Panose is iso-malto-oligosaccharide contained α-1,6 glycosidic bonds, and α-glucosidase is the key enzyme to produce panose. In this study, the α-glucosidase gene in deep-sea hydrothermal vents thermophilic archaea Thermococcus siculi HJ21 was synthesized, and cloned into the vector pET29a. Then expressed in Escherichia coli BL21 and purified by His Trap HP column. The molecular weight of α-glucosidase was determined by SDS-PAGE. The enzymatic properties and transglycosylation were also investigated. The results showed that the gene of α-glucosidase was 729 bp and encoded 242 amino acids. The molecular weight of α-glucosidase was about 27.2 kDa. Its optimal temperature and pH were 40 ℃ and 6.0 respectively. When the receptor was fructose, and the ratio of fructose and maltose was 1:9, the highest yield of panose could reached 79.1% after 10 h reaction. The results provide a basis for producing high-purity panose by α-glucosidase from archaea.

Abstract Purpose Identifying primary hepatic angiosarcoma (PHA) preoperatively is challenging, often relying on postoperative pathology. Invasive biopsy increases bleeding risk, emphasizing the importance of early PHA diagnosis through imaging. However, comprehensive summaries of ultrasound, abdominal computed tomography (CT), magnetic resonance imaging (MRI), and whole- body positron emission tomography-CT (PET-CT) in this context are lacking. This study aimed to investigate the comprehensive imaging characteristics of PHA. Patients and methods Imaging data were collected from 7 patients diagnosed with PHA via pathology between January 2000 and December 2019 in two provincial grade III hospitals. All patients underwent routine color ultrasound examinations before surgery, with 3 patients receiving contrast-enhanced ultrasound (CEUS).CT scans, both plain and enhanced, were performed on 5 patients, and whole-body PET-CT examinations were conducted on 2 patients. Results Among the 7 patients with PHA, 4 presented with a single solid intrahepatic mass (2 of which were large), 1 with a single exophytic macroblock type, 1 with a mixed type featuring multiple masses and nodules, and 1 with a multiple nodule type. Conventional ultrasound of PHA showed uneven echoes within the tumor, potentially accompanied by septal zone echoes, and a blood flow grade of 0-I. CEUS displayed early-stage circular high enhancement, a central non-enhancement area, and a "vascular sign" around the tumor. CT scans revealed low-density shadows in the plain scan stage, high peripheral ring enhancement, and punctate nodular enhancement in the arterial phase, with varying intensities and the presence of a "vascular sign." During the portal vein stage, the interior of the tumor was consistently unfilled and exhibited structural disorder. PET-CT showed low-density lesions in the liver and low fluorodeoxyglucose metabolism. Conclusions Imaging diagnosis plays a crucial role in PHA diagnosis. When liver tumor imaging matches the above characteristics, consider PHA.

Abstract Although previous research has shown that inflammation is associated with development of colorectal cancer (CRC), questions remain about whether inflammatory factor-secreting bacteria play a crucial role in CRC development. The potential role of gut microbiota in secreting inflammatory factors involved in the carcinogenesis of CRC among Chinese patients was explored in this study. 16S rRNA sequencing was utilized to evaluate the distinct microbial characteristics between patients with CRC and colorectal adenoma. The serum levels of TNF-α, IL-6 and IL-10 were measured using Enzyme-linked immunosorbent assay (ELISA), while the expression of LRG1 and TGF-β1 in tissues was evaluated by immunohistochemistry. The correlation between gut microbiota and inflammatory factor signaling was analyzed. Compared with the adenoma group, CRC patients exhibit distinct pathologies. Moreover, elevated levels of CEA, erythrocytes and haemoglobin in the blood of CRC patients were found. In addition, CRC patients have significantly higher levels of TNF-α, IL-6, IL-10, LRG1 and TGF-β1. Spearman correlation analysis revealed that LRG1 was positively related to IL-6 and TNF-α, respectively. The correlation analysis results of TGF-β1 were consistent with the above. The abundance of Blautia and Streptococcus was lower in CRC patients, while the relative abundance of Alistipes, Peptostreptococcus and Porphyromonas was significantly elevated. Moreover, positive correlations between Alistipes and inflammatory factor signaling were also found. Our results suggest that gut commensal Alistipes is a key bacterium with pro-inflammatory properties in the CRC carcinogenesis. TNF-α and IL-6 associated with Alistipes might activate LRG1/TGF-β1 signaling which contributed to the carcinogenesis of CRC.

Abstract Background Acute myeloid leukaemia (AML) comprises a group of heterogeneous and aggressive haematological malignancies with unsatisfactory prognoses and limited treatment options. Treatments targeting B-cell lymphoma-2 (BCL-2) with venetoclax have been approved for patients with AML, and venetoclax-based drug combinations are becoming the standard of care for older patients unfit for intensive chemotherapy. However, the therapeutic duration of either single or combination strategies is limited, and the development of resistance seems inevitable. Therefore, more effective combination regimens are urgently needed. Methods The efficacy of combination therapy with NL101, a SAHA-bendamustine hybrid, and venetoclax was evaluated in preclinical models of AML including established cell lines, primary blasts from patients, and animal models. RNA-sequencing and immunoblotting were used to explore the underlying mechanism. Results NL101 significantly potentiated the activity of venetoclax in AML cell lines, as evidenced by the enhanced decrease in viability and induction of apoptosis. Mechanistically, the addition of NL101 to venetoclax decreased the stability of the antiapoptotic protein myeloid cell leukaemia-1 (MCL-1) by inhibiting ERK, thereby facilitating the release of BIM and triggering mitochondrial apoptosis. Moreover, the strong synergy between NL101 and venetoclax also relied on the downregulation of c-Myc via PI3K/Akt/GSK3β signalling. The combination of NL101 and venetoclax synergistically eliminated primary blasts from 10 AML patients and reduced the leukaemia burden in an MV4-11 cell-derived xenograft model. Conclusions Our results encourage the pursuit of clinical trials of combined treatment with NL101 and venetoclax and provide a novel venetoclax-incorporating therapeutic strategy for AML.

Abstract In the era of 5 G, the rise in power density in miniaturized, flexible electronic devices has created an urgent need for thin, flexible, polymer-based electrically and thermally conductive nanocomposites to address challenges related to electromagnetic interference (EMI) and heat accumulation. However, the difficulties in establishing enduring and continuous transfer pathways for electrons and phonons using solid-rigid conductive fillers within insulative polymer matrices limit the development of such nanocomposites. Herein, we incorporate MXene-bridging-liquid metal (MBLM) solid-liquid bi-continuous electrical-thermal conductive networks within aramid nanofiber/polyvinyl alcohol (AP) matrices, resulting in the AP/MBLM nanocomposite with ultra-high electrical conductivity (3984 S/cm) and distinguished thermal conductivity of 13.17 W m−1 K−1. This nanocomposite exhibits excellent EMI shielding efficiency (SE) of 74.6 dB at a minimal thickness of 22 μm, and maintains high EMI shielding stability after enduring various harsh conditions. Meanwhile, the AP/MBLM nanocomposite also demonstrates promising heat dissipation behavior. This work expands the concept of creating thin films with high electrical and thermal conductivity.

Abstract Hydrogels, provided that they integrate strength and toughness at desired high content of water, promise in load-bearing tissues such as articular cartilage, ligaments, tendons. Many developed strategies impart hydrogels with some mechanical properties akin to natural tissues, but compromise water content. Herein, a strategy deprotonation-complexation-reprotonation is proposed to prepare polyvinyl alcohol hydrogels with water content as high as ~80% and favorable mechanical properties, including tensile strength of 7.4 MPa, elongation of around 1350%, and fracture toughness of 12.4 kJ m−2. The key to water holding yet improved mechanical properties lies in controllable nucleation for refinement of crystalline morphology. With nearly constant water content, mechanical properties of as-prepared hydrogels are successfully tailored by tuning crystal nuclei density via deprotonation degree and their distribution uniformity via complexation temperature. This work provides a nucleation concept to design robust hydrogels with desired water content, holding implications for practical application in tissue engineering.

Abstract This study aimed to develop and validate a predictive model of all-cause mortality risk in American adults aged ≥ 18 years with diabetes. 7918 participants with diabetes were enrolled from the National Health and Nutrition Examination Survey (NHANES) 1999–2016 and followed for a median of 96 months. The primary study endpoint was the all-cause mortality. Predictors of all-cause mortality included age, Monocytes, Erythrocyte, creatinine, Nutrition Risk Index (NRI), neutrophils/lymphocytes (NLR), smoking habits, alcohol consumption, cardiovascular disease (CVD), urinary albumin excretion rate (UAE), and insulin use. Abstract Biological ion channels usually conduct the high-flux transport of 107 ~ 108 ions·s− 1; however, the underlying mechanism is still lacking. Here, by applying the KcsA potassium channel as a typical example, and performing multitimescale molecular dynamics simulations, we demonstrate that there is coherence of the K+ ions confined in biological channels, which determines transport. The coherent oscillation state of confined K+ ions with a nanosecond-level lifetime in the channel dominates each transport event, serving as the physical basis for the high flux of ~108 ions∙s− 1. The coherent transfer of confined K+ ions only takes several picoseconds and has no perturbation effect on the ion coherence, acting as the directional key of transport. Such ion coherence is allowed by quantum mechanics. An increase in the coherence can significantly enhance the ion conductance. These findings provide a potential explanation from the perspective of coherence for the high-flux ion transport with ultralow energy consumption of biological channels.

Objective(s): Sox11, one of the SoxC family members, is an important transcription factor during neural development and neurogenesis. However, there is no report about its function in neural apoptosis. This research aims to examine the function of Sox11 in surgical brain injury (SBI).Materials and Methods: We used 90 Sprague-Dawley rats to develop the SBI models and the siRNA of Sox11 to study the roles of Sox11. Western blot, real-time PCR, immunofluorescence, neuron apoptosis and necrosis, brain edema, and neurological score were determined.Results: The gene and protein amount of Sox11, compared with the Sham group, were increased after SBI, which reached a peak at 12 hr. In addition, following the application of siRNAs, the amount of Sox11 protein was significantly less than that in the SBI group. On the other hand, neuronal apoptosis, necrosis, and brain edema were significantly increased, while neurological scores were decreased.Conclusion: These findings demonstrate the role of Sox11 following nerve injury induced by SBI. Inhibition of Sox11 with siRNA may lead to neuronal injury and cell death, aggravating secondary brain injury after SBI.

Abstract Objective To construct a nomogram for predicting the responsiveness of cardiac resynchronization therapy (CRT) in patients with chronic heart failure and verify its predictive efficacy. Method A retrospective study was conducted including 109 patients with chronic heart failure who successfully received CRT from January 2018 to December 2022. According to patients after six months of the CRT preoperative improving acuity in the left ventricular ejection fraction is 5% or at least improve grade 1 NYHA heart function classification, divided into responsive group and non-responsive group. Clinical data of patients were collected, and LASSO regression analysis and multivariate logistic regression analysis were used to explore relative factors. A nomogram was constructed, and the predictive performance of the nomogram was evaluated using the calibration curve and decision curve analysis (DCA). Results Among the 109 patients, 61 were assigned to the CRT-responsive group, while 48 were assigned to the non-responsive group. LASSO regression analysis showed that left ventricular end-systolic volume, diffuse fibrosis, and left bundle branch block (LBBB) were independent factors for CRT responsiveness in patients with heart failure (P

Abstract Background Antenatal depression is a significant public health issue affecting pregnant women both globally and in China. Using data from a mobile app-based screening programme, this study explored the prevalence and factors associated with antenatal depressive symptoms across different trimesters in Shenzhen. Methods A retrospective cross-sectional study was conducted on pregnant women who gave birth in any hospital in Shenzhen between July 2021 and May 2022 and underwent depression screening using an official maternal and infant health mobile app at least once during pregnancy. Depressive symptoms were evaluated using the 9-item Patient Health Questionnaire (PHQ-9), with cut-off scores of 5 and 10 for mild and high level of symptoms, respectively. The prevalence for each trimester was determined by calculating the proportion of women scoring 5 or higher. A variety of sociodemographic, obstetric, psychological, and lifestyle factors were assessed for their association with depressive symptoms. Chi-square test and multivariate logistic regression were performed to identify significant predictors. Results A total of 110,584 pregnant women were included in the study, with an overall prevalence of depressive symptoms of 18.0% and a prevalence of high-level symptoms of 4.2%. Depressive symptoms were most prevalent in the first trimester (10.9%) and decreased in the second (6.2%) and third trimesters (6.3%). Only a small proportion (0.4%) of women showed persistent depressive symptoms across all trimesters. Anxiety symptoms in early pregnancy emerged as the most significant predictor of depressive symptoms. Other factors linked to an increased risk throughout pregnancy include lower marital satisfaction, living with parents-in-law, experience of negative life events, as well as drinking before and during pregnancy. Factors associated with a reduced risk throughout pregnancy include multiparity and daily physical activity. Conclusions This large-scale study provides valuable insights into the prevalence and factors associated with antenatal depressive symptoms in Shenzhen. The findings underscore the need for targeted interventions for high-risk groups and the integration of mental health care into routine antenatal services. Continuous, dynamic monitoring of depressive symptoms for pregnant women and ensuring at-risk women receive comprehensive follow-up and appropriate psychological or psychiatric care are crucial for effectively addressing antenatal depression and improving maternal and infant health outcomes.

Abstract Biological ion channels exhibit high selectivity and permeability of ions because of their asymmetrical pore structures and surface chemistries. Here, we demonstrate a biomimetic nanofluidic channel (BNC) with an asymmetrical structure and glycyl-L-proline (GLP) -functionalization for ultrafast, selective, and unidirectional Dy3+ extraction over other lanthanide (Ln3+) ions with very similar electronic configurations. The selective extraction mainly depends on the amplified chemical affinity differences between the Ln3+ ions and GLPs in nanoconfinement. In particular, the conductivities of Ln3+ ions across the BNC even reach up to two orders of magnitude higher than in a bulk solution, and a high Dy3+/Nd3+ selectivity of approximately 60 could be achieved. The designed BNC can effectively extract Dy3+ ions with ultralow concentrations and thereby purify Nd3+ ions to an ultimate content of 99.8 wt.%, which contribute to the recycling of rare earth resources and environmental protection. Theoretical simulations reveal that the BNC preferentially binds to Dy3+ ion due to its highest affinity among Ln3+ ions in nanoconfinement, which attributes to the coupling of ion radius and coordination matching. These findings suggest that BNC-based ion selectivity system provides alternative routes to achieving highly efficient lanthanide separation.

The immune microenvironment plays a key role in the development and progression of tumors. In recent years, with the rapid advancement of high-throughput sequencing technologies, researchers have gained a deeper understanding of the composition and function of immune cells in the tumor microenvironment. However, traditional bulk sequencing technologies are limited in resolving heterogeneity at the single-cell level, constraining a comprehensive understanding of the complexity of the tumor microenvironment. The advent of single-cell RNA sequencing technology has brought new opportunities to uncover the heterogeneity of the immune microenvironment in lung cancer. Currently, T-cell-centered immunotherapy in clinical settings is prone to side effects affecting prognosis, such as immunogenic drug resistance or immune-related pneumonia, with the key factor being changes in the interactions between immune cells and tumor cells in the tumor microenvironment. Single-cell RNA sequencing technology can reveal the origins and functions of different subgroups within the tumor microenvironment from perspectives such as intercellular interactions and pseudotime analysis, thereby discovering new cell subgroups or novel biomarkers, providing new avenues for uncovering resistance to immunotherapy and monitoring therapeutic efficacy. This review comprehensively discusses the newest research techniques and advancements in single-cell RNA sequencing technology for unveiling the heterogeneity of the tumor microenvironment after lung cancer immunotherapy, offering insights for enhancing the precision and personalization of immunotherapy.

Abstract To adapt to the complex belowground environment, plants make trade-offs between root resource acquisition and defence ability. This includes forming partnerships with different types of root associating microorganisms, such as arbuscular mycorrhizal and ectomycorrhizal fungi. These trade-offs, by mediating root chemistry, exert legacy effects on nutrient release during decomposition, which may, in turn, affect the ability of new roots to re-acquire resources, thereby generating a feedback loop. However, the linkages at the basis of this potential feedback loop remain largely unquantified. Here, we propose a trait-based root ‘acquisition-defence-decomposition’ conceptual framework and test the strength of relevant linkages across 90 angiosperm tree species. We show that, at the plant species level, the root-fungal symbiosis gradient within the root economics space, root chemical defence (condensed tannins), and root decomposition rate are closely linked, providing support to this framework. Beyond the dichotomy between arbuscular mycorrhizal-dominated versus ectomycorrhizal-dominated systems, we suggest a continuous shift in feedback loops, from ‘high arbuscular mycorrhizal symbiosis-low defence-fast decomposition-inorganic nutrition’ by evolutionarily ancient taxa to ‘high ectomycorrhizal symbiosis-high defence-slow decomposition-organic nutrition’ by more modern taxa. This ‘acquisition-defence-decomposition’ framework provides a foundation for testable hypotheses on multidimensional linkages between species’ belowground strategies and ecosystem nutrient cycling in an evolutionary context.

Abstract TGF-β1 plays a pivotal role in the metastatic cascade of malignant neoplasms. N6-methyladenosine (m6A) stands as one of the most abundant modifications on the mRNA transcriptome. However, in the metastasis of gallbladder carcinoma (GBC), the effect of TGF-β1 with mRNA m6A modification, especially the effect of mRNA translation efficiency associated with m6A modification, remains poorly elucidated. Here we demonstrated a negative correlation between FOXA1 and TGF-β1 expression in GBC. Overexpression of FOXA1 inhibited TGF-β1-induced migration and epithelial-mesenchymal transition (EMT) in GBC cells. Mechanistically, we confirmed that TGF-β1 suppressed the translation efficiency of FOXA1 mRNA through polysome profiling analysis. Importantly, both in vivo and in vitro experiments showed that TGF-β1 promoted m6A modification on the coding sequence (CDS) region of FOXA1 mRNA, which was responsible for the inhibition of FOXA1 mRNA translation by TGF-β1. We demonstrated through MeRIP and RIP assays, dual-luciferase reporter assays and site-directed mutagenesis that ALKBH5 promoted FOXA1 protein expression by inhibiting m6A modification on the CDS region of FOXA1 mRNA. Moreover, TGF-β1 inhibited the binding capacity of ALKBH5 to the FOXA1 CDS region. Lastly, our study confirmed that overexpression of FOXA1 suppressed lung metastasis and EMT in a nude mice lung metastasis model. In summary, our research findings underscore the role of TGF-β1 in regulating TGF-β1/FOXA1-induced GBC EMT and metastasis by inhibiting FOXA1 translation efficiency through m6A modification.

Abstract In cold areas, the steel fiber reinforced rubber concrete (SFRRC) pavement is exposed to natural environment and experiences varying degrees of damage from freezing and thawing. This can have a serious impact on the normal usage and safe operation of the pavement structure. This research examines the impact of varying rubber concentrations on multiple variables, such as the rate of mass reduction, relative dynamic modulus of elasticity, compressive strength, and thickness of the damage layer (H f) during freeze–thaw (F-T) durability testing conducted on SFRRC. Furthermore, an analysis is conducted to determine the degradation pattern exhibited by SFRRC. The internal structure evolution and pore structure characteristics of SFRRC were examined using scanning electron microscopy and mercury intrusion porosimetry techniques, which revealed the underlying damage mechanism in SFRRC during F-T cycles. The results suggest that the addition of an appropriate amount of rubber can effectively enhance the frost resistance of SFRRC in water. A gradual improvement in the frost resistance of SFRRC is observed when increasing the rubber content from 0 to 10%. The optimal frost resistance is observed in SFRRC with 10% rubber content. However, when the rubber content reaches 15%, SFRRC exhibits significant degradation and lower level of resistance to freezing compared to SFRRC without rubber. Microcracks form within SFRRC due to the freezing–thawing forces experienced during the experiment, resulting in the development of a damage layer that extends from the surface to the interior. The compressive strength of the damaged layer significantly decreases as H f increases. The addition of appropriate rubber in SFRRC improves its pore structure, leading to an increased proportion of harmless or less harmful pores and a reduction in average pore size, thereby significantly enhancing its frost resistance.

This study aimed to develop a medium-low glycemic index (GI) cereal meal replacement powder suitable for diabetic patients using selenium-enriched germinated brown rice, red adzuki beans, barley, tartary buckwheat, bean dregs, and mulberry leaf extracts as raw materials. The baking processing technology was optimized through response surface experiments using sensory scores and GI value-weighted scores as indicators. At the same time, the effects of the two processing methods of baking and extrusion on the microstructure, crystal properties, gelatinization properties, gelatinization degree, brewability, caking rate and other physicochemical properties of selenium-enriched germinated brown rice meal replacement powder were discussed. The results showed that the GI value was 62.75, the sensory score was 91.17, and the highest weighted comprehensive score was 60.69 under the optimal conditions of the baking time of 35 min, the baking temperature of 140 ℃ and the liquid-to-material ratio of 1:1 (L/kg). Compared with unprocessed meal replacement powder, the relative crystallinity of baking optimal meal replacement powder and extruded meal replacement powder decreased to 15.15 and 4.28 respectively, and the B-type and V-type crystals increased. The round starch granules in the microstructure of the other two meal replacement powders were reduced and irregular granules were increased. In the electronic nose comprehensive evaluation equation, the comprehensive odor score from high to low was baked optimal meal replacement powder>extruded meal replacement powder>unprocessed meal replacement powder. The experimental conclusion showed that the optimal GI value of baking meal replacement powder was between 55 and 70, which belonged to the medium-low GI range and was suitable for people with diabetes. It can be recommended as the preferred processing parameter for the development of meal replacement powder with a medium and low glycemic index.

Aiming at the problems such as low construction efficiency and high labor intensity in the process of lowering large diameter casing through the broken stratum of complex roof high level drilling in coal mine, a large diameter casing lowering device is designed. This paper introduces the design of large diameter casing lowering device, describes in detail the design process of front inner support assembly, back inner support assembly, cylinder block, slip and other key parts, and describes the technological steps of casing lowering by drilling rig. During the field industrial test, 68 casings with a diameter of ϕ245 mm and a length of 1.5 m were run in 4 directional boreholes with a tilt angle of 17°, totaling 102 m. The large diameter casing lowering device is sensitive and reliable, and can meet the functional requirements of mechanized large diameter casing lowering in complex roof high level drilling.

Abstract The challenges posed by energy and environmental issues have forced mankind to explore and utilize unconventional energy sources. It is imperative to convert the abundant coalbed gas (CBG) into high value-added products, i.e., selective and efficient conversion of methane from CBG. Methane activation, known as the “holy grail”, poses a challenge to the design and development of catalysts. The structural complexity of the active metal on the carrier is of particular concern. In this work, we have studied the nucleation growth of small Co clusters (up to Co6) on the surface of CeO2(110) using density functional theory, from which a stable loaded Co/CeO2(110) structure was selected to investigate the methane activation mechanism. Despite the relatively small size of the selected Co clusters, the obtained Co x /CeO2(110) exhibits interesting properties. The optimized Co5/CeO2(110) structure was selected as the optimal structure to study the activation mechanism of methane due to its competitive electronic structure, adsorption energy and binding energy. The energy barriers for the stepwise dissociation of methane to form CH3*, CH2*, CH*, and C* radical fragments are 0.44, 0.55, 0.31, and 1.20 eV, respectively, indicating that CH* dissociative dehydrogenation is the rate-determining step for the system under investigation here. This fundamental study of metal-support interactions based on Co growth on the CeO2(110) surface contributes to the understanding of the essence of Co/CeO2 catalysts with promising catalytic behavior. It provides theoretical guidance for better designing the optimal Co/CeO2 catalyst for tailored catalytic reactions.

Abstract Messenger RNA (mRNA) therapeutics delivered via lipid nanoparticles hold the potential to treat metabolic diseases caused by protein deficiency, including propionic acidemia (PA), methylmalonic acidemia (MMA), and phenylketonuria (PKU). Herein we report results from multiple independent preclinical studies of mRNA-3927 (an investigational treatment for PA), mRNA-3705 (an investigational treatment for MMA), and mRNA-3210 (an investigational treatment for PKU) in murine models of each disease. All 3 mRNA therapeutics exhibited pharmacokinetic/pharmacodynamic (PK/PD) responses in their respective murine model by driving mRNA, protein, and/or protein activity responses, as well as by decreasing levels of the relevant biomarker(s) when compared to control-treated animals. These preclinical data were then used to develop translational PK/PD models, which were scaled allometrically to humans to predict starting doses for first-in-human clinical studies for each disease. The predicted first-in-human doses for mRNA-3927, mRNA-3705, and mRNA-3210 were determined to be 0.3, 0.1, and 0.4 mg/kg, respectively.

Abstract “National Civilized City” (NCC) is regarded as China’s highest honorary title and most valuable city brand. To win and maintain the “golden city” title, municipal governments must pay close attention to various key appraisal indicators, mainly environmental ones. In this study we verify whether cities with the title are more likely to mitigate SO2 pollution. We adopt the spatial Durbin difference-in-differences (DID) model and use panel data of 283 Chinese cities from 2003 to 2018 to analyze the local (direct) and spillover effects (indirect) of the NCC policy on SO2 pollution. We find that SO2 pollution in Chinese cities is not randomly distributed in geography, suggesting the existence of spatial spillovers and possible biased estimates. Our study treats the NCC policy as a quasi-experiment and incorporates spatial spillovers of NCC policy into a classical DID model to verify this assumption. Our findings show: (1) The spatial distribution of SO2 pollution represents strong spatial spillovers, with the most highly polluted regions mainly situated in the North China Plain. (2) The Moran’s I test results confirms significant spatial autocorrelation. (3) Results of the spatial Durbin DID models reveal that the civilized cities have indeed significantly mitigated SO2 pollution, indicating that cities with the honorary title are acutely aware of the environment in their bid to maintain the golden city brand. As importantly, we notice that the spatial DID term is also significant and negative, implying that neighboring civilized cities have also mitigated their own SO2 pollution. Due to demonstration and competition effects, neighboring cities that won the title ostensibly motivates local officials to adopt stringent policies and measures for lowering SO2 pollution and protecting the environment in competition for the golden title. The spatial autoregressive coefficient was significant and positive, indicating that SO2 pollution of local cities has been deeply affected by neighbors. A series of robustness check tests also confirms our conclusions. Policy recommendations based on the findings for protecting the environment and promoting sustainable development are proposed.

Abstract Aims/Introduction Mitochondrial damage caused by oxidative stress is a main driver of pancreatic β‐cell dysfunction in the pathogenesis of type 2 diabetes mellitus. Prohibitin2 (PHB2) is a vital inner mitochondrial membrane protein that participates in mitophagy to remove the damaged mitochondria. This study aimed to investigate the role and mechanisms of PHB2‐mediated mitophagy in oxidative stress‐induced pancreatic β‐cell dysfunction. Materials and Methods PHB2 and mitophagy‐related protein expression were analyzed by real‐time polymerase chain reaction and western blotting in RINm5F cells treated with H2O2 and islets of diabetic rats. Mitophagy was observed by mitochondrial and lysosome colocalization. RINm5F cells were transfected by phb2 siRNA or overexpression plasmid to explore the role of PHB2 in mitophagy of RINm5F cells. The mechanism of Nrf2 regulating PHB2 was explored by Nrf2 inhibitor and agonist. Results The expression of PHB2, mitophagy related protein PINK1, and Parkin were decreased in RINm5F cells incubated with H2O2 and in islets of diabetic rats. Overexpression of PHB2 protected β‐cells from oxidative stress by promoting mitophagy and inhibiting cell apoptosis, whereas transfection with PHB2 siRNA suppressed mitophagy. Furthermore, PHB2‐mediated mitophagy induced by oxidative stress was through the Nrf2/PHB2 pathway in β‐cells. Antioxidant NAC alleviated oxidative stress injury by promoting PHB2‐mediated mitophagy. Conclusion Our study suggested that PHB2‐mediated mitophagy can protect β‐cells from apoptosis via the Nrf2/PHB2 pathway under oxidative stress. Antioxidants may protect β‐cell from oxidative stress by prompting PHB2‐mediated mitophagy. PHB2‐mediated mitophagy as a potential mechanism takes part in the oxidative stress induced β‐cell injury.

Rationale & Objective: Postpartum renal cortical necrosis (postpartum RCN) is a severe form of obstetric acute kidney injury. This study aimed to identify clinicopathologic features in Chinese postpartum RCN cases to determine how pathologic findings may contribute to the treatment and prognosis. Study Design: Single-center, case series. Setting & Participants: Twelve patients with postpartum RCN had kidney biopsies at Peking University First Hospital between 2014 and 2021. Fat deposition affects beef quantity and quality via preadipocyte proliferation. Beta-sitosterol, a natural small molecular compound, has various functions, such as anti-inflammation, antibacterial, and anticancer properties. The mechanism of action of Beta-sitosterol on bovine preadipocytes remains unclear. This study, based on RNA-seq, reveals the impact of Beta -sitosterol on the proliferation of bovine preadipocytes. Compared to the control group, Beta-sitosterol demonstrated a more pronounced inhibitory effect on cell proliferation after 48 hours of treatment than after 24 hours, as evidenced by the results of EdU staining and flow cytometry. RNA-seq and Western Blot analyses further substantiated these findings. Our results suggest that the impact of Beta-sitosterol on the proliferation of bovine preadipocytes is not significant after a 24-hour treatment. It is only after extending the treatment time to 48 hours that Beta-sitosterol may induce cell cycle arrest at the G2/M phase by suppressing the expression of CCNB1, thereby inhibiting the proliferation of bovine preadipocytes.

Abstract Coastal darkening is emerging as a global threat to fringing reefs. While some reef‐building corals exhibit resistance to low‐light environments, the mechanisms behind this resistance, particularly the role of coral hosts, remain inadequately understood. Here, we investigated variations in underwater photosynthetically active radiation (PAR) and employed the Bayesian stable isotope mixing model (MixSIAR) to estimate the contributions of autotrophic (i.e., dissolved inorganic matter, DIM) and heterotrophic sources (i.e., particulate organic matter, POM, and dissolved organic matter, DOM) to the nutrition of the reef coral Galaxea fascicularis on the Luhuitou turbid reef in the northern South China Sea. Our findings revealed that the heterotrophic contribution to coral nutrition increased to 58.5% with decreasing PAR and that the heterotrophic contribution was significantly negatively correlated with δ13C difference between host and symbiont (δ13Ch–s). Moreover, we observed significant seasonal variations in the respective contributions of POM and DOM to coral nutrition, linked to the sources of these nutrients, demonstrating that G. fascicularis can selectively ingest POM and DOM based on their bioavailability to enhance its heterotrophic contribution. This heterotrophic plasticity improved the low‐light resistance of G. fascicularis and contributed to its prominence within coral communities. However, with a low‐light threshold of approximately 3.73% of the surface PAR for G. fascicularis, our results underscore the need for effective strategies to mitigate low‐light conditions on nearshore turbid reefs. In summary, our study highlights the critical role of heterotrophic plasticity in coral responses to natural low‐light environments, suggesting that some reef‐building corals with such plasticity could become dominant or resilient species in the context of coastal darkening.

BackgroundInsulin resistance (IR) is linked to an increased risk of neurological impairment following a stroke and may contribute to poor neurological prognosis in affected patients. The metabolic score for the insulin resistance index, shortened as the METS-IR, generally serves as a surrogate index for IR. However, its association with the severity of neurological impairment in patients with severe cerebral infarction (CI) in neurological intensive care units (ICU) has not been fully established.MethodsPatients with a diagnosis of CI, admitted to the neurological ICUs of Yangzhou University’s Affiliated Hospital and Xuzhou Medical University’s Affiliated Hospital, were included in the study. A multivariate logistic regression model and restricted cubic splines (RCS) were employed to explore the relationship between the METS-IR index and the severity of neurological impairment in these patients. The predictive capabilities of the METS-IR index and the triglyceride-glucose (TyG) index for outcome measures were compared through the ROC curve. Furthermore, a decision curve analysis was executed, and the integrated discrimination improvement (IDI) index was computed to evaluate the enhancements in predictive performance and clinical utility of various scoring systems with the inclusion of the METS-IR index. Subgroup analysis was conducted regarding age, BMI, and smoking status.ResultsThe study ultimately included 504 participants. Adjusted logistic regression and RCS results showed that as the METS-IR index increases, the risk of neurological impairment in patients with severe CI consistently grows (P for overall = 0.0146, P-nonlinear: 0.0689). The METS-IR index’s predictive capability for neurological impairment (AUC = 0.669) was superior to that of the TyG index (AUC = 0.519).ConclusionFrom the study results, the METS-IR index can serve as an important predictor for neurological impairment in ICU patients with severe CI. It can aid in the identification and early intervention of neurological impairment in these patients.

Robot control in complex and unpredictable scenarios presents challenges such as adaptability, robustness, and human-robot interaction. These scenarios often require robots to perform tasks that involve unknown objects in unstructured environments with high levels of uncertainty. Traditional control methods, such as automatic control, may not be suitable due to their limited adaptability and reliance on prior knowledge. Human-in-the-loop method faces issues such as insufficient feedback, increased failure rates due to noise and delays, and lack of operator immersion, preventing the achievement of human-level performance. This study proposed a shared control framework to achieve a trade-off between efficiency and adaptability by combing the advantages of both teleoperation and automatic control method. The proposed approach combines the advantages of both human and automatic control methods to achieve a balance between performance and adaptability. We developed a linear model to compare three control methods and analyzed the impact of position noise and communication delays on performance. The real-world implementation of the shared control system demonstrates its effectiveness in object grasping and manipulation tasks. The results suggest that shared control can significantly improve grasping efficiency while maintaining adaptability in task execution for practical robotics applications.

Summary: Hyperfunction of the dopamine system has been implicated in manic episodes in bipolar disorders. How dopaminergic neuronal function is regulated in the pathogenesis of mania remains unclear. Histaminergic neurons project dense efferents into the midbrain dopaminergic nuclei. Here, we present mice lacking dopaminergic histamine H2 receptor (H2R) in the ventral tegmental area (VTA) that exhibit a behavioral phenotype mirroring some of the symptoms of mania, including increased locomotor activity and reduced anxiety- and depression-like behavior. These behavioral deficits can be reversed by the mood stabilizers lithium and valproate. H2R deletion in dopaminergic neurons significantly enhances neuronal activity, concurrent with a decrease in the γ-aminobutyric acid (GABA) type A receptor (GABAAR) membrane presence and inhibitory transmission. Conversely, either overexpression of H2R in VTA dopaminergic neurons or treatment of H2R agonist amthamine within the VTA counteracts amphetamine-induced hyperactivity. Together, our results demonstrate the engagement of H2R in reducing VTA dopaminergic activity, shedding light on the role of H2R as a potential target for mania therapy.

Positive interspecific interactions in resource capture and soil property modification benefit the establishment of mixed plantations, contingent on environmental conditions. However, primary species frequently reject companion tree species frequently arises in forest management practices. To understand this contradiction, we investigated the effects of five planting patterns involving two local companion tree species (Phoebe chekiangensis and Torreya grandis) and Phyllostachys edulis on the soil nitrogen (N) and phosphorus (P) content, enzyme activities, net N mineralization rates, microbial composition, fungal functional groups, and the AMO gene abundance under both shade and full light conditions using a pot experiment. Under the full light condition, altered soil properties were predominantly influenced by interspecific interaction compared to intraspecific interactions, leading to an overall enhancement in nutrient availability and enzymatic activities. Under the shade condition, mixing with P. edulis similarly facilitated increased NH4+-N content, ammonification rate, and urease activity. However, P. edulis significantly decreased ammonia monooxygenase and its encoding gene, as evidence by the unbalanced ratio of net ammonification to net nitrification (2.2-fold) under the shade condition, resulting in high NH4+-N/NO3–-N ratios. Additionally, the shade condition induced soil microbial communities in the mixture to resemble those of monocultured dominant species (P. edulis). Our findings also indicated that specific plant-fungal associations in companion species were disrupted when intercropped with P. edulis. Therefore, this study highlights how the performance of companion species, linked to soil NH4+-N/NO3–-N ratios and plant-fungal associations, leads to their suppression by P. edulis under shade conditions.

Charge storage performance of the poly (3,4-ethylenedioxythiophene): poly (styrene sulfonate) (PEDOT: PSS, PH1000)-based supercapacitor is greatly enhanced by dimethyl sulfoxide (DMSO)-treatment and addition of aramid nanofibers (ANFs), but the underlying mechanism is unclear. To reveal it, we synthesized the films of PH1000, DMSO-treated PH1000 (D-P) and DMSO-treated PH1000-20 wt%ANF (D-PA) to construct the in-plane supercapacitors. A comparative study of electrochemical performance was firstly conducted to identify the specific contribution of DMSO-treatment and ANF components. Electrochemical impedance spectroscopy (EIS) was then analyzed, and the results demonstrated that the reduction of the ohmic resistance of the device was mainly attributed to the DMSO-treatment, while the addition of ANFs was found to contribute more to the decrease of Warburg resistance. The effects of DMSO-treatment and ANF components on the electrical propertíe and microstructure of PH1000 film were analyzed based on the square resistance results and AFM/SEM images, respectively. Dynamic permeation processes of the hydrogel electrolyte on the films were further recorded, vividly showing that the electrolyte permeability is highly dependent on the microstructure of the film. Through these analyses, a deep insight into the synergistic effects of DMSO-treatment and ANF addition on the electrochemical performance of the PH1000-based supercapacitor has been reached.

Abstract The emergence of smart windows has been a pivotal innovation in the field of energy efficiency and carbon emission reduction, sparking considerable interest worldwide. This review consolidates the latest research developments in inorganic‐based photochromic materials for smart windows applications, encompassing the design of device architectures, and their implementation in enhancing energy conservation, augmenting comfort levels, and optimizing indoor environmental control. Finally, this review culminates in an insightful analysis of the challenges and constraints in the existing research landscapes, which illuminates guidance for the directions and perspectives of future research.

Abstract For the porous‐membrane‐based osmotic energy generator, the potential synergistic enhancement mechanism of various key parameters is still controversial, especially because optimizing the trade‐off between permeability and selectivity is still a challenge. Here, to construct a permeability and selectivity synergistically enhanced osmotic energy generator, the two‐dimensional porous membranes with tunable charge density are prepared by inserting sulfonated polyether sulfone into graphene oxide. Influences of charge density and pore size on the ion transport are explored, and the ionic behaviors in the channel are calculated by numerical simulations. The mechanism of ion transport in the process is studied in depth, and the fundamental principles of energy conversion are revealed. The results demonstrate that charge density and pore size should be matched to construct the optimal ion channel. This collaborative enhancement strategy of permeability and selectivity has significantly improved the output power in osmotic energy generation; compared to the pure graphene oxide membrane, the composite membrane presents almost 20 times improvement.

Key Clinical Message In recent years, it is necessary to Redo‐TAVR for the patients with bioprosthetic valve degeneration. This case report described a unique instance to successfully Redo‐TAVR a patient with bioprosthetic valve degeneration, in addition, with left cerebral infarction and renal insufficiency. Abstract Over time, more and more patients have bioprosthetic valve degeneration either used in SAVR or TAVR. In order to solve the produced problems due to the degenerated bioprosthetic valve, Redo‐TAVR was increasingly popular due to its safe and efficiency especially for the high risk and complicated symptoms patients. In this case, the patient with left cerebral infarction and renal insufficiency has exhibited severe regurgitation and obvious neoplasm around the previous replaced aortic valve. For the patient with complicated symptoms, we did not image for this patient and only used CT to determine the position and angle for the Redo‐TAVR on the base of metal stent for the previous replaced aortic valve. During the Redo‐TAVR process, for fear of the obvious neoplasm slipping from the previous replaced aortic valve to embolism of important organs, before carrying out the Redo‐TAVR, cerebral protection device, temporary pacemaker, and coronary artery protection device were utilized in order to avoid the damage for the important organs from the obvious neoplasm slipping from the previous replaced aortic valve. The surgery was successful and the patient recovered well. The patient's symptoms of chest tightness and suffocation have been greatly reduced.

Alterations in the tumor microenvironment are closely associated with the metabolic phenotype of tumor cells. Cancer-associated fibroblasts (CAFs) play a pivotal role in tumor growth and metastasis. Existing studies have suggested that lactate produced by tumor cells can activate CAFs, yet the precise underlying mechanisms remain largely unexplored. In this study, we initially identified that lactate derived from lung cancer cells can promote nuclear translocation of NUSAP1, subsequently leading to the recruitment of the transcriptional complex JUNB-FRA1-FRA2 near the DESMIN promoter and facilitating DESMIN transcriptional activation, thereby promoting CAFs' activation. Moreover, DESMIN-positive CAFs, in turn, secrete IL-8, which recruits TAMs or promotes M2 polarization of macrophages, further contributing to the alterations in the tumor microenvironment and facilitating lung cancer progression. Furthermore, we observed that the use of IL-8 receptor antagonists, SB225002, or Navarixin, significantly reduced TAM infiltration and enhanced the therapeutic efficacy of anti-PD-1 or anti-PD-L1 treatment. This finding indicates that inhibiting IL-8R activity can attenuate the impact of CAFs on the tumor microenvironment, thus restraining the progression of lung cancer.

Abstract Purpose This study aimed to evaluate the feasibility of 11C-CFT PET brain imaging in Parkinson’s Disease using a total-body PET/CT scanner and explore the optimal scan duration to guide the clinical practice. Methods Thirty-two patients with Parkinson’s disease (PD) performing 11C-CFT PET/CT brain imaging using a total-body PET/CT scanner were retrospectively enrolled. The PET data acquired over a period of 900 s were reconstructed into groups of different durations: 900-s, 720-s, 600-s, 480-s, 300-s, 180-s, 120-s, 60-s, and 30-s (G900 to G30). The subjective image quality analysis was performed using 5-point scales. Semi-quantitative measurements were analyzed by SUVmean and dopamine transporter (DAT) binding of key brain regions implicated in PD, including the caudate nucleus and putamen. The full-time images (G900) were served as reference. Results The overall G900, G720, and G600 image quality scores were 5.0 ± 0.0, 5.0 ± 0.0, and 4.9 ± 0.3 points, respectively, and there was no significant difference among these groups (P > 0.05). Objective(s): Mitochondrial dysfunction caused by mitochondrial DNA (mtDNA) damage and mutation is widely accepted as one of the pathological processes of neurodegenerative diseases. As an mtDNA binding protein, mitochondrial transcription factor A (TFAM) maintains the integrity of mtDNA through transcription, replication, nucleoid formation, damage perception, and DNA repair. In recent works, the overexpression of TFAM increased the mtDNA copy count, promoted mitochondrial function, and improved the neurological dysfunction of neurodegenerative diseases. The role of TFAM in neurodegenerative diseases has been well explained. However, the role of TFAM after surgical brain injury (SBI) has not been studied. In this work, we aimed to study the role of TFAM in the brain after SBI and its mechanism of action.Materials and Methods: One hour after the occurrence of SBI, tetramethylpyrazine (TMP) was injected into the abdominal cavity of rats, and the brain was collected 48 hr later for testing. The evaluation included neurobehavioral function test, brain water content measurement, immunofluorescence, western blot, TUNEL staining, FJC staining, ROS test, and ATP test. Results: After SBI, the content of TFAM on the ipsilateral side increased and reached a peak at about 48 hr. After intraperitoneal injection of TMP in rats, 48 hr after SBI, the concentration of TFAM, Bcl-2, and adenosine triphosphate (ATP) increased; the content of caspase-3, reactive oxygen species (ROS), and cerebral edema decreased; and the nerve function significantly improved. Conclusion: TMP inhibited cell apoptosis after SBI in rats by up-regulating TFAM and protecting brain tissues.

Abstract Achieving high selectivity of Li+ and Mg2+ is of paramount importance for effective lithium extraction from brines, and nanofiltration (NF) membrane plays a critical role in this process. The key to achieving high selectivity lies in the on-demand design of NF membrane pores in accordance with the size difference between Li+ and Mg2+ ions, but this poses a huge challenge for traditional NF membranes and difficult to be realized. In this work, we report the fabrication of polyamide (PA) NF membranes with ultra-high Li+/Mg2+ selectivity by modifying the interfacial polymerization (IP) process between piperazine (PIP) and trimesoyl chloride (TMC) with an oil-soluble surfactant that forms a monolayer at oil/water interface, referred to as OSARIP. The OSARIP benefits to regulate the membrane pores so that all of them are smaller than Mg2+ ions. Under the solely size sieving effect, an exceptional Mg2+ rejection rate of over 99.9% is achieved. This results in an exceptionally high Li+/Mg2+ selectivity, which is one to two orders of magnitude higher than all the currently reported pressure-driven membranes, and even higher than the microporous framework materials, including COFs, MOFs, and POPs. The large enhancement of ion separation performance of NF membranes may innovate the current lithium extraction process and greatly improve the lithium extraction efficiency.

[Objective] At present, the underground engineering diseases and accidents caused by unbalanced load in urban rail transit are increasing gradually. In order to understand the adverse effect of unbalanced load on the deformation of deep foundation pit in urban rail transit station and reduce the construction risk, it is necessary to analyze the deformation characteristics of station deep foundation pit under unbalanced load. [Method] Based on theoretical analysis and on-site monitoring data, the aforementioned deformation characteristics are studied, the changing law regarding the retaining pile deformation, the horizontal displacement of the deep soil and ground settlement is analyzed, and the interrelation between the retaining pile deformation and the deep soil horizontal displacement, the ground settlement and excavation depth is discussed. [Result & Conclusion] The horizontal displacement curve of the retaining pile is fish-belly shaped, and the formation condition has obvious influence on the deformation of the retaining pile. There is a roughly linear relationship between the deformation of the retaining pile and the horizontal displacement of deep soil, and the empirical coefficient is proposed to be 1.30. Due to partial load, the ground settlement around the deep foundation pit is larger, and the relationship between the maximum horizontal displacement δhm of the retaining pile and the maximum ground settlement δvm is δvm=(0.9~3.0) δhm, and the proportional coefficient between them is greater than the value under symmetric load. The strong deformation resistance of rock strata limits the deformation of the retaining pile. There is a relationship of δhm=(0.009%~0.060%) He between δhm and He, and the proportional coefficient between them is smaller than the value under symmetric load. The impact area of deep foundation pit construction on the surrounding environment is mainly divided into Zone 1 (main impact zone), Zone 2 (secondary impact zone) and Zone 3 (no impact zone), and the impact area of foundation pit construction is about (0~2.5) He.

Abstract Objective To compare neuronavigation-assisted intracerebral hematoma puncture and drainage with neuroendoscopic hematoma removal for treatment of hypertensive cerebral hemorrhage. Method Ninety-one patients with hypertensive cerebral hemorrhage admitted to our neurosurgery department from June 2022 to May 2023 were selected: 47 patients who underwent endoscopic hematoma removal with the aid of neuronavigation in observation Group A and 44 who underwent intracerebral hematoma puncture and drainage in control Group B. The duration of surgery, intraoperative bleeding, hematoma clearance rate, pre- and postoperative GCS score, National Institutes of Health Stroke Scale (NIHSS) score, mRS score and postoperative complications were compared between the two groups. Abstract Background Colorectal cancer (CRC) is a malignancy of remarkable heterogeneity and heightened morbidity. Cancer associated fibroblasts (CAFs) are abundant in CRC tissues and are essential for CRC growth. Here, we aimed to develop a CAF-related classifier for predicting the prognosis of CRC and identify critical pro-tumorigenic genes in CAFs. Method The mRNA expression and clinical information of CRC samples were sourced from two comprehensive databases, The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO). Using a weighted gene co-expression network analysis (WGCNA) approach, CAF-related genes were identified and a CAF risk signature was developed through the application of univariate analysis and the least absolute shrinkage and selection operator (LASSO) Cox regression model. EdU cell proliferation assay, and transwell assay were performed to detect the oncogenic role of KCNE4 in CAFs. Results We constructed a prognostic CAF model consisting of two genes (SFRP2 and KCNE4). CRC patients were classified into low- and high-CAF-risk groups using the median CAF risk score, and patients in the high-CAF-risk group had worse prognosis. Meanwhile, a higher risk score for CAFs was associated with greater stromal and CAF infiltrations, as well as higher expression of CAF markers. Furthermore, TIDE analysis indicated that patients with a high CAF risk score are less responsive to immunotherapy. Our further experiments had confirmed the strong correlation between KCNE4 and the malignant phenotypes of CAFs. Moreover, we had shown that KCNE4 could actively promote tumor-promoting phenotypes in CAFs, indicating its critical role in cancer progression. Conclusion The two-gene prognostic CAF signature was constructed and could be reliable for predicting prognosis for CRC patients. Moreover, KCNE4 may be a promising strategy for the development of novel anti-cancer therapeutics specifically directed against CAFs.

Highlights We present a method that utilizes ionic liquid-enhanced nanomaterial assembly to fabricate highly stable and large-area MXene-silver nanowire electrodes with ordered layered structures. This approach emphasizes the use of hydrophobic and nonvolatile ionic liquids, which form stable interfaces with water by reducing interface energy, preventing the sedimentation loss of nanomaterials during assembly. The prepared electrodes not only exhibit excellent optoelectronic properties (9.4 Ω sq−1 sheet resistance and 93% transmittance), but also have exceptional antioxidant capacity.

Abstract Background This study aimed to determine the incidence and influencing factors of venous thromboembolism (VTE) in patients with traumatic rib fractures. Methods The retrospective study analyzed medical records of patients with traumatic rib fractures from 33 hospitals. Results The overall incidence of VTE in hospitalized patients with traumatic rib fractures was 8.1%. Patients with isolated traumatic rib fractures had a significantly lower incidence of VTE (4.4%) compared to patients with rib fractures combined with other injuries (12.0%). Multivariate analysis identified the number of rib fractures as an independent risk factor for thrombosis. Surgical stabilization of isolated rib fractures involving three or more ribs was associated with a lower VTE incidence compared to conservative treatment. Conclusions Patients with rib fractures have a higher incidence of VTE, positively correlated with the number of rib fractures. However, the occurrence of thrombosis is relatively low in isolated rib fractures. Targeted thromboprophylaxis strategies should be implemented for these patients, and surgical stabilization of rib fractures may be beneficial in reducing the risk of VTE.

Abstract Epidemiological studies suggested an association between omega-3 fatty acids and cognitive function. However, the causal role of the fatty acid desaturase (FADS) gene, which play a key role in regulating omega-3 fatty acids biosynthesis, on cognitive function is unclear. Hence, we used two-sample Mendelian randomization (MR) to estimate the gene-specific causal effect of omega-3 fatty acids (N = 114,999) on cognitive function (N = 300,486). Tissue- and cell type-specific effects of FADS1/FADS2 expression on cognitive function were estimated using brain tissue cis-expression quantitative trait loci (cis-eQTL) datasets (GTEx, N ≤ 209; MetaBrain, N ≤ 8,613) and single cell cis-eQTL data (N = 373), respectively. These causal effects were further evaluated in whole blood cis-eQTL data (N ≤ 31,684). A series of sensitivity analyses were conducted to validate MR assumptions. Leave-one-out MR showed a FADS gene-specific effect of omega-3 fatty acids on cognitive function [β = −1.3 × 10−2, 95% confidence interval (CI) (−2.2 × 10−2, −5 × 10−3), P = 2 × 10−3]. Tissue-specific MR showed an effect of increased FADS1 expression in cerebellar hemisphere and FADS2 expression in nucleus accumbens basal ganglia on maintaining cognitive function, while decreased FADS1 expression in nine brain tissues on maintaining cognitive function [colocalization probability (PP.H4) ranged from 71.7% to 100.0%]. Cell type-specific MR showed decreased FADS1/FADS2 expression in oligodendrocyte was associated with maintaining cognitive function (PP.H4 = 82.3%, respectively). Increased FADS1/FADS2 expression in whole blood showed an effect on cognitive function maintenance (PP.H4 = 86.6% and 88.4%, respectively). This study revealed putative causal effect of FADS1/FADS2 expression in brain tissues and blood on cognitive function. These findings provided evidence to prioritize FADS gene as potential target gene for maintenance of cognitive function.

Background While renal biopsy remains the preferred diagnostic method for assessing proteinuria, hematuria, or renal failure, laparoscopic renal biopsy (LRB) can serve as an alternative for high-risk patients when percutaneous kidney biopsy (PKB) is not recommended. This study was aimed to evaluate the safety of LRB.Methods In study 1, Fourteen patients from January 2021 to January 2023 had a LRB taken for various indications, such as morbid obesity, abnormal kidney construction, uncontrolled hypertension, and coagulopathy. We also conducted a Meta-analysis of the success rate and complication rate of previous LRB in study 2.Results All the patients completed biopsies and adequate renal tissues were obtained. The success rate was 100%. The median number of glomeruli obtained was 22.5 (range:12.0, 45.0). The complication rate was 7.1% (urinary tract infection). There were no significant differences between levels of hemoglobin, serum creatinine, and urinary NAGL before and after surgery. In the meta-analysis, the success rate of operation, satisfactory rate of sample, and complication rate of surgery were 99.9%, 99.1%, and 2.6% respectively.Conclusion LRB can achieve a good success rate and specimen retrieval and does not increase the risk of complications for high-risk patients. It can present as one of the alternative methods for patients with glomerular diseases.

Global warming, primarily driven by emissions of greenhouse gases, particularly carbon dioxide, has emerged as a widely acknowledged concern. Hence, the development of novel carbon capture materials with high capacity and low cost holds substantial practical significance. This study investigates the influence of aging time, pH, and copper salt on the synthesis of Cu2(OH)PO4 and its CO2 adsorption performance. Cu2(OH)PO4 adsorbents were synthesized under various aging time, pH, and copper salt conditions, and their morphology and surface functional groups were characterized. Experimental findings indicate that excessively prolonged or abbreviated aging times adversely affect the formation of distinct, discernible lamellar structures within the material. Different pH levels influence the stacking configuration of the lamellae, impacting both their thickness and size. Under acidic conditions, lamellae exhibit dispersed three-dimensional stacking; under neutral conditions, lamellae notably enlarge and demonstrate two-dimensional stacking; at pH 9, lamellae stack three-dimensionally and aggregate. Additionally, the CO2 adsorption performance of Cu2(OH)PO4 adsorbents synthesized with different copper salts varies. By examining the relationship between surface functional group content and CO2 adsorption capacity, we deduced the mechanism by which various synthesis conditions affect both surface functional groups and adsorption capacity. Cu2(OH)PO4 synthesized with a 24 h aging time, pH 7, and CuSO4 as the copper salt exhibits the highest CO2 adsorption capacity, achieving 1.006 mmol/g.

Background Machine learning classifiers are increasingly used to create predictive models for pathological complete response (pCR) in breast cancer after neoadjuvant therapy (NAT). Few studies have compared the effectiveness of different ML classifiers. This study evaluated radiomics models based on pre- and post-contrast first-phase T1 weighted images (T1WI) in predicting breast cancer pCR after NAT and compared the performance of ML classifiers. Methods This retrospective study enrolled 281 patients undergoing NAT from the Duke-Breast-Cancer-MRI dataset. Radiomic features were extracted from pre- and post-contrast first-phase T1WI images. The Synthetic Minority Oversampling Technique (SMOTE) was applied, then the dataset was randomly divided into training and validation groups (7:3). The radiomics model was built using selected optimal features. Support vector machine (SVM), random forest (RF), decision tree (DT), k-nearest neighbor (KNN), extreme gradient boosting (XGBoost), and light gradient boosting machine (LightGBM) were classifiers. Receiver operating characteristic curves were used to assess predictive performance. Results LightGBM performed best in predicting pCR [area under the curve (AUC): 0.823, 95% confidence interval (CI) [0.743–0.902], accuracy 74.0%, sensitivity 85.0%, specificity 67.2%]. During subgroup analysis, RF was most effective in pCR prediction in luminal breast cancers (AUC: 0.914, 95% CI [0.847–0.981], accuracy 87.0%, sensitivity 85.2%, specificity 88.1%). In triple-negative breast cancers, LightGBM performed best (AUC: 0.836, 95% CI [0.708–0.965], accuracy 78.6%, sensitivity 68.2%, specificity 90.0%). Conclusion The LightGBM-based radiomics model performed best in predicting pCR in patients with breast cancer. RF and LightGBM showed promising results for luminal and triple-negative breast cancers, respectively.

BackgroundAeromonas dhakensis is associated with soft tissue infection, bacteremia and gastroenteritis. Involvement of respiratory system in adults is extremely rare. We report a case of fulminant pneumonia and bacteremia due to A. dhakensis in a patient without underlying diseases.Case presentationA 26-year-old man became ill suddenly with pneumonia after swimming in a river. Despite intensive support measures in the intensive care unit, he died 13 hours after admission and 4 days after his first symptoms. Autopsy showed abundant Gram-negative bacteria, massive inflammatory cell infiltration, edema, necrosis and hemorrhage in lung tissue. A. dhakensis was isolated from blood culture taken at admission and bronchoalveolar lavage fluid (BALF) after intubation. Moreover, A. dhakensis was also detected in lung tissue by metagenomic next-generation sequencing (mNGS) assay. The infection may have come from river water.ConclusionIn patients who develop a fulminant pneumonia after contacting an aquatic environment, A. dhakensis should be alerted and mNGS may aid in the detection of aquatic pathogens by being more sensitive and specific versus traditional bacterial culture.

Observation of coral spawning times is valuable to detect spawning patterns and identify the potential mechanisms behind coral reproduction. Although large amount of data on global coral spawning records have become available over the past decades, information on coral spawning in the northern South China Sea remains scarce. In this study, we investigated the spawning patterns of scleractinian corals in Luhuitou fringing reef, Hainan Island, China, from 2009 to 2021 in relation to lunar cycles (month and day). The spawning times of 22 coral species from five genera (Acropora, Montipora, Platygyra, Dipsastraea, and Galaxea) within three families (Acroporidae, Merulinidae, and Euphylliidae) were recorded, with spawning occurring from lunar February to May 2009–2021. Recorded spawning events started at a period of increasing seawater temperature. Acropora, the most documented genus, spawned between lunar February and April, but primarily in lunar March. Importantly, the spawning time of Acropora was delayed for one lunar month in 2016 most likely due to a rapid decrease in monthly mean seawater temperature in lunar February. Spawning lunar days before, on or after full moon of corals in the Luhuitou Reef, including those of the Acropora species, were highly variable between years even for the same species. No predictable pattern of spawning times can thus be detected. Nonetheless, our results fill up an information gap on coral spawning patterns in the northern South China Sea that may be useful for further understanding of the reproductive biology of scleractinian corals throughout the Indo–West Pacific.
