Background: Different rodent species serve as natural intermediate hosts for carnivore tapeworm Taenia crassiceps. However, this cestode occasionally infects various dead-end hosts including humans and other primates and may cause serious pathological implications with potentially fatal outcome. In this paper, we present subcutaneous cysticercosis caused by T. crassiceps, found in a previously healthy 17-years-old male ring-tailed lemur (Lemur catta) in a Serbian Zoo., Case Presentation: The animal was presented to a veterinarian with a history of periarticular subcutaneous swelling in medial right knee region. After fine needle aspiration revealed cycticerci-like structures, a surgery was performed for complete extraction of the incapsulated multicystic mass containing numerous cysticerci. Collected material was sent for parasitological, histological and molecular analysis. One month after surgery, the lemur died due to respiratory failure unrelated to cysticercosis. Based on morphological features of large and small hooks and characteristic proliferation of cysticerci, a metacestode of T. crassiceps was identified, which was confirmed after sequencing of obtained amplicons and comparing them to the GenBank database., Conclusions: This is one of the few reported cases of T. crassiceps cysticercosis in a ring-tailed lemur, and the first one in Serbia. This endangered species seem to be more sensitive for T. crassiceps than other non-human primates, which represents serious conservation challenge for captive animals. Due to zoonotic nature of the parasite, challenging diagnosis, severity of the disease, difficult treatment and possible fatalities, high biosecurity measures are of particular importance, especially in endemic regions., (© 2023. Gastrointestinal helminth-microbiota associations are shaped by various ecological processes. The effect of the ecological context of the host on the bacterial microbiome and gastrointestinal helminth parasites has been tested in a number of ecosystems and experimentally. This study takes the important step to look at these two groups at the same time and to start to examine how these communities interact in a changing host environment. Fresh faecal samples (N = 335) from eight wild Eulemur populations were collected over 2 years across Madagascar. We used 16S ribosomal RNA gene sequencing to characterise the bacterial microbiota composition, and faecal flotation to isolate and morphologically identify nematode eggs. Infections with nematodes of the genera Callistoura and Lemuricola occurred in all lemur populations. Seasonality significantly contributed to the observed variation in microbiota composition, especially in the dry deciduous forest. Microbial richness and Lemuricola spp. infection prevalence were highest in a previously intensely logged site, whereas Callistoura spp. showed no such pattern. In addition, we observed significant correlations between gastrointestinal parasites and bacterial microbiota composition in these lemurs, with 0.4-0.7% of the variation in faecal bacterial microbiota composition being explained by helminth infections. With this study, we show effects of environmental conditions on gastrointestinal nematodes and bacterial interactions in wild lemurs and believe it is essential to consider the potential role of microbiome-parasite associations on the hosts' GI stability, health, and survival.

Documenting the natural diversity of eukaryotic organisms in the nonhuman primate (NHP) gut is important for understanding the evolution of the mammalian gut microbiome, its role in digestion, health and disease, and the consequences of anthropogenic change on primate biology and conservation. Despite the ecological significance of gut-associated eukaryotes, little is known about the factors that influence their assembly and diversity in mammals. In this study, we used an 18S rRNA gene fragment metabarcoding approach to assess the eukaryotic assemblage of 62 individuals representing 16 NHP species. We find that cercopithecoids, and especially the cercopithecines, have substantially higher alpha diversity than other NHP groups. Gut-associated protists and nematodes are widespread among NHPs, consistent with their ancient association with NHP hosts. However, we do not find a consistent signal of phylosymbiosis or host-species specificity. Rather, gut eukaryotes are only weakly structured by primate phylogeny with minimal signal from diet, in contrast to previous reports of NHP gut bacteria. The results of this study indicate that gut-associated eukaryotes offer different information than gut-associated bacteria and add to our understanding of the structure of the gut microbiome.

Lemurs are primate species that are endemic to Madagascar. At present, about 90% of lemur species are endangered, and 5 species are among the 25 most endangered primates worldwide. Health status is a major factor impacting the viability of wild populations of many endangered species including lemurs. Given this context, we analyzed the microbiome of 24 specimens of Haemaphysalis lemuris, the most common tick parasitizing lemurs in their native habitats. Ticks were collected from 6 lemur species and microbiomes analyzed using next-generation sequencing. Our results show that the H. lemuris microbiome is highly diverse, including over 500 taxa, 267 of which were identified to genus level. Analysis of the microbiome also shows that there is a distinct "host" (lemur species) component when explaining the differences among and between microbial communities of H. lemuris. This "host" component seems to overwhelm any "locality" (geographic origin of the sample) component. In addition to the microbiome data, targeted PCR was used to test for the presence of three pathogens recently detected in the blood of wild lemurs: Borrelia sp., Candidatus Neoehrlichia sp., and Babesia sp. Overall, the presence of DNA of Rickettsia spp., Bartonella spp., Francisella spp., and a Babesia sp., in H. lemuris, is consistent with the hypothesis that these ectoparasites may act as vector for these pathogens. Detecting interaction between species is notoriously difficult, and disentangling species associations in host-related gut communities is especially challenging. Nevertheless, due to contemporary methods, including metabarcoding and 16S sequencing, collecting observational data on community composition has become easier and much more common. We studied the previously collected datasets of intestinal bacterial microbiota and parasite compositions within longitudinally followed mouse lemurs by analysing the potential interactions with diversity metrics and novel joint species distribution modelling. Both methods showed statistical association between certain parasite species and bacterial microbiota composition. Unicellular Eimeria sp. had an effect on diversity of gut microbiota. The cestode Hymenolepis diminuta had negative associations with several bacterial orders, whereas closely related species Hymenolepis nana had positive associations with several bacterial orders. Our results reveal potential interactions between some, but not all, intestinal parasites and gut bacterial microbiota. Host variables contributed over half of the total variation explained with the model, and sex was the most important single host variable; especially with microbiota, there were sex-related differences in the community composition. This study shows how joint species distribution modelling can incorporate both within-host dynamics of several taxa and host characteristics to model potential interactions in intestinal community. The discovery and characterization of emerging tick-borne organisms are critical for global health initiatives to improve animal and human welfare (One Health). It is possible that unknown tick-borne organisms underlie a subset of undiagnosed illness in wildlife, domesticated species, and humans. Our study lends support to the One Health concept by highlighting the prevalence of three blood-borne organisms in wild lemurs living in close proximity to domesticated species and humans. Previously, our team identified three novel, presumably tick-borne, intravascular organisms, belonging to the genera Babesia, Borrelia, and Neoehrlichia, circulating in two of Madagascar's lemur species. Here, we extend our previous observation by developing a targeted molecular surveillance approach aimed at determining the prevalence of these organisms in lemurs. Using quantitative PCR, we provide Babesia, Borrelia, and Neoehrlichia prevalence data for 76 individuals comprising four lemur species located in eastern Madagascar. Our results indicate a high prevalence (96%) of Babesia across sampled individuals with lower prevalences for Neoehrlichia (36%) and Borrelia (14.5%). Amoebiasis, caused by Entamoeba histolytica, affects 50 million people worldwide, and results in 100,000 deaths annually. It is particularly prevalent in developing nations where poverty and poor sanitation contribute to contamination of food and water. E. histolytica is also a zoonotic protozoan parasite with the potential to infect non-human primates. Lemurs, primates endemic to Madagascar, are the most threatened mammalian group in the world due to habitat loss. As forests disappear, humans and lemurs come into more frequent contact, and the potential for E. histolytica to infect lemurs intensifies. Consequently, we screened 176 fecal samples from seven lemur species at eight sites in the rain forests of southeastern Madagascar for E. histolytica to determine if human proximity influenced lemur infection. Of samples examined, 4.0% (from three lemur species) were positive for E. histolytica. Of lemurs infected with E. histolytica, three (43%) exhibited diarrheal feces. Distance to human settlements explained the variation in E. histolytica infection seen in lemurs. These results provide the first evidence of E. histolytica in wild lemurs and highlight the need for additional work to better understand the eco-epidemiology of this potential threat to these species., (Copyright © 2017 Elsevier B.V. Genes of the major histocompatibility complex (MHC) play a central role in adaptive immune responses of vertebrates. They exhibit remarkable polymorphism, often crossing species boundaries with similar alleles or allelic motifs shared across species. This pattern may reflect parallel parasite-mediated selective pressures, either favouring the long maintenance of ancestral MHC allelic lineages across successive speciation events by balancing selection ("trans-species polymorphism"), or alternatively favouring the independent emergence of functionally similar alleles post-speciation via convergent evolution. Here, we investigate the origins of MHC similarity across several species of dwarf and mouse lemurs (Cheirogaleidae). We examined MHC class II variation in two highly polymorphic loci (DRB, DQB) and evaluated the overlap of gut-parasite communities in four sympatric lemurs. We tested for parasite-MHC associations across species to determine whether similar parasite pressures may select for similar MHC alleles in different species. Next, we integrated our MHC data with those previously obtained from other Cheirogaleidae to investigate the relative contribution of convergent evolution and co-ancestry to shared MHC polymorphism by contrasting patterns of codon usage at functional vs. neutral sites. Our results indicate that parasites shared across species may select for functionally similar MHC alleles, implying that the dynamics of MHC-parasite co-evolution should be envisaged at the community level. We further show that balancing selection maintaining trans-species polymorphism, rather than convergent evolution, is the primary mechanism explaining shared MHC sequence motifs between species that diverged up to 30 million years ago., (© 2017 John Wiley & Sons Ltd.)

Here we describe fatal pulmonary cysticercosis caused by Cysticercus longicollis, the larval stage of Taenia crassiceps in a 15-year-old female ring-tailed lemur (Lemur catta) from Sarajevo Zoo. After sudden death, the lemur was subjected to necropsy and large multicystic structure, subdivided with fibrous septa and filled with numerous translucent, oval to ellipsoid bladder-like cysts (cysticerci), almost completely replacing right lung lobe was observed. In addition, numerous free and encysted cysticerci were found in the thoracic cavity. Histopathology revealed connective tissue outlined cavities that compress lung parenchyma. Each cavity contained several thin walled cysticerci with single inverted protoscolex, one or more suckers and rostelum with two rows of hooks. In many of the cysticerci one or several exogenous buds of daughter cysticerci were observed. Based on morphology and microscopic appearance the parasite was identified as C. longicollis. Subsequent molecular analysis and sequencing confirmed presumptive diagnosis. The fox tapeworm Echinococcus multilocularis is the causative agent of alveolar echinococcosis, a severe zoonotic disease that may be fatal if untreated. A broad spectrum of mammalian species may be accidentally infected even in captivity. In April 2011, liver lesions due to E. multilocularis were observed during the necropsy of a captive-born nutria (Myocastor coypus) in a French wildlife park, leading to initiation of a study to survey the parasite's presence in the park. A comparable environmental contamination with fox's feces infected by E. multilocularis was reported inside (17.8%) and outside (20.6%) the park. E. multilocularis worms were found in the intestines of three of the five roaming foxes shot in the park. Coprological analyses of potential definitive hosts in captivity (fox, lynx, wildcat, genet, wolf, bear and raccoon) revealed infection in one Eurasian wolf. Voles trapped inside the park also had a high prevalence of 5.3%. After diagnosis of alveolar echinococcosis in a Lemur catta during necropsy, four other cases in L. catta were detected by a combination of ultrasound and serology. These animals were treated twice daily with albendazole. The systematic massive metacestode development and numerous protoscoleces in L. catta confirmed their particular sensitivity to E. multilocularis infection. The autochthonous origin of the infection in all the captive animals infected was genetically confirmed by EmsB microsatellite analysis. Tapeworms of the genus Echinococcus reside in the small intestine of a number of carnivorous species, predominantly canids. In enzootic areas, hydatidosis caused by taeniid metacestodes can present a significant problem in accidental intermediate hosts, including humans. Whereas the United Kingdom is currently considered free of Echinococcus multilocularis, Echinococcus granulosus sensu stricto (s.s.) and Echinococcus equinus are endemic in the UK and have been reported in a variety of captive mammals. The presentation of echinoccocosis in non-human primates widely parallels disease in humans, and public health concerns are related to the four genera, E. granulosus, E. multilocularis, Echinococcus vogeli and Echinococcus oligarthrus. In contrast, sporadic outbreaks and individual hydatid disease cases in non-human primates have been associated with several Echinococcus and Taenia species. Here we describe three fatal cases of cystic echinococcosis in two captive ring-tailed lemurs (Lemur catta) and one captive red-ruffed lemur (Varecia variegata rubra) and provide molecular tapeworm characterisation. To the best of the authors' knowledge, this includes the first report of Echinococcus ortleppi in a UK born ring-tailed lemur and provides the first in depth case reports of echinococcosis due to E. equinus in UK born ring-tailed and red ruffed lemurs with detailed clinical and pathological findings. Zoonotic diseases are a looming threat to global populations, and nearly 75% of emerging infectious diseases can spread among wildlife, domestic animals and humans. A 'One World, One Health' perspective offers us an ideal framework for understanding and potentially mitigating the spread of zoonoses, and the island of Madagascar serves as a natural laboratory for conducting these studies. Rapid habitat degradation and climate change on the island are contributing to more frequent contact among humans, livestock and wildlife, increasing the potential for pathogen spillover events. Given Madagascar's long geographical isolation, coupled with recent and repeated introduction of agricultural and invasive species, it is likely that a number of circulating pathogens remain uncharacterized in lemur populations. Thus, it is imperative that new approaches be implemented for de novo pathogen discovery. To this end, we used non-targeted deep sequencing of blood transcriptomes from two species of critically endangered wild lemurs (Indri indri and Propithecus diadema) to characterize blood-borne pathogens. Our results show several undescribed vector-borne parasites circulating within lemurs, some of which may cause disease in wildlife, livestock and humans. Baylisascaris procyonis is a well-known ascaridoid nematode that causes larva migrans in humans and many other animal species. The North American raccoon (Procyon lotor) is the definitive host, which has been successfully introduced in the past decades to other geographical regions around the world. Two white-headed lemurs (Eulemuralbifrons) from a Zoological Park in Lugo, Spain, developed severe neurological signs within a brief period after being transferred from exhibit and placed in close contact with three captive raccoons from the same zoo. One lemur was euthanized due to the severity of disease progression and histopathology revealed granulomatous inflammation and ascaridoid larvae in kidneys, lung, spleen and brain. Larvae were identified as B. procyonis larvae by real time PCR. In light of the results, the cage mate with similar neurological signs was put on an albendazole treatment regimen adapted from a human pediatric protocol. The aggressive anthelmintic treatment likely contributed to the arrest of clinical signs and recovery of some motor skills. Background: Marker gene studies often use short amplicons spanning one or more hypervariable regions from an rRNA gene to interrogate the community structure of uncultured environmental samples. Target regions are chosen for their discriminatory power, but the limited phylogenetic signal of short high-throughput sequencing reads precludes accurate phylogenetic analysis. This is particularly unfortunate in the study of microscopic eukaryotes where horizontal gene flow is limited and the rRNA gene is expected to accurately reflect the species phylogeny. A promising alternative to full phylogenetic analysis is phylogenetic placement, where a reference phylogeny is inferred using the complete marker gene and iteratively extended with the short sequences from a metagenetic sample under study., Results: Based on the phylogenetic placement approach we built Séance, a community analysis pipeline focused on the analysis of 18S marker gene data. Séance combines the alignment extension and phylogenetic placement capabilities of the Pagan multiple sequence alignment program with a suite of tools to preprocess, cluster and visualise datasets composed of many samples. We showcase Séance by analysing 454 data from a longitudinal study of intestinal parasite communities in wild rufous mouse lemurs (Microcebus rufus) as well as in simulation. We demonstrate both improved OTU picking at higher levels of sequence similarity for 454 data and show the accuracy of phylogenetic placement to be comparable to maximum likelihood methods for lower numbers of taxa., Conclusions: Séance is an open source community analysis pipeline that provides reference-based phylogenetic analysis for rRNA marker gene studies. Whilst in this article we focus on studying nematodes using the 18S marker gene, the concepts are generic and reference data for alternative marker genes can be easily created. Echinococcus multilocularis is a tapeworm responsible in its larval stage for alveolar echinococcosis, a disease which is lethal when left untreated. Multivesiculated parasitic lesions in the liver were diagnosed at necropsy in a captive-born nutria (Myocastor coypus) and in a ring-tailed lemur (Lemur catta) which had been in a French zoo for 16months. Molecular analyses confirmed the diagnosis of E. multilocularis obtained by histological analyses. These were the first cases of infection by E. multilocularis reported in lemurs in Europe, and the first case in nutria in European enclosures. Lemurs are confirmed to be particularly sensitive to E. multilocularis with a massive infection. In both cases, the infection appears to have been contracted in the zoo indirectly via environmental contamination by feces from roaming foxes. Due to the large endemic area for E. multilocularis, the increasing prevalence in foxes in France, and an increase in awareness of the disease, other cases of infection in captive animals will probably be recorded in France in the coming years., (© 2013.)

The ability of black and brown lemurs (Eulemur macaco and Eulemur fulvus) to make inferences about hidden food was tested using the same paradigm as in Call's (J Comp Psycol 118:232-241, 2004) cup task experiment. When provided with either visual or auditory information about the content of two boxes (one empty, one baited), lemurs performed better in the auditory condition than in the visual one. When provided with visual or auditory information only about the empty box, one subject out of four was above chance in the auditory condition, implying inferential reasoning. No subject was successful in the visual condition. This study reveals that (1) lemurs are capable of inferential reasoning by exclusion and (2) lemurs make better use of auditory than visual information. The results are compared with the performances recorded in apes and monkeys under the same paradigm.

Objective: To observe morphology of Auchenacantha galeopteri (A. galeopteri), Auchenacantha spinosa (A. spinosa) and Auchenacantha parva (A. parva) from Sukabumi and Ujung Kulon, Indonesia using scanning electron microscopy (SEM)., Methods: Specimens for light microscopy examination were fixed with warm 70% alcohol, cleared and mounted in lactophenol for wet mounting. Drawings were made with the aid of a drawing tube attached to a Nikon compound microscope. Specimens for SEM examination were processed according to Bozzola. Measurements were given in micrometers (µm) as the average of findings, followed by the range in parentheses, unless otherwise stated., Results: The measurements of Auchenacantha spp are same with those of previously described. The striations of male and female A. galeopteri are widen towards outside and wavy. The mouth of female with 6 lips, each of them is wider at base than distal end in A. galeopteri and rectangular in A. spinosa. Both species have dentiform protrusion from inner surface of lips, such structure absent in A. parva, but the lips with transverse festoon like pattern at anterior end of each lip., Conclusions: Using SEM, the lip and the striation pattern of three species of Auchenacantha can be clearly distinguished. Sukabumi and Ujung Kulon are new locality of A. galeopteri and A. spinosa, and A. parva is the new record in Indonesia.

Several recent studies of animals in their natural surroundings found evidence for effects of certain major histocompatibility complex (MHC) immune gene alleles on the parasite load. However, in multi-infected individuals the particular selection pressure exerted by specific parasites has rarely been explored. In this study we took advantage of the parasitological and genetic data of two previously investigated Malagasy lemur species (Cheirogaleus medius and Microcebus murinus). We investigated whether the two sympatric and ecologically similar primates are infected by similar parasite species and explored if certain parasites are associated with particular MHC alleles. Our study revealed that most of the parasite egg morphotypes were found in both hosts. In each lemur species we identified one MHC allele which was positively associated with Ascaris-infection. Interestingly, these MHC alleles were very similar to each other but differed from all other investigated MHC alleles in an amino acid substitution in a putative functional important antigen binding site. Thus, our study gives first intriguing evidence for a direct connection between certain antigen binding sites of MHC molecules with a particular parasite in two wild primate populations. Parasites and infectious diseases represent ecological forces shaping animal social evolution. Although empirical studies supporting this link abound in various vertebrate orders, both the study of the dynamics and impact of parasite infections and infectious diseases in strepsirrhine primates have received little empirical attention. We conducted a longitudinal parasitological study on four groups of wild red-fronted lemurs (Eulemur fulvus rufus) at Kirindy Forest, Madagascar, during two field seasons in consecutive years to investigate i) the degree of gastrointestinal parasite infection on population and individual levels and ii) factors potentially determining individual infection risk. Using a comprehensive dataset with multiple individually assignable parasite samples as well as information on age, sex, group size, social rank, and endocrine status (fecal androgen and glucocorticoid), we examined parasite infection patterns and host traits that may affect individual infection risk. In addition, we examined whether parasite infection affects mating and reproductive success. Our results indicated high variability in parasite infection on individual and population levels. Time of year and group size was important determinants of variability in parasite infection. Variation in hormone levels was also associated with parasite species richness and parasite infection intensity. Differences in parasite infection between years indicate a potential immune-enhancing function of steroid hormones on nematode infections, which has not been reported before from other vertebrates studied under natural conditions. We investigated two mitochondrial genes (cytb and cox1), one plastid gene (tufA), and one nuclear gene (ldh) in blood samples from 12 chimpanzees and two gorillas from Cameroon and one lemur from Madagascar. One gorilla sample is related to Plasmodium falciparum, thus confirming the recently reported presence in gorillas of this parasite. The second gorilla sample is more similar to the recently defined Plasmodium gaboni than to the P. falciparum-Plasmodium reichenowi clade, but distinct from both. Two chimpanzee samples are P. falciparum. A third sample is P. reichenowi and two others are P. gaboni. The other chimpanzee samples are different from those in the ape clade: two are Plasmodium ovale, and one is Plasmodium malariae. That is, we have found three human Plasmodium parasites in chimpanzees. Four chimpanzee samples were mixed: one species was P. reichenowi; the other species was P. gaboni in three samples and P. ovale in the fourth sample. The lemur sample, provisionally named Plasmodium malagasi, is a sister lineage to the large cluster of primate parasites that does not include P. falciparum or ape parasites, suggesting that the falciparum + ape parasite cluster (Laverania clade) may have evolved from a parasite present in hosts not ancestral to the primates. If malignant malaria were eradicated from human populations, chimpanzees, in addition to gorillas, might serve as a reservoir for P. falciparum.

Although parasites are important regulatory factors in animal populations, basic knowledge on their fauna in many vertebrate taxa is lacking. In particular, parasite infections of primate species have gained little attention. Here, I present data on the gastro-intestinal fauna of a population of wild red-fronted lemurs ( Eulemur fulvus rufus; Primates: Lemuriformes) monitored over a total of 8 mo during 2 consecutive field seasons in 2006 and 2007 in Kirindy Forest, western Madagascar. Using fecal samples for parasite analyses, I identified 10 parasite species, including 6 nematodes (Lemuricola vauceli, Trichuris sp., 2 species of Callistoura, 1 trichostrongylid, and 1 strongyloid), 1 anoplocephalid cestode, a dicrocoeliid trematode, as well as 2 protozoans (Entamoeba sp. and Balantidium coli). The population in Kirindy Forest had the highest prevalence and number of parasite species ever recorded for species of lemurs. Additionally, prevalence of some parasite species differed between the social groups studied. These findings lead to 2 conclusions. First, it is important to extend a parasitological study to several social groups of a host population, since groups may differ in parasite fauna as a result of minor microclimatic or habitat parameters, and, second, short-term assessments of lemur health might underestimate the real parasite burden.

Complete health assessments were performed on 22 adult red ruffed lemurs (Varecia rubra), comprising nine males and 13 females, found within the Masoala National Park in northeast Madagascar. Each animal was anesthetized using tiletamine and zolazepam and underwent a thorough physical examination, including measurement of its weight and vital signs; blood collection for hematology, plasma total protein concentration, serum chemistries, fat-soluble vitamins, trace minerals, assessment of iron metabolism, toxoplasmosis serology, viral serologies, and examination for hemoparasites; fecal collection for bacterial culture and parasite examination; and collection of a representative number of any ectoparasites. Comparison of blood values with those of captive lemurs demonstrated a number of significant differences thought to be associated with physiologic state (e.g., reproductive stage and stress), hydration, and diet. There was no evidence of serious infectious diseases, and hemoparasites were not detected. The enteric flora appeared unremarkable; however, results may have been skewed toward more cold-tolerant bacteria. The fecal parasite burden was low. Lemurostrongylus spp. was identified in two of the lemurs, and there were moderate numbers of Laelapidae mites present on approximately one third of the lemurs. This study demonstrated the substantial amount of data that can be collected from free-ranging populations, considered invaluable in the management of captive populations, in reducing the incidence of captivity-related diseases, and in the risk assessment associated with reintroduction programs.

Complete physical examinations and biomedical sample collection were performed on 70 free-ranging ring-tailed lemurs (Lemur catta) from three different habitats in the Beza Mahfaly Special Reserve (BMSR), in southern Madagascar, to assess the impact of humans and habitat on lemur health. Lemurs were chemically immobilized with ketamine and diazepam administered via blow darts for concurrent biomedical, morphometric, and behavioral studies. Subsets of the animals had blood analyzed for hematology, serum chemistry, micronutrients, fat-soluble vitamins (vitamins A, D, and E), measures of iron metabolism, and polymerase chain reaction assays (PCR) for Toxoplasma gondii, Hemoplasma spp., Bartonella spp., Ehrlichia spp., Anaplasma phagocytophilum, and Neorickettsia risticii. Results were compared on the basis of gender and the habitats at the study site: reserve (intact gallery forest), degraded (human inhabited and altered), and marginal (dry didieracea forest with heavy grazing and tree cutting). Levels of vitamin D, triglycerides, and cholesterol, and measures of iron metabolism for BMSR lemurs were greater than those previously reported for a free-ranging lemur population (Tsimanampetsotsa Strict Nature Reserve, Madagascar) with less access to foods of anthropogenic origin. BMSR ring-tailed lemurs from a habitat with less water (marginal) had higher sodium (P = 0.051), chloride (P = 0.045), osmolality (P = 0.010), and amylase (P = 0.05) levels than lemurs from other BMSR habitats, suggesting that these lemurs were less hydrated. Vitamin D levels of male lemurs were higher (P = 0.011) than those of females at BMSR, possibly because of differences in sunning behavior or differential selection of food items. The biological significance is uncertain for other parameters with statistically significant differences. All samples tested (n = 20) were negative for the pathogens tested using PCR assays. Continued concurrent biomedical and ecological research is needed at BMSR to confirm these results and determine their association with population mortality and fecundity rates.

In the current study, we determined the seroprevalence of Toxoplasma gondii, Sarcocystis neurona, and Encephalitozoon cuniculi in three species of lemurs from St. Catherines Island, Georgia. Serum samples were tested from 52 ring-tailed lemurs (Lemur catta), six blue-eyed black lemurs (Eulemur macaco flavifrons), and four black and white ruffed lemurs (Varecia variegata variegata) using an agglutination assay. Three ring-tailed lemurs (5.8%) were positive for T. gondii (titer of 1:50); one ring-tailed lemur (1.9%) and one black and white ruffed lemur (25%) were positive for S. neurona (titers of 1:1000); and one ring-tailed lemur (1.9%) was positive for E. cuniculi (titer of 1:400). All blue-eyed black lemurs were negative for antibodies to T. gondii, S. neurona, and E. cuniculi. This is the first detection of antibodies to T. gondii in ring-tailed lemurs and antibodies to S. neurona and E. cuniculi in any species of prosimian.

Complete medical evaluations were performed on 25 wild black lemurs (Eulemur macaco macaco) in Lokobe Reserve, northwestern Madagascar. Each animal received a complete physical examination. Weight, body temperature, heart rate, and respiratory rate were recorded. Blood samples were collected for complete blood cell count, differential white blood cell count, hemoparasite examination, serum biochemical profile, fat-soluble vitamin analysis, trace mineral analysis, and Toxoplasma gondii and viral serology. Fecal samples were collected for bacterial culture and endoparasite examination. Ectoparasites were collected and identified. Values were compared to established ranges from captive black lemurs in North American zoos. Significant differences exist between captive and wild animals for total white blood cell count, segmented neutrophil count, band neutrophil count, eosinophil count, monocyte count, and basophil count, and for serum biochemistry values of aspartate aminotransferase, alanine aminotransferase, total bilirubin, serum alkaline phosphatase, total protein, creatinine, phosphorus, calcium, sodium, potassium, and creatine phosphokinase. One animal had a positive Toxoplasma titer. Detected endoparasites were identified as Lemuricola and Callistroura. Two ectoparasite mites were identified, one within the Psoroptes genus and one within the Laelapidae family. Enteric bacterial flora included Enterococcus sp., Staphylococcus sp., Escherichia coli, Streptococcus sp., Klebsiella ozaenae and Bacillus cereus.

Lemurs on St. Catherines Island, Georgia were tested for Trypanosoma cruzi infection to develop a better understanding of the epizootiology of the parasite in nonhuman primates in the southeastern United States. Fifty-six ring-tailed (Lemur catta), blue-eyed black (Eulemur macaco flavifrons), and black-and-white ruffed (Varecia variegata variegata) lemurs were tested by hemoculture and serology to determine the prevalence of T. cruzi in the population. Of those tested 3 (5%) were identified as culture positive and 25 (44.6%) as seropositive. When hemoculture results were compared with those from a similar study performed in 1997, prevalence remained unchanged. Genetic characterization of the 3 culture isolates indicated they belong to the T. cruzi IIa group, which is identical to strains previously isolated from raccoons on the island. Despite the occurrence of T. cruzi in the population, there was no evidence that the health of the lemurs was compromised as a result of infection. Based upon prevalence and available breeding records we speculate that both vertical and vector-mediated transmission play significant roles in the epidemiology of T. cruzi on the island. This also represents the first report of autochthonous infection in blue-eyed black and black-and-white ruffed lemurs.

Complete medical examinations were performed on 20 wild Decken's sifaka (Propithecus verreauxi deckeni) and 20 wild red-fronted brown lemurs (Eulemurfulvus rufus) from western Madagascar. Each animal received a complete physical examination, and weight, body temperature, heart rate, and respiratory rate were recorded and ectoparasites collected. Blood samples were collected for complete blood cell count, differential white blood cell count, hemoparasite examination, serum biochemical profile, fat-soluble vitamin analysis, trace mineral analysis, and toxoplasmosis and viral serology. Fecal samples were collected for bacterial culture and endoparasite examination. Significant differences exist between the species for serum chemistry values for creatine phosphokinase, glucose, total protein, albumin, globulin, blood urea nitrogen, creatinine, phosphorus, potassium, and chloride; for fat-soluble vitamins 25-hydroxycholecalciferol, retinol, retinyl palmitate, gamma-tocopherol, alpha-tocopherol, and lutein + zeaxanthin; and serum copper. Parasites detected include Lemurostrongylus spp., Lemuricola spp., Trichurus spp., and ectoparasites Haemaphysalis lemuris, Psoroptes, and mites identified to the family Laelapidae. Enteric bacterial flora included Escherichia coli, Citrobacter ssp., Enterococcus faecalis, Bacillus cereus, Klebsiella ozaenae, Acinetobacter lwofii, and Enterobacter amnigenus.

A 3-yr-old secundiparous female ring-tailed lemur presented to the Auburn University Small Animal Clinic with signs of dyspnea, lethargy, and anorexia. The animal died before she could be examined, and a full necropsy was immediately performed. Provisional necropsy findings included moderate pneumonia and hepatopathy. Acute interstitial pneumonia and focal hepatocellular necrosis were confirmed histologically. Lung impression smears, histopathology, electron microscopy, immunohistochemistry, and tissue culture isolation resulted in a diagnosis of acute disseminated Toxoplasma gondii infection, which was confirmed by polymerase chain reaction. The isolate of T. gondii was avirulent for mice and was named AU Tgl and genetically is type II. The source of the infection remains unclear, but speculation suggests contaminated fruit or blackbirds (Passeriformes: Icteridae) acting as transport hosts for oocysts from nondomestic felids and feral cats on the property.

Blood was collected for hematology, determination of plasma total protein concentration, serum chemistry, determination of fat-soluble vitamin and trace mineral concentrations, assessment of iron metabolism, toxoplasmosis and viral serologies, and examination for parasites. Feces were collected for bacterial culture and parasite examination, and representative numbers of ectoparasites were collected. Blood values differed significantly in a number of ways from values in captive lemurs, possibly associated with recent food consumption, hydration, and diet. There was no evidence of serious infectious disease and no hemoparasites. The enteric flora appeared unremarkable, although results may have been skewed toward cold-tolerant bacteria. The fecal parasite burden was low, but there were large numbers of ectoparasites (mites) present. Our study demonstrated that a substantial amount of data can be collected from free-ranging populations, information that is invaluable in the management of captive populations, particularly with regard to disease, and in assessing risks associated with reintroduction programs.

Srivastavanema cynocephali n. sp., a parasite of Cynocephalus variegatus (Dermoptera) from Indonesia, is described. This species is closely related to S. yapi Durette-Desset & Lim Boo Liat, 1975, in the characters of the synlophe at the mid-body and in the shape of the caudal bursa, but it is distinguished by the pattern of the synlophe in the posterior part of the body, very long, thin rays 2 and 3, a less developed genital cone, the absence of a membrane between left and right rays 6, and longer spicules. Its morphology is slightly less specialised than the four previously known species of the genus, which are all parasites of petauristines. The genus Srivastavanema (Singh, 1962) has a systematic position between the Heligmonellinae and the Brevistriatinae. Therefore, it may be a parasite of the Dermoptera which could have secondarily evolved within the Petauristinae. The Dermoptera may have played an important role in the evolution of the Heligmonellinae.

Free-ranging Old World primates released on St. Catherine's Island, Georgia (USA), were tested for infection with Trypanosoma cruzi as part of a study of the epizootiology of sylvatic T. cruzi in the southeastern USA. The parasite was observed in liver infusion tryptose medium cultures of blood from seven of 11 lion-tailed macaques (Macaca silenus) and one of 19 ring-tailed lemurs (Lemur catta). Cultures of blood from 10 black and white ruffed lemurs (Varecia variegata) were all negative. Analysis of small subunit ribosomal RNA gene polymorphisms detected using polymerase chain reaction techniques indicates that the parasites isolated from both the lion-tailed macaques and the ring-tailed lemur are probably the same as T. cruzi parasites isolated from raccoons (Procyon lotor) trapped on St. Catherine's Island and other locations in the southeastern USA. Foraging lion-tailed macaques were observed to handle and partially consume specimens of Triatoma sanguisuga, the triatomine bug thought to be a vector of T. cruzi in the southeastern USA. Oral transmission of the parasite may have occurred as a result of this behavior.

A new species, Blastocystis lemuri sp. n., was found in the Lemur catta. Size of blastocysts in a culture is 10.25-35.25 mkm, the number of nuclei varies from 1 to 2. The optimum temperature for cultivation is +39 degrees.

Classical and morphometric analyses were conducted on several samples of oxyurid nematodes parasitic in strepsirrhine primates: Cheirogaleus major, Galago senegalensis, and Microcebus murinus. A diagnosis of Lemuricola contagiosus Chabaud & Petter, 1959 from C. major is given using syntypes, from which a lectotype is selected. The rest of the specimens were compared to L. contagiosus. From measurements taken from each sex, an initial Principal Component Analysis (PCA) was performed on log-transformed data and a second PCA was performed on log-shape ratios after elimination of the isometric differences. The parasites collected from M. murinus, which can be differentiated from the closely related L. contagiosus by both standard morphological characters and morphometric variables, are described as Lemuricola microcebi n. sp. The parasites collected from Galago are morphologically very similar to L. microcebi n. sp. The most probable explanation for this resemblance is considered to be a transfer from Microcebus to Galago but, due to the small numbers of individuals in our samples, it is impossible to eliminate the possibility that the parasites of Galago belong to an undescribed species. Until more material or information can be obtained, it is proposed to refer to the parasites from Galago as Lemuricola sp.

Lemur macaco macaco from Ambanja region was found polyparasitized by four different species of Plasmodium: --Plasmodium coulangesi recently described by lepers et al. (1989). --P. bucki n. sp.: its main differential characterisitics are the large numvber (32) of merozoites produced in mature schizonts and the stippling, resembling Maurer's dots, in an hypertrophied pinkish erythrocyte. --P. percygarnhami n. sp. (= P. girardi sensu Uilenberg, 1970 pro parte and sensu Garnham et Uilenberg, 1975 pro parte) producing 20 merozoites in mature schizonts and developing inside a deformed corpuscle (holly leaf-shaped or sometimes sea-urchin-shaped) which may also become decolourized when parasitized by older stages. --(?) Plasmodium lemuris: gametocytes are very large (11 microns x 7 micron); the parasitized erythrocyte is much hypertrophied (+/- 10 microns), distored and of a pinkish colour; one schizont only, possibly exo-erythrocytic, was found. The authors hypothesized this parasite to be a Haemoproteid. The analysis of published data led the authors to make the following modifications to the nomenclature previously established: --Plasmodium girardi Buck et al., 1952, sensu Garnham, 1966, sensu Uilenberg, 1970 parte and sensu Garnham and Uilenberg, 1975 pro parte, is refered to as Plasmodium sp., for stages developing in the blood of Lemur fulvus fulvus. The taxon P. percygarnhami is to be employed for stages developing in L. m. macaco and P. girardi Buck et al., 1952 for those developing in Lemur fulvus rufus. --Plasmodium foleyi Buck et al., 1952, sensu Garnham and Uilenberg, 1975 in L. f. fulvus is named Plasmodium uilenbergi n. sp., P. folleyi being a parasite of L.f. rufus. Excluding P. lemuris which probably does not belong to the genus Plasmodium, the morphological analysis led to individualize 7 species, in the three species of Lemurs studied. A phenomenon of "vicariance" thus appears, similar to what is known for the african Rodent Plasmodia, but with a more pronounced speciation. The vicariant species form a pair constituted of: --on the one hand a small species developing in a red blood cell of normal size, P. girardi in L.f. rufus, P. sp. in L.f. fulvus, P. percygarnhami, with also, P. coulangesi, in L. m. macaco; --on the other hand a large species determining an hypertrophy of the erythrocyte, P. foleyi in L.f. rufus, P. uilenbergi in L.f. fulvus and P. bucki in L.m. macaco.

Forests of Ankarana limestone massif in northern Madagascar support one of the largest and least disturbed populations of Crowned Lemurs, Lemur coronatus. This paper reports a preliminary study of the ecology of this species in the Ankarana Special Reserve conducted at the end of the dry season in 1986, with additional information collected a year later. Crowned Lemurs occur in very high densities in the semi-deciduous canopy forest and this probably represents a dry season refuge for the species. They also use more open habitats, including sparsely vegetated limestone and degraded forest. Sanford's Lemur, Lemur fulvus sanfordi, also inhabits the Ankarana forests but is most abundant in degraded habitats. Crowned and Sanford's Lemurs had similar patterns of activity, which included nocturnal travelling and feeding bouts. Crowned Lemurs proved to be unusual among Lemur species in displaying low spatial troop cohesion and a lack of obvious troop hierarchy. Stronglyoides-like enteric helminths infested about one third of Crowned Lemurs but were apparently not causing disease. Crowned Lemurs fall prey to the Fosa, Cryptoprocta ferox, and the young possibly also to the largest raptors. A total of seven living lemur species (including the very rare Propithecus diadema perrieri and Daubentonia madagascariensis) were confirmed at Ankarana by the authors, and three further species have been reported by other observers. In addition to these ten extant lemurs, four subfossil species have been discovered: three of them (Hapalemur simus, Palaeopropithecus and Mesopropithecus) by the authors. The possibility that all 14 lemurs were once sympatric is discussed. For the present, the lemurs of Ankarana are protected from hunting by local taboo. Nevertheless they are under severe threat from habitat destruction, despite Ankarana's Special Reserve status. Given the very restricted distributions of Crowned and Sanford's Lemurs, both must be considered as threatened with extinction.

A review of the Oxyurids parasites of Primates shows that most of the species described in the Malagasy Lemurs and in the Old World Monkeys are not single species, but couples of sister-species. The sister-species of each couple may be differentiated sometimes only by the characteristics of male spicule (less evoluated species), sometimes by other characteristics (cephalic structures, length of oesophagus, etc., in more evoluated species). In the first instance only males can be identified, in the second instance females may also be identified. In the Human parasites, which are very closely related to those of the Old World Monkeys, two types of spicules were described and we consider that each type belongs to a different species: Enterobius vermicularis (L., 1758) for the larger spicule (100 to 122 micrometers), and Enterobius gregorii n. sp. for the shorter spicule (70 to 80 micrometers).

A survey of 17 species of non-human captive primates in Quebec, Canada, revealed that 40% of the animals were infected with the protozoan Entamoeba histolytica. Isoenzyme analysis of lysates prepared from cultured amebic isolates showed them to belong to either a zymodeme III or VIII, and therefore, characteristic of non-pathogenic strains of E. histolytica from humans. These isolates were also of low virulence in gerbil ceca. Serum samples from infected and uninfected primates had negative titres for E. histolytica and all the animals appeared to be healthy.

The splenectomy of a female Lemur macaco macaco, showing a scanty blood infection by Haemosporidians and a Trypanosome was followed by a dramatic increase of the rate of intra-erythrocytic parasitaemia. Three species of Plasmodium where then identified. The first species was unknown; it is described in this paper and named Plasmodium coulangesi n. sp. It is characterised by--the low (6). constant number of merozoites in mature schizonts,--the disposition of the pigment, well apart from the parasitic mass to which it is linked by a tiny wisp of cytoplasm,--the normal host erythrocyte, the shape, size and colour of which are unaltered. The second species was provisionaly designated as Plasmodium sp.; it was previously seen in the same host by Garnham and Uilenberg in 1975 and designated by these authors as Plasmodium girardi Buck, Coudurier et Quesnel, 1952. Only gametocytes of the last species were found; they are similar to those of Plasmodium lemuris Huff and Hoogstraal, 1963.

A morphological study of traumatic insemination in the genera Auchenacantha, a parasite of Dermoptera, and Passalurus, a parasite of Rabbits, revealed that the same phenomenon appeared twice in distinct lineages. In the Auchenacantha, adult females are inseminated in the vagina vera modified into a spermatic pouch. In the Passalurus insemination occurs in the ventral hypodermal field. Adult males probably mate with the female larval, stage four.
