The emergence of new resistant bacterial strains is a worldwide challenge. A resistant bacterial population can emerge from a single cell that acquires resistance or persistence. Hence, new ways of tackling the mechanism of antibiotic response, such as single cell studies are required. It is necessary to see what happens at the single cell level, in order to understand what happens at the population level. To date, linking the heterogeneity of single-cell susceptibility to the population-scale response to antibiotics remains challenging due to the trade-offs between the resolution and the field of view. Here we present a platform that measures the ability of individual E. coli cells to form small colonies at different ciprofloxacin concentrations, by using anchored microfluidic drops and an image and data analysis pipelines. The microfluidic results are benchmarked against classical microbiology measurements of antibiotic susceptibility, showing an agreement between the pooled microfluidic chip and replated bulk measurements. Further, the experimental likelihood of a single cell to form a colony is used to provide a probabilistic antibiotic susceptibility curve. In addition to the probabilistic viewpoint, the microfluidic format enables the characterization of morphological features over time for a large number of individual cells. This pipeline can be used to compare the response of different bacterial strains to antibiotics with different action mechanisms.

Introduction: To mitigate the global health threat posed by climate change, multifaceted responses are required. In previous work, older, male, and highly educated individuals were shown to exhibit greater general readiness to act against climate change; the same is true for people living in larger communities. Yet, the sociodemographic effects may vary for the single facets of the readiness to act against climate change—namely individual climate-friendly behavior, acceptance of climate-protection policies, and political participation. Methods: Data from four waves (2022–2023) of the Planetary Health Action Survey (PACE) were analyzed (N = 3,830, nonprobabilistic representative German sample). Sociodemographic variables (gender, age, community size, education and others) and the three abovementioned facets of the readiness to act against climate change were assessed to examine their relationships. Results: Patterns varied between the indicators. Women and older individuals exhibited more climate-friendly behavior and policy acceptance. In contrast, levels of political participation were higher for men and younger individuals. Higher education was linked to less sustainable behavior, greater policy acceptance, and higher participation, while municipality size was only linked to the latter two. Additional analyses explore the relations between age and the constructs at item level. Conclusions: Findings underscore the importance of a nuanced understanding of people's readiness to act against climate change. Depending on communication objectives, different target groups may be relevant, e.g., do campaigners want to increase political participation vs. change individual behaviors. While older individuals exhibit greater readiness to act via individual behavior and policy acceptance, it is important to include young people in climate-communication efforts.

In der Geldwäschebekämpfung ist der Einsatz von KI mit gravierenden Risiken automatisierter Fehlentscheidungen verbunden, die durch die Privatisierung des geldwäscherechtlichen Verdachtswesens zu massiven Grundrechtsumgehungen führen können. Eine staatliche Einhegung wäre daher wünschenswert (gewesen).

Light transmission aggregometry (LTA) is a method used to investigate platelet functions in platelet-rich plasma (PRP), notably when screening for platelet disorders. Various national guidelines and recommendations help in setting up the LTA test in specialized laboratories. However, due to the nature of the sample matrix and its subsequent specificities, more accurate positions are needed to achieve LTA accreditation according to the standard NF EN ISO 15 189. We reviewed guidelines and recommendations as they can be useful in the accreditation process, and we conducted a survey on LTA practice among members of the Société Française de Thrombose et d’Hémostase (SFTH) in 2021. We formulated 28 proposals, which have been approved by vote within the SFTH. All aspects to take into consideration for the proper conduct of LTA assays and their accreditation have been covered. Notably, preanalytical, analytical and postanalytical aspects are depicted, including blood sampling, PRP preparation, instruments, agonists, performance assessment, personnel training and data interpretation. This document, essentially representing a French position paper on the current recommendations and subsequent proposals for LTA accreditation, might prove useful also outside France for relevant laboratories and auditors involved in LTA accreditation.

Introduction Climate change is a paramount global health threat with multifaceted implications. Societal change is required to mitigate the negative effects of climate change, as well as help people adapt to the associated health risks. This requires situation-specific, large-scale data to help scientists and policymakers understand public perceptions and behaviours and identify the levers to increase public readiness to act against climate change and protect health. The Planetary Health Action Survey (PACE) assesses this readiness to act as a regular monitoring of representative samples in Germany. The ongoing monitoring seeks to develop and refine an integrated conceptual model of the trait-like readiness to act, comprising policy acceptance, political participation and individual behaviours as indicators of the trait. It also proposes as set of determinants to explain different levels of readiness to act. This study protocol provides newly developed valid measurement instruments and the methodological details of the monitoring.Method and analysis PACE assesses indicators of the readiness to act and a set of sociocognitive factors predicting the readiness to act in continuous cross-sectional data collections. The latter comprise climate change knowledge, trust in institutions, perceived health risks, self-efficacy, social norms and perceived effectiveness of policy measures. The online questionnaire is updated regularly. Data collection involves non-probabilistic quota samples from Germany (n≈1000 at each collection).Ethics and dissemination The project adheres to the ethical guidelines of the University of Erfurt and the German Research Foundation. Ethical clearance was granted by the University’s Institutional Review Board (No #20220525/No #2024-01). Participants are guaranteed confidentiality and anonymity, and informed consent is obtained before participation. Results will be published in peer-reviewed journals. Additionally, we aim to inform and empower the public and support stakeholders (media, policymakers, climate protection organisations) in preparing climate communication and assisting policymakers through the project website including an interactive tool, detailed reports and short summaries for practitioners.

Abstract This paper analyses the integration of social considerations into public transport planning. It addresses the challenge of balancing social benefits against objectives such as efficiency and cost minimization. Through a literature review, this paper examines methods for assessing public transport accessibility for potentially disadvantaged groups and identifies knowledge gaps in existing research. The analysis reveals a predominant focus on post-implementation (ex-post) accessibility assessments and a lack of research examining potential impacts (ex-ante) during planning stages. Furthermore, the paper identifies a lack of research on how to weigh social benefits against other more conventional objectives such as efficiency and cost minimization and also against environmental considerations. Research usually proposes changes ex-post based solely on a desire to improve accessibility for potentially disadvantaged groups. Therefore, there is a need for more research on how to manage conflicting interests in planning, for example through composite methods. The paper also highlights the tendency of research to overlook the practical usability of methods developed. The relevance of methods to policy makers and planners responsible for planning public transport systems remains largely unexplored. To develop practically useful methods, research needs a better understanding of the norms governing planning practices. It is common to discuss how the mobility needs of potentially disadvantaged passenger groups should be met in isolation from policy contexts. More research is needed about how the potential tensions between social, economic, and environmental considerations play out in public transport planning practices. All of this leads to a risk of a theory–practice gap, marked by a disconnect between research and the practical needs of the public transport providers. By proposing a more nuanced research approach that better reflects the complexity of real-world planning and the different needs of user groups, research can enable public transport planning for what we term ‘sustainable accessibility’.

Cohesion measure launched from the Family Adaptability and Cohesion Evaluation Scale (FACES) is a well-known instrument to assess family functioning. It is essential to perform a measurement equivalence test of cohesion targeting racially diverse families. The web-based self-administered survey was performed and 291 college/university students’ responses in Ohio were collected (Whites = 37.5%, Hispanics = 23%, Asians = 23%, and Blacks = 16.5%). This study explicated measurement equivalence for family cohesion structure (balanced cohesion, disengagement, enmeshed). The multiple group analysis was performed to investigate whether parameters in the measurement models of the “cohesion sub-structures (balanced cohesion, disengagement, enmeshed; 7 items of each)” were equivalent across the four racial groups. While “balanced cohesion” and “disengagement” structures showed measurement invariance across the groups, the “enmeshed” structure showed significant measurement variance across the groups. Two-items, “Family members feel pressured to spend most free time together” and “We feel too connected to each other” were inadequate for Hispanics’ enmeshment. One-item, “Family members have little need for friends outside the family” was inadequate to explain enmeshment of Blacks and Asians. Professionals should be aware of possible misinterpretations of results from the cohesion measure of FACES IV when they particularly assess the enmeshment status of racially diverse families with closer attention to cross-cultural comparability.

Purpose: To investigate self-reported work ability in patients with musculoskeletal disorders in physiotherapy treatment. What characterizes patients with reduced work ability, whether work ability changes during treatment and characteristics of patients reporting no improvement. Design: Cohort study Material: 1498 patients with musculoskeletal disorders of working age who attend physiotherapy in primary healthcare. Method: Questionnaires at baseline and after three months. Results: 63% reported reduced work ability at baseline. Factors associated with reduced work ability were reduced daily activity level, physical work, psychological complaints, widespread pain, and high pain intensity. Participants with reduced work ability improved their average work ability by 1.7 on the WAS (work ability scale 0-10), however 53% did not achieve improvement. Characteristics of patients without improvement in work ability were long-term complaints, widespread pain, and psychological complaints. Conclusion: It seems difficult to improve work ability for patients with long-term and complex disorders in physiotherapy treatment. Physiotherapists should pay particular attention to psychological factors that inhibit improvement in work ability.

Background: The German status report on climate change and health 2023 identifies numerous health risks that are caused or exacerbated by climate change. One recommendation arising from the report is to strengthen education, information, and communication in the field. This article aims to serve as a basis for this. Methods: Based on four survey waves (2022/2023) of the PACE study (Planetary Health Action Survey, n=3,845, online), the status of risk perception as well as the Readiness to Act against climate change in the adult population in Germany is examined and a target group analysis is carried out. Results: Some health risks due to the climate crisis are perceived as comparatively low (e.g. mental health problems). People with higher risk perception show a higher Readiness to Act. Younger people, men, people with low education, and those living in smaller communities are identified as relevant target groups as they have a lower Readiness to Act. One third state that they never or hardly ever seek out specific information on climate change. Media use differs depending on target group. Conclusions: Target group-specific communication can help to educate people about the health impacts of the climate crisis. In the discussion of this article, implications from existing literature are discussed in detail, which offer practical guidance for effective climate change communication.

A surprising “planar” thermal Hall effect, whereby the field is parallel to the current, has recently been observed in a few magnetic insulators; this effect has been attributed to exotic excitations such as Majorana fermions or chiral magnons. Here, we investigate the possibility of a planar thermal Hall effect in three different cuprate materials, in which the conventional thermal Hall conductivity κ_{xy} (with an out-of-plane field perpendicular to the current) is dominated by either electrons or phonons. Our measurements show that the planar κ_{xy} from electrons in cuprates is zero, as expected from the absence of a Lorentz force in the planar configuration. By contrast, we observe a sizable planar κ_{xy} in those samples where the thermal Hall response is due to phonons, even though it should, in principle, be forbidden by the high crystal symmetry. Our findings call for a careful reexamination of the mechanisms responsible for the phonon thermal Hall effect in insulators.

Abstract Background Cardiovascular disease disproportionately affects African Americans. Psychosocial factors, including the experience of and emotional reactivity to racism and interpersonal stressors, contribute to the etiology and progression of cardiovascular disease through effects on health behaviors, stress-responsive neuroendocrine axes, and immune processes. The full pathway and complexities of these associations remain underexamined in African Americans. The Heart of Detroit Study aims to identify and model the biopsychosocial pathways that influence cardiovascular disease risk in a sample of urban middle-aged and older African American adults. Methods The proposed sample will be composed of 500 African American adults between the ages of 55 and 75 from the Detroit urban area. This longitudinal study will consist of two waves of data collection, two years apart. Biomarkers of stress, inflammation, and cardiovascular surrogate endpoints (i.e., heart rate variability and blood pressure) will be collected at each wave. Ecological momentary assessments will characterize momentary and daily experiences of stress, affect, and health behaviors during the first wave. A proposed subsample of 60 individuals will also complete an in-depth qualitative interview to contextualize quantitative results. The central hypothesis of this project is that interpersonal stressors predict poor cardiovascular outcomes, cumulative physiological stress, poor sleep, and inflammation by altering daily affect, daily health behaviors, and daily physiological stress. Discussion This study will provide insight into the biopsychosocial pathways through which experiences of stress and discrimination increase cardiovascular disease risk over micro and macro time scales among urban African American adults. Its discoveries will guide the design of future contextualized, time-sensitive, and culturally tailored behavioral interventions to reduce racial disparities in cardiovascular disease risk.

Die Nutzung des Internets durchzieht den Alltag Jugendlicher. Diese technologische Entwicklung geht mit einer Vielzahl an positiven neuen Möglichkeiten einher. Allerdings können insbesondere soziale Medien für Jugendliche ein Risiko darstellen, in Radikalisierungsprozesse hineinzugeraten. In der Forschung wurde bislang kaum betrachtet, welche spezifischen Online-Aktivitäten zu einer Radikalisierung führen bzw. mit ihr einhergehen. Der Zusammenhang zwischen rechtsextremen Einstellungen und verschiedenen Online-Aktivitäten wie Konsum von Internetseiten, Vernetzung und Posting rechter Inhalte wird in diesem Beitrag geschlechtsspezifisch untersucht, da sowohl das Online-Verhalten als auch das Ausmass extremistischer Einstellungen bei Jungen und Mädchen unterschiedlich ausgeprägt ist und somit angenommen werden kann, dass auch der Zusammenhang geschlechtsspezifischer Betrachtung bedarf. Es werden Daten einer Schüler:innenbefragung (N = 6.715) herangezogen, die vom Kriminologischen Forschungsinstitut Niedersachsen durchgeführt und vom Bundesministerium für Bildung und Forschung gefördert wurde. Die Ergebnisse zeigen, dass rechtsextreme Einstellungen mit dem Konsum politischer Websites in Verbindung stehen (insbesondere bei Jungen). Vernetzung und Posting-Verhalten weisen (unabhängig vom Geschlecht) nur geringe Zusammenhänge mit rechtsextremen Einstellungen auf.

Abstract Background High-throughput screening (HTS) of small molecule drug libraries has greatly facilitated the discovery of new cancer drugs. However, most phenotypic screening platforms used in the field of oncology are based solely on cancer cell populations and do not allow for the identification of immunomodulatory agents. Methods We developed a phenotypic screening platform based on a miniaturized co-culture system with human colorectal cancer- and immune cells, providing a model that recapitulates part of the tumor immune microenvironment (TIME) complexity while simultaneously being compatible with a simple image-based readout. Using this platform, we screened 1,280 small molecule drugs, all approved by the Food and Drug Administration (FDA), and identified statins as enhancers of immune cell-induced cancer cell death. Results The lipophilic statin pitavastatin had the most potent anti-cancer effect. Further analysis demonstrated that pitavastatin treatment induced a pro-inflammatory cytokine profile as well as an overall pro-inflammatory gene expression profile in our tumor-immune model. Conclusion Our study provides an in vitro phenotypic screening approach for the identification of immunomodulatory agents and thus addresses a critical gap in the field of immuno-oncology. Our pilot screen identified statins, a drug family gaining increasing interest as repurposing candidates for cancer treatment, as enhancers of immune cell-induced cancer cell death. We speculate that the clinical benefits described for cancer patients receiving statins are not simply caused by a direct effect on the cancer cells but rather are dependent on the combined effect exerted on both cancer and immune cells.

The Mekong region consists of six countries linked by the Mekong River, namely Cambodia, China, Laos, Myanmar, Thailand, and Vietnam. Since the early 1990s, regional cooperation in the Mekong region has gained new momentum and become more dynamic. Significant transformations in the trend of regionalism occurred with an increase in the number of multilateral mechanisms. Given the myriad of existing mechanisms, there is a question of whether there is any chance for a new sectoral cooperation mechanism. The following research seeks to answer that question by taking tourism cooperation as a case study. The research will explore current existing tourism cooperation frameworks and factors that influence regional tourism collaboration. It argues that there is room for a new tourism cooperation mechanism. However, there are also push and pull factors which must be taken into consideration as the potential institution is being built. ABSTRACT: The removal (culling) of dairy cows from the farm is a regular and required management practice. The main objectives of this study were to describe the involvement of bovine veterinarians in Ontario, Canada, in dairy cull cow management decisions, their expectations of current producer practices, and their perspectives on welfare issues and best management practices for cull dairy cows. Between February and May 2021, a province-wide survey was conducted among all members of the Ontario Association of Bovine Practitioners (OABP), with questions addressing veterinarians' background and demographics, veterinary clinic characteristics, cull cow management, down cow management, and learning preferences. The response rate for the survey was 25.1%, with 45 responses meeting all eligibility criteria. Dairy veterinarians would like to have greater involvement in cull cow management; when asked for desired involvement in culling decisions, 57.5% of veterinarians would like to be involved in culling decisions most of the time or always. Most (70.0%) veterinarians believed that cull cow welfare has improved over the last decade. However, most respondents also identified at least one area of management (farm of origin, transportation, auction, slaughter) as currently being a high risk to cull cow welfare. Given the frequency of their visits, trust by dairy producers, and knowledge, herd veterinarians are among those in the best position to improve the care of cull dairy cows. Information generated from this survey can be used to inform the development of improved decision-making tools for culling cows, and this, along with increased veterinarians' involvement in cull cow management, could improve the economic and welfare outcomes of culling decisions for dairy producers.

The HIV/AIDS epidemic in Sweden accelerated the hybridisation of welfare, as volunteer organisations underwent rapid processes of professionalization and assumed growing responsibilities for the distribution of welfare services. A sense of urgency prompted by terrifying horror scenarios spread in the media and helped motivate politicians and fund-giving agencies to channel large amounts of money into volunteer organisations like RFSL (the National Union for Sexual Equality), RFSU (the Swedish Association for Sexuality Education) and Noaks Ark, as well as to the various self-help groups that emerged for HIV-positive men who had sex with men, drug users, haemophiliacs, and, to a lesser degree, sex workers. The article focus on how these volunteer organisations – old and new – interacted with the state and the public health sector, how they interacted – and came into conflict – with each other, and how the membership-oriented activities changed over time due to professionalisation and to the introduction of anti-retroviral drugs in 1996.

Abstract Background The study addresses knowledge gaps in research regarding influences of routine health care delivery of physical activity on prescription (PAP). The aim was to investigate if patient and health care characteristics are associated with increased physical activity 1 year after prescription among patients offered counselor support in addition to health care professionals’ prescription. The study was conducted in primary and secondary care in a Swedish health care region. Methods All PAP recipients during 1 year were invited (N = 1503) to participate in this observational prospective study. Data were collected from medical records and questionnaires (baseline and follow-up). Descriptive statistics and multiple logistic regression analysis were used. The outcome variable was increased physical activity after 1 year. Study variables were patient and health care characteristics. Results Three hundred and fifty-five patients with complete follow-up data were included. The mean age was 62 years (SD = 14; range, 18–90) and 68% were females. Almost half (47%) had increased physical activity 1 year after PAP. Multiple logistic regression analysis showed that increased physical activity at follow-up was positively associated with lower baseline activity, counselor use, and positive perception of support. Counselor users with low baseline activity had higher odds ratio for increased physical activity at follow-up than non-users (OR = 7.2, 95% CI = 2.2–23.5 vs. OR = 3.2, 95% CI = 1.4–7.5). Positive perception of support was associated with increased physical activity among counselor users but not among non-users. Conclusions An increase in physical activity after PAP was related to low baseline activity, positive perception of support, and use of counselor support after PAP. Qualified counseling support linked to PAP seems to be important for achieving increased physical activity among patients with lower baseline activity.

Removal of cows from dairy cattle production is a routine and unavoidable practice of the dairy industry and is often referred to as culling. The objectives of this study were to use a survey to describe current on-farm cull cow management, farmers' perception of cull cows' journeys to slaughter, and the adoption of current recommendations and regulations by Ontario dairy farmers. All Ontario dairy farmers were invited to complete a cull cow management survey between December 2020 and March 2021 that included 44 questions covering farmer demographic information, farm characteristics, and cull cow management. The survey response rate was 7.4% (n = 248); a total of 226 of the responses were included in this study for analysis. Most respondents indicated they have a written standard operating procedure (SOP) for cull cows (62%), and 48, 13, and 15% of those identified they use their cull cow SOP “always,” “sometimes,” and “never,” respectively. The more confident respondents were that cull cows arrived at slaughter in the condition they left the farm the less likely they were to have a cull cow SOP [odds ratio (OR) 0.83]. The most important sources of information for the management of cull cows were the herd veterinarian (64%) and members of the marketing/regulatory organization the Dairy Farmers of Ontario (44%). Drug withdrawal time was the only factor most respondents (73%) considered “very important” for the assessment of cull cows prior to transport. Most farmers believe cull cows journey from the farm to slaughter is three or less days (55%), and the confidence of farmers that cull cows arrive at slaughter in the condition they left their farm was generally high. Lastly, most farmers (66%) identified they were familiar with recent regulatory changes around the fitness, duration of transport, and lactation status for cull cows. These results highlight farmers' perceptions of the impacts and durations of the journey of cull cows differs from reality, and there are misunderstandings of the requirements for cull cow management. Further research should investigate how different strategies for training farmers may lead to improved cull cow welfare and regulatory compliance.

Abstract Background The SARS-CoV-2 pandemic is currently leading to increasing numbers of COVID-19 patients all over the world. Clinical presentations range from asymptomatic, mild respiratory tract infection, to severe cases with acute respiratory distress syndrome, respiratory failure, and death. Reports on a dysregulated immune system in the severe cases call for a better characterization and understanding of the changes in the immune system. Methods In order to dissect COVID-19-driven immune host responses, we performed RNA-seq of whole blood cell transcriptomes and granulocyte preparations from mild and severe COVID-19 patients and analyzed the data using a combination of conventional and data-driven co-expression analysis. Additionally, publicly available data was used to show the distinction from COVID-19 to other diseases. Reverse drug target prediction was used to identify known or novel drug candidates based on finding from data-driven findings. Results Here, we profiled whole blood transcriptomes of 39 COVID-19 patients and 10 control donors enabling a data-driven stratification based on molecular phenotype. Neutrophil activation-associated signatures were prominently enriched in severe patient groups, which was corroborated in whole blood transcriptomes from an independent second cohort of 30 as well as in granulocyte samples from a third cohort of 16 COVID-19 patients (44 samples). Comparison of COVID-19 blood transcriptomes with those of a collection of over 3100 samples derived from 12 different viral infections, inflammatory diseases, and independent control samples revealed highly specific transcriptome signatures for COVID-19. Further, stratified transcriptomes predicted patient subgroup-specific drug candidates targeting the dysregulated systemic immune response of the host. Conclusions Our study provides novel insights in the distinct molecular subgroups or phenotypes that are not simply explained by clinical parameters. We show that whole blood transcriptomes are extremely informative for COVID-19 since they capture granulocytes which are major drivers of disease severity.

Abstract Background Several governments apply the policy of external price referencing (EPR), which considers the prices of a medicine in one or more other countries for the purpose of setting the price in the own country. Different methodological choices can be taken to design EPR. The study aimed to analyse whether, or not, and how changes in the methodology of EPR can impact medicine prices. Methods The real-life EPR methodology as of Q1/2015 was surveyed in all European Union Member States (where applicable), Iceland, Norway and Switzerland through a questionnaire responded by national pricing authorities. Different scenarios were developed related to the parameters of the EPR methodology. Discrete-event simulations of fictitious prices in the 28 countries of the study that had EPR were run over 10 years. The continuation of the real-life EPR methodology in the countries as surveyed in 2015, without any change, served as base case. Results In most scenarios, after 10 years, medicine prices in all or most surveyed countries were—sometimes considerably—lower than in the base case scenario. But in a few scenarios medicine prices increased in some countries. Consideration of discounts (an assumed 20% discount in five large economies and the mandatory discount in Germany, Greece and Ireland) and determining the reference price based on the lowest price in the country basket would result in higher price reductions (on average − 47.2% and − 34.2% compared to the base case). An adjustment of medicine price data of the reference countries by purchasing power parities would lead to higher prices in some more affluent countries (e.g. Switzerland, Norway) and lower prices in lower-income economies (Bulgaria, Romania, Hungary, Poland). Regular price revisions and changes in the basket of reference countries would also impact medicine prices, however to a lesser extent. Conclusions EPR has some potential for cost-containment. Medicine prices could be decreased if certain parameters of the EPR methodology were changed. If public payers aim to apply EPR to keep medicine prices at more affordable levels, they are encouraged to explore the cost-containment potential of this policy by taking appropriate methodological choices in the EPR design.

This study aimed to develop a tool to detect mildly lame cows by combining already existing data from sensors, AMSs, and routinely recorded animal and farm data. For this purpose, ten dairy farms were visited every 30–42 days from January 2020 to May 2021. Locomotion scores (LCS, from one for nonlame to five for severely lame) and body condition scores (BCS) were assessed at each visit, resulting in a total of 594 recorded animals. A questionnaire about farm management and husbandry was completed for the inclusion of potential risk factors. A lameness incidence risk (LCS ≥ 2) was calculated and varied widely between farms with a range from 27.07 to 65.52%. Moreover, the impact of lameness on the derived sensor parameters was inspected and showed no significant impact of lameness on total rumination time. Behavioral patterns for eating, low activity, and medium activity differed significantly in lame cows compared to nonlame cows. Finally, random forest models for lameness detection were fit by including different combinations of influencing variables. The results of these models were compared according to accuracy, sensitivity, and specificity. The best performing model achieved an accuracy of 0.75 with a sensitivity of 0.72 and specificity of 0.78. These approaches with routinely available data and sensor data can deliver promising results for early lameness detection in dairy cattle. While experimental automated lameness detection systems have achieved improved predictive results, the benefit of this presented approach is that it uses results from existing, routinely recorded, and therefore widely available data.

Physicians highlight that receiving the flu vaccine is critical, especially during the COVID-19 pandemic period. Younger groups’ flu vaccination coverage is very low, and this tendency is potentially related to a lower level of vaccine literacy and perceptions toward vaccination. This study investigated the relationship between flu vaccine literacy, health beliefs, and flu vaccinations (benefit, barrier, severity, and susceptibility) and their impact on perceived health status controlling for socioeconomic factors. It used the Health Belief Model andHealth Literacy Skills Framework with under/graduate students (N = 382) in Ohio, U.S. Path analyses were performed to examine the causal process using SPSS and Amos 23.0. Indicators of CFI, RMSEA, SRMR, and the chi-square/df of the path models were good–acceptable. Vaccine literacy directly impacted on health beliefs and vaccination. Susceptibility belief directly influenced perceived health status. The mediation effects of health beliefs (benefit, barrier) between vaccine literacy and vaccination were confirmed. The study highlights the need for healthcare providers and governments to work together to improve flu vaccine literacy and reduce negative perceptions toward vaccination among younger populations. Educational programs and official communication channels can be used to address concerns and provide accurate information about vaccines to increase flu vaccination rates and protect public health.

In diesem Beitrag befassen wir uns mit den Herausforderungen qualitativer Forschung im Kontext der Radikalisierungsforschung unter dem Eindruck einer jüngst erfolgten Beschlagnahme von Forschungsdaten und zeichnen die Entwicklungslinien und die Versicherheitlichung des Forschungsfeldes nach. Während sich Forschende oft im Spannungsfeld von Grundlagenforschung und Anwendungsbezug befinden, zeigt sich im Teilbereich der (De-)Radikalisierungsforschung die Tendenz zur Versicherheitlichung wie unter einem Brennglas – und stellt damit Forschende vor besondere Aufgaben; u.a. angesichts des fehlenden Zeugnisverweigerungsrechts. Wir greifen konkrete ethische und rechtliche Aspekte auf und bieten einen Leitfaden mit rechtlichen Fragen, den Forschende bearbeiten sollten, bevor sie ins Feld gehen.

Perilaku belajar yang konsisten bagi anak dengan mild intellectual disability (MID) merupakan hal penting yang perlu dilakukan untuk dapat memaksimalkan kapasitas kemampuan anak, terutama dalam aspek konseptual. Belajar perlu di lakukan tidak hanya di sekolah, namun juga di rumah. Salah satu cara yang dapat dilakukan untuk menerapkan perilaku belajar di rumah adalah menggunakan teknik antecedent control dan token economy. Teknik antecedent control dapat dilakukan untuk mengontrol stimulus anteseden, sedangkan teknik token economy diterapkan untuk memanipulasi konsekuensi yang didapatkan dari munculnya perilaku. Penelitian ini bertujuan untuk mengetahui efektivitas penggunaan teknik antecedent control dan token economy dalam meningkatkan durasi perilaku belajar anak usia sekolah dengan MID. Metode penelitian terbagi menjadi 3 tahap, yaitu tahap baseline, intervensi, dan follow up yang dilakukan dalam 25 sesi pertemuan. Hasil penelitian menunjukkan bahwa terdapat peningkatan durasi perilaku belajar dari sesi baseline hingga sesi follow up. Rata-rata durasi perilaku belajar anak dengan MID meningkat sebanyak 3 kali lebih lama, yaitu dari 22 menit menjadi 66,2 menit. Hasil penelitian membuktikan bahwa teknik antecedent control dan token economy cukup efektif dalam meningkatkan durasi perilaku belajar di rumah pada anak usia sekolah dengan MID, yang juga berimplikasi pada pengembangan kemampuan pemahaman aspek konseptual. Dengan demikian, teknik tersebut dapat menjadi alternatif intervensi yang bisa diterapkan oleh orangtua terhadap anak MID di rumah untuk mengoptimalkan kapasitas yang mereka miliki.

Abstract Background Diabetes during pregnancy is an increasingly common metabolic disorder, associated with significantly increased risks for both mother and child. Aim of this study was to compare maternal and perinatal outcomes in women with pregestational (PDM) type 1 (T1DM), type 2 diabetes (T2DM), gestational diabetes mellitus (GDM) and compare these to pregnancies not complicated with diabetes. This study also evaluated a specifically organized care-model mostly involving specialist diabetes nurses. Methods Retrospective population-based records review 2009–2012. Rates of maternal (preeclampsia, pre-term delivery, cesarean section (CS)) and fetal outcomes (large for gestational age (LGA), macrosomia, congenital malformations/intrauterine death) were assessed and potential predisposing or contributing factors as maternal age, ethnicity, obesity, weight gain, parity, HbA1c levels, insulin types and doses. Results Among 280 pregnancies 48 were PDM, 97 GDM and 135 without diabetes. Within the group with diabetes, early-pregnancy BMI was higher (p = 0.0001), pregnancy weight gain lower (11.1 ± 6.7 kg vs 13.1 ± 7.1 kg, p = 0.005), more delivered preterm (p = 0.0001), by CS (p = 0.05), and had more LGA neonates (p = 0.06) than the group without diabetes. Among pregnancies with diabetes, GDM mothers gained less weight (9.9 kg vs 13.5 kg) (p = 0.006), and rates of CS (p = 0.03), preterm deliveries (p = 0.001) and LGA (p = 0.0001) were not increased compared to PDM; More T1DM infants were LGA, 60% vs. 27% in T2DM. In pregnancies with diabetes obesity, excessive weight gain and multiparity were associated with increased risk of LGA neonates, and mother’s type of diabetes and gestational week were associated with higher rates of CS. Conclusion Weight gain during pregnancy was lower in pregnancies with diabetes and prevalence of LGA, CS and preterm deliveries in GDM was not elevated, also for T2DM, except increased prevalence of LGA in T1DM that warrants increased clinical attention, indicating that this model of antenatal diabetes care may have contributed to improved maternal and fetal outcomes.
