This study characterized important environmental factors that contribute to plant invasion in the forested riparian zones surrounding stream restoration sites. We sampled vegetation and environmental variables (light availability, soil physiochemistry, and site age) across invasion gradients at multiple sites in Virginia, USA. Data analysis involved a multimetric statistical approach combining correlation and Canonical Correspondence Analysis (CCA) to arrive at a plausible model for invasion risk by species. We targeted three of the most problematic invaders in these systems: Lespedeza cuneata (sericea lespedeza), Lonicera japonica (Japanese honeysuckle), and Microstegium vimineum (Japanese stiltgrass). Our analysis revealed species-specific environmental drivers of invasion, with certain factors consistently important across all targeted invaders—notably, canopy cover, nitrogen availability, soil texture, and bioavailable phosphorus, as indicated by the importance of certain proxies (e.g., metal cations). The results of this research have been used to develop a suite of best practices that can be implemented at the outset of a stream restoration project to reduce the risk of invasion in the riparian forests surrounding these sites. Purpose: To evaluate the effect of a mouthwash containing Lespedeza cuneata extract (LCE) on halitosis as an alternative to chemical mouthwashes. The effect of this natural mouthwash on halitosis-causing bacteria (HCB) was clinically evaluated. Materials and Methods: A total of 84 subjects among 103 participants were recruited from the M Dental Clinic (Busan, South Korea) in this randomised, double-blind, placebo-controlled study. The participants were divided into two groups: 41 in the saline-gargle group (saline group) and 43 in the LCE-gargle group (LCE group). A week before the experiment, scaling and oral health education were conducted to standardise the subjects' oral condition. For clinical evaluation, halitosis and HCB were evaluated pre-gargle (baseline), immediately after gargling (treatment), and 5 days after gargling (5 days post-treatment). Statistical analysis was performed using SPSS for Windows. Results: The level of subjective improvement was statistically significantly greater in the LCE group than the saline group (p < 0.05). Compared with the saline group, the LCE group showed reduced halitosis after 5 days of application. Furthermore, halitosis statistically significantly decreased over time (p < 0.05). Moreover, the LCE group showed a statistically significant decrease in the number of all six HCBs (p < 0.05). Conclusion: LCE gargle was effective in reducing halitosis both subjectively and objectively, suggesting an antibacterial effect on HCBs in the oral cavity. Thus, LCE, which can be used as a safe mouthwash ingredient, can promote oral health and will be valuable for the oral healthcare product industry. Lespedeza cuneata (L. cuneata) is an invasive legume threatening grassland ecosystems of several states in the U.S. Mapping the occurrence of invasive species is the necessary first step to control their spread. Imaging spectroscopy or hyperspectral remote sensing imagery has shown its effectiveness in detecting invasive plants. However, challenges such as limited access to hyperspectral imagery, coarse temporal resolution, high costs, and lack of standardized analysis approaches have limited their application in monitoring biological invasions. Here, we aimed to take advantage of fine spatial and temporal resolution of PlanetScope multispectral satellite data and develop an effective approach to detect L. cuneata in a naturally assembled grassland ecosystem. To achieve this objective, we first sampled a suite of foliar functional traits in the field and identified key traits that distinguish L. cuneata from native plants. We then used our airborne hyperspectral data to select vegetation indices that are associated with the key traits while focusing on those indices that are applicable to satellite-derived multispectral data. We developed seven machine learning models and a decision fusion approach using layers of vegetation indices to map L. cuneata during the growing season. Using our approach, we were able to map L. cuneata with high level of accuracy using PlanetScope multispectral imagery. Our results suggested that the optimal period for detecting L. cuneata in our site was mid-to-late growing season, with an overall classification accuracy of 80.49 ± 5.69% (mid-August) and 82.02 ± 4.66% (end of September), respectively. Lespedeza cuneata (LC) is a perennial plant used in herbal medicine to treat numerous diseases, including prostatic hyperplasia, diabetes, early atherosclerosis, and hematuria. Reference collections of bioactive compounds of LC are crucial for the determination of their pharmacological properties. However, little is known regarding its anti-oxidative and anti-inflammatory effects in alveolar macrophage (MH-S) cells. This study examined whether LC can inhibit reactive oxygen species and Coal fly ash (CFA) induced inflammation in MH-S cells. The anti-oxidative effects of LC were evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays, anti-inflammatory effects were examined using nitric oxide (NO) assay, and cytotoxicity was analyzed using methyl thiazolyl tetrazolium assay. The expression of inflammatory cytokine genes was assessed through a reverse-transcription polymerase chain reaction. Our results revealed that LC exhibited high radical scavenging activity and a dose-dependent (7.8–1,000 μg/mL) inhibition of oxidation as compared to ascorbic acid and Trolox. It also inhibited CFA-induced NO production in MH-S cells. Moreover, it suppressed the CFA exposure-mediated expression of pro-inflammatory mediators and cytokines, including inducible nitric oxide synthase (iNOS), cyclooxygenase- 2 (COX-2), interleukin (IL)-1β, IL-6, and tumor necrosis factor-α. Objective: Aging male syndrome is a clinical biochemical syndrome characterized by typical aging symptoms and serum testosterone deficiency. Although it is accompanied by various health problems, directly affects life satisfaction, and requires proper management, no clear prevention or treatment other than hormone replacement therapy is currently available for this syndrome. Here, we aimed to determine the efficacy and safety of the Lespedeza cuneata extract in the management of the aging male syndrome. Methods: Males aged 43-70 years who provided consent for participation and had a total Aging Males' Symptom questionnaire score ≥ 37 and testosterone level ≤ 500 ng/dL were enrolled in this study. This study was conducted in a randomized, double- blind manner. Participants were randomly assigned to either the experimental or control groups and orally administered the assigned product twice a day. Efficacy was evaluated by measuring changes in Aging Males' Symptom score, Androgen Deficiency in the Aging Male questionnaire score, International Index of Erectile Function score, International Prostatic Symptom Score, blood test results, and body mass index at 8 weeks. Results: After 8 weeks, the experimental group had significantly improved symptom scores compared to the control group on the Aging Males' Symptom and Androgen Deficiency in the Aging Male questionnaires. However, no significant differences in the International Index of Erectile Function score, International Prostatic Symptom Score score, blood test results, and body mass index were observed between the experimental and control groups. Conclusion: Lespedeza cuneata extract safely alleviates andropause symptoms without any significant side effects, suggesting its potential for the treatment of the aging male syndrome. Scaling was performed 1 week before the experiment to secure the homogeneity of the subjects’ oral conditions. After gargling with 15 ml of each solution for 1 min, participants spit out the solution to remove any residual mouthwash solution. Then, PD-related bacteria were measured via the O’Leary index, plaque index (PI), and gingival index (GI). Invasive species are suspected to be major contributors to biodiversity declines worldwide. Counterintuitively, however, invasive species effects are likely scale dependent and are hypothesized to be positively related to biodiversity at large spatial scales. Past studies investigating the effect of invasion on biodiversity have been mostly conducted at small scales (<100 m2) that cannot represent large dynamic landscapes by design. Therefore, replicated experimental evidence supporting a negative effect of invasive plants on biodiversity is lacking across many landscape types, including large grasslands.We collected data across eight large (333–809 ha) grassland landscapes managed with pyric herbivory—that is the recoupling of fire and grazing—to test how an invasive legume Lespedeza cuneata affected plant and bird communities at spatial grains ranging from 0.1 m2 to >3,000,000 m2.Lespedeza cuneata invasion effects on grassland plant diversity and composition changed with scale, being negative at small spatial grains (0.1 m2) and neutral or positive at large spatial grains (>3,000,000 m2).Lespedeza cuneata abundance did not significantly affect bird diversity at any spatial grain measured.Lespedeza cuneata may negatively affect biodiversity if abundances are greater than those observed in this study. However, previous research suggests that Lespedeza cuneata may not be capable of exceeding 20% canopy cover across large landscapes (>400 ha). Control and eradication strategies can be costly and are fraught with risk. If data do not clearly support a negative Lespedeza cuneata abundance–biodiversity relationship, and if invasion is spatially limited across large landscapes, ongoing control and eradication efforts may be unwarranted and ineffective.Synthesis and applications: Invasive species effects gleaned from small‐scale studies may not reliably predict their effects at larger scales. Although we recognize the importance of small‐scale studies in potentially isolating individual mechanisms, management strategies based solely on results from small‐scale studies of invasion are unlikely to increase or conserve biodiversity across large landscapes. Rather, processes that generate landscape heterogeneity—like pyric herbivory—are probably more important for promoting biodiversity across all scales. In the recent past, phytomolecules are exponentially applied in discovering the antidiabetic drug due to less adverse effects. This work screened the active solvent fraction of Lespedeza cuneata based on the phytochemical, enzyme inhibition, and antioxidant properties. The antioxidant efficacy of the different fractions of the L. cuneata was assessed by 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), ferric reducing power, hydrogen peroxide, and hydroxyl radical scavenging assays. The digestive enzyme (α-amylase and α-glucosidase) inhibitory activity was also evaluated. The phytochemical composition of ethyl acetate fraction of L. cuneata (Lc-EAF) was studied by UHPLC-QTOF–MS/MS. The effect of Lc-EAF treatments on glucose uptake was studied in insulin resistance HepG2 cells (IR-HepG2). Further, the antidiabetic effect of Lc-EAF in streptozotocin (STZ)-induced diabetic mice were demonstrated. Ethyl acetate, hexane, and methanol fractions of the L. cuneata showed notable antioxidant, α-amylase, and α-glucosidase inhibitory properties. Among the fractions, Lc-EAF was found to be the most potent. The Lc-EAF exhibited an IC50 of 205.32 ± 23.47 µg/mL and 105.32 ± 13.93 µg/mL for α-amylase and α-glucosidase inhibition, respectively. In addition, 75 µg/mL of Lc-EAF exposure enhanced glucose uptake (68.23%) in IR-HepG2 cells. In vivo study indicated that treatment of Lc-EAF (100 mg/kg b.wt) maintained the blood glucose level through reduced insulin level while improving the lipid profile, hepatic, and renal markers. These findings suggest that Lc-EAF could be considered a prominent source for antidiabetic, anti-hyperlipidemic, and anti-ROS potentials. [ABSTRACT FROM AUTHOR]

Invasive plant species present a growing threat to biodiversity. Many invasive plants recruit microbial symbionts in their novel range that influence their growth and fitness. These soil legacy effects can linger after the removal of invasive species and impact restoration efforts. In a growth room experiment to investigate the legacy effects of an invasive legume and herbicide, Lespedeza cuneata was grown with either an intraspecific competitor or a native species in field-collected soils with and without history of an L. cuneata invasion and use of glyphosate herbicide. In separate experiments, we investigated seed germination and seed bank composition in these different soil types. Taken together, results suggest that legacy effects of L. cuneata invasion do not significantly contribute to its growth or spread in a floodplain in eastern North Carolina. The absence of soil legacy effects, which have been documented in other systems, may be attributed to frequent flooding observed in the system and the resulting homogenization of soil biota. Study findings also suggest that the application of glyphosate herbicide creates areas where L. cuneata can readily reinvade, as it significantly reduced the number and diversity of seedlings to emerge from the seed bank while increasing the aboveground biomass and nodule formation of L. cuneata. Concerning the restoration of native flora, data suggest that Chasmanthium latifolium may be negatively impacted by a L. cuneata invasion or glyphosate herbicide, while Solidago altissima and Chamaecrista nictitans could be successful in preventing areas from being reinvaded. Abstract Sericea lespedeza (Lespedeza cuneata) is an invasive nonnative legume that commonly occurs on sites managed for northern bobwhite (Colinus virginianus). Sericea reduces bobwhite habitat availability by outcompeting native plants that provide food and cover. Sericea can be controlled with postemergence herbicides such as glyphosate as well as a mixture of triclopyr with fluroxypyr, but these herbicides do not provide preemergence control. Imazapic has been effective at controlling a limited number of planted sericea seeds, but the efficacy of imazapic at various application rates to control sericea seedbank response following treatment with a postemergence herbicide has not been evaluated. We designed a field experiment to evaluate sericea control and changes in the plant community as related to bobwhite habitat with 2 postemergence herbicides followed by preemergence treatment of imazapic at 3 sites in Tennessee and Alabama, USA, 2018–2022. Specifically, we treated sericea with postemergence broadcast applications of glyphosate or triclopyr with fluroxypyr in 2018 and applied spot treatment with the same herbicides in 2019–2021. We applied imazapic at 4 rates following prescribed fire in 2019 and 2022. We measured coverage of sericea, native forbs, annual grasses, perennial grasses, and bobwhite food plants before treatment in 2018 and after all treatments in 2022. Nearly all postemergence treatments reduced sericea coverage, but imazapic did not increase control compared to postemergence herbicides alone. Perennial grasses used for nesting and bobwhite foraging were maintained following treatments. We recommend managers use either glyphosate or triclopyr with fluroxypyr to control sericea postemergence and consider using a low rate of imazapic to reduce annual grasses if coverage is a problem on the site.

A 6-yr experiment was conducted to determine the effects of prescribed-fire season on stocker cattle growth performance and rangeland plant community characteristics in the Kansas Flint Hills. Eighteen pastures were grouped by watershed and each watershed was randomly assigned to 1 of 3 prescribed-fire treatments: spring (11 April ± 5.7 d), summer (25 August ± 6.2 d), or autumn (2 October ± 9.0 d). All burns were applied prior to grazing in years 1, 2, 3, and 5; however, no burns were applied in year 4 because of unfavorable burn conditions. Over 5 consecutive grazing seasons, 1,939 yearling stocker calves (initial BW = 281 ± 58.9 kg) were grazed from May to August at a targeted stocking density of 280 kg live-weight + ha−1. Beginning in June of 2018 (pretreatment), a permanent 100-m transect was established in each pasture and was used to determine plant-species composition using a modified step-point method. Forage biomass accumulation and root carbohydrate concentrations of 4 native tallgrass plant species were also measured. All data were analyzed as a completely randomized design using a mixed model. Average daily gain (ADG) was 0.05 to 0.07 kg greater (P  = 0.02) for calves grazing spring-burned pastures compared with calves grazing summer- or autumn-burned pastures; however, ADG did not differ (P  ≥ 0.55) between calves assigned to the summer or autumn prescribed-fire treatments. Basal cover of all graminoids and all forbs did not differ (P  ≥ 0.30) among prescribed-fire treatments; however, basal cover of C3 grasses tended (P  = 0.06) to be greater while basal cover of C4 grasses tended (P  = 0.08) to be less in autumn-burned pastures compared with spring-burned pastures. Forage biomass accumulation did not differ (P  = 0.58) among treatments. In addition, root starch or root water-soluble carbohydrate concentrations in big bluestem (Andropogon gerardii), little bluestem (Schizachyrium scoparium), Indiangrass (Sorghastrum nutans), or purple prairieclover (Dalea purpurea) did not differ (P  ≥ 0.26) among prescribed-fire treatments. Overall, we interpreted these data to suggest that prescribed-fire timing had small influences on yearling stocker cattle growth performance and rangeland plant composition but did not influence forage biomass accumulation or root carbohydrate concentrations of key native tallgrass plant species in the Kansas Flint Hills. Mature ewes were used in a 4-yr study to evaluate effects of intensive late-season sheep grazing on vigor of sericea lespedeza in native tallgrass prairie. Pastures (N = 8; 31 ± 3.6 ha) infested with sericea lespedeza (initial basal frequency = 1.4%) were assigned randomly to one of two treatments: early-season beef steer grazing (1.1 ha/steer; initial BW = 258 ± 34 kg) from April 15 to July 15 followed by no grazing for the rest of the year (control; STR) or steer grazing from April 15 to July 15 followed by intensive grazing by mature ewes (0.2 ha/ewe; SHP) from August 1 to October 1. Ewes (initial BW = 65 ± 3.1 kg) were assigned randomly to graze four of eight pastures; remaining pastures were not grazed from August 1 to October 1. Vegetation responses to treatment were measured along four permanent 100-m transects in each pasture. Herbivory on sericea lespedeza was monitored weekly in each pasture from July 21 to October 7. Herbivory on sericea lespedeza in SHP and STR after steer grazing and before sheep grazing was not different (P = 0.51). In contrast, sericea lespedeza herbivory following sheep grazing was greater (P < 0.01) in SHP than in STR. Herbivory of individual sericea plants was greater (P < 0.01) in SHP than in STR by the end of week 1 of the sheep-grazing period (10.6% vs. 0.5%); moreover, herbivory on sericea lespedeza steadily increased (P ≤ 0.01) such that 92.1% of sericea lespedeza plants were grazed in SHP compared to 1.4% in STR by week 8 of the sheep-grazing period. Whole-plant DM weight of sericea lespedeza at dormancy was less (P < 0.01) in SHP than in STR. Additionally, annual seed production by sericea lespedeza was less (P < 0.01) in SHP than in STR (114 vs. 864 seeds/plant). Pasture forage biomass was not different (P = 0.76) between SHP and STR after the steer-grazing period. Conversely, STR had more (P < 0.01) residual forage biomass than SHP at the end of the sheep-grazing period. Growth performance of beef steers grazing from April 15 to July 15 annually was not different (P ≥ 0.59) between treatments. Our results were interpreted to suggest that intensive late-season grazing by sheep decreased vigor of sericea lespedeza. Late-season sheep grazing decreased forage biomass by 904 kg DM/ha compared with late-season rest; however, residual biomass was adequate to prevent soil-moisture loss and erosion during the dormant season. Trial plantings of Castanea dentata (American chestnut) are currently underway using reclaimed surface mines as reintroduction sites for hybrids bred for disease resistance to canker causing Cryphonectria parasitica (chestnut blight). Coalfields of the eastern US coincide with the species range, but little is known of its ecological interactions as it had largely been extirpated from the landscape by the 1940s. Non-native species such as Lespedeza cuneata (sericea lespedeza) have been planted extensively in the region for both agriculture and mine reclamation and often dominate disturbed sites to form novel systems in place of native forests. We investigated the influence of herbaceous species on BC2F3 chestnut hybrid establishment and ectomycorrhizal (ECM) root colonization on three surface mines in eastern Tennessee that were restored using methods to avoid soil compaction. Hybrid chestnut growth, vegetation density, and species composition surrounding each seedling was assessed after eight growing seasons. ECM fungi present on root samples was quantified and identified by DNA sequencing of the internal transcribed spacer (ITS) region. Sites differed significantly in herbaceous cover and seedling growth. Larger seedlings were found on the site that was comprised primarily of L. cuneata. Average ECM colonization was similar across sites, and groundcover vegetation diversity or L. cuneata did not influence ECM species richness. There was a difference in ECM community composition among the three sites with some fungi displaying subtle changes in species rank. Results suggest that in the absence of soil compaction, L. cuneata does not impede hybrid chestnut establishment or ECM root colonization in Appalachian afforestation projects. Service‐learning (SL) is a type of participatory action‐oriented, experiential learning that connects course content with a need in the community. This type of learning is particularly well suited to environmentally themed courses as student action can have a positive impact on the environment while developing students' conservation ethics and "soft skills" required for many conservation jobs. SL was incorporated into one of two sections of an existing, junior‐level elective course in plant biology. As a service activity, undergraduates participated in the removal of an invasive species (Lespedeza cuneata) along a local greenway. We asked whether students who participated in a modest amount of SL demonstrated greater knowledge of plant biology, stronger connections to their class community, and increased self‐efficacy for environmental action than students who do not participate in SL. Data on student content knowledge, perceptions of classroom climate, and views of one's ability to positively impact the environment were collected through surveys given at the beginning and end of the semester. Instructor‐generated content questions and two published, validated surveys—the Connected Classroom Climate Inventory and Self‐Efficacy for Environmental Action scale—were used. We found that SL students showed a slightly increased sense of community with their peers, which has been found to have positive effects on learning, but not an increased perception of their own ability to be an agent of social change. Lespedeza cuneata, belonging to Fabaceae, is well-known as Chinese bushclover, and it has been used in traditional folk medicines for the treatment of disorders, such as diabetes, hematuria, and insomnia. As part of continuing research projects to discover interesting natural compounds with biological activities from Korean medicinal plants, the phytochemical investigation of L. cuneata resulted in the isolation of five chemical constituents: α-tocopherol (1), 7a-methoxy-α-tocopherol (2), 13(R)-hydroxy-octadeca-(9Z,11E,15Z)-trien-oic acid (3), α-dimorphecolic acid (4), and lupeol (5). The structural determination of the isolated compounds was elucidated from data gathered through nuclear magnetic resonance (NMR) spectroscopy and liquid chromatography-mass spectrometry (LC/MS). Until now, this study is the first to report these five compounds from the plant L. cuneata. Moreover, these isolated compounds (1-5) were evaluated for their anti-adipogenesis effects and their role in lipid metabolism during adipocyte maturation. As a result, the upregulation of mRNA expression levels of Fabp4 from 3T3-L1 pre-adipocytes treated with compounds 3 and 4 demonstrated that these compounds efficiently induced adipocyte differentiation. Furthermore, compounds 3 and 4 were found to regulate lipid metabolism by the induction of lipolytic and of lipogenic gene expressions. Therefore, experimental data from these findings supported that the compounds 3 and 4 induce the adipogenesis of 3T3-L1 pre-adipocytes and regulate lipid metabolism. [ABSTRACT FROM AUTHOR]

Effects of invasive exotic plants on plant communities are often profound, but interactions with higher trophic levels are less evident. Postulated effects of invasive plants on arthropods include increased abundances of herbivores through refuge and improved microclimate, facilitation of other exotic species, and altered arthropod diversity. We examined the impact on the arthropod community by Lespedeza cuneata, an exotic legume with dense, chemically defended foliage, by comparing arthropod numbers in unmanipulated 1-m2 plots with L. cuneata shoots (= control) to arthropod numbers in plots from which shoots of L. cuneata were removed. Lespedeza cuneata removal produced no overall effect on arthropod abundance, but an invasive herbivore, Popillia japonica (Japanese Beetle), was twice as abundant in plots with L. cuneata. Lespedeza cuneata removal increased arthropod evenness in May then decreased it through the summer. In a second experiment we quantified chewing herbivory by placing individual potted plants from five native prairie species and L. cuneata within these same removal/no removal plots for 4 weeks. Removal of L. cuneata shoots from plots did not significantly affect herbivory of tagged leaves from the native species. However, entire L. cuneata leaves or plants disappeared more frequently regardless of plot treatment. Invasion by this exotic legume is likely due to direct effects on native plants, but indirect effects through facilitation of an exotic herbivore could contribute to its success in some areas. Most non-native domesticated plants provide benefits without escaping cultivation, but others have become prominent invaders. A better understanding of how domestication might alter plant traits that influence the ability of species to overcome barriers to invasion could improve invasion risk predictions. We explored how variation in foliar chemistry among cultivars of a widespread invader in the U.S. (Lespedeza cuneata) might influence invasion risk through differences in herbivore interactions and a potential tradeoff with competitive ability. In a no-choice feeding bioassay a generalist herbivore performed better when fed cultivars compared to wild genotypes and native congeners, suggesting domesticated L. cuneata may be more vulnerable to herbivory compared to wild genotypes. However, in a factorial greenhouse experiment, with treatments of herbivory and competition, all cultivars had similar seed and biomass production as the wild genotype. Competition with native species reduced productivity of all L. cuneata types, while experimental herbivory alone did not. We also found far less variation in foliar chemistry among L. cuneata types than expected based on domestication history. While our findings from the bioassay suggest potentially lower invasion risk for cultivars if herbivore populations expand more rapidly when feeding on cultivars, we did not find evidence that cultivars had different responses to herbivory or competition with native species in the greenhouse experiment. Altogether, the findings from our multi-pronged approach for evaluating potential factors underlying invasion success of a domesticated species indicate that domestication has not altered invasion risk of this widespread plant invader. Purpose: To analyze the effects of an aqueous extract of Sericea lespedeza (SL) on rat male menopause. Methods: Levels of nitric oxide (NO), endothelial nitric oxide synthase (NOS), cGMP, and prostaglandin E2 (PGE2) in the penile corpus cavernosum of the rats were evaluated using appropriate kits. Serum levels of dihydrotestosterone (DHT), testosterone, sex hormone-binding globulin (SHBG), and 17-beta hydroxysteroid dehydrogenases (17β-HSD) were measured with enzyme-linked immunosorbent assay kits. Total and motile sperms were counted on a hemocytometer. Histological changes in rat testis and epididymis were analyzed with hematoxylin and eosin staining. Results: The levels of NO, NOS, and cGMP (but not PGE2) increased in a dose-dependent manner (p < 0.05) upon administration of an aqueous extract of SL (AESL), while levels of DHT, 17β-HSD, and testosterone increased in the group administered with 300 mg/kg of AESL. Epididymal sperm count increased by 24 % in such rats compared to controls (p < 0.05). Conclusion: The aqueous extract of SL improves sperm count and muscle mass in rats by increasing the levels of NO, NOS, cGMP and testosterone. We examined effects of the invasive species Lespedeza cuneata on native plants and foliar arthropod communities in a tallgrass prairie. Through observational and manipulative experiments, we examined plant and arthropod responses to L. cuneata over one growing season. The observational study found little impact of the invasive plant on arthropods. By the end of the manipulative experiment, the combined biomass of native grasses and forbs was reduced by approximately 50% in Lespedeza-present plots, while total arthropod numbers were only about 15% lower. Seasonal differences were evident; L. cuneata-absent plots showed lower arthropod numbers in May, but higher abundances in June. Some feeding groups, notably carnivorous arthropods, appeared unaffected by L. cuneata. We tested three hypotheses (one bottom-up and two top-down processes) to explain the relatively weak response of the arthropod community to the invasive plant. Nitrogen content of native plants adjacent to L. cuneata areas was significantly higher compared to plants more distant. Ground arthropod predators were higher during mid-summer in L. cuneata areas, which may partially explain seasonal variation in foliar arthropods. Insectivorous birds were unaffected by L. cuneata abundance, suggesting that arthropod predation rates by birds are unchanged. We suggest that while L. cuneata has strong effects on native plants, its ability to increase neighboring plant quality compensates for the lower biomass of native plants in L. cuneata areas, moderating the arthropod response. Invasive species are a common problem in restoration projects. Manipulating soil fertility and species arrival order has the potential to lower their abundance and achieve higher abundances of seeded native species. In a 7‐year experiment in Missouri, United States, we tested how nutrient addition and the timing of arrival of the invasive legume Lespedeza cuneata and seeded native prairie grass and forb species influenced overall community composition. Treatments that involved early arrival of seeded forb and grass species and late arrival of L. cuneata were most successful at creating community structure that fulfilled our restoration goals, displaying high abundance of seeded native forb species, low abundances of L. cuneata, and non‐native species. There were few treatment interactions, with the exception that timing seeded native forbs and timing of L. cuneata arrival interactively influenced the abundance of seeded native forbs. This suggests that the individual treatments are supporting the restoration goals, such as creating a community with low abundance of L. cuneate or high abundance of native seeded species, without restricting each other. This study demonstrates the importance of priority effects in disturbed habitats prone to invasion, the lasting effects of initial seeding on long‐term community composition, and the potential for fertilization to positively benefit restoration of degraded grasslands. Mature Boer goat wethers were supplemented with 0.5% BW rolled corn and consumed pelleted alfalfa (CON), pelleted Sericea lespedeza (HSL; 6.4% condensed tannins), a 1:1 mixture of alfalfa and lespedeza (MSL), or alfalfa with monensin (ION; 22 mg/kg), coconut oil (CCO; 4%), or soybean oil (SBO; 4%). Total DM intake in the 20-wk study (3.86%, 3.75%, 3.52%, 3.69%, and 3.64% BW) and total tract OM digestibility determined every 5 wk (72.8%, 69.5%, 70.3%, 72.0%, and 71.1%) were not affected by treatment, although there were differences in nitrogen digestion (77.5%, 70.7%, 67.0%, 77.0%, 75.7%, and 73.6% for CON, MSL, HSL, ION, CCO, and SBO, respectively; SEM = 1.76). Ruminal methane emission was not influenced by period and was lowest among treatments for CON expressed as percentages of gross (10.3%, 6.8%, 6.3%, 7.2%, 6.5%, and 6.5%; SEM = 0.35) and digestible energy (14.8%, 10.2%, 9.3%, 10.6%, 9.8%, and 10.1% for CON, MSL, HSL, ION, CCO, and SBO, respectively; SEM = 0.62). In conclusion, both levels of lespedeza elicited similar depressions in ruminal methane emission, with a magnitude of change similar to that of an ionophore and coconut and soybean oils, and effects did not vary with week of the study. Two new phenylpropanoid glycosides named cuneataside E (1) and cuneataside F (2), were isolated from the aerial parts of Lespedeza cuneata (Dum. Cours.) G. Don, whose structures were E and Z isomer, respectively. Their structures were elucidated on the basis of comprehensive spectroscopic analysis (UV, IR, HR-ESI-MS, 1D and 2D NMR). In in vitro bioassays at 10 μmol/L, compound 1 showed moderate hepatoprotective activity against N-acetyl-p-aminophenol (APAP)-induced toxicity in HeG2 cells.

The global incidence of breast cancer has increased. However, there are many impediments to the development of safe and effective anticancer drugs. The aim of the present study was to evaluate the effect of aviculin isolated from Lespedeza cuneata (Dum. Cours.) G. Don. (Fabaceae) on MCF-7 human breast cancer cells and determine the underlying mechanism. Using the bioassay-guided isolation by water soluble tetrazolium salt (WST-1)-based Ez-Cytox assay, nine compounds (four lignan glycosides (1–4), three flavonoid glycosides (5–7), and two phenolic compounds (8 and 9)) were isolated from the ethyl acetate (EA) fraction of the L. cuneata methanolic extract. Of these, aviculin (2), a lignan glycoside, was the only compound that reduced metabolic activity on MCF-7 cells below 50% (IC50: 75.47 ± 2.23 μM). The underlying mechanism was analyzed using the annexin V Alexa Fluor 488 binding assay and Western blotting. Aviculin (2) was found to induce apoptotic cell death through the intrinsic apoptosis pathway, as indicated by the increased expression of initiator caspase-9, executioner caspase-7, and poly (ADP-ribose) polymerase (PARP). Aviculin (2)-induced apoptotic cell death was accompanied by an increase in the Bax/Bcl-2 ratio. These findings demonstrated that aviculin (2) could induce breast cancer cell apoptosis through the intrinsic apoptosis pathway, and it can therefore be considered an excellent candidate for herbal treatment of breast cancer.

Purpose: The aim of this study was to investigate the efficacy and safety of a mixed extract of Trigonella foenum-graecum seed and Lespedeza cuneata (TFGL) for the treatment of testosterone deficiency syndrome (TDS). Materials and Methods: Patients were instructed to take a placebo or 200 mg TFGL capsule twice per day for 8 weeks. The primary efficacy variable was the change from baseline in the Aging Males' Symptoms scale (AMS), as well as levels of serum total and free testosterone. Secondary efficacy measurements included changes from baseline in the number of 'yes' answers on the Androgen Deficiency in the Aging Male (ADAM) questionnaire, levels of serum total cholesterol, high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), triglyceride, all domain scores of the International Index of Erectile Function (IIEF), perceived stress scale-10 (PSS-10), as well as changes in body composition. Results: The TFGL group exhibited a significant improvement in the AMS scores at 8 weeks, total testosterone at 8 weeks, and free testosterone at 4 and 8 weeks. At 4 weeks, 25% of the TFGL group changed to negative in terms of ADAM scores and 34.1% of the TFGL group had negative scores at the end of the study. The TFGL group exhibited a significant improvement in total cholesterol, HDL-C, LDL-C, triglyceride, IIEF scores, and PSS-10 scores at 8 weeks. Conclusions: The mixed extract of TFGL resulted in significant improvements in symptoms of TDS, as measured by the AMS, ADAM, PSS-10 and testosterone levels. [ABSTRACT FROM AUTHOR]

Abstract: To develop the ingredient with the cosmeceutical function, the antiwrinkle and antimelanogenesis effects of the ethanol extract of Lespedeza cuneata G. Don were investigated. DPPH radical scavenging activity was significantly increased with the extract of L. cuneata G. Don. Cell viability on CCD986Sk human fibroblast was also increased by the ethanol extract of L. cuneata G. Don. The inhibitory function of the extract of L. cuneata G. Don on collagenase, elastase, and tyrosinase was evaluated. Protein expression level of Claudin‐1, Occludin, and ZO‐1 was up‐regulated in HaCaT human keratinocyte by the extract of L. cuneata G. Don. In addition, the extract of L. cuneata G. Don inhibited melanin synthesis in B16F10 murine melanoma cells by decreasing MITF, TRP1, and TRP2 protein levels and increasing the phosphorylated Erk and Akt. Purpose Bisphenol A (BPA) has been regarded as a possible risk factor for reproductive health. We examined potential reproductive health benefits of Lespedeza cuneata ethanol extract (LCE). Previously, Lespedeza cuneata showed many therapeutic effects. However, the protective effect of LCE on BPA-induced testicular dysfunction and its mechanisms have not been precisely studied. Methods Mice were randomly divided into six groups (n = 7). Sperm counts and motility were measured by light microscope. Testosterone, total cholesterol, triglycerides, HDL, LDL-cholesterol, glucose, free fatty acids, hs-CRP, Angiotensinogen, Angiotensin II, GOT, GPT, TBARS, GSH, CAT, and SOD1 were measured in mouse serum. The potential protective effects of the LCE on mouse sertoli cells were evaluated. Results Oral administration of LCE in BPA-exposed male mice restored testis weight, sperm count, motility, and testosterone levels by inhibiting markers in serum. In addition, treatment with LCE in BPA-treated TM4 sertoli cells recovered cell viability by attenuating Bax expression and activating caspase 3 and PARP. Conclusions These results indicate that LCE prevented BPA-induced testicular dysfunction and cell viability in BPA-treated TM4 sertoli cells. Lespedeza cuneata (Dum. Cours.) G. Don. (Fabaceae), known as Chinese bushclover or sericea lespedeza, has been used in traditional medicine to treat diabetes, hematuria, and insomnia, and it has been reported that bioactive compounds from L. cuneata possess various pharmacological properties. However, there has been no study to determine the active compounds from L. cuneata with potential activity against ovarian cancer. This study aimed to isolate cytotoxic compounds from L. cuneata and identify the molecular mechanisms underlying the apoptosis pathway in ovarian cancer cells. Based on cytotoxic activity identified in the screening test, chemical investigation of the active fraction of L. cuneata led to the isolation of nine compounds including four lignanosides ( 1 – 4 ), three flavonoid glycosides ( 5 – 7 ), and two phenolics ( 8 – 9 ). Cytotoxicity and the molecular mechanism were examined by methyl thiazolyl tetrazolium (MTT) assay and Western blot analysis. Of the isolated compounds, (−)-9′- O -(α- l -rhamnopyranosyl)lyoniresinol ( 3 ) demonstrated the strongest effect in suppressing A2780 human ovarian carcinoma cell proliferation in a dose-dependent manner, with an IC 50 value of 35.40 ± 2.78 μM. Control A2780 cells had normal morphology, whereas cell blebbing, shrinkage, and condensation were observed after treatment with compound 3 . Western blotting analysis showed that compound 3 inhibited A2780 human ovarian cancer cell viability by activating caspase-8, caspase-3, and PARP, which contributed to apoptotic cell death. These results suggest that (−)-9′- O -(α- l -rhamnopyranosyl)lyoniresinol ( 3 ) has potent anticancer activities against A2780 human ovarian carcinoma cells through the extrinsic apoptotic pathway. Therefore, (−)-9′- O -(α- l -rhamnopyranosyl)lyoniresinol is an excellent candidate for the development of novel chemotherapeutics. [ABSTRACT FROM AUTHOR]

Lespedeza cuneata (poorman’s lucerne; sericea lespedeza), a tannin-rich perennial legume, was offered as hay to dry Merino ewes in a confined feeding experiment to evaluate the effect on the level of gastrointestinal parasite infection in sheep. Medicago sativa (a low tannin containing perennial legume) was used as the control treatment. Parameters faecal egg count (FEC), FAMACHA© scores and rectal temperatures were used. FECs were substantially lower (p = 0.05) in the Lespedeza group after 35 days, together with a trend of higher rectal temperatures, compared with the Medicago group. Although non-significant (p > 0.05), the higher rectal temperatures suggested a lower level of anaemia in the sheep on the Lespedeza ration and, therefore, a lower parasite-worm burden. However, FAMACHA© scores showed no significant (p > 0.05) differences between treatments despite the differences in FEC that were recorded, indicating that host homeostasis was possibly mediated by improved nutrition as a result of the high protein content of both experimental diets.

Control of invasive exotic species in restorations without compromising the native plant community is a challenge. Efficacy of exotic species control needs to consider collateral effects on the associated plant community. We asked (1) if short-term control of a dominant exotic invasive, Lespedeza cuneata in grassland restorations allows establishment of a more diverse native plant community, and (2) if control of the exotic and supplemental seed addition allows establishment of native species. A manipulative experiment tested the effects of herbicide treatments (five triclopyr and fluroxypyr formulations plus an untreated control) and seed addition (and unseeded control) on taxonomic and phylogenetic diversity, and community composition of restored grasslands in three sites over three years. We assessed response of L. cuneata through stem density counts, and response of the plant community through estimates of canopy cover. Herbicide treatments reduced the abundance of the exotic in the first field season leading to a less dispersed community composition compared with untreated controls, with the exotic regaining dominance by the third year. Supplemental seed addition did not provide extra resistance of the native community to reinvasion of the exotic. The communities were phylogenetically over-dispersed, but there was a short-term shift to lower phylogenetic diversity in response to herbicides consistent with a decrease in biotic filtering. Native plant communities in these grassland restorations were resilient to short-term reduction in abundance of a dominant invasive even though it was insufficient to provide an establishment window for native species establishment.

While some plants have modified seed structures to facilitate dispersal, many lack such specialised adaptations, making their mode of dispersal unclear. This can be particularly problematic for predicting shifts in species ranges or tracking the spread of invasive plants. As an example, the seed size and shape of the invasive legume, Lespedeza cuneata, suggest that wind and attachment to animals are not important for dispersal, yet populations can spread surprising distances within a few years. Using a series of experiments conducted in the laboratory and field, we tested wind and mammal fur as mediators of seed dispersal. To test wind dispersal, seed traps were arranged radially around a patch of L. cuneata and seeds were collected following dispersal. Attachment to mammal fur was tested by fitting pelts of a deer, coyote and raccoon to artificial torsos and determining seed retention in both the field and the laboratory. Laboratory trials also examined the influence of wet versus dry conditions. Our results showed that wind direction strongly influenced dispersal distance and seeds were readily dispersed by mammal fur. The number of seeds retained was species specific, depending on fur depth and mammal size, with seed retention increasing under wet conditions. Together, these results suggest that both wind and mammal fur contribute to the movement of L. cuneata across grasslands. Invasive plant species can modify soils in a way that benefits their fitness more than the fitness of native species. However, it is unclear how competition among plant species alters the strength and direction of plant-soil feedbacks. We tested how community context altered plant-soil feedback between the non-native invasive forb Lespedeza cuneata and nine co-occurring native prairie species. In a series of greenhouse experiments, we grew plants individually and in communities with soils that differed in soil origin (invaded or uninvaded by L. cuneata) and in soils that were live vs. sterilized. In the absence of competition, L. cuneata produced over 60% more biomass in invaded than uninvaded soils, while native species performance was unaffected. The absence of a soil origin effect in sterile soil suggests that the positive plant-soil feedback was caused by differences in the soil biota. However, in the presence of competition, the positive effect of soil origin on L. cuneata growth disappeared. These results suggest that L. cuneata may benefit from positive plant-soil feedback when establishing populations in disturbed landscapes with few interspecific competitors, but does not support the hypothesis that plant-soil feedbacks influence competitive outcomes between L. cuneata and native plant species. Sericea lespedeza [Lespedeza cuneata (Dumont de Courset) G. Don., SL] is a perennial, warm-season forage legume with wide adaptation, freeze tolerance, establishment ease, and persistence under grazing. It has high condensed tannin (CT) concentrations (g kg−1), which could be useful for methane mitigation and rumen bypass protein, as well as insect pest and gastro-intestinal nematode suppression. However, CT concentration is variable. Our objective was to test CT, crude protein (CP), and fiber concentrations at five locations (Georgia, Alabama, North Carolina, Louisiana, and Texas) during a growing season. Established ‘AU Grazer’ SL regrowth and accumulated herbage in separate trials was sampled starting late April 2010 every 35 d for four dates. Regrowth CP concentration ranged (P ≤ 0.05) from 127 g kg−1 in North Carolina on date 3 up to 221 g kg−1 in Alabama on date 1. Sericea lespedeza regrowth in the warmest latitude (Louisiana) consistently contained among the greatest (P ≤ 0.05) acid detergent fiber. In regrowth herbage, we recorded a wide (P ≤ 0.05) total CT range, from 133.5 mg kg−1 in Texas on date 3 to 46.1 mg kg−1 in Louisiana on date 1, reflecting variability in fiber-bound, protein-bound, and especially extractible CT. Results from accumulated herbage reflected (P ≤ 0.05) herbage ontogenesis with less CP and greater fiber concentration with maturity but showed the same lack of CT concentration pattern as the regrowth herbage. These results indicate that SL herbage nutritive value and CT concentration is variable, so that any hay or pellets sold commercially for CT content should be assayed by environment and crop maturity. Studies have shown that conversion to conservation tillage from conventional tillage does not increase total soil organic carbon (SOC) significantly in the short term (<5 yrs). While no-till increases total SOC in the medium to long term, it was hypothesized that the inclusion of high biomass cover crops and organic mulches may increase total SOC in the short term. The use of invasive, perennial leguminous species as mulch may sustainably increase SOC during limited-input fall vegetable production. The objective of this study, conducted in Alabama (USA), was to quantify total SOC changes due to organic mulches and forage soybean (Glycine max(L.) Merr. cv. Derry) as a summer cover crop after conversion to no-till during limited-input fall collard (Brassica oleraceaL.) production. Forage soybean as a summer cover crop did not increase SOC. No-till without mulch (control treatment) significantly increased SOC at 0–5 cm from 6.3 to 14.0 g kg−1soil in 3 yrs, whereas inclusion of cut-and-carry mulches increased SOC to ≥22.6 g kg−1soil. Treatments did not affect collard yield, which averaged 17,863 kg ha−1yr−1. Purpose: To evaluate the effect of a mouthwash containing Lespedeza cuneata extract (LCE) on halitosis as an alternative to chemical mouthwashes. The effect of this natural mouthwash on halitosis-causing bacteria (HCB) was clinically evaluated., Materials and Methods: A total of 84 subjects among 103 participants were recruited from the M Dental Clinic (Busan, South Korea) in this randomised, double-blind, placebo-controlled study. The participants were divided into two groups: 41 in the saline-gargle group (saline group) and 43 in the LCE-gargle group (LCE group). A week before the experiment, scaling and oral health education were conducted to standardise the subjects' oral condition. For clinical evaluation, halitosis and HCB were evaluated pre-gargle (baseline), immediately after gargling (treatment), and 5 days after gargling (5 days post-treatment). Statistical analysis was performed using SPSS for Windows., Results: The level of subjective improvement was statistically significantly greater in the LCE group than the saline group (p < 0.05). Compared with the saline group, the LCE group showed reduced halitosis after 5 days of application. Furthermore, halitosis statistically significantly decreased over time (p < 0.05). Moreover, the LCE group showed a statistically significant decrease in the number of all six HCBs (p < 0.05)., Conclusion: LCE gargle was effective in reducing halitosis both subjectively and objectively, suggesting an antibacterial effect on HCBs in the oral cavity. Thus, LCE, which can be used as a safe mouthwash ingredient, can promote oral health and will be valuable for the oral healthcare product industry. It might also be helpful for people suffering from halitosis.
