Abstract Negative pressure wound therapy with instillation and dwell time (NPWTi-d) is increasingly used for a diverse range of wounds. Meanwhile, the topical wound irrigation solution consisting of polyhexamethylene biguanide and betaine (PHMB-B) has shown efficacy in managing wound infections. However, the effectiveness of this solution as a topical instillation solution for NPWTi-d in patients with diabetic foot infections (DFIs) has not been thoroughly studied. The objective of this retrospective study was to evaluate the impact of using PHMB-B as the instillation solution during NPWTi-d on reducing bioburden and improving clinical outcomes in patients with DFIs. Between January 2017 and December 2022, a series of patients with DFIs received treatment with NPWTi-d, using either PHMB-B or normal saline as the instillation solution. Data collected retrospectively included demographic information, baseline wound characteristics, and treatment outcomes. The study included 61 patients in the PHMB-B group and 73 patients in the normal saline group, all diagnosed with DFIs. In comparison to patients treated with normal saline, patients with PHMB-B exhibited no significant differences in terms of wound bed preparation time (P = 0.5034), length of hospital stay (P = 0.6783), NPWTi-d application times (P = 0.1458), duration of systematic antimicrobial administration (P = 0.3567), or overall cost of hospitalization (P = 0.6713). The findings of the study suggest that the use of either PHMB-B or normal saline as an instillation solution in NPWTi-d for DFIs shows promise and effectiveness, yet no clinical distinction was observed between the two solutions.

Dahongpao mother tree (Camellia sinensis (L.) O. Ktze) is a representative of Wuyi rock tea. Whether there is a difference in rhizosphere soil microbial diversity and function between asexually propagated cuttings of Dahongpao (PD) and the parent Dahongpao mother tree (MD) has not been reported. In this study, high throughput sequencing technology was used to analyze rhizosphere soil microbial diversity, functions and their relationship with soil available nutrients and enzyme activities in MD and PD. The results showed that available nitrogen, phosphorus and potassium contents and urease, protease, acid phosphatase and sucrase activities of rhizosphere soils in MD were significantly higher than those in PD. Both bacterial and fungal diversity were higher in rhizosphere soils in MD than in PD, and secondly, the bacterial community structure was less stable while the fungal community structure was more stable in PD compared to MD. There were significant differences between MD and PD tea tree rhizosphere soils in 6 genera of characteristic bacteria and 4 genera of characteristic fungi. The results of function and interaction effect analysis showed that the rhizosphere soil available nutrient content and enzyme activities in MD were significantly higher than those in PD, and their contributions mainly originated from Pirellula and Acidisphaera of characteristic bacteria and Alatospora of characteristic fungi. Secondly, MD maybe had a stronger ability to inhibit soil pathogens than PD, with the main contribution coming from Scopulariopsis and Tolypocladium of characteristic fungi. Overall, compared with PD, soil texture in MD was relatively better, and its soil nutrient cycling-related enzyme activities were stronger, which was more favorable to soil nutrient cycling and increased the available nutrient content of the soil, which in turn promoted the growth of tea trees. This study provides an important reference for the planting and management of tea tree cuttings and microbial regulation of tea tree growth.

Aerospace breeding is a breeding technique that utilizes a spacecraft to position plants in a space environment for mutagenesis, which is conducive to rapid mutagenesis for the screening of superior plant varieties. In this study, tea trees with aviation mutagenesis (TM) and those without aviation mutagenesis (CK) were selected as research subjects to analyze the effects of aviation mutagenesis on the growth, physiological properties, and hormone metabolism of tea trees, and to further screen the characteristic hormones and validate their functions. The results showed that the leaf length, leaf width, and leaf area of TM tea trees were significantly larger than those of CK. The growth indexes, the photosynthetic physiological indexes (i.e., chlorophyll content, intercellular CO2 concentration, stomatal conductance, transpiration rate, and photosynthetic rate), and the resistance physiological indexes (i.e., superoxide dismutase, peroxidase, catalase, and soluble sugar) were significantly higher in TM than in CK. Hormone metabolome analysis showed that four characteristic hormones distinguished CK from TM, namely, l-tryptophan, indole, salicylic acid, and salicylic acid 2-O-β-glucoside, all of which were significantly more abundant in TM than in CK. These four characteristic hormones were significantly and positively correlated with the growth indexes, tea yield, and the photosynthetic and resistance physiological indexes of tea trees. The leaf area, chlorophyll content, photosynthetic rate, and superoxide dismutase activity of tea tree seedlings after spraying with the four characteristic hormones were significantly increased, in which salicylic acid and salicylic acid 2-O-β-glucoside were more favorable to increase the leaf area and superoxide dismutase activity, while l-tryptophan and indole were more favorable to increase the leaf chlorophyll content and photosynthetic rate. It can be observed that aviation mutagenesis improves the accumulation of the characteristic hormones of tea trees, enhances their photosynthetic capacity, improves their resistance, promotes their growth, and then improves the tea yield.

With the high intensification of poultry breeding, a series of diseases caused by pathogenic bacteria threaten the health of poultry and human. Among them, poultry diseases induced by Escherichia coli cause significant economic loss every year. The aim of this study was to investigate the effects of dietary supplementation with Artemisia annua L. polysaccharide (AAP) on the growth performance and intestinal barrier function of broilers with Escherichia coli (E. coli) challenge. A total of 256 one-day-old chicks were randomly assigned to four treatment groups: control group (fed basal diet), AAP group (fed basal diet supplemented with AAP), E. coli group (fed basal diet and orally administered E. coli), AAP + E. coli group (fed basal diet supplemented with AAP and orally administered E. coli). Dietary AAP supplementation elevated the BW, ADG and ADFI in non-challenged broilers. AAP also increased the apparent metabolic rate of EE and Ca in E. coli-challenged broilers. Moreover, AAP not only enhanced the serum IgA content but also decreased the serum and jejunum content of IL-6, as well as the jejunum level of IL-1β in non-challenged broilers. AAP also down-regulates the mRNA level of inflammatory factors (IL-1β, IL-6, and TNF-α) by inhibiting the mRNA expression of TLR4 and MyD88 in intestinal NF-κB signaling pathway of E. coli-challenged broilers. Meanwhile, AAP up-regulates the activity and mRNA level CAT by down-regulating the mRNA level of Keap1 in intestinal Nrf2 signaling pathway of E. coli-challenged broilers, and decreased serum MDA concentration. AAP significantly elevated the mRNA level of CAT, SOD and Nrf2 in jejunal of non-challenged broilers. Interestingly, AAP can improve intestinal physical barrier by down-regulating serum ET content, increasing the jejunal villus height/crypt depth (VH/CD) and ZO-1 mRNA level in broilers challenged by E. coli. AAP also elevated the VH/CD and the mRNA level of Occludin, ZO-1, Mucin-2 in non-challenged broilers. Importantly, AAP reshaped the balance of jejunum microbiota in E. coli-challenged broilers by altering α diversity and community composition. In summary, AAP ameliorated the loss of growth performance in broilers challenged with E. coli, probably by regulating the intestinal permeability and mucosa morphology, immune function, antioxidant ability, and microbiota.

Objective: Stent intimal hyperplasia leads to in stent restenosis and thrombosis. This study determined whether Fibulin-1 activity in smooth muscle cells (SMCs) contributes to stent restenosis or thrombosis. Methods: Stent implantation was conducted in a pig model. Target vessel samples were stained and analyzed by protein mass spectrometry. Cell experiments and Fibulin-1 SMC specific knockout mice (Fbln1SMKO) were used to investigate the mechanism of Fibulin-1 induced SMC proliferation and thrombosis. Results: SMC proliferation and phenotypic transition are the main pathological changes of intimal hyperplasia in venous stents. Protein mass spectrometry analysis revealed a total of 67 upregulated proteins and 39 downregulated proteins in intimal hyperplasia after stent implantation compared with normal iliac vein tissues. Among them, Fibulin-1 ranked among the top proteins altered. Fibulin-1 overexpressing human SMCs (Fibulin-1-hSMCs) showed increased migration and phenotypic switching from contractile to secretory type and Fibulin-1 inhibition decreased the activity of SMCs. Mechanistically, Fibulin-1-hSMCs displayed increased levels of angiotensin converting enzyme (ACE) expression and angiotensin II signaling. Inhibition of ACE or angiotensin II signaling alleviated the migration of Fibulin-1-hSMCs. Using Fibulin-1 SMC specific knockout mice (Fbln1SMKO) and venous thrombosis model, we demonstrated that Fibulin-1 deletion attenuated intimal SMCs proliferation and thrombosis. Further, Fibulin-1 concentration was high in iliac vein compression syndrome (IVCS) patients treated with stent and was an independent predictor of venous insufficiency. Conclusions: Fibulin-1 promotes SMC proliferation partially through ACE secretion and angiotensin II signaling after stent implantation. Fibulin-1 plays a role in venous insufficiency syndrome, implicating the protein in the detection and treatment of IVCS.

Abstract Introduction Lung adenocarcinoma (LUAD) is the major type of non‐small cell lung cancer with low a survival rate caused by metastasis. SCN4B encoding voltage‐gated sodium channel β subunit is regarded as a metastasis‐suppressor gene. We aim to explore how SCN4B influences the progression and prognosis of LUAD. Methods The gene expression profiles of 585 LUAD samples in TCGA and GSE31210, GSE116959, and GSE72094 datasets from the GEO database were downloaded for analysis. Differentially expressed genes were obtained through the “limma” package. The “clusterProfiler” package was used to conduct GSEA. Survival analysis was conducted via “survival” and “survminer” packages. Transcription factors regulating SCN4B expression were screened by correlation analysis and further predicted by FIMO. Infiltration of immune cells was analyzed by CIBERSORT. ESTIMATE algorithm was used to evaluate the immune‐related scores. Results SCN4B expressed higher in normal samples than in LUAD samples and higher in female samples than male samples. One hundred and twenty‐six pathways were significantly enriched between high and low SCN4B expression groups. Six transcription factors' expressions were positively related to SCN4B expression, and ChIP‐seq data from “Cistrome” verified that TAL1 and ERG might bind to the upstream sequence of SCN4B. SCN4B expression was significantly correlated with activated memory CD4 T cells, resting mast cells, and monocytes. TMB status, three scores based on ESTIMATE algorithm, and expression of three immune checkpoints showed significant differences between SCN4B high‐ and low‐expression groups. SCN4B could be considered as an independent prognostic signature of LUAD patients that higher expression represents a better prognosis. Conclusion SCN4B expresses higher in normal samples, and SCN4B is able to be an independent prognostic signature for LUAD patients. TAL1 and ERG may regulate the expression of SCN4B by binding its upstream sequences. Our research is valuable in improving the effectiveness of treatment in LUAD.

It is generally recognized that the quality differences in plant germplasm resources are genetically determined, and that only a good “pedigree” can have good quality. Ecological memory of plants and rhizosphere soil fungi provides a new perspective to understand this phenomenon. Here, we selected 45 tea tree germplasm resources and analyzed the rhizosphere soil fungi, nutrient content and tea quality. We found that the ecological memory of tea trees for soil fungi led to the recruitment and aggregation of dominant fungal populations that were similar across tea tree varieties, differing only in the number of fungi. We performed continuous simulation and validation to identify four characteristic fungal genera that determined the quality differences. Further analysis showed that the greater the recruitment and aggregation of Saitozyma and Archaeorhizomyces by tea trees, the greater the rejection of Chaetomium and Trechispora, the higher the available nutrient content in the soil and the better the tea quality. In summary, our study presents a new perspective, showing that ecological memory between tea trees and rhizosphere soil fungi leads to differences in plants’ ability to recruit and aggregate characteristic fungi, which is one of the most important determinants of tea quality. The artificial inoculation of rhizosphere fungi may reconstruct the ecological memory of tea trees and substantially improve their quality.

Objective To evaluate the effects of acupuncture on physical function, depression and quality of life in stroke patients. Methods A systematic search of the publicly available databases such as the Cochrane Library, PubMed, Embase, China Knowledge Network (CNKI), Wan Fang Data, VIP Database (VIP) and China Biomedical Literature Database (CBM) was conducted, and randomized controlled trials on the use of acupuncture in stroke patients from its inception to April 10, 2023 were included for the Meta analysis. Results Thirteen studies were included. The results showed that acupuncture could promoted recovery of limb function ［MD=6. 86, 95%CI (5. 63, 8. 08), P＜0. 01］, improved depressive symptoms common in post-stroke patients ［MD=-6. 95, 95%CI (-9. 32, -4. 57), P＜0. 01］, and improved patients' quality of life ［SMD=1. 83, 95%CI (1. 63, 2. 04), P＜0. 01］. Conclusion Acupuncture can not only promote the recovery of limb function, but also improve depressive symptoms common in post-stroke patients and enhance their quality of life. Tea plant is an acidophilic plant, and soil pH has an important effect on the absorption and enrichment of elements, tea plant growth and quality. In this study, rhizosphere soils and leaves of tea plants from 30 tea plantations were collected to determine soil pH and multi-element content of soil and leaves of tea plants, to obtain and validate key elements that are enriched by pH affecting tea plants, and to analyze the effects of pH on the growth and quality of tea plants. The results showed that soil pH significantly affected the enrichment of 15 elements by tea plants, and the enrichment coefficients of 11 elements (C, Mg, Si, N, P, Mn, Sr, Cd, S, Ca and Sb) tended to increase significantly with the increase of soil pH, while the opposite was true for the other four elements (Cu, Rb, Ba and Al). TOPSIS analysis showed that soil pH had the greatest effect on tea plant enrichment of seven elements, namely N (100%), Mn (43.32%), C (39.22%), P (27.66%), Sr (15.30%), Mg (13.41%) and Ba (10.47%). Pot experiments with tea seedlings also verified that soil pH significantly affected the enrichment of tea leaves for seven key elements. Moreover, with the increase of soil pH, the growth indexes, photosynthesis indexes and quality indexes of tea seedlings showed a significant upward trend. Interaction analysis showed that the enhanced enrichment of N, Mn, C, P, Sr and Mg by tea plants was beneficial to increase the photosynthetic capacity of tea plants, promote the growth of tea plants and improve the quality of tea leaves. This study provides an important theoretical basis for the cultivation and management of tea plants.

There are obvious differences in quality between different varieties of the same plant, and it is not clear whether they can be effectively distinguished from each other from a bacterial point of view. In this study, 44 tea tree varieties (Camellia sinensis) were used to analyze the rhizosphere soil bacterial community using high-throughput sequencing technology, and five types of machine deep learning were used for modeling to obtain characteristic microorganisms that can effectively differentiate different varieties, and validation was performed. The relationship between characteristic microorganisms, soil nutrient transformation, and tea quality formation was further analyzed. It was found that 44 tea tree varieties were classified into two groups (group A and group B) and the characteristic bacteria that distinguished them came from 23 genera. Secondly, the content of rhizosphere soil available nutrients (available nitrogen, available phosphorus, and available potassium) and tea quality indexes (tea polyphenols, theanine, and caffeine) was significantly higher in group A than in group B. The classification result based on both was consistent with the above bacteria. This study provides a new insight and research methodology into the main reasons for the formation of quality differences among different varieties of the same plant.

The quality of the Dahongpao mother tree (Camellia sinensis) remains a mystery to this day. In this study, for the first time, the differences between the Dahongpao mother tree (MD) and Dahongpao cuttings (PD), in terms of odor characteristics and taste characteristics were analyzed by metabomics. The results showed that MD had stronger floral, fruity, green, and woody odor characteristics than PD, and that the contributions were mainly from dihydromyrcenol, methyl salicylate, 2-isobutylpyrazine, 1,6-dihydrocarveol, gamma-terpineol, and linalyl acetate. Further, fresh and brisk taste and mellowness taste characteristics of MD were significantly higher than PD, with contributions mainly from amino acids and derivatives and organic acids. Secondly, bitterness taste characteristics of PD were significantly higher than MD, with contributions from phenolic acids, flavones, and flavonols. This study preliminarily unraveled the legend of the superior quality of the Dahongpao mother tree, and also provided an important reference for the breeding of tea-tree cuttings.

Aviation mutagenesis is a breeding method for the rapid selection of superior plant varieties. In this study, rhizosphere soil chemical indexes, soil enzyme activities, and soil metabolites were measured in Dahongpao tea trees with aviation mutagenesis (TM) and without aviation mutagenesis (CK). The main soil metabolites distinguishing TM and CK and their relationships with soil chemical indexes and soil enzyme activities were analyzed and obtained. The results showed that there was no significant change in the rhizosphere soils’ pH of TM tea trees compared to CK (p = 0.91), while all other chemical indexes of TM were significantly higher than CK (p < 0.05). In addition, the activities of enzymes related to soil nutrient cycling such as urease, protease, sucrase, acid phosphatase and cellulase, and enzymes related to soil antioxidants such as superoxide dismutase, catalase, peroxidase, and polyphenol oxidase were significantly increased (p < 0.05) in the rhizosphere soils of TM tea trees compared to CK. Soil metabolite analysis showed that the main soil metabolites distinguishing CK from TM were carbohydrates, nitrogen compounds, and amines. Of these, carbohydrates and nitrogen compounds were significantly positively correlated with soil chemical indexes and soil enzymes, whereas amine was significantly negatively correlated with soil chemical indexes such as organic matter, total nitrogen, total potassium, available nitrogen, available phosphorus; amine showed significant negative correlation with soil enzymes such as catalase, peroxidase, polyphenol oxidase, and urease. It can be seen that aviation mutagenesis is conducive to improving the ability of tea tree rhizosphere aggregation and transformation of soil nutrients, increasing the total amount of soil nutrients and the content of available nutrients, which is more conducive to promoting the uptake of nutrients by the tea tree, and thus promoting the growth of the tea tree.

Abstract To investigate the ability of the ratio of blood urea nitrogen (BUN) to serum albumin ratio (BAR) in patients with sepsis in intensive care units (ICUs) to predict the prognosis of short-and long-term death. Data are from the Marketplace for Intensive Care Medical Information IV (MIMIC-IV v2.0) database for patients with sepsis as defined by SEPSIS-3. The primary outcome was 30-day mortality and the secondary outcome was 360-day mortality. Kaplan–Meier (KM) survival curves were plotted to describe differences in BAR mortality in different subgroups and area under the curve (AUC) analysis was performed to compare the predictive value of sequential organ failure assessment (SOFA), BAR, blood urea nitrogen (BUN) and albumin. Multivariate Cox regression models and subgroup analysis were used to determine the correlation between BAR and 30-day mortality and 360-day mortality. A total of 7656 eligible patients were enrolled in the study with a median BAR of 8.0 mg/g, including 3837 in the ≤ 8.0 group and 3819 in the BAR > 8.0 group, with 30-day mortality rates of 19.1% and 38.2% (P

BackgroundAcute respiratory distress syndrome (ARDS) is a serious lung ailment marked by significant inflammation and damage in the alveoli and capillaries of the lungs. Recent research suggests a strong correlation between the onset and advancement of ARDS and an imbalance in the gut microbiota (GM).MethodsIn this investigation, Mendelian randomization (MR) analysis was utilized, drawing on data from publicly accessible genome-wide association studies. The primary focus was on examining the interplay between GM, inflammatory factors (IFs) and ARDS. Instrumental variables were established through genetic modifications of GM and IFs. Various statistical analysis methods including the inverse-variance weighted model, MR-Egger method and Wald ratio test were applied for comprehensive data analysis.ResultsEight bacterial taxa within the GM demonstrated a potential causal link with development of ARDS. Notably, the phylum Actinobacteria and the genus Intestinibacter exhibited a negative association with the risk of ARDS. However, Erysipelotrichales (id. 2,148), Victivallis (id. 2,256), Ruminococcaceae UCG014 (id. 11,371), Eubacterium ruminantium group (id. 11,340), Erysipelotrichaceae (id. 2,149) and Erysipelotrichia (id. 2,147) demonstrated a positive association with ARDS risk. Additionally, the study identified a potential causal relationship between the inflammatory factors interleukin-16 and C-C motif chemokine 3 with the occurrence of ARDS.ConclusionThis study strongly suggests that the interaction between gut microbiota (GM) and inflammatory factors (IFs) significantly contributes to the pathogenesis of acute respiratory distress syndrome (ARDS). This underscores their crucial involvement in both the initiation and advancement of this severe lung disorder.

Hashimoto’s thyroiditis (HT) is an autoimmune illness caused by a combination of genetic, epigenetic, and environmental factors. The pathogenesis of HT is not fully elucidated, especially in epigenetics. The epigenetic regulator Jumonji domain-containing protein D3 (JMJD3) has been extensively investigated in immunological disorders. This study has been performed to explore the roles and potential mechanisms of JMJD3 in HT. Thyroid samples from patients and healthy subjects were collected. We first analyzed the expression of JMJD3 and chemokines in the thyroid gland using real-time PCR and immunohistochemistry. In vitro, the apoptosis effect of the JMJD3-specific inhibitor GSK-J4 on the thyroid epithelial cell line Nthy-ori 3-1 was evaluated using FITC Annexin V Detection kit. Reverse transcription-polymerase chain reaction and Western blotting were applied to examine the inhibitory effect of GSK-J4 on the inflammation of thyrocytes. In the thyroid tissue of HT patients, JMJD3 messenger RNA and protein levels were substantially greater than in controls (P < 0.05). Chemokines C–X–C motif chemokine ligand 10 (CXCL10) and C–C motif chemokine ligand 2 (CCL2) were elevated in HT patients, and thyroid cells with stimulation of tumor necrosis factor α (TNF-α). GSK-J4 could suppress TNF-α-induced synthesis of chemokines CXCL10 and CCL2 and prohibit thyrocyte apoptosis. Our results shed light on the potential role of JMJD3 in HT and indicate that JMJD3 may become a novel therapeutic target in HT treatment and prevention.

Objective To establish a fungal image-assisted classification model using deep learning technology. Methods The microscope images of people infected with Aspergillus, Saccharomyces and Cryptococcus neoformans were retrospectively collected from the Eighth Medical Center of PLA General Hospital from September 2020 to April 2021. The images were randomly divided into training set, validation set and test set according to the ratio of 7∶1.5∶1.5. The improved MobileNetV2 network structure was trained using the training set, a convolutional neural network (CNN) fungal image 11 classification model based on multi-scale attention mechanism was constructed and the parameters were debugged based on the validation set. Machine identification results were taken as the gold standard, the performance of the model on 11 fungal image classification tasks was evaluated, and the results were shown by precision, recall and F1 value. In addition, the performance of the proposed model with 5 classic CNN models were compared, and the results were measured in terms of model parameters, memory usage, frames per second (FPS), accuracy, and area under the curve (AUC) of receiver operating characteristic curve. Results A total of 7666 fungal microscope images were collected, including 2781, 4115, and 770 images of Aspergillus, Saccharomyces, and Cryptococcus neoformans, respectively. Among them, there were 5366 training images, 1150 validation images, and 1150 test images. The improved MobileNetV2 model had high performance for the classification of 11 fungal images in the test set. The precision rate was distributed between 96.36% and 100%, the recall rate was distributed between 96.53% and 100%, and the F1 value was distributed between 97.01% and 100%. The parameters, memory usage, FPS, accuracy, and AUC of the improved MobileNetV2 model were 4.22 M, 356.89 M, 573, (99.09±0.18)%, and 0.9944±0.0018, respectively, and the comprehensive performance was better than 5 kinds of classical networks. Conclusion The proposed fungal image classification model based on the improved MobileNetV2 can obtain higher fungal image recognition ability while maintaining low computational cost, with an overall performance better than classical CNN model.

Abstract Background Atherogenic index of plasma (AIP) as a newly discovered blood lipid parameter was shown to be strongly correlated with coronary artery disease (CAD). However, the blood lipid levels changed with age, so we speculated that the correlation between AIP and CAD was also affected by age. Methods A retrospective study was performed on male patients with confirmed or suspected CAD who underwent coronary angiography (CAG) from July 2017 to March 2021. Patients were divided into younger, middle early, middle late, and elderly groups. Patients in each age group were further divided into the non-coronary artery disease (n-CAD) and CAD subgroups according to the CAG diagnostic results. The relationship between blood lipid parameters and CAD was assessed in each age group. Results Age-dependent changes of blood lipid levels were mainly found in CAD patients but not in n-CAD patients. With increased age, the levels of triglyceride, total cholesterol, low-density lipoprotein cholesterol (LDL-C) and AIP were gradually decreased, whereas high-density lipoprotein cholesterol concentration was increased in CAD patients. Multivariate logistic regression analyses showed that AIP was an independent risk factor for CAD in middle early (OR 2.601; CI 1.160–5.832, P = 0.02) and middle late age group (OR 2.707, CI 1.201–6.100, P = 0.016), but not in the younger and elderly groups. LDL-C was an independent risk factor for CAD in all age groups. The areas under curve of AIP for detecting CAD in the middle early and middle late age groups were not higher than that of other blood lipid parameters. Conclusion Although this was a single-center study for males only, the correlation between AIP level and CAD risk was age-dependent. AIP was an independent risk factor for CAD in the middle-aged groups. However, the predictive value of AIP for detecting CAD was not better than that of the traditional blood lipid parameters such as LDL-C.

There are few studies on the shape and structure of the channel-type induction heating tundish on multi-physics field. Computational fluid dynamics has been used to study the influence of the structure of the tundish on the macroscopic transport behavior of the tundish with channel induction heating. The results show that increasing the depth of the molten pool is conducive to dynamic behavior of multiphase, the deeper the molten pool, the larger the active area, the longer residence time, the more inclusions removal and the higher ratio of plug to dead volume. Meanwhile, the larger the channel diameter, the more inclusions removal in the receiving chamber and channel. The channel induction heating has enough ability to increase the superheat and temperature compensate for the heat loss caused by the excessive residence time of the molten steel in the tundish. The change in the channel structure is crucial to the macroscopic transport behavior of the fluid. The change in channel diameter has the greatest effect on the multi-physics field in the molten pool.

Myocardial fibrosis is the characteristic pathology of diabetes-induced cardiomyopathy. Therefore, an in-depth study of cardiac heterogeneity and cell-to-cell interactions can help elucidate the pathogenesis of diabetic myocardial fibrosis and identify treatment targets for the treatment of this disease. In this study, we investigated intercellular communication drivers of myocardial fibrosis in mouse heart with high-fat-diet/streptozotocin-induced diabetes at single-cell resolution. Intercellular and protein–protein interaction networks of fibroblasts and macrophages, endothelial cells, as well as fibroblasts and epicardial cells revealed critical changes in ligand–receptor interactions such as Pdgf(s)–Pdgfra and Efemp1–Egfr, which promote the development of a profibrotic microenvironment during the progression of and confirmed that the specific inhibition of the Pdgfra axis could significantly improve diabetic myocardial fibrosis. We also identified phenotypically distinct Hrchi and Postnhi fibroblast subpopulations associated with pathological extracellular matrix remodeling, of which the Hrchi fibroblasts were found to be the most profibrogenic under diabetic conditions. Finally, we validated the role of the Itgb1 hub gene-mediated intercellular communication drivers of diabetic myocardial fibrosis in Hrchi fibroblasts, and confirmed the results through AAV9-mediated Itgb1 knockdown in the heart of diabetic mice. In summary, cardiac cell mapping provides novel insights into intercellular communication drivers involved in pathological extracellular matrix remodeling during diabetic myocardial fibrosis.

Benshan tea is a kind of oolong tea, and Benshan (Camellia sinensis) tea tree originates from Anxi County of Fujian Province in China, which is a national tea tree breed. Tea processing is the key to the formation of its odor characteristics. It is extremely important to step by step analyze effects of tea processing on aroma intensity and the formation of odor characteristics for optimizing tea processing process and improving tea quality. The results of this study showed that processing resulted in a significant increase in the content of volatile compounds in tea leaves, i.e., from 25.213 μg/kg to 111.223 μg/kg, in which the volatile compounds were mainly terpenoids. Secondly, the analysis found that 20 kinds of key compounds constituted to odor characteristics of Benshan tea leaves, among which geraniol, trans-β-ionone, gerol, citronellol, benzeneacetaldehyde, and trans-nerolidol were the most key six. Floral and fruity aromas, especially floral aroma, mainly formed odor characteristics of Benshan tea after processing, while floral aroma mainly came from the contribution of geraniol, which was the foremost compound in the formation of floral aroma of Benshan tea.

Hematopoietic stem cells (HSCs) are stem cells that can differentiate into various blood cells and have long-term self-renewal capacity. At present, HSC transplantation is an effective therapeutic means for many malignant hematological diseases, such as aplastic hematological diseases and autoimmune diseases. The hematopoietic microenvironment affects the proliferation, differentiation, and homeostasis of HSCs. The regulatory effect of the hematopoietic microenvironment on HSCs is complex and has not been thoroughly studied yet. In this study, we focused on mononuclear cells (MNCs), which provided an important microenvironment for HSCs and established a methodological system for identifying cellular composition by means of multiple technologies and methods. First, single-cell RNA sequencing (scRNA-seq) technology was used to investigate the cellular composition of cells originating from different microenvironments during different stages of hematopoiesis, including mouse fetal liver mononuclear cells (FL-MNCs), bone marrow mononuclear cells (BM-MNCs), and in vitro-cultured fetal liver stromal cells. Second, bioinformatics analysis showed a higher proportion and stronger proliferation of the HSCs in FL-MNCs than those in BM-MNCs. On the other hand, macrophages in in vitro-cultured fetal liver stromal cells were enriched to about 76%. Differential gene expression analysis and Gene Ontology (GO) functional enrichment analysis demonstrated that fetal liver macrophages have strong cell migration and actin skeleton formation capabilities, allowing them to participate in the hematopoietic homeostasis through endocytosis and exocytosis. Last, various validation experiments such as quantitative real-time PCR (qRT-PCR), ELISA, and confocal image assays were performed on randomly selected target genes or proteins secreted by fetal liver macrophages to further demonstrate the potential relationship between HSCs and the cells inhabiting their microenvironment. This system, which integrates multiple methods, could be used to better understand the fate of these specific cells by determining regulation mechanism of both HSCs and macrophages and could also be extended to studies in other cellular models.

Through taking the cold rolling process as the research object, the three-dimensional finite element model of the strip rolling process is established by using ANSYS/LS-DYNA software. The actual rolling product data has strong consistency with the finite element simulation results. The rolling process is dynamically simulated, and the distribution curves of important rolling parameters such as equivalent stress, control efficiency coefficient, transverse rolling pressure, lateral thickness and work roll deflection is obtained. Based on summarizing the influence of rolling parameters on rolling deformation, the research results of this paper can play an important role in the actual rolling process control. The research results have certain guiding significance for the development and optimization of the rolling control system.

Objective: To investigate the outcomes of patients with mucoepidermoid carcinoma of the palate undergoing pedicled facial-submental artery island flap (FSIF) reconstruction following resection. Patients and methods: 41 patients with early stage disease and 9 patients with advanced-stage disease underwent radical excision and neck dissection. 37 IIb, 4 class IIa and 9 IIIb maxillary defects were reconstructed with FSIF, folded FSIF or folded FSIF with titanium mesh respectively. The skin paddles were 3 × 8 to 5 × 15 cm and 3 × 8 to 5 × 14 cm, respectively. 5 patients with high grade disease were treated with cobalt 60 adjuvant radiotherapy after operation. Results: One flap failure occurred, yielding a success rate of 98.0% in the reconstruction of palate II and III defects with FSIF or titanium mesh. The patients were seen for follow-up for 16–60 months postoperative. 76.0% patients alive with no disease (AND); 14.0% patients alive with disease (AD) and 10.0% died of disease (DD). Introduction: Nuclear respiratory factor 1 (NRF1) is an important regulator involved in mitochondrial biogenesis and energy metabolism. However, the specific mechanism of NRF1 in anoikis and epithelial-mesenchymal transition (EMT) remains unclear. Methods: We examined the effect of NRF1 on mitochondria and identified the specific mechanism through transcriptome sequencing, and explored the relationships among NRF1, anoikis, and EMT. Results: We found that upregulated NRF1 expression led to increased mitochondrial oxidative phosphorylation (OXPHOS) and ATP generation. Simultaneously, a significant amount of ROS is generated during OXPHOS. Alternatively, NRF1 upregulates the expression of ROS-scavenging enzymes, allowing tumor cells to maintain low ROS levels and promoting anoikis resistance and EMT. We also found that exogenous ROS was maintained at a low level by NRF1 in breast cancer cells. Conclusion: our study provides mechanistic insight into the function of NRF1 in breast cancer, indicating that NRF1 may serve as a therapeutic target for breast cancer treatment.

Processing is extremely important for the formation of aroma characteristic of tea leaves. In this study, the effects of processing on the content of volatile compounds, aroma intensity and odor characteristic of Shuixian tea were analyzed. The results showed that the content of volatile compounds in Shuixian tea increased significantly after processing, among which terpenoids and esters were the highest. There were 18 key compounds constituting the aroma characteristics of Shuixian tea, among which geraniol and nerol were the most important compounds, which contributed 96.28% to the aroma of Shuixian tea. The odor characteristics of Shuixian tea were mainly floral and fruity and the contribution of floral mainly came from geraniol, while fruity mainly came from nerol. Geraniol and nerol compounds increased rapidly after the withering process of tea leaves. This study provided an important reference for the improvement of processing technology and quality enhancement of Shuixian tea.

Acidification can seriously affect the growth of tea trees and the yield and quality of tea leaves. In this study, we analyzed the effects of acidification on the physicochemical properties, microorganisms and metabolites of tea rhizosphere soils with different pH values, and the results showed that with the increase of soil pH, the organic matter content, cation exchange capacity, microbial biomass carbon, microbial biomass nitrogen, microbial respiration intensity, bacterial number and actinomyces number in tea rhizosphere soil all showed an increasing trend, while the fungi number decreased. The results of soil metabolite analysis showed that 2376, 2377 and 2359 metabolites were detected in tea rhizosphere soil with pH values of 3.29, 4.74 and 5.32, respectively, and the number of similar compounds reached 2331, accounting for more than 98%. The results of soil metabolite content analysis showed that with the increase of soil pH, the total contents of metabolite of tea rhizosphere soil increased significantly. The results of correlation analysis between physicochemical indexes of soil and microorganisms and soil metabolites showed that physicochemical indexes of soil and microorganisms were significantly correlated with 221 soil metabolites, among which 55 were significantly positively correlated and 166 were significantly negatively correlated. Based on correlation interaction network analysis, 59 characteristic compounds were obtained and divided into 22 categories, among which 7 categories compounds showed a significant increasing trend with the increase of soil pH, while the other 15 categories compounds showed the opposite trend. Based on the functional analysis of characteristic metabolites, this study found that with the increase of soil pH in tea rhizosphere, the diversity and number of soil microorganisms increased, and the cyclic ability of C and N of tea rhizosphere soil was enhanced, which in turn might lead to the enhancement of resistance of tea tree and promote the growth of tea tree.

Abstract Head and neck squamous cell carcinoma (HNSCC) is the most common malignant tumor of the head and neck, and the prognosis of patients is poor due to chemotherapeutic resistance. Interestingly, patients with HNSCC induced by human papillomavirus (HPV) infection are more sensitive to chemotherapy and display a better prognosis than HPV‐negative patients. The biological relevance of HPV infection and the mechanism underlying chemosensitivity to cisplatin remain unknown. Herein, SERPINB3 is identified as an important target for regulation of cisplatin sensitivity by HPV‐E6/E7 in HNSCC. Downregulation of SERPINB3 inhibits cisplatin‐induced DNA damage repair and enhances the cytotoxicity of cisplatin. In detail, decreasing SERPINB3 expression reduces the USP1‐mediated deubiquitination of FANCD2–FANCI in the Fanconi anemia pathway, thereby interfering with cisplatin‐induced DNA interstrand crosslinks repair and further contributing to HNSCC cell apoptosis. To translate this finding, pH‐responsive nanoparticles are used to deliver SERPINB3 small interfering RNA in combination with cisplatin, and this treatment successfully reverses cisplatin chemotherapeutic resistance in a patient‐derived xenograft model from HPV‐negative HNSCC. Taken together, these findings suggest that targeting SERPINB3 based on HPV‐positive HNSCC is a potential strategy to overcome cisplatin resistance in HPV‐negative HNSCC and improves the prognosis of this disease.

Abstract Receptor-interacting protein kinase 1 (RIPK1) and 3 (RIPK3) are critical regulators of programmed necrosis or necroptosis. However, the role of the RIPK1/RIPK3 signaling pathway in myocardial fibrosis and related diabetic cardiomyopathy is still unclear. We hypothesized that RIPK1/RIPK3 activation mediated myocardial fibrosis by impairing the autophagic flux. To this end, we established in vitro and in vivo models of type 2 diabetes mellitus with high glucose fat (HGF) medium and diet respectively. HGF induced myocardial fibrosis, and impaired cardiac diastolic and systolic function by activating the RIPK1/RIPK3 pathway, which increased the expression of autophagic related proteins such as LC3-II, P62 and active-cathepsin D. Inhibition of RIPK1 or RIPK3 alleviated HGF-induced death and fibrosis of cardiac fibroblasts by restoring the impaired autophagic flux. The autophagy blocker neutralized the effects of the RIPK1 inhibitor necrostatin-1 (Nec-1) and RIPK3 inhibitor GSK872 (GSK). RIPK1/RIPK3 inhibition respectively decreased the levels of RIPK3/p-RIPK3 and RIPK1/p-RIPK1. P62 forms a complex with RIPK1-RIPK3 and promotes the binding of RIPK1 and RIPK3, silencing of RIPK1 decreased the association of RIPK1 with P62 and the binding of P62 to LC3. Furthermore, inhibition of both kinases in combination with a low dose of Nec-1 and GSK in the HGF-treated fibroblasts significantly decreased cell death and fibrosis, and restored the autophagic flux. In the diabetic rat model, Nec-1 (1.65 mg/kg) treatment for 4 months markedly alleviated myocardial fibrosis, downregulated autophagic related proteins, and improved cardiac systolic and diastolic function. In conclusion, HGF induces myocardial fibrosis and cardiac dysfunction by activating the RIPK1-RIPK3 pathway and by impairing the autophagic flux, which is obviated by the pharmacological and genetic inhibition of RIPK1/RIPK3.

The concentration of atmospheric ozone (O3) pollution is showing a rapid growing tendency, and O3 pollution has become one of the bottleneck issues that restrict the continuous improvement of air quality in China. In this study, we first identified the primary factors based on the source apportionment of O3, then used factor analysis to divide these selected factors into economic and environmental categories. The geographical detector model was used to analyze the impact of factors and their interactions on O3 concentration in 41 cities in the Yangtze River Delta (YRD) region in 2020. The results showed that forest coverage ranked first among all the detected factors, suggesting a strong relationship between the regional O3 concentration and forest coverage. The driving factors of economic activity were ranked as follows: actual utilization of foreign capital (0.400) > gross domestic product (GDP) per capita (0.387) > proportion of tertiary industry (0.360) > urbanization rate (0.327) > per capita consumption expenditure (0.194) > research and development (R&D) of full-time equivalents of industrial enterprises above designated size (0.182) > number of industrial enterprises (0.126). The interaction between any two factors enhanced their influence on O3 concentration more than any single factor, indicating that the variability of regional O3 concentration was an outcome of a combination of multiple factors. This study could provide recommendations for the prevention and control of O3 pollution and the development of ecological integration in the YRD region.

Soil acidification in tea plantation seriously reduced the yield and quality of tea. It was an effective method to use organic fertilizer for acidified soil remediation to ensure tea yield and quality. In this study, different fertilizers were used to treat the acidified tea plantation soils for 4 consecutive years to analyze the remediation effect of different fertilizers on acidified soil and their effects on tea yield and quality. The results showed that during the period of 2017-2021, the soil pH value of tea plantation (S1) with long-term use of chemical fertilizer decreased continuously, from 3.07 to 2.82. In the tea plantation (S2), the soil pH value was stable between 4.26 and 4.65 in the combination of organic fertilizer and chemical fertilizer for a long time. The tea plantation (S3) with long-term use of organic fertilizer has a stable soil pH value between 5.13 and 5.33, which is most suitable for the growth of tea trees. The analysis results of tea yield and quality indicators (tea polyphenols, theanine, amino acids, caffeine, catechin components) showed that after long-term use of chemical fertilizer in S1 tea plantation, soil pH value decreased continuously, soil acidification intensified, tea tree growth was hindered, and tea yield and quality decreased continuously. S2 tea plantation used some organic fertilizer in combination with chemical fertilizer for a long time, the soil pH value gradually improved, soil acidification weakened, and tea yield and quality improved steadily. After long-term use of organic fertilizer in S3 tea plantation, soil acidification was significantly improved, which was conducive to the normal growth of tea trees and the yield and quality of tea reached the maximum. The results of interaction analysis showed that the long-term use of chemical fertilizer had a negative effect on the growth of tea trees, and the combination of organic fertilizer and chemical fertilizer improved the growth of tea trees to some extent, but the effect was poor, while the long-term use of organic fertilizer was the most beneficial to the growth of tea trees and most conducive to ensuring the yield and quality of tea. This study provides important practical significance for the remediation and fertilizer regulation of acidified tea plantation soils. In the process of field experiment, climate is a variable factor, and attention should be paid to the effect of climate change on fertilization efficiency in subsequent experiment.

Fluorescent sensors developed based on nano-materials have penetrated into cancer medical sensing research due to their unique properties, such as high sensitivity, high specificity. The number of cancer patients has continued to rise in recent years, and the limitations of traditional cancer diagnosis methods can no longer meet the demand for accurate early cancer screening. Accurate identification of cancer bio-macromolecular markers can significantly improve early diagnosis and follow-up treatment. To this end, we provide an overview of the properties and uses of different nano-materials and detail the principles and functions of fluorescent nano-biosensor detection for different cancer bio-macromolecular markers. In addition, the prospects and challenges of translating this convenient and visual diagnostic method into the clinical application are discussed.

This study aimed to investigate the distribution of microplastics (MPs) within the Shiwuli River in Hefei, a Chinese inland city. Water and sediment samples were collected during flood season (from May to September) and non-flood season (from October to April) at 10 representative points along the truck stream. The electron microscope, the laser direct infrared chemical imaging system (LDIR), and the scanning electron microscope (SEM) were used to observe and quantify the colour and shape of the MPs, to identify the number, size, and polymer composition of the MPs, and to observe the microstructures of typical MP particles, respectively. The polymer risk index (RI) model and the pollution load index (PLI) model were used to assess the polymer-related risks and the overall extent of MP pollution in the river, respectively. Analysis of MP abundance for different sampling points showed that the water of Shiwuli River had an average abundance of MPs of 8.4 ± 2.5 particles/L during the flood season and 5.8 ± 1.7 particles/L during the non-flood season; the sediment had an average abundance of MPs of 78.9 ± 8.3 particles/kg during the flood season and 63.9 ± 7.1 particles/kg during the non-flood season. The abundance of MPs of different points was investigated. Result show that the more abundances of MPs were found at confluences with tributaries (S4, S5, and S6), where they are also close to the residential and industrial development, while lower values were found in agricultural areas (S8) and wetland ecological regions (S9 and S10). In water, the maximum appeared at S5 with 21.7 ± 4.6 particles/L during the flood season and 15.9 ± 4.2 particles/L during the non-flood season, respectively; the minimum appeared at S9 with 1.8 ± 1.0 particles/L during the flood season and 2.2 ± 0.4 particles/L during the non-flood season, respectively. In sediment, the maximum appeared at S5 with 174.1 ± 10.1 particles/kg during the flood season and 143.6 ± 10.4 particles/kg during the non-flood season, respectively; the minimum appeared at S8 with 10.3 ± 2.8 particles/kg during the flood season and at S9 with 12.1 ± 3.2 particles/kg during the non-flood season, respectively. MP characteristics were also studied. Results show that the MPs mainly exhibited a fibroid morphology (27.90–34%), and red-coloured particles (19.10%) within the smaller size less than 500 μm (38.60%) were more prevalent. Additionally, the result of LDIR scanning shows that a total of eleven types of MP polymers were found in the river water and sediment, including acrylates (ACR), chlorinated polyethylene (CPE), ethylene vinyl acetate (EVA), polyethylene (PE), polyethylene terephthalate (PET), polypropylene (PP), polystyrene (PS), polyurethane (PU), polyvinylchloride (PVC), polyamide (PA), and silicon. The most common particle was PE (19.3–21.6%). Furthermore, the environmental risk assessment demonstrated that the PS polymer posed a Level-III risk in the water samples and a Level-II risk in the sediment samples from the Shiwuli River. The remaining polymer types exhibited Level-I risk. The PLIzone value for water was 2.24 during the flood season, indicating heavy pollution, and 1.66 during the non-flood season, indicating moderate pollution. Similarly, the PLIzone value for sediments was 2.34 during the flood season and 1.91 during the non-flood season, both suggesting a heavy pollution. These findings highlight the potential risk posed by MP pollution in the Shiwuli River to the quality of drinking water sources in Chaohu Lake in Hefei. They provide valuable insights into management, pollution control, and integrated management strategies pertaining to MPs in urban inland rivers in Hefei.

As one of the primary nodes in the flow of micro-plastics (MPs) in the environment, it is critical to examine and assess the Sewage Treatment, occurrence, and removal of MPs in waste treatment plant (WWTP). This research explored the shape, size, and composition of MPs at various stages of the WWTP process in the south of the city of Hefei, China, in dry and rainy weather conditions, as well as the removal effectiveness of MPs in a three-stage process. The collected MPs were quantitatively and qualitatively examined using an Osmosis electron microscope and micro-FTIR. The pollution risk of MPs in WWTP was assessed using the EU classification, labelling and packaging (CLP) standard and the pollution load index (PLI). The findings revealed that the average abundance of fibrous MPs was greatest in WWTP sewage and sludge, 49.3% and 39.7% in dry weather, and 50.1% and 43.2% in rainy weather, respectively. The average distribution of MPs in the 0–500 μm range was highest in WWTP wastewater and sludge, 64.9% and 60.4% in dry weather and 67.9% and 69.0% in rainy weather, respectively. Finally, the overall removal rate was 87.7% and 83.5%. At the same time, it has been demonstrated that MPs with varied compositions are strongly tied to human activities, and environmental conditions (such as rainy weather) also influence their source. In both dry and wet weather, the amount of polymers and the risk score were linked to the pollution risk of MPs in WWTP. In wet weather, the MPS pollution index was more variable. The pollution indices of MPs in row water and tail water were 2.40 and 2.46, respectively, which were heavily contaminated, and 1.0 and 1.2, which were moderately polluted. MPs in dewatered sludge had severely polluted indexes of 3.5 and 3.4, respectively. As a result, there is still MPs efflux or buildup in sludge during and after the WWTP process, which presents an ecological contamination concern.

Abstract Background Deep venous thrombosis (DVT) constitutes a major global disease burden. Endothelial progenitor cells (EPCs) have been described in association with recanalization of venous thrombus. Furthermore, emerging evidence suggests microRNAs are involved in this progression. The goal of this study was to investigate the influence of miR-150 on the behavior of EPCs and its potential contribution in venous thrombosis resolution. Methods We isolated and cultured EPCs from healthy adults. Next, early EPCs or endothelial colony-forming cells (ECFCs or late EPCs) were transfected with miR-150 agomir and antagomir. Gene expression profiles, proliferation, cytokine secretion, and angiogenic capacity of early EPCs and ECFCs were examined. The effects of miR-150 on c-Myb expression and Akt/FOXO1 signaling were also evaluated. Furthermore, a rat model of venous thrombosis was constructed to determine the in vivo function of EPCs. Results Our results showed that miR-150 overexpression in early EPCs significantly promoted differentiation to ECFCs and contributed to proliferation and tube formation. However, suppression of miR-150 in late EPCs inhibited proliferation and tube formation. Moreover, we identified that this progression is regulated by inhibition of c-Myb and activation of the Akt/FOXO1 pathway. Our findings also showed that miR-150 led to the enhanced resolution ability of EPCs in a rat venous thrombosis model. Conclusions In this study, we present a novel mechanism of miRNA-mediated regulation of EPCs and Akt activation in thrombus resolution.

Diabetic cardiomyopathy (DCM) is the leading cause of death in diabetic patients. Folic acid has a protective effect on diabetes-induced cardiomyocyte damage. The aim of this study was to explore the effects of folic acid on cardiomyocytes cultured under high glucose and fat (HGF) conditions and type 2 diabetes mellitus (T2DM) mice, and elucidate the underlying mechanisms. Bioinformatics analysis was used to identify the potential drugs through the Drug-Gene Interaction database. H9C2 cardiomyocytes were cultured with 30 mM glucose and 500 nM palmitic acid in the presence or absence of folic acid or YAP1 inhibitor (verteporfin) or YAP1 siRNA. The cell viability and lactate dehydrogenase (LDH) release were measured using specific assay kits. Pyroptosis was detected by flow cytometry. The concentrations of IL-1β and IL-18 in the supernatants were measured by ELISA. The NLRP3, ASC and caspase-1 mRNA levels were detected by qRT-PCR and that the proteins expression of NLRP3, ASC, cleaved caspase-1 (p10), caspase-1, YAP1, p-YAP1, LATS1 and P-LATS1 were detected by Western blotting. C57BL/6 mice were fed with high fat diet (HFD) combined with streptozotocin (STZ) intraperitoneally to establish a T2DM model, folic acid or PBS treatment for 8 weeks by oral gavage, blood glucose and body weight were measured every 4 weeks, mouse heart tissue was used to detect pyroptosis and hippo signaling pathway related protein expression. We identified 427 differentially expressed genes in the cardiac tissues of high fat diet + streptozotocin mice, among the 30 most significantly DEGs, folic acid was predicted to be the most likely therapeutic drug. Folic acid alleviated HGF-induced cell damage in vitro and in vivo by decreasing activation of the Hippo pathway, as indicated by lower LDH release and increased cell viability, and decreased expression of NLRP3, ASC, cleaved caspase-1, IL-1β, IL-18, p-YAP and p-LATS. Verteporfin or YAP1 siRNA neutralized the protective effect of folic acid by reversing YAP1-induced pyroptosis. Folic acid reduced NLRP3 inflammasome-mediated pyroptosis by down-regulating the Hippo signaling pathway, thereby effectively reducing T2DM-induced damage in H9C2 cells and animals.

In this study, sheep manure fertilizers with different dosages were used for five consecutive years to treat acidified tea plantation soils, and the effects of sheep manure fertilizer on soil pH value, nitrogen transformation, and tea yield and quality were analyzed. The results showed that soil pH value showed an increasing trend after a continuous use of sheep manure fertilizer from 2018 to 2022. After the use of low dosage of sheep manure fertilizer (6 t/hm2–15 t/hm2), tea yield, the content of tea quality indicators (tea polyphenols, theanine, amino acid, and caffeine) and soil ammonium nitrogen content, ammoniating bacteria number, ammoniating intensity, urease activity and protease activity showed increasing trends and were significantly and positively correlated to soil pH value, while the related indexes showed increasing and then decreasing trends after the use of high dosage of sheep manure fertilizer (18 t/hm2). Secondly, the nitrate nitrogen content, nitrifying bacteria number, nitrifying intensity, nitrate reductase activity, and nitrite reductase activity showed decreasing trends after the use of low dosage of sheep manure fertilizer and showed significant negative correlations with soil pH value, while the related indexes showed decreasing trends after the use of high dosage of sheep manure and then increased. The results of principal component and interaction analysis showed that the effects of sheep manure fertilizers with different dosages on tea yield and quality were mainly based on the transformation ability of ammonium nitrogen and nitrate nitrogen in the soil, and the strong transformation ability of ammonium nitrogen and the high ammonium nitrogen content in the soil were conducive to the improvement of tea yield and quality, and vice versa. The results of topsis comprehensive evaluation and analysis showed that the most influential effect on the fertilization effect was the ammonium nitrogen content in the soil and long-term treatment with 15 t/hm2 of sheep manure fertilizer had the highest proximity to the best fertilization effect. This study provided an important practical basis for the remediation and fertilizer management in acidified tea plantation soils.

As a common neurophysiological phenomenon, voluntary muscle fatigue is accompanied by changes in both the central nervous system and peripheral muscles. Considering the effectiveness of the muscle network and the functional corticomuscular coupling (FCMC) in analyzing motor function, muscle fatigue can be analyzed by quantitating the intermuscular coupling and corticomuscular coupling. However, existing coherence-based research on muscle fatigue are limited by the inability of the coherence algorithm to identify the coupling direction, which cannot further reveal the underlying neural mechanism of muscle fatigue. To address this problem, we applied the time-delayed maximal information coefficient (TDMIC) method to quantitate the directional informational interaction in the muscle network and FCMC during a right-hand stabilized grip task. Eight healthy subjects were recruited to the present study. For the muscle networks, the beta-band information flow increased significantly due to muscle fatigue, and the information flow between the synergist muscles were stronger than that between the synergist and antagonist muscles. The information flow in the muscle network mainly flows to flexor digitorum superficialis (FDS), flexor carpi ulnar (FCU), and brachioradialis (BR). For the FCMC, muscle fatigue caused a significant decrease in the beta- and gamma-band bidirectional information flow. Further analysis revealed that the beta-band information flow was significantly stronger in the descending direction [electroencephalogram (EEG) to surface electromyography (sEMG)] than that in the ascending direction (sEMG to EEG) during pre-fatigue tasks. After muscle fatigue, the beta-band information flow in the ascending direction was significantly stronger than that in the descending direction. The present study demonstrates the influence of muscle fatigue on information flow in muscle networks and FCMC. We proposes that beta-band intermuscular and corticomuscular informational interaction plays an adjusting role in autonomous movement completion under muscle fatigue. Directed information flow analysis can be used as an effective method to explore the neural mechanism of muscle fatigue on the macroscopic scale.

Abstract Background Deep venous thrombosis (DVT) of lower extremities is a common thrombotic disease, occurring either in isolation or as a complication of other diseases or procedures. MiR-21 is one of important microRNAs which play critical role in various cellular function. This study aim to determine the effect of miR-21 on endothelial progenitor cells (EPCs) and its role in predicting prognosis of DVT. Methods EPCs was isolated from DVT models and control subjects. miR-21 expression was confirmed by RT-PCR. Potential target mRNA was predicted by bioinformatics analysis. EPCs biological functions were examined by CCK-8 and tube formation assay. Besides, miR-21 expression was determined in DVT patients to investigate the correlation between miR-21 expression and prognosis of DVT. Cox proportional hazard regression analyses were also performed to reveal the risk factors associated with prognosis. Results Here, we found miR-21 was downregulated in EPCs of DVT model rats. Increased miR-21 expression promoted proliferation and angiogenesis of EPCs. Moreover, we demonstrated that FASLG was a target of miR-21 and revealed that FASLG knockdown inhibited function of EPCs. Upregulation of miR-21 led to thrombus resolution in a rat model of venous thrombosis. In addition, lower expression level of miR-21 in DVT patients was associated with an increase of recurrent DVT and post thrombotic syndrome (PTS). Furthermore, Cox proportional hazard regression analyses demonstrated miR-21 expression level as an independent predictor of recurrence of DVT. Conclusions Our data revealed a role of miR-21 in regulating biological function of EPCs and could be a predictor for recurrent DVT or PTS.

Objectives The arthroscopic “lateral gutter drive‐through” (LGDT) sign is reported to diagnose popliteus tendon (PT) injury with high sensitivity and specificity. However, no study has provided a postoperative evaluation of combined posterior cruciate ligament (PCL) and posterolateral corner (PLC) injuries using the LGDT test. Methods From January 2012 to January 2015, a total of 80 consecutive patients who underwent combined PCL reconstruction and PLC surgeries were identified. Fifty eligible patients were included in this study for evaluation with subjective scoring systems, physical examinations, posterior and varus stress radiographs, and second‐look arthroscopic surgeries during hardware removal operation. Results Forty‐nine patients were available for a mean of 31.5 ± 9.3 months follow‐up (range, 24.0–81.0 months). In terms of PLC injury pattern, there were 27 type A, 10 type B, and 12 type C patients, with 21 acute cases and 28 chronic cases. At the final follow‐up, subjective scores were all significantly improved. BackgroundCoronavirus disease 2019 (COVID-19) pandemic has become the most severe global health issue. Abnormal liver functions are frequently reported in these patients. However, liver function abnormality was often overlooked during COVID-19 treatment, and data regarding liver functions after cure of COVID-19 is limited. This study aimed to reveal the changes of liver function tests (LFTs) during hospitalization, and its clinical significance in patients with COVID-19.MethodsIn this retrospective, bi-center study, a total of 158 hospitalized patients diagnosed with COVID-19 in China were included from January 22nd, 2020 to February 20th, 2020. Clinical features, laboratory parameters including LFTs, and treatment data were collected and analyzed. LFTs included alanine transaminase, aspartate aminotransferase, alkaline phosphatase, gamma-glutamyl transferase, and total bilirubin. Patients were considered with abnormal LFTs when any value of these tests was higher than upper limit of normal.ResultsOf 158 patients with COVID-19, 67 (42.41%) patients had abnormal LFTs on admission and another 50 (31.65%) patients developed abnormal LFTs during hospitalization. The incidence of LFTs abnormality in severe COVID-19 cases was significantly higher than non-severe cases. All LFTs in COVID-19 patients were correlated with oxygenation index. There was no statistical difference in treatment between the patients with or without liver test abnormalities. By the time of discharge, there were still 64 (40.50%) patients with abnormal LFTs. Logistic regression analysis identified younger age, hypertension and low lymphocyte counts as independent risk factors for persistent abnormal LFTs during hospitalization.ConclusionLiver function tests abnormality was common in COVID-19 patients and was more prevalent in severe cases than in non-severe cases. A substantial percentage of patients still had abnormal LFTs by the time of discharge.
