Objective Diffuse‐type tenosynovial giant cell tumors (Dt‐TGCTs) commonly occur in the knee joint and tend to recur postoperatively. However, limited clinical data are available on ankle joint involvement especially associated multiportal arthroscopic treatment outcomes. The purpose of this study was to report the clinical results of multiportal arthroscopy‐assisted resection of Dt‐TGCTs of the ankle. Methods We retrospectively reviewed the clinical data of 33 patients with Dt‐TGCT of the ankle who underwent multiportal arthroscopic treatment between August 2011 and December 2020. Clinical follow‐up included the visual analogue scale (VAS) score, American Orthopedic Foot and Ankle Society (AOFAS) score, Kofoed score, and recurrence rate to assess surgical outcomes. The number of patients who achieved the patient acceptable symptom state (PASS) based on the AOFAS score was also examined. Additionally, the patients were categorized into two groups based on the final surgical approach: Group A who underwent multiportal arthroscopic synovectomy and Group AO who underwent combined arthroscopic and open surgical excision. Intergroup comparisons were conducted. Intraoperative characteristics, such as the number of patients with involvement of the tarsal tunnel and fibularis tendon and the Outerbridge grading of cartilage damage, were recorded to assess the selection of surgical procedures. Results Among the 33 patients, 15 were assigned to Group A, and 18 were in Group AO. The median follow‐up duration for the 33 patients was 77 months (range, 28–142 months). The median VAS score was 1 (range, 0–4), the AOFAS score was 96 (range, 65–100), and the Kofoed score was 96 (range, 67–100). A total of 27 patients (82%) achieved PASS based on AOFAS scores, while five patients (15%) had recurrence. No statistically significant difference was observed between the two groups in recurrence rate, follow‐up VAS score, AOFAS score, Kofoed score, or number of patients who reached the PASS (p > 0.05). In the AO group, 16 cases of Dt‐TGCT involved the tarsal tunnel, and 11 cases involved the fibularis tendon. All these patients exhibited extension beyond the joint. Abstract Background Hepatocellular carcinoma (HCC) is a malignant tumor characterized by a high mortality rate. The occurrence and progression of HCC are linked to oxidative stress. Glyoxalase-1 (GLO1) plays an important role in regulating oxidative stress, yet the underlying mechanism remains unclear. GLO1 may serve as a prognostic biomarker and therapeutic target for HCC. Methods Based on TCGA database hepatocellular carcinoma samples, we conducted a bioinformatics analysis to explore the correlation between GLO1 expression and HCC cell proliferation and viability. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis revealed that differentially expressed genes (DEGs) were mainly enriched in the cell cycle pathway. We analyzed the relationships between GLO1 and 24 genes enriched in the cell cycle pathway using a protein-protein interaction (PPI) network. Finally, experimental validation was performed to assess GLO1’s impact on the distribution of cells at different cell cycle stages and on the proliferation and migration of HCC cells. Results Our study demonstrated that GLO1 was overexpressed in HCC tissues and was associated with a poor prognosis. Data analysis indicated that overexpression of GLO1 activated the cell cycle pathway and positively correlated with expression of the majority of key cell cycle genes. Experimental validation showed that GLO1 expression affects the number of HCC cells in G2 and S phases and regulates HCC cell proliferation and migration. Conclusions GLO1 represents a promising therapeutic target for HCC, providing valuable insights into its role in the viability and proliferation of HCC cells.

Altered cell-cell communication is a hallmark of aging, but its impact on bone marrow aging remains poorly understood. Based on a common and effective pipeline and single-cell transcriptome sequencing, we detected 384,124 interactions including 2575 ligand-receptor pairs and 16 non-adherent bone marrow cell types in old and young mouse and identified a total of 5560 significantly different interactions, which were then verified by flow cytometry and quantitative real-time PCR. These differential ligand-receptor interactions exhibited enrichment for the senescence-associated secretory phenotypes. Further validation demonstrated supplementing specific extracellular ligands could modify the senescent signs of hematopoietic stem cells derived from old mouse. Our work provides an effective procedure to detect the ligand-receptor interactions based on single-cell sequencing, which contributes to understand mechanisms and provides a potential strategy for intervention of bone marrow aging.

Abstract Plasmonic materials can generate strong electromagnetic fields to boost the Raman scattering of surrounding molecules, known as surface-enhanced Raman scattering. However, these electromagnetic fields are heterogeneous, with only molecules located at the ‘hotspots’, which account for ≈ 1% of the surface area, experiencing efficient enhancement. Herein, we propose patterned plasmonic trimers, consisting of a pair of plasmonic dimers at the bilateral sides and a trap particle positioned in between, to address this challenge. The trimer configuration selectively directs probe molecules to the central traps where ‘hotspots’ are located through chemical affinity, ensuring a precise spatial overlap between the probes and the location of maximum field enhancement. We investigate the Raman enhancement of the Au@Al2O3-Au-Au@Al2O3 trimers, achieving a detection limit of 10−14 M of 4-methylbenzenethiol, 4-mercaptopyridine, and 4-aminothiophenol. Moreover, single-molecule SERS sensitivity is demonstrated by a bi-analyte method. Benefiting from this sensitivity, our approach is employed for the early detection of lung tumors using fresh tissues. Our findings suggest that this approach is sensitive to adenocarcinoma but not to squamous carcinoma or benign cases, offering insights into the differentiation between lung tumor subtypes.

Background: Bladder cancer (BCa), one of the most common cancers worldwide, is characterized by high rates of recurrence, progression, and mortality. Machine learning algorithms offer promising advancements in enhancing predictive models. This study aims to develop robust machine learning models for predicting BCa survival using clinical and gene expression data. Methods: Clinical data from BCa patients were obtained from the Surveillance, Epidemiology, and End Results database. Cox proportional hazards regression models assessed the association between clinical variables and overall survival. Machine learning algorithms, including logistic regression, random forest, XGBoost, decision tree, and LightGBM, were employed to predict survival at 1, 3, and 5 years. The TAGO database, combined with the data from The Cancer Genome Atlas and four databases from the Gene Expression Omnibus, which have available genomic data and clinical data, were selected. Gene expression data were transformed into gene sets data, and the performance of models based on clinical data and gene sets data and their combination were compared. Furthermore, the impact of model-derived scores on overall survival was evaluated. Results: Among 138,741 BCa patients with available clinical data, key independent predictors of survival included age, race, marital status, surgery, chemotherapy, radiation, and TNM stages. Clinical data machine learning (CML) models used these clinical predictors to achieve AUC values of 0.860, 0.821, and 0.804 in the testing sets for predicting survival at 1, 3, and 5 years, respectively. In the TAGO database, which has 863 patients with clinical and genomic data, the integrated clinical and gene expression machine learning model (IML) outperformed the CML and gene expression machine learning (GML) models in survival prediction. Patients with higher IML and GML model scores exhibited poorer survival outcomes. Conclusions: This study successfully identifies key clinical and genomic predictors, a significant step forward in BCa research. The development of predictive models for BCa survival underscores the potential of integrated data approaches in improving BCa management and treatment strategies.

Essential oils with β-caryophyllene, citral, and linalool as key compounds often exhibit some anti-anxiety like effects in aromatherapy. However, evidence of the effect of these three compounds through human inhalation remains limited. It is worth exploring their potential anxiolytic effect through the olfactory pathway, and finding out whether the three compounds lead to different physiological responses. A total of 48 subjects were randomly assigned to three odor (β-caryophyllene, citral, and linalool) inhalation groups and one control (odorless jojoba oil) group. Stress stimulation was induced using n-back and mental arithmetic tasks. The odor was administered before the task test session. Assessments including the State-Trait Anxiety Inventory (STAI), electroencephalogram (EEG) activities, facial expressions, several physiological indicators, and a self-report scale of subjective perception of the odor environments were carried out. The changes before and after inhalation, as well as the inter-group differences, were analyzed. Both β-caryophyllene and citral inhalation led to a significant decrease in anxiety levels, while only β-caryophyllene resulted in a notable reduction across both sub-scales of STAI. Following the odor inhalation, heart rate significantly decreased in all three groups, with the β-caryophyllene group exhibiting the most pronounced decline. While the systolic blood pressure of the linalool group demonstrated a statistically significant difference. Regarding facial expressions, β-caryophyllene significantly increased the ratio of 'Happiness' and decreased the ratio of ‘Fear'. In the non-task state, citral reduced the power of frontal alpha, delta, and theta waves while β-caryophyllene had a similar effect. All odor inhalation groups showed increased delta and theta waves after the task compared with the control group, with the β-caryophyllene group having notably lower frontal beta waves. β-Caryophyllene and citral exhibited good anti-anxiety effects. Subjects receiving different odors showed different EEG and physiological responses, indicating the differences in emotional regulation ways among the three compounds.

Co-infection with diverse bacteria is commonly seen in patients infected with the novel coronavirus, SARS-CoV-2. This type of co-infection significantly impacts the occurrence and development of novel coronavirus infection. Bacterial co-pathogens are typically identified in the respiratory system and blood culture, which complicates the diagnosis, treatment, and prognosis of COVID-19, and even exacerbates the severity of disease symptoms and increases mortality rates. However, the status and impact of bacterial co-infections during the COVID-19 pandemic have not been properly studied. Recently, the amount of literature on the co-infection of SARS-CoV-2 and bacteria has gradually increased, enabling a comprehensive discussion on this type of co-infection. In this study, we focus on bacterial infections in the respiratory system and blood of patients with COVID-19 because these infection types significantly affect the severity and mortality of COVID-19. Furthermore, the progression of COVID-19 has markedly elevated the antimicrobial resistance among specific bacteria, such as Klebsiella pneumoniae, in clinical settings including intensive care units (ICUs). Grasping these resistance patterns is pivotal for the optimal utilization and stewardship of antibiotics, including fluoroquinolones. Our study offers insights into these aspects and serves as a fundamental basis for devising effective therapeutic strategies. We primarily sourced our articles from PubMed, ScienceDirect, Scopus, and Google Scholar. We queried these databases using specific search terms related to COVID-19 and its co-infections with bacteria or fungi, and selectively chose relevant articles for inclusion in our review.

Abstract Idiopathic pulmonary fibrosis (IPF), often likened to a “tumor‐like disease,” proves more lethal than several malignancies. Although prior studies have demonstrated the lung‐protective efficacy of ginsenosides, the precise mechanism underlying their alleviative effect on IPF remains elusive. In this study, we validated the efficacy of the ginsenoside Rh4 in alleviating IPF and delved into the underlying mechanisms. Our data showed that Rh4 intervention significantly reduced mortality and hydroxyproline (HYP) content in mice. It also alleviated the pathological process of IPF by ameliorating phenomena such as alveolar wall thickening induced by IPF. In addition, in vitro cellular experiments confirmed that ginsenoside Rh4 was effective in alleviating transforming growth factor‐β1‐induced IPF model. Further analysis showed that ginsenoside Rh4 significantly reduced collagen fiber production and deposition while inhibiting the coagulation cascade signaling pathway. In addition, ginsenoside Rh4 inhibited the epithelial‐mesenchymal transition(EMT) pathway by promoting the CXCR3‐CXCL9 axis, which ultimately alleviated IPF. In conclusion, our data strongly suggest that ginsenoside Rh4 has the potential to be an innovative prophylactic drug against IPF.

Abstract Background The current literature shows that dyslipidemia can lead to a higher incidence of rotator cuff tears (RCTs) and an increased retear rate after repair. We aimed to evaluate the influence of preoperative dyslipidemia on postoperative pain, patient-reported outcomes (PROs), active range of motion (ROM), and structural integrity. Methods A cohort of 111 patients who underwent arthroscopic RCT repair between January 2021 and July 2022, and whose complete preoperative serum lipid data were available within one week prior to surgery was retrospectively reviewed. Dyslipidemia was defined as the presence of an increase or decrease in at least one blood lipid profile (triglycerides, total cholesterol, low-density lipoprotein, high-density lipoprotein, or non-high-density lipoprotein). There were 43 patients in the dyslipidemia group and 68 in the ortholiposis group. Patient evaluations, including pain score, PROs, and ROMs, were conducted preoperatively; at 3 and 6 months postoperatively; and at the last follow-up. Structural integrity was assessed by magnetic resonance imaging (MRI) 6 months after surgery if possible, and Sugaya type 4 or 5 was considered a retear. Propensity score matching (PSM) was used to reduce bias. Results The RCT size, surgical technique, preoperative pain status, PROs, and active ROM were comparable between patients with dyslipidemia and those with ortholiposis. Three months after surgery, patients in the dyslipidemia group had worse average PROs (Constant score: P = 0.001; ASES score: P = 0.012; UCLA score: P = 0.015), forward flexion (P = 0.012), and internal rotation (P = 0.001) than patients in the ortholiposis group did. The difference between the two groups persisted after PSM but disappeared at the sixth month after surgery. No significant differences in pain score, PROs, or active ROMs were detected between the dyslipidemia and ortholiposis groups after a mean follow-up of 24 months. Of the 72 patients who underwent MRI, 4 retears (5.6%) were found, and all were in the ortholiposis group. There was no difference in the rate of retears between the two groups (P = 0.291) or with (P = 0.495) PSM. Conclusions In conclusion, we found that perioperative dyslipidemia may impact initial recovery within the first 3 months following arthroscopic rotator cuff repair but may have no effect on pain, PROs, or active ROMs at a mean 2-year follow-up or rotator cuff integrity at 6 months postoperatively. Abstract Ferroptosis is a non-apoptotic form of regulated cell death characterized by the lethal accumulation of iron-dependent membrane-localized lipid peroxides. It acts as an innate tumor suppressor mechanism and participates in the biological processes of tumors. Intriguingly, mesenchymal and dedifferentiated cancer cells, which are usually resistant to apoptosis and traditional therapies, are exquisitely vulnerable to ferroptosis, further underscoring its potential as a treatment approach for cancers, especially for refractory cancers. However, the impact of ferroptosis on cancer extends beyond its direct cytotoxic effect on tumor cells. Ferroptosis induction not only inhibits cancer but also promotes cancer development due to its potential negative impact on anticancer immunity. Thus, a comprehensive understanding of the role of ferroptosis in cancer is crucial for the successful translation of ferroptosis therapy from the laboratory to clinical applications. In this review, we provide an overview of the recent advancements in understanding ferroptosis in cancer, covering molecular mechanisms, biological functions, regulatory pathways, and interactions with the tumor microenvironment. We also summarize the potential applications of ferroptosis induction in immunotherapy, radiotherapy, and systemic therapy, as well as ferroptosis inhibition for cancer treatment in various conditions. We finally discuss ferroptosis markers, the current challenges and future directions of ferroptosis in the treatment of cancer.

Abstract The Ebinur Lake Basin is an ecologically sensitive area in an arid region. Investigating its land use and land cover (LULC) change and assessing and predicting its ecosystem service value (ESV) are of great importance for the stability of the basin's socioeconomic development and sustainable development of its ecological environment. Based on LULC data from 1990, 2000, 2010, and 2020, we assessed the ESV of the Ebinur Lake Basin and coupled the grey multi-objective optimization model with the patch generation land use simulation model to predict ESV changes in 2035 under four scenarios: business-as-usual (BAU) development, rapid economic development (RED), ecological protection (ELP), and ecological–economic balance (EEB). The results show that from 1990 to 2020, the basin was dominated by grassland (51.23%) and unused land (27.6%), with a continuous decrease in unused land and an increase in cultivated land. In thirty years, the total ESV of the study area increased from 18.62 billion to 67.28 billion yuan, with regulation and support services being the dominant functions. By 2035, cultivated land increased while unused land decreased in all four scenarios compared with that in 2020. The total ESV in 2035 under the BAU, RED, ELP, and EEB scenarios was 68.83 billion, 64.47 billion, 67.99 billion, and 66.79 billion yuan, respectively. In the RED and EEB scenarios, ESV decreased by 2.81 billion and 0.49 billion yuan, respectively. In the BAU scenario, provisioning and regulation services increased by 6.05% and 2.93%, respectively. The ELP scenario, focusing on ecological and environmental protection, saw an increase in ESV for all services. This paper can assist policymakers in optimizing land use allocation and provide scientific support for the formulation of land use strategies and sustainable ecological and environmental development in the inland river basins of arid regions.

Multi-channel neural electrodes as a crucial means are of great significance for information exchange between the brain and computers. Herein, we present a 16-channel Si-based active neural probe system that achieves a monolithic integration between the electrodes and circuits in a single probe, making it a standalone integrated electrophysiology recording system. The ASIC prepared on a base (2×2mm2) is a 16-channel analog frontend (AFE) for neural recording, and each channel has a low-noise amplifier (LNA), a bandpass filter (BPF), a buffer and a current bias circuit. The 258 neural signal recording electrodes (22×24μm2) are densely packed on a 50 μm thick, 100 μm wide, and 3 mm long shank. The ASIC of neural probe, internal interconnecting wires are all implemented in commercial SMIC 0.18 μm CMOS technology. The neural probe system achieves a 3.6 μVrms input-referred noise (IRN) in a bandwidth of 1.1Hz-10 kHz, 70.8 μW power consumption, 0.0785 mm2 area per channel, as well as an AFE gain of 58.1 dB Furthermore, the impedances of the Au electrodes can be obtained as 0.5–2.1 MΩ at a frequency of 1 kHz. The functionality of a 16-channel silicon-based neural probe is validated in an in-vivo experiment on lab rats.

AbstractPurpose Remnant cholesterol (RC) is the cholesterol content of triglyceride-rich lipoproteins. This study aimed to investigate the association between RC levels and kidney stones in U.S. adults.Methods Data were obtained from the 2007 to 2016 National Health and Nutrition Examination Survey (NHANES). A total of 10,551 participants with complete data were included and analyzed in this study. Univariate and multivariate logistic regression analysis, restricted cubic spline function, subgroup analysis and mediation analysis were preformed to estimate the independent relationship between RC levels and kidney stones.Results Participants with stone formation had higher levels of RC than those with without stone formation (25.78 ± 13.83 vs 23.27 ± 13.04, p

Abstract Metastasis is a major contributing factor that affects the prognosis of melanoma patients. Nevertheless, the underlying molecular mechanisms involved in melanoma metastasis are not yet entirely understood. Here, we identified ubiquitin‐specific protease 22 (USP22) as a pro‐oncogenic protein in melanoma through screening the survival profiles of 52 ubiquitin‐specific proteases (USPs). USP22 demonstrates a strong association with poor clinical outcomes and is significantly overexpressed in melanoma. Ablation of USP22 expression remarkably attenuates melanoma migration, invasion, and epithelial–mesenchymal transition in vitro and suppresses melanoma metastasis in vivo. Mechanistically, USP22 controls melanoma metastasis through the SIRT1/PTEN/PI3K pathway. In addition, we conducted an United States Food and Drug Administration‐approved drug library screening and identified topotecan as a clinically applicable USP22‐targeting molecule by promoting proteasomal degradation of USP22. Finally, we found that both pharmacological and genetic silence of USP22 sensitize RSL3‐induced ferroptosis through suppressing the PI3K/Akt/mTOR pathway and its downstream SCD, and ferroptosis inhibitor could partly rescued the decreased lung metastasis by topotecan in vivo. Overall, our findings reveal a prometastatic role of USP22 and identify topotecan as a potent USP22‐targeting drug to limit melanoma metastasis.

In this study, polyoxometalates (POMs) as a core-modifying material was used to fabricate the nanofiltration (NF) membrane on the polyvinylidene fluoride (PVDF) microfiltration membrane substrate via a novel interfacial polymerization (IP) method. The formation mechanism of the POMs-modified composite membrane was proposed. The separation and antifouling properties were further investigated. After cross-linking with POMs through the new IP reaction, the modified composite membrane showed improved hydrophilicity, water flux, and salt rejection. In the humic acid fouling experiment, the POMs-modified membrane exhibited the best antifouling performance, with a flux recovery rate of up to 91.3%. Electrochemical impedance spectroscopy was further used to investigate the antifouling performance of the membranes. Nyquist and Bode plots of the POMs-modified membranes showed no significant change before and after fouling compared to the PVDF membrane substrate, indicating reduced fouling attachment on the modified membrane, which was consistent with the fouling index and flux variation observed during the fouling experiment. Our findings provide a simple and valuable route for fabricating POMs-functionalized NF membranes with desirable separation and antifouling performance. HIGHLIGHTS A simple method is proposed to modify POMs onto the NF membrane.; The separation and antifouling performance of the POMs-modified membranes improved evidently.; After simple cleaning, the flux recovery rate of the POMs-modified membrane reached more than 90%.; Improved antifouling performance of the POMs-modified membrane was confirmed by electrochemical impedance spectroscopy analysis.;

Objective There has been long‐standing debate about whether a medial opening wedge high tibial osteotomy (MOWHTO) gap should be filled with autologous bone graft or any other filler to expedite the healing process. The main purpose of this study was to compare the clinical and radiological outcomes of MOWHTO with an opening gap ≥10 mm, utilizing autograft, allograft, or no graft at 1 year postoperatively. Methods A total of 68 patients were included in this retrospective study and divided into three treatment groups: Group A (no bone graft), Group B (autologous iliac crest graft), and Group C (allogenous tibia plateau graft). At postoperative 1‐year follow‐up, the area of callus filling in the most medial side of the knee was measured using anteroposterior radiographs, and a modified van Hemert scoring system was used to evaluate bone union outcomes in five mediolaterally divided zones. Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) scores and relevant complications were assessed. The correlations between the gap width and bone union scores were evaluated. Knee arthroscopy is the replacement choice for patients with symptomatic degenerative arthropathy who have failed conservative treatment, such as intra-articular loose bodies and meniscal pathology. Postoperative joint pain and swelling are the main problems affecting enhanced recovery after surgery, which is thought to be a disorder of the joint environment caused by the trauma of surgery (1, 2, 3). Arthroscopic surgery involves irrigating the joint with a high-pressure flushing solution, which results in the loss of synovial fluid secreted by the joint itself. Synovial fluid has been shown to lubricate and protect the articular cartilage, and it needs to be re-secreted several days after surgery (3). In addition, studies have shown that the lavage fluid could cause damage to articular cartilage, which could also lead to early postoperative joint discomfort (4, 5). Therefore, some scholars have proposed that early postoperative synovial fluid analog supplementation to restore the internal environment may reduce early postoperative joint discomfort and accelerate the rehabilitation process (6, 7, 8, 9).

This article focuses on zircon U-Pb isotope dating and a whole-rock elemental analysis of granodiorites, monzonitic granites, granodioritic porphyries, and alkali feldspar granites in the Yangmugang area of the Lesser Xing’an–Zhangguangcai Range. The zircon U-Pb isotope-dating results revealed that these granitic rocks formed during the late Early Jurassic period (182.9–177.2 Ma). Their geochemical characteristics and zircon saturation temperatures suggest that the granodiorites are moderately differentiated I-type granites and the monzonitic granite, granodioritic porphyries, and alkali feldspar granites are highly differentiated I-type granites. The degree of magma differentiation progressively increased from granodiorites to alkali feldspar granites. By combining the regional Nd and Hf isotope compositions, it was inferred that the magma source involved the melting of lower crustal material from the Mesoproterozoic to the Neoproterozoic eras. By integrating these findings with contemporaneous intrusive rock spatial variations, it was indicated that the late Early Jurassic granitoids in the Lesser Xing’an–Zhangguangcai Range formed within an extensional tectonic setting after the collision and closure of the Songnen–Zhangguangcai Range and Jiamusi blocks. Additionally, this study constrains the closure of the Mudanjiang Ocean to the late Early Jurassic period (177.2 Ma).

Controllable contraception in male animals was demonstrated through the utilization of gold nanorods' photothermal effect to accomplish mild testicular hyperthermia. However, the challenges arising from testicular administration and the non-biodegradability of nanoparticles hinder further clinical implementation. Therefore, a straightforward, non-invasive, and enhanced contraception approach is required. This study explores the utilization of human heavy chain ferritin (HFn) nanocarriers loaded with aggregation-induced emission luminogens (AIEgens) for noninvasive, controllable male contraception guided by Near-Infrared-II (NIR-II) fluorescence imaging. The HFn-caged AIEgens (HFn@BBT) are delivered via intravenous injection and activated by near-infrared irradiation. Lower hyperthermia treatment induces partial damage to the testes and seminiferous tubules, reducing fertility indices by approximately 100% on the 7th day, which gradually recovers to 80% on the 60th day. Conversely, implementation of elevated hyperthermia therapy causes total destruction of both testes and seminiferous tubules, leading to a complete loss of fertility on the 60th day. Additionally, the use of AIEgens in NIR-II imaging offers improved fluorescence efficiency and penetration depth. The findings of this study hold significant promise for the advancement of safe and effective male contraceptive methods, addressing the need for noninvasive and controllable approaches to reproductive health and population control.

Estimating the time of most recent common ancestor (tMRCA) is important to trace the origin of pathogenic viruses. This analysis is based on the genetic diversity accumulated in a certain time period. There have been thousands of mutant sites occurring in the genomes of SARS-CoV-2 since the COVID-19 pandemic started; six highly linked mutation sites occurred early before the start of the pandemic and can be used to classify the genomes into three main haplotypes. Tracing the origin of those three haplotypes may help to understand the origin of SARS-CoV-2. In this article, we present a complete protocol for the classification of SARS-CoV-2 genomes and calculating tMRCA using Bayesian phylodynamic method. Land surface models (LSMs) have prominent advantages for exploring the best agricultural practices in terms of both economic and environmental benefits with regard to different climate scenarios. However, their applications to optimizing fertilization and irrigation have not been well discussed because of their relatively underdeveloped crop modules. We used a CLM5-Crop LSM to optimize fertilization and irrigation schedules that follow actual agricultural practices for the cultivation of maize and wheat, as well as to explore the most economic and environmental-friendly inputs of nitrogen fertilizer and irrigation (FI), in the North China Plain (NCP), which is a typical intensive farming area. The model used the indicators of crop yield, farm gross margin (FGM), nitrogen use efficiency (NUE), water use efficiency (WUE), and soil nitrogen leaching. The results showed that the total optimal FI inputs of FGM were the highest (230 ± 75.8 kg N ha−1 and 20 ± 44.7 mm for maize; 137.5 ± 25 kg N ha−1 and 362.5 ± 47.9 mm for wheat), followed by the FIs of yield, NUE, WUE, and soil nitrogen leaching. After multi-objective optimization, the optimal FIs were 230 ± 75.8 kg N ha−1 and 20 ± 44.7 mm for maize, and 137.5 ± 25 kg N ha−1 and 387.5 ± 85.4 mm for wheat. By comparing our model-based diagnostic results with the actual inputs of FIs in the NCP, we found excessive usage of nitrogen fertilizer and irrigation during the current cultivation period of maize and wheat. The scientific collocation of fertilizer and water resources should be seriously considered for economic and environmental benefits. Overall, the optimized inputs of the FIs were in reasonable ranges, as postulated by previous studies. This result hints at the potential applications of LSMs for guiding sustainable agricultural development.

Greenery is a critical referent for assessing the health and sustainability of urban ecosystems. However, distinct urbanization pathways of cities can be accompanied by complicated encroachment or regeneration of green spaces, thereby leading to certain spatiotemporal diversity in greening signatures at both local and regional scales. Here, this study conducted a long-term (2000–2020) case analysis to evaluate the greening signatures and responsible drivers across the urban–rural gradient of Chinese cities. The major results showed that 1) Most case cities showed general but uneven greening trends. However, the urban–rural gradient shows a Sandwich pattern, i.e., the greening rate is significantly higher in developed and rural areas than in developing regions. 2) Climate change, especially temperature variation, performs as the dominant driver responsible for local greening, followed by anthropogenic-induced landscape alterations. 3) Especially, greening process in urbanized regions was more sensitive to climatic perturbations and less responsive to landscape changes over time, while that in rural regions was relatively stable. In the context of overall greening in Chinese cities, one should be aware of the uneven greening progress along the urban–rural gradient to proactively address the potential challenges of future climate and landscape changes for sustainable urban development.

Abstract Background and aims This study aimed to investigate the association between the Dietary Inflammatory Index (DII) and dyslipidemia, as well as to evaluate the mortality risk associated with DII in participants with dyslipidemia. Methods Data from the National Health and Nutrition Examination Survey database were divided into dyslipidemia and non-dyslipidemia groups. The association between DII and dyslipidemia was investigated using the weighted chi-square test, weighted t-test, and weighted logistic regression. Weighted Cox proportional hazards models were used to estimate the hazard ratios and 95% confidence intervals for all-cause and cardiovascular disease-related mortality within the dyslipidemia group. Results A total of 17,820 participants, including 4,839 without and 12,981 with dyslipidemia were analyzed in this study. In the Yangtze River Delta urban agglomeration, which is the region with the highest urbanization intensity in China, the development of cities leads to changes in land surface temperature (LST), while vegetation phenology varies with LST. To investigate the spatial and temporal changes in vegetation phenology and its response to LST in the study area, this study reconstructed the time series of the enhanced vegetation index (EVI) based on the MODIS EVI product and extracted the vegetation phenology indicators in the study area from 2002 to 2020, including the start of the growing season (SOS), the end of the growing season (EOS), and the growing season length (GSL), and analyzed the temporal–spatial patterns of vegetation phenology and LST in the study area, as well as the correlation between them. The results show that (1) SOS was advanced, EOS was postponed, and GSL was extended in the study area from 2002 to 2020, and there were obvious differences in the vegetation phenology indicators under different land covers and cities; (2) LST was higher in the southeast than in the northwest of the study area from 2002 to 2020, with an increasing trend; and (3) there are differences in the response of vegetation phenology to LST across land covers and cities, and SOS responds differently to LST at different times of the year. EOS shows a significant postponement trend with the annual mean LST increase. Overall, we found differences in vegetation phenology and its response to LST under different land covers and cities, which is important for scholars to understand the response of vegetation phenology to urbanization.

Environmental electromagnetic interference (EMI) has always been a major interference source for multiple-channel neural recording systems, and little theoretical work has been attempted to address it. In this paper, equivalent circuit models are proposed to model both electromagnetic interference sources and neural signals in such systems, and analysis has been performed to generate the design guidelines for neural probes and the subsequent recording circuit towards higher common-mode interference (CMI) rejection performance while maintaining the recorded neural action potential (AP) signal quality. In vivo animal experiments with a configurable 32-channel neural recording system are carried out to validate the proposed models and design guidelines. The results show the power spectral density (PSD) of environmental 50 Hz EMI interference is reduced by three orders from 4.43 × 10−3 V2/Hz to 4.04 × 10−6 V2/Hz without affecting the recorded AP signal quality in an unshielded experiment environment.

Abstract Background One-stage treatment involving rotator cuff repair and shoulder capsule release is mainly used to treat patients with rotator cuff tears (RCTs) and concomitant shoulder stiffness. Despite the increasing attention to the efficacy and safety of one-stage treatment, controversy still remains. Therefore, this systematic review aims to summarize the indications, operation procedure and rehabilitation protocol, and compare the range of motions (ROMs), functional outcomes and retear rates of one-stage treatment for RCTs in stiff shoulders and non-stiff shoulders. Methods Multiple databases (PubMed, the Cochrane Library, Embase and MEDLINE) were searched for studies that investigated outcomes after one-stage treatment for RCTs concomitant with shoulder stiffness compared with rotator cuff repair for RCTs alone, according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses criteria. Descriptive statistics, including range of motion, patient-reported outcome and retear rate, are presented without meta-analysis due to the heterogeneity and low levels of evidence. Results A total of 9 cohort studies were included, with 305 patients treated with one-stage treatment involving rotator cuff repair and simultaneous shoulder capsular release and 1059 patients treated with rotator cuff repair alone. Patients in both groups had significant symptom improvement and functional recovery after the one-stage treatment for the stiffness group and standard repair for the non-stiffness group, and most patients could return to normal life and work within 6 months after the operation. The retear rate in the one-stage treatment group was not higher than that in the rotator cuff repair group. No statistically significant differences between the two groups were observed in terms of range of motion and patient-reported outcomes in the vast majority of studies at the final follow-up, including the visual analog scale for pain, the Constant score, the American Shoulder and Elbow Surgeons score, the University of California Los Angeles Shoulder Score, the Oxford shoulder score and the Simple Shoulder Test. Conclusion One-stage treatment for RCTs in stiff shoulders provides comparable ROM and patient-reported clinical outcomes as rotator cuff repair for non-stiff RCTs. In addition, the rate of postoperative retear in stiff shoulder treated with one-stage treatment was not higher than in non-stiff shoulders.

Abstract Background Complex immune-brain interactions that affect neural development, survival and function might have causal and therapeutic implications for psychiatric illnesses. However, previous studies examining the association between immune inflammation and schizophrenia (SCZ) have yielded inconsistent findings. Methods Comprehensive two-sample Mendelian randomization (MR) analysis was performed to determine the causal association between immune cell signatures and SCZ in this study. Based on publicly available genetic data, we explored causal associations between 731 immune cell signatures and SCZ risk. A total of four types of immune signatures (median fluorescence intensities (MFI), relative cell (RC), absolute cell (AC), and morphological parameters (MP)) were included. Comprehensive sensitivity analyses were used to verify the robustness, heterogeneity, and horizontal pleiotropy of the results. Results After FDR correction, SCZ had no statistically significant effect on immunophenotypes. It was worth mentioning some phenotypes with unadjusted low P-values, including FSC-A on NKT (β = 0.119, 95% CI = 0.044 ~ 0.194, P = 0.002), DN (CD4-CD8-) NKT %T cell (β = 0.131, 95% CI = 0.054 ~ 0.208, P = 9.03 × 10− 4), and SSC-A on lymphocytes (β = 0.136, 95% CI = 0.059 ~ 0.213, P = 5.43 × 10− 4). The causal effect of SCZ IgD on transitional was estimated to 0.127 (95% CI = 0.051 ~ 0.203, P = 1.09 × 10− 3). SCZ also had a causal effect on IgD+ %B cell (β = 0.130, 95% CI = 0.054 ~ 0.207, P = 8.69 × 10− 4), and DP (CD4+CD8+) %T cell (β = 0.131, 95% CI = 0.054 ~ 0.207, P = 8.05 × 10− 4). Furthermore, four immunophenotypes were identified to be significantly associated with SCZ risk: naive CD4+ %T cell (OR = 0.986, 95% CI = 0.979 ~ 0.992, P = 1.37 × 10− 5), HLA DR on CD14− CD16− (OR = 0.738 (95% CI = 0.642 ~ 0.849, P = 2.00 × 10− 5), CD33dim HLA DR+ CD11b− AC (OR = 0.631, 95% CI = 0.529 ~ 0.753, P = 3.40 × 10− 7) and activated & resting Treg % CD4 Treg (OR = 0.937, 95% CI = 0.906 ~ 0.970, P = 1.96 × 10− 4). Conclusions Our study has demonstrated the close connection between immune cells and SCZ by genetic means, thus providing guidance for future clinical research.

Abstract Media convergence is a media change led by technological innovation. Applying media convergence technology to the study of clustering in Chinese medicine can significantly exploit the advantages of media fusion. Obtaining consistent and complementary information among multiple modalities through media convergence can provide technical support for clustering. This article presents an approach based on Media Convergence and Graph convolution Encoder Clustering (MCGEC) for traditional Chinese medicine (TCM) clinical data. It feeds modal information and graph structure from media information into a multi‐modal graph convolution encoder to obtain the media feature representation learnt from multiple modalities. MCGEC captures latent information from various modalities by fusion and optimises the feature representations and network architecture with learnt clustering labels. The experiment is conducted on real‐world multi‐modal TCM clinical data, including information like images and text. MCGEC has improved clustering results compared to the generic single‐modal clustering methods and the current more advanced multi‐modal clustering methods. MCGEC applied to TCM clinical datasets can achieve better results. Integrating multimedia features into clustering algorithms offers significant benefits compared to single‐modal clustering approaches that simply concatenate features from different modalities. It provides practical technical support for multi‐modal clustering in the TCM field incorporating multimedia features.

Mitochondria-associated membrane (MAM) serve as crucial contact sites between mitochondria and the endoplasmic reticulum (ER). Recent research has highlighted the significance of MAM, which serve as a platform for various protein molecules, in processes such as calcium signaling, ATP production, mitochondrial structure and function, and autophagy. Cardiac diseases caused by any reason can lead to changes in myocardial structure and function, significantly impacting human health. Notably, MAM exhibits various regulatory effects to maintain cellular balance in several cardiac diseases conditions, such as obesity, diabetes mellitus, and cardiotoxicity. MAM proteins independently or interact with their counterparts, forming essential tethers between the ER and mitochondria in cardiomyocytes. This review provides an overview of key MAM regulators, detailing their structure and functions. Additionally, it explores the connection between MAM and various cardiac injuries, suggesting that precise genetic, pharmacological, and physical regulation of MAM may be a promising strategy for preventing and treating heart failure.

The ecological security of oasis cities in arid and semi-arid regions is highly susceptible to changes in regional landscape patterns and the degree of coordination between human activities and the environment. At the same time, the ecological security of urban landscapes also profoundly affects the success of regional economic and environmental coordination and development. This study is based on land use data from 1990, 2000, 2010, and 2020, as well as land use data from the natural development scenario (NLD), economic development scenario (ECD), ecological development scenario (ELD) and ecological-economic development scenario (EED) simulated by the patch-generating land use simulation (PLUS) model in 2030. From the perspective of production-living-ecological land (PLEL), it analyzes the changes in the past and future landscape ecological security and coupling coordination characteristics of Bole. The results show that from 1990 to 2020, Bole was mainly dominated by grassland ecological land (GEL) and other ecological land (OEL), accounting for a total proportion of 69.51%, with a large increase in production and living land area; the average value of landscape ecological risk is decreasing, and the landscape ecological security of Bole is developing towards benignity; the area of highly coupled coordination zone is decreasing continuously, while that of basic coordination zone and moderate coordination zone is increasing continuously. Under the 2030 EED scenario, the overall changes in various types of land use are not significant, and the average value of landscape ecological risk is the smallest, but it is higher than that in 2020 as a whole; under EED scenario, the area of highly coordinated zone and moderate coordinated zone is the largest among four scenarios, and the best coupling coordination level among the four scenarios. Landscape ecological security and its coupling coordination will be affected by land use patterns. Optimizing regional land use patterns is of great significance for improving urban landscape ecological security and sustainable high-quality development.

Hypertension constitutes a pervasive chronic ailment on a global scale, frequently inflicting damage upon vital organs, such as the heart, blood vessels, kidneys, brain, and others. And this is a complex clinical dilemma that requires immediate attention. The mitochondria assume a crucial function in the generation of energy, and it is of utmost importance to eliminate any malfunctioning or surplus mitochondria to uphold intracellular homeostasis. Mitophagy is considered a classic example of selective autophagy, an important component of mitochondrial quality control, and is closely associated with many physiological and pathological processes. The ubiquitin-dependent pathway, facilitated by PINK1/Parkin, along with the ubiquitin-independent pathway, orchestrated by receptor proteins such as BNIP3, NIX, and FUNDC1, represent the extensively investigated mechanisms underlying mitophagy. In recent years, research has increasingly shown that mitophagy plays an important role in organ damage associated with hypertension. Exploring the molecular mechanisms of mitophagy in hypertension-mediated organ damage could represent a critical avenue for future research in the development of innovative therapeutic modalities. Therefore, this article provides a comprehensive review of the impact of mitophagy on organ damage due to hypertension.

BackgroundActual motor competence (AMC) and perceived motor competence (PMC), as determinants of children’s active participation in physical activity (PA), were important for children’s healthy development. The correlation between the two had been confirmed. To further understand this relationship, this study investigated the current status of AMC, PMC, and PA in Chinese school-age children, used a person-centered approach to reveal the characteristics of the development of motor competence (MC) in Chinese school-age children and the differences in the level of PA among different MC profiles of children.Materials and methodsA total of 532 children (age: M = 9.37, SD = 1.80 years-old) from grades 1 to 6 participated in this cross-sectional study (male, n = 284, 53.4%; female, n = 248, 46.6%). The Test of Gross Motor Development-3 (TGMD-3) was used to measure children’s AMC, the Pictorial Scale of Perceived Movement Skill Competence (PMSC) to measure children’s PMC, and the revised Chinese version of Physical Activity Questionnaire for Older Children (PAQ-C) to assess children’s PA levels.ResultsThere were some gender differences in AMC but no significant gender differences in PMC and PA in children. AMC and PA levels increased as the children aged, while PMC showed some decline. Cluster analysis identified four groups of children with different MC profiles. Two groups of children had corresponding AMC and PMC levels (the “high-high” cluster, N = 200, 38.91%; the “low-low” cluster, N = 63, 12.26%), and the other two groups were inconsistent in AMC and PMC (the “high-low” cluster, N = 100, 19.46%; the “low-high” cluster, N = 151, 29.38%). Significant differences in PA levels were found between children with different MC profiles. The “high-high” cluster children had the highest PA levels, whereas the “low-low” cluster children demonstrated the lowest PA levels.ConclusionAMC, PMC, and PA in Chinese school-age children were consistent with the pattern of child growth and development. Children with high AMC and high PMC usually had high levels of PA. Therefore, it was recommended to seize the best opportunity to intervene with children, and family, school, and community should synergize to help children improve AMC and PMC, and then actively participate in PA.

Abstract Transit-time ultrasonic flowmeters (TTUFs) are among the most widely used devices for flow measurements. However, traditional TTUFs are usually based on a bulk piezoelectric transducer, which limits their application in small-diameter channels. In this paper, we developed a miniaturized TTUF based on scandium-doped aluminum nitride (ScAlN) piezoelectric micromachined ultrasonic transducers (PMUTs). The proposed TTUF contains two PMUT-based transceivers and a π-type channel. The PMUTs contain 13 × 13 square cells with dimensions of 2.8 × 2.8 mm2. To compensate for the acoustic impedance mismatch with liquid, a layer of polyurethane is added to the surface of the PMUTs as a matching layer. The PMUT-based transceivers show good transmitting sensitivity (with 0.94 MPa/V surface pressure) and receiving sensitivity (1.79 mV/kPa) at a frequency of 1 MHz in water. Moreover, the dimensions of the π-type channel are optimized to achieve a measurement sensitivity of 82 ns/(m/s) and a signal-to-noise ratio (SNR) better than 15 dB. Finally, we integrate the fabricated PMUTs into the TDC-GP30 platform. The experimental results show that the developed TTUF provides a wide range of flow measurements from 2 to 300 L/h in a channel of 4 mm diameter, which is smaller than most reported channels. The accuracy and repeatability of the TTUF are within 0.2% and 1%, respectively. The proposed TTUF shows great application potential in industrial applications such as medical and chemical applications.

Abstract RNF2 is a RING domain-containing E3 ubiquitin ligase that mediate histone H2A mono-ubiquitination to repress gene transcription, but its expression patterns and molecular function in hepatocellular carcinoma (HCC) remain unclear. Herein, we extracted data from TGCA database and validated RNF2 expression in our own cohort, which revealed that RNF2 was highly expressed in HCC and was associated with malignant characteristics and poor prognosis of HCC. Moreover, RNF2 was demonstrated to promote HCC metastasis via enhancing epithelial-mesenchymal transition (EMT) both in vitro and in vivo. Mechanistically, RNF2 repressed E-Cadherin transcription by increasing the deposition of H2K119ub at the E-Cadherin promoter region. In addition, RNF2-regulated crosstalk between H2AK119ub, H3K27me3 and H3K4me3 synergistically reduced E-Cadherin transcription, which promoted EMT and HCC metastasis. These results indicate that RNF2 played an oncogenic role in HCC progression via inducing EMT, and RNF2 could be a potential therapeutic target for HCC.

Abstract Background Knee arthroscopic arthrolysis serves as an effective treatment for knee arthrofibrosis. However, hemarthrosis is the most common complication in arthroscopic surgery, which has potential adverse effects on postoperative rehabilitation. The purpose of this study was to evaluate the effects of topical tranexamic acid (TXA) in knee arthroscopic arthrolysis. Methods A total of 87 patients with knee arthrofibrosis who underwent arthroscopic arthrolysis from September 2019 to June 2021 were eligible for this retrospective review. Patients in the TXA group (n = 47) received topical administration of TXA (50 mL, 10 mg/mL) at the end of the surgery, and patients in the control group (n = 40) received no TXA. The postoperative drainage volumes, hematologic levels, inflammatory marker levels, knee range of motion (ROM), visual analog scale (VAS) pain scores, Lysholm knee scores and complications were compared between the two groups. The curative effect of each group was calculated according to Judet’s criteria. With the development of smart grids and new power systems, the combination of non-intrusive load identification technology and smart home technology can provide users with the operating conditions of home appliances and equipment, thus reducing home energy loss and improving users’ ability to demand a response. This paper proposes a non-intrusive load decomposition model with a parallel multiscale attention mechanism (PMAM). The model can extract both local and global feature information and fuse it through a parallel multiscale network. This improves the attention mechanism’s ability to capture feature information over long time periods. To validate the model’s decomposition ability, we combined the PMAM model with four benchmark models: the Long Short-Term Memory (LSTM) recurrent neural network model, the Time Pooling-based Load Disaggregation Model (TPNILM), the Extreme Learning Machine (ELM), and the Load Disaggregation Model without Parallel Multi-scalar Attention Mechanisms (UNPMAM). The model was trained on the publicly available UK-DALE dataset and tested. The models’ test results were quantitatively evaluated using a confusion matrix. This involved calculating the F1 score of the load decomposition. A higher F1 score indicates better model decomposition performance. The results indicate that the PMAM model proposed in this paper maintains an F1 score above 0.9 for the decomposition of three types of electrical equipment under the same household user, which is 3% higher than that of the other benchmark models on average. In the cross-household test, the PMAM also demonstrated a better decomposition ability, with the F1 score maintained above 0.85, and the mean absolute error (MAE) decreased by 5.3% on average compared with that of the UNPMAM.

The construction of highway microgrids is evolving into a new highway energy system that integrates “Source-Network-Load-Storage”. This paper provides a comprehensive evaluation of expressway microgrids from the perspective of transportation and energy integration. An index model is set up that considers the economy, technology, and environment. The grey evaluation method, on the strength of analytic hierarchy process–entropy weight method, shows that the integrated microgrid of “source-network-load-storage” promotes energy sustainability, supply reliability, and sustainable environmental development. When compared to different evaluation methods for microgrid planning schemes, our method yields the highest score of 0.9127, indicating superior results. This approach is suitable for the multicomponent evaluation system of expressway microgrids and allows for scientifically evaluating microgrid planning schemes.

The global COVID-19 pandemic has presented a significant, ongoing challenge since its emergence in late 2019. Today, the Omicron strain, which is less lethal but more contagious than the original outbreak strain, continues to pose substantial health risks. In this background, the management of psoriatic comorbidities has become even more complex, particularly for patients with underlying inflammatory, metabolic, or cardiovascular diseases. This review aims to summarize current research on comorbid COVID-19 and psoriasis, and provide insights into the development of evidence-based management strategies. By providing appropriate patient instruction, implementing protective measures, and re-evaluating medication prescriptions based on each patient’s unique situation, healthcare professionals can effectively address the challenges faced by patients with comorbid psoriasis in the COVID-19 era.

Polygonum perfoliatum L. is an herbal medicine that has been extensively used in traditional Chinese medicine to treat various health conditions ranging from ancient internal to surgical and gynecological diseases. Numerous studies suggest that P. perfoliatum extract elicits significant anti-tumor, anti-inflammatory, anti-bacterial, and anti-viral effects. Nevertheless, the underlying mechanisms of its anti-liver cancer effects remain poorly understood. Our study suggests that P. perfoliatum stem extract (PPLA) has a favorable safety profile and exhibits a significant anti-liver cancer effect both in vitro and in vivo. We identified that PPLA activates the cGMP-PKG signaling pathway, and key regulatory genes including ADRA1B, PLCB2, PRKG2, CALML4, and GLO1 involved in this activation. Moreover, PPLA modulates the expression of genes responsible for the cell cycle. Additionally, we identified four constituents of PPLA, namely taxifolin, myricetin, eriodictyol, and pinocembrin, that plausibly act via the cGMP-PKG signaling pathway. Both in vitro and in vivo experiments confirmed that PPLA, along with its constituting compounds taxifolin, myricetin, and eriodictyol, exhibit potent anti-cancer activities and hold the promise of being developed into therapeutic agents.

Highlights A new class of γ-cyclodextrin based metal–organic framework -derived strategy was used to fabricate highly active M–N-C catalysts containing both single-atom sites and nanoparticles. The obtained Co@C-CoNC exhibits remarkable bifunctional oxygen reduction (ORR)/oxygen evolution reactions (OER) performance, which further delivered a high power density and excellent cyclic stability in rechargeable zinc-air batteries. Density functional theory calculation suggests that the mutual self-regulation of d-electron density of both Co NP and Co SAC conjointly reduce the reaction energy barriers and thus boost the ORR/OER kinetics through their fast adsorption/desorption ability of reaction intermediates.

Abstract Background Cuproptosis is a new type of copper-induced cell death that is characterized by the aggregation of lipoylated tricarboxylic acid (TCA) cycle proteins. However, its role in hepatocellular carcinoma (HCC) remains unclear. The goal of this research is to develop a cuproptosis-related signature predicting the prognosis of HCC. Methods The cuproptosis-related genes were defined using Pearson correlation coefficients. LASSO-Cox regression analysis was used to evaluate the prognostic values of cuproptosis-related genes to construct a cuproptosis-related prognostic model. The immune microenvironment analysis was performed by “ssGSEA” to reveal the associated immune cell infiltration patterns with the cuproptosis-related genes signature. The expression levels of one of the prognostic genes PDXK were then verified in HCC samples by Western Blot and immunohistochemistry. The potential roles of target genes in cuproptosis were further explored during in-vitro experiments. Results A total of 136 cuproptosis-related genes were discovered using Pearson correlation analysis in HCC. A cuproptosis-related signature that included 5 cuproptosis-related genes (PDXK, HPN, SLC25A28, RNFT1, CLEC3B) was established in the TCGA-LIHC training cohort. TCGA validation cohort and another two external validation cohorts confirmed the robustness of the signature’s predictive value. Moreover, a nomogram using the risk score was created to best predict the survival of HCC patients. The immune microenvironment analysis revealed distinct immune infiltrations patterns between different risk groups based on the signature model. Furthermore, the upregulation of PDXK was confirmed in HCC tumor tissues in 30 clinical HCC specimens. The knockdown of PDXK reduced the proliferation, migration and invasion of HCC cells. Besides, the expression of PDXK was upregulated after the induction of cuproptosis by elesclomol–CuCL2, which could be suppressed when pretreated with a copper ion chelator. And PDXK deficiency increased the sensitivity of HCC cells to cuproptosis inducer. Conclusion Our study identified a new cuproptosis-related gene signature that could predict the prognosis of HCC patient. Besides, the upregulated PDXK could promote the proliferation and metastasis of HCC. And PDXK deficiency facilities cuproptosis in HCC. Therefore, these fundings highlighted that PDXK might serve as a potential diagnostic and therapeutic target for HCC.

The LD-14 radar used can realize the functions of autonomous follow-up and pedestrian obstacles. When a sensor in the system fails or the detection deviation is too large, the multi-sensor fusion technology can ensure the normal operation of the system due to a certain degree of redundant. Context Salvianolic acid B (SalB) can attenuate myocardial ischemia/reperfusion (I/R) injury, but the mechanisms are not entirely known.Objective Our study investigates if SalB protects cardiomyocytes against I/R injury by regulating Tripartite motif (TRIM) protein.Materials and methods AC16 cardiomyocytes were treated with I/R, and then with SalB (10, 25 and 50 μM) for 24 h, while control cells were cultured under normal conditions. Female Sprague-Dawley rats were subjected to I/R injury, and then intravenously injected with 20, 40, or 60 mg/kg SalB or saline, as a control, rats received sham operation and saline injection.Results Upon treatment, apoptotic rate, reactive oxygen species (ROS), and malondialdehyde (MDA) were increased 10-, 3.8-, and 1.3-fold, respectively, while superoxide dismutase (SOD) activity was reduced by 62.1% compared to control cells. I/R treatment elevated the mRNA and protein expression of TRIM8. SalB treatment remarkably abolished the above-mentioned effects of I/R treatment. TRIM8 knock-down could partially alleviate I/R-induced myocardial injury. TRIM8 overexpression promoted cardiomyocyte injury, which was alleviated by SalB. Moreover, TRIM8 negatively regulated protein expression of antioxidant enzyme glutathione peroxidase 1 (GPX1). TRIM8 protein interacted with GPX1 and TRIM8 overexpression promoted GPX1 ubiquitnation. GPX1 knock-down abolished the protective effects of SalB on I/R-injured cardiomyocytes. Our in vivo experiments confirmed the effects of SalB on I/R-induced myocardial injury.Discussion and conclusions SalB protected cardiomyocytes from I/R-induced apoptosis and oxidative stress in vitro and in vivo, which was partly mediated by the TRIM8/GPX1 axis. This suggests that down-regulation of TRIM8 expression may ameliorate I/R-induced myocardial injury.

IntroductionIn urban road scenes, due to the small size of traffic signs and the large amount of surrounding interference information, current methods are difficult to achieve good detection results in the field of unmanned driving.MethodsTo address the aforementioned challenges, this paper proposes an improved E-YOLOv4-tiny based on the YOLOv4-tiny. Firstly, this article constructs an efficient layer aggregation lightweight block with deep separable convolutions to enhance the feature extraction ability of the backbone. Secondly, this paper presents a feature fusion refinement module aimed at fully integrating multi-scale features. Moreover, this module incorporates our proposed efficient coordinate attention for refining interference information during feature transfer. Finally, this article proposes an improved S-RFB to add contextual feature information to the network, further enhancing the accuracy of traffic sign detection.Results and discussionThe method in this paper is tested on the CCTSDB dataset and the Tsinghua-Tencent 100K dataset. The experimental results show that the proposed method outperforms the original YOLOv4-tiny in traffic sign detection with 3.76% and 7.37% improvement in mAP, respectively, and 21% reduction in the number of parameters. Compared with other advanced methods, the method proposed in this paper achieves a better balance between accuracy, real-time performance, and the number of model parameters, which has better application value.

ObjectiveTo explore the pattern of empathy characteristics in male patients with schizophrenia (SCH) and to examine whether empathy deficit is associated with impulsivity and premeditated violence.MethodsOne hundred and fourteen male SCH patients were enrolled in this study. The demographic data of all patients were collected and the subjects were divided into two groups, namely, the violent group, including 60 cases, and the non-violent group, comprising 54 cases, according to the Modified Overt Aggression Scale (MOAS). The Chinese version of the Interpersonal Reactivity Index-C (IRI-C) was used to evaluate empathy and the Impulsive/Predicted Aggression Scales (IPAS) was employed to assess the characteristics of aggression.ResultsAmong the 60 patients in the violent group, 44 patients had impulsive aggression (IA) and 16 patients had premeditated aggression (PM) according to the IPAS scale. In the violent group, the scores of the four subfactors of the IRI-C, i.e., perspective taking (PT), fantasy (FS), personal distress (PD), and empathy concern (EC), were significantly lower than in the non-violent group. Stepwise logistic regression showed that PM was independent influencing factor for violent behaviors in SCH patients. Correlation analysis revealed that EC of affective empathy was positively correlated with PM but not with IA.ConclusionSCH patients with violent behavior had more extensive empathy deficits compared with non-violent SCH patients. EC, IA and PM are independent risk factors of violence in SCH patients. Empathy concern is an important index to predict PM in male patients with SCH.

The increasing number of electric vehicles (EVs) on the roads has led to a rise in the number of batteries reaching the end of their first life. Such batteries, however, still have a capacity of 75–80% remaining, creating an opportunity for a second life in less power-intensive applications. Utilising these second-life batteries (SLBs) requires specific preparation, including grading the batteries based on their State of Health (SoH); repackaging, considering the end-use requirements; and the development of an accurate battery-management system (BMS) based on validated theoretical models. In this paper, we conduct a technical review of mathematical modelling and experimental analyses of SLBs to address existing challenges in BMS development. Our review reveals that most of the recent research focuses on environmental and economic aspects rather than technical challenges. The review suggests the use of equivalent-circuit models with 2RCs and 3RCs, which exhibit good accuracy for estimating the performance of lithium-ion batteries during their second life. Furthermore, electrochemical impedance spectroscopy (EIS) tests provide valuable information about the SLBs’ degradation history and conditions. For addressing calendar-ageing mechanisms, electrochemical models are suggested over empirical models due to their effectiveness and efficiency. Additionally, generating cycle-ageing test profiles based on real application scenarios using synthetic load data is recommended for reliable predictions. Artificial intelligence algorithms show promise in predicting SLB cycle-ageing fading parameters, offering significant time-saving benefits for lab testing. Our study emphasises the importance of focusing on technical challenges to facilitate the effective utilisation of SLBs in stationary applications, such as building energy-storage systems and EV charging stations.

The frequency of compound hot–dry events ( f _HD ) is projected to increase significantly with future warming, yet associated uncertainties remain considerable and poorly constrained. In this study, we constrain future projections of f _HD (2070–2099) using observations of recent trends in temperature ( T ) and precipitation ( P ) (1980–2014) during the warm-seasons. The physical mechanism is that the variance of f _HD across climate models is dominated by their projected changes in P (Δ P ), which can be constrained by recent trends in T and P . Compared to the raw projections, the observationally constrained f _HD is reduced by 9.68%–18.74%, with uncertainty narrowed by 3.79%–10.66% under the high emission scenario. The highest decline of f _HD is located in regions with low population and gross domestic product (GDP), and globally, population and GDP exposures to f _HD are reduced by 6.02%–10.73% and 6.51%–12.03%, respectively. The observationally constrained f _HD with lower uncertainty provides more reliable information for risk management under climate change.
