Abstract

Neurodegenerative diseases are characterized by oxidative stress, mitochondrial damage, and death of neuronal cells. To counteract such damage and to favor neurogenesis, neurotrophic factors could be used as therapeutic agents. Octadecaneuropeptide (ODN), produced by astrocytes, is a potent neuroprotective agent. In N2a cells, we studied the ability of ODN to promote neuronal differentiation. This parameter was evaluated by phase contrast microscopy, staining with crystal violet, cresyl blue, and Sulforhodamine 101. The effect of ODN on cell viability and mitochondrial activity was determined with fluorescein diacetate and DiOC(6)(3), respectively. The impact of ODN on the topography of mitochondria and peroxisomes, two tightly connected organelles involved in nerve cell functions and lipid metabolism, was evaluated by transmission electron microscopy and fluorescence microscopy: detection of mitochondria with MitoTracker Red, and peroxisome with an antibody directed against the ABCD3 peroxisomal transporter. The profiles in fatty acids, cholesterol, and cholesterol precursors were determined by gas chromatography, in some cases coupled with mass spectrometry. Treatment of N2a cells with ODN (10(-14) M, 48 h) induces neurite outgrowth. ODN-induced neuronal differentiation was associated with modification of topographical distribution of mitochondria and peroxisomes throughout the neurites and did not affect cell viability and mitochondrial activity. The inhibition of ODN-induced N2a differentiation with H89, U73122, chelerythrine and U0126 supports the activation of a PKA/PLC/PKC/MEK/ERK-dependent signaling pathway. Although there is no difference in fatty acid profile between control and ODN-treated cells, the level of cholesterol and some of its precursors (lanosterol, desmosterol, lathosterol) was increased in ODN-treated cells. Abstract

The Concise Guide to PHARMACOLOGY 2021/22 is the fifth in this series of biennial publications. The Concise Guide provides concise overviews, mostly in tabular format, of the key properties of nearly 1900 human drug targets with an emphasis on selective pharmacology (where available), plus links to the open access knowledgebase source of drug targets and their ligands (www.guidetopharmacology.org), which provides more detailed views of target and ligand properties. Although the Concise Guide constitutes over 500 pages, the material presented is substantially reduced compared to information and links presented on the website. It provides a permanent, citable, point-in-time record that will survive database updates. The full contents of this section can be found at http://onlinelibrary.wiley.com/doi/bph.15538. G protein-coupled receptors are one of the six major pharmacological targets into which the Guide is divided, with the others being: ion channels, nuclear hormone receptors, catalytic receptors, enzymes and transporters. These are presented with nomenclature guidance and summary information on the best available pharmacological tools, alongside key references and suggestions for further reading. The landscape format of the Concise Guide is designed to facilitate comparison of related targets from material contemporary to mid-2021, and supersedes data presented in the 2019/20, 2017/18, 2015/16 and 2013/14 Concise Guides and previous Guides to Receptors and Channels. It is produced in close conjunction with the Nomenclature and Standards Committee of the International Union of Basic and Clinical Pharmacology (NC-IUPHAR), therefore, providing official IUPHAR classification and nomenclature for human drug targets, where appropriate.

15. Venom Peptide Repertoire of the European Myrmicine Ant Manica rubida: Identification of Insecticidal Toxins.

Abstract

Using an integrated transcriptomic and proteomic approach, we characterized the venom peptidome of the European red ant, Manica rubida. We identified 13 "myrmicitoxins" that share sequence similarities with previously identified ant venom peptides, one of them being identified as an EGF-like toxin likely resulting from a threonine residue modified by O-fucosylation. Furthermore, we conducted insecticidal assays of reversed-phase HPLC venom fractions on the blowfly Lucilia caesar, permitting us to identify six myrmicitoxins (i.e., U3-, U10-, U13-, U20-MYRTX-Mri1a, U10-MYRTX-Mri1b, and U10-MYRTX-Mri1c) with an insecticidal activity. Chemically synthesized U10-MYRTX-Mri1a, -Mri1b, -Mri1c, and U20-MYRTX-Mri1a irreversibly paralyzed blowflies at the highest doses tested (30-125 nmol·g-1). U13-MYRTX-Mri1a, the most potent neurotoxic peptide at 1 h, had reversible effects after 24 h (150 nmol·g-1). Finally, U3-MYRTX-Mri1a has no insecticidal activity, even at up to 55 nmol·g-1. Thus, M. rubida employs a paralytic venom rich in linear insecticidal peptides, which likely act by disrupting cell membranes.

Abstract

The Concise Guide to PHARMACOLOGY 2019/20 is the fourth in this series of biennial publications. The Concise Guide provides concise overviews of the key properties of nearly 1800 human drug targets with an emphasis on selective pharmacology (where available), plus links to the open access knowledgebase source of drug targets and their ligands (www.guidetopharmacology.org), which provides more detailed views of target and ligand properties. Although the Concise Guide represents approximately 400 pages, the material presented is substantially reduced compared to information and links presented on the website. It provides a permanent, citable, point-in-time record that will survive database updates. The full contents of this section can be found at http://onlinelibrary.wiley.com/doi/10.1111/bph.14748. G protein-coupled receptors are one of the six major pharmacological targets into which the Guide is divided, with the others being: ion channels, nuclear hormone receptors, catalytic receptors, enzymes and transporters. These are presented with nomenclature guidance and summary information on the best available pharmacological tools, alongside key references and suggestions for further reading. The landscape format of the Concise Guide is designed to facilitate comparison of related targets from material contemporary to mid-2019, and supersedes data presented in the 2017/18, 2015/16 and 2013/14 Concise Guides and previous Guides to Receptors and Channels. It is produced in close conjunction with the International Union of Basic and Clinical Pharmacology Committee on Receptor Nomenclature and Drug Classification (NC-IUPHAR), therefore, providing official IUPHAR classification and nomenclature for human drug targets, where appropriate.

Abstract

Despite the growing knowledge with regard to the immunomodulatory properties of host defense peptides, their impact on macrophage differentiation and on its associated microbicidal functions is still poorly understood. Here, we demonstrated that the P17, a new cationic antimicrobial peptide from ant venom, induces an alternative phenotype of human monocyte-derived macrophages (h-MDMs). This phenotype is characterized by a C-type lectin receptors (CLRs) signature composed of mannose receptor (MR) and Dectin-1 expression. Concomitantly, this activation is associated to an inflammatory profile characterized by reactive oxygen species (ROS), interleukin (IL)-1 beta, and TNF-alpha release. P17-activated h-MDMs exhibit an improved capacity to recognize and to engulf Candida albicans through the overexpression both of MR and Dectin-1. This upregulation requires arachidonic acid (AA) mobilization and the activation of peroxisome proliferator-activated receptor gamma (PPAR.) nuclear receptor through the leukotriene B4 (LTB4) production. AA/LTB4/PPAR gamma/Dectin-1-MR signaling pathway is crucial for P17-mediated anti-fungal activity of h-MDMs, as indicated by the fact that the activation of this axis by P17 triggered ROS production and inflammasome-dependent IL-1 beta release. Moreover, we showed that the increased anti-fungal immune response of h-MDMs by P17 was dependent on intracellular calcium mobilization triggered by the interaction of P17 with pertussis toxin-sensitive G-protein-coupled receptors on h-MDMs. Finally, we also demonstrated that P17-treated mice infected with C. albicans develop less severe gastrointestinal infection related to a higher efficiency of their macrophages to engulf Candida, to produce ROS and IL-1 beta and to kill the yeasts. Altogether, these results identify P17 as an original activator of the fungicidal response of macrophages that acts upstream PPAR gamma/CLRs axis and offer new immunomodulatory therapeutic perspectives in the field of infectious diseases.

35. Core build-up resin composites: an in-vitro comparative study.

Abstract

Aim: Resin composite (RC) are commonly used under full crowns. However, independent information is lacking to guide practitioners regarding core RC material selection. This study aimed at comparing the flexural properties of a large selection of commercially-available core build-up RCs (CBU-RC), either light-, self- or dual-cure, to conventional light-cure RCs. Methods: RCs were injected into a 25x2x2mm Teflon mold, and either light-cured during 20 s (materials with claimed light-cure characteristics) or covered by aluminum during 10 min (dualand self-cure CBU-RCs). They were subjected after a one-week water storage at 37.5°C to threepoint bending, and Flexural modulus (Eflex) and Flexural Strength (σ f) were calculated (n=20). Thermogravimetric analysis (n=3) was performed to determine inorganic filler content (%). Results: For dual-cure CBU-RCs, both RC (p<.0001) and light-curing (p=.0007) had a significant influence on Eflex, while only RC was significant for σ f (p<.0001). Between all conventional RCs and CBU-RCs, significant differences were observed (p<.0001), both regarding Eflex and σf, with values ranging from 3.9 to 15.5 GPa and from 76 to 130.3MPa, respectively. Higher Eflex values were observed for light-cure RCs than for self- and dual-cure ones, while no clear trend was noticed regarding σ f. Good linear correlation was found between inorganic filler content and Eflex (R2=0.85, p<.0001), but not with σ f (R2=0.08, p=.1609). Conclusion: This work demonstrated a positive influence of light-curing on dual-cure CBU-RC's Eflex. It also highlighted large differences in flexural properties (especially Eflex) among the investigated materials, questioning the use of some CBU-RCs as dentin replacement in case of large tissue loss. Abstract

La recherche a porté, plus particulièrement, sur l’analyse du discours du juge administratif dans la prise en compte des discriminations. Pour mener à bien cette réflexion, ont été mobilisés des juristes, des sociologues, des linguistes et des spécialistes de science politique. Ce travail s’est déroulé sur une période de deux ans. La première année a permis de cerner comment le juge administratif appréhendait le principe de non-discrimination. Afin d’atteindre cet objectif, il a fallu mobiliser la jurisprudence sur la période 2000-2015 (environ 7000 décisions et conclusions). Très vite, deux approches ont été empruntées, la première, afin d’être exhaustif, a consisté à examiner l’ensemble des critères de discrimination et à analyser comment le juge les exploitait, la seconde a consisté à s’intéresser aux instruments et aux modes de preuves que le juge utilisait pour sanctionner les discriminations. A la suite de cette étude, il ressort que le juge opte clairement pour le principe d’égalité et rechigne à mettre en oeuvre le principe de non-discrimination, même si sous l’influence des Cours européennes il l’admet mais en le liant au principe d’égalité à la française. La seconde année de la recherche a permis de mettre en lumière la singularité du juge administratif. En effet, par une technique comparatiste, le discours du juge administratif a été confronté à celui des cours judiciaires françaises, révélant, une attitude toute différente notamment de la Cour de cassation que ce soit dans la réception du principe de nondiscrimination ou dans la technique d’administration de la preuve. Prolongeant cette méthode, a été également constatée une influence réduite des Cours européennes et une démarche totalement différente des cours constitutionnelles italiennes et allemandes. En définitive, le principe de non discrimination apparaît comme une greffe prenant mal dans le discours du juge administratif qui lui préfère le principe d’égalité.

Abstract

We investigate the strain evolution in low strained gratings before and after embedding in the substrate material by high-resolution grazing incidence X-ray diffraction and elasticity theory. For the experiments, we use a non-coplanar multiple crystal arrangement. A suitable selection of reflections allows us to characterise the grating shape independently of the strain state. Depth selective measurements give evidence for a non-uniform strain relaxation in the surface grating and a complete planarization of the sample surface by the burying process. In the buried grating a counteraction of the embedding material is found. The counteraction only leads to a partial restraining of the grating. There remains a laterally periodic strain modulation in the grating, which extends into the non-patterned material below and above the grating and reaches the free surface of the embedding material. The experimental findings are confirmed by calculations of the strain field based on elasticity theory. The subsequent simulations of the diffraction patterns show a good quantitative agreement with experiment.

International audience; Les polymères utilisés en odontologie sont nombreux et leurs applications sont spécifiques. Ils peuvent être thermoplastiques ou thermodurcissables ou encore faire partie de la famille particulière des élastomères. Le cas particulier des résines composites est plus particulièrement développé dans cet article. Les résines composites ont suivi de très nombreuses améliorations au cours de ces dernières années. Les défauts majeurs ont été largement corrigés : l'usure est devenue proche de celle de l'émail, la rétraction de prise a été ramenée à 1,5 %, les propriétés mécaniques sont suffisantes pour répondre à la majorité des restaurations. Cependant, les échecs cliniques restent élevés. Une des principales causes est la récidive de carie qui est en relation avec la rétraction de prise encore trop élevée, le différentiel de coefficient d'expansion thermique, la forte variabilité de l'adhésion dentinaire mettant en évidence le rôle déterminant du praticien. Le matériau reste, en effet, très sensible à la technique de mise en oeuvre qui doit être respectée pour lui permettre d'exprimer ses qualités intrinsèques. Il reste donc très important pour le praticien de sélectionner, parmi le très et même trop large éventail de produits commercialisés, les composites suffisamment performants, de respecter les indications de chaque type de matériaux et de contrôler rigoureusement les nombreuses étapes du processus de collage-pose du composite.

Abstract

In the central nervous system of mammals, the gene encoding diazepam-binding inhibitor (DBI) is exclusively expressed in glial cells. Previous studies have shown that central administration of a DBI processing product, the octadecaneuropeptide ODN, causes a marked inhibition of food consumption in rodents. Paradoxically, however, the effect of food restriction on DBI gene expression has never been investigated. Here, we show that in mice, acute fasting dramatically reduces DBI mRNA levels in the hypothalamus and the ependyma bordering the third and lateral ventricles. I.p. injection of insulin, but not of leptin, selectively stimulated DBI expression in the lateral ventricle area. These data support the notion that glial cells, through the production of endozepines, may relay peripheral signals to neurons involved in the central regulation of energy homeostasis.
