- Abstract
BACKGROUND: TRK fusions are detected in less than 3% of CNS tumors. Given their rarity, there are limited data on the clinical course of these patients. METHOD(S): We contacted 166 oncology centers worldwide to retrieve data on patients with TRK fusion-driven CNS tumors. Data extracted included demographics, histopathology, NTRK gene fusion, treatment modalities and outcomes. Patients less than 18 years of age at diagnosis were included in this analysis. RESULT(S): Seventy-three pediatric patients with TRK fusion-driven primary CNS tumors were identified. Median age at diagnosis was 2.4 years (range 0.0-17.8) and 60.2 % were male. NTRK2 gene fusions were found in 37 patients (50.7%), NTRK1 and NTRK3 aberrations were detected in 19 (26.0%) and 17 (23.3%), respectively. Tumor types included 38 high-grade gliomas (HGG; 52.1%), 20 low-grade gliomas (LGG; 27.4%), 4 embryonal tumors (5.5%) and 11 others (15.1%). Median follow-up was 46.5 months (range 3-226). During the course of their disease, a total of 62 (84.9%) patients underwent surgery with a treatment intent, 50 (68.5%) patients received chemotherapy, 35 (47.9%) patients received radiation therapy, while 34 (46.6%) patients received NTRK inhibitors (3 as first line treatment). Twenty-four (32.9%) had no progression including 9 LGG (45%) and 9 HGG (23.6%). At last follow-up, only one (5.6%-18 evaluable) patient with LGG died compared to 11 with HGG (35.5%-31 evaluable). For LGG the median progression-free survival (PFS) after the first line of treatment was 17 months (95% CI: 0.0-35.5) and median overall survival (OS) was not reached. For patients with HGG the median PFS was 30 months (95% CI: 11.9-48.1) and median OS was 182 months (95% CI 20.2-343.8). CONCLUSION(S): We report the largest cohort of pediatric patients with TRK fusion-driven primary CNS tumors. These results will help us to better understand clinical evolution and compare outcomes with ongoing clinical trials.

Exceptional preservation of aragonite in a circa 3.3 billion year old microbial mat from the Barberton greenstone belt, South Africa Frances Westall, Barbara Cavalazzi, Laurence Lemelle, Yves Marrocchi, Jean-Noël Rouzaud, Alexandre Simionovici, Murielle Salomé, Smail Mostefaoui, Caroline Andreazza, Frédéric Foucher, Jan Toporski, Andrea Jauss, Volker Thiel, Axel Hofmann, Anders Meibom, François Robert Aragonite occurs as a biologically-formed mineral precipitate within modern calcifying microbial mats. It is, however, rarely preserved in the geological record because, as one of the least stable polymorphs of calcium carbonate, it readily converts to calcite in present environmental conditions at the Earth's surface. In an in situ investigation at the micro- to nanometer-scale, we show that 5-10 nm sized nanocrystals of aragonite are preserved within the organic framework of a partially calcified microbial mat from the ~ 3.3 billion year-old Josefsdal Chert in the Barberton greenstone belt, South Africa. Transformation of the aragonite to calcite was blocked by a combination of chemical inhibitors within the crystal lattice, organic molecules coating the nanocrystals and, in particular, to the precocious permeation of the mat by hydrothermal silica. Apart from its exceptional preservation for 3.3 billion years, the identification of aragonite in the Josefsdal microbial mat is the earliest evidence for in situ calcification of a microbial mat. Furthermore, the indications of associated sulphur-reducing bacteria (SRB) activity with calcification strongly support a photosynthetic origin for the mat. This is the most direct evidence for photosynthesis in early Archaean rocks.

- Abstract
Seven particles captured by the Stardust Interstellar Dust Collector and returned to Earth for laboratory analysis have features consistent with an origin in the contemporary interstellar dust stream. More than 50 spacecraft debris particles were also identified. The interstellar dust candidates are readily distinguished from debris impacts on the basis of elemental composition and/or impact trajectory. The seven candidate interstellar particles are diverse in elemental composition, crystal structure, and size. The presence of crystalline grains and multiple iron-bearing phases, including sulfide, in some particles indicates that individual interstellar particles diverge from any one representative model of interstellar dust inferred from astronomical observations and theory.

Microorganisms were searched for among the complex microstructures observed on the surface of a fragment of the Tatahouine meteorite inherited from the Tunisian soil in which they were buried. In this view, the chemical compositions, particularly the nitrogen, phosphorus, and sulphur compositions, including the sulphur speciation, were investigated using scanning electron microscopy (SEM), electron probe microanalysis (EPMA) mapping, and scanning X-ray microscopy (SXM). A few 2-μm-thick filaments, partly covered by patches of calcite ensuring they were not deposited by a laboratory contamination, were observed by SEM. The EPMA maps show that the portions free of calcite of the filaments have low but constant contents of nitrogen, sulphur, and phosphorus. The SXM maps were recorded at 2473.5, 2478, and 2482.2 eV, which are respectively characteristic for amino acid linked sulphur, sulphite (SO32−), and sulphate (SO42−). The portions of the filaments detected by EPMA are also those that are enriched in amino acid linked sulphur. The calculated (N/S) elemental ratio is consistent with the one of the dehydrated Escherichia coli matter, contrary to the much lower (P/S) elemental ratio. In living cells, the bulk N and S elements are mainly located in large polymers by covalent bonds, whereas a significant amount of P belongs to small and reactive molecules. We thus can propose that the observed microstructures are dehydrated microorganisms, in which most of the elements that were composing the polymers were retained, whereas the small electrolytes and molecules were removed. The use of gold nanoparticles as biological probes requires the improvement of colloidal stability. Dihydrolipoic acid (DHLA), a dithiol obtained by the reduction of thioctic acid, appears therefore very attractive for the stabilization and the further functionalization of gold nanoparticles because DHLA is characterized by a carboxylic acid group and two thiol functions. The ionizable carboxylic acid groups ensure, for pH ≥ 8, the water solubility of DHLA-capped gold (Au@DHLA) nanoparticles, prepared by the Brust protocol, and the stability of the resulting colloid by electrostatic repulsions. Moreover almost all DHLA, adsorbed onto gold, adopts a conformation allowing their immobilization by both sulfur ends. It is proved by sulfur K-edge X-ray absorption near edge structure spectroscopy, which appears as an appropriate tool for determining the chemical form of sulfur atoms present in the organic monolayer. Such a grafting renders the DHLA monolayers more resistant to displacement by dithiothreitol than mercaptoundecanoic acid monolayers. The presence of DHLA on gold particles allows their functionalization by the electroluminescent luminol through amine coupling reactions assisted by 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride and N-hydroxysuccinimide. As a luminol-functionalized particle is nine times as bright as a single luminol molecule, the use of the particles as a biological probe with a lower threshold of detection is envisaged.

X-ray imaging techniques at the best spatial resolution and using synchrotron facilities are put forth as powerful techniques for the search of small life forms in extraterrestrial rocks under quarantine conditions. Absorption and fluorescence X-ray microtomographies on sub-millimeter silicate assemblages inside a container reveal the mineralogical microenvironments were life should be looked for in priority. Limitations with respect to bacterial detection are due to the difficulties to obtain information about light elements (Z less than or equal to 14), major constituents of biological and silicate samples. The X-ray signature of a "present" bacteria on a silicate surface was defined by X-ray mapping, out of a container, as coincident micrometer and oval zones having strong P and S fluorescence lines (S-fluorescence being slightly lower than P-fluorescence) and an amino-linked sulfur redox speciation. The detection of a single cell along with new procedures to calculate tomographic views will allow considerable improvements of 3D detection of life by X-ray techniques.

The goal of this work is to present the current state of the X-ray Fluorescence Computed Microtomography. The advantages of both the experimental techniques and the data processing (the ESRF ID22 beamline facilities [1]) are considered. The latter one includes the spectra treatment, reconstruction of the atomic density distribution function of the element under consideration and/or calculation of the element effective concentration map and calculation of the element ratio maps. Possible ways of estimating the space resolution and the accuracy are discussed. Results of the tomographic investigation of the NWA817 meteorite micro-grain are presented.
