Abstract

International audience; Bacteria tumble periodically, following environmental cues. Whether they can tumble near a solid surface is a basic issue for the inception of infection or mineral biofouling. Observing freely swimming Escherichia coli near and parallel to a glass surface imaged at high magnification (Â100) and high temporal resolution (500 Hz), we identified tumbles as events starting (or finishing, respectively) in abrupt deceleration (or reacceleration, respectively) of the body motion. Selected events show an equiprobable clockwise (CW) or counterclockwise change in direction that is superimposed on a surface CW path because of persistent propulsion. These tumbles follow a common long (about 300 5 100 ms, N ¼ 52) deceleration-reorientation acceleration pattern. A wavelet transform multiscale analysis shows these tumbles cause in-plane diffusive reorientations with 1.5 rad 2 /s rotational diffusivity, a value that compares with that measured in bulk tumbles. In half of the cases, additional few-millisecond bursts of an almost equiprobable CW or counterclockwise change of direction (12 5 90 , N ¼ 89) occur within the reorientation stage. The highly dispersed absolute values of change of direction (70 5 66 , N ¼ 89) of only a few bursts destabilize the cell-swimming direction. These first observations of surface tumbles set a foundation for statistical models of run-and-tumble surface motion different from that in bulk and lend support for chemotaxis near solid surface.

18. A new nondestructive X-ray method for the determination of the 3D mineralogy at the micrometer scale

Abstract

The combination of synchrotron-based X-ray absorption and fluorescence computed tomographies (CT) is a new method allowing a noninvasive and nondestructive determination of the three-dimensional (3D) mineralogy with micrometer resolution of sub-millimeter silicate grains, possibly stored in a silica holder. These CT were performed with beams of a few tens of keV from a third-generation synchrotron source on one olivine grain of the NWA817 Martian meteorite presenting a reddish alteration phase. The reconstructed sections show a network of fractures and a few micrometer-thick layers formed on one grain. The 3D facet orientation and the X-ray attenuation coefficient indicate that this grain is an [Fo.sub.44[+ or -]9] olivine crystal. The fluorescence section reveals rims enriched in Fe (a major element) or depleted in Ca (a minor element). This CT combination shows that the micrometer-thick layer is preferentially formed on the (010) olivine face and has a lower density (3.5 [+ or -] 0.4 g/[cm.sup.3]) than the olivine, even though it is enriched in Fe. Its complex nano-petrography and the distributions of nanometer-sized voids and fractures in such a micrometer thick layer, first observed by scanning electron microscopy on focused ion-beam cuts, is not shown by CT. The precision presently achieved, although moderate, is sufficient to obtain a 3D semi-quantitative view of the mineralogy consistent with the one previously established by electron probe microanalyses (Sautter et al. 2002).

Abstract

Under the auspices of the Stardust Interstellar Preliminary Examination, picokeystones extracted from the Stardust Interstellar Dust Collector were examined with synchrotron Fourier transform infrared (FTIR) microscopy to establish whether they contained extraterrestrial organic material. The picokeystones were found to be contaminated with varying concentrations and speciation of organics in the native aerogel, which hindered the search for organics in the interstellar dust candidates. Furthermore, examination of the picokeystones prior to and post X-ray microprobe analyses yielded evidence of beam damage in the form of organic deposition or modification, particularly with hard X-ray synchrotron X-ray fluorescence. From these results, it is clear that considerable care must be taken to interpret any organics that might be in interstellar dust particles. For the interstellar candidates examined thus far, however, there is no clear evidence of extraterrestrial organics associated with the track and/or terminal particles. However, we detected organic matter associated with the terminal particle in Track 37, likely a secondary impact from the Al-deck of the sample return capsule, demonstrating the ability of synchrotron FTIR to detect organic matter in small particles within picokeystones from the Stardust interstellar dust collector.

20. Study of the (010) olivine surface by Rutherford backscattering spectrometry in channeling geometry

Abstract

The (010) surfaces of forsterite ([FO.sub.100]) and natural forsteritic olivine ([FO.sub.90]) single crystals have been studied by Rutherford backscattering spectrometry (RBS). Spectra were collected either in channeling geometry with the incident beam parallel to the [010] zone axis, or in a random orientation. In both materials, two surface preparations were examined: (1) mechanical polishing and (2) chemical etching by hydrofluoric acid after mechanical polishing. Composition profiles extending to several hundreds of nanometers below the surface were probed by RBS in random mode. Simulations of all the spectra indicated constant major element compositions, equal to the bulk compositions of the crystals. Characteristics of the few top atomic layers were probed by RBS in channeling mode. The crystalline quality of the surface of chemically etched samples has been evaluated quantitatively and is shown to be much better than that of mechanically polished samples. Given the energy resolution, we estimate that the bulk crystalline quality is perturbed for more than 40 nm below the surface in the case of mechanical polishing whereas disorder is limited to a topmost layer thinner than 1 nm in the case of chemical etching. On average, for the chemically etched samples, less than one atom per [010] atomic row is displaced from its mean crystallographic position. The bulk stoichiometry is preserved in the topmost layer of pure forsterite whereas a slightly higher Fe/Mg ratio is detected at the surface of chemically etched [FO.sub.90]. A method for the quantitative assessment of surface quality of olivine is thus proposed, constituting a useful preliminary step before any study of surface modifications of olivine subjected to various geological conditions.

Abstract

NASA's interstellar collector from the Stardust mission captured several particles that are now thought to be of interstellar origin. We analyzed two of these via nanodiffraction at the European Synchrotron Radiation Facility (ESRF) and found them to contain crystalline components. The unit cell of the crystalline material is determined from the diffraction patterns and the most likely mineral components are identified as olivine and spinel.

Abstract

International audience; Metals are pre-requisites for life, fingerprints of life and tracers of metabolic processes of biomineralisation in living organisms. To study metals at macromolecular interaction scales requires state of the art analytical capabilities down to trace element levels and nanometer spatial resolutions. Fortunately, in the last decade, highly sensitive X-ray probes have been developed at worldwide 3rd generation synchrotron facilities. To date, a limited number of these highly specific nanoimaging probes have been deployed, and the ESRF ID16A/B and ID21 ones are among the leaders. X-rays are non (or least) destructive, noninvasive, penetrative and highly sensitive probes of solid samples and metals are their ideal targets. X-ray fluorescence has reached few tens of nm resolutions and a world record for absolute mass of e.g. 50 zg for Fe, whichcan be recognized as only about 600 atoms. Over the past decade, we have developed a multi-analyses methodology of nanoimaging mid-Z elements in low-Z matrices as a direct application to Life and/or Planetary Sciences of trace metals in biological matrices or in C/Si-based ones.X-ray fluorescence and absorption spectroscopies, deployed at the few nanometer scales, are sensitive probes but they require a re-assessment of the analytical requirements previously established at the few micron levels [1]. We will present some specific analytical developments and methodologies optimized for such applications. They are centered onto absolute quantification of metals using both the Fundamental Parameter Approximation and reference materials, after spectra analysis using the PyMCA package [2] coupled to Monte-Carlo simulations of sample compositions [3] and geometries, prerequisites for 2D/3D elemental imaging. Examples based on metal detection were applied to either environmental key organisms such as foraminifers or coccolithofores [4, 5] but also to the oldest living organisms tracedback to life's origins on Earth [1]. Finally, our search for life being also aimed at planetary and astronomical scales on exoplanets, we will present our patented methodology and sample holder [6] for Quarantine Extraterrestrial Sample Analyses (QESA) for the upcoming Returned Sample Missions from Mars and asteroids [7].[1] – L. Abstract

Conventional x-ray transmission tomography provides the spatial distribution of the absorption coefficient inside a sample. Other tomographic techniques, based on the detection of photons coming from fluorescent emission, Compton and Rayleigh scattering, are used for obtaining information on the internal elemental composition of the sample. However, the reconstruction problem for these techniques is generally much more difficult than that of transmission tomography, mainly due to self-absorption effects in the sample. In this article an approach to the reconstruction problem is presented, which integrates the information from the three types of signals. This method provides the quantitative spatial distribution of all elements that emit detectable fluorescent lines (Z≥15 in usual experimental conditions), even when the absorption effects are strong, and the spatial distribution of the global density of the lighter elements. The use of this technique is demonstrated on the reconstruction of a grain of the martian meteorite NWA817, mainly composed of low Z elements not measured in fluorescence and for which this method provides a unique insight. 31. Tumble Kinematics of Escherichia colinear a Solid Surface

Abstract

Bacteria tumble periodically, following environmental cues. Whether they can tumble near a solid surface is a basic issue for the inception of infection or mineral biofouling. Observing freely swimming Escherichia colinear and parallel to a glass surface imaged at high magnification (×100) and high temporal resolution (500 Hz), we identified tumbles as events starting (or finishing, respectively) in abrupt deceleration (or reacceleration, respectively) of the body motion. Selected events show an equiprobable clockwise (CW) or counterclockwise change in direction that is superimposed on a surface CW path because of persistent propulsion. These tumbles follow a common long (about 300 ± 100 ms, N = 52) deceleration-reorientation-acceleration pattern. A wavelet transform multiscale analysis shows these tumbles cause in-plane diffusive reorientations with 1.5 rad2/s rotational diffusivity, a value that compares with that measured in bulk tumbles. In half of the cases, additional few-millisecond bursts of an almost equiprobable CW or counterclockwise change of direction (12 ± 90°, N = 89) occur within the reorientation stage. The highly dispersed absolute values of change of direction (70 ± 66°, N = 89) of only a few bursts destabilize the cell-swimming direction. These first observations of surface tumbles set a foundation for statistical models of run-and-tumble surface motion different from that in bulk and lend support for chemotaxis near solid surface.

32. An experimental study of the external reduction of olivine single crystals

Abstract

Single crystals of San Carlos olivine in contact with graphite were annealed at P = 1 bar, T = 1373 K, for studying the reaction of extraction of (Fe, Ni) metal. Scanning electron microscopy and transmission electron microscopy were performed on samples recovered after the experiments. Precipitates of (Fe, Ni) and thin amorphous layer of silica were identified, exclusively on the surface of the single crystals. Mass balance indicates that volatilization of Fe, Mg, and Si is negligible under these conditions. The reaction can be summarized as: [MATHEMATICAL EXPRESSION NOT REPRODUCIBLE IN ASCII] which occurs at the crystal surface without affecting the interior of the crystal, except for an [Fe.sup.2+] and [Mg.sup.2+] compositional profile in the olivine matrix. These chemical profiles are consistent with measured values of [Fe.sup.2+]-[Mg.sup.2+] interdiffusion coefficients, in agreement with the fact that Si and O are relatively immobile in olivine under such conditions. This study shows that annealing at relatively moderate temperature under reducing conditions can cause surface modifications and thus probably can strongly influence the surface evolution of planetary objects exposed directly to space environments (regoliths, surfaces of asteroids, or interplanetary dust particles).

73rd Annual Meeting of the Meteoritical-Society : New York, NY, July 26-30, 2010; International audience; In preparation for the upcoming international Mars Sample Return mission, bringing to Earth samples containing potential biohazards, we have implemented a hyperspectral method of analysis of grains performed in BSL4 quarantine conditions,by combining several non-destructive imaging diagnostics. This methodology was tested on meteorites [1, 2] and cometary grains from the recent NASA Stardust mission [3-6].

Abstract

73rd Annual Meeting of the Meteoritical-Society, New York, NY, July 26-30, 2010; International audience; Synchrotron radiation X-ray fluorescence (SR-XRF) is a method of choice to analyze fragile, unique meteoritic samples, requiring no sample preparation. It is a nondestructive, multielemental, quantitative method, easily coupled to diffraction and speciation for a detailed sample characterization. The composition of samples thicker than a few microns is however difficult to obtain due to the high attenuation of the characteristic X-rays resulting in non-detection of low-Z elements (Z≤ 14).

Abstract

On the basis of an interstellar dust model compatible with Ulysses and Galileo observations, we calculate and predict the trajectories of interstellar dust (ISD) in the solar system and the distribution of the impact speeds, directions, and flux of ISD particles on the Stardust Interstellar Dust Collector during the two collection periods of the mission. We find that the expected impact velocities are generally low (<10 km s?1) for particles with the ratio of the solar radiation pressure force to the solar gravitational force ? > 1, and that some of the particles will impact on the cometary side of the collector. If we assume astronomical silicates for particle material and a density of 2 g cm?3, and use the Ulysses measurements and the ISD trajectory simulations, we conclude that the total number of (detectable) captured ISD particles may be on the order of 50. In companion papers in this volume, we report the discovery of three interstellar dust candidates in the Stardust aerogel tiles. The impact directions and speeds of these candidates are consistent with those calculated from our ISD propagation model, within the uncertainties of the model and of the observations.

Abstract

International audience; In preparation for the upcoming sample return missions containing potential biohazards which may have withstood the rigors of space travel we present a hyperspectral method of in-situ analysis of grains combining several non-destructive imaging diagnostics, performed in BSL4 quarantine conditions. This offers an alternative to the analyses in facilities at large, using optimized experimental setups while keeping the samples in conditions of quarantine. Our methodology was tested during analyses of meteorites and cometary and interstellar grains from the recent NASA Stardust mission.

Abstract

CI, CM, CR and ungrouped C2 carbonaceous chondrites contain a few percents of insoluble organic matter (IOM), which could have a presolar origin and provide clues on carbon chemistry in interstellar medium of protostellar environments. However, these chondrites have experienced complex hydrothermal processes in their parent bodies [1-3]. A major issue is the understanding of chemical changes in composition and structure of IOM due to aqueous alteration. Chemical variations have been reported among various chondrites, and so far interpreted as the consequence of oxidation processes by some authors [4; 5]. Variations among the precursors accreted can also be suspected [6]. We measured by micro-infrared spectroscopy (at LPG, Grenoble) and sulfur XANES at the K-edge (at ESRF, ID21, Grenoble) a series of IOMs extracted from CI1 (Ivuna, Orgueil, Alais), CM2 (Murchison, Cold Bokkeveld, QUE99355, QUE97990, Mighei and Murray), CR2 (Renazzo) and C2 ungrouped (Tagish Lake) chondrites. Only CIs and Cold Bokkeveld have strong carboxylic IR bands and oxidised organic sulfur XANES features. Neither XANES and IR show major variability within one group. These results evidence systematic differences between C1 and other C2 chondrites. Cold Bokkeveld appears as a peculiar object, and should be considered as an anomalous CM2. The lack of organic oxidation trend going with alteration gradient makes us say : i) hydrothermalism was very stronger on C1 parent body than on C2's and ii) oxidation of different precursors of C1 and C2 may have an effect on the IOM.

Abstract

The study of very ancient microfossils has recently raised contentious issues regarding interpretation of the biogenicity of the structures. In situ investigation of certain elements such as sulfur within potential microfossils is a powerful complement to other methods of investigation that can provide valuable information on biogenicity. We present here a first such study on Precambrian microfossils from the 700–800-My-old Neoproterozoic Draken Formation, Svalbard, using scanning X-ray microscopy (SXM) in the fluorescence mode and X-ray absorption near edge spectroscopy (XANES) at the sulfur K-edge. SXM allowed mapping of up to 300 ppm of probably endogenous sulfur within the kerogenous walls of Myxococcoides chlorelloidea microfossils. XANES showed that the sulfur is most likely contained in heterocyclic organic compounds, such as thiophene(s).
