magnetic drug targeting (MDT) as a clinically relevant technique is designing systems that can apply sufficient magnetic force to actuate magnetic drug carriers at useful tissue depths. In this study an optimisation routine was developed to generate designs of Halbach arrays consisting of multiple layers of high grade, cubic, permanent magnet elements, configured to deliver the maximum pull or push force at a position of interest between 5 and 50 mm from the array, resulting in arrays capable of delivering useful magnetic forces to depths past 20 mm. The optimisation routine utilises a numerical model of the magnetic field and force generated by an arbitrary configuration of magnetic elements. Simulated field and force profiles of optimised arrays were evaluated, also taking into account the forces required for assembling the array in practice. The resultant selection for the array, consisting of two layers, was then constructed and characterised to verify the simulations. Finally the array was utilised in a set of in vitro experiments to demonstrate its capacity to separate and retain microbubbles loaded with magnetic nanoparticles against a constant flow. The optimised designs are presented as light-weight, inexpensive options for applying high-gradient, external magnetic fields in MDT applications.

Blood-brain barrier (BBB) disruption mediated by ultrasound and microbubbles (US-BBBD) is a promising strategy for non-invasive and targeted delivery of therapeutics to the brain. In US-BBBD, treatment control is achieved by externally monitoring acoustic emissions (AE) and adjusting ultrasound parameters in real-time to avoid AE associated with damage. Recent work suggests that AE may also provide insight regarding the extent of BBB opening and BBB recovery time. The mechanisms underlying BBB opening and recovery, however, are largely not understood. To investigate US-BBBD mechanisms with regard to AE, we developed an in vitro platform for monitoring both BBB integrity and AE during US-BBBD. Temporally resolved BBB integrity monitoring was achieved using a microfluidic BBB-on-a-chip device with integrated trans-endothelial electrical resistance (TEER) measurements. Well-characterized ultrasound exposure and AE monitoring were achieved using a focally aligned high-intensity focused ultrasound transducer and passive cavitation detector. In addition to recording TEER and AE data, our platform is compatible with fluorescence microscopy during ultrasound exposure, providing further insight into US-BBBD mechanisms. The magnetic hysteresis loops of an aged Cu(81.2)Mn(18.8) alloy sample exhibit significant asymmetric reversal at low temperatures, with high sensitivity to the cooling field. Much of the observed behaviour was explained by considering an ensemble of coherent, ferromagnetically aligned clusters interacting with a randomized spin glass component. A modified Stoner-Wohlfarth model was successfully applied to the data using Monte Carlo simulations, in order to gain insight into the dependence of the cluster shape anisotropy and exchange anisotropy on the cooling field. This model suggested that ferromagnetic clusters grow as the cooling field increases.
