Abstract

This paper investigates the radio resource management (RRM) design for multiuser rate-splitting multiple access (RSMA), accounting for various characteristics of practical wireless systems, such as the use of discrete rates, the inability to serve all users, and the imperfect successive interference cancellation (SIC). Specifically, failure to consider these characteristics in RRM design may lead to inefficient use of radio resources. Therefore, we formulate the RRM of RSMA as optimization problems to maximize respectively the weighted sum rate (WSR) and weighted energy efficiency (WEE), and jointly optimize the beamforming, user admission, discrete/continuous rates, accounting for imperfect SIC, which result in nonconvex mixed-integer nonlinear programs that are challenging to solve. Despite the difficulty of the optimization problems, we develop algorithms that can find high-quality solutions. We show via simulations that carefully accounting for the aforementioned characteristics, can lead to significant gains. Precisely, by considering that transmission rates are discrete, the transmit power can be utilized more intelligently, allocating just enough power to guarantee a given discrete rate. Additionally, we reveal that user admission plays a crucial role in RSMA, enabling additional gains compared to random admission by facilitating the servicing of selected users with mutually beneficial channel characteristics. Furthermore, provisioning for possibly imperfect SIC makes RSMA more robust and reliable.

2. High energy and high power primary Li-CF$_x$ batteries enabled by the combined effects of the binder and the electrolyte

Abstract

Several effective methods have been developed recently to demonstrate simultaneous high energy and high power density in Li - carbon fluoride (CF$_x$) batteries. These methods can achieve as high as 1000 Wh/kg energy density at 60-70 kW/kg power density (40-50 C rate) in coin cells and 750 Wh/kg energy density at 12.5 kW/kg power density (20 C rate) in pouch cells. This performance is made possible by ingenious nano-architecture design, controlled porosity, boron doping and electrolyte additives. In the present study, we show that a similarly great performance, 931 Wh/kg energy density at 59 kW/kg power density, can be achieved by using a polyacrylonitrile binder and a LiBF4 electrolyte in Li - graphite fluoride coin cells. We also demonstrate that the observed effect is the result of the right combination of the binder and the electrolyte. We propose that the mechanistic origin of the observed phenomena is an electro-catalytic effect by the polyacrylonitrile binder. While our proposed method has a competitive performance, it also offers a simple implementation and a scalable production of high energy and high power primary Li-CF$_x$ cells., Comment: 13 pages, 8 figures, 1 table

3. Alpha-effect in three-dimensional coherent vortex of conducting rotating liquid

Abstract

We study one-point statistics of spiral turbulent pulsations on the background of three-dimensional large-scale vortex in a rotating fluid. Assuming that the helical flow is created by a statistically axially symmetric random force with broken mirror symmetry, we analytically calculate velocity-vorticity mean including its magnitude and the anisotropy. For electrically conducting liquid, we examine the $\alpha$-effect in the system. The found elements of $\alpha$-matrix strongly depend on relation between Rossby $\mathrm{Ro}$ and magnetic Prandtl $\mathrm{Pr}_{\mathrm{m}}$ numbers in considered region $\mathrm{Ro}\lesssim 1$, $\mathrm{Pr}_{\mathrm{m}} \lesssim 1$. Abstract

ABSTRACT The pathogenesis of COVID-19 is associated with a hyperinflammatory immune response. Monocytes and macrophages play a central role in this hyperinflammatory response to SARS-CoV-2. NLRP3 inflammasome activation has been observed in monocytes of patients with COVID-19, but the mechanism and consequences of inflammasome activation require further investigation. In this study, we inoculated a macrophage-like THP-1 cell line, primary differentiated human nasal epithelial cell (hNEC) cultures, and primary monocytes with SARS-CoV-2. We found that the activation of the NLRP3 inflammasome in macrophages does not rely on viral replication, receptor-mediated entry, or actin-dependent entry. SARS-CoV-2 productively infected hNEC cultures without triggering the production of inflammasome cytokines IL-18 and IL-1β. Importantly, these cytokines did not inhibit viral replication in hNEC cultures. SARS-CoV-2 inoculation of primary monocytes led to inflammasome activation and induced a macrophage phenotype in these cells. Monocytic cells from bronchoalveolar lavage (BAL) fluid, but not from peripheral blood, of patients with COVID-19, showed evidence of inflammasome activation, expressed the proinflammatory marker CD11b, and displayed oxidative burst. These findings highlight the central role of activated macrophages, as a result of direct viral sensing, in COVID-19 and support the inhibition of IL-1β and IL-18 as potential therapeutic strategies to reduce immunopathology without increasing viral replication.IMPORTANCEInflammasome activation is associated with severe COVID-19. The impact of inflammasome activation on viral replication and mechanistic details of this activation are not clarified. This study advances our understanding of the role of inflammasome activation in macrophages by identifying TLR2, NLRP3, ASC, and caspase-1 as dependent factors in this activation. Further, it highlights that SARS-CoV-2 inflammasome activation is not a feature of nasal epithelial cells but rather activation of bystander macrophages in the airway. Finally, we demonstrate that two pro inflammatory cytokines produced by inflammasome activation, IL-18 and IL-1β, do not restrict viral replication and are potential targets to ameliorate pathological inflammation in severe COVID-19.

Abstract

Natural killer (NK) cells are a major subset of innate immune cells that are essential for host defense against pathogens and cancer. Two main classes of inhibitory NK receptors (NKR), KIR and CD94/NKG2A, play a key role in suppressing NK activity upon engagement with tumor cells or virus-infected cells, limiting their antitumor and antiviral activities. Here, we find that single-chain NKR antagonists linked to a VHH that binds the cell surface phosphatase CD45 potentiate NK and T activities to a greater extent than NKR blocking antibodies alone in vitro. We also uncovered crosstalk between NKG2A and Ly49 that collectively inhibit NK cell activation, such that CD45-NKG2A and CD45-Ly49 bispecific molecules show synergistic effects in their ability to enhance NK cell activation. The basis of the activity enhancement by CD45 ligation may reflect greater antagonism of inhibitory signaling from engagement of MHC I on target cells, combined with other mechanisms, including avidity effects, tonic signaling, antagonism of weak inhibition from engagement of MHC I on non-target cells, and possible CD45 segregation within the NK cell-target cell synapse. These results uncover a strategy for enhancing the activity of NK and T cells that may improve cancer immunotherapies.

Abstract

Millimeter-wave self-backhauled small cells are a key component of next-generation wireless networks. Their dense deployment will increase data rates, reduce latency, and enable efficient data transport between the access and backhaul networks, providing greater flexibility not previously possible with optical fiber. Despite their high potential, operating dense self-backhauled networks optimally is an open challenge, particularly for radio resource management (RRM). This paper presents, RadiOrchestra, a holistic RRM framework that models and optimizes beamforming, rate selection as well as user association and admission control for self-backhauled networks. The framework is designed to account for practical challenges such as hardware limitations of base stations (e.g., computational capacity, discrete rates), the need for adaptability of backhaul links, and the presence of interference. Our framework is formulated as a nonconvex mixed-integer nonlinear program, which is challenging to solve. To approach this problem, we propose three algorithms that provide a trade-off between complexity and optimality. Furthermore, we derive upper and lower bounds to characterize the performance limits of the system. Abstract

A specimen of common thresher shark Alopias vulpinus (Bonnaterre 1788) was caught in a fishing net in the waters off Piran (Gulf of Trieste, northern Adriatic Sea). It was a juvenile male with a total length of 2841 mm. In recent decades, other cases of juvenile specimens of common thresher shark have been recorded in Slovenian coastal waters. Despite the drastic decline in the population of this species, thresher shark continues to be caught in local Adriatic fisheries.

13. Velocity structure function in a geostrophic coherent vortex under strong rotation

Abstract

We consider analytically pair structure function of turbulent pulsations on the background of a coherent geostrophic vortex in a fast rotating fluid. The statistics of the turbulent pulsation is determined by their dynamics which is the dynamics of inertia waves affected by the differential rotation in the vortex and weak viscous damping. Our consideration is restricted by the smallest scales, where the velocity field remains smooth. We establish the anisotropy of the structure function. The velocity gradient of the turbulent pulsations achieves its largest value for the radial direction and its smallest value in the streamwise direction, resembling its behaviour in turbulent flow with mean shear component without imposed rotation., Comment: 4 pages, 0 figures

Abstract

Synthetic receptor signalling has the potential to endow adoptively transferred T cells with new functions that overcome major barriers in the treatment of solid tumours, including the need for conditioning chemotherapy1,2. Here we designed chimeric receptors that have an orthogonal IL-2 receptor extracellular domain (ECD) fused with the intracellular domain (ICD) of receptors for common γ-chain (γc) cytokines IL-4, IL-7, IL-9 and IL-21 such that the orthogonal IL-2 cytokine elicits the corresponding γc cytokine signal. Of these, T cells that signal through the chimeric orthogonal IL-2Rβ-ECD-IL-9R-ICD (o9R) are distinguished by the concomitant activation of STAT1, STAT3 and STAT5 and assume characteristics of stem cell memory and effector T cells. Compared to o2R T cells, o9R T cells have superior anti-tumour efficacy in two recalcitrant syngeneic mouse solid tumour models of melanoma and pancreatic cancer and are effective even in the absence of conditioning lymphodepletion. Therefore, by repurposing IL-9R signalling using a chimeric orthogonal cytokine receptor, T cells gain new functions, and this results in improved anti-tumour activity for hard-to-treat solid tumours.

The Beijing genotype is the leading resistant lineage in Asia and western countries. M. tuberculosis (Mtb) (sub) lineages responsible for most drug resistance in Ethiopia are not well described. Hence, this study aimed to identify the leading drug resistance sub-lineages and characterize first-line anti-tuberculosis drug resistance-associated single nucleotide polymorphisms (SNPs).Methods: A facility-based cross-sectional study was conducted in 2020– 2022 among new and presumptive multidrug resistant-TB (MDR-TB) cases in Northwest Ethiopia. Whole-genome sequencing (WGS) was performed on 161 isolates using Illumina NovaSeq 6000 technology. The SNP mutations associated with drug resistance were identified using MtbSeq and TB profiler Bioinformatics softwares.Results: Of the 146 Mtb isolates that were successfully genotyped, 20 (13.7%) harbored one or more resistance-associated SNPs. L4.2.2.ETH was the leading drug-resistant sub-lineage, accounting for 10/20 (50%) of the resistant Mtb. MDR-TB isolates showed extensive mutations against first-line anti-TB drugs. Ser450Leu/(tcg/tTg) for Rifampicin (RIF), Ser315Thr/(agc/aCc) for Isoniazid (INH), Met306Ile/(atg/atA(C)) for Ethambutol (EMB), and Gly69Asp for Streptomycin (STR) were the leading resistance associated mutations which accounted for 56.5%, 89.5%, 47%, and 29.4%, respectively. The presence of both clustered and non-clustered drug resistance (DR) isolates indicated that the epidemics is driven by both new DR development and acquired resistance.Conclusion: The high prevalence of drug-resistant TB due to geographically restricted sub-lineages (L4.2.2.ETH) indicates the ongoing local micro epidemics. The Mtb drug resistance surveillance system must be improved. Abstract

BACKGROUND:. This case series explores the management of respiratory failure in patients with large anterior tracheal thyroid tumors where tracheostomy is not an option. To our knowledge, this study is the first to address the challenges associated with caring for such patients. CASE SUMMARY:. We present the clinical courses of four intubated adults with advanced thyroid cancer and complex airway issues that preclude surgical tracheostomy. Interventions included custom airway stents, long-term intubation, and oncological therapies. Ethical quandaries around patient autonomy and capacity emerged, exacerbated by the absence of viable exit strategies for prolonged intubation, notably the performance of a tracheostomy, causing emotional distress in patients, families, and staff. CONCLUSIONS:. This study showcases the multifaceted challenges in medical, ethical, and emotional domains associated with managing intubated patients with complex disease precluding tracheotomies. We advocate for a nuanced, multidisciplinary, and personalized approach to confront unique issues in airway management, ethical considerations, and disposition.

Abstract

Liquid Chromatography coupled to Mass Spectrometry (LC-MS) based methods are commonly used for high-throughput, quantitative measurements of the proteome (i.e. the set of all proteins in a sample at a given time). Targeted LC-MS produces data in the form of a two-dimensional time series spectrum, with the mass to charge ratio of analytes (m/z) on one axis, and the retention time from the chromatography on the other. The elution of a peptide of interest produces highly specific patterns across multiple fragment ion traces (extracted ion chromatograms, or XICs). In this paper, we formulate this peak detection problem as a multivariate time series segmentation problem, and propose a novel approach based on the Transformer architecture. Here we augment Transformers, which are capable of capturing long distance dependencies with a global view, with Convolutional Neural Networks (CNNs), which can capture local context important to the task at hand, in the form of Transformers with Convolutional Self-Attention. We further train this model in a semisupervised manner by adapting state of the art semisupervised image classification techniques for multi-channel time series data. Experiments on a representative LC-MS dataset are benchmarked using manual annotations to showcase the encouraging performance of our method; it outperforms baseline neural network architectures and is competitive against the current state of the art in automated peak detection.

We investigated the possibility of diagnosing the degree of departure from the Maxwellian distribution using the Fe XVII - Fe XVIII spectra originating in plasmas in collisional ionization equilibrium, such as in the cores of solar active regions or microflares. The original collision strengths for excitation are integrated over the non-Maxwellian electron $\kappa$-distributions characterized by a high-energy tail. Synthetic X-ray emission line spectra were calculated for a range of temperatures and $\kappa$. We focus on the 6-24 A spectral range to be observed by the upcoming Marshall Grazing-Incidence X-ray Spectrometer MaGIXS. We find that many line intensity ratios are sensitive to both $T$ and $\kappa$. Best diagnostic options are provided if a ratio involving both Fe XVII and Fe XVIII is combined with another ratio involving lines formed within a single ion. The sensitivity of such diagnostics to $\kappa$ is typically a few tens of per cent. Much larger sensitivity, of about a factor of two to three, can be obtained if the Fe XVIII 93.93 A line observed by SDO/AIA is used in conjuction with the X-ray lines. Abstract

Background: Literature suggests colonic resection and primary anastomosis (RPA) instead of Hartmann’s procedure (HP) for the treatment of left-sided colonic emergencies. We aim to evaluate the surgical options globally used to treat patients with acute left-sided colonic emergencies and the factors that leading to the choice of treatment, comparing HP and RPA. Methods: This is a prospective, international, multicenter, observational study registered on ClinicalTrials.gov. A total 1215 patients with left-sided colonic emergencies who required surgery were included from 204 centers during the period of March 1, 2020, to May 31, 2020. with a 1-year follow-up. Results: 564 patients (43.1%) were females. The mean age was 65.9 ± 15.6 years. HP was performed in 697 (57.3%) patients and RPA in 384 (31.6%) cases. Complicated acute diverticulitis was the most common cause of left-sided colonic emergencies (40.2%), followed by colorectal malignancy (36.6%). Severe complications (Clavien-Dindo ≥ 3b) were higher in the HP group (P < 0.001). 30-day mortality was higher in HP patients (13.7%), especially in case of bowel perforation and diffused peritonitis. 1-year follow-up showed no differences on ostomy reversal rate between HP and RPA. (P = 0.127). A backward likelihood logistic regression model showed that RPA was preferred in younger patients, having low ASA score (≤ 3), in case of large bowel obstruction, absence of colonic ischemia, longer time from admission to surgery, operating early at the day working hours, by a surgeon who performed more than 50 colorectal resections. Conclusions: After 100 years since the first Hartmann’s procedure, HP remains the most common treatment for left-sided colorectal emergencies. Treatment’s choice depends on patient characteristics, the time of surgery and the experience of the surgeon. RPA should be considered as the gold standard for surgery, with HP being an exception.

Abstract

Summary: Interleukin-21 (IL-21) plays a critical role in generating immunological memory by promoting the germinal center reaction, yet clinical use of IL-21 remains challenging because of its pleiotropy and association with autoimmune disease. To better understand the structural basis of IL-21 signaling, we determine the structure of the IL-21–IL-21R–γc ternary signaling complex by X-ray crystallography and a structure of a dimer of trimeric complexes using cryo-electron microscopy. Guided by the structure, we design analogs of IL-21 by introducing substitutions to the IL-21–γc interface. These IL-21 analogs act as partial agonists that modulate downstream activation of pS6, pSTAT3, and pSTAT1. These analogs exhibit differential activity on T and B cell subsets and modulate antibody production in human tonsil organoids. These results clarify the structural basis of IL-21 signaling and offer a potential strategy for tunable manipulation of humoral immunity.

Abstract

Li3BN2 is investigated for the first time as a transition metal free, high capacity cathode material for Li-ion batteries. It is shown that α-Li3BN2 can exhibit a specific capacity of 890 mA h g−1 with the charge storage mechanism associated with the valence state change of N ions in the BN2 anion. The specific capacity demonstrated in this study is the highest one ever reported in literature for an intercalation-type cathode material. Further, using the valence state change of N ions as a charge storage mechanism opens the door for designing additional high performance, transition metal free electrodes in the future.

As such, preservice physical education teachers (PPETs) must be trained appropriately for effective youth fitness testing, with careful consideration dedicated to PPETs perceptions, since many purported attitudes to have an effect on behavior (Ajzen 1985. "From intentions to actions: A theory of planned behavior." In "Action control: From cognition to behavior," edited by J. Kuhl, and J. Beckmann. Berlin: Springer-Verlag). Our understanding on PPETs' attitudes toward fitness testing is limited, with no research found on the topic for Chinese PPETs. Purposes: The primary purpose of the current study was to examine the overall attitudes of Chinese PPETs' toward school-based fitness testing. The secondary purpose was to test differences of attitudes in gender and years in college, and interaction between these two independent variables. Method: A total of 1012 PPETs from 11 Chinese universities participated in the study. Participants consisted of 28.3% of freshmen, 37.0% of sophomore, 19.5% of junior, and 15.2% of senior, with an average age of 21.2. Participants were recruited via emails and asked to complete a previously validated 16-item survey in English, measuring PPETs' attitudes toward fitness tests (Keating et al. 2008. "Physical Education Teacher Attitudes toward Fitness Tests Scale: Cross-Revalidation and Modification." "Measurement in Physical Education and Exercise Science" 12 (2): 72. https://doi.org/10.1080/10913670801903969). The survey consisted of two domains (i.e. beliefs of the importance of fitness testing and effects of fitness test implementation with two subdomains: enjoyment of fitness test implementation and enjoyment of using fitness test results). The survey was translated into Chinese for distribution. ANOVA was performed to test overall attitude differences in gender and year in college, while two-way MANOVA was used to examine attitude subdomain differences in gender and year in college. Results: The overall attitudes of Chinese PPETs were slightly positive, and consistent with existing literature. Only significant gender differences in both overall attitudes and the three subdomains were found. It was also found that year in college was not significant, nor was the interaction between gender and year in college. These results may suggest that PETE programs have little effect on improving PPETs' attitudes toward fitness testing. Conclusions: Chinese PPETs' attitudes toward youth fitness testing were only slightly positive. Gender may affect Chinese PPETs' attitudes, while surprisingly PETE programs may not focus on PPETs' attitudes toward fitness testing. Future interventions on improving Chinese PPETs' attitudes toward fitness tests are needed, with specialized interests targeting female candidates during PETE training programs.

Abstract

The synthesis of vitamin D is strongly linked to the availability of solar energy. For a long time, it was clear that using the erythemal irradiances is a good choice to calculate the exposure time. Several authors argue that this method of minimum exposure time overestimates the calculation of the solar irradiance of vitamin D. In this paper, the vitamin D and erythemal irradiances are calculated on the basis of the spectral solar irradiance in Urcuqu\'i (Ecuador). From these results, we obtain the minimal exposure time. It was found that there is a difference between the use of the erythemal irradiance and the vitamin D one; namely, the exposure times were too large (almost a 100% difference) for the erythemal irradiance., Comment: 6 pages, 3 figures

Abstract

The demand for high-energy-density lithium-ion batteries (LIBs) has driven intensive research into cathode materials that exhibit both superior performance and stability over multiple charge–discharge cycles. This work focuses on enhancing the dynamic stability of single-crystal (SC) low-cobalt (Co) nickel-rich (Ni-rich) cathode materials, crucial for the advancement of LIB technology. The proposed strategy involves co-doping of iron (Fe) and aluminum (Al) with an optimized composition to mitigate the capacity degradation and voltage fading observed for traditional Ni-rich (≥90%) cathodes. Through a comprehensive investigation combining theoretical modeling, material synthesis, and electrochemical characterization, the synergistic effects of Fe/Al co-doping are elucidated. The presence of Fe and Al ions in the crystal lattice not only stabilizes the structural integrity but also facilitates the suppression of phase transformation and surface degradation during cycling. Moreover, the incorporation of Fe and Al ions optimizes the lithium (Li)-ion diffusion kinetics and enhances the electronic conductivity, leading to improved electrochemical performance. The achieved dynamic stability of the co-doped SC cathode material enables prolonged cycle life and high-rate capability, making it a promising candidate for next-generation LIBs.
