Leptospirosis is an important worldwide zoonosis, and it has also been reported in Slovenia. The cultivation of Leptospira from human material is difficult. Despite that, we successfully isolated 12 human Leptospira strains isolated from patients between 2002 and 2020 and used various methods for the phenotypic and genotypic characterization of the strains, including matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF) using our own MALDI-TOF data library, melting temperature analysis of the amplified lfb1 gene, determination of Leptospira serogroups using rabbit immune sera, NotI-RFLP of the whole Leptospira genome, multilocus sequence typing (MLST) of seven housekeeping genes, and whole-genome sequencing (WGS)-based typing. We confirmed the presence of four pathogenic Leptospira species (L. kirschneri, L. interrogans, L. borgpetersenii, and L. santarosai) and three serogroups: Grippotyphosa, Icterohaemorrhagiae, and Sejroe. MALDI-TOF identified three of seven isolates at the species level and four isolates at the genus level. Serovars of 8 of the 10 strains were determined using NotI-RFLP. MLST showed that the clinical isolates belonged to sequence types ST17, ST110, and ST155. WGS confirmed the analysis of Leptospira strains using conventional methods. Animal hoarding disorder, especially dogs and cats, is a complex issue that occurs in almost all communities, and it is necessary to collect data that contribute to the understanding of leptospirosis within the One Health initiative. In order to determine the seroprevalence of Leptospira sp., 71 dogs and 39 cats from 33 people with animal hoarding profile in the municipality of Patos, state of Paraíba, semi-arid region of Northeastern Brazil were evaluated by using the microscopic serum agglutination test (MAT) as diagnostic test. Thirty-eight animals (34.5%; 95% CI = 25.7 – 43.4%) were seroreactive, with 46.5% in dogs and 12.8% in cats (P < 0.01), with the highest frequency of the Icterohaemorrhagiae serogroup in both species. The high seroprevalence reported suggests the need for public policies for this vulnerable population in order to prevent the transmission of zoonoses. [ABSTRACT FROM AUTHOR]

Background: Leptospirosis is a health problem that causes death in Indonesia. In 2017, Boyolali District was reported that the number of leptospirosis cases reached 40.62 per 100,000 population with a CFR of 23.52%. The determination of risk factors and Leptospira bacteria's presence in the body of water plays an essential role in the transmission of leptospirosis. Design and methods: This study aims to determine the risk factors and Leptospira bacteria's presence in water bodies in Boyolali District. This research is descriptive research with a survey method using a cross-sectional design and an analytical study using an observational method with a case-control approach. The sample was 100 water samples from wells, rivers, and paddy fields in endemic and non-endemic areas of leptospirosis. This study's population was 34 people with leptospirosis in Boyolali Regency in January 2017 - August 2018. Results: There was a significant relationship between leptospirosis incidence in the Boyolali Regency with garbage, the pet presence, a history of injuries, and field activity. Leptospira bacteria are found in rivers (18.18%) and rice fields (6.67%), while in sub-districts with cases occur almost every year. Leptospira are found in wells (18.18%) and rice fields (6.67%). The most common presentation of animal leptospirosis is the subclinical and silent chronic form, that can lead to important reproductive disorders. The diagnosis of this chronic form remains a challenge. The aim of the present study is to gather and critically analyse the current information about molecular tools applied to animal leptospirosis diagnosis, particularly the silent chronic presentation of the infection. Regarding clinical specimens, samples from urinary tract were the most used (69/102, 67·7%), while few studies (12/102, 11·8%) investigated samples from reproductive tract. Concerning the molecular methods applied, the most used is still the conventional polymerase chain reaction (PCR) (46/102, 45%), followed by real‐time PCR (38/102, 37·2%). The lipL32 gene is currently the most common target used for Leptospira detection, with 48% of studies applying this genetic marker. From all the studies, only few (21/102, 20·5%) performed gene sequencing. According to the majority of authors, current evidence suggests that lipL32‐PCR is useful for an initial screening for Leptospira DNA detection in animal clinical samples. Posteriorly, if DNA sequencing could be performed on positive lipL32‐PCR samples, we encourage the use of secY gene as a genetic marker. The molecular methods appear as the most important tools for the diagnosis of the chronic silent leptospirosis on domestic animals, reinforcing its evident impact not only on animal reproduction but also on a One Health context. The Fiocruz Mata Atlântica Campus (CFMA) represents one of the main urban growth fronts in Rio de Janeiro city. The present study evaluates the environment associated with the seroprevalence of canine visceral leishmaniasis (CVL), toxoplasmosis and leptospirosis in dogs and of toxoplasmosis in cats and production poultry belonging to the residents of the CFMA. Blood samples obtained by the census were used to assess frequency of positivity through serological assays. The proportion of seroprevalence of toxoplasmosis, leptospirosis and CVL were respectively 37.2%, 6.2% and 1.2%. The association with the environment had different results depending on the infection. Seroprevalence of leptospirosis was significantly lower (P=0.03) in dogs from anthropized areas (3%), but this was the only region with cases of leishmaniasis. For toxoplasmosis, there was a higher seroprevalence (50%) in rural scenarios. In cats, the seroprevalence of toxoplasmosis was 32.1% and in poultry, it was 18.1%. The seroprevalence in these animals were similar in the studied environments. The results demonstrate the occurrence of Leishmania sp., Toxoplasma gondii and Leptospira spp. in dogs, cats, hens and ducks living in anthropic environments of the Atlantic Forest, in Rio de Janeiro city, being possible to correlate this occurrence between these domestic animal species and the living environment. Leptospirosis is a health problem in Indonesia. In the year 2016 cases of leptospirosis in Klaten reported death rate 15.38%. The presence of Leptospira bacteria in the body of water plays an important role in the transmission of leptospirosis. This study aims to determine the presence of Leptospira bacteria in water bodies in Klaten Regency. This research is descriptive research with survey method using cross sectional design. The sample was 100 water samples from wells, rivers and paddy fields in endemic and non endemic areas of Leptospirosis. Polymerase Chain Reaction examination there were 6 positive water samples of rpoB leptospira gene on well water (6.7%), irrigation channel (6.7%) and paddy field (5%). Leptospira is found at temperatures ranging from 24-31º C, pH 6.8-7.5 and dissolved oxygen 4-12 ppm. [ABSTRACT FROM AUTHOR]

The aim of the present study was to describe the strategies of the control of an outbreak of leptospiral infection in dairy cattle in Maranhão State, Northeastern Brazil. In the period from January to July 2015, 18 (17%) out of 106 cows presented abortion, six (5.7%) stillbirth, and 12 (11.3%) repeated estrus, totaling 24 animals with reproductive problems. The diagnosis of leptospirosis was based on serology (microscopic agglutination test—MAT), bacteriological culture, and polymerase chain reaction (PCR). Antibiotic therapy, vaccination protocols, and changes in management practices were suggested as control measures. Of all animals on the farm (n = 280), 136 (48.6%) were seropositive for at least one serovar of Leptospira sp. No pure leptospiral culture was obtained. Eight of the animals with reproductive problems yielded positive PCR results (vaginal fluid of seven animals and urine and vaginal fluid of one animal). Genetic sequencing of a vaginal fluid/urine PCR-positive sample revealed Leptospira borgpetersenii. One year after the adoption of control measures, no reproductive problems were observed. Thus, leptospirosis probably caused the reproductive failures in the herd, and the control and prevention measures implemented were efficient in controlling the disease. [ABSTRACT FROM AUTHOR]

Foi realizada uma análise espacial da ocorrência de leptospirose humana e canina na Supervisão de Vigilância em Saúde do Butantã, situada no município de São Paulo, no ano de 2007, associada a variáveis ambientais de risco, tais como: focos de enchente e áreas de desratização. Foram encontrados aglomerados espaciais de pontos de alagamentos em 12 setores censitários e de casos de leptospirose humana em quatro setores censitários, sem correlação entre ambos. Não foram encontrados agrupamentos de casos em cães, possivelmente devido à subnotificação. As proporções casos humanos de leptospirose : população humana dentro e fora da área de desratização foram 7:199.600 e 9:257.980, respectivamente. Conclui-se que medidas de controle de roedores como a desratização foram responsáveis pela minimização dos efeitos dos fatores de risco para a transmissão de leptospirose para humanos.

Over the last decade, an escalating number of reported cases of leptospirosis in Malaysia has resulted in a significant number of deaths. Leptospirosis is caused by Leptospira sp., which is disseminated by soil and water. Due to the absolute dependence of microbes on this biosynthesis pathway coupled with its non-existence in humans, the structural analysis of riboflavin synthase as a potential target for the development of antimicrobial agents is very significant. Riboflavin synthase (E.C 2.5.1.9) is an important enzyme that catalyzes the last stage of riboflavin biosynthesis. Riboflavin synthase from the locally isolated pathogenic bacterium Leptospira kmetyi was cloned and expressed in Escherichia coli. The protein was purified and crystallized for X-ray analysis. The crystal structure of riboflavin synthase from L. kmetyi has been determined at 3.2 Å and belonged to the orthorhombic space group P212121, with an R-factor of 21.8%. The calculated Matthews coefficient (VM) was 2.94 Å3Da−1 with a solvent content of 58.24 %. There are three molecules present in the asymmetric unit. The structure was analyzed with potential inhibitors via molecular docking and molecular dynamics simulation. The complex with compound 1 had the best possible interaction and stability for a 100 ns simulation. The existence of this crystal structure enables its structural properties to be unveiled and facilitates the application of structure-based drug discovery approach. This study sheds light on the development of new potential antibacterial drugs that may be beneficial for the treatment of leptospirosis or other infectious diseases associated with it. Background Leptospirosis, a spirochaetal zoonotic disease of worldwide distribution, endemic in Europe, has been recognized as an important emerging infectious disease, though yet it is mostly a neglected disease which imparts its greatest burden on impoverished populations from developing countries. Leptospirosis is caused by the infection with any of the more than 230 serovars of pathogenic Leptospira sp. In this study we aimed to implement the MALDI-TOF mass spectrometry (MS) database currently available in our laboratory with Leptospira reference pathogenic (L. interrogans, L. borgpetersenii, L. kirschneri, L. noguchii), intermediate (L. fainei) and saprophytic (L. biflexa) strains of our collection in order to evaluate its possible application to the diagnosis of leptospirosis and to the typing of strains. This was done with the goal of understanding whether this methodology could be used as a tool for the identification of Leptospira strains, not only at species level for diagnostic purposes, but also at serovar level for epidemiological purposes, overcoming the limits of serological and molecular conventional methods. Twenty Leptospira reference strains were analysed by MALDI-TOF MS. Statistical analysis of the protein spectra was performed by ClinProTools software. Results The spectra obtained by the analysis of the reference strains tested were grouped into 6 main classes corresponding to the species analysed, highlighting species-specific protein profiles. Moreover, the statistical analysis of the spectra identified discriminatory peaks to recognize Leptospira strains also at serovar level extending previously published data. Conclusions In conclusion, we confirmed that MALDI-TOF MS could be a powerful tool for research and diagnostic in the field of leptospirosis with broad applications ranging from the detection and identification of pathogenic leptospires for diagnostic purposes to the typing of pathogenic and non-pathogenic leptospires for epidemiological purposes in order to enrich our knowledge about the epidemiology of the infection in different areas and generate control strategies. In order to determine and compare the prevalence of antibodies to Brucella abortus, Leptospira sp. and Neospora caninum, two cattle, two buffaloes and three mixed (both species) farms, were researched, besides including three buffaloes sale facilities. Antibody prevalence was higher in buffaloes than in cattle for the three studied agents, Leptospira sp. (37,3% vs 28%), Neospora caninum (45.4% vs. 12.5%) and Brucella abortus (11,9% vs. 5,3%). Antibodies were present at six serovars in both, cattle and water buffaloes and seroprevalence values for Hardjo and Grippotyphosa serovars were superior in both species. The prevalence of antibodies to leptospirosis was higher in simple premises in both cattle and buffaloes, and the seroprevalence to brucellosis in cattle of mixed farms was higher when compared with that obtained in simple farms with a prevalence ratio (PR) of 6,8; (CI, 1,58 to 29,6), whereas in buffaloes, the prevalence of antibody was higher in those single (OR, 2,8, CI = 1,18 to 5,71 ). In cattle, there was not an association between the seroprevalence to Neospora caninum and the herd type, but in buffaloes seroprevalence was higher in simple farms with a RP of 2,3 (CI= 1,75 to 3,01). It is concluded that buffaloes and cattle have a high prevalence of antibodies to Neospora caninum and Leptospira sp. in all herds and sale facilities and there are still herds affected by Brucella abortus in spite of having the control programs established. Abstract: We evaluated the seroprevalence of 10 zoonotic agents among the general population (15 years old and over) of Eastmain and Wemindji, James Bay, Quebec, in 2007. Overall seroprevalence rates were similar between the 2 communities. Nearly half the individuals tested (n = 251; 146 women, 105 men) were seropositive (n = 115) for at least one zoonosis. The highest seroprevalence rates were for Leptospira sp. (23%), Francisella tularensis (17%), and the California serogroup viruses (JC and SSH viruses) (10%). The other zoonoses (Toxoplasma gondii, Coxiella burnetii, Echinococcus granulosus, Toxocara canis, and Trichinella sp.) had seroprevalence rates ≤5%; no exposures were identified to hantaviruses (Sin Nombre virus). Overall, seropositivity was related to age, gender, hunting, and owning a dog. There was no medical history suggestive of overt diseases. Nonetheless, physicians should consider these agents when confronted with difficult or confusing diagnoses. Abstract: Neosporosis is an infection of animals caused by an intracellular coccidian parasite, Neospora caninum, closely related to Toxoplasma gondii. The parasite is one of important abortifacient agents of bovine abortions worldwide. The aim of the study was to detect the prevalence of anti-Neospora antibodies in dairy aborting sheep from two eastern Slovak regions and to compare it with the occurrence of other potential abortifacient agents. Sera of 382 sheep, mainly the Improved Valachian and Merino breed, were tested for the presence of anti-Neospora and anti-Toxoplasma antibodies by ELISA, anti-Leptospira sp. by micro-agglutination-assay and anti-Chlamydophila antibodies using the complement fixation test. The mean seroprevalence of N. caninum was 3.7% and of T. gondii, 24.3%. This phenomenon of higher susceptibility of sheep to T. gondii is in the opposite of N. caninum infection in cattle. Anti-Leptospira antibodies were observed in 2.9% of serum samples with titres from 800 to 1600, whereas IgG antibodies against Chlamydophila abortus were found in 13.6% with titres from 64 to 1024. Half of N. caninum positive sera were simultaneously positive for T. gondii and one sample for C. abortus. From examined abortifacient agents the most important, from the frequency point of view, were toxoplasmosis (24.3%) and chlamydiosis (13.6%). No significant association between the frequencies of the abortions and mean seroprevalence of the abortifacient agents in Košice region was determined. Likewise, no significant differences between the mean seroprevalence of neosporosis and toxoplasmosis in the two regions were detected. The first survey of neosporosis in aborting sheep from eastern Slovakia revealed a low prevalence resulting in a lower impact on reproduction losses in these small ruminants. One kidney was collected from each of 32 fattening pigs at an abattoir in southern Vietnam in 2001 in order to demonstrate infecting Leptospira serovar and to associate renal macro- and microscopic findings with the presence of renal leptospires. Leptospires were demonstrated in 22 (69%) of the investigated kidneys by immunofluorescence. Multifocal interstitial nephritis (MFIN) and gross renal lesions (white spots) were each demonstrated in 24 (75%) kidneys. Leptospira interrogans serovar bratislava was isolated from one kidney. There was no association between presence of leptospires and MFIN (P=0.19), respectively and white spots (P=0.98), respectively. These data suggest that Leptospira infection is common among fattening pigs in the study area and that these animals may be considered as an occupational human health hazard. It is also suggested that the presence of white spots is an unreliable indicator of the presence of renal leptospires. [Copyright &y& Elsevier]

The article discusses a study which evaluated the prevalence of Leptospira infection in rodents that were trapped in the port of Toulon, France. Kidney samples were taken from fifty-two rodents and real time polymerase chain reaction (PCR) amplification was performed. It found that ten kidney tissue specimens were found positive only with the first system and amplified fragments were evidenced with the rpoB system in nine cases. The study showed that the natural high infection rate of rodents living in an inner city implies a potential hazard for human health.

Serum samples from 73 lion tamarins of three species living at the Centro de Primatologia do Rio de Janeiro were tested for antibodies against serovars of Leptospira interrogans by microscopic agglutination test. Antibodies were detected in serum samples of 15% of tested samples. Seroreactivity prevalence was lower than that described in other non-human primates and could be explained by the arboreal behaviour of these animals. RESUMO: A leptospirose é uma doença infecciosa causada por bactérias do gênero Leptospira, que afeta animais domésticos, selvagens e também humanos. De outubro a novembro de 2014, numa propriedade rural localizada em Glorinha, RS, em que bovinos eram mantidos em resteva de arroz, 13 bezerros manifestaram hemoglobinúria e apatia, nove dos quais morreram em menos de 24 horas após o início dos sinais clínicos. Foram necropsiados quatro bezerros (A, B, C e D). Fragmentos de tecido foram fixados em formalina a 10%. Amostras de rim, fígado e pulmão dos Bezerros B, C e D foram enviadas para análise de PCR para RNA ribossômico 16S e a proteína Lip 32 de Leptospira. No exame macroscópico foram observados mucosas e tecido subcutâneo amarelados, fígado alaranjado, pulmões com múltiplas petéquias, predominantemente nos lóbulos craniais. A cavidade torácica do Bezerro A estava repleta de um líquido vermelho-escuro. À avaliação microscópica foi observada hemorragia acentuada nos pulmões; no fígado havia necrose e vacuolização hepatocelular centrolobular difusa moderada, além de infiltrado linfocítico periportal discreto. Nos rins observou-se nefrite intersticial linfoplasmocítica discreta multifocal. A análise por PCR teve resultado positivo para os Bezerros B e D. O diagnóstico de leptospirose nos bezerros foi baseado nos achados epidemiológicos, clínicos e patológicos, associados ao resultado positivo na PCR. Este estudo demonstra a importância da investigação da doença quando animais jovens são criados em áreas inundadas e têm manifestações clínicas de doença septicêmica aguda.

Due to the complex and dynamic epidemiology of leptospirosis on livestock, control is still controversial and frustrating. In this context, this paper discusses the main challenges and perspectives for the control of bovine leptospirosis, particularly under tropical conditions. In order to reduce the effects of the disease in cattle, it has been proposed that the control should integrate the trinomial antibiotic therapy (mainly streptomycin); vaccination (whole-cell bacterins); and environmental management. This last element should be carefully considered in tropics. Despite the enormous economic impact of the disease, mainly on its chronic and silent reproductive presentation, research on control programs is not proportional.
