Research on the assessment and management of pain in cows following difficult or assisted calving is still limited, especially on the effects of analgesics intended to mitigate this pain. The purpose of this study was to assess the effects of flunixin meglumine on the health and production of Holstein cows after calving. In total, 34 flunixin-treated and 38 placebo-treated animals were enrolled in a precalving treatment trial. A total of 633 animals given flunixin and 632 animals administered a placebo were enrolled in a postcalving treatment trial. In both cases, animals were randomly assigned to treatment, and researchers were blind to treatment condition until after analysis. A total of 1,265 animal records were analyzed for milk production for the first 14 d in milk and health outcomes for the first 30 d in milk. Animals treated with flunixin meglumine before calving had a significantly increased risk of stillbirth. Animals treated immediately after calving had increased odds of having a retained placenta and, in turn, increased risk of a high temperature, decreased milk production, and an increased risk of developing metritis. The objectives of this study were to evaluate the effects of free-access acidified milk replacer feeding on the pre- and postweaning health of dairy and veal calves. Individually housed calves were systematically assigned at birth to 1 of 2 feeding programs: free-access feeding (ad libitum) of acidified milk replacer (ACD, n = 249) or traditional restricted feeding (3 L fed twice daily) of milk replacer (RES, n = 249). Calves were fed milk replacer containing 24% crude protein and 18% fat. Acidified milk replacer was prepared to a target pH between 4.0 and 4.5 using formic acid. Calves were weaned off milk replacer at approximately 6 wk of age. Weaning occurred over 5 d, and during this weaning period, ACD calves had access to milk replacer for 12 h/d and RES calves were offered only one feeding of milk replacer (3 L) daily. Calves were monitored daily for signs of disease. Fecal consistency scores were assigned each week from birth until weaning. A subset of calves was systematically selected for fecal sampling at 3 time points between 7 and 27 d of age. Fecal samples were analyzed for enterotoxigenic Escherichia coli F5, Cryptosporidium parvum, rotavirus, and coronavirus. Hip width, hip height, body length, heart girth, and body weight were measured at birth and weaning. Postweaning body weight measurements were collected from the heifers at approximately 8 mo of age. Postweaning body weight and carcass grading information was collected from the veal calves at slaughter once a live weight between 300 and 350 kg had been achieved. The odds of ACD calves being treated for a preweaning disease event tended to be lower than that of the RES calves (1.2 vs. 5.2%, respectively). Preweaning mortality, postweaning disease treatment, and postweaning mortality did not differ between feeding treatments. The ACD feeding treatment supported greater preweaning average daily gain (0.59 vs. 0.43 kg/d) and structural growth than RES feeding. Postweaning average daily gain and carcass characteristics were similar for ACD and RES calves. These results indicate that free-access acidified milk replacer feeding tended to support improved health, and greater body weight gain and structural growth during the preweaning period; these effects did not persist in the postweaning period. Recent research suggests that circulating β-hydroxybutyrate (BHB) levels may be a meaningful indicator of grain intake and rumen development in pre-ruminant calves. As such, BHB levels may be a surrogate measure of rumen function to contribute to minimal weaning stress during the transition from liquid to solid feed. The primary objective of this study was to determine the optimal cut-point of circulating BHB levels that would be predictive of sufficient grain intake and rumen development for a successful transition from liquid to solid feed at the time of weaning. An additional objective was to validate the Precision Xtra (Abbott Diabetes Care, Abingdon, UK) calf-side test for determination of BHB in whole blood in calves around weaning, as compared with the gold standard laboratory method. A total of 20 Holstein female calves were randomly assigned at birth to be weaned at 6 wk (n = 10) or 8 wk (n = 10) of age. Milk replacer (mixed at 150 g/L) was offered at 1.2 kg/calf per d in 2 meals until a 1-wk step down, when milk meals were reduced by 50% 1 wk before weaning. Daily measurements included the intakes of starter grain, chopped straw, and water. Weekly measurements included body weight and blood BHB, until 70 d of life. To assess digestive tract development, rumen fluid samples were taken before and after weaning (d 35, 49, and 63) and analyzed for ruminal short-chain fatty acids. Whole blood was collected by jugular venipuncture, and BHB was determined by the Precision Xtra test at calf-side. In addition, serum was separated from a clotted sample, frozen, and stored until laboratory analysis was performed. Laboratory BHB results were correlated with both the Precision Xtra test (r = 0.95) and starter intake over 1 d (r = 0.89), a 3-d average (r = 0.90), and a 7-d average (r = 0.90). Additionally, laboratory BHB results were associated with total ruminal volatile fatty acids (r = 0.82), ruminal butyrate (r = 0.77), and body weight (r = 0.69). A receiver operating characteristic curve was created to plot the true positive rate against the false positive rate at 10 μmol/L BHB intervals to determine the optimal cut-point of circulating BHB that is predictive of an average starter intake of 1,000 g/d over a 3-d period. The optimal combination of sensitivity (95.7%) and specificity (96.1%) was at 100 μmol of BHB/L of blood. A value of 0.2 mmol/L on the Precision Xtra test yielded a sensitivity and specificity of 84.0 and 97.2%, respectively, over the 3-d average period of starter intake. These results show considerable promise for use of the Precision Xtra whole blood BHB test in the decision-making process of determining sufficient starter grain intake and rumen development for a successful transition from liquid to solid feed, and indicate that this test conducted at calf-side is highly accurate. The effect of administering recombinant bovine somatotropin (rbST) to cows with hyperketonemia during the early postpartum period on health, metabolic parameters, milk production, and early reproductive performance was evaluated in a double-blinded clinical trial. Cows from 8 dairy herds in New York State were tested weekly between 3 and 16 d in milk for elevated serum β-hydroxybutyrate. Cows were enrolled in the study when blood β-hydroxybutyrate was ≥1.3 mmol/L for the first time. Enrolled cows were randomly assigned to a treatment (n = 273) or placebo control (n = 270) group. Treated cows were given 325 mg of rbST subcutaneously on the day of enrollment and again 14 d later. Control cows received the same regimen except the syringe contained only the carrier without somatotropin. After enrollment, blood samples were collected weekly for 4 wk and submitted to the laboratory to be analyzed for selected metabolites. Risk ratios for clinical diseases subsequent to treatment were calculated using Poisson regression. Continuous data were analyzed using linear mixed models. Time to first insemination was assessed with survival analysis. In the 42 d following the first administration of rbST, incidence risks of displaced abomasum, clinical ketosis, metritis, clinical mastitis, and lameness were not different between treatment groups. Cows treated with rbST had a slightly lower body condition score 28 d after enrollment compared with control cows. In the 4 wk following enrollment, serum nonesterified fatty acids and aspartate amino-transferase were slightly higher for treated than control cows, respectively. Serum glucose, calcium, haptoglobin, and β-hydroxybutyrate were similar between groups. Treatment had no effect on resolution of hyperketonemia in any of the 4 wk after enrollment. Milk production in either of the 2-wk periods after each treatment was not different between treated and control cows. Furthermore, milk production was not different between groups from enrollment to 98 d in milk (42.6 ± 0.6 and 42.1 ± 0.7 kg/d for treatment and control groups, respectively). Treatment had no effect on time to first insemination (83 and 74 d in milk for treatment and control groups, respectively; hazard ratio = 0.72) or first insemination pregnancy risk (27 and 29% for treatment and control groups, respectively; risk ratio = 0.92). Bovine respiratory disease (BRD) is one of the most common infectious causes of morbidity and mortality in young dairy cattle. The objective of this randomized clinical trial was to determine the effectiveness of 1 or 2 doses of a 5-way, modified-live viral vaccine, administered to heifer calves before weaning to aid in the prevention of BRD. The hypotheses were that vaccination would reduce the incidence of BRD and mortality and that 2 doses would be more effective than 1. A total of 2,874 heifer calves from 19 commercial dairy farms in Minnesota and Ontario were enrolled at 1 to 7 d of age and were followed until 3 mo of age. Calves were randomly assigned to receive a commercial, intramuscular, modified-live vaccine against bovine vira diarrhea virus types 1 and 2, bovine respiratory syncytial virus, bovine herpesvirus type 1, and parainfluenza virus type 3 at 15 to 21 d of age (2 wk only), 35 to 42 d (5 wk only), both 2 and 5 wk, or sterile saline at both times (unvaccinated controls). The incidence of failure of passive transfer was 11 or 32%, using cut-points of serum total protein of 5.2 and 5.7 g/dL, respectively Overall, 22% of calves were treated at least once for BRD. The incidence risk of naturally occurring BRD was 7.7% before 2 wk of age, 8.0% between 2 and 5 wk and 9.5% between 5 wk and 3 mo of age, and was not different between vaccination groups. Overall mortality throughout the 3-mo study period was 3.5%. Mortality was 1.6% before 2 wk of age, 0.5% between 2 and 5 wk, and 1.2% between 5 wk and 3 mo of age. The risk of mortality was not affected by vaccination. Mean average daily gain of 1.07 kg/d from 5 wk to 3 mo of age was not different between vaccine groups. In this population of commercial, home-raised calves, with an overall low incidence of failure of passive transfer intramuscular vaccination with a multivalent, modified live viral vaccine at 2 or 5 wk of age or both was not associated with a decreased risk of BRD or mortality, or with growth until 3 mo of age. Reasons for these findings may include interference by maternal antibodies, unresponsiveness of the neonatal immune system, timing of immunity relative to pathogen exposure, disease caused by pathogens other than the viruses in the vaccine, or herd immunity. However, in populations with higher incidence of failure of passive transfer or risk of BRD, calves with low levels of specific antibodies may respond differently to vaccination. The objective was to compare reproductive performance with management programs based on an automated activity monitoring (AAM) system or a synchronized breeding program under field conditions. In total, 1,429 Holstein cows from 3 commercial herds in Ontario, Canada, were enrolled over 1 yr in a randomized controlled trial. At each farm, primiparous and multiparous animals were housed in separate pens. At the pen level, cows were assigned to reproductive management primarily using an AAM system based on monitoring activity levels (Heatime, SCR Engineers Ltd., Netanya, Israel) or a timed artificial insemination (TAI) program. A crossover occurred after 6 mo of the trial to avoid confounding treatment with parity. Insemination based on additional detection of estrus by visual observation was practiced in all pens. At the individual cow level, time to pregnancy throughout the study (n = 1,985 cow-6 mo periods), time to first service, and time to second service were analyzed with a Cox proportional hazards models accounting for herd, and did not differ overall between the AAM and TAI treatment groups. However, we observed an interaction between herd and breeding program, such that association of AAM- or TAI-based program with these outcomes differed between farms. Time to pregnancy was not different in herd A [median = 151 and 136 d; hazard ratio (HR) = 0.93] and herd C (median = 99 and 124, HR = 1.24), whereas herd B had a median time to pregnancy of 119 d and 146 d (HR = 1.3) in the AAM and TAI groups, respectively. Under conditions in which 19 to 32% of artificial inseminations in both groups were based on visually detected estrus, herd pregnancy rate and cow-level time to pregnancy did not differ overall between TAI- and AAM-based programs, but the effect of reproductive management approach depended on herd. Considering 924 cow-6 mo periods with artificial inseminations only by the assigned program, we also found an interaction between herd and breeding program. Stratified analysis showed no difference in time to pregnancy in herd A (HR = 1.3), whereas in herds B (HR = 1.7) and C (HR = 2.8), cows in the AAM treatment group became pregnant sooner compared with those in the TAI group. Factors that influence the variability in relative performance of these management systems between herds require further investigation. The use of flunixin meglumine (FM), a nonsteroidal antiinflammatory drug, during experimentally induced Escherichia coli mastitis was evaluated. Twenty-four primiparous and multiparous lactating dairy cows were challenged with 1 x 102 cfu of E. coli 727 in 1 uninfected quarter. Of the 24 E. coli-challenged animals, 12 were administered FM [ECF; 100 mg (2 cc)/45.5 kg of body weight) at the onset of clinical mastitis signs. The remaining 12 challenged cows were untreated (EC). An additional 11 cows were infused with 1 mL of sterile phosphate-buffered saline and served as the nonchallenged control (CTL) group. Activity measures, dry matter intake (DMI), milk production, milk bacterial counts from challenged mammary glands, and somatic cell score (SCS) were collected on all animals. Activity measurements were collected using both a behaviormonitoring system and data loggers. Activity was summarized by day (behavior-monitoring system) and in 3-h time periods (data loggers). An examination of animal activity indicated that EC and ECF cows stood more and lay less as compared with the CTL animals in the first 6 h after FM administration. When DMI was analyzed, CTL and ECF animals had greater DMI than the EC animals on d 1 postchallenge. However, by d 2 postchallenge, DMI for ECF and EC cows was significantly less than for the CTL cows. The ECF cows had greater milk yield than did EC animals by d 3 and 4 postchallenge, and no significant difference in yield was observed between the ECF and CTL animals. No differences in SCS were observed between the parity groups. Yet, bacterial counts in milk were greater in multiparous animals compared with the primiparous cows. Therefore, it can be concluded that E. coli mastitis does alter animal activity and may have a negative effect on animal well-being. However, the improvement in DMI and milk production for ECF animals provides evidence for using a nonsteroidal antiinflammatory drug as supportive therapy in alleviating the adverse effects associated with E. coli mastitis. Captive cynomolgus macaques are prone to obesity, increasing their risk for developing hyperglycemia and type 2 diabetes mellitus (T2DM). Social rank may be a contributing risk factor predisposing macaques to adverse health events. Using retrospective health records from 259 animals, a matched case-control study was conducted to assess risk factors for developing hyperglycemia in group-housed, adult females aged 10 or older. Univariable exact and conditional logistic regression models were used to analyze the data. The odds of developing hyperglycemia were significantly greater in animals with more frequent counts of injury. Similarly, subordinate animals had higher odds of developing hyperglycemia than affiliates. Subordinate social status may increase the risk of hyperglycemia in mature female cynomolgus macaques. Opportunities for subordinates to alter feeding strategies are reduced in captivity. This may be associated with increased social stress around feeding, and for animals housed long-term could predispose them to obesity and hyperglycemia. [ABSTRACT FROM AUTHOR]

The objective of this study was to determine how different feeding methods may affect the learning of feeding, sorting, and competitive behavior of growing dairy heifers. We hypothesized that heifers previously fed a total mixed ration (TMR) would distribute their feeding time more evenly throughout the day, sort the new ration less, compete less for feed, maintain a more solid fecal consistency, and continue to grow rapidly compared with heifers previously fed a top-dressed ration (TDR). Thirty-two Holstein heifers (237.2 × 21.9 d of age) were divided into 8 groups of 4 and exposed to 1 of 2 treatments for 13 wk: 1) TMR or 2) TDR, with each containing 65% grass/alfalfa haylage and 35% textured concentrate on a dry matter (DM) basis. Following this feeding period, all heifers were switched to an unfamiliar TMR containing 56.1% grass/alfalfa haylage, 21.0% corn silage, 21.0% high-moisture corn, and 1.9% mineral supplement (DM basis) for 7 wk. Group DM intakes were recorded daily throughout the experiment. Feeding behavior, recorded using timelapse video, and sorting behavior were measured for 7 d during wk 1, 4, and 7 after the dietary change. Feeding competition was measured on d 2, 4, and 6 of each recording week. Sorting activity was determined through particle size analysis of the fresh feed and orts. The particle size separator separated feed into 4 fractions (long, medium, short, and fine). Sorting of each fraction was calculated as actual intake expressed as a percentage of predicted intake. Animals were scored for fecal consistency twice weekly, using a scale from 1 (liquid) to 4 (solid). Heifers were weighed every 2 wk. Neither DM intake (9.0 kg/d) nor average daily gain (1.2 kg/d) differed between treatments. Sorting also did not differ between treatments. Heifers tended to spend more time feeding if they had previously been fed a TDR (198.8 vs. 186.8 min/d). As they had done before the dietary change, heifers previously fed the TDR spent more time at the bunk in the 2 h following feed delivery (40.6 vs. 25.9 min/d). Heifers previously fed the TDR were displaced from the feed bunk more frequently than heifers previously fed the TMR (23.0 vs. 13.1 times/d), particularly during the 2-h period following feed delivery. Fecal scores were lower (more fluid in consistency) for heifers originally fed the TDR (3.2 vs. 3.7). The continued difference in feed bunk competition suggests that heifers previously fed the TDR had learned these patterns in the past, retaining them even when switched to an unfamiliar ration. Furthermore, lower fecal scores for heifers previously fed the TDR suggest altered rumen fermentation, possibly because of altered diurnal consumption patterns. These results show that feeding a TMR to replacement dairy heifers from a young age promotes a more even diurnal feeding pattern, minimizes feed bunk competition, and promotes a more solid fecal consistency. Thirty-two Holstein heifers (146.2 ± 21.9 d of age) were divided into 8 groups of 4 and exposed to 1 of 2 feed delivery treatments for 13 wk. The treatment rations contained 65% grass/alfalfa haylage and 35% textured concentrate (on a dry matter basis) fed as a 1) total mixed ration (TMR) or 2) top-dressed ration (TDR). Group dry matter intakes were recorded daily throughout the experiment. Feeding behavior, recorded using time-lapse video, and sorting behavior were measured for 7 d during each of wk 1, 5, 9, and 13. Sorting activity was determined through particle size analysis of the fresh feed and orts. The particle size separator separated feed into 4 fractions (long, medium, short, and fine). Sorting of each fraction was calculated as actual intake expressed as a percentage of predicted intake. Heifers were fecal scored for consistency of stool twice weekly using a scale from 1 (liquid) to 4 (solid); heifers were weighed every 2 wk. Neither dry matter intake (7.3 kg/d) nor average daily gain (1.3 kg/d) differed between treatments. Heifers fed the TDR tended to consume less neutral detergent fiber than heifers fed the TMR (4.77 vs. 4.91 kg/d). Heifers fed the TDR sorted against long particles (98.9 vs. 96.0%) and consumed short particles (100.3 vs. 101.1%) to a greater extent than did heifers fed the TMR. Daily feeding time did not differ between treatments (201.0 min/d), but heifers on the TDR did spend more time at the bunk in the 2 h following feed delivery (50.1 vs. 32.0 min/d). Heifers fed the TDR were displaced from the feed bunk more frequently than heifers fed the TMR (17.6 vs. 8.6 times/d), particularly during the 2-h period following feed delivery. Fecal scores were lower for heifers on the TDR (2.7 vs. 3.4). These results suggest that feeding a TMR to replacement dairy heifers may promote a more even diurnal feeding pattern, minimize feed sorting and feed bunk competition, and promote more solid fecal consistency. Adequate passive transfer of maternal immunoglobulin is important for optimal health and performance in newborn dairy calves. From June to October 2003 and January to April 2004, blood samples were collected from 961 dairy calves 0 to 8 d of age on 11 farms in southwestern Ontario. This was followed by a second study conducted from May to October 2004, in which similar samples were taken from 422 calves up to 8 d of age on 119 dairy farms throughout southern Ontario. For each sample collected, serum refractometry was used to evaluate serum total protein (TP) as a measure of passive transfer of maternal immunity. During each study, producers were asked to provide information on calf management practices, including details of colostrum feeding. Data were analyzed to assess the levels of maternal immunity present in the calves, and to investigate whether these were associated with any calf management or colostrum feeding practices used on the farms. Serum TP readings ranged from 3.5 to 9.8 g/dL. Controlling for any effects of variation between farms, we found no statistically significant difference in serum TP levels, or risk of failure of passive transfer (FPT), between heifer and bull calves. The odds of FPT in calves on farms where more than 75% of cows were usually allowed to remain with their calves for more than 3 h after calving were significantly higher than the odds of FPT in calves on farms where dams and calves were separated within 3 h of the birth. Furthermore, an increased volume of colostrum given to calves within 6 h of birth was significantly associated with a reduced risk of FPT in calves. An investigation was conducted to compare the effects of the monensin controlled-release capsule, monensin sodium in feed, and a negative control on feed intake and metabolic parameters in a randomized and blinded clinical trial. A total of 136 Holstein cows and heifers were assigned to a negative control group, administered a monensin controlled-release capsule (CRC) or administered 22 mg/kg of dry matter of monensin sodium in the total mixed ration (premix). Cows were enrolled 3 wk prior to expected calving; at this time monensin treatment began. Cows were located at the Elora Dairy Research Centre (Elora, Ontario, Canada). Blood samples were obtained at enrollment, at 1 wk prior to expected calving date, at calving, and at 1 and 2 wk postpartum. Sera from these samples were analyzed for/%hydroxybutyrate (BHBA), nonesterified fatty acids, glucose, urea, bilirubin, aspartate aminotransferase activity, insulin, and cortisol. Cows were assigned a body condition score upon enrollment and upon completion of the trial. The dry matter intake was measured for all cows for the entire experimental period (12.0, 11.7, and 11.3 kg/d for control, premix, and CRC groups, respectively). However, no differences in dry matter intake between treatment groups were noted. The interaction of experimental group and sampling time was significant for serum concentration of BHBA and urea. Both monensin delivery methods significantly decreased serum BHBA postpartum. Urea concentrations were increased in the postpartum period compared with the prepartum samples. The CRC group had a significant impact on reducing the loss in body condition over the study period. Serum concentrations of all measured metabolic parameters varied over the peripartum period. The efficacy and adherence of an external teat sealant applied at drying offwas evaluated in 2 studies between 1997 and 1999. At drying off, 2 quarters were randomized to receive intramammary dry-cow antibiotic therapy, and the remaining 2 quarters were treated with either a single or double application of external teat sealant. Approximately 3 d before calving, all teats that had been dipped at drying off were redipped in a single coating of teat sealant. Adherence of the teat sealant was scored for the first 2 wk after drying off, and physical traits of the teat skin and teat ends were recorded. Quarter milk samples were collected 1 wk before drying off, at drying off, 0 to 7 d, and 14 to 21 d postcalving. Somatic cell counts were determined from quarter samples taken at d 7 and 14 to 21 d after calving. Data were analyzed from 172 dry periods of 162 cows. The mean time of sealant adherence following drying off application was 3 ± 0.13 d. Double sealant application significantly increased the duration of adherence by 0.67 d. Teats that had teat sealant applied twice at drying off and that had up to 3 d of adherence had the lowest linear score (LS) at 14 to 21 d (1.89 ± 0.31) of all quarters. The LS of quarters that received antibiotic therapy only was 2.27 ± 0.19. The majority of intramammary infections identified at drying off were caused by the minor pathogens, coagulase-negative staphylococci and Corynebacterium bovis (51 and 23%, respectively). The results from this study indicate that duration of sealant adherence to the teat-end should be considered when evaluating the impact of teat sealant treatment at drying off on the level of infection after calving. During 1998 and 1999, a purposive sample of Ontario dairy herds was enrolled in a study to examine management factors associated with adherence of an external teat sealant. A total of 74 herds were recruited that had previously complied with a provincial Sentinel Herd mastitis study. All herds were sent a management survey and a commercially available external teat sealant product. The sealant was applied to all cows in first lactation or greater that were scheduled to go dry. Adherence of the teat sealant was scored on a scale of 1 to 5 (1 = sealant completely removed) for the first 12 d following dry off. Complete data were analyzed from 806 cows in 48 herds (mean of 17 cows/ herd). Mean duration of adherence of the teat sealant was 4 d (range 1 to 7 d). Of the management factors recorded, only changing the feed to reduce milk production prior to drying off was significantly associated with 0.78-d prolonged adherence. No factors related to dry cow housing, bedding material, or floor surface were associated with the duration of adherence. The variance around the duration of adherence was examined. The median variance of adherence was 3.0 d2, which was equivalent to a standard deviation of 2.6 d. Herds in which dry cows were housed in tie-stall barns, and the herds that used a penicillin and novobiocin combination dry cow antibiotic had the greatest variation. We concluded that modifying the ration to reduce milk production enhanced adherence of an external teat sealant. Teat-end preparation prior to application of the teat sealant is an important factor to consider when choosing to adopt this dry period mastitis prevention strategy. An experimental teat dip containing 0.5% hydrogen peroxide as the active ingredient was compared with a teat disinfectant also containing 0.5% hydrogen peroxide that is commercially available throughout North America. The study was conducted under conditions of experimental challenge with a positive control following the guidelines recommended by the National Mastitis Council. The efficacy of the test product and the positive control product were compared in 45 cows, with 89 total teats receiving each product after milking during a 10-wk study period. There was no significant difference between the experimental hydrogen peroxide product and the positive control in new intramammary infections caused by Staphylococcus aureus (27.0 and 18.0% in the treatment groups, respectively). Additionally, there was no significant difference between the experimental product (6.7%) and the positive control groups (4.5%) in new intramammary infections caused by Streptococcus agalactiae. Traditional analysis of teat skin condition changes supported improved teat skin condition with the test disinfectant. After accounting for the correlation of teats within cow, significant differences were also observed between the treatment groups for teat skin condition. The experimental hydrogen peroxide-based teat disinfectant provided efficacy similar to that of the positive control teat disinfectant, with significant improvement in teat skin condition and no adverse effects on teat end condition. A 1.0% iodophore teat disinfectant (Full-Bac) was evaluated in comparison with a positive control (Bovadine), a commercially available 1.0% iodophore teat disinfectant. The study was conducted under conditions of experimental challenge, following the guidelines recommended by the National Mastitis Council. The test product and a positive control were compared in 41 cows, with 82 teats receiving each product at milking, during a 10-wk study period. There were no differences between the test product and the positive control in new intramammary infections due to Staphylococcus aureus, which averaged 13.4% in each of the 2 treatment groups. Additionally, no statistical difference was seen between the test product and positive control in new intramammary infections by Streptococcus agalactiae, which averaged 8.5 and 6.1% for the Full-Bac and Bovadine groups, respectively. Teat skin and teat end condition scores were statistically evaluated at wk 1, 5, and 9 of the study, and no significant differences were observed between the treatment groups. The test teat disinfectant provided similar germicidal activity to that of the positive control teat disinfectant with no adverse effects on teat skin or teat end condition during the warm-season study period. The objective of this field study was to identify metabolic tests available in clinical practice that identified cows at increased risk of left displaced abomasum (LDA). A technician visited 1044 cows in 20 herds weekly from 1 wk before expected calving until 1 wk postpartum. Cows were assigned a body condition score and samples were collected at each visit for measurement of serum nonesterified fatty acids (NEFA), cholesterol, β-hydroxybutyrate (BHBA), glucose, urea, calcium, and phosphorus, and a milk sample was collected postpartum for measurement of BHBA. The probability of LDA was modeled with multivariable logistic regression accounting for clustering. There were 53 cases of LDA (incidence risk = 5.1%) and the median time of diagnosis was 11 d in milk. In cows with LDA, mean NEFA concentrations began to diverge from the mean in cows without LDA 14 d before calving, whereas mean serum BHBA concentrations did not diverge until the day of calving. Prepartum, only NEFA concentration was associated with risk of subsequent LDA. Between 0 and 6 d before calving, cows with NEFA concentration ≥0.5 mEq/L were 3.6 times more likely to develop LDA after calving. For prospective application, among samples taken 4 to 10 d before expected calving, the optimum NEFA cut-point remained 0.5 mEq/L. The sensitivity, specificity, and likelihood ratio (LR) were 46%, 82%, and 2.6, respectively. Between 1 and 7 d postpartum, retained placenta, metritis, and increasing serum concentrations of BHBA and NEFA were associated with increased risk of subsequent LDA. However, considered separately, postpartum serum BHBA was a more sensitive and specific test than NEFA concentration. The odds of LDA were 8 times greater in cows with serum BHBA ≥1200 µmol/L (LR = 3.5). Cows with milk BHBA concentration ≥200 µmol/L were 3.4 times more likely to develop LDA. An axial first-order gradiometer is formed by coupling the input coil of a flexible high-temperature superconducting flux transformer inductively to a directly coupled superconducting quantum interference device magnetometer. The transformer is patterned in a single-layer YBa2Cu3O7-x film on a flexible Hastelloy tape. The tape is bent such flint file two outer pickup loops of the transformer are facing each other while perpendicular to the magnetometer plane resulting in a gradiometer baseline of 35 mm. Conception to synchronized ovulation (Ovsynch) using injections of GnRH and PGF[sub2α] and timed artificial insemination has been shown to be maximized when the program is initiated 5 to 12 d after estrus. The objective of this double-blinded field trial was to assess the effect of one injection of PGF[sub2α], 10 d before the Ovsynch program, on the probability of pregnancy at first insemination in lactating dairy cows. The hypothesis was that cows that underwent luteolysis in response to PGF[sub2α] would be between 5 and 8 d postestrus at the start of Ovsynch. In five commercial dairy herds in Ontario, Canada, at 52 ± 12 d in milk, 506 cows were assigned at random to receive either one i.m. injection of 500 µg of cloprostenol or saline. Ten days later, all cows received 100 µg of GnRH i.m., followed in 7 d by 500 µg of cloprostenol i.m. and 100 & micro;g of GnRH i.m. 48 h later. All cows were artificially inseminated 0 to 20 h after the second injection of GnRH, without regard to detection of estrus. Pregnancy was diagnosed by transrectal palpation at least 35 d after insemination. The probability of pregnancy after first insemination was modeled with logistic regression, accounting for the correlation of cows with herd and the effect of season of calving. There was no difference in pregnancy risk between cows that received PGF[sub2α], presynchronization and controls (37.3 and 36.6%, respectively; odds ratio = 1.03, 95% confidence interval, 0.88 to 1.20). High ambient heat and humidity have profound effects on the production, health, profitability, and welfare of dairy cattle. To describe the relationship between summer temperature and relative humidity in the barn and determine the appropriateness of using meteorological station data as a surrogate for on-farm environmental monitoring, a study was conducted on 48 farms in Ontario, Canada, over the summer (May through September) of 2013. Within-barn environmental conditions were recorded using remote data loggers. These values were compared with those of the closest official meteorological station. In addition, farm-level characteristics and heat-abatement strategies were recorded for each farm. Environmental readings within the barn were significantly higher than those of the closest meteorological station; however, this relationship varied greatly by herd. Daily temperature-humidity index (THI) values within the barn tended to be 1 unit higher than those of the closest meteorological station. Numerically, 1.5 times more mean daily THI readings were in excess of 68 (heat stress threshold for lactating dairy cows) in the barn, relative to the closest meteorological station. In addition, tiestalls, herds that were allowed access to pasture, and herds that had no permanent cooling strategy for their cows had the highest mean and maximum daily THI values. Minimum daily THI values were almost 4 units higher for tiestall relative to freestall herds. Overall, due to farm-specific and unpredictable variability in magnitude of environmental differences between on-farm and meteorological station readings, researchers attempting to study the effects of environment on dairy cows should not use readings from meteorological stations because these will often underestimate the level of heat stress to which cows are exposed. The forced cessation of milk production, or dry-off, is a routine management practice in dairy cattle, sheep, and goats. This practice initiates a dry period, during which the animal is not milked. Milking begins again after parturition. Most of the literature on the dry period has focused on how various drying-off strategies affect milk production and disease; little work to date has addressed how dry-off affects the overall welfare of the dairy animal. The first aim of this review was to present an overview of the importance of dry-off and how it is commonly achieved. Our review shows that much scientific progress has been made in improving health status between lactations. The second aim was to identify important gaps in the literature, of which 2 key research disparities have been identified. We find that much of the work to date has focused on cattle and very little research has examined dry-off in dairy sheep and goats. We also find a lack of research addressing how common dry-off methodologies affect animal welfare on more than just a biological level, regardless of species. [ABSTRACT FROM AUTHOR]

Recent research has revealed potential advantages of feeding an elevated plane of nutrition to calves during the preweaning period. However, calves fed more nutrients preweaning may be more susceptible to depressed growth and weaning stress during the transition from liquid to solid feed. The objective of this study was to investigate the relationship between the age of weaning and feed intake, and its influence on growth, gastrointestinal development, and behavioral indicators in dairy calves fed an elevated plane of nutrition during the preweaning period. To meet this objective, 20 female Holstein calves were randomly assigned at birth to be weaned at 6 or 8 wk. Milk replacer (mixed at 150 g/L) was offered at 1.2 kg/calf per day in 2 meals until a 1-wk step-down, when meals were reduced by 50% 1 wk before weaning. Daily starter, chopped oat straw, water intake, and weekly body weights were measured until d 70 of life. To assess digestive tract development, rumen fluid, fecal, and blood samples were taken before and after weaning (d 35, 49, and 63) and analyzed for ruminal short-chain fatty acids, blood β-hydroxybutyrate, and fecal starch, respectively. Behavioral indicators of weaning stress, including vocalizing and non-nutritive oral behavior, were measured by visual observation for 1 h, 3 times per week, before the second feeding of the day during the period from 2 wk before weaning to 2 wk after weaning. The calves weaned at 8 wk compared with 6 wk had higher average daily gain for the week preweaning (0.79 ± 0.09 vs. 0.34 ± 0.10 kg/d) and postweaning (1.05 ± 0.09 vs. 0.35 ± 0.11 kg/d), and were heavier at d 70 (99.9 ± 1.81 vs. 91.0 ± 2.26 kg). From 5 to 8 wk of age, starter and water intakes were lower in calves weaned at 8 wk of age. However, overall starter intake did not differ during the last week of the experiment. Furthermore, calves weaned at 8 wk compared with 6 wk had higher starter intake for 1 wk preweaning (1.36 ± 0.13 vs. 0.40 ± 0.08 kg/d) and postweaning (2.51 ± 0.20 vs. 1.16 ± 0.15 kg/d). In both treatments, weaning increased ruminal short-chain fatty acids, blood β-hydroxybutyrate, and fecal starch, yet the differences between the week before and after weaning were greater for calves weaned at 6 wk compared with those weaned at 8 wk. Treatment × week relative to weaning interactions indicated that several behaviors varied between early- and later-weaned calves during the week before weaning; calves weaned at 6 wk tended to exhibit 75% more non-nutritive oral behavior and spent 55% less time ruminating, and 36% less time lying compared with calves weaned at 8 wk. Under the conditions of this study, the results suggest that calves fed an elevated plane of nutrition preweaning have higher starter intakes and average daily gain during the weaning period when weaning is extended from 6 to 8 wk of age. Regionally aggregated bulk milk somatic cell count (BMSCC) data from around the world shows a repeatable cyclicity, with the highest levels experienced during warm, humid seasons. No studies have evaluated this seasonal phenomenon at the herd level. The objectives of this study were to define summer seasonality in BMSCC on an individual herd basis, and subsequently to describe the characteristics and dynamics of herds with increased BMSCC in the summer. The data used for this analysis were from all dairy farms in Ontario, Canada, between January 2000 and December 2011 (n ≈ 4,000 to 6,000 herds/yr). Bulk milk data were obtained from the milk marketing board and consisted of bulk milk production, components (fat, protein, lactose, other solids), and quality (BMSCC, bacterial count, inhibitor presence, freezing point), total milk quota of the farm, and milk quota and incentive fill percentage. A time-series linear mixed model, with random slopes and intercepts, was constructed using sine and cosine terms as predictors to describe seasonality, with herd as a random effect. For each herd, seasonality was described with reference to 1 cosine function of variable amplitude and phase shift. The predicted months of maximal and minimal BMSCC were then calculated. Herds were assigned as low, medium, and high summer increase (LSI, MSI, and HSI, respectively) based on percentiles of amplitude in BMSCC change for each of the 4 seasons. Using these seasonality classifications, 2 transitional repeated measures logistic regression models were built to assess the characteristics of MSI and HSI herds, using LSI herds as controls. Based on the analyses performed, a history of summer BMSCC increases increased the odds of experiencing a subsequent increase. As herd size decreased, the odds of experiencing HSI to MSI in BMSCC increased. Herds with more variability in daily BMSCC were at higher odds of experiencing MSI and HSI in BMSCC, as were herds with lower annual mean BMSCC. Finally, a negative association was noted between filling herd production targets and experiencing MSI to HSI in BMSCC. These findings provide farm advisors direction for predicting herds likely to experience increases in SCC over the summer, allowing them to proactively focus udder health prevention strategies before the high-risk summer period. This study investigated how the feeding behavior of pair-housed calves develops in response to reduced teat and feed place availability. Twenty Holstein bull calves were pair housed and provided with milk replacer (MR) and grain concentrate ad libitum via either (1) 1 teat and feed bucket/pen, such that calves could not feed simultaneously [competitive feeding (CF)] or (2) 2 teats and feed buckets/pen [noncompetitive feeding (NCF)]. The calves were weaned during wk 7 of life by incrementally diluting the MR. Postweaning, all pens were managed identically and offered a complete pelleted diet ad libitum via 2 feed buckets/pen (NCF) in period 1 (wk 8 and 9) and period 3 (wk 12 and 13) and exposed to a competitive challenge with CF in period 2 (wk 10 and 11). Feeding times and competitive interactions were recorded from video for 3 d/wk in wk 2, 4, and 6 and 2 d/wk in wk 8 to 13. Meal criteria were used to calculate daily meal frequency, meal time, and synchronized meal time (the percentage of meal time when calves within the pen were engaged in simultaneous meals). Milk replacer intake was subject to a treatment x week interaction, with calves in CF pens having lower MR intake (wk 2: 8.3 vs. 10.0 L/calf per day for CF vs. NCF), but compensating by increasing intake to a greater extent over time (wk 4-5: 13.3 vs. 11.7 L/d for CF vs. NCF). Corresponding to MR intake, meal frequencies and feeding times evolved differently over this period, increasing in CF pens and decreasing in NCF pens (wk 2: 5.8 vs. 11.1 and wk 4-6: 6.2 vs. 5.1 meals/d for CF vs. NCF). Calves in CF pens also spent less time engaged in synchronized meals (28 vs. 51% of meal time; standard error = 7.1) and displaced each other 5 times more frequently during synchronized eating. Postweaning, calves in previously CF pens maintained 5 times greater displacement frequencies and had fewer overlapping meals than calves in previously NCF pens (34.5 vs. 40.7% of meals, respectively). Postweaning, when calves were all fed noncompetitively (2 buckets/pen in periods 1 and 3), calves previously fed CF had greater intakes in period 3 (4.87 vs. 4.44 kg/d) and a tendency for greater intake in period 1 (2.80 vs. 2.63 kg/d), greater rates of intake in period 3 (87.8 vs. 72.0 g/min) and a tendency for greater rates of intake in period 1 (44.3 vs. 38.9 g/min), and greater meal frequencies in both periods (11.1 vs. 9.9 meals/d). In the competitive challenge period (1 bucket/pen in period 2), intake, rates of intake, and meal frequencies were subject to treatment x week interactions: calves in NCF pens had lower intake and meal frequencies, but increased meal frequency, intake, and rate of intake over time to a greater extent than calves previously in CF pens. Recent evidence exists to suggest that the risk of subclinical mastitis, particularly those infections caused by environmental pathogens, in dairy cows is related to standing and lying patterns. The objective of this study was to determine the association between postmilking standing duration (PMSD) of dairy cows milked 3x/d and risk of intramammary infection (IMI). Four commercial freestall dairy herds in Eastern Ontario, milking 3x/d, were enrolled in a longitudinal study. Forty Holstein-Friesian cows per herd were randomly selected as focal animals from those cows in each herd that met our selection criteria of days in milk (<200 d) and somatic cell count (<100,000 cells/mL). The study consisted of three 28-d periods. The study began following a regularly scheduled Dairy Herd Improvement test with the collection of quarter-level milk samples from all focal animals. Bacteriology was used to confirm infection status at the start of the study and for determination of incidence of IMI throughout the study. A new IMI was defined as having a culture-positive quarter-level sample when the previous sample (28 d prior) had been culture negative for the pathogen of interest. Four sets of quarter-level milk samples were obtained for each focal animal. Lying behavior was recorded for 5 d after each milk sampling using data loggers. For these 5 d, individual milking times, production, and feeding times were also recorded. Postmilking standing duration was analyzed by milking event, with increased PMSD being positively associated with provision of fresh feed or freshly pushed-up feed around the time of milking, greater feed bunk space per cow, and lower freestall stocking density. Over the study period, 456 new IMI were detected, resulting in a mean herd incidence rate of 3.22 IMI per quarter year. Coagulase-negative staphylococci (CNS) and Corynebacterium spp. IMI were statistically analyzed to determine relationship with PMSD; they were the 2 predominant pathogens representing 45 and 31% of IMI, respectively. Only CNS IMI was associated with PMSD. A nonlinear relationship between PMSD and incidence of CNS IMI was found; cows with a PMSD of 90 to 120 min were at a reduced risk for CNS IMI. The risk of experiencing CNS IMI was also reduced with increased frequency of feed push-ups and provision of fresh feed 60 min before to 90 min after milking and >540 min after milking. These results indicate that management practices that promote PMSD of 90 to 120 min, such as the provision of fresh feed or freshly pushed-up feed around the time of milking, providing ample feed bunk space per cow, and keeping freestall stocking density low, should be encouraged to reduce the risk of CNS IMI in freestall-housed cows milked 3x/d. We present the design and implementation of a high-temperature superconducting (HTS) Josephson junction detector array for terahertz (THz) imaging. The array device is made of ten YBa2Cu3Ox−7 (YBCO) step-edge junctions coupled to gold thin-film ring-slot antennas on a MgO substrate. The design and characterization of the detector array in response to a 0.6 THz signal are presented. The objectives of this study were to determine the effect of feed presentation on meal frequency and duration, as well as diurnal feeding patterns of dairy calves, and to assess any longer-term differences in feeding patterns resulting from previous experience. Twenty Holstein bull calves were exposed from wk 1 to 8 of life to 1 of 2 feed presentation treatments: concentrate and chopped grass hay (<2.5 cm) offered at a ratio of 7:3 as a mixture (MIX), or as separate components (COM). Feed was provided ad libitum. Calves received 8 L/d of milk replacer (1.2 kg of dry matter), with the amount progressively reduced after 5 wk to facilitate weaning by the end of wk 7. At the beginning of wk 9, all calves received the MIX diet and remained on trial for an additional 3 wk. Feeding behavior was recorded from video for 4 d during wk 6, 8, 9, and 11. In wk 6, calves fed MIX spent more time feeding than calves fed COM (56.7 vs. 46.8 min/d). In wk 8, calves fed MIX spent more time feeding (174.0 vs. 139.1 min/d) and had a lower rate of intake (11.5 vs. 14.7 g/min) compared with calves fed COM. Meal frequency was similar between treatments (12.2 meals/d). Diurnal feeding patterns in wk 8 were also affected by feed presentation, with calves fed MIX spending less time feeding at time of feed delivery and more time feeding throughout the rest of the daylight hours than calves fed COM. Diurnal feeding patterns of hay and concentrate in wk 8 differed for calves fed COM, with more time spent consuming hay at time of feed delivery and less time spent consuming hay throughout the rest of the day. Once calves previously fed COM were transitioned to the MIX diet in wk 9, meal frequency, meal duration, and diurnal feeding patterns were similar between treatments: both treatments spent similar amounts of time feeding (173.9 min/d) and had similar peaks in feeding activity at time of feed delivery, sunrise, and sunset. Provision of hay and concentrate to young calves as a mixed ration, compared with separate components, increases time spent feeding and results in more evenly distributed diurnal feeding patterns. This study examined how early exposure to different feed presentations affects development of feed sorting in dairy calves. Twenty Holstein bull calves were exposed for the first 8 wk of life to 1 of 2 feed presentation treatments: concentrate and chopped grass hay (<2.5 cm) offered ad libitum at a ratio of 7:3 as a mixture (MIX), or as separate components (COM). Calves received 8 L/d of milk replacer (1.2 kg of dry matter), with the amount progressively reduced after 5 wk to facilitate weaning by the end of wk 7. All calves received the MIX diet in wk 9 to 11 and, subsequently, a novel total mixed ration (TMR; containing 40.5% corn silage, 22.0% haylage, 21.5% high-moisture corn, and 16.0% protein supplement) in wk 12 to 13. Intake was recorded daily and calves were weighed twice a week. Fresh feed and orts were sampled on d 1 to 4 of wk 6, 8, 9, 11, 12, and 13 for analysis of feed sorting, which was assessed through nutrient analysis for the MIX diet and particle size analysis for the TMR. The particle separator had 3 screens (19, 8, and 1.18 mm) producing long, medium, short, and fine particle fractions. Sorting of nutrients or particle fractions was calculated as the actual intake as a percentage of predicted intake; values >100% indicate sorting for, whereas values <100% indicate sorting against. Feed presentation did not affect dry matter intake or growth. Prior to weaning, all calves selected in favor of hay; MIX calves consumed more neutral detergent fiber (NDF) than predicted (103.6%) and less nonfiber carbohydrates (NFC) than predicted (92.6%), and COM calves consumed, as a percentage of dry matter intake, 40.3% hay (vs. 30% offered rate). In wk 8, calves fed COM consumed more NFC than calves fed MIX (1.0 vs. 0.95 kg/d) and less NDF (0.43 vs. 0.54 kg/d), indicating greater selection in favor of concentrate. However, when provided the MIX diet, calves previously fed COM did not sort, whereas calves previously fed MIX consumed more NFC intake than predicted (103.2%) and less NDF intake than predicted (97.6%). Calves previously fed MIX maintained increased sorting after transition to the novel TMR, sorting against long particles (86.5%) and for short (101.8%) and fine (101.2%) particles. These results indicate that initially providing dairy calves with solid feeds as separate components, compared with as a mixed ration, reduces the extent of feed sorting in the weeks after transition to a common ration. The objectives of this study were to evaluate changes in weight shifting between legs while standing on a weighing platform in response to endotoxin-induced clinical mastitis, and to evaluate the effect of the non-steroidal antiinflammatory drug flunixin meglumine on weight distribution between legs while standing in dairy cattle with endotoxin-induced clinical mastitis. Clinical mastitis was induced in 10 primiparous and 9 multiparous lactating dairy cows (days in milk = 55 ± 12; mean ± standard deviation) by intramammary infusion of 100 µg of Escherichia coli lipopolysaccharide (LPS) into the right rear quarter. Four hours later, 10 animals were randomly assigned to receive flunixin meglumine intravenously (2.2 mg/kg of body weight; treated group) and 9 received an equivalent volume of sterile isotonic saline solution (control group). Body temperature was monitored rectally 3 d before LPS infusion, immediately before LPS infusion, and 4, 7, 10, 13, 16, and 28 h after LPS infusion. The weight applied to each leg was recorded while cows were standing on a weighing platform on the day before the challenge and 7, 10, 13, 16, and 28 h after LPS infusion. Two measures of weight shifting between the rear legs were calculated for each recording session: the standard deviation of the weight applied to the legs over time and the frequency of steps. The LPS infusion resulted in a consistent case of clinical mastitis approximately 4 h after the LPS infusion, as assessed by the presence of visible swelling and elevated rectal temperature in all cows. However, control animals had a higher temperature 7 h after LPS infusion compared with treated animals (40.8 vs. 39.0°C; standard error of the difference = 0.2). Overall, weight shifting between the rear legs was decreased 7 h after the LPS infusion compared with baseline, and this decrease was not affected by treatment with flunixin meglumine. It is likely that weight shifting increases friction between the swollen udder and the legs, increasing the pain experienced by the cow. Thus, cows with endotoxin-induced mastitis avoided shifting weight, particularly at the times when the most severe signs of inflammation occurred. Further research is needed to assess the efficacy of flunixin meglumine in mitigating udder pain and the accuracy of behavioral measures such as weight shifting in assessing analgesia in cows with mastitis. Evidence exists that early life experiences may influence development of characteristic feeding patterns in dairy cattle. The objective of this study was to determine the effect of milk feeding level on performance and development of feeding patterns in dairy calves. Twenty Holstein bull calves were assigned at birth to a milk replacer feeding level, fed via a teat: (1) ad libitum (ADL) or (2) restricted feeding at 5 L/d (0.75 kg of milk replacer) in 2 feedings/d (RES). All calves were offered concentrate ad libitum during the milk feeding stage. Calves were weaned gradually during wk 7 and then fed a pelleted diet ad libitum for 7 wk. Calves were weighed 2 times/wk. Feed and milk intakes were recorded daily. Feeding behavior was recorded from video for 5 d in each of wk 3, 6, 8, 11, and 14. Calves fed ADL consumed 2.6 times more milk, had greater weight gain (1.2 vs. 0.6 kg/d), and consumed less solid feed (45.2 vs. 98.0 g/d) during the milk feeding period than did calves fed RES. As expected, providing milk ADL resulted in more frequent meals (in wk 6, 7.1 vs. 2.0) and more evenly distributed diurnal patterns of feeding activity compared with the RES milk level. When considering all sucking bouts (milk feeding and non-nutritive sucking), no difference was observed between treatments in daily bout frequency or sucking time/ bout. Postweaning, calves fed RES had initially greater rate of intake (24.9 vs. 17.8 g/min) and meal size (160.8 vs. 117.2 g/meal). Diurnal feeding patterns also differed in the first week after weaning, with ADL calves feeding less continuously at the time of feed delivery. Meal characteristics and diurnal patterns of feeding activity were similar between calves by the end of the trial. However, milk feeding level had a longer-term effect on variability in feeding behavior, with calves fed RES having greater day-to-day variability in feeding time and meal size. Variability of meal characteristics also decreased over time across treatments. These results suggest that milk feeding level may have a short-term influence on postweaning feeding behavior. The objective of this cross-sectional study was to describe the housing, feeding management, and characteristics (parity and stage of lactation) of cows on commercial automatic milking system (AMS) dairies and their associations with the standing and lying behavior patterns and milking activity (frequency and yield) of lactating dairy cows. Thirteen AMS herds were en-rolled in the study, with an average herd size of 71 ± 30 (mean ± SD; range: 34 to 131) lactating cows. All of the herds used freestall barns, each set up for free cow traffic to the AMS. On-farm measurements were taken to determine stocking density at the freestalls (0.9 ± 0.1 cows/stall; mean ± SD), feed bunk (0.66 ± 0.17 m of feed bunk space/cow; mean ± SD), and AMS units (55 ± 11 cows/AMS; mean ± SD). A random sample of 30 cows/herd was selected to monitor standing and lying behavior for 4 d using electronic data loggers. Times of feed delivery and feed push-up were recorded daily by the herd managers. Milking times, frequency, and yield were automatically recorded by the AMS units. Data were analyzed in a multivariable mixed regression model to determine which herd-level (housing and feeding management) and cow-level (parity, DIM, and milk yield) factors were associated with behavior and milking activity measures. Lying bout lengths were found to be negatively associated with milk yield and tended to be positively associated with more space at the feed bunk. Increased lying duration was associated with cows of lower milk production, increased space at the feed bunk, and increased frequency of feed push-up. Longer postmilking standing durations were associated with cows of higher parity. An association existed between cows milking less frequently when they were further in lactation, were of higher parity, and as stocking density at the AMS (cows/AMS) increased. Milk yield was positively associated with increased space at the feed bunk and higher parity and negatively associated with DIM. From this study, it can be concluded that increased milking frequency may be achieved in AMS herds by reducing stocking density at the AMS unit. Further, in AMS systems, greater milk yield and lying duration may be achieved by ensuring that cows have ample feed bunk space and have their feed readily available to them in the bunk. Similar to other primate species, captive cynomolgus macaques (Macaca fascicularis) are prone to becoming overweight. The relationship between body condition and feeding behaviour in group-housed animals has not been reported. This study evaluated the effect of daily feeding routines on behaviour patterns in cynomolgus macaques to determine whether overweight macaques displayed different behaviours and activity levels. In this prospective observational study, 16 macaques (m = 4, f = 12) from four separate troops (n = 4 per troop) were selected from a colony of 165 animals. Observational data were collected over six months during morning and afternoon feedings by scan sampling. Behaviours of interest included foraging, eating, aggressive and positive social interactions, inactivity and physical activities. Multivariable mixed logistic regression modelling was used for data analysis. Results indicated that overweight animals were more likely to be inactive, dominant animals had increased probabilities of eating compared with non-dominants, and aggressive behaviours were more likely to occur in the morning and before feeding, suggesting feeding anticipation. Positive social interaction before feeding was seen and may be a strategy used to avoid aggressive encounters around food resources. Individual animal caregivers had an unintentional impact on behaviour, as decreased eating and an increase in inactivity were noted when certain individuals fed animals. These findings illustrate the complexities of feeding group-housed cynomolgus macaques to avoid overweight body condition. Clinical mastitis is a commonly occurring and economically important problem in the dairy industry. Researchers have suggested that changes in lying behavior could be useful as early indicators of cow discomfort and poor welfare. The objective of this study was to determine the associations between the onset of illness resulting from experimentally induced clinical mastitis and measures of lying behavior. Clinical mastitis was induced in 21 lactating dairy cows (parity = 2.0 ± 1.0, range = 1 to 4; days in milk = 61 ± 18) by intramammary infusion of 25 or 100 µg of Escherichia coli lipopolysaccharide (LPS) into 1 uninfected mammary quarter. Lying behavior was monitored from 2 d before through 3 d after the LPS challenge by fitting each cow with a data logger. Calculated outcome measures were total lying time, lying time on the side of the intramammary infusion, number of lying bouts, and average lying bout duration. Cows spent less time lying down on the day of the challenge compared with the 2 d before (633.3 vs. 707.0 min/d; SE = 29.6), particularly during the 4 to 7 h following LPS infusion. However, no significant relationship was found between the mammary quarter challenged and cow preference for lying side throughout the episode of induced clinical mastitis. Given that lying is a high-priority behavior in dairy cows and that increased lying time is an adaptive sickness behavior to facilitate recovery, we infer that this reduction in lying time may present a concern for cows with clinical mastitis. Although additional studies with larger numbers of animals are needed, automated monitoring of lying behavior could be an important component of the on-farm early detection of health problems, such as mastitis, in the future. The objectives were to identify risk factors for and to quantify the effect of postpartum anovulation on reproductive performance in dairy cows. Data from 2,178 Holstein cows in 6 commercial herds enrolled in a randomized clinical trial were used. Data on periparturient disease incidence, calving history, and body condition score were collected. Cows were examined at wk 5 postpartum for reproductive tract disease; cytological endometritis was defined as ≥6% polymorphonuclear cells in endometrial cytology, and purulent vaginal discharge was defined as the presence of mucopurulent or purulent vaginal discharge. Cows were followed until 300 d in milk (DIM) for reproductive performance. Serum nonesterified fatty acids (NEFA) concentration was measured once during the week before expected calving. Serum β-hydroxybutyrate, NEFA, and haptoglobin were measured at wk 1, 2, and 3 postpartum. Serum progesterone (P4) was measured at wk 3, 5, 7, and 9 postpartum. The end of the postpartum anovulation period was defined as the first sampling time at which P4 was >1 ng/mL. Statistical analyses were performed using logistic regression models and Cox proportional hazard models. The prevalence of anovulation was 72, 44, 26, and 17% at wk 3, 5, 7, and 9, respectively. Cows were classified according to their ovulatory status as having luteal function at 21 DIM (Cyc21), as having low P4 at 21 DIM but having luteal function at least once at 35 or 49 or 63 DIM (Cyc63), or being anovulatory at 63 DIM (Anov63; no samples with P4 >1 ng/ mL). Factors associated with early ovulation (Cyc21) included season, parity, decreased haptoglobinemia, and decreased serum NEFA concentration before and after parturition. Risk factors for prolonged anovulation (Anov63) included cytological endometritis, increased haptoglobinemia, and greater serum NEFA concentrations before and after parturition. Cows classified as Anov63 had an increased median time to first breeding compared with Cyc63 (74.1 vs. 73.2 d). The effect of prolonged postpartum anovulation on median time to pregnancy was conditional on parity group; a detrimental effect was present in cows of parity ≥3 (129 d for Cyc21, 151 d for Cyc63, and 180 d for Anov63), but no effect was observed in cows of parity ≥2. Overall, these findings suggest that postpartum anovulation was associated with indicators of energy balance and uterine inflammation, and with detrimental effects on reproductive performance. Choline and monensin may be supplemented during the transition period with the objectives of aiding in fat metabolism and improving energy balance, respectively. The objectives of this study were to determine the effects of supplementing rumen-protected choline (RPC) and monensin in a controlled-release capsule (CRC) on metabolism, dry matter intake, milk production, and liver function in transition dairy cattle. Three weeks before expected calving, 182 Holsteins were randomly assigned to receive one of the following: a monensin CRC, 56 g/d of RPC until 28 d in milk, CRC + RPC, or neither supplement (control). Blood samples were collected at enrollment, 1 wk before calving, and in the first and second weeks after calving. Liver biopsies were obtained from multiparous cows randomly selected from each treatment group within 24 h and again 3 wk postpartum. Daily milk production was recorded through 60 d in milk. There were no interactions of the effects of RPC and CRC on any of the outcomes measured. Overall, cows that received RPC produced 1.2 kg/d more milk in the first 60 d of lactation, but this effect was attributable to an increase in milk production of 4.4 kg/d among cows with a body condition score ≥4 at 3 wk before calving; fat cows that received RPC ate 1.1 kg of DM/d more from wk 3 before calving through wk 4 after calving. Monensin supplementation significantly increased serum concentrations of glucose and urea, lowered concentrations of β-hydroxybutyric acid and aspartate aminotransferase in the peripartum period, and increased liver glycogen content at 3 wk into lactation. The metabolic effects of CRC are consistent with previous studies, and the effects on liver are novel. Aims: To understand the potential use of bacteriophage K to treat bovine Staphylococcus aureus mastitis, we studied the role of whey proteins in the inhibition of the phage–pathogen interaction in vitro. Methods and Results: The interaction of bacteriophage K and S. aureus strain Newbould 305 was studied in raw bovine whey and serum. Incubation of S. aureus with phage in whey showed that the bacteria are more resistant to phage lysis when grown in whey and also bovine serum. Whey collected from 23 animals showed a wide variation in the level of phage-binding inhibition. The role of the protein component of milk whey in this inhibition was established; treatment of the whey by heat, proteases and ultrafiltration removed the inhibitory activity. Brief exposure of S. aureus cells to whey, followed by resuspension in broth, also reduced phage binding. Microscopy showed the adhesion of extracellular material to the S. aureus cell surface following exposure to whey. Chromatographic fractionation of the whey demonstrated that the inhibitory proteins were present in the high molecular weight fraction. Conclusions: The adsorption of whey proteins to the S. aureus cell surface appeared to inhibit phage attachment and thereby hindered lysis. The inhibitory whey proteins are of high molecular weight in their native form and may sterically block phage attachment sites on the cell surface. Significance and Impact of the Study: These findings have implications for any future use of phage therapy in the treatment of mastitis, and other diseases, caused by S. aureus. Preparturient heifers (n = 561) from 9 herds in 6 US states and 1 Canadian province were enrolled in a study to test the hypothesis that prepartum intramammary therapy would cure existing intramammary infections (IMI) and lead to increased milk production, reduced linear somatic cell count (LSCC), and improved reproductive performance. Mammary secretions were collected 10 to 21 d before expected calving from each quarter. Heifers were then assigned by identification number to receive intramammary therapy consisting of infusion of one tube per mammary quarter of a lactating cow commercial antibiotic preparation containing cephapirin or to a nontreated control group. Overall, 34.1% of mammary quarters were infected with a mastitis pathogen before parturition and 63.4% of heifers had at least one mammary quarter infected. The coagulase-negative staphylococci (CNS) caused the majority (74.8%) of prepartum IMI. Coagulase-positive staphylococci, environmental streptococci, and coliforms accounted for 24.5% of prepartum infections. Treatment had a significant effect on the cure rate of infected mammary quarters. Mammary quarters that were infected prepartum and treated with antibiotics had a 59.5% efficacy of cure rate and the percentage reduction in heifers with IMI was 51.9. Control quarters had a spontaneous cure rate of 31.7%. Treatment did not significantly affect milk production or LSCC in the first 200 d of lactation; however, there was a significant treatment by herd interaction for milk production. Quarters cured of either CNS or major pathogens had a lower LSCC in the first 200 d of lactation. No significant effect on services per conception or days open between treatment and control groups was observed. This trial demonstrated that prepartum intramammary antibiotic therapy did reduce the number of heifer IMI postpartum. Milk production, LSCC, and reproductive performance during the first 200 d of the first lactation were not significantly affected by treatment. Given these results, use of prepartum intramammary antibiotic therapy in heifers as a universal strategy to increase milk production in first-lactation dairy cows may not be warranted. The objective of this study was to determine effects of monensin on ovarian follicular development and reproductive performance in postpartum dairy cows. Forty-eight multiparous Holstein cows were randomly assigned to receive either a control total mixed ration (n = 24) or the same diet plus 22 mg of monensin/kg (n = 24) from 21 d before anticipated calving until cows were either confirmed pregnant or were >180 d postpartum. Monensin had no effect on development of the first dominant follicle postpartum or the numbers of class 1 (3 to 5 mm), 2 (6 to 9 mm), or 3 (10 to 15 mm) follicles. Control cows had more class 4 (>15 mm) follicles at 10 to 13 d postpartum than cows in the monensin group. The first dominant follicle postpartum ovulated, regressed, or became cystic unrelated to differences between diets. However, the first ovulation postpartum occurred earlier in monensin-fed cows than in the control group (27.2 ± 2.1 d vs. 32.4 ± 1.5 d), with no dietary effects on the diameter of the ovulating follicle. Similarly, treatments did not differ in the proportion of cows with 2 or 3 waves of ovarian follicular development per cycle, nor in the number of follicles of all classes during the breeding period. Times of ovulation following treatment with prostaglandin F[sub2α] were not different between dietary groups. Pregnancy rates after timed artificial insemination were similar between diets. We report the performance of twenty-four high temperature superconducting dc superconducting quantum interference devices (SQUIDs) whose Josephson junctions were formed from a YBa2Cu3O7-x (YBCO) grain boundary on an engineered MgO step-edge substrate. Each device has the same SQUID design with an inductance of 65 pH, and consisted of two SQUIDs with different junction widths, connected in series, and directly coupled to a large pick-up loop of 7.5 mm × 8.8 mm on a 1 × 1 cm2 chip. 58% of the devices had a peak-to-peak modulation voltage, ΔV, larger than 30 μV. The typical values of the white flux and field noise were 4.7 μ[uppercase_phi_synonym]o and 56 fT/[formula], respectively, in a magnetically shielded environment with 1/f corner frequency at 15-20 Hz. Mastitis is a common and costly production disease on dairy farms. In Canada, the incidence rate of clinical mastitis (IRCM) has been determined for conventionally managed dairy farms; however, no studies to date have assessed rates in organically managed systems. The objectives of this observational study were (1) to determine the producer-reported IRCM and predominant pathogen types on conventional and organic dairy farms in Southern Ontario, Canada, and (2) to evaluate the association of both mean overall IRCM and pathogen-specific IRCM with management system, housing type, and pasture access. Data from 59 dairy farms in Southern Ontario, Canada, distributed across conventional (n = 41) and organic management (n = 18) systems, were collected from April 2011 to May 2012. In addition to management system, farms were categorized by housing method (loose or tie-stall) and pasture access for lactating cows. Participating producers identified and collected samples from 936 cases of clinical mastitis. The most frequently isolated mastitis pathogens were coagulase-negative staphylococci, Bacillus spp., Streptococcus spp., Staphylococcus aureus, and Escherichia coli. The IRCM was higher on conventional farms than organic (23.7 vs. 13.2 cases per 100 cow-years) and was not associated with housing type (loose or tie-stall), pasture access, or herd-average milk yield. Bulk tank somatic cell count tended to be lower on conventional farms than organic (222,000 vs. 272,000 cells/mL). Pathogen-specific IRCM attributed to Staph. aureus, Bacillus spp., and E. coli was greater on conventional than organic farms, but was not associated with housing or any other factors. The objective of this research was to investigate factors associated with serum haptoglobin (Hp) levels in newborn calves. In addition, the associations between serum Hp levels in newborn calves with growth, morbidity, and mortality in calves <4 mo of age were investigated. A total of 1,365 Holstein heifer calves from 15 dairy farms were enrolled in this study from January to December, 2008. Following calving, a birth record was completed, including information on the calving event, colostrum administration, and other details. During weekly farm visits, each calf was assessed at 1 to 8 d, 5 to 21 d, 36 to 42 d, and 90 to 120 d of age. At these sampling times, each calf was assessed using a standardized clinical score for general health, and height and weight were measured. At 1 to 8 d of age, a blood sample was collected to measure serum total protein and Hp concentrations. Treatment events and death loss were recorded throughout the study by the farm staff. Serum Hp concentration in the first week of life was not significantly associated with the degree of calving difficulty. However, serum Hp was higher in calves with a higher rectal temperature and depressed attitude at the first sampling time. Furthermore, the association between serum Hp and the severity of nasal discharge varied by age at first sampling time. Calves with higher Hp in their first week of life had significantly higher total health scores throughout the entire sampling period. Haptoglobin was not significantly associated with average daily gain or treatment for bovine respiratory disease. Yet, for every 1 g/L increase in serum Hp in the first week of life, the odds of being treated for any ther disease during the study period increased by 7.6 times. Treatment for bovine respiratory disease, diarrhea, or any other disease resulted in increased odds of calf mortality. In addition, Hp concentration in the first week of life was associated with mortality in calves <4 mo of age. The optimal cut point for Hp was determined to be 0.13 g/L for the prediction of disease and death, although the sensitivity of Hp concentration alone as a diagnostic test for individual calves was low. The objective of this study was to evaluate the utility of a digital Brix refractometer for the assessment of success of passive transfer of maternal immunoglobulin compared with the measurement of serum total protein (STP) by refractometry. Blood samples (n = 400) were collected from calves at 3 to 6 d of age. Serum IgG concentration was determined by radial immunodiffusion (RID), and STP and percentage Brix (%Brix) were determined using a digital refractometer. The mean IgG concentration was 24.1 g/L [standard deviation (SD) ± 10.0] with a range from 2.1 to 59.1 g/L. The mean STP concentration was 6.0 g/dL (SD ± 0.8) with a range from 4.4 to 8.8 g/dL. The mean %Brix concentration was 9.2% (SD ± 0.9) with a range of 7.3 to 12.4%. Brix percentage was highly correlated with IgG (r = 0.93). Test characteristics were calculated to assess failure of passive transfer (FPT; serum IgG <10 g/L). The sensitivity and specificity of STP at 5.5 g/dL were 76.3 and 94.4%, respectively. A receiver operating characteristic curve was created to plot the true positive rate against the false positive rate for consecutive %Brix values. The optimal combination of sensitivity (88.9%) and specificity (88.9%) was at 8.4% Brix. Serum total protein was also positively correlated with %Brix (r = 1.00) and IgG (r = 0.93). Dairy producers can successfully monitor their colostrum management and the overall success of passive transfer using a digital Brix refractometer to estimate IgG concentration of colostrum and calf serum. The objectives of this study were to (1) evaluate the use of a pressure algometer and an automated rumination monitoring system to assess changes in pain sensitivity and rumination time in response to endotoxin-induced clinical mastitis and (2) evaluate the effect of the nonsteroidal antiinflammatory drug meloxicam on pain sensitivity and rumination time, as well as other clinical signs, in dairy cattle with endotoxin-induced clinical mastitis. Clinical mastitis was induced in 12 primiparous and 12 multiparous lactating dairy cows by intramammary infusion of 25 µg of Escherichia coli lipopolysaccharide (LPS) into 1 uninfected quarter. Immediately after, half the cows were injected subcutaneously with meloxicam (treated group) and half with the same volume of a placebo solution (control group). Pain sensitivity was assessed by measuring the difference in pressure required to elicit a response on the control and challenged quarter using an algometer 3 d before, immediately before, and at 3, 6, 12, and 24 h after LPS infusion and either meloxicam or placebo injection. Rumination was continuously monitored from 2 d before to 3 d after LPS infusion using rumination loggers. Udder edema, body temperature, somatic cell score, and dry matter intake were also monitored to evaluate the occurrence and the duration of the inflammation after LPS infusion. In control animals, the difference in the pressure applied to the control and challenged quarters (control -- challenged quarter) increased by 1.1 ± 0.4 kg of force 6 h after LPS infusion compared with the baseline, suggesting an increase in pain sensitivity in the challenged quarter. Neither the LPS infusion nor the meloxicam treatment had an effect on daily rumination time. However, the rumination diurnal pattern on the day of LPS infusion showed an overall deviation from the baseline pattern. Cows spent less time ruminating in the hours following LPS infusion and more time ruminating later in the day. Meloxicam did not alter somatic cell score or dry matter intake. However, meloxicam-treated animals had less udder edema and a lower body temperature in the hours following LPS infusion compared with control animals. In conclusion, pressure algometers and rumination loggers show promise as tools to detect mastitis and monitor recovery on farm. The objective of the present study was to investigate health and survival in heifer calves, from 16 farms in southwestern Ontario. Research technicians visited the farms on a weekly schedule for 16 mo, starting in January 2008, and collected blood samples and various measurements to determine the overall health status of the calves. Data of 1,588 Holstein heifer calves from birth until 90 to 120 d of age were analyzed. Initially, a survival analysis was carried out to determine which factors surrounding birth increased a calf's risk of mortality. The factors examined were type of birth, calving ease, season of birth, serum total protein, weight during first week of life, treatment status during first week of life, total number of treatments, and bovine respiratory disease status. In general, calves born in fall (October-December), low serum total protein, low weight, and calves treated for illness during the first week of life, increasing number of treatments, and being treated for bovine respiratory disease all increased a calf's risk of mortality. Genetic parameters for requiring treatment for illness, survival, bovine respiratory disease, other diseases, height, and weight were estimated. Heritabilities for treatment, survival, and diseases were low, ranging from 0.04 to 0.07, and were not significant. Moderate and significant heritabilities were found for height (0.33 to 0.45) and weight (0.24 to 0.44). Treatment and survival to 90 to 120 d of age were favorably correlated, indicating that calves requiring less treatment have a higher survival rate and vice versa. Further, body weight was found to have a tendency of favorable genetic correlation (-0.22 to -0.53) with treatment for diseases, indicating that calves with higher body weight in their first month of life were genetically more resistant to diseases than those with average body weight. Overall, our results show the potential of using data on calfhood disease and survival in future dairy cattle breeding programs. Poor dairy cow hygiene has been consistently associated with elevated somatic cell count (SCC) and the risk of subclinical mastitis. The objective of this study was to determine the associations between dairy cow standing and lying behavior, barn hygiene, cow hygiene, and the risk of experiencing elevated SCC. Lactating Holstein dairy cows (n = 69; 86 ± 51 DIM; parity: 2.0 ± 1.2; means ± SD), kept in 1 of 2 groups, were monitored over a 4-mo period. Each group contained 61 ± 1 (mean ± SD) cows over the study period; complete data were obtained from 37 and 32 animals within each respective group. Cows were housed in a sand-bedded, freestall barn with 2 symmetrical pens, each with a free cow traffic automatic milking system. To vary barn hygiene, in 4 consecutive 28-d periods, alley manure scrapers in each of the 2 pens were randomly assigned to frequencies of operation of 3, 6, 12, and 24 times per day. During the last 7 d of each period, cow hygiene (upper leg/flank, lower legs, and udder; scale of 1 = very clean to 4 = very dirty) and stall hygiene (number of 0.15 × 0.15-m squares contaminated with manure in a 1.20 × 1.65-m grid) were recorded. Standing and lying behavior of the cows were collected during those days using data loggers. Individual-cow SCC was recorded at the beginning and end of each 28-d period. Elevated SCC was used as an indicator of subclinical mastitis; incidence of elevated SCC was defined as having a SCC >200,000 cells/mL at the end of each 28-d period, when SCC was <100,000 cells/mL at the beginning of the period. Less frequent scraping of the barn alleys was associated with cows having poorer hygiene. Poor udder hygiene was associated with poor stall hygiene. Longer lying duration was associated with poor hygiene of the upper legs/flank and udder. Greater premilking standing duration was associated with poor udder hygiene and decreased frequency of lying bouts was associated with poor hygiene of the lower legs. Higher milk yield was associated with poor hygiene of the udder and lower legs; multiparous cows had poorer hygiene of the upper legs/flank and udder. Over the study period, 24 new cases of elevated SCC were detected. No associations existed for the risk of experiencing an elevated SCC with alley scraping frequency or cow behavior patterns. However, increased odds of occurrence of elevated SCC were noted for cows of lower milk yield as well as for multiparous cows. In summary, these results show that cow hygiene is affected by the standing and lying behavior of cows and by the cleanliness of the cow's environment. These findings emphasize the need for cows to be provided clean standing and lying environments. The results also show that frequent cleaning of barn alley floors will help improve cow hygiene. The objective was to identify herd-level indicators expressed as a proportion of sampled animals with increased nonesterified fatty acids (NEFA) or β -hydroxybutyric acid (BHBA), or decreased calcium in wk -1 and wk +1 relative to calving that were associated with herd-level incidence of retained placenta, metritis and displaced abomasum, milk production, and probability of pregnancy at the first artificial insemination (AI). Fifty-five Holstein freestall dairy herds in the United States and Canada were visited weekly. Blood was collected from 2,365 cows around parturition, and serum concentrations of NEFA, BHBA, and calcium were determined. Different cow-level metabolite thresholds associated with detrimental health or productivity in previous studies were used to classify animals into high- and low-risk metabolite concentration groups. For wk -1 and wk +1 relative to calving, a herd-level threshold was determined as the proportion of sampled animals in the high-risk metabolite concentration groups with the strongest association with increased incidence of disease, milk loss, or decreased pregnancy at the first AI. The odds of displaced abomasum after calving were higher in herds that had ≥ 25% of the animals with BHBA ≥ 1,400 μ mol/L in wk +1 [odds ratio (OR) = 2.1; 95% confidence interval (CI) = 1.0-4.2)] or ≥ 35% of the animals with calcium ≤ 2.1 mmol/L in wk +1 (OR = 2.4; CI = 1.3-4.3). Herd-level thresholds of ≥ 15% of the cows with BHBA ≥ 800 μ mol/L in wk -1 and ≥ 15% of the cows with calcium ≤ 2.1 mmol/L in wk +1 were associated with milk loss (±SE) of 4.4 ± 1.7 and 3.8 ± 1.4 kg/d per cow, respectively. When only multiparous cows were considered, herds with ≥ 30% of the multiparous cows with NEFA ≥ 0.5 mEq/L in wk -1 were associated with a 3.0 ± 1.5 kg/d per cow milk loss. The odds of pregnancy at first AI were lower in herds that had ≥ 5% of the cows with calcium ≤ 2.1 mmol/L in wk -1 (OR = 0.7; CI = 0.5-1.0), or ≥ 30% of the cows with NEFA ≥ 1.0 mEq/L (OR = 0.6; CI = 0.4-0.9) or ≥ 25% of the cows with calcium ≤ 2.1 mmol/L in wk +1 (OR = 0.7; CI = 0.5-0.9). When only multiparous cows were considered, the odds of pregnancy at first AI were lower in herds that had ≥ 50% of multiparous cows with NEFA ≥ 0.5 mEq/L in wk -1 (OR = 0.5; CI = 0.2-0.9). In conclusion, several herd-level thresholds for the proportion of cows with increased NEFA or BHBA, or decreased calcium in the week before and after calving were associated with higher risk of displaced abomasum, milk loss at the first Dairy Herd Improvement Association test, and decreased pregnancy at first AI. The association found between precalving BHBA and milk production is promising due to the availability of several cow-side tests for measuring BHBA. Some of the herd-level associations differed from the previously described cow-level associations, suggesting the potential of interpreting periparturient metabolic challenges at the herd level, where changes in diet and management are generally implemented. Prototheca spp. are algae that cause incurable acute or chronic mastitis in dairy cows. The aim of this casecontrol study was the identification of cow- and herdlevel risk factors for this unusual mastitis pathogen. Aseptically collected composite milk samples from 2,428 milking cows in 23 case and 23 control herds were collected between January and May 2011. A questionnaire was administered to the producers, and cow-level production and demographic data were gathered. In 58 of 64 isolates, Prototheca spp. and Prototheca zopfii genotypes were differentiated using PCR and matrixassisted laser desorption/ionization time-of-flight mass spectrometry. All isolates were identified as Prototheca zopfii genotype 2. The mean within-herd prevalence for Prototheca spp. was 5.1% (range 0.0-12.5%). Case herds had a significantly lower herd-level prevalence of Staphylococcus aureus and a higher prevalence of yeasts than did control herds. The final logistic regression model for herd-level risk factors included use of intramammary injections of a non-intramammary drug [odds ratio (OR) = 136.8], the number of different injectable antibiotic products being used (OR = 2.82), the use of any dry cow teat sealant (external OR = 80.0; internal OR = 34.2), and having treated 3 or more displaced abomasums in the last 12 mo OR = 44.7). The final logistic regression model for cow-level risk factors included second or greater lactation (OR = 4.40) and the logarithm of the lactation-average somatic cell count (OR = 2.99). Bovine respiratory disease complex (BRD) is a common disease in weaned dairy calves that incurs economic and welfare costs. This study was an extension of a randomized clinical trial in which a single injection of tulathromycin (TUL) or oxytetracycline (TET) was administered at first movement to group housing for the prevention of BRD in the 60 d following antimicrobial treatment (BRD60). Calves treated with TUL were 0.5 times [95% confidence interval (CI): 0.4 to 0.7] as likely to be treated for BRD60 as calves treated with TET. The objectives of the current study were to evaluate the long-term effects of BRD and antibiotic treatment on growth of heifers until breeding age, age at first calving, incidence of dystocia, milk production, and mortality before first calving and mortality before 120 d in milk. At entry to the breeding barn (382 d of age), calves that experienced BRD60 weighed 16.0 ± 2.3 kg less than calves that did not. Survival to first calving was recorded for 98% (1,343/1,392) of the heifers on this trial. For TET and TUL heifers with BRD60, 63% (94/150) and 73% (64/88) survived to first lactation, respectively. For TET and TUL calves without BRD60, 84% (436/517) and 84% (494/588) survived to first lactation, respectively. The median age at first calving for heifers with and without BRD60 was 714 (95% CI: 705-723) and 702 (95% CI: 699-705) days, respectively. Heifers with BRD60 were 1.5 (95% CI: 1.1-2.2) times more likely to have a calving ease score ≥2 at their first calving compared with heifers without BRD60. The objective was to examine the associations of peripartum concentrations of nonesterified fatty acids (NEFA), β-hydroxybutyrate (BHBA), and calcium with milk production in early lactation and pregnancy at the first artificial insemination (AI) across different management systems. Fifty-five Holstein freestall dairy herds located across the United States and Canada were visited weekly for blood sample collection from 2,365 cows. For each week of sampling (from wk .1 through wk 3 relative to calving) and for each metabolite, serum concentrations were dichotomized at various thresholds to identify the thresholds with the best negative associations with milk production and pregnancy at first AI. These thresholds were used to categorize the serum concentrations into higher and lower risk categories. Repeated-measures ANOVA and multivariable logistic regression were conducted for milk production and pregnancy at the first AI data, respectively, considering cow as the experimental unit and herd as a random effect. In the week before calving, serum NEFA .0.5 mEq/L, BHBA ≥600 µmol/L, and calcium ≤2.1 mmol/L were associated with 1.6 to 3.2 kg/d milk loss across the first 4 Dairy Herd Improvement Association (DHIA) milk tests. High levels of NEFA and BHBA in wk 1 and 2 after calving (≥0.7 and ≥1.0 mEq/L for NEFA, and ≥1,400 and ≥1,200 µmol/L for BHBA), and low levels of calcium (≤2.1 mmol/L) in wk 1, 2 and 3 after calving were associated with milk loss at the first DHIA milk test. Serum concentrations of NEFA and BHBA were not associated with pregnancy at first AI in any sampling week, whereas calcium <2.2 to 2.4 mmol/L from wk 1 through wk 3 postpartum were associated with reduced pregnancy at first AI. In conclusion, high serum concentrations of NEFA, BHBA, and low concentrations of calcium around parturition were associated with early lactation milk loss, and low calcium concentration around parturition was associated with impaired early lactation reproduction. The objectives of this study were to estimate the genetic parameters of preweaning undifferentiated bovine respiratory disease (BRD), umbilical diseases (UMB), and bloat (BLT) for a population of Holstein calves from New York State, as well as to associate the estimated breeding values determined in the current study with traits from ongoing genetic evaluations used in Canada and the United States. Data were recorded for 7,372 heifer calves at a commercial rearing facility in New York State, from arrival at 1 to 7 d of age for the duration of stay at the facility. Performance and disease up to weaning and mortality before and after weaning were recorded. The 3 traits of interest, BRD, UMB, and BLT, were scored as 0 or 1 and analyzed using a multivariate linear sire model. The model included fixed effects of arrival weight, serum total protein, weaning weight, and season and year of birth; herd and sire were included as random effects. The heritabilities of the 3 health traits of interest were estimated at 0.09 for BRD, 0.14 for UMB, and 0.04 for BLT. The genetic correlation between the calf health traits BRD and BLT was 0.62. Correlations between BRD and UMB and between BLT and UMB were close to zero. Breeding values were estimated for the 3 calf health traits and correlated with routinely evaluated traits from Canadian and US genetic evaluations (correlations ranged from -0.42 to 0.32).
